  1. Multiphase flow modeling in centrifugal partition chromatography.

    The separation efficiency in Centrifugal Partition Chromatography (CPC) depends on selection of a suitable biphasic solvent system (distribution ratio, selectivity factor, sample solubility) and is influenced by hydrodynamics in the chambers. Especially the stationary phase retention, the interfacial area for mass transfer and the flow pattern (backmixing) are important parameters. Their relationship with physical properties, operating parameters and chamber geometry is not completely understood and predictions are hardly possible. Experimental flow visualization is expensive and two-dimensional only. Therefore we simulated the flow pattern using a volume-of-fluid (VOF) method, which was implemented in OpenFOAM®. For the three-dimensional simulation of a rotating FCPC®-chamber, gravitational centrifugal and Coriolis forces were added to the conservation equation. For experimental validation the flow pattern of different solvent systems was visualized with an optical measurement system. The amount of mobile phase in a chamber was calculated from gray scale values of videos recorded by an image processing routine in ImageJ®. To visualize the flow of the stationary phase polyethylene particles were used to perform a qualitative particle image velocimetry (PIV) analysis. We found a good agreement between flow patterns and velocity profiles of experiments and simulations. By using the model we found that increasing the chamber depth leads to higher specific interfacial area. Additionally a circular flow in the stationary phase was identified that lowers the interfacial area because it pushes the jet of mobile phase to the chamber wall. The Coriolis force alone gives the impulse for this behavior. As a result the model is easier to handle than experiments and allows 3D prediction of hydrodynamics in the chamber.     Deep eutectic solvents (DESs) were evaluated as solvents in centrifugal partition chromatography, a liquid-liquid chromatography separation technology. To this end, the partition coefficients of ten natural compounds of different hydrophobicity were determined in non-aqueous biphasic systems containing DES. The influence of the composition of DESs and the presence of water in the biphasic system on the partition coefficient were also examined. In addition, several process relevant physical properties of the biphasic system, such as the density and viscosity of the phases, were measured. A mixture of three to four hydrophobic compounds was successfully separated in a centrifugal partition extractor using a heptane/ethanol/DES biphasic system. PMID:26810802

  3. Methodology for optimally sized centrifugal partition chromatography columns.

    Centrifugal Partition Chromatography (CPC) is a separation process based on the partitioning of solutes between two partially miscible liquid phases. There is no solid support for the stationary phase. The centrifugal acceleration is responsible for both stationary phase retention and mobile phase dispersion. CPC is thus a process based on liquid-liquid mass transfer. The separation efficiency is mainly influenced by the hydrodynamics of the phases in each cell of the column. Thanks to a visualization system, called "Visual CPC", it was observed that the mobile phase can flow through the stationary phase as a sheet, or a spray. Hydrodynamics, which directly governs the instrument efficiency, is directly affected during scale changes, and non-linear phenomena prevent the successful achievement of mastered geometrical scale changes. In this work, a methodology for CPC column sizing is proposed, based on the characterization of the efficiency of advanced cell shapes, taking into account the hydrodynamics. Knowledge about relationship between stationary phase volume, cell efficiency and separation resolution in CPC allowed calculating the optimum cell number for laboratory and industrial scale CPC application. The methodology is highlighted with results on five different geometries from 25 to 5000 mL, for two applications: the separation of alkylbenzene by partitioning with heptane/methanol/water biphasic system; and the separation of peptides by partitioning with n-butanol/acetic acid/water (4/1/5) biphasic system.   4. Solanidine isolation from Solanum tuberosum by centrifugal partition chromatography.

    The aim of this investigation was the preparative isolation of solanidine (aglycone of the two main potato glycoalkaloids: α-chaconine and α-solanine) from fresh Solanum tuberosum (cv. Pompadour) material by implementing a new preparation scheme using centrifugal partition chromatography (CPC). A setup for obtaining solanidine by hydrolysis of the glycoalkaloids found in the skin and sprouts of S. tuberosum was first developed. Then its isolation was carried out by the development of CPC conditions: the solvent system used for separation was ethyl acetate/butanol/water in the ratio 42.5:7.5:50 v/v/v, 0.6 g of crude extract were separated with a 8 mL/min flow rate of mobile phase while rotating at 2500 rpm. A run yielded 98 mg of solanidine (86.7% recovery from the crude extract) in a one-step separation. The purity of the isolated solanidine was over 98%. Thus, CPC has proven to be the method of choice to get solanidine of very high purity from S. tuberosum biomass in large quantities. PMID:23640901

    Centrifugal partition chromatography is a liquid-liquid separation method well adapted for the fractionation or purification of natural compounds from plant extracts. However, following the preparative isolation, the fractions collected must be analysed by high-performance thin-layer chromatography or high-performance liquid chromatography to evaluate their composition and/or their purity. These additional steps are time-consuming and increase the risk of compound degradation. In order to get an instantaneous analysis of fraction content with structural information on the phytochemicals eluted, it is possible to hyphenate on-line centrifugal partition chromatography with mass spectrometry. Depending on the complexity of the extract, two different kinds of centrifugal partition chromatography-mass spectrometry coupling can be performed: centrifugal partition chromatography-mass spectrometry or centrifugal partition chromatography-high-performance liquid chromatography-mass spectrometry coupling. In the first case, one part of the centrifugal partition chromatography effluent is directly introduced in the mass spectrometry ionisation source to identify the eluted compounds, while in the second case, it is directed to a high-performance liquid chromatography-mass spectrometry system where compounds are first separated thanks to high-performance liquid chromatography and then identified using mass spectrometry.   6. Extraction of cocoa proanthocyanidins and their fractionation by sequential centrifugal partition chromatography and gel permeation chromatography.

    Cocoa beans contain secondary metabolites ranging from simple alkaloids to complex polyphenols with most of them believed to possess significant health benefits. The increasing interest in these health effects has prompted the need to develop techniques for their extraction, fractionation, separation, and analysis. This work provides an update on analytical procedures with a focus on establishing a gentle extraction technique. Cocoa beans were finely ground to an average particle size of <100 μm, defatted at 20 °C using n-hexane, and extracted three times with 50 % aqueous acetone at 50 °C. Determination of the total phenolic content was done using the Folin-Ciocalteu assay, the concentration of individual polyphenols was analyzed by electrospray ionization high performance liquid chromatography-mass spectrometry (ESI-HPLC/MS). Fractions of bioactive compounds were separated by combining sequential centrifugal partition chromatography (SCPC) and gel permeation column chromatography using Sephadex LH-20. For SCPC, a two-phase solvent system consisting of ethyl acetate/n-butanol/water (4:1:5, v/v/v) was successfully applied for the separation of theobromine, caffeine, and representatives of the two main phenolic compound classes flavan-3-ols and flavonols. Gel permeation chromatography on Sephadex LH-20 using a stepwise elution sequence with aqueous acetone has been shown for effectively separating individual flavan-3-ols. Separation was obtained for (-)-epicatechin, proanthocyanidin dimer B2, trimer C1, and tetramer cinnamtannin A2. The purity of alkaloids and phenolic compounds was determined by HPLC analysis and their chemical identity was confirmed by mass spectrometry. PMID:27318471

    Three phenylpropanoids, myristicin [1], elemicin [2], and trans-isoelemicin [3], together with two furanocoumarins, oxypeucedanin [4] and oxypeucedanin hydrate [5], have been isolated from the leaves of Diplolophium buchanani by a separation strategy involving the almost exclusive use of centrifugal partition chromatography. All five compounds were antifungal in a tlc bioautography test using Cladosporium cucumerinum. Compounds 1-4 exhibited larvicidal activity against Aedes aegypti. PMID:7760070

    Summary Jungia sellowii (Asteraceae) is a shrub that grows in Southern Brazil and polar extract of its leaves presents anti-inflammatory properties. Cyperane, guaiane, nortrixane, and trixane sesquiterpene types were reported as the main metabolites in Jungia species. This work aims to describe the isolation and identification of sesquiterpenes in the leaves of J. sellowii using liquid–liquid partition and centrifugal partition chromatography. Thus, the crude extract of fresh leaves of J. sellowii was partitioned with hexane, dichloromethane, ethyl acetate and butanol, respectively. The butanol fraction was then subjected to a selected ternary system optimized for the CPC (centrifugal partition chromatography): ethyl acetate–ethanol–water (9:2:10, v/v/v). The separation was carried out isocratically at a flow rate of 25 mL/min at 1200 rpm, affording seven fractions A to G. TLC of fractions B, C and F displayed a single spot corresponding to three new glycosylated sesquiterpenoids. Their structures were established by using spectroscopic data in comparison to those reported in the literature. Furthermore, the isolates were evaluated for their leishmanicidal and cytotoxic effects. No cytotoxic effect was observed against the three cancer cell lines (HL60, JURKAT and REH), but compound 1 showed a weak antiprotozoal activity. Liquid–liquid partition and CPC turned to be a versatile technique of glycoside purification which is environmentally friendly and requires a limited amount of organic solvents. PMID:27340460

    The aim of this study was to investigate the species Symphonia globulifera, a source of polycyclic polyprenylated acyl phloroglucinols such as guttiferone A, which is known to exhibit a variety of biological activities including noticeable antileishmanial properties. Our goal was the identification and the quantification of guttiferone A in different renewable parts of S. globulifera and its preparative isolation. To the best of our knowledge, there is no data concerning its mechanism of action. Consequently, it is particularly interesting to isolate it in gram quantities in order to establish structure activity relationship studies. After performing high-performance liquid chromatography profiles detecting the presence of guttiferone A and proceeding to its quantification, a centrifugal partition chromatography methodology using a two-phase solvent system of cyclohexane/ethyl acetate/methanol/water (20 :  1 :  20 : 1, v/v/v/v) was applied to each extract. In conclusion, a centrifugal partition chromatography system has been developed to ensure a fast, reliable, and scalable way to isolate, with a high level of purity, guttiferone A from five renewable parts of S. globulifera.   10. Alternative separation steps for monoclonal antibody purification: combination of centrifugal partitioning chromatography and precipitation.

    Protein drugs continue to grow both in medicinal importance as in scale of their production. This furthers the interest in separation technologies that have the potential to replace chromatographic steps in a protein purification process. Two such unit operations that are employed in large scale in the chemical industry are extraction and precipitation. Their usefulness for the purification of proteins has been demonstrated, but the integration of such unit operations in a way that generate an output stream of high protein concentration and low process related impurities was missing. In this work, we employ centrifugal partitioning chromatography ('CPC') in combination with precipitation of the protein of interest to purify a cell culture supernatant of a monoclonal antibody producing cell line. Centrifugal partitioning chromatography was used as means of multi-step extraction using aqueous two-phase systems and was able to remove up to 88.2% of host cell protein ('HCP'). The following PEG driven precipitation and resolubilization of the protein of interest was use to condition the CPC output stream to suit subsequent chromatographic steps, to increase mAb concentration, remove the phase forming polymer, further improve HCP clearance, and integrate a low pH hold step for viral clearance. The entire process reduced HCP content by 99.4% while recovering 93% of the protein of interest. High throughput screening techniques were extensively employed during the development of the process.   11. Rapid purification method for fumonisin B1 using centrifugal partition chromatography.

    Fumonisin B1 (FB1) is a highly toxic mycotoxin produced by fungal strains belonging to the Fusarium genus, which can be found mainly in maize products, and is gaining interest in food safety. To produce large amounts of pure FB1, a novel purifying method was developed by using centrifugal partition chromatography, which is a prominent member of the liquid-liquid chromatographic techniques. Rice cultured with Fusarium verticillioides was extracted with a mixture of methanol/water and found to contain 0.87 mg of FB1 per gram. The crude extracts were purified on a strong anion-exchange column and then separated by using a biphasic solvent system consisting of methyl-tert-butyl-ether-acetonitrile-0.1% formic acid in water. The collected fractions were analysed by flow injection-mass spectrometry and high-performance liquid chromatography coupled with Corona-charged aerosol detector and identified by congruent retention time on high-performance liquid chromatography and mass spectrometric data. This method produced approximately 120 mg of FB1 with a purity of more than 98% from 200 g of the rice culture. The whole purification process is able to produce a large amount of pure FB1 for analytical applications or for toxicological studies. PMID:23043634

    Aeruginosin-865 was isolated from cultivated soil cyanobacteria using a combination of centrifugal partition chromatography (CPC) and gel permeation chromatography. The solubility of Aer-865 in different solvents was evaluated using the conductor-like screening model for real solvents (COSMO-RS). The CPC separation was performed in descending mode with a biphasic solvent system composed of water-n-BuOH-acetic acid (5:4:1, v/v/v). The upper phase was used as a stationary phase, whereas the lower phase was employed as a mobile phase at a flow rate of 10 mL/min. The revolution speed and temperature of the separation column were 1700 rpm and 25 degrees C, respectively. Preparative CPC separation followed by gel permeation chromatography was performed on 50 mg of crude extract yielding Aer-865 (3.5 mg), with a purity over 95% as determined by HPLC. The chemical identity of the isolated compound was confirmed by comparing its spectroscopic data (UV, HRESI-MS, HRESI-MS/MS) with those of an authentic standard and data available in the literature.     An innovative procedure coupling pressurized solvent extraction and centrifugal partition chromatography (CPC) used in linear gradient elution mode was developed to isolate two pure germacranolides (9α-hydroxyparthenolide and 9β-hydroxyparthenolide) and to separate flavonoids (nepetin, isorhamnetin and jaceosidin) and chlorophyll pigments from aerial parts of Anvillea radiata (Coss.&Durieu). The two main germacranolides recovered using this method represent 2 and 5% of the dried plant material respectively. These molecules were extracted using accelerated solvent extraction with chloroform. After optimization of the CPC method, a two-phase solvent system composed of heptane/ethyl acetate/methanol/water (1:5:1:5 v/v/v/v) was employed in descending mode to isolate the germacranolides. Then the lower phase of a heptane/ethyl acetate/methanol/water (6:5:6:5 v/v/v/v) system was pumped in descending mode to generate a linear elution gradient, progressively decreasing the mobile phase polarity, that enabled the flavonoid compounds to be separated in the same run. The efficiency of the preparative separation was controlled through RP-HPLC analysis of the obtained fractions using UV, evaporative light scattering and mass spectrometry detection. The structural identification of the two germacranolides purified over 99% was established by (1)H NMR and (13)C NMR. The least abundant flavonoids were identified by mass spectrometry.   14. Purification of vandaterosides from Vanda teres (Orchidaceae) by stepwise gradient centrifugal partition chromatography.

    Vandaterosides are polar glucosyloxybenzyl eucomate derivatives found in Vanda teres (Orchidaceae), which display biological activities that slow the skin ageing process. In order to obtain larger quantities to allow us to go further in the bioassays, the hydroalcoholic extract of aerial parts (leaves and stems) of V. teres were fractionated by centrifugal partition chromatography, combining isocratic, gradient, and dual elution modes. The first fractionation was performed on the extract maintained in the stationary phase as water saturated in butanol, while increasing the polarity of the mobile phase by changing the proportions of ethyl acetate/1-butanol/water, in order to obtain two enriched fractions. Vandateroside I was then purified by isocratic mode with ethyl acetate/ethanol/water (46:14:40), while vandateroside II was obtained by combining isocratic elution with ethyl acetate/isopropanol/water (30:20:50) followed by a multiple dual mode with ethyl acetate/ethanol/water (46:14:40). In this manner, hundreds of milligrams of vandateroside I and II were recovered from 10 g of V. teres extract. PMID:26104238

  15. Preparative separation of major xanthones from mangosteen pericarp using high-performance centrifugal partition chromatography.

    Mangosteen fruit pericarp (MFP) is a rich source of xanthones, which has shown remarkable pharmacological activities. To isolate xanthones, previous methods included labor intensive and time-consuming solid-phase extractions (Sephadex LH20, silica gel) and sequential solvent extraction. In this study, major xanthones (alpha- and gamma-mangostins) in MFP were isolated at high purity in one step utilizing high-performance centrifugal partition chromatography with solvent system composed of petroleum ether, ethyl acetate, methanol and water (10:5:5:1). In one run, 200 mg crude extract of MFP was injected and 55.4 mg alpha-mangostin and 12.4 mg gamma-mangostin were obtained with the purity of 93.6 and 98.4%, respectively. The yields of them were 86.3 and 76.3%, respectively. As alpha- and gamma-mangostins are reported to show potent antioxidant, anti-inflammatory and anticancer activities, this method can be used for the large-scale production of them for future in vitro and in vivo biological studies.   16. Preparative purification of plasmin activity stimulating phenolic derivatives from Gastrodia elata using centrifugal partition chromatography.

    Gastrodia rhizome, a dried and steamed tuber of Gastrodia elata Blume (Orchidaceae), has been traditionally used in Korea, China and Japan for the treatment of neurological and nervous disorders such as headaches, dizziness, vertigo and convulsive illnesses. The ethyl acetate and water extracts of G. elata stimulated plasmin activity. The active ethyl acetate fraction was subjected to centrifugal partition chromatography (CPC) with a two-phase solvent system, composed of n-hexane-ethyl acetate-methanol-water (3:7:4:6, v/v) followed by semi-preparative HPLC purification to separate active compounds and the water fraction was purified by Diaion HP-20 resin and semi-preparative HPLC. In ethyl acetate extract, 4-hydroxybenzyl alcohol (1), 4-hydroxybenzoic acid (2), 4-hydroxybenzaldehyde (3), 4-ethoxymethylphenol (4), 4,4'-oxybis(methylene)diphenol (5) and 4,4'-methylenediphenol (6) were obtained with high purities. Parishin (7) and parishin B (8) were isolated from water extract. Among isolated compounds, 4-hydroxybenzyl alcohol (1), 4-hydroxybenzaldehyde (3) and 4-ethoxymethylphenol (4) significantly stimulated plasmin activity. Copyright © 2015 John Wiley & Sons, Ltd.   17. Pilot-scale ion-exchange centrifugal partition chromatography: purification of sinalbin from white mustard seeds.

