    The purpose of an Environmental Assessment (EA) is to provide the DOE with sufficient evidence and analysis to determine whether to prepare an Environmental Impact Statement (EIS) or a Finding of No Significant Impact (FONSI). Additional considerations (such as costs, timing, or non-environmental legal issues) that influence DOE decisions are not analyzed in this EA. As part of its initiative to fulfill its responsibilities to provide support for the County of Los Alamos (the County), in northern New Mexico, the US Department of Energy (DOE) proposes to transfer ownership of the undeveloped, so called, DP Road property to the County. Transfer of this tract would permanently reduce the size of Los Alamos National Laboratory (LANL) by approximately 0.1%. Approximately 12 hectares (28 acres) would be changed from an undeveloped to a developed status. This would result in an equivalent loss of wildlife habitat. A hypothetical accident was analyzed that evaluated potential radiological dose to the public at the DP Road tract from LANL operations. The dose to the hypothetical worker population of 450 new employees could result in an increase of approximately three latent cancer fatalities in the population. The DOE finds that there would be no significant impact from proceeding with the transfer of the 28-acre tract for development and use as a business park or for light industrial purposes.

    The purpose of an Environmental Assessment (EA) is to provide the DOE with sufficient evidence and analysis to determine whether to prepare an Environmental Impact Statement (EIS) or a Finding of No Significant Impact (FONSI). In this case, the DOE decision to be made is whether or not to transfer its ownership of the subject land tract by deed. This type of action does not in and of itself result in environmental effects; however, the DOE has considered the planned use of the land and the ensuing potential environmental effects subsequent to the transfer of ownership in its decision making process. Therefore, DOE is evaluating its decision in light of the contemplated land use as outlined by County officials that could only occur if the DOE decides to transfer its ownership of the subject land tract. The objectives of this EA are (1) to describe the baseline environmental conditions at the tract location involved in the Proposed Action, (2) to analyze potential generic effects to the baseline environment from land development activities and future occupants' operations occurring at the tract location, and (3) to identify and characterize cumulative effects of future anticipated uses of the tract involved in the Proposed Action together with those of surrounding properties. In addition, the EA provides DOE with environmental information that would be used in preparing appropriate notices which would serve to preserve the integrity of the human environment and natural ecosystems should the DOE decide to proceed with the Proposed Action.

    One of the most common causes of illnesses in humans is from respiratory tract infections caused by bacterial, viral or fungal pathogens. Inhaled anti-infective drugs are crucial for the prophylaxis and treatment of respiratory tract infections. The benefit of anti-infective drug delivery via inhalation is that it affords delivery of sufficient therapeutic dosages directly to the primary site of infection, while minimizing the risks of systemic toxicity or avoiding potential suboptimal pharmacokinetics/pharmacodynamics associated with systemic drug exposure. This review provides an up-to-date treatise of approved and novel developmental inhaled anti-infective agents, with particular attention to effective strategies for their use, pulmonary pharmacokinetic properties and safety.     Galectins are an evolutionarily ancient and widely expressed family of lectins that have unique glycan-binding characteristics. They are pleiotropic regulators of key biological processes, such as cell growth, proliferation, differentiation, apoptosis, signal transduction, and pre-mRNA splicing, as well as homo- and heterotypic cell-cell and cell-extracellular matrix interactions. Galectins are also pivotal in immune responses since they regulate host-pathogen interactions, innate and adaptive immune responses, acute and chronic inflammation, and immune tolerance. Some galectins are also central to the regulation of angiogenesis, cell migration and invasion. Expression and functional data provide convincing evidence that, due to these functions, galectins play key roles in shared and unique pathways of normal embryonic and placental development as well as oncodevelopmental processes in tumorigenesis. Therefore, galectins may sometimes act as double-edged swords since they have beneficial but also harmful effects for the organism. Recent advances facilitate the use of galectins as biomarkers in obstetrical syndromes and in various malignancies, and their therapeutic applications are also under investigation. This review provides a general overview of galectins and a focused review of this lectin subfamily in the context of inflammation, infection and tumors of the female reproductive tract as well as in normal pregnancies and those complicated by the great obstetrical syndromes.   5. Biological assessment for the transfer of the DP land tract

    The Department of Energy (DOE) is proposing to transfer to the County of Los Alamos up to 10-ha (25-ac) of federal land located in Technical Area-21 to be developed for commercial uses. Previous studies for the proposed land transfer area indicate that potential habitat for four threatened, endangered, and sensitive species occurs in or adjacent to the proposed land transfer area. These include the northern goshawk (federal species of concern), Mexican spotted owl (federal threatened), the spotted bat (federal species of concern, state threatened), die peregrine falcon (federal endangered, state endangered), and the. In order to determine the possible influences of the land transfer on these organisms, information from species-specific surveys was collected. These surveys were used to confirm the presence of these species or to infer their absence in or near the project area. It was concluded that none of die above mentioned species occur in the project area. Stretches of the stream channel within Los Alamos Canyon have been identified as palustrine and riverine, temporarily flooded wetlands. The proposed land transfer should not affect these wetlands.

    The dystrophin gene consists of 79 exons and encodes tissue-specific isoforms. Mutations in the dystrophin gene cause Duchenne muscular dystrophy, of which a substantial proportion of cases are complicated by non-progressive mental retardation. Abnormalities of Dp71, an isoform transcribed from a promoter in intron 62, are a suspected cause of mental retardation. However, the roles of Dp71 in human brain have not been fully elucidated. Here, we characterized dystrophin in human HEK293 cells with the neuronal lineage. Reverse transcription-PCR amplification of the full-length dystrophin transcript revealed the absence of fragments covering the 5' part of the dystrophin cDNA. In contrast, fragments covering exons 64-79 were present. The Dp71 promoter-specific exon G1 was shown spliced to exon 63. We demonstrated that the Dp71 transcript comprised two subisoforms: one lacking exon 78 (Dp71b) and the other lacking both exons 71 and 78 (Dp71ab). Western blotting of cell lysates using an antibody against the dystrophin C-terminal region revealed two bands, corresponding to Dp71b and Dp71ab. Immunohistochemical examination with the dystrophin antibody revealed scattered punctate signals in the cytoplasm and the nucleus. Western blotting revealed one band corresponding to Dp71b in the cytoplasm and two bands corresponding to Dp71b and Dp71ab in the nucleus, with Dp71b being predominant. These results indicated that Dp71ab is a nucleus-specific subisoform. We concluded that Dp71, comprising Dp71b and Dp71ab, was expressed exclusively in HEK293 cells and that Dp71ab was specifically localized to the nucleus. Our findings suggest that Dp71ab in the nucleus contributes to the diverse functions of HEK293 cells. PMID:27109495

  8. Rescattering effects in B{sub u,d,s}{yields}DP,DP decays

    We study quasielastic rescattering effects in B{sub u,d,s}{yields}DP,DP decays, where P is a light pseudoscalar. The updated measurements of B{sub u,d}{yields}DP decays are used to extract the effective Wilson coefficients a{sub 1}{sup eff}{approx_equal}0.90 and a{sub 2}{sup eff}{approx_equal}0.23, three strong phases {delta}{approx_equal}56 deg., {theta}{approx_equal}18 deg., and {sigma}{approx_equal}-104 deg., and the mixing angle {tau}{approx_equal}7 deg. This information is used to predict rates of 19 B{sub s}{yields}DP and B{sub u,d,s}{yields}DP decay modes, including modes of interests in the {gamma}/{phi}{sub 3} program. Many decay rates are found to be enhanced. In particular, the B{sub s}{yields}D{sup 0}K{sup 0} rate is predicted to be 7x10{sup -4}, which could be measured soon. The rescattering effects on the corresponding B{sub u,d,s}{yields}DP,DP amplitude ratios r{sub B} and r{sub B{sub s}}, and the relative strong phases {delta}{sub B} and {delta}{sub B{sub s}} are studied. Although the decay rates are enhanced in most cases, r{sub B,B{sub s}} values are similar to factorization expectation.

