    Ben H. Moore, emeritus professor of physics, astronomy and earth sciences at St. Cloud State University, Minnesota, died 7 November 2003, in South Padre Island, Texas. Ben was born 18 March 1921, in Kansas City, Missouri, to Fraser D. and Cora R. (Hawkins) Moore. Though his parents provided a strong guiding influence on Ben's development, Ben's career was impacted most clearly by his work as a student and research assistant for Allen Basset (Ben's father-in-law) at Park College. This relationship turned Ben's early interest in chemistry and biology toward a focus on physics. Ben received his undergraduate degree from Park College where he graduated Phi Delta Kappa. He received a MS in physics from Kansas State University. He also did post-masters work at the University of Kansas, the University of Colorado, the University of Washington and Temple University. In addition to his work as a research assistant, Ben taught at Park College as well as Washington Kansas High School, Wyandotte High School, and Kansas City Kansas Junior College before moving to St. Cloud State University in 1960. He retired from this teaching position in May 1982 but remained involved in activities of his department, including some teaching, through most of his years in retirement. Ben's accomplishments were mainly centered on his teaching. His development of courses and his rapport with students consistently brought praise from both his colleagues and his students. Above all, his work involved innovative development of the curriculum in the sciences at St. Cloud State. Soon after his arrival at the university, Ben took over the fledgling field geology course and continued to shape this offering into a program in earth sciences. The popularity of his classes, which attracted both general students and a growing number of majors, finally enabled the university to establish an earth sciences department in the late 1960's and Ben was the first chair of that department.     This is an aerial view of the deep-sea research submarine 'Ben Franklin' at dock. Named for American patriot and inventor Ben Franklin, who discovered the Gulf Steam, the 50-foot Ben Franklin was built between 1966 and 1968 in Switzerland for deep-ocean explorer Jacques Piccard and the Grumman Aircraft Engineering Corporation. The submersible made a famous 30-day drift dive off the East Coast of the United States and Canada in 1969 mapping the Gulf Stream's currents and sea life, and also made space exploration history by studying the behavior of aquanauts in a sealed, self-contained, self-sufficient capsule for NASA. On July 14, 1969, the Ben Franklin was towed to the high-velocity center of the Stream off the coast of Palm Beach, Florida. With a NASA observer on board, the sub descended to 1,000 feet off of Riviera Beach, Florida and drifted 1,400 miles north with the current for more than four weeks, reemerging near Maine. During the course of the dive, NASA conducted exhaustive analyses of virtually every aspect of onboard life. They measured sleep quality and patterns, sense of humor and behavioral shifts, physical reflexes, and the effects of a long-term routine on the crew. The submarine's record-shattering dive influenced the design of Apollo and Skylab missions and continued to guide NASA scientists as they devised future marned space-flight missions.

    BEN is a primary, chronic tubulointerstitial nephritis characterized with chronic anemia, absence of edema, xantoderma, normal blood pressure and normal findings on the fundus oculi. The disease is distributed in restricted areas in Bulgaria, Romania, Croatia, Bosnia, Former Yugoslavia. Despite numerous studies on genetic and environmental factors and their possible involvement in BEN, its etiopathogenesis still remains elusive. Our recent study aim to elucidate the possible epigenetic component in BEN development. Whole genome DNA array methylation analysis was applied to compare the methylation profiles of male and female BEN patients from endemic regions in Bulgaria and Serbia and healthy controls. All three most prominent candidate genes with aberrations in the epigenetic profile discovered with this study are involved in the inflammatory/immune processes and oncogenesis. These data are in concordance with the reported pathological alterations in BEN. This research supports the role of epigenetic changes in BEN pathology. Exome sequencing of 22.000 genes with Illumina Nextera Exome Enrichment Kit revealed three mutant genes (CELA1, HSPG2, and KCNK5) in BEN patients which encode proteins involved in basement membrane/extracellular matrix and vascular tone, tightly connected to process of angiogenesis. We suggest that an abnormal process of angiogenesis plays a key role in the molecular pathogenesis of BEN. PMID:27442376

    The Agency developed the BEN computer model to calculate the economic benefit a violator derives from delaying or avoiding compliance with environmental statutes. In general, the Agency uses the BEN computer model to assist its own staff in developing settlement penalty figures. While the primary purpose of the BEN model is to calculate the economic benefit of noncompliance, the model may also be used to calculate the after tax net present value of a pollution prevention or mitigation project and to calculate 'cash outs' in Superfund cases. The document, the BEN User's Manual, contains all the formulas that make up the BEN computer model and is freely available to the public upon request.

  3. When autonomy kills: the case of Sami Mbarka Ben Garci.

    Foucault suggested that foreigners and criminals are treated in a particularly unfavourable way by the law. We find arguable support for that proposition in the case of Sami Mbarka Ben Garci. He was a Tunisian Muslim prisoner, charged with rape, held in an Italian prison. He went on a hunger strike, protesting his innocence. He was not force-fed, and was allowed to die. Hunger strikers are commonly force fed. We ask why he was not, and although the reasons in his case are not clear, we suggest that many prisoners perceived as being 'undesirable' (in the sense of being foreigners, or facing particularly serious allegations) are allowed to die (the rhetoric being that their autonomy is being respected), while other prisoners' autonomy would be violated in order to ensure survival. We explore the European and some domestic jurisprudence surrounding force-feeding, and conclude that the law is applied in a worryingly inconsistent way. PMID:23447905

    Jiu huang ben cao (Materia Medica for Relief of Famines) was the first monograph on famines herbal in the history of China, which creates a new research field of edible plants. Around the middle and late 17th century, Jiu huang ben cao was spread to Japan and aroused great attention of famous Japanese herbalists. Thus, all versions of different edition systems were circulated in Japan. Later, some famous Japanese scholar ssuccessively quoted texts of Jiu huang ben cao from the Nong zheng quan shu (Whole book on Agricultural Administration) spread in Japan, and block-printed it as an independent work. As a result, Jiu huang ben cao virtually circulated widely in Japan. PMID:25620360

  5. BenR, a XylS Homologue, Regulates Three Different Pathways of Aromatic Acid Degradation in Pseudomonas putida

    Pseudomonas putida converts benzoate to catechol using two enzymes that are encoded on the chromosome and whose expression is induced by benzoate. Benzoate also binds to the regulator XylS to induce expression of the TOL (toluene degradation) plasmid-encoded meta pathway operon for benzoate and methylbenzoate degradation. Finally, benzoate represses the ability of P. putida to transport 4-hydroxybenzoate (4-HBA) by preventing transcription of pcaK, the gene encoding the 4-HBA permease. Here we identified a gene, benR, as a regulator of benzoate, methylbenzoate, and 4-HBA degradation genes. A benR mutant isolated by random transposon mutagenesis was unable to grow on benzoate. The deduced amino acid sequence of BenR showed high similarity (62% identity) to the sequence of XylS, a member of the AraC family of regulators. An additional seven genes located adjacent to benR were inferred to be involved in benzoate degradation based on their deduced amino acid sequences. The benABC genes likely encode benzoate dioxygenase, and benD likely encodes 2-hydro-1,2-dihydroxybenzoate dehydrogenase. benK and benF were assigned functions as a benzoate permease and porin, respectively. The possible function of a final gene, benE, is not known. benR activated expression of a benA-lacZ reporter fusion in response to benzoate. It also activated expression of a meta cleavage operon promoter-lacZ fusion inserted in an E. coli chromosome. Third, benR was required for benzoate-mediated repression of pcaK-lacZ fusion expression. The benA promoter region contains a direct repeat sequence that matches the XylS binding site previously defined for the meta cleavage operon promoter. It is likely that BenR binds to the promoter region of chromosomal benzoate degradation genes and plasmid-encoded methylbenzoate degradation genes to activate gene expression in response to benzoate. The action of BenR in repressing 4-HBA uptake is probably indirect. PMID:11053377

    Military training, soil texture, and ground cover influence ant communities at Fort Benning, a military installation in west-central Georgia. We sampled 81,237 ground-dwelling ants (47 species in 20 genera) with pitfall traps at 40 sites on a continuum from nearly pristine forest to highly disturbed training areas. We also measured 15 environmental variables related to vegetation and soil. Sites disturbed by military training had fewer trees, less canopy cover, more bare ground, and more compact soils with shallower A-horizons than comparable undisturbed sites. Pheidole bicarinata, Dorymyrmex smithi, and Pogonomyrmex badius dominated the most highly disturbed sites. Competitively submissive myrmicines, such as Aphaenogaster and Crematogaster, and formicines, such as Camponotus and Formica, were abundant in the undisturbed sites. Solenopsis invicta occurred in all but the least disturbed sites. Ant community composition was a useful indicator of disturbance at Fort Benning.

  3. Evaluation of pest vulnerability of 'Benning' soybean value added and insect resistant near isogenic lines.

    Crop enhancement with value added traits may affect vulnerability to insects, and evaluating the susceptibility levels of the various value added traits in elite germplasm would aid in developing integrated pest management strategies. During 2007-2008, five 'Benning' soybean (Glycine max (L.) Merr) lines with different value added nutritional traits and four insect resistant quantitative trait loci (QTL) lines were evaluated in an effort to determine their pest vulnerability under artificial and natural insect pest populations. The lines showed variable susceptibility to lepidopterous insect pests classified as defoliators and stem feeders in replicated greenhouse and field tests. The study was carried out in Athens and Midville, GA. The green cloverworm (Hypena scabra (F.)) was the most common lepidopteran defoliator occurring in the fields. Other caterpillar pests found included the soybean looper (Pseudoplusia includens (Walker)), the bollworm (Helicoverpa zea (Boddie)), and the velvetbean caterpillar (Anticarsia gemmatalis (Hübner)). Data indicated that there was no significantly increased pest susceptibility among the value added cultivars with improved nutritional qualities, with the insect resistant quantitative trait loci lines Benning M and Benning MGH consistently being less susceptible to lepidopterous (Noctuidae) leaf injury.   4. Land Cover Differences in Soil Carbon and Nitrogen at Fort Benning, Georgia

    Land cover characterization might help land managers assess the impacts of management practices and land cover change on attributes linked to the maintenance and/or recovery of soil quality. However, connections between land cover and measures of soil quality are not well established. The objective of this limited investigation was to examine differences in soil carbon and nitrogen among various land cover types at Fort Benning, Georgia. Forty-one sampling sites were classified into five major land cover types: deciduous forest, mixed forest, evergreen forest or plantation, transitional herbaceous vegetation, and barren land. Key measures of soil quality (including mineral soil density, nitrogen availability, soil carbon and nitrogen stocks, as well as properties and chemistry of the O-horizon) were significantly different among the five land covers. In general, barren land had the poorest soil quality. Barren land, created through disturbance by tracked vehicles and/or erosion, had significantly greater soil density and a substantial loss of carbon and nitrogen relative to soils at less disturbed sites. We estimate that recovery of soil carbon under barren land at Fort Benning to current day levels under transitional vegetation or forests would require about 60 years following reestablishment of vegetation. Maps of soil carbon and nitrogen were produced for Fort Benning based on a 1999 land cover map and field measurements of soil carbon and nitrogen stocks under different land cover categories.

    Gram-negative bacteria typically overcome poor permeability of outer membranes through general porins like OmpF and OmpC, which form water-filled transmembrane pores permitting diffusion of hydrophilic molecules with no particular selectivity. Many bacteria lacking such general porins use substrate-specific porins to overcome growth-limiting conditions and facilitate selective transport of metabolites. Exclusive reliance on substrate-specific porins yields lower membrane permeability to small molecules (<600 Da) versus that seen for Escherichia coli. In Pseudomonads, transit of most small molecules across the cell membrane is thought to be mediated by substrate-specific channels of the OprD superfamily. This property explains, at least in part, the high incidence of Pseudomonas aeruginosa antibiotic resistance. High-throughput DNA sequencing of the P. aeruginosa chromosome revealed the presence of 19 genes encoding structurally related, substrate-specific porins (with 30-45% pairwise amino acid sequence identity) that mediate transmembrane passage of small, water-soluble compounds. The OprD superfamily encompasses the eponymous OprD subfamily, which includes 9 P. aeruginosa proteins that convey basic amino acids and carbapenem antibiotics, and the OpdK subfamily, which includes 11 P. aeruginosa proteins that convey aromatic acids and other small aromatic compounds. Genome sequencing of other gram-negative bacteria has revealed additional members of the OprD and OpdK subfamilies in various organisms, including other pseudomonads. Among the many bacteria in which OprD superfamily members have been identified are P. putida, P. fluorescens Pf-5, P. syringae, and Azotobacter vinelandii, all of which share closely related genes that encode the so-called BenF-like porins. In P. putida, benF is part of an operon involved in benzoate catabolism regulated by benR.     BenM and CatM control transcription of a complex regulon for aromatic compound degradation. These Acinetobacter baylyi paralogues belong to the largest family of prokaryotic transcriptional regulators, the LysR-type proteins. Whereas BenM activates transcription synergistically in response to two effectors, benzoate and cis,cis-muconate, CatM responds only to cis,cis-muconate. Here, site-directed mutagenesis was used to determine the physiological significance of an unexpected benzoate-binding pocket in BenM discovered during structural studies. Residues in BenM were changed to match those of CatM in this hydrophobic pocket. Two BenM residues, R160 and Y293, were found to mediate the response to benzoate. Additionally, alteration of these residues caused benzoate to inhibit activation by cis,cis-muconate, positioned in a separate primary effector-binding site of BenM. The location of the primary site, in an interdomain cleft, is conserved in diverse LysR-type regulators. To improve understanding of this important family, additional regulatory mutants were analysed. The atomic-level structures were characterized of the effector-binding domains of variants that do not require inducers for activation, CatM(R156H) and BenM(R156H,T157S). These structures clearly resemble those of the wild-type proteins in their activated muconate-bound complexes. Amino acid replacements that enable activation without effectors reside at protein interfaces that may impact transcription through effects on oligomerization.

    We recently reported that Drosophila Insensitive (Insv) promotes sensory organ development and has activity as a nuclear corepressor for the Notch transcription factor Suppressor of Hairless [Su(H)]. Insv lacks domains of known biochemical function but contains a single BEN domain (i.e., a “BEN-solo” protein). Our chromatin immunoprecipitation (ChIP) sequencing (ChIP-seq) analysis confirmed binding of Insensitive to Su(H) target genes in the Enhancer of split gene complex [E(spl)-C]; however, de novo motif analysis revealed a novel site strongly enriched in Insv peaks (TCYAATHRGAA). We validate binding of endogenous Insv to genomic regions bearing such sites, whose associated genes are enriched for neural functions and are functionally repressed by Insv. Unexpectedly, we found that the Insv BEN domain binds specifically to this sequence motif and that Insv directly regulates transcription via this motif. We determined the crystal structure of the BEN–DNA target complex, revealing homodimeric binding of the BEN domain and extensive nucleotide contacts via α helices and a C-terminal loop. Point mutations in key DNA-contacting residues severely impair DNA binding in vitro and capacity for transcriptional regulation in vivo. We further demonstrate DNA-binding and repression activities by the mammalian neural BEN-solo protein BEND5. Altogether, we define novel DNA-binding activity in a conserved family of transcriptional repressors, opening a molecular window on this extensive gene family. PMID:23468431

  8. Territoriality of feral pigs in a highly persecuted population on Fort Benning, Georgia

    We examined home range behavior of female feral pigs (Sus scrofa) in a heavily hunted population on Fort Benning Military Reservation in west-central Georgia, USA. We used Global Positioning System location data from 24 individuals representing 18 sounders (i.e., F social groups) combined with markrecapture and camera-trap data to evaluate evidence of territorial behavior at the individual and sounder levels. Through a manipulative experiment, we examined evidence for an inverse relationship between population density and home range size that would be expected for territorial animals. Pigs from the same sounder had extensive home range overlap and did not have exclusive core areas. Sounders had nearly exclusive home ranges and had completely exclusive core areas, suggesting that female feral pigs on Fort Benning were territorial at the sounder level but not at the individual level. Lethal removal maintained stable densities of pigs in our treatment area, whereas density increased in our control area; territory size in the 2 areas was weakly and inversely related to density of pigs. Territorial behavior in feral pigs could influence population density by limiting access to reproductive space. Removal strategies that 1) match distribution of removal efforts to distribution of territories, 2) remove entire sounders instead of individuals, and 3) focus efforts where high-quality food resources strongly influence territorial behaviors may be best for long-term control of feral pigs.