    The purification of p-hydroxybenzylglucosinolate (sinalbin) on a multigram scale from a crude aqueous extract of white mustard seeds (Sinapis alba var. concerta) was successfully achieved by scaling up a strong ion-exchange centrifugal partition chromatography (SIXCPC) laboratory procedure. Thus, the one-step sinalbin purification was performed with 2.35 g of crude extract in approximately 170 min (830 mg/h) up to 70.3 g in approximately 160 min (26.3 g/h) by switching from a 200 mL laboratory scale column to a 5.7 L pilot-scale column. The required biphasic solvent system contained ethyl acetate, n-butanol, and water in 3:2:5 v/v/v proportions, Aliquat 336 (trioctylmethyl ammonium chloride) was added to the organic stationary phase (80 mM) and acted as ion-exchanger. Potassium iodide in the aqueous mobile phase (80 mM) was used as sinalbin displacer. The 28.5 mass scale factor arose from the increase in mobile phase flow-rate (from 2 to 50 mL/min), from the higher mass of injected white mustard seed extract (from 12 to 350 g), and from the calculated productivity (from 830 mg to 26.3 g). These results demonstrate that industry scale production of glucosinolates is easily performed by SIXCPC, thus providing pure reference standards for pharmacology studies. PMID:19479767

  18. Intensified Separation of Steviol Glycosides from a Crude Aqueous Extract of Stevia rebaudiana Leaves Using Centrifugal Partition Chromatography.

    Aqueous extracts of Stevia rebaudiana leaves have been approved since 2008 by the Joint Expert Committee for Food Additives as sugar substitutes in many food and beverages in Western and Far East Asian countries. The compounds responsible for the natural sweetness of Stevia leaves include a diversity of diterpenoid glycosides derived from a steviol skeleton. These steviol glycosides also exhibit a low calorific value as well as promising therapeutic applications, particularly for the treatment of sugar metabolism disturbances. In this work, centrifugal partition chromatography is proposed as an efficient technical alternative to purify steviol glycosides from crude aqueous extracts of Stevia leaves on a multigram scale. Two different commercial instruments, including an ASCPC250® and a FCPE300® made of columns containing 1890 and 231 twin-cells, respectively, were evaluated and compared. All experiments were performed with a polar biphasic solvent system composed of ethyl acetate, n-butanol, and water in a gradient elution mode. When using the 1890 partition cell centrifugal partition chromatography column of 250 mL, 42 mg of stevioside, 68 mg of dulcoside A, and 172 mg of rebaudioside A, three major constituents of the initial extract were obtained from 1 g of the initial mixture at purities of 81%, 83%, and 99%, respectively. The productivity was further improved by intensifying the procedure on the 231 partition cell centrifugal partition chromatography column of 303 mL with the sample mass loading increased up to 5 g, resulting in the recovery of 1.2 g of stevioside, 100 mg of dulcoside A, and 1.1 g of rebaudioside A at purities of 79%, 62%, and 98%, respectively. The structures of the isolated compounds were validated by HPLC-UV, ESI-MS, (1)H, and (13)C NMR analyses.     A critical step in the bioprocessing of sustainable biomass feedstocks, such as sugar beet pulp (SBP), is the isolation of the component sugars from the hydrolysed polysaccharides. This facilitates their subsequent conversion into higher value chemicals and pharmaceutical intermediates. Separation methodologies such as centrifugal partition chromatography (CPC) offer an alternative to traditional resin-based chromatographic techniques for multicomponent sugar separations. Highly polar two-phase systems containing ethanol and aqueous ammonium sulphate are examined here for the separation of monosaccharides present in hydrolysed SBP pectin: l-rhamnose, l-arabinose, d-galactose and d-galacturonic acid. Dimethyl sulfoxide (DMSO) was selected as an effective phase system modifier improving monosaccharide separation. The best phase system identified was ethanol:DMSO:aqueous ammonium sulphate (300gL(-1)) (0.8:0.1:1.8, v:v:v) which enabled separation of the SBP monosaccharides by CPC (200mL column) in ascending mode (upper phase as mobile phase) with a mobile phase flow rate of 8mLmin(-1). A mixture containing all four monosaccharides (1.08g total sugars) in the proportions found in hydrolysed SBP was separated into three main fractions; a pure l-rhamnose fraction (>90%), a mixed l-arabinose/d-galactose fraction and a pure d-galacturonic acid fraction (>90%). The separation took less than 2h demonstrating that CPC is a promising technique for the separation of these sugars with potential for application within an integrated, whole crop biorefinery.     Using both library paper copies and modern electronic copies, every known, published, English-language journal paper that employs either countercurrent or centrifugal partition chromatography solvent systems for natural product purifications has been studied and the solvent systems classified in a comprehensive database. Papers were studied from the earliest found examples containing natural product separations in 1984 until the end of 2014. In total, 2594 solvent systems have been classified, of which 272 are gradient systems. To observe any trends or patterns in the data, the natural product solutes were divided into 21 classes and the solvent systems into 7 different types. The complete database, sorted according to natural product class, is available for download to assist separation scientists in future liquid-liquid chromatography purifications. PMID:26219437

    For the production of bio active compounds, e.g., active enzymes or antibodies, a conserved purification process with a minimum loss of active compounds is necessary. In centrifugal partition chromatography (CPC), the separation effect is based on the different distribution of the components to be separated between two immiscible liquid phases. Thereby, one liquid phase is kept stationary in chambers by a centrifugal field and the mobile phase is pumped through via connecting ducts. Aqueous two phase systems (ATPS) are known to provide benign conditions for biochemical products and seem to be promising when used in CPC for purification tasks. However, it is not known if active biochemical compounds can "survive" the conditions in a CPC where strong shear forces can occur due to the two-phasic flow under centrifugal forces. Therefore, this aspect has been faced within this study by the separation of active laccases from a fermentation broth of Pleurotus sapidus. After selecting a suitable ATPS and operating conditions, the activity yield was calculated and the preservation of the active enzymes could be observed.     In centrifugal partition chromatography (CPC) the separation efficiency is mainly influenced by the hydrodynamic of mobile and stationary phase in the chambers. Thus, the hydrodynamic has to be investigated and understood in order to enhance a CPC separation run. Different chamber geometries have been developed in the past and the influence of several phase systems and CPC operating conditions were investigated for these chambers. However, a direct comparison between the different chamber types has not been performed yet. In order to investigate the direct influence of the chamber design on the hydrodynamic, several chamber designs - partially similar in geometry to commercial available designs - are investigated under standardized conditions in the present study. The results show the influence of geometrical aspects of the chamber design on the hydrodynamic and therewith, on the separation efficiency.     The glucosinolates sinalbin and glucoraphanin were purified by strong ion-exchange displacement centrifugal partition chromatography (SIXCPC). The optimized conditions involved the biphasic solvent system ethyl acetate/n-butanol/water (3:2:5, v/v), the lipophilic anion-exchanger Aliquat 336 (trioctylmethylammonium chloride, 160 and 408 mM) and a sodium iodide solution (80 and 272 mM) as displacer. Amounts as high as 2.4 g of sinalbin and 2.6g of glucoraphanin were obtained in one step in 2.5 and 3.5h respectively, starting from 12 and 25 g of mustard and broccoli seed aqueous extracts, using a laboratory scale CPC column (200 mL inner volume).     Various bioactive phlorotannins of Ecklonia cava (e.g., dieckol, eckol, 6,6-bieckol, phloroglucinol, phloroeckol, and phlorofucofuroeckol-A) are reported. However, their isolation and purification are not easy. Centrifugal partition chromatography (CPC) can be used to efficiently purify the various bioactive-compounds efficiently from E. cava. Phlorotannins are successfully isolated from the ethyl acetate (EtOAc) fraction of E. cava by CPC with a two-phase solvent system comprising n-hexane:EtOAc:methanol:water (2:7:3:7, v/v) solution. The dieckol (fraction I, 40.2mg), phlorofucofuroeckol-A (fraction III, 31.1mg), and fraction II (34.1mg) with 2,7-phloroglucinol-6,6-bieckol and pyrogallol-phloroglucinol-6,6-bieckol are isolated from the crude extract (500 mg) by a one-step CPC system. The purities of the isolated dieckol and phlorofucofuroeckol-A are ⩾90% according to high performance liquid chromatography (HPLC) and electrospray ionization multi stage tandem mass spectrometry analyses. The purified 2,7-phloroglucinol-6,6-bieckol and pyrogallol-phloroglucinol-6,6-bieckol are collected from fraction II by recycle-HPLC. Thus, the CPC system is useful for easy and simple isolation of phlorotannins from E. cava. PMID:24731366

    Separation of potentially bioactive components from foods and plant extracts is one of the main challenges for their study. Centrifugal partition chromatography has been a successful technique for the screening and identification of molecules with bioactive potential, such as steroidal saponins. Agave is a source of steroidal saponins with anticancer potential, though the activity of these compounds in concentrated agave sap has not been yet explored. In this study, fast centrifugal partition chromatography (FCPC) was used coupled with in vitro tests on HT-29 cells as a screening procedure to identify apoptotic saponins from an acetonic extract of concentrated agave sap. The three most bioactive fractions obtained by FCPC at partition coefficients between 0.23 and 0.4 contained steroidal saponins, predominantly magueyoside b. Flow cytometry analysis determined that the fraction rich in kammogenin and manogenin glycosides induced apoptosis, but when gentrogenin and hecogenin glycosides were also found in the fraction, a necrotic effect was observed. In conclusion, this study provides the evidence that steroidal saponins in concentrated agave sap were potential inductors of apoptosis and that it was possible to separate them using fast centrifugal partition chromatography.     Xanthones are well known for their interesting phytochemical properties, which make them attractive to the pharmaceutical and medicinal industry. We have therefore developed a method to analyse the major xanthones in Garcina mangostana. The xanthones were extracted by pressurized liquid extraction with ethanol and separated at the semi-preparative scale by centrifugal partition chromatography (CPC) with a biphasic solvent system consisting of heptane/ethyl acetate/methanol/water (2:1:2:1, v/v/v/v). A CPC-electrospray ionisation MS coupling was performed and used to simultaneously separate and identify the compounds. Thanks to a variable flow splitter and an additional stream of ethanol/1 mol L(-1) ammonium acetate (95:5, v/v), all the compounds were ionised, detected and monitored whatever the solvents used in mobile phase for the CPC separation.     Barks from conifers and broadleaved trees constitute abundant wastes generated from wood harvesting and logging activities. Extracts of such residues obtained from Alnus trees have been reported as interesting resources with potent antibacterial activities. The present study aims to determine the antimicrobial activity of a crude methanol extract prepared from the bark of Alnus glutinosa against a panel of 22 bacteria and yeasts and to optimize a purification method enabling the high production of the most active substances. Fractionation of the crude extract was performed by Centrifugal Partition Chromatography (CPC) using a three-phase solvent system composed of n-heptane, methyl-ter-butyl ether, acetonitrile and water. The major known compounds contained in the fractions produced by CPC were chemically profiled by (13)C NMR dereplication, resulting in the unambiguous identification of oregonin, hirsutanonol, betulinic acid, and alusenone 1a. The antibacterial evaluation of the fractions by bioautography on Staphylococcus aureus revealed that oregonin, in addition to being the major metabolite of the crude extract (∼32% w/w), was the most active with an antibacterial inhibitory effect comparable to antibiotics. The purification of oregonin was optimized at the laboratory-scale by CPC. A single injection of 3.7g of crude extract resulted in a recovery of 72% (850mg) of the available oregonin at purity higher than 94%. PMID:27428455

    Nowadays, a large portion of synthetic products (active cosmetic and therapeutic ingredients) have their origin in natural products. Kniphofia uvaria is a plant from Africa which has proved in the past by in-vivo tests an antioxidant activity due to compounds present in roots. Recently, we have observed anthraquinones in K. uvaria seeds extracts. These derivatives are natural colorants which could have interesting bioactive potential. The aim of this study was to obtain an extract enriched in anthraquinones from K. uvaria seeds which mainly contains glycerides. First, the separation of the seed compounds was studied by using supercritical fluid chromatography (SFC) in the goal to provide a rapid quantification method of these bioactive compounds. A screening of numerous polar stationary phases was achieved for selecting the most suited phase to the separation of the four anthraquinones founded in the seeds. A gradient elution was optimized for improving the separation of the bioactive compounds from the numerous other families of major compounds of the extracts (fatty acids, di- and triglycerides). Besides, a non-selective and green Supercritical Fluid Extraction (SFE) with pure CO2 was applied to seeds followed by a Centrifugal Partition Chromatography (CPC). The CPC system was optimized by using the Arizona phase system, to enrich the extract in anthraquinones. Two systems were selected to isolate the bioactive compounds from the oily extract with varied purity target. The effect of the injection mode for these very viscous samples was also studied. Finally, in order to directly apply a selective process of extraction to the seeds, the super/subcritical fluid extraction was optimized to increase the anthraquinone yield in the final extract, by studying varied modifier compositions and nature, as well as different temperatures and backpressures.     Aqueous two-phase systems composed of imidazolium-based ionic liquids and phosphate salts were evaluated for their applicability in liquid-liquid chromatography. The influence of the nature of ionic liquid anion and cation on the partitioning of bovine serum albumin, lysozyme and myoglobin was investigated. A mixture of K2HPO4 and KH2PO4 in a ratio of 1.82:1 wt/wt was used in all of the tested biphasic systems to adjust the pH to a range of 7-8. The results show that more hydrophobic cations decrease the partition coefficients of the proteins in the biphasic systems and outweigh the effect of the anion on the distribution of the macromolecules. Viscosities and densities of the biphasic systems were in a suitable range for liquid-liquid chromatography. Even though the partition coefficients were too high for a conventional batch operation mode, these aqueous two-phase systems show favorable properties for protein capturing in liquid-liquid chromatographic columns. Additionally, the possible application of ionic liquids as modifiers in polyethylene glycol (PEG)-based aqueous two-phase systems was investigated.     Effective column space and stationary phase retention have been improved by changing the configuration of the helical column originally used for toroidal coil countercurrent chromatography. The use of an equilateral triangular core for the helix column doubles effective column space and retains the stationary phase over 40% of the total column capacity without increasing the column pressure. The present results suggest that the stationary phase retention and the peak resolution will be further improved using new column designs fabricated by a new technology called "laser sintering for rapid prototyping."     The aim of this article was to evaluate the influence of the column design of a hydrostatic support-free liquid-liquid chromatography device on the process efficiency when the strong ion-exchange (SIX) development mode is used. The purification of p-hydroxybenzylglucosinolate (sinalbin) from a crude aqueous extract of white mustard seeds (Sinapis alba L.) was achieved on two types of devices: a centrifugal partition chromatograph (CPC) and a centrifugal partition extractor (CPE). They differ in the number, volume and geometry of their partition cells. The SIX-CPE process was evaluated in terms of productivity and sinalbin purification capability as compared to previously optimized SIX-CPC protocols that were carried out on columns of 200 mL and 5700 mL inner volume, respectively. The objective was to determine whether the decrease in partition cell number, the increase in their volume and the use of a "twin cell" design would induce a significant increase in productivity by applying higher mobile phase flow rate while maintaining a constant separation quality. 4.6g of sinalbin (92% recovery) were isolated from 25 g of a crude white mustard seed extract, in only 32 min and with a purity of 94.7%, thus corresponding to a productivity of 28 g per hour and per liter of column volume (g/h/LV(c)).     An improved apparatus and a procedure are described by which the migration of sample components in column chromatography is accelerated by centrifugal force, thereby making it possible to use beds of densely packed gel prepared with ultrafine silica. This technique was used to resolve components of certain lipid mixtures where other methods have failed, and it has been found generally useful as an adjunct to other methods for the fractionation of lipids. Biologically active phosphoglycolipids from Mycobacterium tuberculosis and a phosphatidylglycerol-like substance from Mycoplasma pneumoniae which formed single spots on thin-layer chromatographic plates were each found to contain a major and several minor components by centrifugal chromatography. The method enabled us to isolate individual components of Wax D from M. tuberculosis rather than a spectrum of components. Minor components were resolved which, although present in insufficient quantity to influence results of chemical analyses, may be responsible for biological activity. The apparatus provides an essentially closed system which reduces highly volatile solvents to minimal evaporation during the chromatographic process. Samples are applied in solution and are not allowed to dry on the columns until after separation has been achieved. Consequently, polar, labile microbial lipids can be resolved without the use of harsh reagents which destroy some of their properties. Single components may be harvested by cutting and removing appropriate segments of the larger chromatograms or by eluting them from the columns. Images PMID:4314479