    Understanding the impact of road investments on deforestation is part of a complete evaluation of the expansion of infrastructure for development. We find evidence of spatial spillovers from roads in the Brazilian Amazon: deforestation rises in the census tracts that lack roads but are in the same county as and within 100 km of a tract with a new paved or unpaved road. At greater distances from the new roads the evidence is mixed, including negative coefficients of inconsistent significance between 100 and 300 km, and if anything, higher neighbor deforestation at distances over 300 km.

  15. Effects of Dilute Acid Pretreatment on Cellulose DP and the Relationship Between DP Reduction and Cellulose Digestibility

    The degree of polymerization(DP) of cellulose is considered to be one of the most important properties affecting the enzymatic hydrolysis of cellulose. Various pure cellulosic and biomass materials have been used in a study of the effect of dilute acid treatment on cellulose DP. A substantial reduction in DP was found for all pure cellulosic materials studied even at conditions that would be considered relatively mild for pretreatment. The effect of dilute acid pretreatment on cellulose DP in biomass samples was also investigated. Corn stover pretreated with dilute acid under the most optimal conditions contained cellulose with a DPw in the range of 1600{approx}3500, which is much higher than the level-off DP(DPw 150{approx}300) obtained with pure celluloses. The effect of DP reduction on the saccharification of celluloses was also studied. From this study it does not appear that cellulose DP is a main factor affecting cellulose saccharification.

  16. Noncommutative Tachyon Kinks as D(p-1)-branes from Unstable Dp-brane

    We study noncommutative (NC) field theory of a real NC tachyon and NC U(1) gauge field, describing the dynamics of an unstable Dp-brane. For every given set of diagonal component of open string metric G 0 , NC parameter θ0 , and interpolating electric field hat E, we find all possible static NC kinks as exact solutions, in spite of complicated NC terms, which are classified by an array of NC kink-antikink and topological NC kinks. By computing their tensions and charges, those configurations are identified as an array of D0bar D0 and single stable D0 from the unstable D1, respectively. When the interpolating electric field has critical value as G 0 2 = hat E2 , the obtained topological kink becomes a BPS object with nonzero thickness and is identified as BPS D0 in the fluid of fundamental strings. Particularly in the scaling limit of infinite θ0 and vanishing G 0 and hat E, while keeping G 0θ0 = hat Eθ0 = 1, finiteness of the tension of NC kink corresponds to tensionless kink in ordinary effective field theory. An extension to stable D(p-1) from unstable Dp is straightforward for pure electric cases with parallel NC parameter and interpolating two-form field.

    Up to one-third of community participants report being perpetrators of road rage, indicating that various forms of road rage are relatively commonplace. However, only two percent or less of incidents culminate in serious damage to persons or vehicles. The most common offenders appear to be young and male. A number of factors may contribute to road rage, including environmental factors (e.g., greater number of miles driven per day, traffic density), nonspecific psychological factors (e.g., displaced aggression, attribution of blame to others), and bona fide Axis I and II disorders. The most common Axis I disorders appear to be related to alcohol and substance misuse, whereas possible Axis II disorders include borderline and antisocial personality disorders. Being aware of these contributory factors to road rage may improve general clinical awareness of the nature and treatment of perpetrators. PMID:20805914

We previously reported the existence of a novel short isoform of Dp, Dp40, in adult mouse brain. However, the exact biochemical expression profile and cytological distribution of the Dp40 protein remain unknown. In this study, we generated a polyclonal antibody against the NH{sub 2}-terminal region of the Dp40 and identified the expression profile of Dp40 in the mouse brain. Through an analysis using embryonic and postnatal mouse cerebrums, we found that Dp40 emerged from the early neonatal stages until adulthood, whereas Dp71, an another Dp short isoform, was highly detected in both prenatal and postnatal cerebrums. Intriguingly, relative expressions of Dp40 and Dp71 were prominent in cultured dissociated neurons and non-neuronal cells derived from mouse hippocampus, respectively. Furthermore, the immunocytological distribution of Dp40 was analyzed in dissociated cultured neurons, revealing that Dp40 is detected in the soma and its dendrites, but not in the axon. It is worthy to note that Dp40 is localized along the subplasmalemmal region of the dendritic shafts, as well as at excitatory postsynaptic sites. Thus, Dp40 was identified as a neuron-type Dp possibly involving dendritic and synaptic functions.

    A system that detects damage on or below the surface of a paved structure or pavement is provided. A distributed road assessment system includes road assessment pods and a road assessment server. Each road assessment pod includes a ground-penetrating radar antenna array and a detection system that detects road damage from the return signals as the vehicle on which the pod is mounted travels down a road. Each road assessment pod transmits to the road assessment server occurrence information describing each occurrence of road damage that is newly detected on a current scan of a road. The road assessment server maintains a road damage database of occurrence information describing the previously detected occurrences of road damage. After the road assessment server receives occurrence information for newly detected occurrences of road damage for a portion of a road, the road assessment server determines which newly detected occurrences correspond to which previously detected occurrences of road damage.

We previously reported the existence of a novel short isoform of Dp, Dp40, in adult mouse brain. However, the exact biochemical expression profile and cytological distribution of the Dp40 protein remain unknown. In this study, we generated a polyclonal antibody against the NH2-terminal region of the Dp40 and identified the expression profile of Dp40 in the mouse brain. Through an analysis using embryonic and postnatal mouse cerebrums, we found that Dp40 emerged from the early neonatal stages until adulthood, whereas Dp71, an another Dp short isoform, was highly detected in both prenatal and postnatal cerebrums. Intriguingly, relative expressions of Dp40 and Dp71 were prominent in cultured dissociated neurons and non-neuronal cells derived from mouse hippocampus, respectively. Furthermore, the immunocytological distribution of Dp40 was analyzed in dissociated cultured neurons, revealing that Dp40 is detected in the soma and its dendrites, but not in the axon.   11. Extracellular Polyhydroxyalkanoate Depolymerase by Acidovorax sp. DP5

    Bacteria capable of degrading polyhydroxyalkanoates (PHA) by secreting extracellular depolymerase enzymes were isolated from water and soil samples collected from various environments in Malaysia. A total of 8 potential degraders exhibited clear zones on poly(3-hydroxybutyrate) [P(3HB)] based agar, indicating the presence of extracellular PHA depolymerase. Among the isolates, DP5 exhibited the largest clearing zone with a degradation index of 6.0. The highest degradation activity of P(3HB) was also observed with depolymerase enzyme of DP5 in mineral salt medium containing P(3HB). Based on biochemical characterization and 16S rRNA cloning and sequencing, isolate DP5 was found to belong to the genus Acidovorax and subsequently named as Acidovorax sp. DP5. The highest extracellular depolymerase enzyme activity was achieved when 0.25% (w/v) of P(3HB) and 1 g/L of urea were used as carbon and nitrogen source, respectively, in the culture media. The most suitable assay condition of the depolymerase enzyme in response to pH and temperature was tested. The depolymerase produced by strain Acidovorax sp. DP5 showed high percentage of degradation with P(3HB) films in an alkaline condition with pH 9 and at a temperature of 40°C. PMID:26664741