  9. Modeling soil quality thresholds to ecosystem recovery at Fort Benning, GA, USA

    The objective of this research was to use a simple model of soil carbon (C) and nitrogen (N) dynamics to predict nutrient thresholds to ecosystem recovery on degraded soils at Fort Benning, Georgia, in the southeastern USA. Artillery, wheeled, and tracked vehicle training at military installations can produce soil disturbance and potentially create barren, degraded soils. Ecosystem reclamation is an important component of natural resource management at military installations. Four factors were important to the development of thresholds to recovery of aboveground biomass on degraded soils: (1) initial amounts of aboveground biomass, (2) initial soil C stocks (i.e., soil quality), (3) relative recovery rates of biomass, and (4) soil sand content. Forests and old fields on soils with varying sand content had different predicted thresholds for ecosystem recovery. Soil C stocks at barren sites on Fort Benning were generally below predicted thresholds to 100% recovery of desired future ecosystem conditions defined on the basis of aboveground biomass. Predicted thresholds to ecosystem recovery were less on soils with more than 70% sand content. The lower thresholds for old field and forest recovery on more sandy soils were apparently due to higher relative rates of net soil N mineralization. Calculations with the model indicated that a combination of desired future conditions, initial levels of soil quality (defined by soil C stocks), and the rate of biomass accumulation determine the predicted success of ecosystem recovery on disturbed soils.

    The article describes the programme „Kids’ Skills“ by the Finnish psychiatrist Ben Furman. „Kids’ Skills“ was developed to address behavioural issues in children. It is based on the assumption that children’s behavioural problems should not be pathologized, but can instead be corrected by learning a corresponding skill. The programme is characterised by its focus on strengths and its humorous and playful approach. The 15 steps of “Kids’ Skills” are intended to identify the specific skill, help generate a learning process and continue motivating the child. The authors describe the steps of the programme using a case study. They also address the limited number of existing studies, which have included a telephone and an online survey of practitioners using the programme, as well as case studies. The results of these studies are discussed with regard to their basis in evidence and practical relevance. Continuing research is recommended and possible implementations are suggested.     Based on the meteorological data over a period of 4 years (1980-3), the macro-environment of BenCat Farm situated in the southern part of Vietnam (27 m above mean sea-level, 11° N and 106° E) was categorized as a “monsoon tropical climate”, due to heavy rainfall (annual mean 2028.96 mm) and about 32% wet days (annual mean 116.52 days) together with high air temperature (annual mean daily temperature 28.58, max. 32.33 and min 24.85° C). April was the hottest (monthly mean >35°C) and January the coldest month (monthly mean <22° C) of the year. The maximum number of wet days were during September and October (mean 18 days.month), whereas the minimum number of wet days were during January and February (mean <1 day/month). The months of December and January at Ben-Cat buffalo farm were categorized as the “comfortable (moderate-Dry) period” as the mean daily temperature was <27° C, while the remaining 10 months of the Calender year (February-November) were categorized as the “hot period” (mean daily temperature >27° C). On the basis of rainfall and the number of wet days, the hot period was further subdivided into a “hot-dry period” (February-April, mean of 1.67 wet days/month and mean rainfall 19.43 mm/month) and a “hot-humid period” (May-November, mean of 15.57 wet days/month and mean rainfall 276.28 mm/month).

    Recently, the BEN (BANP, E5R, and NAC1) domain was recognized as a new class of conserved DNA-binding domain. The fly genome encodes three proteins that bear only a single BEN domain (“BEN-solo” factors); namely, Insensitive (Insv), Bsg25A (Elba1), and CG9883 (Elba2). Insv homodimers preferentially bind CCAATTGG palindromes throughout the genome to mediate transcriptional repression, whereas Bsg25A and Elba2 heterotrimerize with their obligate adaptor, Elba3 (i.e., the ELBA complex), to recognize a CCAATAAG motif in the Fab-7 insulator.     Recently, the BEN (BANP, E5R, and NAC1) domain was recognized as a new class of conserved DNA-binding domain. The fly genome encodes three proteins that bear only a single BEN domain ("BEN-solo" factors); namely, Insensitive (Insv), Bsg25A (Elba1), and CG9883 (Elba2). Insv homodimers preferentially bind CCAATTGG palindromes throughout the genome to mediate transcriptional repression, whereas Bsg25A and Elba2 heterotrimerize with their obligate adaptor, Elba3 (i.e., the ELBA complex), to recognize a CCAATAAG motif in the Fab-7 insulator.   19. Full-Length Structures of BenM and Two Variants Reveal Different Oligomerization Schemes for LysR-Type Transcriptional Regulators

    BenM, a LysR-type transcriptional regulator (LTTR) from the bacterium Acinetobacter baylyi, responds synergistically to benzoate and cis,cis-muconate. With these effectors, BenM activates gene expression during benzoate consumption. Without effectors, BenM represses transcription. Here, X-ray crystallography was used to determine the full-length structures of BenM and two variants that activate transcription without benzoate or cis,cis-muconate: BenM(R156H) and BenM(E226K). Previous studies indicate that these regulators function as tetramers. Here, interconnections between subunits in the crystals prevented the formation of a closed oligomer and highlighted the inherent flexibility of this multidomain regulator. Nevertheless, analysis of subunit interfaces suggested the functional significance of key interactions. The structures of BenM and its variants were nearly identical, implying that transcriptional differences rely on factors beyond major conformational changes defined solely by sequence. Comparisons of BenM with other LTTRs, including unpublished structures in the Protein Data Bank, revealed extensive variation in the relative orientations of DNA-binding domains (DBDs) and effector-binding domains (EBDs). To form dimers, different LTTRs used similar interfaces between two EBDs, each containing two subdomains: EBD-I and EBD-II. Surprisingly, the dimers used three substantially different schemes to form higher-order oligomers. In one scheme used by BenM, oligomer assembly involved contacts between the EBD-II regions and the DBD regions of adjacent subunits. In another scheme, there were no contacts between the EBDs; only the DBDs were involved in tetramer formation. In the third scheme, the oligomer interface involved DBD and EBD-I/EBD-II contacts. These diverse schemes demonstrate novel variation in the oligomeric structures of individual LTTRs within this large and important family.

  20. Effects of Heavy, Tracked-Vehicle Disturbance on Forest Soil Properties at Fort Benning, Georgia

    The purpose of this report is to describe the effects of heavy, tracked-vehicle disturbance on various measures of soil quality in training compartment K-11 at Fort Benning, Georgia. Predisturbance soil sampling in April and October of 2002 indicated statistically significant differences in soil properties between upland and riparian sites. Soil density was less at riparian sites, but riparian soils had significantly greater C and N concentrations and stocks than upland soils. Most of the C stock in riparian soils was associated with mineral-associated organic matter (i.e., the silt + clay fraction physically separated from whole mineral soil). Topographic differences in soil N availability were highly dependent on the time of sampling. Riparian soils had higher concentrations of extractable inorganic N than upland soils and also exhibited significantly greater soil N availability during the spring sampling. The disturbance experiment was performed in May 2003 by driving a D7 bulldozer through the mixed pine/hardwood forest. Post-disturbance sampling was limited to upland sites because training with heavy, tracked vehicles at Fort Benning is generally confined to upland soils. Soil sampling approximately one month after the experiment indicated that effects of the bulldozer were limited primarily to the forest floor (O-horizon) and the surface (0-10 cm) mineral soil. O-horizon dry mass and C stocks were significantly reduced, relative to undisturbed sites, and there was an indication of reduced mineral soil C stocks in the disturbance zone. Differences in the surface (0-10 cm) mineral soil also indicated a significant increase in soil density as a result of disturbance by the bulldozer. Although there was some tendency for greater soil N availability in disturbed soils, the changes were not significantly different from undisturbed controls.     The objective of this research was to use a simple model of soil C and N dynamics to predict nutrient thresholds to ecosystem recovery on degraded soils at Fort Benning, Georgia, in the southeastern USA. The model calculates aboveground and belowground biomass, soil C inputs and dynamics, soil N stocks and availability, and plant N requirements. A threshold is crossed when predicted soil N supplies fall short of predicted N required to sustain biomass accrual at a specified recovery rate. Four factors were important to development of thresholds to recovery: (1) initial amounts of aboveground biomass, (2) initial soil C stocks (i.e., soil quality), (3) relative recovery rates of biomass, and (4) soil sand content. Thresholds to ecosystem recovery predicted by the model should not be interpreted independent of a specified recovery rate. Initial soil C stocks influenced the predicted patterns of recovery by both old field and forest ecosystems. Forests and old fields on soils with varying sand content had different predicted thresholds to recovery. Soil C stocks at barren sites on Fort Benning generally lie below predicted thresholds to 100% recovery of desired future ecosystem conditions defined on the basis of aboveground biomass (18000 versus 360 g m{sup -2} for forests and old fields, respectively). Calculations with the model indicated that reestablishment of vegetation on barren sites to a level below the desired future condition is possible at recovery rates used in the model, but the time to 100% recovery of desired future conditions, without crossing a nutrient threshold, is prolonged by a reduced rate of forest growth. Predicted thresholds to ecosystem recovery were less on soils with more than 70% sand content. The lower thresholds for old field and forest recovery on more sandy soils are apparently due to higher relative rates of net soil N mineralization in more sandy soils.   2. Assessing the Health Impacts of Air Pollution Regulations Using BenMAP, the Environmental Benefits Mapping and Analysis Program

    The U.S. EPA Office of Air and Radiation has developed BenMAP, the environmental Benefits Mapping and Analysis Program, a new software tool for estimating the health and environmental impacts of environmental regulations. BenMAP is the US EPA's premier tool for estimating benefits associated with air pollution reduction strategies, and has recently been used in evaluating US EPA's Proposed Non-Road Diesel Vehicle Standards and the proposed Clear Skies Act legislation. BenMAP is a geographic information system (GIS) that uses modeled and monitored air quality data combined with population forecasts to develop estimates of changes in community level exposure to ambient environmental pollution (currently ambient air pollution, e.g. ozone and PM). These estimated changes in exposure to ambient pollution are used as inputs to concentration-response functions derived from the epidemiological literature, along with data on baseline incidence of health effects, i.e. county level age and cause specific mortality rates. The resulting point estimates of changes in incidence of health effects, along with their associated uncertainty distributions (currently based on reported standard errors in the literature), are then multiplied by economic unit values (represented by distributions), i.e. the cost of a hospital admission, to derive dollar estimates of health benefits. BenMAP is unique in that it uses highly detailed census and health data matched with spatially detailed environmental quality data to estimate health benefits. The program can also provide estimates of the uncertainty associated with the estimated benefits. Other features of the program include the ability to pool results from multiple studies of the same endpoint, using fixed or random effects weights, or user supplied weights.     In eukaryotes, higher order chromatin structure governs crucial cellular processes including DNA replication, transcription and post-transcriptional gene regulation. Specific chromatin-interacting proteins play vital roles in the maintenance of chromatin structure. We have identified BEND3, a quadruple BEN domain-containing protein that is highly conserved amongst vertebrates. BEND3 colocalizes with HP1 and H3 trimethylated at K9 at heterochromatic regions in mammalian cells. Using an in vivo gene locus, we have been able to demonstrate that BEND3 associates with the locus only when it is heterochromatic and dissociates upon activation of transcription. Furthermore, tethering BEND3 inhibits transcription from the locus, indicating that BEND3 is involved in transcriptional repression through its interaction with histone deacetylases and Sall4, a transcription repressor. We further demonstrate that BEND3 is SUMOylated and that such modifications are essential for its role in transcriptional repression. Finally, overexpression of BEND3 causes premature chromatin condensation and extensive heterochromatinization, resulting in cell cycle arrest. Taken together, our data demonstrate the role of a novel heterochromatin-associated protein in transcriptional repression. PMID:21914818

  5. Geomorphological activity during the Lateglacial in the corrie complex of Ben Hope, NW Highlands, Scotland

    The Lateglacial Period in Europe is characterised by pronounced climatic fluctuations and encompasses the time from Devensian (Weichselian) ice sheet retreat to the beginning of the Holocene. The most notable period in terms of geomorphological activity is the Younger Dryas (12.7-11.5 ka BP), during which an ice cap centred on the main watershed covered c. 350 km2 of the NW Highlands (Lukas and Bradwell, 2009); only one individual corrie glacier existed on an isolated mountain massif (Arkle) to the north of the main watershed in this area during the Younger Dryas (Carr et al., 2009). The study site at Ben Hope, c. 10 km east of the main ice cap, is the most prominent and highest mountain in this area (927 m asl). At this site, Sissons (1977) suggested the presence of small glaciers in the three corries on the eastern side of the mountain, based on the identification of lateral moraines and a series of hummocky moraines. However, recent mapping of the field evidence by the present authors suggests that the landform assemblages in the corrie might have been misinterpreted from aerial photographs: The undulating ground at the toe of the corrie (A) resembles openwork blocks with a-axes of up to 5 m that are angular to subangular. The corrie headwall contains (B) an elongated accumulation of large blocks (a-axes up to 10 m) between 644 m and 740 m a.s.l. This feature displays a steep front (35-40°) and a central depression that is elongated downslope. Above this feature, a truncated spur displaying antiscarps can be observed (C). The centre of the corrie is occupied by a lochan and two coherent, 15 m-high, blocky ridges that are covered by up to 2 m of peat (D). The evidence is at present interpreted as (A) undulating rockfall debris, possibly on to former glacier ice which melted during retreat, due to the presence of depressions and lack of spatial order.     We develop a three-dimensional lattice Boltzmann (LB) model accounting for directional interactions between water-like molecules, based on the so-called Ben-Naim (BN) potential [A. Ben-Naim, Molecular Theory of Water and Aqueous Solutions: Part I: Understanding Water (World Scientific Publishing Company, 2010); A. Ben-Naim, "Statistical mechanics of `waterlike' particles in two dimensions. I. Physical model and application of the Percus-Yevick equation," J. Chem. Phys. 54, 3682 (1971)], 10.1063/1.1675414. The water-like molecules are represented by rigid tetrahedra, with two donors and two acceptors at the corners and interacting with neighboring tetrahedra, sitting on the nodes of a regular lattice. The tetrahedra are free to rotate about their centers under the drive of the torque arising from the interparticle potential. The orientations of the water molecules are evolved in time via an overdamped Langevin dynamics for the torque, which is solved by means of a quaternion technique. The resulting advection-diffusion-reaction equation for the quaternion components is solved by a LB method, acting as a dynamic minimizer for the global energy of the fluid. By adding thermal fluctuations to the torque equation, the model is shown to reproduce some microscopic features of real water, such as an average number of hydrogen bonds per molecules (HBs) between 3 and 4, in a qualitative agreement with microscopic water models. Albeit slower than a standard LB solver for ordinary fluids, the present scheme opens up potentially far-reaching scenarios for multiscale applications based on a coarse-grained representation of the water solvent.