    The toroidal coil using an equilateral triangular core has improved both retention of the stationary phase and peak resolution of the conventional toroidal coil in centrifugal countercurrent chromatography. To further improve the retention of stationary phase and peak resolution, a novel zigzag toroidal coil was designed and the performance of the system was evaluated at various flow rates. The results indicated that both retention of stationary phase and peak resolution were improved as the flow rate was decreased. Modification of the tubing by pressing at given intervals with a pair of pliers improved the peak resolution without increasing the column pressure. All these separations were performed under low column pressure indicating the separation can be further improved by increasing the column length and/or revolution speed without damaging the separation column.     A compact type-I coil planet centrifuge has been developed for performing counter-current chromatography. It has a revolution radius of 10 cm and a column holder height of 5 cm compared with 37 cm and 50 cm in the original prototype, respectively. The reduction in the revolution radius and column length permits application of higher revolution speed and more stable balancing of the rotor which leads us to learn more about its performance and the future potential of type-I coil planet centrifuge. The chromatographic performance of this apparatus was evaluated in terms of retention of the stationary phase (Sf), peak resolution (Rs), theoretical plate (N) and peak retention time (tR). The results of the experiment indicated that increasing the revolution speed slightly improved both the retention of the stationary phase and the peak resolution while the separation time is remarkably shortened to yield an excellent peak resolution at a revolution speed of 800 rpm. With a 12 ml capacity coiled column, DNP-glu, DNP-β-ala and DNP-ala were resolved at Rs of 2.75 and 2.16 within 90 min at a flow rate of 0.4 ml/min.     High level liquid waste (HLLW) produced from the reprocessing of the spent nuclear fuel still contains moderate amounts of uranium, transuranium (TRU) actinides, (90)Sr, (137)Cs, etc., and thus constitutes a permanent hazard to the environment. The partitioning and transmutation (P&T) strategy has increasingly attracted interest for the safe treatment and disposal of HLLW, in which the partitioning of HLLW is one of the critical technical issues. An improved total partitioning process, including a TRPO (tri-alkylphosphine oxide) process for the removal of actinides, a CESE (crown ether strontium extraction) process for the removal of Sr, and a CECE (calixcrown ether cesium extraction) process for the removal of Cs, has been developed to treat Chinese HLLW. A 160-hour hot test of the improved total partitioning process was carried out using 72-stage 10-mm-dia annular centrifugal contactors (ACCs) and genuine HLLW. The hot test results showed that the average DFs of total α activity, Sr and Cs were 3.57 × 10(3), 2.25 × 10(4) and 1.68 × 10(4) after the hot test reached equilibrium, respectively. During the hot test, 72-stage 10-mm-dia ACCs worked stable, continuously with no stage failing or interruption of the operation. PMID:25016455

    The partitioning of star branched polymers into a slit pore at three different chromatography conditions, namely, size exclusion chromatography (SEC), liquid chromatography at the critical condition (LCCC), and liquid adsorption chromatography (LAC) have been investigated with lattice Monte Carlo simulations. Two different chain models are used: random walks (RW) that have no excluded volume interaction and self-avoiding walks (SAW) that have excluded volume interaction. The simulation data obtained for the two chain models are compared to illustrate the effect of excluded volume interactions on the partitioning of star branched polymers. The two most outstanding effects observed due to the introduction of excluded volume interactions are: (i) stars with a high number of arms can be excluded from the pore at condition corresponding to the LCCC of the linear polymers; (ii) the partition coefficient of stars in LAC mode is not dependent only on the total number of monomers on the chain. These effects illustrated by the current study should be taken into account when interpreting experimental chromatography data for branched polymers. PMID:20728895

    A novel ellipsoid column was designed for centrifugal counter-current chromatography. Performance of the ellipsoid column with a capacity of 3.4 mL was examined with three different solvent systems composed of 1-butanol-acetic acid-water (4:1:5, v/v) (BAW), hexane-ethyl acetate-methanol-0.1 M HCl (1:1:1:1, v/v) (HEMH), and 12.5% (w/w) PEG1000 and 12.5% (w/w) dibasic potassium phosphate in water (PEG-DPP) each with suitable test samples. In dipeptide separation with BAW system, both stationary phase retention (Sf) and peak resolution (Rs) of the ellipsoid column were much higher at 0° column angle (column axis parallel to the centrifugal force) than at 90° column angle (column axis perpendicular to the centrifugal force), where elution with the lower phase at a low flow rate produced the best separation yielding Rs at 2.02 with 27.8% Sf at a flow rate of 0.07 ml/min. In the DNP-amino acid separation with HEMW system, the best results were obtained at a flow rate of 0.05 ml/min with 31.6% Sf yielding high Rs values at 2.16 between DNP-DL-glu and DNP-β-ala peaks and 1.81 between DNP-β-ala and DNP-L-ala peaks. In protein separation with PEG-DPP system, lysozyme and myolobin were resolved at Rs of 1.08 at a flow rate of 0.03 ml/min with 38.9% Sf.     The performance of protein separation using the figure-8 column configuration in centrifugal counter-current chromatography was investigated under various flow rates and revolution speeds. The separation was performed with a two-phase solvent system composed of polyethylene glycol 1000/potassium phosphate each at 12.5% (w/w) in water and with lysozyme and myoglobin as test samples. In order to improve tracing of the elution curve, a hollow fiber membrane dialyzer was inserted at the inlet of the UV detector. The results showed that the retention of stationary phase (Sf) and resolution (Rs) increased with decreased flow rate and increased revolution speed. The highest Rs of approximately 1 was obtained at a flow rate of 0.01 mL/min under a revolution speed of 1200 rpm with a 3.4 ml capacity column. PMID:22100551

    The toroidal coil using an equilateral triangular core and zigzag pattern column have improved both retention of the stationary phase and peak resolution of the conventional toroidal coil in centrifugal counter-current chromatography. To further improve the retention of stationary phase and peak resolution, a novel saw tooth column was designed and the performance of the system was evaluated at various flow rates. The results indicated that both retention of the stationary phase and peak resolution were improved as the flow rate was decreased and at a flow rate of 0.005 ml/min the resolution is remarkably increased. Modification of the tubing called flat-twisted tubing further improved the peak resolution without increasing the column pressure. With a decreased column length at a capacity of about 0.2 ml, resolution of the saw tooth column was 1.02. PMID:20543965

    The conventional toroidal coil in centrifugal countercurrent chromatography has a low level of stationary phase retention, since a half of each helical turn is entirely occupied by the mobile phase. In order to cope with this problem, several new column designs including zigzag, saw-tooth and figure-8 patterns have been introduced and their performance was compared in terms of retention of the stationary phase (Sf), peak resolution (Rs), theoretical plate number (N) and column pressures. Overall results of experiments indicate that the figure-8 column yields the highest Rs when the lower phase is used as the mobile phase. Since the column pressure of all these new columns are much lower than that in the traditional toroidal coil column, the separation efficiency can be improved using a long separation column without a risk of column damage by high back pressure.     Plants have evolved a unique way of partitioning the cytoplasm of dividing cells: Instead of forming a contractile ring that constricts the plasma membrane, plant cells target membrane vesicles to the plane of division where the vesicles fuse with one another to form the partitioning membrane. Plant cytokinesis starts in the centre and progresses towards the periphery, culminating in the fusion of the partitioning membrane with the parental plasma membrane. This membrane dynamics is orchestrated by a specific cytoskeletal array named phragmoplast that originates from interzone spindle remnants.     Partition coefficients of organic compounds in four ionic liquids: 1-ethanol-3-methylimidazolium tetrafluoroborate, 1-ethanol-3-methylimidazolium hexafluorophosphate, 1,3-dimethylimidazolium dimethylphosphate and 1-ethyl-3-methylimidazolium diethylphosphate were measured using inverse gas chromatography from 303.3 to 332.55K. The influence of gas-liquid and gas-solid interfacial adsorption of different solutes on ionic liquids was also studied. Most of the polar solutes were retained largely by partition while light hydrocarbons were retained predominantly by interfacial adsorption on the ionic liquids studied in this work.     Anion exchange chromatography (AEX) operated under weak partitioning mode has been proven to be a powerful polishing step as well as a robust viral clearance step in Pfizer's monoclonal antibody (mAb) platform purification process. A multivariate design of experiment (DoE) study was conducted to understand the impact of operating parameters and feedstream impurity levels on viral clearance by weak partitioning mode AEX. Bacteriophage was used initially as a surrogate for neutral and acidic isoelectric point mammalian viruses (e.g., retrovirus and parvovirus). Five different mAbs were used in the evaluation of process parameters such as load challenge (both product and impurities), load pH, load conductivity, and contact time (bed height and flow-rate). The operating ranges obtained from phage clearance studies and Pfizer's historical data were used to define an appropriate operating range for a subsequent clearance study with model retrovirus and parvovirus. Both phage and virus clearance evaluations included feedstreams containing different levels of impurities such as high molecular mass species (HMMS), host cell proteins (HCPs), and host cell DNA. For all the conditions tested, over 5 log10 of clearance for both retrovirus and parvovirus was achieved. The results demonstrated that weak partitioning mode AEX chromatography is a robust step for viral clearance and has the potential to be included as part of the modular viral clearance approach.   10. Effect of electric field on the partitioning behavior of solutes in entropic interaction chromatography.

    In this study, a novel column design with a round cross-section was proposed to be suitable for a transverse electric field (EF). Additionally, two beads for entropic interaction chromatography (EIC) were prepared by grafting glycidyl methacrylate onto Toyopearl HW-65F (T65F) beads. Solute partitioning was then investigated to elucidate the role of graft polymerization with and without an EF. In a T65F column, solute partitioning was attributed to the distinct pore structure in the beads and was governed by pore flow. Under EF, partition coefficients (Kp) for solutes decreased with increasing EF strength. In the two EIC columns, a decrease of Kp was also observed without an EF while the fractionation windows were extended. It was more pronounced in the EIC column with a high grafting density (T65F-H). This was explained by the decrease in the effective pore size of solutes caused by the steric hindrance of polymer chains. Under an EF, the solutes showed different partitioning behaviours in the T65F-H column. With increasing EF strength, Kp for vitamin B12 and myoglobin was decreased. In contrast, Kp for large solutes increased as a result of concentration polarization on the bead surface. Both behaviors were related to the modulation of graft polymerization to residual charge on the matrix and the pore size of the solutes. PMID:23857725

    A method of preparation of rotors with a reversed phase (RP) solid silica gel sorbent layer has been developed for centrifugal preparative chromatography (CPC), also known as rotational planar chromatography (RPC). The rotors consist of binder free RP solid SiO2 layers of different thicknesses packed between two supported circular glass discs and can be used in any appropriate device for centrifugal chromatography, like Chromatotron and CycloGraph. Polar and /or semi-polar compounds with close R(f) values, as well as extracts and column fractions were separated and/or purified in a preparative and/or semi-preparative scale using the RP rotors, eluted with mixtures of aqueous-based solvents.   13. Centrifugal partition extraction in the pH-zone-refining displacement mode: an efficient strategy for the screening and isolation of biologically active phenolic compounds.

    Centrifugal partition extraction (CPE) was developed for the first time in the pH-zone-refining mode to fractionate a crude bark extract of the African tree Anogeissus leiocarpus Guill. & Perr. (Combretaceae). The fractionation process was performed at a flow rate of 20mL/min using a biphasic solvent system composed of methyl tert-butyl ether/acetonitrile/water (4:1:5, v/v/v) in the ascending mode. Sodium hydroxide (40mM) and trifluoroacetic acid (30mM) were used as retainer and displacer agents, respectively. In a single run of 67min, 3g of the initial crude extract were successfully separated into fractions selectively enriched in ionizable triterpenes, ellagic acid derivatives and flavonoids. The antioxidant potential of the initial crude extract, isolated compounds and fraction pools was also evaluated by using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) stable free radical scavenging assay, providing an interesting view about the effect of the degree of substitution of ellagic acid derivatives on their radical scavenging activity. This study will demonstrate that centrifugal partition extraction used in the pH-zone-refining mode can be proposed as an efficient strategy for the rapid screening of natural phenolic compounds.     A simple and fast method namely in-coupled syringe assisted octanol-water partition microextraction combined with high performance liquid chromatography (HPLC) has been developed for the extraction, preconcentration and determination of neonicotinoid insecticide residues (e.g. imidacloprid, acetamiprid, clothianidin, thiacloprid, thiamethoxam, dinotefuran, and nitenpyram) in honey. The experimental parameters affected the extraction efficiency, including kind and concentration of salt, kind of disperser solvent and its volume, kind of extraction solvent and its volume, shooting times and extraction time were investigated. The extraction process was carried out by rapid shooting of two syringes. Therefore, rapid dispersion and mass transfer processes was created between phases, and thus affects the extraction efficiency of the proposed method. The optimum extraction conditions were 10.00 mL of aqueous sample, 10% (w/v) Na2SO4, 1-octanol (100µL) as an extraction solvent, shooting 4 times and extraction time 2min. No disperser solvent and centrifugation step was necessary. Linearity was obtained within the range of 0.1-3000 ngmL(-1), with the correlation coefficients greater than 0.99. The high enrichment factor of the target analytes was 100 fold and low limit of detection (0.25-0.50 ngmL(-1)) could be obtained. This proposed method has been successfully applied in the analysis of neonicotinoid residues in honey, and good recoveries in the range of 96.93-107.70% were obtained. PMID:25882403

    The toroidal column using a zigzag pattern has been improved in both retention of the stationary phase and peak resolution. To further improve the retention of stationary phase and peak resolution, a series of novel geometric designs of tubing (plain, mid-clamping, flattened and flat-twisted tubing) was evaluated their performance in CCC. The results showed that the tubing which was flattened vertically against centrifugal force (vert-flattened tubing) produced the best peak resolution among them. Using vert-flattened tubing a series of experiments was performed to study the effects of column capacity and sample size. The results indicated that a 0.25 ml capacity column is ideal for analysis of small amount samples. PMID:20454530

  16. Analysis of steric partition behavior of molecules in membranes using statistical physics. Application to gel chromatography and electrophoresis.

    The principles of statistical physics are used to formulate general expressions for the steric partition behavior of molecules in both random and ordered membrane structures that may be applied to any shape of the solute and/or the volume-excluding element of the membrane. These expressions fully define partitioning in terms of the volume excluded to point molecules and to finite-sized molecules. The mean effective exclusion volume for a molecule is calculated as a function of a global interaction energy, which varies with position, conformation, and orientation of the molecule. It allows consideration of electrostatic and other nonsteric factors. To test the model, specific partition functions are derived for several simple geometries describing the membrane and solute. Frequently, the derived expressions agree with past analyses; however, a new expression describing partitioning within an random network of fibers is derived. It agrees with past results only in the limit of low exclusion volumes. With greater volume exclusions, past results greatly overestimate the partition function. It is applied to gel electrophoresis and chromatography and survives testing with available experimental data. Unlike past analyses, it predicts nonlinear Ferguson plots for agarose gel electrophoresis. In addition, an analytical expression predicting the minimum radius of a sphere excluded from a random fiber matrix is derived, tested, and found to agree with experimental data. PMID:3148335

  17. Determination of octanol-water partition coefficients for carbonate esters and other small organic molecules by microemulsion electrokinetic chromatography.

    Microemulsion electrokinetic chromatography (MEEKC) was assessed as a tool for determination of octanol-water partition coefficients using 34 solutes encompassing 8 carbonate esters. It was confirmed that microemulsions containing 1.44-2.88% w/w SDS, 6.49% w/w 1-butanol, and 0.82% w/w n-heptane constitute a good model of octanol-water partitioning in the pH range of 1.4-7.4. Use of the migration index concept led to improved repeatability of the MEEKC method compared to the use of retention factors. Using a dynamical coating, a high electroosmotic flow at pH 1.4 and 4.75 was achieved expanding the practical pH working range of the MEEKC system. The correlation obtained between the migration index and log P was unaffected by pH indicating that the properties of the microemulsion droplets and, thus, partitioning are independent of pH. No evidence for congeneric behavior was found for the sample set comprising solutes with different hydrogen bonding properties suggesting that simple reference compounds can be used as calibrators. Lipophilicity estimates for the series of carbonate esters were obtained. The increase in lipophilicity with chain length was smaller than expected from the Hansch substituent constant, pi.     Two novel methods are reported for measuring octanol/water partition rates of pesticides. A liquid scintillation counting (LSC) method was developed for automated monitoring of /sup 14/C-labeled pesticides partitioning in biphasic water/octanol cocktail systems with limited success. A high performance liquid chromatography (HPLC) method was developed for automated partition rate monitoring of several constituents in a pesticide mixture, simultaneously. The mean log Kow +/- SD determined from triplicate experimental runs were for: 2,4-D-DMA (2,4-dichlorophenoxyacetic acid dimethylamine), 0.65 +/- .17; Deet (N,N-diethyl-m-toluamide), 2.02 +/- .01; Guthion (O,O-dimethyl-S-(4-oxo-1,2,3-benzotriazin-3(4H)-ylmethyl) phosphorodithioate), 2.43 +/- .03; Methyl-Parathion (O,O-dimethyl-O-(p-nitrophenyl) phosphorothioate), 2.68 +/- .05; and Fenitrothion (O,O-dimethyl O-(4-nitro-m-tolyl) phosphorothioate), 3.16 +/- .03. A strong positive linear correlation (r = .9979) was obtained between log Kow and log k' (log Kow = 2.35 (log k') + 0.63). The advantages that this automated procedure has in comparison with the standard manual shake-flask procedure are discussed.

    The quantification of proteins adsorbed at the oil-in-water interface is often difficult since it requires separation of fat globules from the aqueous phase that may damage the fat globule size and/or modify the interfacial composition. Front-face fluorescence spectroscopy was used to characterize the protein partitioning between the aqueous and oil phases of emulsions without separating these two phases. Different emulsions based on skim milk powder (SMP), two mono- and di-glyceride (MDG) mixtures (saturated and partially unsaturated), and three fats (hydrogenated and refined coconut oils and refined palm oil) were studied. The impact of an ageing period (24 h at 4 degrees C) was also investigated to typify the first step of ice cream processing. The emulsions were characterized for protein partitioning, immediately following emulsification and after ageing, using the Bradford spectrophotometric method, applied to the aqueous phase recovered after emulsion centrifugation. In parallel, the emulsions were characterized by their tryptophan emission fluorescence spectra. The area of the peaks at 333 nm, of the fourth-derivative fluorescence spectra corresponding to the amount of proteins present in the aqueous phase of emulsions, was well correlated with the Bradford measurements (r2=0.91). This amount was also calculated from the fluorescence calibration curve obtained with SMP in solution. In conclusion, front-face fluorescence spectroscopy appeared to be a powerful and simple technique allowing the quantification of different populations of protein in an emulsified system, i.e., in the aqueous phase and loaded at the fat globule interface.     Two solvent extraction technologies under development in Russia for the partitioning of radionuclides from radioactive wastes were tested at the Idaho Chemical Processing Plant (ICPP) with simulated high-activity liquid waste (HAW) on a continuous basis using 24 stages of 2-cm diameter centrifugal contactors. Two flowsheet tests were conducted with chlorinated cobalt dicarbollide (ChCoDiC) to evaluate the separation of cesium and strontium from ICPP HAW. Also, a flowsheet test was performed with a derivative of phosphine oxide (POR) to evaluate the separation of actinides, rare earths, and technetium from ICPP HAW. All experiments utilized a non-radioactive HAW simulant prepared to emulate the macro (or matrix) constituents of actual ICPP HAW at their average tank composition. The behavior of the species of interest was monitored using the stable forms of Sr and Cs, europium as a surrogate for americium, and rhenium as a surrogate for technetium. Removal efficiencies and distribution coefficients were determined for each flowsheet at steady-state conditions. Results of this testing indicate the POR and ChCoDiC processes can be used to effectively treat ICPP HAW. This series of tests is a continuation of ongoing efforts to evaluate the applicability of these Russian developed technologies to U.S. nuclear wastes under the auspices of a joint program between the U.S. Department of Energy and the Russian Ministry of Atomic Energy.