    The tendency of unpaved road surfaces to develop lateral ripples (``washboard'' or ``corrugated'' road) is annoyingly familiar to drivers on dry gravel roads. Similar ripples are well known on railroad tracks and many other rolling or sliding, load bearing surfaces. Our approach combined laboratory experiments, soft-particle direct numerical simulations and simple nonlinear dynamics models. The experiment consisted of a rotating table 60 cm in radius with a thick layer of sand forming a roadbed around the circumference. A 6 cm radius hard rubber wheel, with a support stationary in the lab frame, rolled on the sand layer. We varied the speed of the table and the details of the suspension of the wheel. The onset of the ripple pattern exhibits a sharp threshold and was strongly subcritical with a large hysteresis as a function of the speed of the table. The ripple pattern appears as small patches of travelling waves which eventually spread to the entire circumference. The ripples move slowly in the driving direction. Interesting secondary dynamics of the saturated ripples were observed. All of these effects are captured qualitatively by a 2D soft particle simulations. The simulations clearly indicate that neither compaction nor particle size segregation are crucial for the appearance of the ripples, and we present a simple model to describe the wavelength and amplitude of the ripples.

  13. Sex Hormones Regulate Tenofovir-Diphosphate in Female Reproductive Tract Cells in Culture

    The conflicting results of recent pre-exposure prophylaxis (PrEP) trials utilizing tenofovir (TFV) to prevent HIV infection in women led us to evaluate the accumulation of intracellular TFV-diphosphate (TFV-DP) in cells from the female reproductive tract (FRT) and whether sex hormones influence the presence of TFV-DP in these cells. Following incubation with TFV, isolated epithelial cells, fibroblasts, CD4+ T cells and CD14+ cells from the FRT as well as blood CD4+ T cells and monocyte-derived macrophages convert TFV to TFV-DP. Unexpectedly, we found that TFV-DP concentrations (fmol/million cells) vary significantly with the cell type analyzed and the site within the FRT. Epithelial cells had 5-fold higher TFV-DP concentrations than fibroblasts; endometrial epithelial cells had higher TFV-DP concentrations than cells from the ectocervix. Epithelial cells had 125-fold higher TFV-DP concentrations than FRT CD4+ T cells, which were comparable to that measured in peripheral blood CD4+ T cells. These findings suggest the existence of a TFV-DP gradient in the FRT where epithelial cells > fibroblasts > CD4+ T cells and macrophages. In other studies, estradiol increased TFV-DP concentrations in endometrial and endocervical/ectocervical epithelial cells, but had no effect on fibroblasts or CD4+ T cells from FRT tissues. In contrast, progesterone alone and in combination with estradiol decreased TFV-DP concentrations in FRT CD4+ T cells. Our results suggest that epithelial cells and fibroblasts are a repository of TFV-DP that is under hormonal control. These cells might act either as a sink to decrease TFV availability to CD4+ T cells and macrophages in the FRT, or upon conversion of TFV-DP to TFV increase TFV availability to HIV-target cells. In summary, these results indicate that intracellular TFV-DP varies with cell type and location in the FRT and demonstrate that estradiol and/or progesterone regulate the intracellular concentrations of TFV-DP in FRT epithelial cells

    The Data Processing software for ESA's Herschel Space Observatory is written in JAVA as a joint effort of ESA and the three instrument teams. The observers as well as the instrument specialists will use a jython environment to reduce the science observations and to analyse the calibration measurements of the instrument. We describe the development of an image display package for Herschel DP (Data Processing) based on JSky, which is being developed as part of the Gemini Observing Tool and the difficulties we encountered in adapting the JSky library to the needs of Herschel DP. We give insights in the adaption to the Herschel DP data structures and the possibility to display both two- and three dimensional arrays and images. We describe the power of using Jython as a frontend and the possibility to use this image display in GUI's written by or for observers.

    In this work we explain the construction of the thermal vacuum for the bosonic string, as well that of the thermal boundary state interpreted as a Dp-brane at finite temperature. In both case we calculate the respective entropy using the entropy operator of the Thermo Field Dynamics theory. We show that the contribution of the thermal string entropy is explicitly present in the Dp-brane entropy. Furthermore, we show that the Thermo Field approach is suitable to introduce temperature in boundary states.

    The objective of this study was to compare the microstructural response of various DP 600 products subjected to low velocity, dynamic impact tests, typically encountered in a car crash. Since the response of steel is sensitive to its microstructure as controlled by the alloying elements, phase content, and processing; various DP 600 products may respond differently to crashes. The microstructure before and after dynamic impact deformation at 5 and 10 mph was characterized with regards to grain size, morphology, and phase content among vendors A, B, and C to evaluate efficiency in absorbing energy mechanisms during a crash simulated by dynamic impact testing in a drop tower.

    In this paper, we investigate the performance of dual polarization orthogonal frequency division multiplexing (DP-OFDM) signal generation when the signal is generated by a DP-IQ optical modulator. The DP-IQ optical modulator is made of four parallel Mach-Zehnder modulators (MZMs) which have nonlinear responses and limited extinction ratios. We analyze the effects of the MZM in the DP-OFDM signal generation by numerical simulation. The operating conditions of the DP-IQ modulator are optimized to have the best performance of the DP-OFDM signal.

  2. Friction Stir Spot Welding of DP780 Carbon Steel

    Friction stir spot welds were made in uncoated and galvannneled DP780 sheets using polycrystalline boron nitride stir tools. The tools were plunged at either a single continuous rate or in two segments consisting of a relatively high rate followed by a slower rate of shorter depth. Welding times ranged from 1-10 s. Increasing tool rotation speed from 800 to 1600 rpm increased strength values. The 2-segment welding procedures also produced higher strength joints. Average lap-shear strengths exceeding 10.3 kN were consistently obtained in 4 s on both the uncoated and the galvannealed DP780. The likelihood of diffusion and mechanical interlocking contributing to bond formation was supported by metallographic examinations. A cost analysis based on spot welding in automobile assembly showed that for friction stir spot welding to be economically competitive with resistance spot welding the cost of stir tools must approach that of resistance spot welding electrode tips.

    Abstract Probabilistic methods have the potential to generate multiple and complex white matter fiber tracts in diffusion tensor imaging (DTI). Here, a method based on dynamic programming (DP) is introduced to reconstruct fibers pathways whose complex anatomical structures cannot be resolved beyond the resolution of standard DTI data. DP is based on optimizing a sequentially additive cost function derived from a Gaussian diffusion model whose covariance is defined by the diffusion tensor. DP is used to determine the optimal path between initial and terminal nodes by efficiently searching over all paths, connecting the nodes, and choosing the path in which the total probability is maximized. An ex vivo high-resolution scan of a macaque hemi-brain is used to demonstrate the advantages and limitations of DP. DP can generate fiber bundles between distant cortical areas (superior longitudinal fasciculi, arcuate fasciculus, uncinate fasciculus, and fronto-occipital fasciculus), neighboring cortical areas (dorsal and ventral banks of the principal sulcus), as well as cortical projections to the hippocampal formation (cingulum bundle), neostriatum (motor cortical projections to the putamen), thalamus (subcortical bundle), and hippocampal formation projections to the mammillary bodies via the fornix. Validation is established either by comparison with in vivo intracellular transport of horseradish peroxidase in another macaque monkey or by comparison with atlases. DP is able to generate known pathways, including crossing and kissing tracts. Thus, DP has the potential to enhance neuroimaging studies of cortical connectivity. PMID:23879573