The extant first volume of the original block - printed edition, revised by Lü Fu, was collected in the Library of China Academy of Military Medical Science. There is a preface written by Jie Hong, a Director Ministry of Techndogy in 1366 at the front of this volume. The red seal in the first page showed that it had been collected by Yun Xiang, Pan Zuyin et al. It is highly possible that this volume is the only existing copy. PMID:12015054

    As a result of globalization, the number of multinational mergers and acquisitions across the globe has grown steadily over the years. In this article I have conducted research based on two key studies: the failure and success of two companies, BenQ and Foxconn, through the mergers and acquisitions. I gain the conclusion that effective cross-border culture communication is an absolute, decisive factor in building successful relationships among companies; enterprises must develop a "cross-border culture communication motivation function" from both the acquiring and the acquired parts of the partnership. It is only then, that enterprises can exert their market and financial synergy influence to the fullest extents.

  9. Federal land management, carbon sequestration, and climate change in the Southeastern U.S.: a case study with fort benning

    Land use activities can have a major impact on the temporal trendsandspatialpatternsofregionalland-atmosphereexchange of carbon. Federal lands generally have substantially different land management strategies from surrounding areas, and the carbon consequences have rarely been quantified and assessed. Using the Fort Benning Installation as a case study, we used the General Ensemble biogeochemical Modeling System (GEMS) to simulate and compare ecosystem carbon sequestration between the U.S. Army's Fort Benning and surrounding areas from 1992 to 2050. Our results indicate that the military installation sequestered more carbon than surrounding areas from 1992 to 2007 (76.7 vs 18.5 g C m-2 yr-1), and is projected to continue sequestering more carbon from 2008 to 2050 (75.7 vs 25.6 g C m-2 yr-1), mostly because of the proactive management approaches adopted on military training lands. Our results suggest that federal lands might play a positive and important role in sequestering and conserving atmospheric carbon because some anthropogenic disturbances (e.g., urbanization, forest harvesting, and agriculture) can be minimized or prevented on federal lands

    The nucleotide sequences of the Acinetobacter calcoaceticus benABC genes encoding a multicomponent oxygenase for the conversion of benzoate to a nonaromatic cis-diol were determined. The enzyme, benzoate 1,2-dioxygenase, is composed of a hydroxylase component, encoded by benAB, and an electron transfer component, encoded by benC. Comparison of the deduced amino acid sequences of BenABC with related sequences, including those for the multicomponent toluate, toluene, benzene, and naphthalene 1,2-dioxygenases, indicated that the similarly sized subunits of the hydroxylase components were derived from a common ancestor. Conserved cysteine and histidine residues may bind a [2Fe-2S] Rieske-type cluster to the alpha-subunits of all the hydroxylases. Conserved histidines and tyrosines may coordinate a mononuclear Fe(II) ion. The less conserved beta-subunits of the hydroxylases may be responsible for determining substrate specificity. Each dioxygenase had either one or two electron transfer proteins. The electron transfer component of benzoate dioxygenase, encoded by benC, and the corresponding protein of the toluate 1,2-dioxygenase, encoded by xylZ, were each found to have an N-terminal region which resembled chloroplast-type ferredoxins and a C-terminal region which resembled several oxidoreductases. These BenC and XylZ proteins had regions similar to certain monooxygenase components but did not appear to be evolutionarily related to the two-protein electron transfer systems of the benzene, toluene, and naphthalene 1,2-dioxygenases.     This paper examines the history of weather and climate since 1706 along three intertwined analytical axes: technology (killer apps), science (cutting edges), and social issues (tipping points). For example, Franklin's best-known killer app, the lightning rod, gains added significance when seen in light of his cutting edge contributions to the science of electricity, his lifelong promotion of useful knowledge, and the societal tipping point his work triggered in our relationship to the sky. Subsequently, other major tipping points and conceptual shifts followed the introduction of telegraphy, radio, television, digital computers, and rocketry into meteorology. Following an analysis of the career and contributions of Benjamin Franklin (1706-1790), the paper examines later historical moments and watersheds, not merely in retrospect, but from the perspective of leading participants at the time. It focuses on technologies of significant promise, especially those involving electro- magnetism, up to and including the dawn of the twenty-first century, and asks playfully, "What would Ben think?"

  17. Expression of the cell adhesion proteins BEN/SC1/DM-GRASP and TAG-1 defines early steps of axonogenesis in the human spinal cord.

    We have studied the expression pattern of two cell adhesion proteins of the immunoglobin (Ig) superfamily, BEN/SC1/DM-GRASP (BEN) and the transient axonal glycoprotein TAG-1, during the development of the human nervous system. This study was performed by immunocytochemistry on sections of human embryos ranging from 4 to 13 weeks postconception. The overall distribution of the two proteins during development is very similar to that reported in other vertebrate species, but several important differences have been observed. Both proteins exhibit a transient expression on selected neuronal populations, which include the motor and the sensory neurons. In addition, BEN was also detected on virtually all neurons derived from the neural crest as well as in nonneuronal tissues. A major difference of expression with the chick embryo is that, in the motor neurons, BEN expression was not observed at early stages of development, thus arguing against a role of this molecule in pathfinding and fasciculation. BEN was observed to be restricted to subsets of motor neurons, such as the medial column at the upper limb level. Expression was also detected in a laterodorsal population of the ventral horn cells, which are likely to correspond to migrating preganglionic neurons that originate from the motor pool at the thoracic level. TAG-1 was found on commissural neurons and weakly on the sympathetic neurons; it was also detected on restricted nonneuronal populations. In addition, we observed TAG-1 expression in fibers that could correspond either to subsets of dorsal root ganglia (DRGs) central afferences (including the Ia fibers) or to the axons of association interneurons and in scattered motoneurons likely to correspond either to preganglionic neurons, to gamma-motoneurons, or to late-born motoneurons.   18. A Honeycomb BeN2 Sheet with a Desirable Direct Band Gap and High Carrier Mobility.

    Using global particle-swarm optimization method, we report, for the first time, a BeN2 sheet (h-BeN2) with a graphene-like honeycomb lattice but displaying a direct band gap. Symmetry group analysis indicates that the dipole transition is allowed between the conduction band minimum and the valence band maximum. Although the direct band gap of 2.23 eV is close to that (2.14 eV) of MoS2 sheet, the h-BeN2 sheet has additional advantages: the direct band gap feature of the h-BeN2 sheet is quite insensitive to the layer stacking pattern and layer number, in contrast to the well-known direct-to-indirect band gap transition observed in TMDs and h-BN sheets. When rolled up, all the resulting h-BeN2 nanotubes have direct band gaps independent of chirality and diameter. Furthermore, the intrinsic acoustic-phonon-limited carrier mobility of the h-BeN2 sheet can reach ∼10(5) cm(2) V(-1) s(-1) for electron and ∼10(4) cm(2) V(-1) s(-1) for hole, which are higher than that of MoS2 and black phosphorus.     The responses of species and ecosystems to future climate changes will present challenges for conservation and natural resource managers attempting to maintain both species populations and essential habitat. This report describes projected future changes in climate and vegetation for three study areas surrounding the military installations of Fort Benning, Georgia, Fort Hood, Texas, and Fort Irwin, California. Projected climate changes are described for the time period 2070–2099 (30-year mean) as compared to 1961–1990 (30-year mean) for each study area using data simulated by the coupled atmosphere-ocean general circulation models CCSM3, CGCM3.1(T47), and UKMO-HadCM3, run under the B1, A1B, and A2 future greenhouse gas emissions scenarios. These climate data are used to simulate potential changes in important components of the vegetation for each study area using LPJ, a dynamic global vegetation model, and LPJ-GUESS, a dynamic vegetation model optimized for regional studies. The simulated vegetation results are compared with observed vegetation data for the study areas. Potential effects of the simulated future climate and vegetation changes for species and habitats of management concern are discussed in each study area, with a particular focus on federally listed threatened and endangered species.

  20. Similarities between the antABC-Encoded Anthranilate Dioxygenase and the benABC-Encoded Benzoate Dioxygenase of Acinetobacter sp. Strain ADP1

    Acinetobacter sp. strain ADP1 can use benzoate or anthranilate as a sole carbon source. These structurally similar compounds are independently converted to catechol, allowing further degradation to proceed via the β-ketoadipate pathway. In this study, the first step in anthranilate catabolism was characterized. A mutant unable to grow on anthranilate, ACN26, was selected. The sequence of a wild-type DNA fragment that restored growth revealed the antABC genes, encoding 54-, 19-, and 39-kDa proteins, respectively. The deduced AntABC sequences were homologous to those of class IB multicomponent aromatic ring-dihydroxylating enzymes, including the dioxygenase that initiates benzoate catabolism. Expression of antABC in Escherichia coli, a bacterium that normally does not degrade anthranilate, enabled the conversion of anthranilate to catechol. Unlike benzoate dioxygenase (BenABC), anthranilate dioxygenase (AntABC) catalyzed catechol formation without requiring a dehydrogenase. In Acinetobacter mutants, benC substituted for antC during growth on anthranilate, suggesting relatively broad substrate specificity of the BenC reductase, which transfers electrons from NADH to the terminal oxygenase. In contrast, the benAB genes did not substitute for antAB. An antA point mutation in ACN26 prevented anthranilate degradation, and this mutation was independent of a mucK mutation in the same strain that prevented exogenous muconate degradation. Anthranilate induced expression of antA, although no associated transcriptional regulators were identified. Disruption of three open reading frames in the immediate vicinity of antABC did not prevent the use of anthranilate as a sole carbon source. The antABC genes were mapped on the ADP1 chromosome and were not linked to the two known supraoperonic gene clusters involved in aromatic compound degradation. PMID:9721284

    Ben Shabat et al. (Int J Biometeorol 56(4):639-51, 2013) present revised charts for wind chill equivalent temperatures (WCET) and facial skin temperatures (FST) that differ significantly from currently accepted charts. They credit these differences to their more sophisticated calculation model and to the human-based equation that it used for finding the convective heat transfer coefficient (Ben Shabat and Shitzer, Int J Biometeorol 56:639-651, 2012). Because a version of the simple model that was used to create the current charts accurately reproduces their results when it uses the human-based equation, the differences that they found must be entirely due to this equation. In deriving it, Ben Shabat and Shitzer assumed that all of the heat transfer from the surface of their cylindrical model was due to forced convection alone. Because several modes of heat transfer were occurring in the human experiments they were attempting to simulate, notably radiation, their coefficients are actually total external heat transfer coefficients, not purely convective ones, as the calculation models assume. Data from the one human experiment that used heat flux sensors supports this conclusion and exposes the hazard of using a numerical model with several adjustable parameters that cannot be measured. Because the human-based equation is faulty, the values in the proposed charts are not correct. The equation that Ben Shabat et al. (Int J Biometeorol 56(4):639-51, 2013) propose to calculate WCET should not be used.

  8. Predicted Effects of Prescribed Burning and Timber Management on Forest Recovery and Sustainability at Fort Benning, Georgia

    The objective of this work was to use a simple compartment model of soil carbon (C) and nitrogen (N) dynamics to predict forest recovery on degraded soils and forest sustainability, following recovery, under different regimes of prescribed fire and timber management. This report describes the model and a model-based analysis of the effect of prescribed burning and forest thinning or clearcutting on stand recovery and sustainability at Fort Benning, GA. I developed the model using Stella{reg_sign} Research Software (High Performance Systems, Inc., Hanover, NH) and parameterized the model using data from field studies at Fort Benning, literature sources, and parameter fitting. The model included (1) a tree biomass submodel that predicted aboveground and belowground tree biomass, (2) a litter production submodel that predicted the dynamics of herbaceous aboveground and belowground biomass, (3) a soil C and N submodel that predicted soil C and N stocks (to a 30 cm soil depth) and net soil N mineralization, and (4) an excess N submodel that calculated the difference between predicted plant N demands and soil N supplies. There was a modeled feedback from potential excess N (PEN) to tree growth such that forest growth was limited under conditions of N deficiency. Two experiments were performed for the model-based analysis. In the first experiment, forest recovery from barren soils was predicted for 100 years with or without prescribed burning and with or without timber management by thinning or clearcutting. In the second experiment, simulations began with 100 years of predicted forest growth in the absence of fire or harvesting, and sustainability was predicted for a further 100 years either with or without prescribed burning and with or without forest management.     As an American icon, Benjamin Franklin is often portrayed as wise and canny in business and politics, earnestly pursuing and extolling diligence, sensible conduct, pragmatism, and good works. Also legendary are some of his inventions, particularly the lightning rod, bifocals, and an efficient wood-burning stove. The iconic image is misleading in major respects. Today, surprisingly few people appreciate that, in the 18th century, Franklin was greatly esteemed throughout Europe as a scientist (termed then a "natural philosopher.") He was hailed as the "Newton of Electricity." Indeed, until Franklin, electricity seemed more mysterious than had gravity in Newton's time, and lightning was considered the wrath of God. By his own account, Franklin's studies of electricity and many other phenomena were prompted not by practical aims, but by his playful curiosity--which often became obsessive. Also not generally appreciated is the importance of Franklin's scientific reputation in enhancing his efforts to obtain French support for the American Revolution.

