The environmental impact of using recombinant bovine somatotropin (rbST) in dairy production was examined on an individual cow, industry-scale adoption, and overall production system basis. An average 2006 U.S. milk yield of 28.9 kg per day was used, with a daily response to rbST supplementation of 4.5 kg per cow. Rations were formulated and both resource inputs (feedstuffs, fertilizers, and fuels) and waste outputs (nutrient excretion and greenhouse gas emissions) calculated. The wider environmental impact of production systems was assessed via acidification (AP), eutrophication (EP), and global warming (GWP) potentials. From a producer perspective, rbST supplementation improved individual cow production, with reductions in nutrient input and waste output per unit of milk produced. From an industry perspective, supplementing one million cows with rbST reduced feedstuff and water use, cropland area, N and P excretion, greenhouse gas emissions, and fossil fuel use compared with an equivalent milk production from unsupplemented cows. Meeting future U.S. milk requirements from cows supplemented with rbST conferred the lowest AP, EP, and GWP, with intermediate values for conventional management and the highest environmental impact resulting from organic production. Overall, rbST appears to represent a valuable management tool for use in dairy production to improve productive efficiency and to have less negative effects on the environment than conventional dairying. Two groups of five crossbred 87.5% Holstein cattle were housed in normal shade only (NS) as non-cooled cows and in shaded housing with misty-fan cooling (MF) as cooled cows. The cows were treated with recombinant bovine somatotropin (rbST) in early, mid and late lactation with three consecutive injections of rbST in every 14 days. Ambient temperatures and the temperature humidity index in the NS barn were significantly higher than those of the MF barn, whereas relative humidity in MF was higher than that of NS barn. The DMI of cooled cows were higher than those of non-cooled cows, and cooled cows exhibited more response to rbST treatment. Exogenous rbST significantly increased milk yield throughout lactation. The rbST-treated cows had higher total ruminal fermentation products as volatile fatty acid and ammonia nitrogen than the non-rbST treated cows and associated changes were greater in cooled animals in all stages of lactation. Exogenous rbST increased the concentrations of milk urea nitrogen in both groups. These results suggest that the changes in ruminal fermentation with greater production of total VFA and NH(3)N in response to rbST in crossbred cows whether under misty-fan cooling or not, is in part through an increase in feed intake, thereby making more substrate available to the mammary gland for milk synthesis. Our objective in this study was to determine the effect of recombinant bovine somatotropin (rbST) on indices of muscle development in creep-fed beef calves. Crossbred steer calves were assigned to one of two treatment groups: control (sham-injected; n = 12) or rbST-treated (.09 mg x kg(-1) x d(-1); n = 12). Calves were injected every 14 d starting at d 28 of age and were weaned at 205 d of age. Supplemental creep feed was supplied free access to all calves to compensate for an expected increased protein and energy requirement in calves given rbST. Biopsy (d 100) and slaughter (d 206) samples of semitendinosus muscle were evaluated for satellite cell, myofiber nuclei numbers, and myosin light chain (MLC-1f) mRNA quantification. Myofiber nuclei and satellite cell numbers per 100 myofibers and MLC-1f mRNA:rRNA ratios at 100 and 206 d of age were not different (P > .10) between control and rbST-treated calves. Total gain, ADG, quality grade, femur length, percentage kidney, pelvic, and heart fat, dressing percentage, plasma IGF-I, and plasma urea nitrogen concentrations did not differ (P > .10) between control and rbST-treated calves. However, rbST-treated calves had larger longissimus muscle areas (P < .03), less marbling (P < .001), higher carcass conformation scores (P < .04), greater mass of separated muscle (P < .03), more ground meat (P < .01), and heavier carcass weights (P < .05) than control calves. Thus, rbST treatment increased muscle characteristics while nuclei number and MLC-1f mRNA concentrations remained the same, implying that the additional muscle growth was in a normal fashion.

The aim of this experiment was to determine effects of treating peripartum dairy cows with body condition score ≥3.75 with recombinant bovine somatotropin (rbST) on immune, inflammatory, and metabolic responses. Holstein cows (253±1d of gestation) were assigned randomly to 1 of 3 treatments: untreated control (n=53), rbST87.5 (n=56; 87.5mg of rbST), and rbST125 (n=57; 125mg of rbST). Cows in the rbST87.5 and rbST125 treatments received rbST weekly from -21 to 28d relative to calving. Growth hormone, insulin-like growth factor 1, haptoglobin, tumor necrosis factor α, nonesterified fatty acids, β-hydroxybutyrate, glucose, and cortisol concentrations were determined weekly from -21 to 21d relative to calving. Blood sampled weekly from -14 to 21d relative to calving was used for hemogram and polymorphonuclear leukocyte (PMNL) expression of adhesion molecules, phagocytosis, and oxidative burst. Cows were vaccinated with ovalbumin at -21, -7, and 7d relative to calving, and blood was collected weekly from -21 to 21d relative to calving to determine IgG anti-ovalbumin concentrations. A subsample of cows had liver biopsied -21, -7, and 7d relative to calving to determine total lipids, triglycerides, and glycogen content. Growth hormone concentrations prepartum (control=11.0±1.2, rbST87.5=14.1±1.2, rbST125=15.1±1.3ng/mL) and postpartum (control=14.4±1.1, rbST87.5=17.8±1.2, rbST125=21.8±1.1ng/mL) were highest for rbST125 cows. Cows treated with rbST had higher insulin-like growth factor 1 concentrations than control cows (control=110.5±4.5, rbST87.5=126.2±4.5, rbST125=127.2±4.5ng/mL) only prepartum. Intensity of L-selectin expression was higher for rbST125 than for control and rbST87.5 cows [control=3,590±270, rbST87.5=3,279±271, rbST125=4,371±279 geometric mean fluorescence intensity (GMFI)] in the prepartum period. The PMNL intensities of phagocytosis (control=3,131±130, rbST87.5=3,391±133, rbST125=3,673±137 GMFI) and oxidative burst (control=9,588±746