    Prostaglandin D₂ (PGD₂) acts through two G protein-coupled receptors (GPCRs), the prostanoid DP receptor and CRTH2 also known as DP1 and DP2, respectively. Several previously characterized GPCR antagonists are now classified as inverse agonists and a number of GPCR ligands are known to display pharmacochaperone activity towards a given receptor. Here, we demonstrate that a DP1 specific antagonist, MK-0524 (also known as laropiprant), decreased basal levels of intracellular cAMP produced by DP1, a Gα(s)-coupled receptor, in HEK293 cells. This reduction in cAMP levels was not altered by pertussis toxin treatment, indicating that MK-0524 did not induce coupling of DP1 to Gα(i/o) proteins and that this ligand is a DP1 inverse agonist. Basal ERK1/2 activation by DP1 was not modulated by MK-0524. Interestingly, treatment of HEK293 cells expressing Flag-tagged DP1 with MK-0524 promoted DP1 cell surface expression time-dependently to reach a maximum increase of 50% compared to control after 24 h. In contrast, PGD₂ induced the internalization of 75% of cell surface DP1 after the same time of stimulation. The increase in DP1 cell surface targeting by MK-0524 was inhibited by Brefeldin A, an inhibitor of transport from the endoplasmic reticulum-Golgi to the plasma membrane. Confocal microscopy confirmed that a large population of DP1 remained trapped intracellularly and co-localized with calnexin, an endoplasmic reticulum marker. Redistribution of DP1 from intracellular compartments to the plasma membrane was observed following treatment with MK-0524 for 24 h. Furthermore, MK-0524 promoted the interaction between DP1 and the ANKRD13C protein, which we showed previously to display chaperone-like effects towards the receptor. We thus report that MK-0524 is an inverse agonist and a pharmacochaperone of DP1. Our findings may have important implications during therapeutic treatments with MK-0524 and for the development of new molecules targeting DP1. PMID:23762421

    We study the ground state of the two-dimensional d-p model in the oxide superconductors by using the variational Monte Carlo method. We employ the Gutzwiller-projected BCS and SDW wave functions in search for possible ground states with respect to dependence on parameters included in the d-p model. Near half-filling the strong antiferromagnetic correlations exist and the SDW phase extends up to 20 percent doping. It is shown that the d-wave state is possible away from half-filling for both the hole and electron doping cases. Overall structure of the phase diagram obtained by our calculations coincides with experimental indications. The superconducting condensation energy agrees well with the experimental value obtained from specific heat and critical magnetic field measurements. A possibility of stripe phase is also examined by using the SDW wave functions with incommensurate spin structures. The distance between stripes depends on the hole concentration in the underdoped region. We show that a stability of the stripe state is dependent on the transfer parameter tpp between oxygen sites.

    The function of dystrophin Dp71 in neuronal cells remains unknown. To approach this issue, we have selected the PC12 neuronal cell line. These cells express both a Dp71f cytoplasmic variant and a Dp71d nuclear isoform. In this study, we demonstrated by electron and confocal microscopy analyses of in situ nuclear matrices and Western blotting evaluation of cell extracts that Dp71d associates with the nuclear matrix. Interestingly, this binding is modulated during NGF-induced neuronal differentiation of PC12 cells with a twofold increment in the differentiated cells, compared to control cells. Also, distribution of Dp71d along the periphery of the nuclear matrix observed in the undifferentiated cells is replaced by intense fluorescent foci localized in Center of the nucleoskeletal structure. In summary, we revealed that Dp71d is a dynamic component of nuclear matrix that might participate in the nuclear modeling occurring during neuronal differentiation.

    DP2 caused by IMF southward/northward variations has important information about how the solar wind effects are transferred into the magnetosphere, and ionosphere, and on the ground. Since dayside ground magnetic field variations are significantly enhanced during DP2, dayside DP2 fluctuations have been investigated for ages. But night-side DP2 variations are not investigated enough yet. As a new approach to DP2, in this study we examined night-side magnetic variations when DP2 occurred in dayside. Ground data from MAGnetic Data Acqisition System and Circum-pan Pacific Magnetometer Network (MAGDAS/CPMN) stations were analyzed. To identify dayside DP2 events, we used the records of stations located at the dip equator. The association of DP2 with the solar wind variations, magnetic and velocity data from the ACE satellite were investigated. The obtained results by statistical analysis can be summarized as follows. (1) About half (82/153) of the dayside DP2 events are found to accompany the night-side DP2 with similar wave form. (2) The amplitude of the night-side variation (which correlates with the dayside DP2) shows a good correlation with the intensity of the dawn-to-dusk electric field in the solar wind. In the present paper, we will discuss a generation mechanism of the night-side DP2 for a case study. Acknowledgments. PGD2 signaling through DP1 and DP2 mediates different and often opposite effects in many cell types of the immune system. Although mast cells (MC) are the largest source of PGD2 in the body, there is little information about their potential expression of DP2 and its functional significance. In this study, we show that tissue MC in human nasal polyps express DP2 protein, and that human MC lines and primary cultured human MC express mRNA as well as protein of DP2. By immunohistochemistry, we detected that 34% of MC in human nasal polyps expressed DP2. In addition, flow cytometry showed that 87% of the LAD2 human MC line and 98% of primary cultured human MC contained intracellular DP2. However, we could not detect surface expression of DP2 on human MC by single cell analysis using imaging flow cytometry. Blocking of endogenous PGD2 production with aspirin did not induce surface expression of DP2 in human MC. Two DP2 selective agonists, DK-PGD2 and 15R-15-methyl PGD2 induced dose-dependent intracellular calcium mobilization that was abrogated by pertussis toxin, but not by three DP2 selective antagonists. MC mediator release including degranulation was not affected by DP2 selective agonists. Thus, human MC express DP2 intracellularly rather than on their surface, and the function of DP2 in human MC is different than in other immune cells such as Th2 cells, eosinophils and basophils where it is expressed on the cell surface and induces Th2 cytokine and/or granule associated mediator release. Further studies to elucidate the role of intracellular DP2 in human MC may expand our understanding of this molecule and provide novel therapeutic opportunities. PMID:25268140

  18. Microstructure and Mechanical Properties of Friction Stir Spot-Welded IF/DP Dissimilar Steel Joints

    Interstitial-free (IF) and dual-phase (DP) steel sheets of 1-mm thickness were joined by friction stir spot welding with a convex shoulder tool. Two different combinations were used; one with IF as top sheet (IF/DP) and another with DP as top sheet (DP/IF). Material intermixing between the overlapping sheets takes place within the stirred zone. The truncated sheet interface curls upward into the top sheet, more so in case of IF/DP, due to lower resistance offered by the top (IF) sheet to the upward migrating bottom (DP) sheet material. Material from the IF steel contains ferrite phases, while that from the DP steel contains acicular ferrite and lath martensite. Under quasi-static loading, the crack passes along the dissimilar interface and into the top sheet thickness, resulting in pull-out failure. Under cyclic loading, the failure is brought about by the initiation of kinked fatigue cracks and their subsequent propagation through the top and bottom sheet thickness. The dominant fatigue crack moves through the reduced top sheet thickness. The mechanical performance of DP/IF is better than IF/DP owing to higher strength of the stirred zone. The mechanical performances of the dissimilar joints are intermediate to that of the similar material joints.