    Dengue has emerged in Vietnam 50 years ago and since has become endemo-epidemic throughout the whole country. Each year, major epidemics of dengue fever (DF) and dengue hemorrhagic fever (DHF) hit South Vietnam during the rainy season, causing significant morbidity and mortality, especially among young children. The only preventive measure is vector control, but it is often implemented too late or indiscriminately. The aim of this study was to investigate, in the pre-epidemic stage, the existence of significant changes in vector indices, which will predict DF/DHF outbreaks. We conducted a descriptive transversal study, repeated once a month for four months (March to June) in the village of Locthuan (province Ben Tre) in the Mekong's delta. Adult mosquitoes were caught in 30 houses, and larvae were collected in water holding containers of 50 houses. The houses were randomly selected. Vector densities were calculated according to the indices recommended by WHO. Virological analysis was carried out on lots of female Aedes and larvae in order to determine viral infection rates. Catches of adult mosquitoes collected 496 specimens including 329 Aedes, 139 Culex and 28 Anopheles. Aedes aegypti was present in 63% of visited homes that is an average density of 1.8 mosquitoes per house. The increase in imaginal indices during the 4 months was not significant. The survey of breeding sites of Ae. aegypti identified 1292 water containers in which 71,569 larval specimens were collected. The values of house index, container index [CI] and Breteau index [BI] increased each month, the latter from 166 to 442. This increase was significant for CI and BI. Breeding sites were mostly intra-home, mainly consisting of large and small ceramic jars. Larval density of Ae. aegypti in the containers also increased significantly over the 4 months.     Ben cao tu jing (Illustrated Classic of Materia Medica) is the earliest extant atlas book of materia medica in China, with 933 attached drawings. Among them, the largest portion, amounting to 670, are herbaceous plants, mostly commonly used, with definite marks of the origin producing areas, distributed across 149 administrative divisions(prefectures and counties) of the Song Dynasty, most of them in Northern area which were distributed denser than those in Southern area. The densest ones were located in Southern Shanxi, Eastern Sichuan and Eastern Anhui.     The xylXYZ DNA region is carried on the TOL pWW0 plasmid in Pseudomonas putida and encodes a benzoate dioxygenase with broad substrate specificity. The DNA sequence of the region is presented and compared with benABC, the chromosomal region encoding the benzoate dioxygenase of Acinetobacter calcoaceticus. Corresponding genes from the two biological sources share common ancestry: comparison of aligned XylX-BenA, XylY-BenB, and XylZ-BenC amino acid sequences revealed respective identities of 58.3, 61.3, and 53%. The aligned genes have diverged to assume G+C contents that differ by 14.0 to 14.9%. Usage of the unusual arginine codons AGA and AGG appears to have been selected in the P. putida xylX gene as it diverged from the ancestor it shared with A. calcoaceticus benA. Homologous A. calcoaceticus and P. putida genes exhibit different patterns of DNA sequence repetition, and analysis of one such pattern suggests that mutations creating different DNA slippage structures made a significant contribution to the evolutionary divergence of xylX.     The ancient carving workers have made a great contribution to the xylographic printing art in ancient China, so the studies on them are significant for a survey of ancient Chinese printing history, and for the identification of ancient Chinese books edition. Zheng lei ben cao published by Huiming Xuan (Huiming Sanctum) in the Jin and Yuan dynasties, which is the earliest extant edition of Zhenghe version system of Zheng lei ben cao and has important literature value. Thirty carving workers were involved in its printing process. On the whole, these workers had a relatively high technique and completed a remarkably fine work. In addition to lettering, 28 persons of them also made a total of 536 pages with 900 exquisite engraving illustrations on Chinese materia medica included in this book. Because of the high levels on carving, this precious book has been the representative of Pingshui edition, which has a great reputation but has very few works now. PMID:23363847

  16. Chinese herbal medicine formula Gu-Ben-Fang-Xiao-Tang attenuates airway inflammation by modulating Th17/Treg balance in an ovalbumin-induced murine asthma model

    Gu-Ben-Fang-Xiao-Tang (GBFXT) is a traditional Chinese medicine formula consisting of 11 medicinal plants, which has been used in the treatment of asthma. The present study aimed to determine the protective effects and the underlying mechanisms of GBFXT on ovalbumin (OVA)-induced allergic inflammation in a mouse model of allergic asthma. A total of 50 mice were randomly assigned to the following five experimental groups: Normal, model, montelukast (2.6 mg/kg), 12 g/kg GBFXT and 36 g/kg GBFXT groups. Airway responsiveness was measured using the forced oscillation technique, while differential cell count in the bronchoalveolar lavage fluid (BALF) was measured by Wright-Giemsa staining. Histological assessment was performed by hematoxylin and eosin staining, while BALF levels of Th17/Treg cytokines were measured by enzyme-linked immunosorbent assay, and the proportions of Th17 and Treg cells were evaluated by flow cytometry. The results showed that GBFXT suppressed airway hyperresponsiveness during methacholine-induced constriction, reduced the percentage of leukocytes and eosinophils, and resulted in decreased absolute neutrophil infiltration in lung tissue. In addition, GBFXT treatment significantly decreased the IL-17A cytokine level and increased the IL-10 cytokine level in the BALF. Furthermore, GBFXT significantly suppressed Th17 cells and increased Treg cells in asthmatic mice. In conclusion, the current results demonstrated that GBFXT may effectively inhibit the progression of airway inflammation in allergic asthma, partially by modulating the Th17/Treg cell balance.

The air quality in Guangzhou typically worsens in November due to unfavorable meteorological conditions for pollutant dispersion. During the Guangzhou Asian Games in November 2010, the Guangzhou government carried out a number of emission control measures that significantly improved the air quality. In this paper, we estimated the acute health outcome changes related to the air quality improvement during the 2010 Guangzhou Asian Games using a next-generation, fully-integrated assessment system for air quality and health benefits. This advanced system generates air quality data by fusing model and monitoring data instead of using monitoring data alone, which provides more reliable results. The air quality estimates retain the spatial distribution of model results while calibrating the value with observations. The results show that the mean PM2.5 concentration in November 2010 decreased by 3.5 μg/m(3) compared to that in 2009 due to the emission control measures. From the analysis, we estimate that the air quality improvement avoided 106 premature deaths, 1869 cases of hospital admission, and 20,026 cases of outpatient visits. The overall cost benefit of the improved air quality is estimated to be 165 million CNY, with the avoided premature death contributing 90% of this figure. The research demonstrates that BenMAP-CE is capable of assessing the health and cost benefits of air pollution control for sound policy making. PMID:27090690

    In recent years, levels of particulate matter (PM) air pollution in China have been relatively high, exceeding China's Class II standards in many cities and impacting public health. This analysis takes Chinese health impact functions and underlying health incidence, applies 2010-2012 modeled and monitored PM air quality data, and estimates avoided cases of mortality and morbidity in Shanghai, assuming achievement of China's Class II air quality standards. In Shanghai, the estimated avoided all cause mortality due to PM10 ranged from 13 to 55 cases per day and from 300 to 800 cases per year. The estimated avoided impact on hospital admissions due to PM10 ranged from 230 cases to 580 cases per day and from 5400 to 7900 per year. The estimated avoided impact on all cause mortality due to PM2.5 ranged from 6 to 26 cases per day and from 39 to 1400 per year. The estimated impact on all cause mortality of a year exposure to an annual or monthly mean PM2.5 concentration ranged from 180 to 3500 per year. In Shanghai, the avoided cases of all cause mortality had an estimated monetary value ranging from 170 million yuan (1 US dollar=4.2 yuan Purchasing Power Parity) to 1200 million yuan. Avoided hospital admissions had an estimated value from 20 to 43 million yuan. Avoided emergency department visits had an estimated value from 5.6 million to 15 million yuan. Avoided outpatient visits had an estimated value from 21 million to 31 million yuan. In this analysis, available data were adequate to estimate avoided health impacts and assign monetary value. Sufficient supporting documentation was available to construct and format data sets for use in the United States Environmental Protection Agency's health and environmental assessment model, known as the Environmental Benefits Mapping and Analysis Program - Community Edition ("BenMAP-CE").     The NAHB Research Center partnered with production builder Winchester/Camberley Homes to build a DOE Building America New Construction Test House (NCTH). This single family, detached house, located in the mixed-humid climate zone of Silver Spring, MD, was completed in June 2011. The primary goal for this house was to improve energy efficiency by 30% over the Building America B10 benchmark by developing and implementing an optimized energy solutions package design that could be cost effectively and reliably constructed on a production basis using quality management practices. The intent of this report is to outline the features of this house, discuss the implementation of the energy efficient design, and report on short-term testing results. During the interactive design process of this project, numerous iterations of the framing, air sealing, insulation, and space conditioning systems were evaluated for energy performance, cost, and practical implementation. The final design featured numerous advanced framing techniques, high levels of insulation, and the HVAC system entirely within conditioned space. Short-term testing confirmed a very tight thermal envelope and efficient and effective heating and cooling. In addition, relevant heating, cooling, humidity, energy, and wall cavity moisture data will be collected and presented in a future long-term report.

    To assist in understanding sediment loadings and the management of water resources, a bathymetric survey was conducted in the part of Lake Linganore between Boyers Mill Road Bridge and Bens Branch in Frederick County, Maryland. The bathymetric survey was performed in January 2012 by the U.S. Geological Survey, in cooperation with the City of Frederick and Frederick County. A separate, but related, field effort to collect 18 sediment cores was conducted in March and April 2012. Depth and location data from the bathymetric survey and location data for the sediment cores were compiled and edited by using geographic information system (GIS) software. A three-dimensional triangulated irregular network (TIN) model of the lake bottom was created to calculate the volume of stored water in the reservoir. Large-scale topographic maps of the valley prior to inundation in 1972 were provided by the Frederick County Division of Utilities and Solid Waste Management and digitized for comparison with current (2012) conditions in order to calculate sediment volume. Cartographic representations of both water depth and sediment accumulation were produced, along with an accuracy assessment for the resulting bathymetric model. Vertical accuracies at the 95-percent confidence level for the collected data, the bathymetric surface model, and the bathymetric contour map were calculated to be 0.64 feet (ft), 1.77 ft, and 2.30 ft, respectively. A dry bulk sediment density was calculated for each of the 18 sediment cores collected during March and April 2012, and used to determine accumulated sediment mass. Water-storage capacity in the study area is 110 acre-feet (acre-ft) at a full-pool elevation 308 ft above the National Geodetic Vertical Datum of 1929, whereas total sediment volume in the study area is 202 acre-ft. These totals indicate a loss of about 65 percent of the original water-storage capacity in the 40 years since dam construction. This corresponds to an average rate of sediment

    Many X-ray accreting pulsars have a soft excess below 10 keV. This feature was also detected in faint sources and at low luminosity levels, suggesting that it is an ubiquitous phenomenon. For the high luminosity pulsars (LX > 1036 erg s-1), the fit to this component with thermal emission models usually provides low temperatures (kT < 0.5 keV) and large emission regions (R ≥ a few hundred km), hence it is referred to as a "soft" excess. Nevertheless, we previously found that in persistent, low-luminosity (LX ~ 1034 erg s-1) and long-period (P > 100 s) Be accreting pulsars the observed excess can be modeled with a rather hot (kTBB > 1 keV) black-body component of small area (RBB < 0.5 km), which can be interpreted as emission from the NS polar caps. In this paper, we present an analysis of an XMM-Newton observation of the Galactic Be pulsar RX J0440.9+4431, which is a poorly studied member of this class of sources. We find a best-fit period of P = 204.96 ± 0.02 s, which implies an average pulsar spin-down over the past 13 years of Ṗ ≃ 6 × 10-9 s s-1. The estimated source luminosity is LX ~ 8 × 1034 erg s-1, which is higher by a factor of less than ten compared to those obtained in the first source observations, but almost two orders of magnitude lower than those measured during the few outbursts that have been detected most recently. The source spectrum can be described with a power-law and black-body model, with kTBB = 1.34 ± 0.04 keV and RBB = 273 ± 16 m, suggesting a polar-cap origin of this component. Our results support the classification of RX J0440.9+4431 as a persistent Be/NS pulsar, and confirm that the hot black-body spectral component is a common property of this class of sources.

    The Triassic outcrops of the Debadib-Ben Gasseur area, located in the Kef area (northwestern Tunisia) have recently been interpreted as submarine 'salt glaciers' interbedded within the Albian deposits (J.-M. Vila et al., C. R. Acad. Sci. Paris, Ser. II 318 (1994) 1661-1667). New cartographic, drills and gravimetric data recognised evidence for an Albian diapiric stage for the Triassic rocks. The Triassic rocks as pulling out of the sedimentary cover could spread laterally as small size 'salt glaciers' before they have been deformed during the Tertiary compressional deformations. In this scenario, Ben Amar and Bianca [2] give us a broad picture of the existing mathematical and computational tools that have been used to model fibrotic processes at the molecular, cellular, and tissue levels. Among such techniques, agent based models (ABM) can give a valuable contribution in the understanding and better management of fibrotic diseases.

    The development of anti-fibrotic therapies in diversities of diseases becomes more and more urgent recently, such as in pulmonary, renal and liver fibrosis [1,2], as well as in malignant tumor growths [3]. As reviewed by Ben Amar and Bianca [4], various theoretical, experimental and in-silico models have been developed to understand the fibrosis process, where the implication on therapeutic strategies has also been frequently demonstrated (e.g., [5-7]). In [4], these models are analyzed and sorted according to their approaches, and in the end of [4], a unified multi-scale approach was proposed to understand fibrosis. While one of the major purposes of extensive modeling of fibrosis is to shed light on therapeutic strategies, the theoretical, experimental and in-silico studies of anti-fibrosis therapies should be conducted more intensively.

    Mathematical and computational models in biology and medicine help to improve diagnostics and medical treatments. Modeling of pathological fibrosis is reviewed by M. Ben Amar and C. Bianca in [4]. Pathological fibrosis is the process when excessive fibrous tissue is deposited on an organ or tissue during a wound healing and can obliterate their normal function. In [4] the phenomena of fibrosis are briefly explained including the causes, mechanism and management; research models of pathological fibrosis are described, compared and critically analyzed. Different models are suitable at different levels: molecular, cellular and tissue. The main goal of mathematical modeling of fibrosis is to predict long term behavior of the system depending on bifurcation parameters; there are two main trends: inhibition of fibrosis due to an active immune system and swelling of fibrosis because of a weak immune system.

DOI:10.1016/j.atmosres.2010.06.011.), reanalyzed the results of the operational seeding in northern Israel between 1975 and 2007 and the preceding Israel 2 cloud seeding experiment (1969-1975) and concluded that there is no net increase in precipitation over the target areas. Our analysis revealed that a synoptic bias during Israel 2 is one of the reasons for the apparent positive effect of seeding in the northern target area and the negative effect in the southern area both of which disappeared in the following experiment in the south (Israel 3; 1975-1995) and the operational seeding in the north. Ben-Zvi et al. (2010;hereafter BRG) criticized our paper primarily on the ground that we did not consider the positive results of Israel 1 experiment (1960-1967). It should be noted that in Israel 1 different seeding lines were used from those in both Israel 2 and the operational period. In addition, its raw data is not accessible anymore for reanalysis. Furthermore, Israel 2 had been designed as a confirmatory cross-over experiment to Israel 1 and failed to reproduce its promising results with double ratio (DR) of ~ 1.00, namely, zero rainfall enhancements. The same DR values were also found in Israel 3 and in the operational seeding. Therefore, because of the differences in the two experiments, the lack of access to the raw data and the disappointing results of the confirmatory experiment, we decided to concentrate our analysis on the more recent seeding activities. The attempt by BRG to explain the reduction of the DR to ~ 1.00 in the operational seeding period by the suppression due to pollution have been disproved by Alpert et al. (2008, 2009) and also fail to explain the sharp decline of the target/control ratio right at the beginning of the operational seeding period when

    At the age of 42, Franklin retired from his prosperous printing business, ``to have leisure to read, study, and make experiments." The scope of his scientific work was remarkable. Beyond his key contributions to understanding electrcity, Franklin wrote major papers on population growth, on meterology, on heat conduction and evaporation, charted the Gulf Stream, studied bioluminescence and the stilling of water waves by a surface layer of oil. He also advanced arguments in favor of conservation of mass and the wave theory of light. Although always alert for practical applications, his style was that of an explorer, eager for adventure and insight. However, Franklin did not consider science as important as public service. He promoted many civic projects in Philadelphia, including a circulating library, fire department, paving of streets, hospital, and was the prime mover in founding the American Philosophical Society and an academy that evolved into the University of Pennsylvania. As well as publishing the most widely read newspaper in the colonies and his bestselling almanac, he became public printer for several of the colonies and postmaster. He lived in England 14 years as a trade representative, largely struggling in vain. At the age of 70, he undertook a decade of service as minister to France, achieving against great odds crucial diplomatic triumphs.