A comprehensive econometric model was developed to evaluate potential impacts of recombinant bovine somatotropin (rbST) approval in Japan. Three novel features of the analyses include 1) investigation of impact of rbST on herd-size structure, 2) determination of economic feasibilities of rbST adoption by herd size, and 3) evaluation of policies to mitigate negative effects of rbST approval. Simulation analysis was conducted over a 10-yr projected period assuming rbST was approved in Japan in 2001. Nine hypothetical scenarios were simulated to examine sensitivity of simulation results. Simulation results indicate that rbST approval would accelerate structural change in Japan's dairy industry toward fewer, larger farms. Negative effects of rbST on farm income are projected to be more severe for smaller farms, because of higher costs, lower profit-earning ability, lower milk yields, and lower adoption rates of rbST. Larger farms benefit from rbST adoption if milk demand is maintained. However, if concerns about rbST induce significant milk demand decreases, even the largest farms' income and cow numbers will decrease. Thus, Japan's dairy industry could be caught in a double downward spiral of declining milk prices and production. Assuming rbST is approved, small farms would benefit by using the technology, but they fare best if rbST is not approved. Two policies could be effective in mitigating possible farm income losses. First, lost farm income can be offset if dairy cooperatives can exercise greater market power to control fluid milk marketings. Second, because generic milk advertising has positive effects on both milk demand and farm income, increasing check-off rates to fund more advertising could ease farm income losses. PMID:15291007

We present preliminary results on the dynamical influence of an adiabatically grown central black hole upon its surroundings. Young (1980), in his pioneer work, found an r-3/2 density profile for the neighborhood of a black hole, a result not confirmed by observation. In this work, we extend Young's result to a more realistic case, namely an inhomogeneous galactic center, although keeping the spherical symmetry hypothesis, in order to verify whether the final density profile changes.

Recombinant bovine somatotropin (rbST) is a peptide hormone used to increase milk yield in cows and buffalos. In Europe, its use has been banned. However, rbST is sometimes illegally included in zootechnical practices for profit purposes, undermining the fair trade and the law prescriptions. For this reason, efficient and reliable analytical techniques are required to contrast rbST misuse. A few LC-MS-MS methods have been developed to detect, in cow serum, methyonil-rbST, one of the two main rbST forms available on the market. The other form, which is widespread, is identical to the most abundant variant of bovine somatotropin (bST) and differs from the buffalo somatotropin for one amino acid in the N-terminus. For this reason, it is technically possible to distinguish both rbST forms in serum of buffalos. In this work, we describe a novel LC-MS-MS-based method, capable to quantify, with a high sensitivity and selectivity, the methyonil-rbST and the other bST-identical recombinant form in buffalo serum, previously purified using a solid-phase extraction procedure. The method was internally validated and used to analyse 152 serum samples, collected from eight buffalos administered with rbST for a period of 3 months, according to conventional protocols. The obtained results confirmed the suitability of the method in the detection of illegal hormonal treatments. Graphical abstract ᅟ. PMID:27146507

The use of recombinant bovine somatotropin (rbST) to enhance milk production is approved in several countries, but it is prohibited in the European Union. According to EU legislation, it is necessary to confirm positive screening results prior to enforcement. Although adequate screening assays are available nowadays, development of liquid chromatography tandem mass spectrometry (LC-MS/MS) confirmatory methods to detect low levels of rbST is still a challenge. Here, we present a novel approach using immuno-affinity enrichment on monolithic micro-columns in combination with state-of-the-art ultra-high pressure LC-MS/MS (UHPLC-MS/MS) detection. The developed approach enables detection and confirmation of rbST in serum at a decision limit (CCα) concentration of 0.8 ng mL(-1). Furthermore, the method is easy to handle, robust and reproducible. We successfully applied the confirmatory method to serum samples from rbST treated cows that were found suspect after immunoassay-based screening. The use of rbST could be confirmed over 1 week after treatment, and the developed method demonstrated the sensitivity needed for effective control. The effect of supplementary administration of recombinant bovine somatotrophin (rbST) on the renal tubular handling of sodium in crossbred 87.5% Holstein cattle housed in normal shade (NS) or mist-fan cooled (MF) barns was evaluated. The cows were injected with 500 mg rbST at three different stages of lactation. The MF barn housed cows showed a slightly decreased ambient temperature and temperature humidity index, but an increased relative humidity. Rectal temperature and respiration rates were significantly lower in cooled cows. The rbST treated cows, housed in NS or MF barns, showed markedly increased milk yields, total body water, extracellular fluid and plasma volume levels, along with a reduced rate of urine flow and urinary excretion of sodium, potassium and chloride ions and osmolar clearance, in all three stages of lactation. Renal tubular sodium and water reabsorption were increased after rbST administration without any alteration in the renal hemodynamics. Lithium clearance data suggested that the site of response is in the proximal nephron segment, which may be mediated via increases in the plasma levels of aldosterone and IGF-1, but not vasopressin, during rbST administration. The 20 yr of commercial use of recombinant bovine somatotropin (rbST) in the United States provide the backdrop for reviewing the outcome of use on human health issues by the upcoming 78th meeting of the Joint Food and Agriculture Organization of the United Nations (FAO)/World Health Organization (WHO) Expert Committee on Food Additives. These results and further advancements in scientific knowledge indicate there are no new human health issues related to the use of rbST by the dairy industry. Use of rbST has no effect on the micro- and macrocomposition of milk. Also, no evidence exists that rbST use has increased human exposure to antibiotic residues in milk. Concerns that IGF-I present in milk could have biological effects on humans have been allayed by studies showing that oral consumption of IGF-I by humans has little or no biological activity. Additionally, concentrations of IGF-I in digestive tract fluids of humans far exceed any IGF-I consumed when drinking milk. Furthermore, chronic supplementation of cows with rbST does not increase concentrations of milk IGF-I outside the range typically observed for effects of farm, parity, or stage of lactation. Use of rbST has not affected expression of retroviruses in cattle or posed an increased risk to human health from retroviruses in cattle. Furthermore, risk for development of type 1 or type 2 diabetes has not increased in children or adults consuming milk and dairy products from rbST-supplemented cows. Overall, milk and dairy products provide essential nutrients and related benefits in health maintenance and the prevention of chronic diseases. PMID:24663163