    The DP4 parameter, which provides a confidence level for NMR assignment, has been widely used to help assign the structures of many stereochemically-rich molecules. We present an improved version of the procedure, which can be downloaded as Python script instead of running within a web-browser, and which analyses output from open-source molecular modelling programs (TINKER and NWChem) in addition to being able to use output from commercial packages (Schrodinger's Macromodel and Jaguar; Gaussian). The new open-source workflow incorporates a method for the automatic generation of diastereomers using InChI strings and has been tested on a range of new structures. This improved workflow permits the rapid and convenient computational elucidation of structure and relative stereochemistry. PMID:27045792

  20. Using Dynamic Programming and Genetic Algorithms to Reduce Erosion Risks From Forest Roads

    Many anadromous fisheries streams in the Pacific Northwest have been damaged by various land use activities, including timber harvest and road construction. Unpaved forest roads can cause erosion and downstream sedimentation damage in anadromous fish-bearing streams. Although road decommissioning and road upgrading activities have been conducted on many of these roads, these activities have usually been implemented and evaluated on a site-specific basis without the benefit of a watershed perspective. Land managers still struggle with designing the most effective road treatment plan to minimize erosion while keeping costs reasonable across a large land base. Trade-offs between costs of different levels of treatment and the net effect on reducing sediment risks to streams need to be quantified. For example, which problems should be treated first, and by what treatment method? Is it better to fix one large problem or 100 small problems? If sediment reduction to anadromous fish-bearing streams is the desired outcome of road treatment activities, a more rigorous evaluation of risks and optimization of treatments is needed. Two approaches, Dynamic Programming (DP) and Genetic Algorithms (GA), were successfully used to determine the most effective treatment levels for roads and stream crossings in a pilot study basin with approximately 200 road segments and stream crossings and in an actual watershed with approximately 600 road segments and crossings. The optimization models determine the treatment levels for roads and crossings that maximize the total sediment saved within a watershed while maintaining the total treatment cost within the specified budget. The optimization models import GIS data on roads and crossings and export the optimal treatment level for each road and crossing to the GIS watershed model.

    Urinary tract infections occur very frequently in the community and in hospitalized patients and are mainly caused by Escherichia (E.) coli. Depending on virulence determinants of uropathogenic microorganisms and host-specific defense mechanisms, urinary tract infections can manifest as cystitis, pyelonephritis (bacterial interstitial nephritis), bacteremia or urosepsis. Uncomplicated urinary tract infections in otherwise healthy women should be treated for 3-7 days depending on the antibiotic therapy chosen, even if spontaneous remission rates of up to 40% have been reported. Antibiotics of the first choice for empirical treatment of uncomplicated urinary tract infection are fluoroquinolones, pivmecillinam and fosfomycin. A huge problem is the increasing antimicrobial resistance of uropathogenic microorganisms. Complicated urinary tract infections associated with anatomical and/or functional abnormalities of the urinary tract and/or comorbidities such as diabetes or immunosuppressive therapy, need longer antibiotic treatment (e.g. 10-14 days) as well as interdisciplinary diagnostic procedures. Treatment of community acquired urosepsis includes cephalosporins of the third generation, piperacillin/tazobactam or ciprofloxacin. For nosocomial urosepsis the combination with an aminoglycoside or a carbapenem is recommended. PMID:21850538

    Despite advancement in breast cancer treatment, 30% of patients with early breast cancers experience relapse with distant metastasis. It is a challenge to identify patients at risk for relapse; therefore, the identification of markers and therapeutic targets for metastatic breast cancers is imperative. Here, we identified DP103 as a biomarker and metastasis-driving oncogene in human breast cancers and determined that DP103 elevates matrix metallopeptidase 9 (MMP9) levels, which are associated with metastasis and invasion through activation of NF-κB. In turn, NF-κB signaling positively activated DP103 expression. Furthermore, DP103 enhanced TGF-β-activated kinase-1 (TAK1) phosphorylation of NF-κB-activating IκB kinase 2 (IKK2), leading to increased NF-κB activity. Reduction of DP103 expression in invasive breast cancer cells reduced phosphorylation of IKK2, abrogated NF-κB-mediated MMP9 expression, and impeded metastasis in a murine xenograft model. In breast cancer patient tissues, elevated levels of DP103 correlated with enhanced MMP9, reduced overall survival, and reduced survival after relapse. Together, these data indicate that a positive DP103/NF-κB feedback loop promotes constitutive NF-κB activation in invasive breast cancers and activation of this pathway is linked to cancer progression and the acquisition of chemotherapy resistance. Furthermore, our results suggest that DP103 has potential as a therapeutic target for breast cancer treatment. PMID:25083991

  8. On Key Factors Influencing Ductile Fractures of Dual Phase (DP) Steels

    In this paper, we examine the key factors influencing ductile failure of various grades of dual phase (DP) steels using the microstructure-based modeling approach. Various microstructure-based finite element models are generated based on the actual microstructures of DP steels with different martensite volume fractions. These models are, then, used to investigate the influence of ductility of the constituent ferrite phase and also the influence of voids introduced in the ferrite phase on the overall ductility of DP steels. It is found that with volume fraction of martensite in the microstructure less than 15%, the overall ductility of the DP steels strongly depends on the ductility of the ferrite matrix, hence pre-existing micro voids in the microstructure significantly reduce the overall ductility of the steel. When the volume fraction of martensite is above 15%, the preexisting voids in the ferrite matrix does not significantly reduce the overall ductility of the DP steels, and the overall ductility is more influenced by the mechanical property disparity between the two phases. The applicability of the phase inhomogeneity driven ductile failure of DP steels is then discussed based on the obtained computational results for various grades of DP steels, and the experimentally obtained scanning electron microscopy (SEM) pictures of the corresponding grades of DP steels near fracture surface are used as evidence for result validations.

    Understanding retinal vascular development is crucial because many retinal vascular diseases such as diabetic retinopathy (in adults) or retinopathy of prematurity (in children) are among the leading causes of blindness. Given the localization of the protein Dp71 around the retinal vessels in adult mice and its role in maintaining retinal homeostasis, the aim of this study was to determine if Dp71 was involved in astrocyte and vascular development regulation. An experimental study in mouse retinas was conducted. Using a dual immunolabeling with antibodies to Dp71 and anti-GFAP for astrocytes on retinal sections and isolated astrocytes, it was found that Dp71 was expressed in wild-type (WT) mouse astrocytes from early developmental stages to adult stage. In Dp71-null mice, a reduction in GFAP-immunopositive astrocytes was observed as early as postnatal day 6 (P6) compared with WT mice. Using real-time PCR, it was showed that Dp71 mRNA was stable between P1 and P6, in parallel with post-natal vascular development. Regarding morphology in Dp71-null and WT mice, a significant decrease in overall astrocyte process number in Dp71-null retinas at P6 to adult age was found. Using fluorescence-conjugated isolectin Griffonia simplicifolia on whole mount retinas, subsequent delay of developing vascular network at the same age in Dp71-null mice was found. An evidence that the Dystrophin Dp71, a membrane-associated cytoskeletal protein and one of the smaller Duchenne muscular dystrophy gene products, regulates astrocyte morphology and density and is associated with subsequent normal blood vessel development was provided.     The mammalian transcription factor E2F plays an important role in regulating the expression of genes that are required for passage through the cell cycle. This transcriptional activity is inhibited by association with the retinoblastoma tumor suppressor protein (pRB) or its relatives p107 and p103. The first cDNA from the E2F family to be cloned was designated E2F-1, and multiple E2F family members have now been identified. They bind to DNA as heterodimers, interacting with proteins known as DP. Here we demonstrate that DP is also a family of polypeptides with at least two members (hDP-1 and hDP-2). Both hDP-1 and hDP-2 bind to all E2F family members in vivo, and each complex is capable of activating transcription. However, the various E2F/DP complexes display strong differences in the ability to bind to either pRB or p107 in vivo, and the specificity of pRB or p107 binding is mediated by the E2F subunit.     The nearr-Earth Asteroid (NEA) 2006 DP14 was discovered by the LINEAR asteroid discovery program on February 23, 2006 (MPEC 2006-D40). With a likely diameter on the order of ~500 meters and a Minimum Orbit Intersection Distance MOID = 0.016 AU, the object has been designated as a Potentially Hazardous Asteroid (PHA) by the Minor Planet Center. Delay-doppler radar imaging was obtained by the JPL Planetary Radar Team at Goldstone on February 12, 2014 (http://echo.jpl.nasa.gov/asteroids/2006DP14/2006DP14_planning.html).