Herb properties were evaluated mainly according to record and clinical practice with emphasis on property-effect (effect and adverse effect) relationship. Logical deduction was also used to deduce herb toxicity and quality according to herb flavor, nature, tastes and their growing environment. The book described in detail the influence of processing on the herb properties and toxicity.     L. Benjamin Wyckoff's seminal contributions to both psychological theory and application are the subject of this review. Wyckoff started his academic career as a graduate student at Indiana University, where he developed the observing-response procedure under the guidance of B. F. Skinner and C. J. Burke. At the University of Wisconsin–Madison, Wyckoff refined his mathematical theory of secondary reinforcement. This theory was the impetus for his creation of an electronic simulation of a rat running a T maze, one of the first “computer models” of learning. Wyckoff next went to Emory University, leaving there to help create two of the most successful companies dedicated to the advancement of programmed instruction and teaching machines: Teaching Machines, Inc. and the Human Development Institute. Wyckoff's involvement in these companies epitomizes the application of basic behavior-analytic principles in the development of technology to improve education and human relationships. The emergent picture of Wyckoff is that of a man who, through his research, professional work in educational applications of behavioral principles, and active involvement in the civil rights movement of the 1960s, was strongly committed to applying behavioral science to positively influence human behavior change.   8. Observing ben wyckoff: from basic research to programmed instruction and social issues.

    L. Benjamin Wyckoff's seminal contributions to both psychological theory and application are the subject of this review. Wyckoff started his academic career as a graduate student at Indiana University, where he developed the observing-response procedure under the guidance of B. F. Skinner and C. J. Burke. At the University of Wisconsin-Madison, Wyckoff refined his mathematical theory of secondary reinforcement. This theory was the impetus for his creation of an electronic simulation of a rat running a T maze, one of the first "computer models" of learning. Wyckoff next went to Emory University, leaving there to help create two of the most successful companies dedicated to the advancement of programmed instruction and teaching machines: Teaching Machines, Inc. and the Human Development Institute. Wyckoff's involvement in these companies epitomizes the application of basic behavior-analytic principles in the development of technology to improve education and human relationships. The emergent picture of Wyckoff is that of a man who, through his research, professional work in educational applications of behavioral principles, and active involvement in the civil rights movement of the 1960s, was strongly committed to applying behavioral science to positively influence human behavior change.     The modern and medieval meanings of words reporting ill health often bear little resemblance to one another. This article compares the use of ‘diseased’ and ‘sick’ in the fifteenth-century Stonor family letters. It examines the word ‘crased’, which implies physical ill health most directly, but also suggests emotional, psychological, or spiritual distress in female family members especially. The article then turns to the practical implications of poor health, asking how and why it affected the day-to-day concerns of the Stonors and their associates. It uncovers compelling evidence for resilience in the face of many and competing calls of duty. Finally, the article presents unique palaeographical evidence for the impact of illness, where a correspondent is so ‘seke’ that he can scarcely hold his pen.

    In the summer of 1969, a deep submersible drifted for 30 days below the surface of the Gulf Stream, while operated by a six man crew. The main purpose of the mission was oceanographic research. The crew's activities and completely self-contained environment resembled those of a space station such as Skylab. Because of these similarities aspects of onboard vehicle maintenance during the actual conduct of a scientific mission were investigated. The maintainability study was accomplished in six distinct phases. Two useful plots of manpower distribution were developed. A maintenance action summary is presented in a table.

  17. Promoting healthy eating, active play and sustainability consciousness in early childhood curricula, addressing the Ben10™ problem: a randomised control trial

    Background This paper details the research protocol for a study funded by the Australian Research Council. An integrated approach towards helping young children respond to the significant pressures of ‘360 degree marketing’ on their food choices, levels of active play, and sustainability consciousness via the early childhood curriculum is lacking. The overall goal of this study is to evaluate the efficacy of curriculum interventions that educators design when using a pedagogical communication strategy on children’s knowledge about healthy eating, active play and the sustainability consequences of their toy food and toy selections. Methods/Design This cluster-randomised trial will be conducted with 300, 4 to 5 year-old children attending pre-school. Early childhood educators will develop a curriculum intervention using a pedagogical communication strategy that integrates content knowledge about healthy eating, active play and sustainability consciousness and deliver this to their pre-school class. Children will be interviewed about their knowledge of healthy eating, active play and the sustainability consequences of their food and toy selections. Parents will complete an Eating and Physical Activity Questionnaire rating their children’s food preferences, digital media viewing and physical activity habits. All measures will be administered at baseline, the end of the intervention and 6 months post intervention. Informed consent will be obtained from all parents and the pre-school classes will be allocated randomly to the intervention or wait-list control group. Discussion This study is the first to utilise an integrated pedagogical communication strategy developed specifically for early childhood educators focusing on children’s healthy eating, active play, and sustainability consciousness. The significance of the early childhood period, for young children’s learning about healthy eating, active play and sustainability, is now unquestioned. The specific teaching and learning practices used by early childhood educators, as part of the intervention program, will incorporate a sociocultural perspective on learning; this perspective emphasises building on the play interests of children, that are experienced within the family and home context, as a basis for curriculum provision.   18. Variation of a benthic heterotrophic bacteria community with different respiratory metabolisms in Coyuca de Benítez coastal lagoon (Guerrero, Mexico).

    The fluctuations of the number, biomass and composition of the heterotrophic community were studied daily for two days, according to depth, pH, Eh, O2 and organic carbon concentration within a zone of the canal between the Coyuca de Benitez lagoon (Guerrero, Mexico) and the coastal waters. At the three moments of the day studied (6 am, 2 pm and 10 pm), the oxygen concentrations in the overlying water and in the superficial sediment layer were near air-saturation in the diurnal samplings (582 microM at 6 am and 665 microM at 2 pm), and sub-satured during the night (158 microM). In the sediments, the models of vertical distribution of Eh and organic carbon distributions were very irregular due to the bio-perturbation of the benthic, meio- and macrofauna, whose activity allows the superficial organic carbon to migrate towards sediment deeper layers. Vertical distribution of the different viable bacteria populations seems to be related to the hydrodynamic patterns of the communicating canal and sediments heterogeneity. In the sediment column, the heterotrophic bacteria total number varied from 6.8 to 20.3 x 108 cells cm(-3). The highest heterotrophic bacterial biomass values were encountered during the diurnal samplings (39.2 microgC.l(-1) at 6 am and 34.4 microgC.(l(-1) at 2 pm) and the lowest during the night (9.7 microgC.l(-1). The fluctuations of viable heterotrophic bacteria populations with different respiratory metabolisms (aerobic, microaerophilic and anaerobic) can be explained by the existence of suboxic microniches that appear when particles of sediment are resuspended due to the water circulation and the benthic infauna excavating activity, that allows the supernatant water oxygen to penetrate through its galleries towards deeper sediment zones. The statistical analysis (Multiple lineal regression model r2 > or = 0.5) showed that the on the whole, the hydrological parameters are not influence over the bacterial number and bacterial biomass distribution (r2 < or = 0.5), Nevertheless, the variations of the heterotrophic bacteria community observed in the two days sampling, seem to be governed (with F-values of 0.6 to 0.9) by the irregular flows of bio-available organic material and the sediment porosity.     This symposium will explore simulation methods for the preclinical evaluation of novel 3D and 4D x-ray breast imaging systems – the subject of AAPM taskgroup TG234. Given the complex design of modern imaging systems, simulations offer significant advantages over long and costly clinical studies in terms of reproducibility, reduced radiation exposures, a known reference standard, and the capability for studying patient and disease subpopulations through appropriate choice of simulation parameters. Our focus will be on testing the realism of software anthropomorphic phantoms and virtual clinical trials tools developed for the optimization and validation of breast imaging systems. The symposium will review the stateof- the-science, as well as the advantages and limitations of various approaches to testing realism of phantoms and simulated breast images. Approaches based upon the visual assessment of synthetic breast images by expert observers will be contrasted with approaches based upon comparing statistical properties between synthetic and clinical images. The role of observer models in the assessment of realism will be considered. Finally, an industry perspective will be presented, summarizing the role and importance of virtual tools and simulation methods in product development. The challenges and conditions that must be satisfied in order for computational modeling and simulation to play a significantly increased role in the design and evaluation of novel breast imaging systems will be addressed.     The process of inflammation tries to protect the body after an injury due to biological causes such as the presence of pathogens or chemicals, or to physical processes such as burns or cuts. The biological rationale for this process has the main goal of eliminating the cause of the injury and then repairing the damaged tissues. We can distinguish two kinds of inflammations: acute and chronic. In acute inflammation, a series of events involving the local vascular systems, the immune system and various cells within the injured tissue work together to eradicate the harmful stimuli. If the inflammation does not resolve the problem, it can evolve into a chronic inflammation, where the type of cells involved changes and there is a simultaneous destruction and healing of the tissue from the inflammation process.

    The elastic scattering cross-sections of 10,11Be on protons and 12C at energy E < 100 MeV/nucleon using microscopically calculated optical potentials (OP) are presented. The real OP is obtained by a folding procedure with effective NN interactions, while the imaginary OP is estimated within the high energy approximation (HEA). The spin-orbit part of the OP is also included. The characteristics of the breakup processes of 11Be on different nuclear targets are also considered. The cross-sections of diffractive breakup and stripping reactions of 11Be on 9Be, 93Nb, 181Ta and 238U at energy E = 63 MeV/nucleon and the longitudinal momentum distributions of 10Be fragments produced in the breakup of 11Be on these nuclei are presented. The results are in a good agreement with the available experimental data, in particular the obtained widths of about 50 MeV/c are closed to the empirical ones.

    Climate proxy records from the middle to high latitude Southern Hemisphere indicate that a Lateglacial (15,000-11,500 years ago) climate reversal, approximately coeval with the Antarctic Cold Reversal (ACR), interrupted a warming trend during deglaciation. In New Zealand, some palaeoclimate proxy records indicate a cool episode during the ACR (ca 14,500-12,500 years ago), while others do not express a significant change in climate. Recently published moraine maps and ages present an opportunity to improve the palaeoclimate interpretation through numerical modelling of glaciers. We use a coupled energy-balance and ice-flow model to quantify palaeoclimate from past glacier extent constrained by mapped and dated moraines in the headwaters of Irishman Stream, a high-elevation catchment in the Southern Alps. First, a suite of steady-state model runs is used to identify the temperature and precipitation forcing required to fit the modelled glacier to well-dated Lateglacial moraine crests. Second, time-dependent glacier simulations forced by a nearby proxy temperature record derived from chironomids are used to assess the fit with the glacial geomorphic record. Steady-state experiments using an optimal parameter set demonstrate that the conditions under which the 13,000 year old moraine formed were 2.3-3.2 °C colder than present with the range in temperature corresponding to a ±20% variance in precipitation relative to the present-day. This reconstructed climate change relative to the present-day corresponds to an equilibrium-line altitude of ca 2000 ± 40 m above sea level (asl), which is ca 400 m lower than present. Time-dependent simulations of glacier length produce ice advance to within 100 m of the 13,000 year old terminal moraine, indicating that the chironomid-based temperature forcing and moraine record provide consistent information about past climate. Our results, together with other climate proxy reconstructions from pollen records and marine sediment cores, support the notion that temperatures during the ACR in New Zealand were ˜2-3 °C cooler than today.

    Clozapine is the treatment of choice for treatment-resistant schizophrenia, but it is associated with a risk of neutropaenia and agranulocytosis. Clozapine use is regulated by mandatory blood monitoring in the UK, requiring cessation of treatment should the absolute neutrophil count (ANC) drop below specified values. Benign reductions in the ANC in non-white populations are common, and this can preclude a patient from receiving treatment with clozapine. A diagnosis of benign ethnic neutropaenia can reduce these treatment restrictions (UK specific), but the degree of neutropaenia can be significant enough to still prevent treatment. In this report, we show that response to granulocyte colony stimulating factor (G-CSF) may be quite variable and difficult to predict, but with careful monitoring it can be used to increase the ANC count and allow continued treatment with clozapine.     The C14:C12 ratio of atmospheric CO2 (expressed as Delta 14C) is the gold standard measurement to derive the portion of observed atmospheric CO2 gradients resulting from combustion of fossil fuels (CO2-ff). This is because fossil fuels are devoid of 14C, unlike all other sources and sinks that impact atmospheric Delta14C. With enough 14C measurements, independent, 'top-down', estimation of US fossil fuel-CO2 emissions should be possible. However, our ability to make carbon-14 measurements is severely constrained by cost, accessibility to accelerator mass spectrometers (AMS) and the volume of air required to make high precision (~0.2 %) measurements of 14CO2 (mixing ratio is ~ 4e-16 mol/mol). Thus, Delta 14C is currently measured in only a small subset of NOAA/ESRL Global Monitoring Division (GMD) tall-tower and aircraft air samples. Here, we present a Projection Pursuit Regression (PPR) model to predict CO2-ff measured at different times and altitudes in terms of surrogate gases that are more widespread and relatively inexpensive to measure. This method would, in effect, allow expansion of Delta 14C measurements by factor of ~3 or 4 throughout North America. To create a proxy for CO2-ff, we take advantage of the observed correlations between (Delta 14C-derrived) CO2-ff and regional-scale enhancements of a wide array of anthropogenic gases, like CO, SF6, and halo- and hydro-carbons. We select the complexity and form of the PPR model by cross validation where validation data prediction error is minimized. In cross validation, the prediction model is based on the training data and not the validation data. We quantify prediction model performance with test data excluded from the model development process. According to cross validation, the PPR model is superior to a simpler linear model. Comparison with test CO2-ff data shows that CO2-ff can be predicted with a root mean square error of 1.1 ppm, only slightly higher than the Delta 14C-precision limit for CO2-ff of 1 ppm. Applying the PPR model to the large NOAA/ESRL/GMD database of surrogate gases, we can create a large set of synthetic CO2-ff data for the US. Finally, we will discuss the application of this large data set of lower-precision CO2-ff to top down determination of US fossil fuel CO2 emissions by presenting results from an Observation System Simulation Experiment (OSSE).

    The topic of the review article [1] is the derivation of a multiscale paradigm for the modeling of fibrosis. Firstly, the biological process of the physiological and pathological fibrosis including therapeutical actions is reviewed. Fibrosis can be a consequence of tissue damage, infections and autoimmune diseases, foreign material, tumors. Some questions regarding the pathogenesis, progression and possible regression of fibrosis are lacking. At each scale of observation, different theoretical tools coming from computational, mathematical and physical biology have been proposed. However a complete framework that takes into account the different mechanisms occurring at different scales is still missing. Therefore with the main aim to define a multiscale approach for the modeling of fibrosis, the authors of [1] have presented different top-down and bottom-up approaches that have been developed in the literature. Specifically, their description refers to models for fibrosis diseases based on ordinary and partial differential equation, agents [2], thermostatted kinetic theory [3-5], coarse-grained structures [6-8] and constitutive laws for fibrous collagen networks [9]. A critical analysis has been addressed for all frameworks discussed in the paper. Open problems and future research directions referring to both biological and modeling insight of fibrosis are presented. The paper concludes with the ambitious aim of a multiscale model.

    The review paper [1] is devoted to the survey of different structures that have been developed for the modeling and analysis of various types of fibrosis. Biomathematics, bioinformatics, biomechanics and biophysics modeling have been treated by means of a brief description of the different models developed. The review is impressive and clearly written, addressed to a reader interested not only in the theoretical modeling but also in the biological description. The models have been described without recurring to technical statements or mathematical equations thus allowing the non-specialist reader to understand what framework is more suitable at a certain observation scale. The review [1] concludes with the possibility to develop a multiscale approach considering also the definition of a therapeutical strategy for pathological fibrosis. In particular the control and optimization of therapeutics action is an important issue and this article aims at commenting on this topic.