Extraction was carried out by low temperature partitioning and analyzed by ultrahigh performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS). For a favor extraction yield, the parameters such as temperature and solvent were optimized. The result showed that sample would be easily frozen and separated using acetonitrile under -20°C for 10 min. The limit of detection (LOD) was 0.5 μg/kg, and the limit of quantification (LOQ) was 2 and 5 μg/kg for potato and soil, respectively. The influence of paclobutrazol residue in potato was evaluated. The possible contamination of paclobutrazol from surface can be rinsed by distilled water or peeled off, but the paclobutrazol in potato harvest comes mainly from absorption and transport, which could not be removed by peeling. The half-life of paclobutrazol in soil was 20.64 days, and the residue was below 0.22 mg/kg on 50th day after spraying. According to the risk assessment with Need Maximum Daily Intake (NEDI) and Acceptable Daily Intake (ADI), a Maximum Residue Limit (MRL) of paclobutrazol in potato was recommended as 1.0 mg/kg.   6. Isolation of α-Amylase Inhibitors from Kadsura longipedunculata Using a High-Speed Counter-Current Chromatography Target Guided by Centrifugal Ultrafiltration with LC-MS.

    In this study, a high-speed counter-current chromatography (HSCCC) separation method target guided by centrifugal ultrafiltration with high-performance liquid chromatography-mass spectrometry (CU-LC-MS) was proposed. This method was used to analyze α-amylase inhibitors from Kadsura longipedunculata extract. According to previous screening with CU-LC-MS, two screened potential α-amylase inhibitors was successfully isolated from Kadsura longipedunculata extract using HSCCC under the optimized experimental conditions. The isolated two target compounds (with purities of 92.3% and 94.6%) were, respectively, identified as quercetin-3-O-rhamnoside (1) and protocatechuic acid (2) based on the MS, UV, and ¹H-NMR spectrometry data. To verify the inhibition of screened compounds, the inhibitory activities of quercetin-3-O-rhamnoside (1) and protocatechuic acid (2) on α-amylase were tested, and it demonstrated that the experimental IC50 values of quercetin-3-O-rhamnoside (1) and protocatechuic acid (2) were 28.8 and 12.5 μmol/L. These results proved that the hyphenated technique using CU-LC-MS and HSCCC was a rapid, competent, and reproductive method to screen and separate potential active compounds, like enzyme inhibitors from the extract of herbal medicines.     Centrifugal precipitation chromatography (CPC) is a separation system that mainly employs a moving concentration gradient of precipitating agent along a channel and solutes of interest undergo repetitive precipitation-dissolution, fractionate at different locations, and elute out from the channel according to their solubility in the precipitating agent solution. We report here for the first time the use of a CPC system for fractionation of protein, RNA, and plasmid DNA in clarified lysate produced from bacterial culture. The cationic surfactant cetyltrimethylammonium bromide (CTAB) was initially used as a precipitating agent; however, all biomolecules showed no differential solubility in the moving concentration gradient of this surfactant and, as a result, no separation of protein, RNA, and plasmid DNA occurred. To overcome this problem, inorganic salts such as NaCl and NH(4)Cl were introduced into solution of CTAB. The protein and RNA were found to have higher solubility with the addition of these salts and separated from the plasmid DNA. Decreasing surface charge density of CTAB upon addition of NaCl and NH(4)Cl was believed to lead to lower surfactant complexation, and therefore caused differential solubility and fractionation of these biomolecules. Addition of CaCl(2) did not improve solubility and separation of RNA from plasmid DNA. PMID:15384057

    The TRUEX process is being evaluated at the Idaho Chemical Processing Plant (ICPP) for the separation of the actinides from acidic radioactive wastes stored at the ICPP. These efforts have culminated in recent demonstrations of the TRUEX process with actual tank waste. The first demonstration was performed in 1996 using 24 stages of 2-cm diameter centrifugal contactors and waste from tank WM-183. Based on the results of this flowsheet demonstration, the flowsheet was optimized and a second flowsheet demonstration was performed. This test also was performed using 2-cm diameter centrifugal contactors and waste from tank WM-183. However, the total number of contactor stages was reduced from 24 to 20. Also, the concentration of HEDPA in the strip solution was reduced from 0.04 M to 0.01 M in order to minimize the amount of phosphate in the HLW fraction, which would be immobilized into a glass waste form. This flowsheet demonstration was performed using centrifugal contactors installed in the shielded hot cell at the ICPP Remote Analytical Laboratory. The flowsheet tested consisted of six extraction stages, four scrub stages, six strip stages, two solvent was stages, and two acid rinse stages. An overall removal efficiency of 99.79% was obtained for the actinides. As a result, the activity of the actinides was reduced from 540 nCi/g in the feed to 0.90 nCi/g in the aqueous raffinate, which is well below the NRC Class A LLW requirement of 10 nCi/g for non-TRU waste. Removal efficiencies of 99.84%, 99.97%, 99.97%, 99.85%, and 99.76% were obtained for {sup 241}Am, {sup 238}Pu, {sup 239}Pu, {sup 235}U, and {sup 238}U, respectively.

    The use of micelles in ionic liquid based gas-chromatography stationary phases was evaluated using equations derived for a "three-phase" model. This model allows the determination of all three partition coefficients involved in the system, and elucidates the micellar contribution to retention and selectivity. Four types of micellar-ionic liquid columns were examined in this study: 1-butyl-3-methylimidazolium chloride with sodium dodecylsulfate or dioctyl sulfosuccinate, and 1-butyl-3-methylimidazolium hexafluorophosphate with polyoxyethylene-100-stearyl ether or polyoxyethylene-23-lauryl ether. The partition coefficients were measured for a wide range of probe molecules capable of a variety of types and magnitudes of interactions. In general, most probe molecules preferentially partitioned to the micellar pseudophase over the bulk ionic liquid component of the stationary phase. Therefore, addition of surfactant to the stationary phase usually resulted in greater solute retention. It is also shown that the selectivity of the stationary phase is significantly altered by the presence of micelles, either by enhancing or lessening the separation. The effects of surfactant on the interaction parameters of the stationary phase are determined using the Abraham solvation parameter model. The addition of sodium dodecylsulfate and dioctyl sulfosuccinate to 1-butyl-3-methylimidazolium chloride stationary phases generally increased the phase's hydrogen bond basicity and increased the level of dispersion interaction. Polyoxyethylene-100-stearyl ether and polyoxyethylene-23-lauryl ether surfactants, however, enhanced the pi-pi/n-pi, polarizability/dipolarity, and hydrogen bond basicity interactions of 1-butyl-3-methylimidazolium hexafluorophosphate to a greater degree than the ionic surfactants with 1-butyl-3-methylimidazolium chloride.     On a partition chromatographic column in which the support is Kieselguhr and the stationary phase is sulfuric acid solution (2 mol/L), three components of compound theophylline tablet were simultaneously eluted by chloroform and three other components were simultaneously eluted by ammonia-saturated chloroform. The two mixtures were determined by computer-aided convolution curve method separately. The corresponding average recovery and relative standard deviation of the six components were as follows: 101.6, 1.46% for caffeine; 99.7, 0.10% for phenacetin; 100.9, 1.31% for phenobarbitone; 100.2, 0.81% for theophylline; 99.9, 0.81% for theobromine and 100.8, 0.48% for aminopyrine.

    An ion-pair reversed-phase partition high-performance liquid chromatography-spectrophotometric method is described for the determination of aluminum in human serum, based on its complexation with 2,2'-dihydroxyazobenzene. The chelate is separated on C18-bonded silica packing by using an aqueous methanol mobile phase containing tetrabutylammonium bromide and is detected with 0.005 AU full-scale at 510 nm. The proposed system offers a simple, sensitive, and selective method. The detection limits defined as twice the standard deviation of the blank signal, are 0.2 micrograms L-1 in pure solution and 6 micrograms L-1 in serum. The sample solution is prepared from 0.4 mL of serum after protein precipitation with hydrochloric acid and methanol. There was good agreement between the values obtained by this method and graphite furnace atomic absorption spectrophotometry. The aluminum concentrations in 121 patients on hemodialysis were 6-201 micrograms L-1. PMID:1759707

  16. Linear modeling of the soil-water partition coefficient normalized to organic carbon content by reversed-phase thin-layer chromatography.

    Soil-water partition coefficient normalized to the organic carbon content (KOC) is one of the crucial properties influencing the fate of organic compounds in the environment. Chromatographic methods are well established alternative for direct sorption techniques used for KOC determination. The present work proposes reversed-phase thin-layer chromatography (RP-TLC) as a simpler, yet equally accurate method as officially recommended HPLC technique. Several TLC systems were studied including octadecyl-(RP18) and cyano-(CN) modified silica layers in combination with methanol-water and acetonitrile-water mixtures as mobile phases. In total 50 compounds of different molecular shape, size, and various ability to establish specific interactions were selected (phenols, beznodiazepines, triazine herbicides, and polyaromatic hydrocarbons). Calibration set of 29 compounds with known logKOC values determined by sorption experiments was used to build simple univariate calibrations, Principal Component Regression (PCR) and Partial Least Squares (PLS) models between logKOC and TLC retention parameters. Models exhibit good statistical performance, indicating that CN-layers contribute better to logKOC modeling than RP18-silica. The most promising TLC methods, officially recommended HPLC method, and four in silico estimation approaches have been compared by non-parametric Sum of Ranking Differences approach (SRD). The best estimations of logKOC values were achieved by simple univariate calibration of TLC retention data involving CN-silica layers and moderate content of methanol (40-50%v/v). They were ranked far well compared to the officially recommended HPLC method which was ranked in the middle. The worst estimates have been obtained from in silico computations based on octanol-water partition coefficient.   18. High-Throughput Determination of Octanol-Water Partition Coefficients by Ultrasound-Assisted Liquid-Phase Microextraction.

    A method of high-throughput determination, which is based on ultrasound-assisted liquid-phase microextraction, was developed to measure directly the partition coefficients of n-octanol-water. In ultrasound-assisted liquid-phase microextraction, ultrasonic energy can facilitate the mass transfer process of six or more microextractors simultaneously. Therefore, high-throughput determination of n-octanol-water partition coefficients can be performed favorably, and the equilibrium time of each microextractor can be decreased effectively. Several experimental parameters including ultrasonic power and frequency, centrifugation conditions, extractant volume and sample concentration were analyzed and optimized at 25°C. Under the optimum conditions, it only takes 2 min to reach extraction equilibrium, and the solutions of sample can be separated by centrifugation in 4 min. After centrifugation, the concentrations in n-octanol phases are analyzed with gas chromatography. The method was further evaluated with eight reference compounds and the findings demonstrated that this method is suitable to determine the partition coefficients of organic compounds accurately and quickly. Next, the method was exploited to measure the partition coefficients of n-octanol-water containing 20 organic compounds, which cover the [Formula: see text] values from 0.05 to 4.36, with comparatively low relative standard deviation (RSD) directly.     Mitochondria are an important intracellular pharmacological target because damage to this organelle results in a variety of human disorders and because mitochondria are involved in complex processes such as energy generation, apoptosis and lipid metabolism. To expedite the search for natural bioactive compounds targeting mitochondria, we initially developed an efficient mitochondria-based screening method by combining centrifugal ultrafiltration (CU) with liquid chromatography/mass spectrometry (LC/MS), which is called screening method for mitochondria-targeted bioactive constituents (SM-MBC) and is compatible with the search of mitochondria-targeted compounds from complex matrixes such as herbal medicines extracts. Functionally active, structurally intact and pure mitochondria were obtained from rat myocardium using an optimized protocol for mitochondrial isolation comprising organelle release followed by differential and Nycodenz density gradient centrifugation. After evaluating the reliability of the method using thiabendazole (TZ), rotenone (RN), amiodarone (AR) and trimetazidine (TD) as positive controls, this method was successfully applied to screen bioactive constituents from extracts of Polygoni Cuspidati Rhizoma et Radix (PCRR) and Scutellariae Radix (SR). Nineteen active compounds were detected and identified by LC/MS, of which 17 were new mitochondria-targeted compounds. The activity of 9 of the 19 hit compounds was confirmed by in vitro pharmacological trials. These results demonstrate that SM-MBC can be used for the efficient screening of mitochondria-targeted constituents in complex preparations used to treat mitochondrial disorders, such as PCRR and SR. The results may be meaningful for an in-depth understanding of drug mechanism of action and drug discovery from medicinal herbs. PMID:26306914

    Polymer-water partition coefficients (Kpw) of ten DDT-related compounds were determined in pure water at 25 °C using commercial polydimethylsiloxane-coated optical fiber. Analyte concentrations were measured by thermal desorption-gas chromatography/full scan mass spectrometry (TD-GC/MSFS; fibers) and liquid injection-gas chromatography/selected ion monitoring mass spectrometry (LI-GC/MSSIM; water). Equilibrium was approached from two directions (fiber uptake and depletion) as a means of assessing data concordance. Measured compound-specific log Kpw values ranged from 4.8 to 6.1 with an average difference in log Kpw between the two approaches of 0.05 log units (∼ 12% of Kpw). Comparison of the experimentally-determined log Kpw values with previously published data confirmed the consistency of the results and the reliability of the method. A second experiment was conducted with the same ten DDT-related compounds and twelve selected PCB (polychlorinated biphenyl) congeners under conditions characteristic of a coastal marine field site (viz., seawater, 11°C) that is currently under investigation for DDT and PCB contamination. Equilibration at lower temperature and higher ionic strength resulted in an increase in log Kpw for the DDT-related compounds of 0.28-0.49 log units (61-101% of Kpw), depending on the analyte. The increase in Kpw would have the effect of reducing by approximately half the calculated freely dissolved pore-water concentrations (Cfree). This demonstrates the importance of determining partition coefficients under conditions as they exist in the field. PMID:26898149

    We introduce partition equilibrium and study its existence in resource selection games (RSG). In partition equilibrium the agents are partitioned into coalitions, and only deviations by the prescribed coalitions are considered. This is in difference to the classical concept of strong equilibrium according to which any subset of the agents may deviate. In resource selection games, each agent selects a resource from a set of resources, and its payoff is an increasing (or non-decreasing) function of the number of agents selecting its resource. While it has been shown that strong equilibrium exists in resource selection games, these games do not possess super-strong equilibrium, in which a fruitful deviation benefits at least one deviator without hurting any other deviator, even in the case of two identical resources with increasing cost functions. Similarly, strong equilibrium does not exist for that restricted two identical resources setting when the game is played repeatedly. We prove that for any given partition there exists a super-strong equilibrium for resource selection games of identical resources with increasing cost functions; we also show similar existence results for a variety of other classes of resource selection games. For the case of repeated games we identify partitions that guarantee the existence of strong equilibrium. Together, our work introduces a natural concept, which turns out to lead to positive and applicable results in one of the basic domains studied in the literature.

    32P-Postlabeling has emerged as a major tool for detecting DNA adducts resulting from exposure to complex carcinogen mixtures. An integral component of this assay is multi-directional PEI-cellulose TLC in which lipophilic 32P-adducts are resolved in high-salt, high-urea solvents following removal of the bulk of non-adduct radioactivity. This TLC system is very effective for adducts formed following exposure to individual carcinogens; however, adducts resulting from exposure to complex mixtures (e.g. cigarette smoke) generally appear in the form of the so-called diagonal radioactive zones. By using mixtures of polycyclic aromatic hydrocarbon- and aromatic amine-DNA adducts as well as adducts in mouse skin treated with cigarette smoke condensate, we have demonstrated that a combination of 0.3-0.4 M NH4OH and isopropanol-4 M NH4OH (1-1.4:1) solvents can provide more sharply defined adduct spots than the commonly used urea solvents. The non-urea solvents also result in excellent resolution of many adducts which otherwise may remain buried in diagonal radioactive zones when using the urea solvents. In addition, the signal-to-noise ratio is increased 2- to 5-fold over the urea solvents enabling detection of discrete adducts at < or = 3 adducts per 10(10) nucleotides. These partition TLC solvents also involve fewer manipulations (e.g. no water washes to remove salt and urea), and are likely to be more informative with regards to the type of individual adducts detected in the biomonitoring of humans than has hitherto been possible.   8. In vitro prediction of human intestinal absorption and blood-brain barrier partitioning: development of a lipid analog for micellar liquid chromatography.