The Archean East Indian cratonic margin was affected by the Kerguelen plume (KP) ˜117 Ma, causing flood-basalt eruptions of the Rajmahal-Bengal-Sylhet Traps (RBST). The RBST cover ˜one million km2 in and around the Bengal Basin as alkalic-ultrabasic intrusives in the west and Sikkim in the north, and Sylhet basalts and alkalic-carbonatitic-ultramafic complexes in the Shillong plateau - Mikir hills farther east of the Rajmahal-Bengal Traps. We provide new Nd-Sr-Pb-isotopic and trace element data on 21 unreported discrete lava flows of the Rajmahal Traps, 56 alkalic-carbonatitic-mafic-ultramafic rocks from four alkalic complexes, and three dikes from the Gondwana Bokaro coalfields, all belonging to the RBST. The data allow geochemical correlation of the RBST with some contemporaneous Kerguelen Plateau basalts and KP-related volcanics in the southern Indian Ocean. Specifically, the new data show similarity with previous data of Rajmahal group I-II basalts, Sylhet Traps, Bunbury basalts, and lavas from the southern Kerguelen Plateau, indicating a relatively primitive KP source, estimated as: ɛNd(I) = +2, 87Sr/86Sr(I) = 0.7046, with a nearly flat time-integrated rare earth element (REE) pattern. We model the origin of the uncontaminated RBST basalts by ˜18% batch melting with a 2× chondritic KP source in the spinel-peridotite stability depths of 60-70 km in the mantle. The new geochemical data similar to the Rajmahal group II basalts indicate a light REE enriched average source at ɛNd(I) = -5, 87Sr/86Sr(I) = 0.7069. Our geochemical modeling indicates these lavas assimilated granulites of the Eastern Ghats, reducing the thickness of the continental Indian lithosphere. Lack of an asthenospheric MORB component in the RBST province is indicated by various trace element ratios as well as the Nd-Sr isotopic ratios. Three alkalic complexes, Sung, Samchampi, and Barpung in NE India, and one in Sikkim to the north are of two groups: carbonatites, pyroxenites, lamproites

Recombinant bovine somatotrophin (rbST) is widely used in some countries to increase milk production. Since 1994, both marketing and use of this substance have been prohibited within the European Union. In this context, the targeted plasma biochemical and hormonal profiling was assessed as a potential screening strategy to highlight rbST (ab)use in cattle. Twenty-one routinely measured clinical blood parameters, representative of main biological profiles (energetic, proteic, etc.), were measured in the plasma of six lactating cows before and after rbST treatment throughout a 23-day study period. Appropriate multivariate statistical analyses [principal component analysis (PCA) and orthogonal partial least square (OPLS)] enabled discriminating animal samples before and after treatment (days 0 vs. 2 to 9, P = 2.10(-9)) and highlighted the five most relevant blood parameters in this discrimination. Based on each five-analyte contribution, a simple mathematically weighted equation was suggested to predict the status of samples. A suspicious threshold was proposed, and the model was further tested with the status prediction of the supplementary samples from untreated (n = 20) and treated cows (n = 22). The calculated false-positive (10%) and false-negative (4.5%) rates were in accordance with the EU requirements for screening methods. Although the model needs to be further validated with additional samples, such targeted plasma biochemical and hormonal profiling already appears as a potential promising screening strategy to highlight rbST (ab)use in cattle. The effect of administering recombinant bovine somatotropin (rbST) to cows with hyperketonemia during the early postpartum period on health, metabolic parameters, milk production, and early reproductive performance was evaluated in a double-blinded clinical trial. Cows from 8 dairy herds in New York State were tested weekly between 3 and 16d in milk for elevated serum β-hydroxybutyrate. Cows were enrolled in the study when blood β-hydroxybutyrate was ≥1.3mmol/L for the first time. Enrolled cows were randomly assigned to a treatment (n=273) or placebo control (n=270) group. Treated cows were given 325mg of rbST subcutaneously on the day of enrollment and again 14d later. Control cows received the same regimen except the syringe contained only the carrier without somatotropin. After enrollment, blood samples were collected weekly for 4wk and submitted to the laboratory to be analyzed for selected metabolites. Risk ratios for clinical diseases subsequent to treatment were calculated using Poisson regression. Continuous data were analyzed using linear mixed models. Time to first insemination was assessed with survival analysis. In the 42d following the first administration of rbST, incidence risks of displaced abomasum, clinical ketosis, metritis, clinical mastitis, and lameness were not different between treatment groups. Cows treated with rbST had a slightly lower body condition score 28d after enrollment compared with control cows. In the 4wk following enrollment, serum nonesterified fatty acids and aspartate amino-transferase were slightly higher for treated than control cows, respectively. Serum glucose, calcium, haptoglobin, and β-hydroxybutyrate were similar between groups. Treatment had no effect on resolution of hyperketonemia in any of the 4wk after enrollment. Milk production in either of the 2-wk periods after each treatment was not different between treated and control cows. Furthermore, milk production was not different between groups from enrollment

The study objective was to determine if feeding the rumen-degradable AA Val can increase milk production comparable to recombinant bovine somatotropin (bST). Eight multiparous late-lactating (255±26.4 d in milk) Holstein dairy cows were blocked by milk yield (34.1±8.25 kg/d) and randomly assigned to 1 of 4 treatments in a replicated 4×4 Latin square design with 21-d periods (7 d for dietary adaptation and 14 d for data collection). Treatments were control (CON), a single injection of recombinant bST (rbST), and Val fed at 40 (V40) and 80 g/d (V80). Cows were fed a total mixed ration with a distillers dried grains carrier at 113.4 g/d containing none or added AA. Dry matter intake (21.3, 22.0, 22.8, and 21.5 kg/d for CON, rbST, V40, and V80, respectively) was similar among treatments, except cows receiving V40 had greater dry matter intake than cows receiving V80. Milk yield (22.0, 26.1, 25.2, and 24.9 kg/d), 3.5% fat-corrected milk (22.1, 25.4, 24.4, and 24.3 kg/d), and energy-corrected milk (22.7, 26.1, 25.1, and 24.9 kg/d) were increased at similar amounts for cows receiving rbST, V40, and V80 compared with CON cows. Milk fat percentages (3.51, 3.36, 3.32, and 3.38%) were greatest for CON cows compared with cows receiving V40, whereas cows receiving other treatments were intermediate and similar. Milk protein percentages (3.20, 3.12, 3.15, and 3.13%) were greater for CON cows compared with cows receiving rbST and V40, whereas cows receiving V80 were intermediate and similar. Ruminal isobutyrate (1.19, 1.24, 1.44, and 1.74 mol/100 mol) concentrations were increased for cows receiving V40 and V80 compared with CON and rbST cows, with cows receiving V80 having greater concentrations than cows receiving V40. Twenty bulls (Aberdeen Angus and Brangus) were divided by breed into two groups. Placebo group was injected with NaCl 0.9% (s.c.) and treatment group with rbST (s.c., 500 mg) at days 0 and 14 of the experiment. Immediately after semen collection, blood samples were taken on days 0, 14, 28, 42 and 56 of the experiment. Semen was also collected on day 70 of the experiment. Evaluation of sperm motility was performed at pre-freezing and post-thawing stage, whereas assessment of sperm membrane integrity was performed after freezing and thawing. Analysis of data revealed that the effect of treatment and treatment-by-collection day on plasma concentrations of IGF-I and insulin was not significant. However, mean plasma concentrations of IGF-I and insulin were affected (p < 0.0001) by days of blood sampling. Effect of treatment and treatment-by-collection day on motility of spermatozoa was similar (p > 0.05) at pre-freezing and post-thawing stage. Intactness of plasmalemma and tail membrane of spermatozoa at post-thawing stage was higher (p < 0.05) in rbST-treated group than in control. In conclusion, rbST did not affect plasma concentrations of IGF-I and insulin, however, it did improve post-thaw sperm membrane integrity. PMID:19663813