  5. Removal of rhodamine B using iron-pillared bentonite.

    The iron-pillared bentonite (Fe-Ben) was prepared by ion-exchange using the natural bentonite (GZ-Ben) from Gaozhou, China, at room temperature without calcination. Both Fe-Ben and GZ-Ben were characterized by X-ray diffraction, N(2) adsorption and Fourier transform infrared spectroscopy. The results show that the d(001) value and surface area of the bentonite material increased after iron pillaring. Fe-Ben adsorbed much more Rhodamine B (RhB) than GZ-Ben, which can be ascribed to the special surface properties and large surface area of Fe-Ben. The optimum pH value for the adsorption of RhB on Fe-Ben is 5.0. The adsorption of RhB onto Fe-Ben can be well described by the pseudo-second-order kinetic model and the intraparticle diffusion kinetic model. The adsorption isotherm of RhB onto Fe-Ben matches well with the Langmuir model. PMID:21168960

Using resting state functional magnetic resonance imaging (rsfMRI), reliable and spatially distributed BEN patterns have been identified in normal brain, suggesting a potential use in clinical populations since temporal brain dynamics and entropy may be altered in disease conditions. The purpose of this study was to characterize BEN in multiple sclerosis (MS), a neurodegenerative disease that affects millions of people. Since currently there is no cure for MS, developing treatment or medication that can slow down its progression represents a high research priority, for which validating a brain marker sensitive to disease and the related functional impairments is essential. Because MS can start long time before any measurable symptoms and structural deficits, assessing the dynamic brain activity and correspondingly BEN may provide a critical way to study MS and its progression. Because BEN is new to MS, we aimed to assess BEN alterations in the relapsing-remitting MS (RRMS) patients using a patient versus control design, to examine the correlation of BEN to clinical measurements, and to check the correlation of BEN to structural brain measures which have been more often used in MS studies. As compared to controls, RRMS patients showed increased BEN in motor areas, executive control area, spatial coordinating area, and memory system. Increased BEN was related to greater disease severity as measured by the expanded disability status scale (EDSS) and greater tissue damage as indicated by the mean diffusivity. Patients also showed decreased BEN in other places, which was associated with less disability or fatigue, indicating a disease-related BEN re-distribution. Our results suggest BEN as a novel and useful tool for characterizing RRMS. PMID:26727514

    Balkan endemic nephropathy (BEN) is a chronic familial form of interstitial nephritis that might eventually lead to end stage renal disease. This nephropathy affects individuals living along of the Danube River and its tributaries in Serbia, Bosnia, Croatia, Bulgaria and Romania. The increased incidence of urinary tract tumors in the BEN areas is well described, but its specific genetic predisposition is still unclear. Certain nephrocarcinogenic compounds, including those associated with BEN, are metabolized by glutathione S-transferase (GST) superfamily of phase II detoxication enzymes. Importantly, the GST-mediated detoxification may result in formation of more toxic compounds. We examined the association of common GST polymorphisms and bladder cancer (BC) risk in individuals from BEN areas in Serbia. A hospital-based case-control study included 201 BC cases (67 from BEN region) and 122 controls. Each polymorphism was identified by a PCR-based method. Individuals from BEN region with low-expression GSTA1 genotype (AB+BB) exhibited a 2.6-fold higher BC risk compared to those with GSTA1 (AA) genotype who were from non-BEN region (OR = 2.60, p = 0.015). In contrast, carriers of GSTM1-active genotype from BEN region had a 2.9-fold increased BC risk compared to those with GSTM1-active genotype from non-BEN region (OR = 2.90, p = 0.010). Likewise, carriers with GSTT1-active genotype from BEN region exhibited 2.1-fold higher BC risk compared to those from non-BEN region with GSTT1-active genotype (OR = 2.10, p = 0.027). Thus, common polymorphisms in GSTA1, GSTM1 and GSTT1 are associated with susceptibility to BC in individuals from BEN areas of Serbia. PMID:27568660

  9. Offspring of parents with Balkan Endemic Nephropathy have higher C-reactive protein levels suggestive of inflammatory processes: a longitudinal study

    Background Despite the characteristic extensive tubulointerstitial fibrosis, Balkan Endemic Nephropathy (BEN) is usually considered a non-inflammatory disease. Methods We examined a marker of inflammation, C-reactive protein (CRP), in the offspring of patients with BEN, a population at risk for BEN, prior to development of established disease to determine if an inflammatory process could be identified in the early stages of the disease. In 2003/04, 102 adult offspring whose parents had BEN and a control group of 99 adult offspring of non-BEN patients were enrolled in this prospective study. This cohort was re-examined yearly for four consecutive years. Levels of serum CRP were measured in years 3 and 4 and compared between groups. The data were analyzed with mixed models. Results Compared to controls, offspring of BEN parents had statistically higher CRP levels in two consecutive years, suggestive of early inflammatory reactivity. Whenever the mother was affected by BEN (both parents, or mother only), serum CRP was significantly increased, but not if only the father had BEN. CRP was inversely related to kidney cortex width but not to markers or renal function. Conclusion Early stages of BEN may involve inflammatory processes. The observation of a maternal involvement supports the concept of fetal programming, which has been implicated in the pathogenesis of other chronic kidney diseases. PMID:19400955

    Benign ethnic neutropenia (BEN) is an asymptomatic condition reported in adults of African and Middle Eastern descent. The clinical description in children is currently lacking. In our urban outpatient pediatric hematology clinic, the median neutrophil count of children with BEN was lower than previous reports in adults at 893×10 cells/L, but increased with older age. There was an equal male to female ratio and 24% of our BEN children reported ethnicities other than African or Middle Eastern. Children with BEN had a clinical course comparable with other healthy children including otherwise normal blood counts, except for mild anemia. PMID:26925714

  11. Suitability of Soybean Meal from Insect-Resistant Soybeans for Broiler Chickens.

    Benning(M) and Benning(MGH) are near-isogenic lines (NILs) of the soybean cultivar Benning, which contain insect-resistance quantitative trait loci (QTLs) from the soybean accession PI 229358. Benning(M) contains QTL-M, which confers antibiosis and antixenosis. In addition to QTL-M, Benning(MGH) contains QTL-G, which confers antibiosis, and QTL-H, which confers antixenosis. Soybean meal was produced from Benning and the NILs. Nutritional composition, digestible amino acid content, and nitrogen-corrected true metabolizable energy (TMEN) were equivalent among soybean meals. A 21-day broiler feeding trial was carried out to determine if the QTLs affect soybean meal quality. Weight gain and feed-to-gain ratio were evaluated. No biologically significant differences were detected for broilers fed Benning, Benning(M), and Benning(MGH). This demonstrates that soybean meal produced from the insect-resistant NILs is equivalent to soybean meal produced from their non-insect-resistant parent cultivar for broiler weight gain.     Cigarette smoking is a chronic relapsing brain disorder, and remains a premier cause of morbidity and mortality. Functional neuroimaging has been used to assess differences in the mean strength of brain activity in smokers’ brains, however less is known about the temporal dynamics within smokers’ brains. Temporal dynamics is a key feature of a dynamic system such as the brain, and may carry information critical to understanding the brain mechanisms underlying cigarette smoking. We measured the temporal dynamics of brain activity using brain entropy (BEN) mapping and compared BEN between chronic non-deprived smokers and non-smoking controls. Because of the known sex differences in neural and behavioral smoking characteristics, comparisons were also made between males and females. Associations between BEN and smoking related clinical measures were assessed in smokers. Our data showed globally higher BEN in chronic smokers compared to controls. The escalated BEN was associated with more years of smoking in the right limbic area and frontal region. Female nonsmokers showed higher BEN than male nonsmokers in prefrontal cortex, insula, and precuneus, but the BEN sex difference in smokers was less pronounced. These findings suggest that BEN mapping may provide a useful tool for probing brain mechanisms related to smoking. PMID:27377552

    1H NMR spectroscopy of urine has been applied to exploring metabolomic differences between people diagnosed with Balkan endemic nephropathy (BEN), and treated by haemodialysis, and those without overt renal disease in Romania and Bulgaria. Convenience sampling was made from patients receiving haemodialysis in hospital and healthy controls in their village. Principal component analysis clustered healthy controls from both countries together. Bulgarian BEN patients clustered separately from controls, though in the same space. However, Romanian BEN patients not only also clustered away from controls but also clustered separately from the BEN patients in Bulgaria. Notably, the urinary metabolomic data of two people sampled as Romanian controls clustered within the Romanian BEN group. One of these had been suspected of incipient symptoms of BEN at the time of selection as a ‘healthy’ control. This implies, at first sight, that metabolomic analysis can be predictive of impending morbidity before conventional criteria can diagnose BEN. Separate clustering of BEN patients from Romania and Bulgaria could indicate difference in aetiology of this particular silent renal atrophy in different geographic foci across the Balkans. Functional neuroimaging has been used to assess differences in the mean strength of brain activity in smokers' brains, however less is known about the temporal dynamics within smokers' brains. Temporal dynamics is a key feature of a dynamic system such as the brain, and may carry information critical to understanding the brain mechanisms underlying cigarette smoking. We measured the temporal dynamics of brain activity using brain entropy (BEN) mapping and compared BEN between chronic non-deprived smokers and non-smoking controls. Because of the known sex differences in neural and behavioral smoking characteristics, comparisons were also made between males and females. Associations between BEN and smoking related clinical measures were assessed in smokers. Our data showed globally higher BEN in chronic smokers compared to controls. The escalated BEN was associated with more years of smoking in the right limbic area and frontal region. Female nonsmokers showed higher BEN than male nonsmokers in prefrontal cortex, insula, and precuneus, but the BEN sex difference in smokers was less pronounced. These findings suggest that BEN mapping may provide a useful tool for probing brain mechanisms related to smoking. PMID:27377552

A pathogenic strain BEN-1 was isolated from bovine lung and underwent continuous passages in rabbits for 468 generations. During this process, the strain’s strong virulence became weak and, gradually, it lost the ability to confer protective immunity in cattle but developed virulence in rabbits. In order to gain insight into the mechanisms behind the reduction in virulence and the loss of immunogenicity, we sequenced five representative strains of the BEN series, including the original strain (BEN-1), the strain generation that first acquired virulence in rabbits (BEN-50), the two vaccine strain generations (BEN-181 and BEN-326), and the strain generation showing the greatest loss of immunogenicity (BEN-468). The gene mutation rate in the four different propagation stages varied greatly, and over half of variations observed in each generation were removed during the propagation process. However, the variation maintained in the BEN-468 generation might contribute to its changes in virulence and immunogenicity. We thus identified 18 genes associated with host adaptation, six genes contributing to virulence in cattle, and 35 genes participating in conferring immunity in cattle.   2. The role of lecithin cholesterol acyltransferase and organic substances from coal in the etiology of Balkan endemic nephropathy: A new hypothesis

    Balkan endemic nephropathy (BEN) occurs in Serbia, Bulgaria, Romania, Bosnia and Herzegovina, and Croatia. BEN has been characterized as a chronic, slowly progressive renal disease of unknown etiology. In this study, we examined the influence of soluble organic compounds in drinking water leached from Pliocene lignite from BEN-endemic areas on plasma lecithin-cholesterol acyltransferase (LCAT) activity. We found that changes for all samples were the most prominent for the dilution category containing 90% plasma and 10% of diluting media. Water samples from BEN villages from Serbia and Romania showed higher LCAT inhibiting activity (p = 0.02) and (p = 0.003), respectively, compared to deionised water and non-endemic water. A secondary LCAT deficiency could result from this inhibitory effect of the organic compounds found in endemic water supplies and provide an ethiopathogenic basis for the development of BEN in the susceptible population. ?? 2007 Elsevier Ltd.     Balkan endemic nephropathy (BEN) is a fatal kidney disease that is known to occur only in clusters of villages in alluvial valleys of tributaries of the Danube River in Bulgaria, Romania, Yugoslavia, Bosnia, and Croatia. The confinement of this disease to a specific geographic area has led to speculation that an environmental factor may be involved in the etiology of BEN. Numerous environmental factors have been suggested as causative agents for producing BEN, including toxic metals in drinking water, metal deficiency in soils of BEN areas, and environmental mycotoxins to name a few. These hypotheses have either been disproved or have failed to conclusively demonstrate a connection to the etiology of BEN, or the clustering of BEN villages. In previous work, we observed a distinct geographic relationship between the distribution of Pliocene lignites in the Balkans and BEN villages. We hypothesized that the long-term consumption of well water containing toxic organic compounds derived from the leaching of nearby Pliocene lignites by groundwater was a primary factor in the etiology of BEN. In our current work, chemical analysis using 13C nuclear magnetic resonance (13CNMR) spectroscopy indicated a high degree of organic functionality in Pliocene lignite from the Balkans, and suggested that groundwater can readily leach organic matter from these coal beds. Semi-quantitative gas chromatography/mass spectroscopy analysis of solvent extracts of groundwater from shallow wells in BEN villages indicated the presence of potentially toxic aromatic compounds, such as napthalene, fluorene, phenanthrene, and pyrene at concentrations in the ppb range. Laboratory leaching of Balkan Pliocene lignites with distilled water yielded soluble organic matter (> 500 MW) containing large amounts of aromatic structures similar to the simple/discrete aromatic compounds detected in well water from BEN villages.     Balkan endemic nephropathy (BEN), originally described in the late 1950s as a chronic tubulointerstitial kidney disease, is identified by its unique epidemiological features. The most remarkable characteristic of BEN is the focal topographical nature that characterizes its occurrence at the global, national, and even household level. BEN affects only certain endemic rural foci along tributaries of the Danube River in the Balkan countries of Bosnia, Bulgaria, Croatia, Romania, and Serbia. The spatial distribution has remained astonishingly unchanged with time because the disease affects the same endemic clusters as 50 years ago. The natural course of the disease is characterized by universal development of end-stage renal disease and the frequent development of upper urinary tract tumors, posing a substantial disease burden to the afflicted areas. The greatest challenge in the study of BEN has been the elucidation of its cause. The unique features of the disease, in particular its endemic nature and the long incubation period required for the disease to develop, have led to the proposal that BEN represents a unique environmental disease. The quest for the responsible environmental factor has been long and diverse, and although no definitive answer has been provided to date, converging lines of evidence support the theory that long-term consumption of food contaminated with aristolochic acid underlies the pathogenesis of BEN. The present review describes the evolution of our knowledge of BEN in relation to the development of the main theories for its pathogenesis. PMID:18725017

  5. Correlation Of Balkan Endemic Nephropathy With Fluorescent Organic Compounds In Shallow Groundwater

    Balkan endemic nephropathy (BEN) is a disease of intersitial nephropathy leading to end-stage renal failure. The disease occurs in persons living in villages on alluvial valleys of streams tributary to the Danube River in Rumania, Bulgaria and former Yugoslavia. The etiologic agent is not known, but a contaminant in shallow groundwater has become suspect. In this study, samples of drinking water from endemic and non-endemic village water supplies were analyzed by excitation/emission matrix (EEM) fluorescence spectroscopy. Spectra characteristic of groundwater from BEN households show elongated teardrop shapes in the fluorescence excitation/emission matrix. A sharp rise occurs in fluorescence emission between 380 and 400 nanometers (nm) and a trailing emission intensity from 400 to 550 nm. Spectra of groundwater samples from some BEN households have an additional excitation maxima at 300 nm, which further contributes to the emission intensity at 400 nm. Spectra of water samples from non-BEN households located in endemic villages show characteristics of BEN household waters, exhibiting the 250-nm excitation peak, even though the fluorophoric intensity is much less than that in samples from BEN household waters. Samples from non-endemic villages do not show the characteristic EEM spectra described as "teardrop shaped". The non-BEN households have lower concentrations of these fluorophores in the drinking water than the endemic households; hence, one of the factors in contracting the disease may be the concentration of these fluorescent materials in drinking water.