    Over the past decades, several in vitro methods have been tested for their ability to predict either human intestinal absorption (HIA) or penetration across the blood-brain barrier (BBB) of drugs. Micellar liquid chromatography (MLC) has been a successful approach for retention time measurements of drugs to establish models together with other molecular descriptors. Thus far, MLC approaches have only made use of commercial surfactants such as sodium dodecyl sulfate (SDS) and polyoxyethylene (23) lauryl ether (Brij35), which are not representative for the phospholipids present in human membranes. Miltefosine, a phosphocholine-based lipid, is presented here as an alternative surfactant for MLC measurements. By using the obtained retention factors and several computed descriptors for a set of 48 compounds, two models were constructed: one for the prediction of HIA and another for the prediction of penetration across the BBB expressed as log BB. All data were correlated to experimental HIA and log BB values, and the performance of the models was evaluated. Log BB prediction performed better than HIA prediction, although HIA prediction was also improved a lot (from 0.5530 to 0.7175) compared to in silico predicted HIA values. PMID:26277183

    The solution behaviour of selected proteins has been studied under conditions promoting precipitation, binding to mildly hydrophobic adsorbents or partition. Solvophobic theory may be used to describe these forms of protein partition. The tendency of a protein to partition therein is dependent upon surface properties of the protein solute mediated by the concentration and nature of added salts. As applied to partitioning in poly(ethylene glycol) (PEG)-salt systems this implies that linear (Brönsted) relationships apply only to proteins partitioned close to the critical point. At longer tie-line lengths protein partitioning is increasingly influenced by salting-out forces. This is confirmed by the observed behaviour of the proteins. The point at which this behaviour changes has been unambiguously defined enabling the direct comparison of phase transition of proteins during partition in all systems. The results obtained show that phase transition during adsorption and partition occur at similar concentrations of salt. This is less than that required to promote precipitation. It appears, from these limited studies, that top-phase preferring proteins are partitioned at salt concentrations above those required to cause adsorption. Proteins preferring the lower phase are partitioned at salt concentrations close to or below those required for adsorption. This raises questions regarding the solvated molecular form of the partitioned proteins and the definition of the partition coefficient. PMID:8798879

    An accurate closed-form expression for the rotational partition function of linear polyatomic molecules in 1Sigma electronic states is derived, including the effect of nuclear spin (significant at very low temperatures) and of quartic and sextic centrifugal distortion terms (significant at moderate and high temperatures). The proper first-order quantum correction to the classical rigid-rotator partition function is shown to yield Qr = about 1/beta exp beta/3, where beta is defined as hcB / kT and B is the rotational constant in per cm; for beta of 0.2 or greater additional power-series terms in beta are necessary. Comparison between the results of this treatment and exact summations are made for HCN and C2H2 at temperatures from 2 to 5000 K, including separate evaluation of the conributions of nuclear spin and centrifugal distortion.

    High performance liquid chromatography (HPLC) has many applications in food chemistry. Food components that have been analyzed with HPLC include organic acids, vitamins, amino acids, sugars, nitrosamines, certain pesticides, metabolites, fatty acids, aflatoxins, pigments, and certain food additives. Unlike gas chromatography, it is not necessary for the compound being analyzed to be volatile. It is necessary, however, for the compounds to have some solubility in the mobile phase. It is important that the solubilized samples for injection be free from all particulate matter, so centrifugation and filtration are common procedures. Also, solid-phase extraction is used commonly in sample preparation to remove interfering compounds from the sample matrix prior to HPLC analysis.

    The TLC-based Generally Useful Estimate of Solvent Systems (GUESS) method was employed for countercurrent chromatography solvent system selection, in order to separate the three synthetic isomers: 3-O-methylpyridoxine, 4'-O-methylpyridoxine (ginkgotoxin), and 5'-O-methylpyridoxine. The Rf values of the three isomers indicated that ChMWat+2 (chloroform-methanol-water 10:5:5, v/v/v) was appropriate for the countercurrent separation. The isomer separation was highly selective and demonstrated that the TLC-based GUESS method can accelerate solvent system selection for countercurrent separation. Accordingly, the study re-emphasizes the practicality of TLC as a tool to facilitate the rapid development of new countercurrent and centrifugal partition chromatography methods for this solvent system.     A seal is described for a high speed centrifuge wherein the centrifugal force of rotation acts on the gasket to form a tight seal. The cylindrical rotating bowl of the centrifuge contains a closure member resting on a shoulder in the bowl wall having a lower surface containing bands of gasket material, parallel and adjacent to the cylinder wall. As the centrifuge speed increases, centrifugal force acts on the bands of gasket material forcing them in to a sealing contact against the cylinder wall. This arrangememt forms a simple and effective seal for high speed centrifuges, replacing more costly methods such as welding a closure in place.

    The Monsanto Chocolate Bayou plant has had an aggressive and successful energy conservation program. The combined efforts have resulted in a 80% reduction in unit energy consumption compared to 1972. The approach of using system audits to optimize fluid systems was developed. Since most of the fluid movers are centrifugal, the name Centrifugal Savings Task Force was adopted. There are three tools that are particularly valuable in optimizing fluid systems. First, a working level understanding of the Affinity Laws seems a must. In addition, the performance curves for the fluid movers is needed. The last need is accurate system field data. Systems effectively managed at the Chocolate Bayou plant were process air improvement, feed-water pressure reduction, combustion air blower turbine speed control, and cooling tower pressure reduction. Optimization of centrifugal systems is an often-overlooked opportunity for energy savings. The basic guidelines are to move only the fluid needed, and move it at as low a pressure as possible.

    Modifications are underway at the National Aeronautics and Space Administration (NASA) Ames Research Center in California to transform a centrifuge used in the Apollo space program to the largest geotechnical centrifuge in the free world. The centrifuge, to be finished in August and opened next January, following check out and tuning, will enable geoscientists to model stratigraphic features down to 275 m below the earth's surface. Scientists will be able to model processes that are coupled with body force loading, including earthquake response of earth structures and soil structure interaction; rubbled-bed behavior during in situ coal gasification or in oil shale in situ retorts; behavior of frozen soil; frost heave; behavior of offshore structures; wave-seabed interactions; explosive cratering; and blast-induced liquefaction.The centrifuge will have a load capacity of 900-g-tons (short); that is, it will be able to carry a net soil load of 3 short tons to a centripetal acceleration of 300 times the acceleration caused by gravity. Modified for a total cost of $2.4 million, the centrifuge will have an arm with a 7.6-m radius and a swinging platform or bucket at its end that will be able to carry a payload container measuring 2.1×2.1 m. An additional future input of $500,000 would enable the purchase of a larger bucket that could accommodate a load of up to 20 tons, according to Charles Babendreier, program director for geotechnical engineering at the National Science Foundation. Additional cooling for the motor would also be required. The centrifuge has the capability of accelerating the 20-ton load to 100 g.

    A major challenge in countercurrent chromatography (CCC), the technique that works with a support-free biphasic liquid system, is to retain the liquid stationary phase inside the CCC column (Sf parameter). Two solutions are commercially available: the hydrostatic CCC columns, also called centrifugal partition chromatographs (CPC), with disks of interconnected channels and rotary seals, and the hydrodynamic CCC columns with bobbins of coiled open tube and no rotary seals. It was demonstrated that the amount of liquid stationary phase retained by a coiled tube was higher with larger bore tubing than with small bore tubes. At constant column volume, small bore tubing will be longer producing more efficiency than larger bore tube that will better retain the liquid stationary phase. Since the resolution equation in CCC is depending on both column efficiency and stationary phase retention ratio, the influence of the tubing bore should be studied. This theoretical work showed that there is an optimum tubing bore size depending on solute partition coefficient and mobile phase flow rate. The interesting result of the theoretical study is that larger tubing bores allow for dramatically reduced experiment durations for all solutes: in reversed phase CCC (polar mobile phase), hydrophobic solutes are usually highly retained. These apolar solutes can be separated by the same coil at high flow rates and reduced Sf with similar retention times as polar solutes separated at smaller flow rates and much higher Sf.     Centrifugally decoupling mechanical bearing systems provide thin tensioned metallic ribbons contained in a support structure. This assembly rotates around a stationary shaft being centered at low speeds by the action of the metal ribbons. Tension springs are connected on one end to the ribbons and on the other end to the support structure. The ribbons pass through slots in the inner ring of the support structure. The spring preloading thus insures contact (or near-contact) between the ribbons and the shaft at rotation speeds below the transition speed. Above this speed, however, the centrifugal force on the ribbons produces a tensile force on them that exceeds the spring tensile force so that the ribbons curve outward, effectively decoupling them from mechanical contact with the shaft. They still remain, however, in position to act as a touchdown bearing in case of abnormally high transverse accelerations.

    In ordinary centrifugation, a suspended particle that is heavier than the displaced fluid migrates away from the rotation axis when the fluid-filled container rotates steadily about that axis. In contrast a particle that is lighter than the displaced fluid (e.g., a bubble) migrates toward the rotation axis in a centrifuge. In this paper, we show theoretically that if a fluid-filled container rotates in an oscillatory manner as a rigid body about an axis, at high enough oscillation frequencies, the sense of migration of suspended particles is reversed. That is, in that case particles denser than the fluid migrate inward, while those that are lighter than the fluid move outward. We term this unusual phenomenon "Oscillatory Counter-Centrifugation" or OCC, for short. Through application of the method of averaging to the equations of motion, we derive a simple criterion to predict the occurrence of OCC. The analysis also reveals that the time-average of the Coriolis force in the radial direction is the term that is responsible for this effect. In addition, we analyze the effects of the Basset history force and the Rubinow-Keller lift force on particle trajectories and find that OCC persists even when these forces are active. The phenomenon awaits experimental verification.

    Gas centrifuges have been an ideal enrichment method for a wide variety of countries. Many countries have built gas centrifuges to make enriched uranium for peaceful nuclear purposes. Other countries have secretly sought centrifuges to make highly enriched uranium for nuclear weapons. In more recent times, several countries have secretly sought or built gas centrifuges in regions of tension. The main countries that have been of interest in the last two decades have been Pakistan, Iraq, Iran, and North Korea. Currently, most attention is focused on Iran, Pakistan, and North Korea. These states did not have the indigenous abilities to make gas centrifuges, focusing instead on illicit and questionable foreign procurement. The presentation covered the following main sections: Spread of centrifuges through illicit procurement; Role of export controls in stopping proliferation; Increasing the transparency of gas centrifuge programs in non-nuclear weapon states; and, Verified dismantlement of gas centrifuge programs. Gas centrifuges are important providers of low enriched uranium for civil nuclear power reactors. They also pose special nuclear proliferation risks. We all have special responsibilities to prevent the spread of gas centrifuges into regions of tension and to mitigate the consequences of their spread into the Middle East, South Asia, and North Asia.

    The overall project consists of several integrated research phases related to the applicability, continued development, demonstration, and commercialization of the SpinTek centrifugal membrane filtration process. Work performed during this reporting period consisted of Phase 2 evaluation of the SpinTek centrifugal membrane filtration technology and Phase 3, Technology Partnering. During Phase 1 testing conducted at the EERC using the SpinTek ST-IIL unit operating on a surrogate tank waste, a solids cake developed on the membrane surface. The solids cake was observed where linear membrane velocities were less than 17.5 ft/s and reduced the unobstructed membrane surface area up to 25%, reducing overall filtration performance. The primary goal of the Phase 2 research effort was to enhance filtration performance through the development and testing of alternative turbulence promoter designs. The turbulence promoters were designed to generate a shear force across the entire membrane surface sufficient to maintain a self-cleaning membrane capability and improve filtration efficiency and long-term performance. Specific Phase 2 research activities included the following: System modifications to accommodate an 11-in.-diameter, two-disk rotating membrane assembly; Development and fabrication of alternative turbulence promoter designs; Testing and evaluation of the existing and alternative turbulence promoters under selected operating conditions using a statistically designed test matrix; and Data reduction and analysis; The objective of Phase 3 research was to demonstrate the effectiveness of SpinTek's centrifugal membrane filtration as a pretreatment to remove suspended solids from a liquid waste upstream of 3M's WWL cartridge technology for the selective removal of technetium (Tc).

    The centrifugal adsorption cartridge system (CACS) is an apparatus that recovers one or more bioproduct(s) from a dilute aqueous solution or suspension flowing from a bioreactor. The CACS can be used both on Earth in unit gravity and in space in low gravity. The CACS can be connected downstream from the bioreactor; alternatively, it can be connected into a flow loop that includes the bioreactor so that the liquid can be recycled. A centrifugal adsorption cartridge in the CACS (see figure) includes two concentric cylinders with a spiral ramp between them. The volume between the inner and outer cylinders, and between the turns of the spiral ramp is packed with an adsorbent material. The inner cylinder is a sieve tube covered with a gas-permeable, hydrophobic membrane. During operation, the liquid effluent from the bioreactor is introduced at one end of the spiral ramp, which then constrains the liquid to flow along the spiral path through the adsorbent material. The spiral ramp also makes the flow more nearly uniform than it would otherwise be, and it minimizes any channeling other than that of the spiral flow itself. The adsorbent material is formulated to selectively capture the bioproduct(s) of interest. The bioproduct(s) can then be stored in bound form in the cartridge or else eluted from the cartridge. The centrifugal effect of the spiral flow is utilized to remove gas bubbles from the liquid. The centrifugal effect forces the bubbles radially inward, toward and through the membrane of the inner cylinder. The gas-permeable, hydrophobic membrane allows the bubbles to enter the inner cylinder while keeping the liquid out. The bubbles that thus enter the cylinder are vented to the atmosphere. The spacing between the ramps determines rate of flow along the spiral, and thereby affects the air-bubble-removal efficiency. The spacing between the ramps also determines the length of the fluid path through the cartridge adsorbent, and thus affects the bioproduct

This demonstration is part of the Chicago Pile-5 (CP-5) Large-Scale Demonstration Project (LSDP) sponsored by the US Department of Energy (DOE), office of Science and Technology (OST), Deactivation and Decommissioning Focus Area (DDFA). The objective of the LSDP is to select and demonstrate potentially beneficial technologies at the Argonne National Laboratory-East (ANL) CP-5 Research Reactor. The purpose of the LSDP is to demonstrate that using innovative and improved decontamination and decommissioning (D and D) technologies from various sources can result in significant benefits, such as decreased cost and increased health and safety, as compared with baseline D and D technologies. Potential markets exist for the innovative centrifugal shot blast system at the following sites: Fernald Environmental Management Project, Los Alamos, Nevada, Oak Ridge Y-12 and K-25, Paducah, Portsmouth Gaseous Diffusion site, and the Savannah River Site.   20. Rat growth during chronic centrifugation

    Female weanling rats were chronically centrifuged at 4.15 G with controls at terrestrial gravity. Samples were sacrificed for body composition studies at 0, 28, 63, 105 and 308 days of centrifugation. The centrifuged group approached a significantly lower mature body mass than the controls (251 and 318g) but the rate of approach was the same in both groups. Retirement to 1G on the 60th day resulted in complete recovery. Among individual components muscle, bone, skin, CNS, heart, kidneys, body water and body fat were changed in the centrifuged group. However, an analysis of the growth of individual components relative to growth of the total fat-free compartment revealed that only skin (which increased in mass) was responding to centrifugation per se.

  1. Unshrouded Centrifugal Turbopump Impeller

    The ratio of rocket engine thrust to weight is a limiting constraint in placing more payload into orbit at a low cost. A key component of an engine's overall weight is the turbopump weight, Reducing the turbopump weight can result in significant engine weight reduction and hence, increased delivered payload. There are two main types of pumps: centrifugal and axial pumps. These types of pumps can be further sub-divided into those with shrouds and those without shrouds (unshrouded pumps). Centrifugal pumps can achieve the same pump discharge pressure as an axial pump and it requires fewer pump stages and lower pump weight than an axial pump. Also, with unshrouded centrifugal pumps (impeller), the number of stages and weight can be further reduced. However. there are several issues with regard to using an unshrouded impeller: 1) there is a pump performance penalty due to the front open face recirculation flow; 2) there is a potential pump axial thrust problem from the unbalanced front open face and the back shroud face; and, 3) since test data is very linu'ted for this configuration, there is uncertainty in the magnitude and phase of the rotordynamics forces due to the front impeller passage. The purpose of the paper is to discuss the design of an unshrouded impeller and to examine the design's hydrodynamic performance, axial thrust, and rotordynamics performance. The design methodology will also be discussed. This work will help provide some guidelines for unshrouded impeller design. In particular, the paper will discuss the design of three unshrouded impellers - one with 5 full and 5 partial blades (5+5). one with 6+6 blades and one with 8+8 blades.     Aralia continentalis (Araliaceae) is widely used as a medicinal plant in East Asia. Previous studies have indicated that diterpenoid isomers (kaurenoic acid, continentalic acid, and ent-continentalic acid) are the major bioactive compounds of this plant. A new strategy was developed to alleviate difficulties in the separation of these isomers from this plant. A three-phase solvent system was applied to separate the isomers, and furthermore, the distribution ratio (Kc) was introduced as a substitute for the partition coefficient (KD). For compounds exhibiting a single equilibrium, their distributions in two immiscible phases were only affected by the partition coefficient of each solute. However, compounds that have a dissociating functional group (e.g., -COOH) are involved in two types of equilibrium in the two-phase system. In this case, the partitioning behaviors of the solutes are greatly affected by the pH of the solution. A mathematical prediction was applied for adjusting the solutions to the proper pH values. To prevent non-used phase (medium phase) waste, both the stationary phase (upper phase) and mobile phase (lower phase) were prepared on-demand without pre-saturation with the application of (1)H NMR. Each fraction obtained was collected and dried, yielding the following diterpenoid isomers from the 50mg injected sample: kaurenoic acid (19.7mg, yield: 39%) and ent-continentalic acid (21.3mg, yield: 42%).     The Centrifuge Rotor (CR) is a large life science experiment facility which will be installed in the International Space Station (ISS). It will provide artificial gravity of 2g or less by rotating up to 4 science habitats, and it will be the first such machinery to be used in space. To prevent vibration disturbance exchanges between the CR and the ISS, a soft 5 dof vibration isolation mechanism is used which cannot support the CR weight on the ground. Therefore, the CR on-orbit performance must be predicted by integrated analysis which must model all of the equipment including sensors, actuators, flexible structure, gyroscopic effects, and controllers. Here, we introduce the CR mechatronics, a verification procedure, and examples of the application of the integrated analysis which is based on the general-purpose mechanism analysis software ADAMS.