Records representing data from 1,500 barren Holstein cows over an 8-yr period from a large commercial dairy farm in northern Mexico were analyzed to determine the effects of lactation number and season and year of initiation of lactation on milk production of cows induced hormonally into lactation and treated with recombinant bovine somatotropin (rbST) throughout lactation. Peak and 305-d milk yields were also assessed as predictors of total milk yield in cows induced into lactation. A significant quadratic relationship was found between 305-d milk yield and number of lactation [7,607±145 and 9,548±181 kg for first- and ≥6-lactation cows, respectively; mean ± standard error of the mean (SEM)] with the highest production occurring in the fifth lactation. Total milk yields of cows with ≤2 lactations were approximately 4,500 kg less than milk yields of adult cows (the overall average ± standard milk yield was 13,544±5,491 kg per lactation and the average lactation length was 454±154 d). Moreover, 305-d milk production was depressed in cows induced into lactation in spring (8,804±153 kg; mean ± SEM) and summer (8,724±163 kg) than in fall (9,079±151 kg) and winter (9,085±143 kg). Partial regression coefficients for 305-d milk yield and peak milk yield indicated an increment of 157 kg of milk per lactation per 1-kg increase in peak milk yield (r(2)=0.69). Neither peak milk yield (r(2)=0.18) nor 305-d milk yield (r(2)=0.29) was accurate for predicting total milk yield per lactation. Year, parity, and season effects had significant influence on milk yield of cows induced into lactation and treated with rbST throughout lactation, and peak milk yield can assist in the prediction of 305-d milk yield but not total milk yield. This study also showed that hormonal induction of lactation in barren high-yielding cows is a reliable, practical, and affordable technique in countries where rbST treatment and prolonged steroid administration of dairy cows are legally

A total of 140 true anestrous buffalo were divided on the basis of receiving short-term (20 days) nutritional supplementation (N, n=80) or not (WN, n=60). The animals in N group were subdivided into NQ (n=40) where the quantity of the offered diet was increased by 20% and NF (n=40) where the offered diet was supplemented by 3% of dry protected fat. Buffaloes in either NQ or NF were equally allotted on the following treatment regimens: Insulin/GnRH (NQi or NFi, n=10 for each); rbST/GnRH (NQr or NFr, n=10 for each); GnRH alone treated (NQG or NFG, n=10 for each) and saline-treated control (NQc or NFc, n=10 for each). Insulin-treated subgroups (NQi or NFi) received s/c injection of insulin at a dose of 0.25 I.U./kg on Days 21, 22 and 23 while rbST-treated subgroups (NQr or NFr) received single IM injection of rbST (500 mg Sometribove) on Day 21. GnRH was injected at a dose of 0.020 mg buserelin (5 ml Receptal(®)) on Day 24 in all subgroups except NQCand NFC where Day 1 was the first day of the short-term nutritional improvement. Buffalo in the WN (n=60) were equally allotted on the same treatment regimens applied in the first group insulin/GnRH (WNi, n=15), rbST/GnRH (WNr, n=15); GnRH alone treated (WNG, n=15) and saline-treated control (WNC, n=15). Ultrasonic scanning of ovaries was conducted on Day 24 to measure largest follicle diameter (LFD). The results showed increases (P<0.05) in the LFD following nutritional supplementation with insulin or rbST. The recorded EIRs for GnRH pre-treated with nutritional improvement - metabolic hormones combinations (9/10 and 8/10 for NQi and NFi or 8/10 for NQr) were greater (P<0.05) than those pre-treated with either metabolic hormone alone (7/15 for WNi and/or WNr) or nutritional improvement alone (6/10 for NQG and/or NFG) and control as well. The greatest CR was recorded in the NQi group. It could be concluded that pre-GnRH nutritional improvement plus administration of insulin or rbST increases LFD in true anestrous buffalo

The dairy industry in the United States has undergone many changes over the past century. Adulteration and contamination of milk were rampant before the passage and enforcement of the Pure Food and Drug Act of 1906, and the introduction and eventual acceptance of certified and pasteurized milk have provided consumers with a consistently safe product. Homogenization and advances in the packaging and transport of milk gradually took hold, improving the milk supply. Other developments included the concentration of milk and whey, lactose-reduced milk, and the popularization of yogurt. Consumers have benefited from advances in butter packaging, low-fat ice cream, cheese manufacture, and yogurt technology, which has helped create the large demand for dairy products in the United States. Current trends and issues, including the increasing popularity of organic and artisanal products and the use of rBST, will shape the future of the dairy industry. PMID:19719132