    A field study was conducted to measure dust from haul trucks at a limestone quarry and a coal preparation plant waste hauling operation. The study found that primarily wind, distance and road treatment conditions notably affected the dust concentrations at locations next to, 50 ft from, and 100 ft away from the unpaved haulage road. Airborne dust measured along the unpaved haul road showed that high concentrations of fugitive dust can be generated with these concentrations rapidly decreasing to nearly background levels within 100 ft of the road. Instantaneous respirable dust measurements illustrated that the trucks generate a real-time dust cloud that has a peak concentration with a time-related decay rate as the dust moves past the sampling locations. The respirable dust concentrations and peak levels were notably diminished as the dust cloud was transported, diluted, and diffused by the wind over the 100 ft distance from the road. Individual truck concentrations and peak levels measured next to the dry road surface test section were quite variable and dependent on wind conditions, particularly wind direction, with respect to reaching the sampling location. The vast majority of the fugitive airborne dust generated from unpaved and untreated haulage roads was non-respirable. 6 figs.

    We present a one dimensional model for the development of corrugations in roads subjected to compressive forces from a flux of cars. The cars are modeled as damped harmonic oscillators translating with constant horizontal velocity across the surface, and the road surface is subject to diffusive relaxation. We derive dimensionless coupled equations of motion for the positions of the cars and the road surface H( x, t), which contain two phenomenological variables: an effective diffusion constant Δ( H) that characterizes the relaxation of the road surface, and a function a( H) that characterizes the plasticity or erodibility of the road bed. Linear stability analysis shows that corrugations grow if the speed of the cars exceeds a critical value, which decreases if the flux of cars is increased. Modifying the model to enforce the simple fact that the normal force exerted by the road can never be negative seems to lead to restabilized, quasi-steady road shapes, in which the corrugation amplitude and phase velocity remain fixed.

    Dystrophin Dp71 is the main product of the Duchenne muscular dystrophy gene in the brain; however, its function is unknown. To study the role of Dp71 in neuronal cells, we previously generated by antisense treatment PC12 neuronal cell clones with decreased Dp71 expression (antisense-Dp71 cells). PC12 cells express two different splicing isoforms of Dp71, a cytoplasmic variant called Dp71f and a nuclear isoform called Dp71d. We previously reported that antisense-Dp71 cells display deficient adhesion to substrate and reduced immunostaining of β1-integrin in the cell area contacting the substrate. In this study, we isolated additional antisenseDp71 clones to analyze in detail the potential involvement of Dp71f isoform with the β1-integrin adhesion system of PC12 cells. Immunofluorescence analyses as well as immunoprecipitation assays demonstrated that the PC12 cell β1-integrin adhesion complex is composed of β1-integrin, talin, paxillin, α-actinin, FAK and actin. In addition, our results showed that Dp71f associates with most of the β1-integrin complex components (β1-integrin, FAK, α-actinin, talin and actin). In the antisense-Dp71 cells, the deficiency of Dp71 provokes a significant reduction of the β1-integrin adhesion complex and, consequently, the deficient adhesion of these cells to laminin. In vitro binding experiments confirmed the interaction of Dp71f with FAK and β1-integrin. Our data indicate that Dp71f is a structural component of the β1-integrin adhesion complex of PC12 cells that modulates PC12 cell adhesion by conferring proper complex assembly and/or maintenance. The association (K+1) and dissociation (K−1) rate constants of binding were 0.758±0.064 min−1, mmol and 0.0042±0.0002 min−1, respectively, yielding dissociation constants (KDs) of 5.66±0.44 nM (n=4).Specific [3H]-BWA868C bound to DP receptors with a high affinity (KD=1.45±0.01 nM, n=3) and to a finite, saturable number of binding sites (Bmax=21.1±0.6 nmol g−1 wet weight).DP receptor class prostanoids (e.g. ZK118182, BW245C, BWA868C, PGD2) exhibited high (nanomolar) affinities for [3H]-BWA868C binding, while prostanoids selective for EP, FP, IP and TP receptors showed a low (micromolar) affinity.Specific DP receptor binding sites were autoradiographically localized on the ciliary epithelium/process, longitudinal and circular ciliary muscles, retinal choroid and iris in human eye sections using [3H]-BWA868C. While [3H]-PGD2 yielded similar quantitative distribution of DP receptors as [3H]-BWA868C, the level of non-specific binding observed with [3H]-PGD2 was significantly greater than that observed with [3H]-BWA868C.It is concluded that [3H]-BWA868C is a high-affinity and very specific DP receptor radioligand capable of selectively labelling the DP receptor. [3H]-BWA868C may prove useful for future homogenate-based and autoradiographic studies on the DP receptor. PMID:11082108

    Prostaglandin D2 (PGD2) plays a key role in mediating allergic reactions seen in asthma, allergic rhinitis, and atopic dermatitis. PGD2 exerts its activity through two G protein-coupled receptors (GPCRs), prostanoid D receptor (DP or DP1), and chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2 or DP2). We report the optimization of a series of phenylacetic acid derivatives in an effort to improve the dual activity of AMG 009 against DP and CRTH2. These efforts led to the discovery of AMG 853 (2-(4-(4-(tert-butylcarbamoyl)-2-(2-chloro-4-cyclopropylphenyl sulfonamido)phenoxy)-5-chloro-2-fluorophenyl)acetic acid), which is being evaluated in human clinical trials for asthma.     A DNA binding protein with a molecular weight of 64,000, designated 64DP, has been purified and characterized. This protein was isolated from adult human pooled serum by DEAE Sephadex column Chromatography, DNA cellulose affinity column chromatography, ammonium sulfate fractionation and Sephadex G-150 gel filtration. The final preparation of 64DP was homogeneous, as judged from polyacrylamide gel electrophoresis with or without sodium dodecyl sulfate and sedimentation experiments. Physicochemical and immunochemical properties of this protein were very similar or identical to those of alpha-1-antichymotrypsin with some differences in electric mobility and th pattern of isoelectric focusing. Furthermore, the general properties of 64DP from various sera were practically similar with the exception that isolectric focusing analysis showed microheterogeneity among 64DP purified from various sera.     Microstructure failure mechanisms and void nucleation in dual-phase (DP) steels during deformation have been studied using a combination of in situ tensile testing in a scanning electron microscope (SEM), digital image correlation (DIC) and finite element (FE) modelling. SEM images acquired during in situ tests were used to follow the evolution of damage within the microstructure of a DP1000 steel. From these images, strain maps were generated using DIC and used as boundary conditions for a FE model to investigate the stress state of martensite and ferrite before the onset of the martensite phase cracking. Based on the simulation results, a maximum principal stress of about 1700 MPa has been estimated for crack initiation in the martensite of the investigated DP1000 steel. The SEM image observations in combination with the FE analyses provide new insights for the development of physically-based damage models for DP-steels.