    This work intends to focus on Galileo's study of what is now called "centrifugal force," within the framework of the Second Day of his Dialogo written in 1632, rather than on the previously published commentaries on the topic. Galileo proposes three geometrical demonstrations in order to prove that gravity will always overcome centrifugalforce, and that the potential rotation of the Earth, whatever its speed, cannot in any case project objects beyond it. Each of these demonstrations must consequently contain an error and it has seemed to us that the first one had not been understood up until now. Our analysis offers an opportunity to return to Galileo's geometrical representation of dynamical questions; actually, we get an insight into the sophistication of Galileo's practices more than into his mistakes. Our second point, concerning the historiography of the problem, shows an evolution from anachronic critics to more contextual considerations, in the course of the second half of the twentieth century.   7. Determination of Trichlorfon Pesticide Residues in Milk via Gas Chromatography with μ-Electron Capture Detection and GC-MS.

    The pesticide trichlorfon is readily degraded under experimental conditions to dichlorvos. A method has therefore been developed by which residues of trichlorfon in milk are determined as dichlorvos, using gas chromatography with μ-electron capture detection. The identification of dichlorvos was confirmed by mass spectrometry. Milk was extracted with acetonitrile followed by centrifugation, freezing lipid filtration, and partitioning into dichloromethane. The residue after partitioning of dichloromethane was dissolved in ethyl acetate for gas chromatography. Recovery concentration was determined at 0.5, 1.0, and 2.0 of times the maximum permitted residue limits (MRLs) for trichlorfon in milk. The average recoveries (n = 6) ranged from 92.4 to 103.6%. The repeatability of the measurements was expressed as relative standard deviations (RSDs) ranging from 3.6%, to 6.7%. Limit of detection (LOD) and limit of quantification (LOQ) were 3.7 and 11.1 μg/l, respectively. The accuracy and precision (expressed as RSD) were estimated at concentrations from 25 to 250 μg/l. The intra- and inter-day accuracy (n = 6) ranged from 89.2%to 91% and 91.3% to 96.3%, respectively. The intra- and inter-day precisions were lower than 8%. The developed method was applied to determine trichlorfon in real samples collected from the seven major cities in the Republic of Korea. No residual trichlorfon was detected in any samples.     Dynamics of waves generated by scopes in gas centrifuges (GC) for isotope separation is considered. The centrifugal acceleration in the GC reaches values of the order of 106g. The centrifugal and Coriolis forces modify essentially the conventional sound waves. Three families of the waves with different polarization and dispersion exist in these conditions. Dynamics of the flow in the model GC Iguasu is investigated numerically. Comparison of the results of the numerical modeling of the wave dynamics with the analytical predictions is performed. New phenomena of the resonances in the GC is found. The resonances occur for the waves polarized along the rotational axis having the smallest dumping due to the viscosity.

The effect that centrifugal distortion of the rotating molecules has on these rigid-rotator formulations is considered. The nuclear-spin correction is generalized to symmetric-top molecules of other symmetries. The treatment is extended to nonplanar molecules that exhibit inversion doubling, with particular attention given to NH3.

  16. Screen bowl centrifuge: a high-efficiency particle size separator

    Over the years, screen bowl centrifuges have been widely used for dewatering fine coal in coal preparation plants in the United States and elsewhere. It is generally recognized in the engineering and scientific communities that screen bowl centrifuges provide some degree of particle size separation while dewatering fine coal in a common application. However, the extent of differential partitioning of coarse and fine particles achievable by a screen bowl centrifuge has not been systematically studied in the past. The present investigation was aimed at conducting a parametric study using a statistically designed experimental program to better understand and optimize the size classification performance of a screen bowl centrifuge. A continuously operating screen bowl centrifuge having a bowl diameter of 0.5 m was used for this study at the Illinois Coal Development Park. Three key operating parameters, i.e., feed flow rate, feed solid content and pool depth, were varied to conduct a total of 17 experiments using a three-level factorial test matrix. Some of the best size separation performances achieved in this study may be described as having an imperfection value of 0.13 at an effective separation size (d(50c)) of 38 mu m and an imperfection value of 0.27 at an effective separation size (d(50c)) of 2.8 mu m. Due to an effective separation of ultrafine high ash materials, the ash content of the screen bowl feed was reduced from 22.3% to a minimum of 8.84% with a combustible recovery of 84.1% and an ash rejection of 71.6%. A higher combustible recovery of 92.1% was achieved at a product ash content of 12.5% with a d(50c) of 2.8 mu m and imperfection of 0.27.

    SpinTek Membrane Systems, Inc., the developer of a centrifugal membrane filtration technology, has engineered and developed a system for use within the U.S. Department of Energy (DOE) Environmental Management (EM) Program. The technology uses supported microporous membranes rotating at high rpm, under pressure, to separate suspended and colloidal solids from liquid streams, yielding a solids-free permeate stream and a highly concentrated solids stream. This is a crosscutting technology that falls under the Efficient Separations and Processing Crosscutting Program, with potential application to tank wastes, contaminated groundwater, landfill leachate, and secondary liquid waste streams from other remediation processes, including decontamination and decommissioning systems. SpinTek II High Shear Rotary Membrane Filtration System is a unique compact crossflow membrane system that has large, demonstrable advantages in performance and cost compared to currently available systems: (1) High fluid shear prevents membrane fouling even with very high solids content; hazardous and radioactive components can be concentrated to the consistency of a pasty slurry without fouling. (2) Induced turbulence and shear across the membrane increases membrane flux by a factor of ten over existing systems and allows operation on fluids not otherwise treatable. (3) Innovative ceramic membrane and mechanical sealing technology eliminates compatibility problems with aggressive DOE waste streams. (4) System design allows rapid, simple disassembly for inspection or complete decontamination. (5) Produces colloidal- and suspended-solids-free filtrate without the addition of chemicals. The first phase of this project (PRDA maturity stage 5) completed the physical scale-up of the SpinTek unit and verified successful scale-up with surrogate materials. The applied rotating wall electric field provides a positive torque that counteracts background drags, resulting in radial compression or steady-state confinement in near-thermal equilibrium states. The last part of the chapter discusses centrifugal separation in a rotating multi-species non-neutral plasma.     An apparatus is presented for remotely determining the alignment of a centrifuge. The centrifage shaft is provided with a shoulder, upon which two followers ride, one for detecting radial movements, and one upon the shoulder face for determining the axial motion. The followers are attached to separate liquid filled bellows, and a tube connects each bellows to its respective indicating gage at a remote location. Vibrations produced by misalignment of the centrifuge shaft are transmitted to the bellows, and tbence through the tubing to the indicator gage. This apparatus is particularly useful for operation in a hot cell where the materials handled are dangerous to the operating personnel.

    The partition coefficients of three homologous anticonvulsant phenylalkylamides [racemic alpha-hydroxy-alpha-ethyl-alpha-phenylacetamide (HEPA); beta-hydroxy-beta-ethyl-beta-phenylpropionamide (HEPP); and gamma-hydroxy-gamma-ethyl-gamma-phenylbutyramide (HEPB)] were determined by reversed-phase high-performance liquid chromatography (RP-HPLC). The log kw values (methanol:water, extrapolated to 100% water) were 1.260 for HEPA, 1.670 for HEPP, and 1.852 for HEPB. From these results, the partition coefficients (log P) were calculated by regression as 1.20, 1.83, and 2.11, respectively. The log P values were essentially equal to those calculated by the Leo-Hansch fragmental method. Since the potency of the three anticonvulsants is approximately the same in a variety of tests, no dependence on lipophilicity could be established. PMID:2292764

    Speciation of trialkylated arsenic compunds in natural gas, pressurized and stable condensate samples from the same gas well was performed using (Cryotrapping) Gas Chromatography-Inductively Coupled Plasma Mass Spectrometry. The major species in all phases investigated was found to be trimethylarsine with a highest concentration of 17.8 ng/L (As) in the gas phase and 33.2 μg/L (As) in the stable condensate phase. The highest amount of trimethylarsine (121 μg/L (As)) was found in the pressurized condensate, along with trace amounts of non-identified higher alkylated arsines. Volatile arsenic species in natural gas and its related products cause concern with regards to environment, safety, occupational health and gas processing. Therefore, interest lies in a fast and simple field method for the determination of volatile arsenicals. Here, we use simple liquid and solid sorption techniques, namely absorption in silver nitrate solution and adsorption on silver nitrate impregnated silica gel tubes followed by total arsenic determination as a promising tool for field monitoring of volatile arsenicals in natural gas and gas condensates. Preliminary results obtained for the sorption-based methods show that around 70% of the arsenic is determined with these methods in comparison to volatile arsenic determination using GC-ICP-MS. Furthermore, an inter-laboratory- and inter-method comparison was performed using silver nitrate impregnated silica tubes on 14 different gas samples with concentrations varying from below 1 to 1000 μg As/m 3 natural gas. The results obtained from the two laboratories differ in a range of 10 to 60%, but agree within the order of magnitude, which is satisfactory for our purposes.

    Rat liver plasma membranes were separated from other cellular membranes by affinity partitioning in an aqueous polymer two-phase system by using the lectin wheat-germ agglutinin covalently bound to dextran as the affinity ligand. In borate buffer the bulk of membranes partitioned in the poly(ethylene glycol)-rich top phase, whereas plasma membranes were pulled selectively into the dextran-rich bottom phase in the presence of ligand. The purity and yield of plasma membranes prepared by lectin affinity partitioning and by conventional sucrose-density-gradient centrifugation was similar, as judged from marker-enzyme activities. The affinity procedure, not dependent on lengthy centrifugations, is fast and gentle and will be advantageous when studying labile components. PMID:1703408

  19. Partitioning Breaks Communities

    Considering a clique as a conservative definition of community structure, we examine how graph partitioning algorithms interact with cliques. Many popular community-finding algorithms partition the entire graph into non-overlapping communities. We show that on a wide range of empirical networks, from different domains, significant numbers of cliques are split across the separate partitions produced by these algorithms. We then examine the largest connected component of the subgraph formed by retaining only edges in cliques, and apply partitioning strategies that explicitly minimise the number of cliques split. We further examine several modern overlapping community finding algorithms, in terms of the interaction between cliques and the communities they find, and in terms of the global overlap of the sets of communities they find. We conclude that, due to the connectedness of many networks, any community finding algorithm that produces partitions must fail to find at least some significant structures. Moreover, contrary to traditional intuition, in some empirical networks, strong ties and cliques frequently do cross community boundaries; much community structure is fundamentally overlapping and unpartitionable in nature.

    Partition density functional theory (PDFT) is a method for dividing a molecular electronic structure calculation into fragment calculations. The molecular density and energy corresponding to Kohn Sham density-functional theory (KS-DFT) may be exactly recovered from these fragments. Each fragment acts as an isolated system except for the influence of a global one-body 'partition' potential which deforms the fragment densities. In this work, the developments of PDFT are put into the context of other fragment-based density functional methods. We developed three numerical implementations of PDFT: One within the NWChem computational chemistry package using basis sets, and the other two developed from scratch using real-space grids. It is shown that all three of these programs can exactly reproduce a KS-DFT calculation via fragment calculations. The first of our in-house codes handles non-interacting electrons in arbitrary one-dimensional potentials with any number of fragments. This code is used to explore how the exact partition potential changes for different partitionings of the same system and also to study features which determine which systems yield non-integer PDFT occupations and which systems are locked into integer PDFT occupations. The second in-house code, CADMium, performs real-space calculations of diatomic molecules. Features of the exact partition potential are studied for a variety of cases and an analytical formula determining singularities in the partition potential is derived. We introduce an approximation for the non-additive kinetic energy and show how this quantity can be computed exactly. Finally a PDFT functional is developed to address the issues of static correlation and delocalization errors in approximations within DFT. The functional is applied to the dissociation of H2 + and H2.

    We utilize the formal equivalence between the number-partitioning problem and a harmonically trapped ideal Bose gas within the microcanonical ensemble for characterizing the probability distribution which governs the number of addends occurring in an unrestricted partition of a natural number n. By deriving accurate asymptotic formulae for its coefficients of skewness and excess, it is shown that this distribution remains non-Gaussian even when n is made arbitrarily large. Both skewness and excess vary substantially before settling to their constant-limiting values for n > 1010.

Plant managers are thus entitled to a thorough analysis of whether reduced expenditures truly lower costs or actually hinder maintenance and increase costs. Absence of such an analysis hides the fact that proper and timely maintenance has a double effect: it not only reduces power consumption but also extends equipment life, and thus reduces the frequency of labor and material expenditures for scheduled or crisis maintenance. Centrifugal pump maintenance can demonstrate well the validity of this observation.     Rotating stall and the inlet vortex in centrifugal fans with inlet vane control has been studied. The advances in stall research in aero-engine compressors are discussed. The present study shows that stall in centrifugal fans can be quite different from that in axial compressors, in that stall can occur in a progressive and intermittent fashion. The study also shows that a discontinuity in the fan characteristic is not necessarily accompanied by rotating stall, unlike the axial machines. Experimental results indicate that the positive prewhirl created by inlet vanes tends to delay the occurrence of stall. Also, dorsal fin devices that are used to control the inlet vortex do not seem to affect the stall point unfavorably. The inlet vortex frequency was found to invariably exhibit a linear relation with the flow rate even when dorsal fins were used. This offers a practical method to distinguish between the inlet vortex and rotating stall.

  6. Twinning of amphibian embryos by centrifugation

    In the frog Xenopus laevis, the dorsal structures of the embryonic body axis normally derive from the side of the egg opposite the side of sperm entry. However, if the uncleaved egg is inclined at lg or centrifuged in an inclined position, this topographic relationship is overridden: the egg makes its dorsal axial structures according to its orientation in the gravitational/centrifugal field, irrespective of the position of sperm entry. Certain conditions of centrifugation cause eggs to develop into conjoined twins with two sets of axial structures. A detailed analysis of twinning provided some insight into experimental axis orientation. First, as with single-axis embryos, both axes in twins are oriented according to the direction of centrifugation. One axis forms at the centripetal side of the egg and the other forms at the centrifugal side, even when the side of sperm entry is normal to the centrifugal force vector. Second, if eggs are centrifuged to give twins, but are inclined at lg to prevent post-centrifugation endoplasmic redistributions, only single-axis embryos develop. Thus, a second redistribution is required for high-frequency secondary axis formation. This can be accomplished by lg (as in the single centrifugations) or by a second centrifugation directed along the egg's animal-vegetal axis.

  7. New Aperture Partitioning Element

    Postprocessing in an optical system can be aided by adding an optical element to partition the pupil into a number of segments. When imaging through the atmosphere, the recorded data are blurred by temperature-induced variations in the index of refraction along the line of sight. Using speckle imaging techniques developed in the astronomy community, this blurring can be corrected to some degree. The effectiveness of these techniques is diminished by redundant baselines in the pupil. Partitioning the pupil reduces the degree of baseline redundancy, and therefore improves the quality of images that can be obtained from the system. It is possible to implement the described approach on an optical system with a segmented primary mirror, but not very practical. This is because most optical systems do not have segmented primary mirrors, and those that do have relatively low bandwidth positioning of segments due to their large mass and inertia. It is much more practical to position an active aperture partitioning element at an aft optics pupil of the optical system. This paper describes the design, implementation and testing of a new aperture partitioning element that is completely reflective and reconfigurable. The device uses four independent, annular segments that can be positioned with a high degree of accuracy without impacting optical wavefront of each segment. This mirror has been produced and is currently deployed and working on the 3.6 m telescope.

    The feasibility of using computational fluid dynamics to guide the design of better centrifuges for processing shed blood is explored here. The velocity field and the rate of protein removal from the shed blood have been studied. The results indicate that computational fluid dynamics could help screen preliminary centrifuge bowl designs thus reducing the number of initial experimental tests required when developing new centrifuge bowls. Though the focus of this work is on washing shed blood the methods developed here are applicable to the design of centrifuge bowls for other blood processing applications. PMID:15133962

  10. Design Attributes and Scale Up Testing of Annular Centrifugal Contactors

    Annular centrifugal contactors are being used for rapid yet efficient liquid- liquid processing in numerous industrial and government applications. Commercialization of this technology began eleven years ago and now units with throughputs ranging from 0.25 to 700 liters per minute are readily available. Separation, washing, and extraction processes all benefit from the use of this relatively new commercial tool. Processing advantages of this technology include: low in-process volume per stage, rapid mixing and separation in a single unit, connection-in-series for multi-stage use, and a wide operating range of input flow rates and phase ratios without adjustment. Recent design enhancements have been added to simplify maintenance, improve inspection ability, and provide increased reliability. Cartridge-style bearing and mechanical rotary seal assemblies that can include liquid-leak sensors are employed to enhance remote operations, minimize maintenance downtime, prevent equipment damage, and extend service life. Clean-in-place capability eliminates the need for disassembly, facilitates the use of contactors for feed clarification, and can be automated for continuous operation. In nuclear fuel cycle studies, aqueous based separations are being developed that efficiently partition uranium, actinides, and fission products via liquid-liquid solvent extraction. Thus, annular centrifugal contactors are destined to play a significant role in the design of such new processes. Laboratory scale studies using mini-contactors have demonstrated feasibility for many such separation processes but validation at an engineering scale is needed to support actual process design.