Here we present the concept of a protein microarray-based fluorescence immunoassay for multiple biomarker detection in milk extracts by an ordinary smartphone. A multiplex immunoassay was designed on a microarray chip, having built-in positive and negative quality controls. After the immunoassay procedure, the 48 microspots were labelled with Quantum Dots (QD) depending on the protein biomarker levels in the sample. QD-fluorescence was subsequently detected by the smartphone camera under UV light excitation from LEDs embedded in a simple 3D-printed opto-mechanical smartphone attachment. The somewhat aberrant images obtained under such conditions, were corrected by newly developed Android-based software on the same smartphone, and protein biomarker profiles were calculated. The indirect detection of recombinant bovine somatotropin (rbST) in milk extracts based on altered biomarker profile of anti-rbST antibodies was selected as a real-life challenge. RbST-treated and untreated cows clearly showed reproducible treatment-dependent biomarker profiles in milk, in excellent agreement with results from a flow cytometer reference method. In a pilot experiment, anti-rbST antibody detection was multiplexed with the detection of another rbST-dependent biomarker, insulin-like growth factor 1 (IGF-1). Milk extract IGF-1 levels were found to be increased after rbST treatment and correlated with the results obtained from the reference method. These data clearly demonstrate the potential of the portable protein microarray concept towards simultaneous detection of multiple biomarkers. We envisage broad application of this ‘protein microarray on a smartphone’-concept for on-site testing, e.g., in food safety, environment and health monitoring. PMID:26308444

This research used surveys of the public and dairy farmers in the United States to assess perceptions and attitudes related to dairy cattle welfare. Sixty-three percent of public respondents indicated that they were concerned about dairy cattle welfare. Most public respondents agreed that animal welfare was more important than low milk prices but that the average American did not necessarily agree. Most public respondents had not viewed media stories related to dairy cattle welfare. Respondents who had viewed these stories did so on television or Internet. The United States Department of Agriculture (USDA) was viewed as the most accurate source of information related to dairy cattle welfare, followed by the Humane Society of the United States (HSUS) and the American Veterinary Medicine Association (AVMA). Both public and dairy farmer respondents viewed farmers as having the most influence on dairy cattle welfare. However, there was a general pattern of public respondents indicating that groups including USDA, HSUS, and AVMA had a relatively larger influence on dairy cattle welfare than did farmer respondents. In contrast, dairy farmers indicated that individual actors-farmers, veterinarians, consumers-had more influence than the public indicated. When asked about production practices, most public respondents indicated that they would vote for a ban on antibiotic use outside of disease treatment or for the mandated use of pain control in castration. However, a minority indicated they would vote to ban the use of recombinant bovine somatotropin (rbST) or to pay a premium for milk produced without rbST. With respect to explaining public support for the production practice bans and limits, respondents were more likely to vote for the restrictions if they were older, female, had higher income, or had viewed animal welfare stories in the media. A survey of management practices was conducted to investigate potential associations with groupings of herds formed by cluster analysis (CA) of Dairy Herd Improvement (DHI) data of 557 Upper Midwest herds of 200 cows or greater. Differences in herd management practices were identified between the groups, despite underlying similarities; for example, freestall housing and milking in a parlor. Group 6 comprised larger herds with a high proportion of primiparous cows and most frequently utilized practices promoting increased production [e.g., 84.4% used recombinant bovine somatotropin (rbST)], decreased lameness (e.g., 96.9% used routine hoof trimming for cows), and improved efficiency in reproduction [e.g., 93.8% synchronized the first breeding in cows (SYNCH)] and labor (e.g., mean ± SD, 67 ± 19 cows per 50-h per week full-time equivalent worker). Group 1 had the best mean DHI performances and followed most closely group 6 for the rate of adoption of intensive management practices while tending to outperform group 6 despite a generally smaller mean herd size (e.g., 42.3 ± 3.6 kg vs. 39.9 ± 3.6 kg of energy-corrected milk production; 608 ± 352 cows vs. 1,716 ± 1,405 cows). Group 2 were smaller herds with relatively high levels of performance that used less intensive management (e.g., 100% milked twice daily) and less technology (33.3 vs. 73.0% of group 1 used rbST). Group 4 were smaller but poorer-performing herds with low turnover and least frequently used intensive management practices (e.g., 39.1% SYNCH; 30.4% allowed mature, high-producing cows access to pasture). Group 5 used modern technologies and practices associated with improved production, yet had the least desirable mean DHI performance of all 6 groups. This group had the lowest proportion of deep loose-bedded stalls (only 52.2% used sand bedding) and the highest proportion (34.8%) of herds not using routine hoof trimming. The survey of group 3 herds did not reveal strong trends in management. The

We are developing a computer-aided diagnosis (CAD) system to classify malignant and benign lung nodules found on CT scans. A fully automated system was designed to segment the nodule from its surrounding structured background in a local volume of interest (VOI) and to extract image features for classification. Image segmentation was performed with a three-dimensional (3D) active contour (AC) method. A data set of 96 lung nodules (44 malignant, 52 benign) from 58 patients was used in this study. The 3D AC model is based on two-dimensional AC with the addition of three new energy components to take advantage of 3D information: (1) 3D gradient, which guides the active contour to seek the object surface, (2) 3D curvature, which imposes a smoothness constraint in the z direction, and (3) mask energy, which penalizes contours that grow beyond the pleura or thoracic wall. The search for the best energy weights in the 3D AC model was guided by a simplex optimization method. Morphological and gray-level features were extracted from the segmented nodule. The rubber band straightening transform (RBST) was applied to the shell of voxels surrounding the nodule. Texture features based on run-length statistics were extracted from the RBST image. A linear discriminant analysis classifier with stepwise feature selection was designed using a second simplex optimization to select the most effective features. Leave-one-case-out resampling was used to train and test the CAD system. The system achieved a test area under the receiver operating characteristic curve (Az) of 0.83±0.04. Our preliminary results indicate that use of the 3D AC model and the 3D texture features surrounding the nodule is a promising approach to the segmentation and classification of lung nodules with CAD. The segmentation performance of the 3D AC model trained with our data set was evaluated with 23 nodules available in the Lung Image Database Consortium (LIDC). The lung nodule volumes segmented by the 3D AC

If plotted on mass vs time diagrams, geologic entities (for example, continental and oceanic crust, sediments, and mineral resources) display an exponential (power law) relationship, with entity per unit time increasing toward the present. This relationship is consistent with the concept of recycling and can be simulated mathematically. The approach is based on the plate tectonic theory and considers area-age or mass-age distributions of crystalline basement and sediments for major global tectonic realms. Each tectonic realm is characterized by a specific lifespan, which is an inverse function of its recycling rate. The estimated average half-area of half-mass ages are given. The corresponding parameters for continental crust are 690 Ma for K/Ar, and approximately 1200 Ma for Rb/St and U-Th/Pb dating pairs. Tectonic diversity preserved in the geologic record is therefore a function of time, with oceanic tectonic realms, because of their rapid recycling, underrepresented in the rocks older than approximately 300 Ma. The Sm/Nd isotopic systematic of sediments suggest that, for a near steady-state post-Archean sedimentary mass, recycling is approximately 90 + or - 5 percent cannibalistic. This yields an estimated upper limit on crust-mantle exchange via sediment subduction of approximately 1.1 + or - 0.5 x 10 g a(sup -1) considerably less than demanded by isotopic constraints. The discrepancy may indicate the existence of additional loci, such as orogenic belts, for significant crust-mantle interaction.