    Hydrodynamic formulas for mass-loaded spring-driven liquid-surface waves, together with cochlear parameters derived from experimental literature values of single-tone BM-vibration phases, are used to predict the phases of cubic DP's and DPOAE's. These predictions depend on the primary-tone level and on the category (low- or high-side) of the distortion; they agree [disagree] with recent fixed- f2 low-side DPOAE experiments by two different groups in Mongolian gerbil if the backward cochlear DP-wave is assumed to be a slow surface wave [a fast compression wave]. The weakness of DPOAE's, particularly of high-side ones, is argued to be due to destructive interference among slow backward DP-TW's from different parts of the DP generation zone.

    This thesis investigated the science behind polarimetric road ice detection systems. Laboratory Mueller matrix measurements of a simulated road under differing surface conditions were collected searching for a discriminatory polarization property. These Mueller matrices were decomposed into depolarization, diattenuation, and retardance. Individual sample surface polarization properties were then calculated from these three unique matrices and compared. Specular and off-specular reflection responses of each sample were collected. Four polarization properties stood out for having high separation between dry and iced measurements: Depolarization Index, Linear Diattenuation, Linear Polarizance, and Linear Retardance. Through our investigation polarimetric ice detection is possible. Continued research of the polarization properties of road ice can result in the development of a road ice detection system. Proposed deployment methods of such a system have been outlined following the analysis of the data collected in this experiment.

    A method for extracting road boundaries using the monochrome image of a visual road scene is presented. The statistical information regarding the intensity levels present in the image along with some geometrical constraints concerning the road are the basics of this approach. Results and advantages of this technique compared to others are discussed. The major advantages of this technique, when compared to others, are its ability to process the image in only one pass, to limit the area searched in the image using only knowledge concerning the road geometry and previous boundary information, and dynamically adjust for inconsistencies in the located boundary information, all of which helps to increase the efficacy of this technique.

    Dystrophin Dp71, the smallest product encoded by the Duchenne muscular dystrophy gene, is ubiquitously expressed in all non-muscle cells. Although Dp71 is involved in various cellular processes, the mechanisms underlying its expression have been little studied. In hepatic cells, Dp71 expression is down-regulated by the xenobiotic β-naphthoflavone. However, the effectors of this regulation remain unknown. In the present study we aimed at identifying DNA elements and transcription factors involved in Dp71 expression in hepatic cells. Relevant DNA elements on the Dp71 promoter were identified by comparing Dp71 5'-end flanking regions between species. The functionality of these elements was demonstrated by site-directed mutagenesis. Using EMSAs and ChIP, we showed that the Sp1 (specificity protein 1), Sp3 (specificity protein 3) and YY1 (Yin and Yang 1) transcription factors bind to the Dp71 promoter region. Knockdown of Sp1, Sp3 and YY1 in hepatic cells increased endogenous Dp71 expression, but reduced Dp71 promoter activity. In summary, Dp71 expression in hepatic cells is carried out, in part, by YY1-, Sp1- and Sp3-mediated transcription from the Dp71 promoter. PMID:27143785

  2. The role of the prostaglandin D2 receptor, DP, in eosinophil trafficking.

    Prostaglandin (PG) D2 is a major mast cell product that acts via two receptors, the D-type prostanoid (DP) and the chemoattractant receptor-homologous molecule expressed on Th2 cells (CRTH2) receptors. Whereas CRTH2 mediates the chemotaxis of eosinophils, basophils, and Th2 lymphocytes, the role of DP has remained unclear. We report in this study that, in addition to CRTH2, the DP receptor plays an important role in eosinophil trafficking. First, we investigated the release of eosinophils from bone marrow using the in situ perfused guinea pig hind limb preparation. PGD2 induced the rapid release of eosinophils from bone marrow and this effect was inhibited by either the DP receptor antagonist BWA868c or the CRTH2 receptor antagonist ramatroban. In contrast, BWA868c did not inhibit the release of bone marrow eosinophils when this was induced by the CRTH2-selective agonist 13,14-dihydro-15-keto-PGD2. In additional experiments, we isolated bone marrow eosinophils from the femoral cavity and found that these cells migrated toward PGD2. We also observed that BWA868c inhibited this response to a similar extent as ramatroban. Finally, using immunohistochemistry we could demonstrate that eosinophils in human bone marrow specimens expressed DP and CRTH2 receptors at similar levels. Eosinophils isolated from human peripheral blood likewise expressed DP receptor protein but at lower levels than CRTH2. In agreement with this, the chemotaxis of human peripheral blood eosinophils was inhibited both by BWA868c and ramatroban. These findings suggest that DP receptors comediate with CRTH2 the mobilization of eosinophils from bone marrow and their chemotaxis, which might provide the rationale for DP antagonists in the treatment of allergic disease.     We previously reported the identification of DP-1 isoforms ({alpha} and {beta}), which are structurally C-terminus-deleted ones, and revealed the low-level expression of these isoforms. It is known that wild-type DP-1 is degraded by the ubiquitin-proteasome system, but few details are known about the domains concerned with the protein stability/instability for the proteolysis of these DP-1 isoforms. Here we identified the domains responsible for the stability/instability of DP-1. Especially, the DP-1 'Stabilon' domain was a C-terminal acidic motif and was quite important for DP-1 stability. Moreover, we propose that this DP-1 Stabilon may be useful for the stability of other nuclear proteins when fused to them.

  6. Conventional and Multiscale Modeling of Microstructure Evolution During Laminar Cooling of DP Steel Strips

    Physical and numerical simulations of the hot rolling and laminar cooling of DP steel strips are presented in the paper. The objectives of the paper were twofold. Physical simulations of hot plastic deformation were used to identify and validate numerical models. Validated models were applied to simulate the manufacturing of DP steel strips. Conventional flow stress model and microstructure evolution model were used in the hot deformation part. The approach to the complex systems analysis based on global thermodynamic characterization and detailed microstructure characterization was applied to determine equilibrium state at various temperatures. Finally, two numerical models were used to simulate kinetics of austenite decomposition at varying temperatures: the first, conventional model based on the Avrami equation, and the second, the discrete Cellular Automata approach. Plastometric tests and stress relaxation tests were used for identification of the hot rolling model for the DP steel. Dilatometric tests were performed to identify the phase transformation models. Verification confirmed good accuracy of all models. Validated models were applied to simulate the manufacturing of DP steel strips. Influence of technological parameters ( e.g., strip thickness and velocity, active sections in the laminar cooling, and water flux in the sections) on the DP microstructure was analyzed. The cooling schedules, which give required microstructures were proposed. The numerical tool, which simulates manufacturing chain for DP steel strips is the main output of the paper.

  7. Applicability of a Micromechanics Model Based on Actual Microstructure for Failure Prediction of DP Steels

    In this paper, various micromechanics models based on actual microstructures of DP steels are examined in order to determine the reasonable range of martensite volume fraction where the methodology described in this study can be applied. For this purpose, various micromechanics-based finite element models are first created based on the actual microstructures of DP steels with different martensite volume fractions. These models are, then, used to investigate the influence of ductility of the constituent ferrite and martensite phases and also the influence of voids in the ferrite phase on the overall ductility of DP steels. The computational results indicate that there is a range of martensite volume fraction where the phase inhomogeneity between the ferrite and martensite phases has dominant effect on the overall ductility of DP steels, defeating the influence of the ductility of each phase and the voids in the ferrite phase, and that this phase inhomogeneity dominant region includes the range of marteniste volume fraction between 15% and 40%. Therefore, the methodology, adopted in this study, may be applied to DP steels within the phase inhomogeneity dominant region in tailoring the DP steel design for its intended purpose and desired properties.