    A centrifugal contactor that performs oxidation and reduction exchange reactions between molten metals and salts at 500 degrees Centigrade has been tested successfully at Argonne National Laboratory (ANL). The design is based on contactors for aqueous- organic systems operation near room temperature. In tests to demonstrate the performance of the pyrocontactor, cadmium and LICl- KCl eutectic salt were the immiscible solvent phases, and rare earths were the distributing solutes. The tests showed that the pyrocontactor mixed and separated the phases well, with stage efficiencies approaching 99% at rotor speeds near 2700 rpm. The contactor ran smoothly and reliably over the entire range of speeds that was tested.

    Dual high-speed countercurrent chromatography (dual CCC) literally permits countercurrent flow of two immiscible solvent phases continuously through the coiled column for separation of solutes according to their partition coefficients. Application of this technique has been successfully demonstrated by separation of analytes by gas-liquid and liquid-liquid two-phase systems. However, the method cannot be directly applied to the system with a set of coiled columns connected in series, since the countercurrent process is interrupted at the junction between the columns. However, this problem can be solved by intermittent dual CCC by eluting each phase alternately through the opposite ends of the separation column. This mode of application has an advantage over the conventional dual CCC in that the method can be applied to all types of CCC systems including hydrostatic equilibrium systems such as toroidal coil CCC and centrifugal partition chromatography. Recently, the application of this method to high-speed CCC (hydrodynamic system) has been demonstrated for separation of natural products by Hewitson et al. using a set of conventional multilayer coil separation columns connected in series. Here, we have developed a mathematical model for this intermittent dual CCC system to predict retention time of the analytes, and using a simplified model system the validity of the model is justified by a series of basic studies on both hydrodynamic and hydrostatic CCC systems with a computer-programmed single sliding valve. The present method has been successfully applied to spiral tube assembly high-speed CCC (hydrodynamic system) and toroidal coil CCC (hydrostatic system) for separation of DNP-amino acid samples with two biphasic solvent systems composed of hexane-ethyl acetate-methanol-0.1M hydrochloric acid (1:1:1:1 and 4:5:4:5, v/v).     Damping bearings for use on the shafts of an ultracentrifuge were designed which are capable of passing through critical angular speeds. The shaft extending from one end of the rotor is journaled in fixed-plain bearings mounted on annular resilient shock-absorbing elements to dampen small vibrations. The shaft at the other end of the rotor is journaled in two damper-bearing assemblies which are so spaced on the shaft that a vibration node can at no time exist at both bearing assemblies. These bearings are similar to the other bearings except that the bearing housings are slidably mounted on the supporting structure for movement transverse to the rotational axis of the rotor.

  15. Sediment/Aqueous Partitioning of Fecal Indicator Bacteria in Spring Meltwater

    This study was designed to elucidate the partitioning behavior of fecal indicator bacteria Enterococcus and E. coli in spring snowmelt. Two snowmelt lysimeters were constructed and inoculated with agriculturally-derived manure prior to the first snowfall. After approximately six months, the lysimeter runoff was collected daily and transported to the lab for analysis. Particle-attached and disassociated bacteria were separated via centrifugation, and quantified via the most probable number technique. Results revealed significant differences between the behaviors of Enterococcus compared to E. coli. First, Enterococcus exhibited a significantly higher survival rate over the winter months compared to E. coli. Second, Enterococcus partitioned almost exclusively with the sediment in the meltwater, whereas E. coli partitioned variably between the aqueous and particulate phases. These results demonstrated that water quality managers must understand the specific partitioning behavior of the indicator organisms tested in order to effectively interpret water quality monitoring results.

    A common characteristic of mass flows like debris flows, rock avalanches and mudflows is that gravity is their main driving force. Gravity defines the intensity and duration of the main interactions between particles and their surrounding media (particle-particle, particle-fluid, fluid-fluid). At the same time, gravity delimits the occurrence of phase separation, inverse segregation, and mass consolidation, among other phenomena. Therefore, in the understanding of the flow physics it is important to account for the scaling of gravity in scaled models. In this research, a centrifuge model is developed to model free surface granular flows down an incline at controlled gravity conditions. Gravity is controlled by the action of an induced inertial acceleration field resulting from the rotation of the model in a geotechnical centrifuge. The characteristics of the induced inertial acceleration field during flow are discussed and validated via experimental data. Flow heights, velocity fields, basal pressure and impact forces are measured for a range of channel inclinations and gravity conditions. Preliminary results enlighten the flow characteristics at variable gravity conditions and open a discussion on the simulation of large scale processes at a laboratory scale. Further analysis on the flow physics brings valuable information for the validation of granular flows rheology.

    Pellet injectors are needed to fuel long pulse tokamak plasmas and other magnetic confinement devices. For this purpose, an apparatus has been developed that forms 1.3-mm-diam pellets of frozen deuterium at a rate of 40 pellets per second and accelerates them to a speed of 1 km/s.     Pellet injectors are needed to fuel long pulse tokamak plasmas and other magnetic confinement devices.     A recent study has argued that the California Undercurrent, and poleward eastern boundary currents in general, generate mixing events through centrifugal instability (CI). Conditions favorable for CI are created by the strong horizontal shears developed in turbulent bottom layers of currents flowing in the direction of topographic waves. At points of abrupt topographic change, like promontories and capes, the coastal current separates from the boundary and injects gravitationally stable but dynamically unstable flow into the interior. The resulting finite amplitude development of the instability involves overturnings and diabatic mixing. The purpose of this study is to examine the energetics of CI in order to characterize it as has been done for other instabilities and develop a framework in which to estimate its regional and global impacts. We argue that CI is roughly twice as efficient at mixing as is Kelvin-Helmholtz instability, and that roughly 10% of the initial energy in a CUC-like current is lost to either local mixing or the generation of unbalanced flows. The latter probably leads to non-local mixing. Thus centrifugal instability is an effective process by which energy is lost from the balanced flow and spent in mixing neighboring water masses. We argue the importance of the mixing is regional in nature, but of less importance to the global budgets given its regional specificity.

    Simulator sickness is now a well-recognized entity. It is recognized as a form of motion sickness, having a higher incidence in the more sophisticated simulators. Human centrifuges (dynamic simulators) are the newest innovation in aircrew training devices. Simulator sickness has never been reported in human centrifuges. We are reporting on a case of delayed simulator sickness in a pilot-subject after a centrifuge experience. A review of the "psycho-physiological" problems routinely experienced by subjects on human centrifuges indicates such problems are due to simulator sickness, although they are not reported as such. In this paper, we give a brief overview of simulator sickness and briefly discuss simulator sickness, as related to the human centrifuge experience. PMID:1749507

    Procedures for the micro-determination of beta-cyclodextrin (beta-CyD) in plasma were investigated by four methods using high-performance liquid chromatography (HPLC). In methods A and B, underivatized beta-CyD was detected with a refractive index detector and determined by the absolute calibration graph method. An NH2-bonded silica/acetonitrile-water system was used in A and a C18-bonded silica/methanol-water system in B. In method C, the percarbanilate of beta-CyD was separated on a C8-bonded silica column with acetonitrile-water and determined using gamma-CyD as the internal standard with a UV detector at 231 nm. In method D, the per[1-14C]acetate of beta-CyD was fractionated on a silica column with n-hexane-ethanol containing 1% of water and the radioactivity of each fraction was measured with a liquid scintillation counter. gamma-CyD was used as the internal standard. Interfering plasma proteins were removed by centrifugal ultrafiltration with an MPS-1 micro-partition system. Method B was superior to the other methods with respect to ease of sample preparation, sensitivity and time required for analysis. The cumulative amount of beta-CyD in the mesenteric vein absorbed from the rat intestinal lumen after administration of phenobarbital-beta-CyD complex in a closed loop method was determined by the use of method B.     This paper restricts itself to radial impellers with cylindrical blades since, as Prasil has shown, the flow about an arbitrarily curved surface of revolution may be reduced to this normal form we have chosen by a relatively simple conformal transformation. This method starts from the simple hypotheses of the older centrifugal impeller theory by first assuming an impeller with an infinite number of blades. How the flow is then modified is then investigated. For the computation of flow for a finite number of blades, the approximation method as developed by Munk, Prandtl and Birnbaum, or Glauert is found suitable. The essential idea of this method is to replace the wing by a vortex sheet and compute the flow as the field of these vortices. The shape of the blades is then obtained from the condition that the flow must be along the surface of the blade.

    An experimental study is described to explore the dominant sound generation mechanisms of the spectral components governing the overall noise level of centrifugal compressors. At the design speed with supersonic flow conditions in the rotor blade channels, blade tone noise and buzz-saw noise are the main contributors. On the inlet, rotor-alone noise is the main source while rotor-stator interaction noise dominates on the outlet side in case of vaned outlet diffusers. Over a large range of rotor speeds with subsonic flow conditions, radial compressor noise is dominated by tip clearance noise which is produced by the secondary flow through the gap between rotor blade tips and the casing wall which in turn gives rise to the rotating instability phenomena observed earlier in axial-flow machines.

    Data from space flight and ground based experiments have clearly demonstrated the importance of Earth gravity for normal physiological function in man and animals. Gravitational Physiology is concerned with the role and influence of gravity on physiological systems. Research in this field examines how we perceive and respond to gravity and the mechanisms underlying these responses. Inherent in our search for answers to these questions is the ability to alter gravity, which is not physically possible without leaving Earth. However, useful experimental paradigms have been to modify the perceived force of gravity by changing either the orientation of subjects to the gravity vector (i.e., postural changes) or by applying inertial forces to augment the magnitude of the gravity vector. The later technique has commonly been used by applying centripetal force via centrifugation.

    The Energy and Environmental Research Center (EERC) has teamed with SpinTek Membrane Systems, Inc., the developer of a centrifugal membrane filtration technology, to demonstrate applications for the SpinTek technology within the US Department of Energy (DOE) Environmental management (EM) Program. The technology uses supported microporous membranes rotating at high rpm, under pressure, to separate suspended and colloidal solids from liquid streams, yielding a solids-free permeate stream and a highly concentrated solids stream. This is a crosscutting technology that falls under the Efficient Separations and Processing Crosscutting Program, with potential application to tank wastes, contaminated groundwater, landfill leachate, and secondary liquid waste streams from other remediation processes, including decontamination and decommissioning systems. Membrane-screening tests were performed with the SpinTek STC-X4 static test cell filtration unit, using five ceramic membranes with different pore size and composition. Based on permeate flux, a 0.25-{micro}m TiO{sub 2}/Al{sub 2}O{sub 3} membrane was selected for detailed performance evaluation using the SpinTek ST-IIL centrifugal membrane filtration unit with a surrogate tank waste solution. An extended test run of 100 hr performed on a surrogate tank waste solution showed some deterioration in filtration performance, based on flux, apparently due to the buildup of solids near the inner portion of the membrane where relative membrane velocities were low. Continued testing of the system will focus on modifications to the shear pattern across the entire membrane surface to affect improved long-term performance.

    Phase II of the Centrifuge Workers Study was a follow-up to the Phase I efforts. The Phase I results had indicated a higher risk than expected among centrifuge workers for developing bladder cancer when compared with the risk in the general population for developing this same type of cancer. However, no specific agent could be identified as the causative agent for these bladder cancers. As the Phase II Report states, Phase I had been limited to workers who had the greatest potential for exposure to substances used in the centrifuge process. Phase II was designed to expand the survey to evaluate the health of all employees who had ever worked in Centrifuge Program Departments 1330-1339 but who had not been interviewed in Phase I. Employees in analytical laboratories and maintenance departments who provided support services for the Centrifuge Program were also included in Phase II. In December 1989, the Oak Ridge Associated Universities (ORAU), now known as Oak Ridge Institute for Science and Education (ORISE), was contracted to conduct a follow-up study (Phase II). Phase H of the Centrifuge Workers Study expanded the survey to include all former centrifuge workers who were not included in Phase I. ORISE was chosen because they had performed the Phase I tasks and summarized the corresponding survey data therefrom.

  17. Contaminated vadose zone characterization using partitioning gas tracers

    This paper describes laboratory research conducted to investigate the performance of partitioning tracers for the detection of nonaqueous-phase liquids (NAPLs) in vadose zones. Once evaluated, the tracers may be used for volume estimation of NAPLs and remediation performance assessment of vadose zones. These laboratory studies used glass chromatography columns packed with: (1) Ottawa sand; and then (2) in a separate experiment, soil extracted from the Chemical Waste Landfill at Sandia National Laboratories. The columns were prepared in a manner that resulted in a three-phase system of air, water, and NAPL in the columns. Conservative and partitioning gas tracers were injected into the columns, and their elution concentrations were analyzed. The method of moments was used to estimate partition coefficients between the air and NAPL phases for each of the tracers. The partition coefficients and retardation factors, also estimated during the study, are used to select appropriate tracers for NAPL detection. This research identified several suitable perfluorocarbon tracers and demonstrated the feasibility of using partitioning tracers as a tool for NAPL detection in the vadose zone.

    An analytical equation defining separative power of an optimized concurrent gas centrifuge is obtained for an arbitrary binary mixture of isotopes. In the case of the uranium isotopes the equation gives δU= 12.7(V/700 m/s)2(300 K/T)L, kg SWU/yr, where L and V are the length and linear velocity of the rotor of the gas centrifuge, T is the temperature. This formula well agrees with an empirical separative power of counter current gas centrifuges.

    Dynamics of waves generated by scopes in gas centrifuges (GC) for isotope separation is considered. The centrifugal acceleration in the GC reaches values of the order of 106g. The centrifugal and Coriolis forces modify essentially the conventional sound waves. Three families of the waves with different polarisation and dispersion exist in these conditions. Dynamics of the flow in the model GC Iguasu is investigated numerically. Comparison of the results of the numerical modelling of the wave dynamics with the analytical predictions is performed. New phenomena of the resonances in the GC is found. The resonances occur for the waves polarized along the rotational axis having the smallest dumping due to the viscosity.

    Based on the steady flow in a tube, a mathematical model has been established for the consideration of centrifuging force field by combining the equations of continuity, conservation of momentum and general energy. Effects of centrifugal field on the filling and solidification are modeled by two accessional terms: centrifugal force and Chorios force. In addition, the transfer of heat by convection is considered to achieve a coupling calculation of velocity field and temperature field. The solution of pressure item is avoided by introducing the stream function ψ(x,y) and the eddy function ξ(x,y). Corresponding difference formats for the simultaneous equations of centrifugal filling, the accessional terms and the solidifying latent heat have been established by the finite difference technique. Furthermore, the centrifugal filling and solidification processes in a horizontal tube are summarized to interpret the mechanism by which internal defects are formed in centrifugal castings.

    Gas chromatography (GC) has many applications in the analysis of food products. GC has been used for the determination of fatty acids, triglycerides, cholesterol, gases, water, alcohols, pesticides, flavor compounds, and many more. While GC has been used for other food components such as sugars, oligosaccharides, amino acids, peptides, and vitamins, these substances are more suited to analysis by high performance liquid chromatography. GC is ideally suited to the analysis of volatile substances that are thermally stable. Substances such as pesticides and flavor compounds that meet these criteria can be isolated from a food and directly injected into the GC. For compounds that are thermally unstable, too low in volatility, or yield poor chromatographic separation due to polarity, a derivatization step must be done before GC analysis. The two parts of the experiment described here include the analysis of alcohols that requires no derivatization step, and the analysis of fatty acids which requires derivatization. The experiments specify the use of capillary columns, but the first experiment includes conditions for a packed column.

The elutriated cells were shown to be viable by photosynthetic incorporation of /sup 14/CO/sub 2/ and low release of photosynthetic products into the incubation medium. The level of contamination by clinging debris was low and by host solids was negligible.

    Previously reported effects of chronic centrifugation on renal function in mammals are contradictory. The present study was conducted as an effort to provide a comprehensive analysis of renal response to chronic centrifugation (12 days at +2 Gz). Sixteen male Sprague-Dawley rats (210-230 g) were used: eight centrifuged (EC) and eight off centrifuge controls (OCC). During centrifugation EC had lower body weight and food consumption. EC showed a decrease (72%) in water intake for the first two days (T1 and T2) followed by significant increases from T4-T6. EC urine output increased two-fold over the first four days, returning to baseline by T9. EC urea excretion was elevated on T3 through T5. Creatinine, Na(+), K(+), and osmolar excretion were lower than OCC over the last four days of the study. Assuming constant plasma osmolarity and creatinine levels, EC free water clearance (C(sub H2O)) was elevated significantly on T4 when the peak urine output was exhibited. EC also had a greater C(sub H2O) over the last four days, associated with a significantly lower osmolar clearance and GFR. The initial diuresis exhibited during centrifugation can be attributed to a reduced water resorption and increased urea excretion. This diuresis was mediated independent of changes in GFR over the first eight days. However, differences in excretion seen after eight days of centrifugation are probably GFR mediated which would imply animals established a new homeostatic setpoint by that time. Centrifugation elicites an acute alteration in fluid homeostasis followed by adaptation within a week.

    Fan designs are often constrained by envelope, rotational speed, weight, and power. Aerodynamic performance and motor electrical performance are heavily influenced by rotational speed. The fan used in this work is at a practical limit for rotational speed due to motor performance characteristics, and there is no more space available in the packaging for a larger fan. The pressure rise requirements keep growing. The way to ordinarily accommodate a higher DP is to spin faster or grow the fan rotor diameter. The invention is to put two radially oriented stages on a single disk. Flow enters the first stage from the center; energy is imparted to the flow in the first stage blades, the flow is redirected some amount opposite to the direction of rotation in the fixed stators, and more energy is imparted to the flow in the second- stage blades. Without increasing either rotational speed or disk diameter, it is believed that as much as 50 percent more DP can be achieved with this design than with an ordinary, single-stage centrifugal design. This invention is useful primarily for fans having relatively low flow rates with relatively high pressure rise requirements.