Village dwellers in Central Ukraine have access to various types of therapy that comprise diverse medical landscapes. Patients' movements within these landscapes are possible thanks to each person's web of relations. Medical landscapes are not fixed, but vary and dynamically change for each person, depending on their fluid and interchanging, hierarchical webs of mutual relations with other people, personal bodies, institutions, discourses, political powers, other non-human organisms, or objects such as medicines. This paper was inspired by the medicoscape concept (Hörbst and Krause 2004 ) as well as Ingold's idea of meshwork analyses of relations between various actors: in this case, patients, healers, a weak state, official healthcare providers, pharmacists and medicinal plants, in the context of patients' therapeutic choices. Self-medication based on herbal remedies is a very important feature of people's medical landscapes in Central Ukraine and usually the first therapy choice for most interlocutors. That is why this paper is focused on the presentation of the means through which people acquire knowledge about medicinal plants, and the ways they interact with plants and plants interact with them. In this way, showing the complexity of villagers' webs of relations is possible. The analysis is based on ethnographic research conducted between 2009 and 2013 in the Vinnytsia region (Central Ukraine). PMID:27362484

The synergism of excellent properties of carbon nanotubes and gold nanoparticles is used in this work for bio-sensing of recombinant bovine growth hormones (rbST) by making Multi Wall Carbon Nanotubes (MWCNT) locally optically responsive by augmenting it optical properties through Localized Surface Plasmon Resonance (LSPR). To this purpose, locally gold nano particles decorated gold-MWCNT composite was synthesized from a suspension of MWCNT bundles and hydrogen chloroauric acid in an aqueous solution, activated ultrasonically and, then, drop-casted on a glass substrate. The slow drying of the drop produces a “coffee ring” pattern that is found to contain gold-MWCNT nanocomposites, accumulated mostly along the perimeter of the ring. The reaction is studied also at low-temperature, in the vacuum chamber of the Scanning Electron Microscope and is accounted for by the local melting processes that facilitate the contact between the bundle of tubes and the gold ions. Biosensing applications of the gold-MWCNT nanocomposite using their LSPR properties are demonstrated for the plasmonic detection of traces of bovine growth hormone. The sensitivity of the hybrid platform which is found to be 1 ng/ml is much better than that measuring with gold nanoparticles alone which is only 25 ng/ml.

A double-blind, randomized clinical trial was conducted in 5 commercial dairy herds in southern Ontario with 1,362 cows enrolled to evaluate the effect of prepartum administration of recombinant bovine somatotropin (rbST) on health and performance. Cows were randomly assigned to receive either 325 mg of sometribove zinc suspension (n=680) or a placebo injection (n=682; control) subcutaneously every 14 d until calving. Treatments started 28 to 22 d before expected calving, with a maximum of 3 treatments per cow. Serum samples taken at the time of enrollment, 1 wk before calving, and weekly for 3 wk after calving were analyzed for nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), glucose, aspartate aminotransferase, calcium, and haptoglobin. Diseases were recorded by farm staff. Incidences of clinical ketosis, clinical mastitis, displaced abomasum, metritis, retained placenta, milk fever, and lameness were similar between groups. Body condition score was lower for treated than for control cows at 3 wk after calving (3.13 and 3.17, respectively). Serum NEFA tended to be higher for treated than for control cows by 0.01 mmol/L. Overall BHBA was not different between groups, but BHBA for treated cows was higher in wk 1 after calving (750 and 698 μmol/L, respectively) and tended to be higher in wk 2 after calving (779 and 735 μmol/L, respectively). Incidence of hyperketonemia was similar between groups. Treated cows had higher serum glucose compared with control cows (2.8 and 2.7 mmol/L, respectively). We detected no differences in serum aspartate aminotransferase, calcium, or haptoglobin between groups. Milk yield was recorded daily for each cow for 63 d, and did not differ between groups (37.1 ± 0.5 kg and 36.7 ± 0.5 kg, respectively) but we detected a tendency for treated cows to produce 0.8 kg/d more milk than control cows in wk 1 after calving. We observed no difference between groups in the time from calving to first insemination or the probability of

Purpose: In digital breast tomosynthesis (DBT), quasi-three-dimensional (3D) structural information is reconstructed from a small number of 2D projection view (PV) mammograms acquired over a limited angular range. The authors developed preliminary computer-aided diagnosis (CADx) methods for classification of malignant and benign masses and compared the effectiveness of analyzing lesion characteristics in the reconstructed DBT slices and in the PVs. Methods: A data set of MLO view DBT of 99 patients containing 107 masses (56 malignant and 51 benign) was collected at the Massachusetts General Hospital with IRB approval. The DBTs were obtained with a GE prototype system which acquired 11 PVs over a 50 deg. arc. The authors reconstructed the DBTs at 1 mm slice interval using a simultaneous algebraic reconstruction technique. The region of interest (ROI) containing the mass was marked by a radiologist in the DBT volume and the corresponding ROIs on the PVs were derived based on the imaging geometry. The subsequent processes were fully automated. For classification of masses using the DBT-slice approach, the mass on each slice was segmented by an active contour model initialized with adaptive k-means clustering. A spiculation likelihood map was generated by analysis of the gradient directions around the mass margin and spiculation features were extracted from the map. The rubber band straightening transform (RBST) was applied to a band of pixels around the segmented mass boundary. The RBST image was enhanced by Sobel filtering in the horizontal and vertical directions, from which run-length statistics texture features were extracted. Morphological features including those from the normalized radial length were designed to describe the mass shape. A feature space composed of the spiculation features, texture features, and morphological features extracted from the central slice alone and seven feature spaces obtained by averaging the corresponding features from three to 19