    Upper Tract Urothelial Carcinoma (UTUC) is an uncommon disease which occurs more frequently in some regions of Balkan countries than in other areas in the world. Investigation of UTUC in the South Morava River basin and its tributaries where BEN is endemic revealed increased frequency not only of tumour of the renal pelvis and ureter but also of urinary bladder tumours. A comparative morphological and immunohistochemical study of UTUC in the BEN region and control rural and city populations free of BEN, identify growth pattern as the best morphological characteristic which differentiated BEN and control tumours, i.e. solid growth for BEN tumours and papillary for control tumours. Overexpression of tumour suppressor p53 as well as decreased expression of E-CD was detected in BEN tumours. Other cells cycle related molecular markers--Cyclin D1, p16, and HER-2 showed no difference in expression between groups, as well as the proliferative marker Ki-67. Investigation of apoptosis-related markers identifies Bax as a specific marker of BEN-associated UTUC. Decrease of the pro-apoptotic protein Bax together with alteration of Survivin may be indicative of specific disturbances of an intrinsic apoptotic pathway in UTUC arising in endemic areas.     Aim. The aim of this study was to compare urinary alpha 1 microglobulin (A1MG) in healthy individuals with and without family burden for Balkan endemic nephropathy (BEN) in an endemic village. Methods. Otherwise healthy inhabitants with microalbuminuria or proteinuria were divided into two groups: with (n = 24) and without (n = 32) family BEN burden and screened for urinary A1MG and A1MG/urine creatinine ratio. Results. Average value of urinary A1MG was 10.35 ± 7.01 mg/L in group with and 10.79 ± 8.27 mg/L in group without family history for BEN (NS, P = 0.87). A1MG was higher than 10 mg/L in eight (33.33%) inhabitants with family history and in 12 (37.5%) without (NS, P = 0.187). Average values of urinary A1MG/creatinine ratio were 1.30 ± 1.59 and 0.94 ± 0.78 in group with and group without family BEN history (NS, P = 0.39, resp.). Elevated values of this ratio were found in 13 (54.17%) inhabitants with and 14 (43.75%) without family history for BEN (NS, P = 0.415). Conclusion. We did not find statistically significant difference in the examined markers between healthy individuals with and without family burden for BEN. We concluded that these markers are not predictive of risk for BEN. PMID:24563783

    Balkan endemic nephropathy (BEN), originally described in 1956, is a unique familial, chronic renal disease encountered with a high-prevalence rate in Serbia, Bulgaria, Romania, Croatia and Bosnia and Herzegovina. The most prominent features of the disease are its endemic nature, long-incubation period, familial clustering of the disease and an unusually high incidence of associated upper urothelial cancer (UUC). There are no clear-cut data on BEN incidence and prevalence, since the studies carried out in different endemic areas yielded contradictory information. In spite of intermittent variations, the incidence of new cases has remained stable over time. It has been estimated that almost 100 000 people are at risk of BEN, whereas 25 000 have the disease. The clinical signs and symptoms of BEN are non-specific and often remain unrecognized for years. There are no pathognomonic diagnostic features of BEN, but the set of epidemiological, clinical and biochemical data along with the pattern of pathologic injury in the absence of any other renal diseases are highly suggestive of this entity. Although the aetiology has been extensively studied, fostering the publication of various hypotheses, only one of them has provided conclusive evidence related to the aetiology of BEN. Studies conducted over the past decade have provided particularly strong arguments that BEN and UUC are caused by chronic poisoning with aristolochic acids (AAs). In light of these later studies, one can raise the question whether AAs could be responsible for previously and currently widespread unrecognized global renal disease and UUC. PMID:26064484

  9. Cloning and Characterization of Benzoate Catabolic Genes in the Gram-Positive Polychlorinated Biphenyl Degrader Rhodococcus sp. Strain RHA1

    Benzoate catabolism is thought to play a key role in aerobic bacterial degradation of biphenyl and polychlorinated biphenyls (PCBs). Benzoate catabolic genes were cloned from a PCB degrader, Rhodococcus sp. strain RHA1, by using PCR amplification and temporal temperature gradient electrophoresis separation. A nucleotide sequence determination revealed that the deduced amino acid sequences encoded by the RHA1 benzoate catabolic genes, benABCDK, exhibit 33 to 65% identity with those of Acinetobacter sp. strain ADP1. The gene organization of the RHA1 benABCDK genes differs from that of ADP1. The RHA1 benABCDK region was localized on the chromosome, in contrast to the biphenyl catabolic genes, which are located on linear plasmids. Escherichia coli cells containing RHA1 benABCD transformed benzoate to catechol via 2-hydro-1,2-dihydroxybenzoate. They transformed neither 2- nor 4-chlorobenzoates but did transform 3-chlorobenzoate. The RHA1 benA gene was inactivated by insertion of a thiostrepton resistance gene. The resultant mutant strain, RBD169, neither grew on benzoate nor transformed benzoate, and it did not transform 3-chlorobenzoate. It did, however, exhibit diminished growth on biphenyl and growth repression in the presence of a high concentration of biphenyl (13 mM). These results indicate that the cloned benABCD genes could play an essential role not only in benzoate catabolism but also in biphenyl catabolism in RHA1. Six rhodococcal benzoate degraders were found to have homologs of RHA1 benABC. In contrast, two rhodococcal strains that cannot transform benzoate were found not to have RHA1 benABC homologs, suggesting that many Rhodococcus strains contain benzoate catabolic genes similar to RHA1 benABC.     β(S) haplotypes were studied in 47 non-related patients with sickle-cell anemia from the state of Rio Grande do Norte, Brazil. Molecular analysis was conducted by PCR/RFLP using restriction endonucleases XmnI, HindIII, HincII and HinfI to analyze six polymorphic sites from the beta cluster. Twenty-seven patients (57.5%) were identified with genotype CAR/CAR, 9 (19.1%) CAR/BEN, 6 (12.8%) CAR/CAM, 1 (2.1%) BEN/BEN, 2 (4.3%) CAR/Atp, 1 (2.1%) BEN/Atp and 1 (2.1%) with genotype Atp/Atp. The greater frequency of Cameroon haplotypes compared to other Brazilian states suggests the existence of a peculiarity of African origin in the state of Rio Grande do Norte. PMID:21931513

  18. Dual delivery systems based on polyamine analog BENSpm as prodrug and gene delivery vectors

    Combination drug and gene therapy shows promise in cancer treatment. However, the success of such strategy requires careful selection of the therapeutic agents, as well as development of efficient delivery vectors. BENSpm (N 1, N11-bisethylnorspermine), a polyamine analogue targeting the intracellular polyamine pathway, draws our special attention because of the following reasons: (1) polyamine pathway is frequently dysregulated in cancer; (2) BENSpm exhibits multiple functions to interfere with the polyamine pathway, such as to up-regulate polyamine metabolism enzymes and down-regulate polyamine biosynthesis enzymes. Therefore BENSpm depletes all natural polyamines and leads to apoptosis and cell growth inhibition in a wide range of cancers; (3) preclinical studies proved that BENSpm can act synergistically with various chemotherapy agents, making it a promising candidate in combination therapy; (4) multiple positive charges in BENSpm enable it as a suitable building block for cationic polymers, which can be further applied to gene delivery. In this dissertation, our goal was to design dual-function delivery vector based on BENSpm that can function as a gene delivery vector and, after intracellular degradation, as an active anticancer agent targeting dysregulated polyamine metabolism. We first demonstrated strong synergism between BENSpm and a potential therapeutic gene product TRAIL. Strong synergism was obtained in both estrogen-dependent MCF-7 breast cancer cells and triple-negative MDA-MB-231 breast cancer cells. Significant dose reduction of TRAIL in combination with BENSpm in MDA-MB-231 cells, together with the fact that BENSpm rendered MCF-7 cells more sensitive to TRAIL treatment verified our rationale of designing BENSpm-based delivery platform. This was expected to be beneficial for overcoming drug resistance in chemotherapy, as well as boosting the therapeutic effect of therapeutic genes.     βS haplotypes were studied in 47 non-related patients with sickle-cell anemia from the state of Rio Grande do Norte, Brazil. Molecular analysis was conducted by PCR/RFLP using restriction endonucleases XmnI, HindIII, HincII and HinfI to analyze six polymorphic sites from the beta cluster. Twenty-seven patients (57.5%) were identified with genotype CAR/CAR, 9 (19.1%) CAR/BEN, 6 (12.8%) CAR/CAM, 1 (2.1%) BEN/BEN, 2 (4.3%) CAR/Atp, 1 (2.1%) BEN/Atp and 1 (2.1%) with genotype Atp/Atp. The greater frequency of Cameroon haplotypes compared to other Brazilian states suggests the existence of a peculiarity of African origin in the state of Rio Grande do Norte. PMID:21931513

    The 2005 L'ORÉAL-UNESCO award for women in physics recognized Zohra Ben Lakhdar's contributions to research in Tunisia. But when Professor Ben Lakhdar was a young girl in 1950s Tunisia, girls did not go to school beyond the elementary grades, and she found herself under the tutelage of her mother learning how to take care of a family and home. Tunisia's independence in 1956 changed that, and Professor Ben Lakhdar soon became the only girl in an all-boys' college. In 1978 when she returned to Tunisia after earning her PhD in Paris, fewer than 10 Tunisians were doing research. But the number of students in the country was increasing and trained teachers were needed. Developing the capability to do research in Tunisia was urgent. So Professor Ben Lakhdar built a research laboratory in Tunisia. This paper tells the story.

    Mycoplasma mycoides subsp. mycoides (Mmm) is the causative agent of contagious bovine pleuropneumonia (CBPP). The virulent Mmm Ben-1 strain was isolated from the lung of a CBPP-infected cow in China in the 1950s. To attenuate the virulence of the Ben-1 strain and preserve its protective ability, the isolate was re-isolated after inoculation into the testicles of rabbits and into the rabbit thorax. As a result, after the subsequent isolates were continuously passaged 468 times in rabbits, its pathogenicity to cattle decreased. However, the molecular mechanisms leading to attenuation of the Mmm Ben-1 remain unknown. We compared the entire genomes of the Ben-1 strain and the 468 th generation strain passaged in rabbits (Ben-468) and discovered that a putative protein gene named p19 was absent from the Ben-468 strain. The p19 gene was cloned and expressed in Escherichia coli to obtain recombinant P19 (rP19). Western blot analysis demonstrated that the P19 protein is detected in the cell-membrane fraction, the cell-soluble cytosolic fraction and whole-cell lysate of the Mmm Ben-1 strain. The rP19 can interact with international standard serum against CBPP. Immunostaining visualised via confocal laser scanning microscopy indicated that P19 is able to adhere to embryonic bovine lung (EBL) cells, and this finding was also confirmed by a sandwich ELISA. We also found that anti-rP19 serum could inhibit the adhesion of the Mmm Ben-1 total proteins to EBL cells. PMID:26806796

  17. Evaluating Weight of Evidence in the Mystery of Balkan Endemic Nephropathy

    Balkan Endemic Nephropathy (BEN) is a chronic, progressive wasting disease of the kidneys, endemic in certain rural regions of the Balkan nations Croatia, Serbia, Bulgaria, and Romania. It is irreversible, and ultimately fatal. Though this disease was first described in the 1920s, its causes have been a mystery and a source of much academic and clinical contention. Possible etiologic agents that have been explored include exposure to metals and metalloids, viruses and bacteria, and the environmental toxins aristolochic acid (AA) and ochratoxin A (OTA). Aristolochic acid is a toxin produced by weeds of the genus Aristolochia, common in Balkan wheat fields. Aristolochia seeds may intermingle with harvested grains and thus inadvertently enter human diets. Ochratoxin A is a mycotoxin (fungal toxin) common in many foods, including cereal grains. In this study, we analyzed the weight of evidence for each of the suspected causes of BEN using the Bradford Hill Criteria (BHC): nine conditions that determine weight of evidence for a causal relationship between an agent and a disease. Each agent postulated to cause BEN was evaluated using the nine criteria, and for each criterion was given a rating based on the strength of the association between exposure to the substance and BEN. From the overall available scientific evidence for each of these suspected risk factors, aristolochic acid is the agent with the greatest weight of evidence in causing BEN. We describe other methods for testing causality from epidemiological studies, which support this conclusion of AA causing BEN. PMID:24954501

  18. Betaine protects cerebellum from oxidative stress following levodopa and benserazide administration in rats

    Objective(s): The aim of the present study was to evaluate antioxidant and methyl donor effects of betaine in cerebellum following levodopa and benserazide administration in rats. Materials and Methods: Sprague-Dawley male rats were treated with levodopa (LD), betaine (Bet), levodopa plus betaine (LD/Bet), levodopa plus benserazide (LD/Ben), levodopa plus betaine-benserazide (LD/Bet-Ben), and the controls with vehicle for 10 consecutive days, orally. Results: Treatment of rats with LD and benserazide significantly increased total homocysteine in plasma of the LD/Ben group when compared to the other groups. Lipid peroxidation of cerebellum increased significantly in LD-treated rats when compared to the other groups. In contrast, glutathione peroxidase activity and glutathione content in cerebellum were significantly higher in the betaine-treated rats when compared to the LD and LD/Ben groups. Serum dopamine concentration increased significantly in LD-treated rats in comparison with the LD/Ben group. LD/Bet-treated rats also demonstrated significantly higher dopamine levels when compared to the LD/Ben group. Conclusion: We observed valuable effects of Bet in combination with LD and benserazide, which routinely were used for Parkinson’s disease (PD) treatment, in experimentally-induced oxidative stress and hyperhomocysteinemia in rats. Therefore, it seems that Bet is a vital and promising agent regarding PD for future clinical trials in humans. PMID:26730328

  19. Angiogenesis in upper tract urothelial carcinoma associated with Balkan endemic nephropathy

    Upper tract urothelial carcinoma (UTUC) associated with Balkan endemic nephropathy (BEN) is characterized by a number of aberrations in cell-cycle regulation and apoptosis. The aim of this study was to detect angiogenesis-related marker(s) specific for BEN UTUC, and to examine the influence of HIF 1α upon angiogenesis and apoptosis in UTUC. Present investigation included 110 patients with UTUC, 50 from BEN region and 60 control tumors. Altered expression of VEGFR1 was more often present in control UTUC than in BEN tumors (p<0.005). It was associated with high grade, low and high stage, solid growth, and metaplastic change of control UTUC. Microvessel density assessed by CD31 (MVD CD31) was significantly higher in UTUC with lymphovascular invasion (p<0.05), and in BEN tumors with papillary growth (p<0.05). Discriminant analysis indicated that BEN and control tumors do not differ significantly in expression of angiogenesis related markers. The most important discriminant variable that determined control UTUC was expression of VEGFR1 (p=0.002). HIF 1α in UTUC significantly correlated with the low stage, papillary growth and expression of Bcl-2, Caspase-3 index, and MVD CD34 (p<0.001; 0.0005; 0.01; 0.005; 0.01, respectively). HIF-1α may be helpful marker in evaluation of UTUC, especially when combined with angiogenesis and apoptosis. PMID:22977664

    The locus control region (LCR), composed of four hypersensitive sites (HS1-4) 5{prime} of the {epsilon} globin gene, confers strong, copy-number dependent expression on globin genes in transgenic mice. Several {beta}-globin gene cluster haplotypes carry the sickle cell gene, and show variable levels of fetal hemoglobin (Hb F) expression in association with DNA sequence differences in HS2, {gamma} and {beta} globin promoters, and {gamma}IVSII: The Senegal (SEN or No. 3) haplotype generally has high (>10%) Hb F, Benin (BEN or No. 19) has intermediate Hb F (but some low and some high), and Banu (BAN or No. 20) generally has low Hb F. Huisman and colleagues have proposed that `factors produced under conditions of hematopoietic stress, together with genetic determinants on the haplotype-3 like LCR sequences, allow for high level expression of {gamma} globin genes`. We have now used slot blot to screen high Hb F (>9.5%) and low Hb F cases for two of the three HS2 point mutations described by Oener et al. Comparing eight high Hb F BEN/BEN with two low Hb F BEN/BEN, all ten had the BEN mutations considered by Oener et al. to be associated with low Hb F. Comparing three high Hb F BEN/BAN with two low Hb F BEN/BAN, all five were heterozygous at three positions; this is consistent with BEN having G and T and BAN having A at both positions. DNA sequencing of HS2 for BAN, which is generally associated with low HB F, showed that the point mutations at all three positions were those seen in SEN (generally high Hb F); only the AT repeat region showed major differences, confirming results of Huisman and colleagues. Hence, if there is any effect of HS2 of the Senegal sickle cell haplotype in causing elevated Hb F under hematopoietic stress, it must be due to specific variation in the AT repeat region, which Oener et al. have suggested may bind a silencer.