    One interesting class of gravitational radiation sources includes rapidly rotating astrophysical objects that encounter dynamical instabilities. We have carried out a set of simulations of rotationally induced instabilities in differentially rotating polytropes. An n=1.5 polytrope with the Maclaurin rotation law will encounter the m=2 bar instability at T/{vert_bar}W{vert_bar} {ge} 0.27. Our results indicate that the remnant of this in-stability is a persistent bar-like structure that emits a long-lived gravitational radiation signal. Furthermore, dynamical instability is shown to occur in n=3.33 polytropes with the j-constant rotation law at T/{vert_bar}W{vert_bar} {ge} 0:14. In this case, the dominant mode of instability is m=1. Such instability may allow a centrifugally-hung core to begin collapsing to neutron star densities on a dynamical timescale. If it occurs in a supermassive star, it may produce gravitational radiation detectable by LISA.

    Given a large and complex network, we would like to find the partition of this network into a small number of clusters. This question has been addressed in many different ways. In a previous paper, we proposed a deterministic framework for an optimal partition of a network as well as the associated algorithms. In this paper, we extend this framework to a probabilistic setting, in which each node has a certain probability of belonging to a certain cluster. Two classes of numerical algorithms for such a probabilistic network partition are presented and tested. Application to three representative examples is discussed.

    Decline in the abundance of renewable natural resources (RNRs) coupled with increasing demands of an expanding human population will greatly intensify competition for Earth's natural resources during this century, yet curiously, analytical approaches to the management of productive ecosystems (ecological theory of wildlife harvesting, tragedy of the commons, green economics, and bioeconomics) give only peripheral attention to the driving influence of competition on resource exploitation. Here, I apply resource competition theory (RCT) to the exploitation of RNRs and derive four general policies in support of their sustainable and equitable use: (1) regulate resource extraction technology to avoid damage to the resource base; (2) increase efficiency of resource use and reduce waste at every step in the resource supply chain and distribution network; (3) partition ecosystems with the harvesting niche as the basic organizing principle for sustainable management of natural resources by multiple users; and (4) increase negative feedback between consumer and resource to bring about long-term sustainable use. A simple policy framework demonstrates how RCT integrates with other elements of sustainability science to better manage productive ecosystems. Several problem areas of RNR management are discussed in the light of RCT, including tragedy of the commons, overharvesting, resource collapse, bycatch, single species quotas, and simplification of ecosystems.   17. Centrifugal acceleration in the magnetotail lobes

    Combined Cluster EFW and EDI measurements have shown that cold ion outflow in the magnetospheric lobes dominates the hydrogen ion outflow from the Earth's atmosphere. The ions have too low kinetic energy to be measurable with particle instruments, at least for the typical spacecraft potential of a sunlit spacecraft in the tenuous lobe plasmas outside a few RE. The measurement technique yields both density and bulk velocity, which can be combined with magnetic field measurements to estimate the centrifugal acceleration experienced by these particles. We present a quantitative estimate of the centrifugal acceleration, and the velocity change with distance which we would expect due to centrifugal acceleration. It is found that the centrifugal acceleration is on average outward with an average value of about of 5 m s-2. This is small, but acting during long transport times and over long distances the cumulative effect is significant, while still consistent with the relatively low velocities estimated using the combination of EFW and EDI data. The centrifugal acceleration should accelerate any oxygen ions in the lobes to energies observable by particle spectrometers. The data set also put constraints on the effectiveness of any other acceleration mechanisms acting in the lobes, where the total velocity increase between 5 and 19 RE geocentric distance is less than 5 km s-1.

    A bifurcation theory and associated computational model are developed to account for abrupt transitions observed recently on the Maryland Centrifugal eXperiment (MCX) [R. F. Ellis et al. Phys. Plasmas 8, 2057 (2001)], a supersonically rotating magnetized plasma that relies on centrifugal forces to prevent thermal expansion of plasma along the magnetic field. The observed transitions are from a well-confined, high-rotation state (HR-mode) to a lower-rotation, lesser-confined state (O-mode). A two-dimensional time-dependent magnetohydrodynamics code is used to simulate the dynamical equilibrium states of the MCX configuration. In addition to the expected viscous drag on the core plasma rotation, a momentum loss term is added that models the friction of plasma on the enhanced level of neutrals expected in the vicinity of the insulators at the throats of the magnetic mirror geometry. At small values of the external rotation drive, the plasma is not well-centrifugally confined and hence experiences the drag from near the insulators. Beyond a critical value of the external drive, the system makes an abrupt transition to a well-centrifugally confined state in which the plasma has pulled away from the end insulator plates; more effective centrifugal confinement lowers the plasma mass near the insulators allowing runaway increases in the rotation speed. The well-confined steady state is reached when the external drive is balanced by only the viscosity of the core plasma. A clear hysteresis phenomenon is shown.

Overpressured layer chromatography gives a widely used special chapter of forced-flow planar chromatography, a special bridge between high-performance liquid column chromatography and thin-layer chromatography (TLC). A simple and special rule characterizes the progress of mobile phase. Optimal efficiency is composed by the doubled effect of flow resulting from the pump-forced mobile phase (convex profile of laminar flow) and capillary forces on the dry stationary phase (concave laminar flow).     Large-eye impellers for high energy centrifugal pumps were developed to meet the specification of reduced NPSH{sub r} at rated flow conditions. Unfortunately, this improved NPSH performance was not without adverse tradeoffs because an abnormal increase in noise, vibration and cavitation erosion were experienced at low flows. Centrifugal pumps are often used under widely varying and adverse conditions, and in the case of high energy and large-eye impeller pumps, these conditions should be evaluated very carefully. At petrochemical complexes in Secunda in the Republic of South Africa, a centrifugal pump application for lean carbonate solution experienced frequent failures. An investigation and literature survey indicated the cause was low flow cavitation. The purpose of this article is but to give design guidelines as obtained from literature and experience.

  2. Effects of chronic centrifugation on mice

    Previous studies have shown that exposure to excess gravity in vitro alters the developmental sequence in embryonic mouse limbs and palates (Duke, Janer and Campbell, 1984; Duke, 1983). The effects of excess gravity on in vivo mammalian development was investigated using a small animal centrifuge. Four-week old female mice exposed to excess gravities of 1.8-3.5 G for eight weeks weighed significantly less than controls. Mice were mated after five weeks of adaptation to excess G, and sacrificed either at gestational day 12 or 18. There were fewer pregnancies in the centrifuged group (4/36) than in controls (9/31), and crown rump lengths (CRL) of embryos developing in the centrifuge were less than CRLs of 1-G embryos. These results show that although immersed in amniotic fluid, embryos are responsive to Delta-G.

    Electrically assisted boost is a prominent method to solve the issues of transient lag in turbocharger and remains an optimized operation condition for a compressor due to decoupling from turbine. Usually a centrifugal compressor for gasoline engine boosting is operated at high rotational speed which is beyond the ability of an electric motor in market. In this paper a centrifugal compressor with rotational speed as 120k RPM and pressure ratio as 2.0 is specially developed for electrically assisted boost. A centrifugal compressor including the impeller, vaneless diffuser and the volute is designed by meanline method followed by 3D detailed design. Then CFD method is employed to predict as well as analyse the performance of the design compressor. The results show that the pressure ratio and efficiency at design point is 2.07 and 78% specifically.

    The possibility of the segregation under solidification process using a centrifugal casting machine was investigated using an electron probe microanalyzer with elemental distribution map, line analysis and quantitative analysis. When a very small quantity of platinum was added to local molten titanium during the casting process, macroscopic segregation was observed under conditions of density difference of 0.1 g/cm3 at the most, confirming that the centrifugal force of the casting machine is extremely strong. When a Ti-6Al-4V alloy was cast, however, no macroscopic segregation was observed. The centrifugal force of the casting machine examined in the present study hardly results in the body-force segregation in this titanium alloy. PMID:9550020

    Over the last eight years, Retech has developed a plasma-powered system for destroying organics and stabilizing metal oxides in a non-leaching slag. The system, termed Plasma Arc Centrifugal Treatment (PACT), can handle a variety of waste streams such as contaminated soils, sludges, ion-exchange resins, incinerator bottom and fly-ash and drummed waste among others. This paper will review recent commercial applications of the technology. Three Plasma Arc Centrifugal Treatment systems having an eight-foot diameter centrifuge (PACT-8) are in the construction phase. One will be used in the Lockheed Environmental Systems and Technologies (LESAT) system for remediating Pit 9 at the Idaho National Engineering Lab (INEL). A second unit will be located at the Retech plant in Ukiah, California. The third unit will be located at a site in Munster, Germany.

  7. Reduced-order modeling for mistuned centrifugal impellers with crack damages

    An efficient method for nonlinear vibration analysis of mistuned centrifugal impellers with crack damages is presented. The main objective is to investigate the effects of mistuning and cracks on the vibration features of centrifugal impellers and to explore effective techniques for crack detection. Firstly, in order to reduce the input information needed for component mode synthesis (CMS), the whole model of an impeller is obtained by rotation transformation based on the finite element model of a sector model. Then, a hybrid-interface method of CMS is employed to generate a reduced-order model (ROM) for the cracked impeller. The degrees of freedom on the crack surfaces are retained in the ROM to simulate the crack breathing effects. A novel approach for computing the inversion of large sparse matrix is proposed to save memory space during model order reduction by partitioning the matrix into many smaller blocks. Moreover, to investigate the effects of mistuning and cracks on the resonant frequencies, the bilinear frequency approximation is used to estimate the resonant frequencies of the mistuned impeller with a crack. Additionally, statistical analysis is performed using the Monte Carlo simulation to study the statistical characteristics of the resonant frequencies versus crack length at different mistuning levels. The results show that the most significant effect of mistuning and cracks on the vibration response is the shift and split of the two resonant frequencies with the same nodal diameters. Finally, potential quantitative indicators for detection of crack of centrifugal impellers are discussed.

Centrifuge diameter (1.8m, 2.5m or 6.0m) and schedule of operation (Daily or weekly stop) varied between groups. Body mass, food consumption, water consumption and neurovestibular function were measured weekly. Body temperature and activity were continuously monitored using telemetry. A subset of subjects were videotaped (50 minutes per day) to allow for movement analysis. Exposure to a hyperdynamic field of this magnitude did cause the expected depression in the physiological variables monitored. Recovery was accomplished within a relatively rapid time frame; all variables returned to precentrifugation levels. In general, the magnitudes of the changes and the rate of recovery were similar at different centrifuge diameters and stopping frequency. There were cases, however, in which the magnitude of the response and/or the rate of recovery to a new steady-state were altered as a result of centrifuge diameter. In summary, these results indicate that stopping frequency has little, if any, effect on adaptation to chronic centrifugation. However, the angular velocity (omega), and therefore centrifuge diameter is an important consideration in the adaptation of an organism to chronic centrifugation.

Natural gas liquids that condense under pressure in the separating drum pass through openings in the drum into the space between the drum and housing. In this space, a series of openings, or throttling restrictors, allows the liquids to expand and return to gas. The gaseous component that does not liquefy in the drum remains separate for drawing off.

    System analyses of cylindrical plasma centrifuges are presented, for which the velocity field and electromagnetic fields are calculated. The effects of different electrode geometrics, induced magnetic fields, Hall-effect, and secondary flows are discussed. It is shown that speeds of 10000 m/sec can be achieved in plasma centrifuges, and that an efficient separation of U238 and U235 in uranium plasmas is feasible. The external boundary-value problem for the deposition of sputtering products is reduced to a Fredholm integral equation, which is solved analytically by means of the method of successive approximations.

    Centrifugal mirrors use supersonic rotation to provide axial confinement and enhanced stability. Usually the rotation is produced using electrodes, but these electrodes have limited the rotation to the Alfven critical ionization velocity, which is too slow to be useful for fusion. Instead, the rotation could be produced using radio frequency waves. A fixed azimuthal ripple is a simple and efficient wave that could produce rotation by harnessing alpha particle energy. This is an extension of the alpha channeling effect. The alpha particle power and efficiency in a simulated devices is sufficient to produce rotation without external energy input. By eliminating the need for electrodes, this opens new opportunities for centrifugal traps.

    We are proposing a new approach to investigate the physical effects of the centrifugal acceleration in an inertial reference system. Our proposal is based primarily on geometrical considerations, built on the extension of the equivalence principle. The equivalence principle is then used to derive an effective Schwarzschild radius. After having developed the equations, we propose an experiment to detect the effect of the centrifugal acceleration on the time. We compute also the consequence on the unstable particle's lifetime. The testability of our model is well reachable from our present technology.

    Centrifuges are devices that separate particles of different densities and sizes through the application of a centrifugal force. If a centrifuge could be operated under atmospheric conditions, all vacuum-related components such as the vacuum chamber, vacuum pump, diffusion pump, and sealing could be removed from a conventional centrifuge system. The design and manufacturing procedure for centrifuges could then be greatly simplified to facilitate the production of lightweight centrifuge systems of smaller volume. Furthermore, the maintenance costs incurred owing to wear and tear due to conventional ball bearings would be eliminated. In this study, we describe a novel vacuum chamber-free centrifuge supported by magnetic bearings. We demonstrate the feasibility of the vacuum chamber-free centrifuge by presenting experimental results that verify its high-speed support capability and motoring power capacity.

    Compressor units up to 2 MW are typically dominated by piston compressors driven by spark-ignited reciprocating piston engines, called recips. Waertsilae Compression Systems competes in the output range of 1--3 MW from other piston engine manufacturers and says its larger prime movers like the 255G, 285G and 345G will match gas turbine-driven turbo compressors, called centrifugals. The centrifugals dominate the business worldwide today in unit sizes above 3 MW. Technically a centrifugal compressor is very much like a gas turbine and so is its performance. It is a good machine when the operation conditions are ideal for it, i.e., compression ratio is low (<1:1.5), gas pressure and temperature are constant and it operates only on full load, at sea level and at an ambient temperature below 15 degrees C. This is seldom the case. To establish the most economical compression solution for a customer, compare the lifetime costs to compress the planned amount of gas in real operating and site conditions. The lifecycle costs of recips driven by spark-ignited gas engines were compared to the corresponding costs of centrifugal compressor units driven by gas turbines. Cases with typical operation profiles in realistic site conditions were analyzed.

The model assumes 1) a definition of an escaping particle as the one that flows in every operational point from the trailing edge towards the leading edge of the impeller blades, and 2) a characterization of flow where an operational point is said to have a theoretical flow pattern if it is not possible to establish a fully-reversed escaping particle on it. Therefore, the first part of the article is focused on defining an escaping particle for a centrifugal compressor. The model locates over the map of a centrifugal impeller the line that splits the map in two regions: the region on the right hand side, where a theoretical flow pattern can exist, and the region on the left, where a theoretical flow pattern cannot exist. Therefore, the locus of this line marks a frontier where the expected performance of the impeller cannot be sustained as high as expected. The second part of the article uses a high-performance commercial centrifugal impeller wheel for contrasting the model. A qualitative characterization of the surge line, conclusions and discussions are presented.

    Equations are developed for liquid metal pressure in centrifugal dental casting, given the instantaneous rotational velocity, density, and certain dimensions of the casting machine and casting pattern. A "reference parabola" is introduced making the fluid pressure concept more understandable. A specially designed specimen demonstrates experimentally the reference parabola at freezing. PMID:355283

    A Type III gold alloy, a silver-palladium alloy, and a base metal alloy were cast in two different centrifugal casting machines. With the number of complete cast mesh squares as an indicator of castability, the Airspin casting machine produced superior castings with all three alloys. The base metal alloy produced the greatest number of complete squares with both casting machines. PMID:3889295

    A prototype flow visualization system is constructed to examine buoyancy driven flows during centrifugation in space. An axial density gradient is formed by imposing a thermal gradient between the two ends of the test cell. Numerical computations for this geometry showed that the Prandtl number plays a limited part in determining the flow.

    A magnetized, centrifugally confined plasma is subjected to a 3D MHD stability test. Ordinarily, the system is expected to be grossly unstable to "flute" interchanges of field lines. Numerical simulation shows though that the system is stable on account of velocity shear. This allows consideration of a magnetically confined plasma for thermonuclear fusion that has a particularly simple coil configuration. PMID:11736455

  4. Differential white cell count by centrifugal microfluidics.

    We present a method for counting white blood cells that is uniquely compatible with centrifugation based microfluidics. Blood is deposited on top of one or more layers of density media within a microfluidic disk. Spinning the disk causes the cell populations within whole blood to settle through the media, reaching an equilibrium based on the density of each cell type. Separation and fluorescence measurement of cell types stained with a DNA dye is demonstrated using this technique. The integrated signal from bands of fluorescent microspheres is shown to be proportional to their initial concentration in suspension. Among the current generation of medical diagnostics are devices based on the principle of centrifuging a CD sized disk functionalized with microfluidics. These portable 'lab on a disk' devices are capable of conducting multiple assays directly from a blood sample, embodied by platforms developed by Gyros, Samsung, and Abaxis. [1,2] However, no centrifugal platform to date includes a differential white blood cell count, which is an important metric complimentary to diagnostic assays. Measuring the differential white blood cell count (the relative fraction of granulocytes, lymphocytes, and monocytes) is a standard medical diagnostic technique useful for identifying sepsis, leukemia, AIDS, radiation exposure, and a host of other conditions that affect the immune system. Several methods exist for measuring the relative white blood cell count including flow cytometry, electrical impedance, and visual identification from a stained drop of blood under a microscope. However, none of these methods is easily incorporated into a centrifugal microfluidic diagnostic platform.

    One eight-stage unit (8-pack) of centrifugal contactors was tested in both extraction and stripping modes. Efficiencies approaching 100 percent were obtained in both modes. The contactors were operated successfully at a wide range of combined flow rates, including the HEF conditions. This report discusses the results of that test.