Mycobacterium bovis BCG has the potential to be an effective live vector for multivalent vaccines. However, most mycobacterial cloning vectors rely on antibiotic resistance genes as selectable markers, which would be undesirable in any practical vaccine. Here we report the use of auxotrophic complementation as a selectable marker that would be suitable for use in a recombinant vaccine. A BCG auxotrophic for the amino acid leucine was constructed by knocking out the leuD gene by unmarked homologous recombination. Expression of leuD on a plasmid not only allowed complementation, but also acted as a selectable marker. Removal of the kanamycin resistance gene, which remained necessary for plasmid manipulations in Escherichia coli, was accomplished by two different methods: restriction enzyme digestion followed by re-ligation before BCG transformation, or by Cre-loxP in vitro recombination mediated by the bacteriophage P1 Cre Recombinase. Stability of the plasmid was evaluated during in vitro and in vivo growth of the recombinant BCG in comparison to selection by antibiotic resistance. The new system was highly stable even during in vivo growth, as the selective pressure is maintained, whereas the conventional vector was unstable in the absence of selective pressure. This new system will now allow the construction of potential recombinante vaccine strains using stable multicopy plasmid vectors without the inclusion of antibiotic resistance markers. PMID:17888740

Fourteen Holstein-Friesian heifers between 15 and 22 months of age with normal reproductive tracts were used in an experimental set up to investigate the effect of a long term rBST-treatment on the follicular population prior to transvaginal ovum pick-up (OPU). The estrous cycle of the animals was synchronized by means of a double injection of 2 ml cloprostenol 11 days apart. The heifers were divided in 2 equal groups (n = 7) in which the animals had the same average body weight, one group receiving a weekly subcutaneous injection of 640 mg recombinantly derived bovine somatotropin (rBST) on Mondays (rBST-treatment group) and a control group, being injected with 10 ml of saline. Heifers in both groups were submitted to OPU twice a week on Mondays and Thursdays using a 5 Mhz transducer and a disposable, 55 mm long, 20-g short bevelled needle at a vacuum pressure corresponding to approximately 13 ml water/min. The experimental period lasted for 10 weeks (April to June), each animal receiving a total of 10 injections and being submitted to OPU for 20 times. Oocytes were subsequently matured and cultured in a separate drop per cow following conventional IVF procedures. A blood sample was taken on heparin immediately after each OPU session, for determination of blood progesterone concentrations to assess the influence of treatment and OPU procedure on the cow's estrous cycle. Although results show a significant increase in the total number of follicles and medium sized follicles in the rBST-treated group, no statistically significant different number of retrieved oocytes between the rBST-treated and nontreated group could be detected. The average number of retrieved oocytes per session per cow was comparable, being 6.4 for the treated and 6.0 for the control group. Additionally, the average number of blastocysts per cow per session did not differ significantly between groups, being 1.41 in the rBST-treated group and 1.53 in the control group. The number of cultured

The aim of this study was to determine, using multiple logistic models, factors affecting pregnancy per AI (P/AI) following fixed-time artificial insemination (FTAI) in repeat breeders cows (RBC) treated with rbST throughout lactation. 498 RBC received a CIDR device and 100μg of GnRH on day 0. CIDR removal and PGF(2α) (25mg) treatment were done concurrently on day 7. Estradiol benzoate (EB, 1mg) was injected on day 8 and GnRH on day 9; cows were inseminated 16-20h later. Cows with an average milk protein <3% were 43% more likely (P<0.05) to become pregnant at FTAI than cows with milk protein ≥3%. Cows with <6 services had significantly increased chances of becoming pregnant than cows with ≥6 services at FTAI (P/AI 36 vs. 27%; P<0.05). CIDR-treated cows with less than three lactations were 1.7 times more likely (P/AI 35 vs. 21%; P<0.05) to become pregnant than cows in third or greater lactation. Cows with peak milk yields lower than 55kg were 1.5 times more likely to get pregnant than cows with peak milk yields greater than 55kg (P/AI 37 vs. 28%; P<0.05). P/AI was lower (30 vs. 35%; P<0.01) for cows with dry periods <62 days than cows with dry periods ≥62 days. Cows subjected to FTAI with a temperature-humidity index (THI) <76 were 45% more likely (P<0.05) to become pregnant than cows inseminated at a THI≥76. It was concluded that an acceptable proportion (32%) of RBC can become pregnant with the protocol used in the present study. Also, subfertility in CIDR-treated cows was associated with high peak yields, high milk protein, increased service, increased lactation, high THI at AI and short dry periods. PMID:22917878

Background Chronic rhinosinusitis with nasal polyposis (CRSwNP) in Western countries is characterized by eosinophilia, IgE production, and TH2 cytokine expression. Type 2 innate lymphoid cells from polyps produce IL-5 and IL-13 in response to IL-25 and IL-33, although the relevance of this axis to local mucosal T-cell responses is unknown. Objective We sought to investigate the role of the IL-25/IL-33 axis in local mucosal T-cell responses in patients with CRSwNP. Methods Polyp tissue and blood were obtained from patients undergoing nasal polypectomy. Control nasal biopsy specimens and blood were obtained from healthy volunteers. Tissue was cultured in a short-term explant model. T-cell surface phenotype/intracellular cytokines were assessed by means of flow cytometry. T-cell receptor variable β-chain analysis was performed with the immunoSEQ assay. Microarrays were performed for gene expression analysis. Results IL-25 receptor (IL-17RB)–expressing TH2 effector cells were identified in nasal polyp tissue but not the healthy nasal mucosa or periphery. IL-17RB+CD4+ polyp–derived TH2 cells coexpressed ST2 (IL-33 receptor) and responded to IL-25 and IL-33 with enhanced IL-5 and IL-13 production. Within IL-17RB+CD4+ T cells, several identical T-cell receptor variable β-chain complementarity-determining region 3 sequences were identified in different subjects, suggesting clonal expansion driven by a common antigen. Abundant IL-17–producing T cells were observed in both healthy nasal mucosal and polyp populations, with TH17-related genes the most overexpressed compared with peripheral blood T cells. Conclusion IL-25 and IL-33 can interact locally with IL-17RB+ST2+ polyp T cells to augment TH2 responses in patients with CRSwNP. A local TH17 response might be important in healthy nasal mucosal immune homeostasis. PMID:26684290