  1. Constant sediment budget of Mekong delta shorelines: implications from long-term shoreline changes

    The sustainability of delta shorelines requires the sufficient sediment supply that is equal to or more than the sediment removal by tides and waves. The Mekong River delta, southern Vietnam, is a mixed-energy, complicated system with multiple delta plain shorelines that share the sediment supply from the river. We reconstructed the long-term shoreline changes of the Mekong delta over the last 2500 years based on the architecture and chronology of beach ridges on three delta plain in Ben Tre (North, Central, and South Ben Tre), and a delta plain in Tra Vinh to consider the sediment flux and its contribution to the growth of individual delta plains. While each delta plain shows temporal changes in growth rate, the sum of these four plains is nearly constant. This implies that the net sediment supply to the shorelines is constant, and that the geomorphological changes at distributary branches cause changes in sediment supply to each shoreline compartment. The distributary between North and Central Ben Tre was choked sometime between 500 and 1000 years ago, causing the rapid and stagnant shoreline progradation in North and Central Ben Tre, respectively. The shoreline of Central Ben Tre however has prograded slowly even after the choking, suggesting the beach sand can be supplied beyond the distributary or onshore from the delta front. Pre-industrial changes in delta geomorphology, based on geological records, improve the understanding of the delta systems and when compared with historical changes, diagnose the post-industrial state of delta.

Here, we identified and characterized the HYPERSENSITIVE TO LATRUNCULIN B1 (HLB1) protein isolated through a forward-genetic screen in Arabidopsis thaliana for mutants with heightened sensitivity to actin-disrupting drugs. HLB1 is a plant-specific tetratricopeptide repeat domain-containing protein of unknown function encoded by a single Arabidopsis gene. HLB1 associated with the trans-Golgi network (TGN)/early endosome (EE) and tracked along filamentous actin, indicating that it could link post-Golgi traffic with the actin cytoskeleton in plants. HLB1 was found to interact with the ADP-ribosylation-factor guanine nucleotide exchange factor, MIN7/BEN1 (HOPM INTERACTOR7/BREFELDIN A-VISUALIZED ENDOCYTIC TRAFFICKING DEFECTIVE1) by coimmunoprecipitation. The min7/ben1 mutant phenocopied the mild root developmental defects and latrunculin B hypersensitivity of hlb1, and analyses of a hlb1/ min7/ben1 double mutant showed that hlb1 and min7/ben1 operate in common genetic pathways. Based on these data, we propose that HLB1 together with MIN7/BEN1 form a complex with actin to modulate the function of the TGN/EE at the intersection of the exocytic and endocytic pathways in plants.     Background: Balkan endemic nephropathy (BEN) is a chronic, tubulointerstitial renal disease often accompanied by urothelial cancer that has a lethality of nearly 100%. Introduction: One of the many factors that have been proposed to play an etiological role in BEN is exposure to organic compounds from Pliocene lignite coal deposits via the drinking water in endemic areas. Objectives: The objective of this study was to systematically evaluate the role of the tenets of the Pliocene lignite hypothesis in the etiology of BEN in order to provide an improved understanding of the hypothesis for colleagues and patients alike. Methods: A comprehensive compilation of the possible limitations of the hypothesis, with each limitation addressed in turn is presented. Results: The Pliocene lignite hypothesis can best account for, is consistent with, or has the potential to explain the evidence associated with the myriad of factors related to BEN. Conclusions: Residents of endemic areas are exposed to complex mixtures containing hundreds of organic compounds at varying doses and their potentially more toxic (including nephrotoxic) and/or carcinogenic metabolites; however, a multifactorial etiology of BEN appears most likely.   5. (abstract) Line Mixing Behavior of Hydrogen-Broadened Ammonia Under Jovian Atmospheric Conditions

    Laboratory spectral data reported last year have been used to investigate the line mixing behavior of hydrogen-broadened ammonia inversion lines. The data show that broadening parameters appearing in the modified Ben-Reuven opacity formalism of Berge and Gulkis (1976) cannot maintain constant values over pressure ranges that include low to moderate pressures and high pressures. Also, they cannot change drastically in value, as in the Spilker (1990) revision of the Berge and Gulkis formalism. It has long been recognized that at low pressures, less than about 1 bar of a Jovian atmospheric mixture, a VVW formalism yields more accurate predictions of ammonia opacity than Ben-Reuven formalisms. At higher pressures the Ben-Reuven formalisms are more accurate. Since the Ben-Reuven lineshape collapses to a VVW lineshape in the low pressure limit, this low pressure inaccuracy of the Ben-Reuven formalisms is surprising. By incorporating various behavior, a new formalism is produced that is more accurate than previous formalisms, particularly in the critical 'transition region' from 0.5 to 2 bars, and that can be used without discontinuity from pressures of zero to hundreds of bars. The new formalism will be useful in such applications as interpretation of radio astronomical and radio occultation data on giant planet atmospheres, and radiative transfer modeling of those atmospheres.

    Broad bean wilt virus 1 (BBWV-1), genus Fabavirus, has a genome composed of two single-stranded positive-sense RNAs of ∼5.8 (RNA1) and 3.4kb (RNA2). Full-length cDNA clones of both genomic RNAs (pBenR1 and pBenR2) from BBWV-1 isolate Ben were constructed under the control of the T7 promoter. In vitro derived capped transcripts were infectious in Nicotiana benthamiana, Chenopodium quinoa and Vicia faba plants. The biological activity of viral transcripts was not affected by extra bases at the 5'-terminus introduced during in vitro transcription. Virions derived from the infectious cDNA clones displayed similar viral infectivity and accumulation, as well as symptom induction as the wild-type BBWV-1 isolate. PMID:26951858

  9. Natural Compounds as Potential Treatments of NF2-Deficient Schwannoma and Meningioma: Cucurbitacin D and Goyazensolide

    Hypothesis Cucurbitacin D and goyazensolide, two plant-derived natural compounds, possess potent growth-inhibitory activity in schwannoma and meningioma cells. Background Currently, no FDA-approved drugs are available for neurofibromatosis type 2 (NF2)-associated schwannomas and meningiomas. Selected natural compounds with antineoplastic activity, such as cucurbitacin and goyazensolide, may be developed as potential treatments for these tumors. Methods The Nf2-deficient mouse schwannoma Sch10545 and human benign meningioma Ben-Men-1 cells were treated with various concentrations of cucurbitacin D and goyazensolide. The effect on cell proliferation was determined using resazurin assays. Flow cytometry was used to assess the cell cycle profiles. Western blot analysis was performed to investigate the expression of various signal molecules related to the cell cycle and the AKT pathway. Results Cucurbitacin D inhibited proliferation of Sch10545 cells (IC50 ~0.75 μM) and Ben-Men-1 cells (IC50 ~0.2 μM). Goyazensolide also reduced cell proliferation of Sch10545 cells (IC50 ~0.9 μM) and Ben-Men-1 cells (IC50 ~1 μM). The G2/M population increased in both Sch10545 and Ben-Men-1 cells treated with cucurbitacin D or goyazensolide around the IC50. Cucurbitacin and goyazensolide substantially reduced the levels of cyclins E and A in treated Sch10545 and Ben-Men-1 cells. Cucurbitacin D also inhibited cyclin B, phospho-AKT and phospho-PRAS40 expression. In addition, goyazensolide reduced the levels of phospho-AKT and NFκB and increased the expression of pro-apoptotic Bim in Sch10545 and Ben-Men-1 cells. Conclusions Both cucurbitacin D and goyazensolide effectively inhibit proliferation of NF2-deficient schwannoma and meningioma cells, suggesting that these natural compounds should be further evaluated as potential treatments for NF2-related tumors.     Advances in haploidentical bone marrow transplantation (h-BMT) have drastically broadened the treatment options for patients requiring BMT. The possibility of significantly reducing the complications resulting from graft-versus-host disease (GvHD) with the administration of post-transplant cyclophosphamide (PT-CY) has substantially improved the efficacy and applicability of T cell-replete h-BMT. However, higher frequency of disease recurrence remains a major challenge in h-BMT with PT-CY. There is a critical need to identify novel strategies to prevent GvHD while sparing the graft-versus-leukaemia (GvL) effect in h-BMT. To this end, we evaluated the impact of bendamustine (BEN), given post-transplant, on GvHD and GvL using clinically relevant murine h-BMT models. We provide results indicating that post-transplant bendamustine (PT-BEN) alleviates GvHD, significantly improving survival, while preserving engraftment and GvL effects. We further document that PT-BEN can mitigate GvHD even in the absence of Treg. Our results also indicate that PT-BEN is less myelosuppressive than PT-CY, significantly increasing the number and proportion of CD11b(+) Gr-1(hi) cells, while decreasing lymphoid cells. In vitro we observed that BEN enhances the suppressive function of myeloid-derived suppressor cells (MDSCs) while impairing the proliferation of T- and B-cells. These results advocate for the consideration of PT-BEN as a new therapeutic platform for clinical implementation in h-BMT. PMID:27030315

    Broadband electrostatic noise (BEN) bursts whose amplitude sometimes reaches about 100 mV m{sup {minus}1} have been observed by the Viking satellite in the dayside auroral zone. These emissions have been shown to be greatly influenced by nonlinear effects and to occur simultaneously with the observation of particle distributions favouring the destabilization of the electron acoustic mode. It is shown that electron acoustic solitons passing by the satellite would generate spectra that can explain the high-frequency part of BEN, above the electron plasma frequency.

    Benjamin P. Flower, a gifted paleoceanographer and marine geologist, supportive colleague, and dedicated educator at the University of South Florida (USF) College of Marine Science (CMS) passed away on 1 July 2012 from complications related to a rare genetic immune dysfunction, Common Variable Immunodeficiency. He was 49 years old. During his brief illness, Ben's love of life and boundless high spirits were an inspiration to his family, friends, and colleagues. He exhibited remarkable courage and kept his sense of humor in face of adversity. Ben's intellectualism and enduring love of science remained intact, even in his last hours.

    Ben Hogan Company's Golf Ball Division, which is based in Elyria, Ohio, had developed concepts and prototypes for new golf balls but was unable to determine exact performance characteristics. Specifically, the company's R&D department wanted to measure the spin rates of experimental golf balls. After the Golf Ball Division requested assistance, researchers and technicians from the NASA Lewis Research Center went to Elyria and conducted several days worth of tests. Ben Hogan is using the test results to improve the spin characteristics of a new ball it plans to introduce to the market.

  14. Dual delivery systems based on polyamine analog BENSpm as prodrug and gene delivery vectors

    Combination drug and gene therapy shows promise in cancer treatment. However, the success of such strategy requires careful selection of the therapeutic agents, as well as development of efficient delivery vectors. BENSpm (N 1, N11-bisethylnorspermine), a polyamine analogue targeting the intracellular polyamine pathway, draws our special attention because of the following reasons: (1) polyamine pathway is frequently dysregulated in cancer; (2) BENSpm exhibits multiple functions to interfere with the polyamine pathway, such as to up-regulate polyamine metabolism enzymes and down-regulate polyamine biosynthesis enzymes. Therefore BENSpm depletes all natural polyamines and leads to apoptosis and cell growth inhibition in a wide range of cancers; (3) preclinical studies proved that BENSpm can act synergistically with various chemotherapy agents, making it a promising candidate in combination therapy; (4) multiple positive charges in BENSpm enable it as a suitable building block for cationic polymers, which can be further applied to gene delivery. In this dissertation, our goal was to design dual-function delivery vector based on BENSpm that can function as a gene delivery vector and, after intracellular degradation, as an active anticancer agent targeting dysregulated polyamine metabolism. We first demonstrated strong synergism between BENSpm and a potential therapeutic gene product TRAIL. Strong synergism was obtained in both estrogen-dependent MCF-7 breast cancer cells and triple-negative MDA-MB-231 breast cancer cells. Significant dose reduction of TRAIL in combination with BENSpm in MDA-MB-231 cells, together with the fact that BENSpm rendered MCF-7 cells more sensitive to TRAIL treatment verified our rationale of designing BENSpm-based delivery platform. This was expected to be beneficial for overcoming drug resistance in chemotherapy, as well as boosting the therapeutic effect of therapeutic genes.     Laboratory experiments with sediments from three shallow Danish lakes were conducted to evaluate the effects of chemical lake restoration products during resuspension. Phosphorus (P) removal, sediment stability, sediment consolidation and color reduction were studied over time. The investigated products were aluminum (Al), Phoslock (a commercial bentonite product coated with lanthanum) and a combination of Al covered with bentonite (Al/Ben). All treatments effectively reduced the P concentration in the water. However, the treatments containing Al reduced the P concentration immediately after resuspension, whereas Phoslock required several days after resuspension to reduce the P concentration. Especially Phoslock, but also Al/Ben, increased the sediment stability threshold by 265% and 101%, respectively, whereas Al had no stabilizing effect. The fresh Al floc was resuspended 5x easier than untreated sediment. The largest consolidation of the sediment occurred with addition of Phoslock, followed by Al/Ben, while Al alone had no effect. Enhanced consolidation may be of importance for macrophyte colonisation of organic sediment. Phoslock improved the light climate moderately by removing color, whereas Al was very effective in removing color. Ben/Al showed intermediate effects on color reduction. These findings are important when decisions are made on restoration method for a specific lake, which may be more or less wind exposed. PMID:20055487