Thymidine glycol residues in DNA are biologically active oxidative molecular damage sites caused by ionizing radiation and other factors. One or two thymidine glycol residues were incorporated in 19- to 31-mer DNA fragments during automatic oligonucleotide synthesis. These oligonucleotide models were used to estimate the effect of oxidized thymidines on the thermodynamic, substrate and interfacial acoustic properties of DNA. UV-monitoring melting data revealed that modified residues in place of thymidines destabilize the DNA double helix by 8-22 degrees C, depending on the number of lesions, the length of oligonucleotide duplexes and their GC-content. The diminished hybridizing capacity of modified oligonucleotides is presumably due to the loss of aromaticity and elevated hydrophilicity of thymine glycol in comparison to the thymine base. According to circular dichroism (CD) data, the modified DNA duplexes retain B-form geometry, and the thymidine glycol residue introduces only local perturbations limited to the lesion site. The rate of DNA hydrolysis by restriction endonucleases R.MvaI, R.Bst2UI, R.MspR9I and R.Bme1390I is significantly decreased as the thymidine glycol is located in the central position of the double-stranded recognition sequences 5'-CC / WGG-3' (W = A, T) or 5'-CC / NGG-3' (N = A, T, G, C) adjacent to the cleavage site. On the other hand, the catalytic properties of enzymes R.Psp6I and R.BstSCI recognizing the similar sequence are not changed dramatically, since their cleavage site is separated from the point of modification by several base-pairs. Data obtained by gel-electrophoretic analysis of radioactive DNA substrates were confirmed by direct spectrophotometric assay developed by the authors. The effect of thymidine glycol was also observed on DNA hybridization at the surface of a thickness-shear mode acoustic wave device. A 1.9-fold decrease in the rate of duplex formation was noted for oligonucleotides carrying one or two thymidine glycol

A trend in food labeling is to make claims related to agricultural management, and this is occurring with dairy labels. A survey study was conducted to compare retail milk for quality (antibiotics and bacterial counts), nutritional value (fat, protein, and solids-not-fat), and hormonal composition (somatotropin, insulin-like growth factor-1 [IGF-1], estradiol, and progesterone) as affected by three label claims related to dairy-cow management: conventional, recombinant bovine somatotropin (rbST)-free (processor-certified not from cows supplemented with rbST), or organic (follows US Department of Agriculture organic practices). Retail milk samples (n=334) from 48 states were collected. Based on a statistical analysis that reflected the sampling schema and distributions appropriate to the various response variables, minor differences were observed for conventional, rbST-free, and organic milk labels. Conventionally labeled milk had the lowest (P<0.05) bacterial counts compared to either milk labeled rbST-free or organic; however, these differences were not biologically meaningful. In addition, conventionally labeled milk had significantly less (P<0.05) estradiol and progesterone than organic milk (4.97 vs 6.40 pg/mL and 12.0 vs 13.9 ng/mL, respectively). Milk labeled rbST-free had similar concentrations of progesterone vs conventional milk and similar concentrations of estradiol vs organic milk. Concentrations of IGF-1 in milk were similar between conventional milk and milk labeled rbST-free. Organic milk had less (P<0.05) IGF-1 than either conventional or rbST-free milk (2.73 ng/mL vs 3.12 and 3.04 ng/mL, respectively). The macronutrient profiles of the different milks were similar, except for a slight increase in protein in organic milk (about 0.1% greater for organic compared to other milks). Label claims were not related to any meaningful differences in the milk compositional variables measured. It is important for food and nutrition professionals to know that

PMID:25328251

The title points to the joint effort of all societies involved to rethink their approaches and to signal the need to move on from silo thinking by exploring the complexity of health together although from different perspectives. Complex systems often involve heterogeneous objects and multifaceted interactions. The health domain is recognised to be particularly complex; hence, we need to identify models that are able to integrate all the various aspects. We need to find new ways for collaboration of different scientific disciplines as well as for establishing comprehensive methodological approaches. The HEC2016 conference is a step forward. The current volume supports this effort by documenting the results of this event. The HEC2016 received 833 contributions from 47 countries.

The ferroelectric response of bismuth titanate Bi4Ti3O12 (BIT) thin film is studied through a Monte Carlo simulation of hysteresis loops. The ferroelectric system is described by using a Diffour Hamiltonian with three terms: the electric field applied in the z direction, the nearest dipole-dipole interaction in the transversal (x-y) direction, and the nearest dipole-dipole interaction in the direction perpendicular to the thin film (the z axis). In the sample construction, we take into consideration the dipole orientations of the monoclinic and orthorhombic structures that can appear in BIT at low temperature in the ferroelectric state. The effects of temperature, stress, and the concentration of pinned dipole defects are assessed by using the hysteresis loops. The results indicate the changes in the hysteresis area with temperature and stress, and the asymmetric hysteresis loops exhibit evidence of the imprint failure mechanism with the emergence of pinned dipolar defects. The simulated shift in the hysteresis loops conforms to the experimental ferroelectric response.

Andalusia region (Southern Spain) is the largest olive oil producer in the world with 40% of the total production (1.1millions tons; FAOSTAT, 2013). Predicting flowering dates of olive under future climate conditions has a critical importance as flowering is a critical stage for olive as heat and water stresses during this period have a significant impact on final olive oil yield. The aim of this study was to evaluate the olive flowering dates under future climate conditions. Climate data for the future period was obtained from the ENSEMBLES European Project (http://www.ensembles-eu.org/) with a bias correction in temperature and precipitation with regard with the SPAIN02 dataset (Herrera et al., 2012) (ENS-SP). Flowering of ten different olive cultivars were evaluated under current and forced (greenhouse) climate conditions, applying the bias in temperatures comparing the current period (1981-2010) with the future period (2071-2100) to the current conditions in Cordoba (Andalusia, Spain). These observations allowed obtaining a flowering approach which was applied to the whole Andalusia region. The results showed an average advance in flowering dates about 16 days at the end of the 21st century. With these results different areas within Andalusia region were classified based on the vulnerability caused by the lack of chilling hours accumulation (coastal areas) or by high temperatures during flowering (north and northeast regions).