    The aims of the present study were to determine whether the estimations of the frontal area of the combined cyclist-bicycle (APCB) obtained with the Heil's non-logarithmic prediction equations (NPE) in the stem position (SP), brake hoods position (BHP) and drops position (DP) are comparable to the measured APCB with the computerized planimetry (CP) method, and to analyse with the CP method and the NPE the influence of the body position on the APCB. Nineteen participants competing in the Spanish Road Cycling First division took part in the study. The NPE overestimated the APCB in the BHP and in the DP compared with the measured APCB with the CP method (6.9% and 5.1%, respectively; p<0.05). Significant differences among the three positions were obtained with the CP method. The overestimation of the APCB with the NPE in the BHP and in the DP and the less sensitivity of the NPE to show significant differences between the SP and DP suggest that the NPE are not appropriate to accurately predict the APCB.   9. Effect of the PGD2-DP signaling pathway on primary cultured rat hippocampal neuron injury caused by aluminum overload

    In the present study, the agonists and antagonists of DP receptor were used to examine whether the PGD2-DP signaling pathway affects neuronal function. Primary cultured hippocampal neuron was prepared and treated with aluminum maltolate (100 μM) to establish the neuronal damage model. PGD2 and cAMP content was detected by ELISA. L-PGDS and DPs mRNA and protein expression were measured by RT-PCR and Western blotting, respectively. The aluminium-load neuron was treated with the DP1 agonist BW245C, the DP1 antagonist BWA868C, the DP2 agonist DK-PGD2, and the DP2 antagonist CAY10471, respectively. Neuronal pathomorphology was observed using H-E staining. The cell viability and the lactate dehydrogenase leakage rates of neurons were measured with MTT and LDH kit, respectively. Ca2+ level was detected by Fluo-3/AM. In the model group, the MTT values obviously decreased; LDH leakage rates and PGD2 content increased significantly; L-PGDS, DP1 mRNA and protein expressions increased, and DP2 level decreased. BW245C reduced the Ca2+ fluorescence intensity and protected the neurons. DK-PGD2 increased the intensity of Ca2+ fluorescence, while CAY10471 had the opposite effect. In conclusion, contrary to the effect of DP2, the PGD2-DP1 signaling pathway protects against the primary cultured rat hippocampal neuronal injury caused by aluminum overload. PMID:27089935

    Susceptibility to chronic beryllium disease (CBD) is linked to certain HLA-DP molecules, including HLA-DP2. To elucidate the molecular basis of this association, we exposed mice transgenic (Tg) for HLA-DP2 to beryllium oxide (BeO) via oropharyngeal aspiration. As opposed to WT mice, BeO-exposed HLA-DP2 Tg mice developed mononuclear infiltrates in a peribronchovascular distribution that were composed of CD4(+) T cells and included regulatory T (Treg) cells. Beryllium-responsive, HLA-DP2-restricted CD4(+) T cells expressing IFN-γ and IL-2 were present in BeO-exposed HLA-DP2 Tg mice and not in WT mice. Using Be-loaded HLA-DP2-peptide tetramers, we identified Be-specific CD4(+) T cells in the mouse lung that recognize identical ligands as CD4(+) T cells derived from the human lung. Importantly, a subset of HLA-DP2 tetramer-binding CD4(+) T cells expressed forkhead box P3, consistent with the expansion of antigen-specific Treg cells. Depletion of Treg cells in BeO-exposed HLA-DP2 Tg mice exacerbated lung inflammation and enhanced granuloma formation. These findings document, for the first time to our knowledge, the development of a Be-specific adaptive immune response in mice expressing HLA-DP2 and the ability of Treg cells to modulate the beryllium-induced granulomatous immune response.   11. Proximity of California public schools to busy roads.

    Residential proximity to busy roads has been associated with adverse health outcomes, and school location may also be an important determinant of children's exposure to traffic-related pollutants. The goal of this study was to examine the characteristics of public schools (grades K-12) in California (n = 7,460) by proximity to major roads. We determined maximum daily traffic counts for all roads within 150 m of the school using a statewide road network and a geographic information system. Statewide, 173 schools (2.3%) with a total enrollment of 150,323 students were located within 150 m of high-traffic roads (greater than or equal to 50,000 vehicles/day); 536 schools (7.2%) were within 150 m of medium-traffic roads (25,000-49,999 vehicles/day). Traffic exposure was related to race/ethnicity. For example, the overall percentage of nonwhite students was 78% at the schools located near high-traffic roads versus 60% at the schools with very low exposure (no streets with counted traffic data within 150 m). As the traffic exposure of schools increased, the percentage of both non-Hispanic black and Hispanic students attending the schools increased substantially. Traffic exposure was also related to school-based and census-tract-based socioeconomic indicators, including English language learners. The median percentage of children enrolled in free or reduced-price meal programs increased from 40.7% in the group with very low exposure to 60.5% in the highest exposure group. In summary, a substantial number of children in California attend schools close to major roads with very high traffic counts, and a disproportionate number of those students are economically disadvantaged and nonwhite.     Clinical presentation helps differentiate between upper and lower urinary tract infections (UTIs). UTIs are classified as either complicated or uncomplicated. A complicated UTI is associated with an underlying condition that increases the risk of failing therapy. Primary laboratory tests for UTIs consist of urinalysis and urine culture. The most common pathogen for uncomplicated cystitis and pyelonephritis is Escherichia coli. Nitrofurantoin, fosfomycin, and trimethoprim-sulfamethoxazole are first-line therapies for acute uncomplicated cystitis. Decisions regarding antibiotic agents should be individualized based on patients' allergies, tolerability, community resistance rates, cost, and availability. PMID:23958364

    Disclosed are improvments to a rapid road repair vehicle comprising an improved cleaning device arrangement, two dispensing arrays for filling defects more rapidly and efficiently, an array of pre-heaters to heat the road way surface in order to help the repair material better bond to the repaired surface, a means for detecting, measuring, and computing the number, location and volume of each of the detected surface imperfection, and a computer means schema for controlling the operation of the plurality of vehicle subsystems. The improved vehicle is, therefore, better able to perform its intended function of filling surface imperfections while moving over those surfaces at near normal traffic speeds.

    The response of equatorial ionosphere to the magnetospheric origin DP 2 current system fluctuations is examined using ground-based multiinstrument observations. The interaction between the solar wind and magnetosphere generates a convection electric field that can penetrate to the ionosphere and cause the DP 2 current system. The quasiperiodic DP 2 current system, which fluctuates coherently with fluctuations of the interplanetary magnetic field (IMF) Bz, penetrates nearly instantaneously to the dayside equatorial region at all longitudes and modulates the electrodynamics that governs the equatorial density distributions. In this paper, using magnetometers at high and equatorial latitudes, we demonstrate that the quasiperiodic DP 2 current system penetrates to the equator and causes the dayside equatorial electrojet (EEJ) and the independently measured ionospheric drift velocity to fluctuate coherently with the high-latitude DP 2 current as well as with the IMF Bz component. At the same time, radar observations show that the ionospheric density layers move up and down, causing the density to fluctuate up and down coherently with the EEJ and IMF Bz.

    Platelet adhesion is a critical step due to its hemostatic role in stopping bleeding after vascular damage. Short dystrophins are the most abundant dmd gene products in nonmuscle tissues, and in association with cytoskeleton proteins contribute to their intrinsic function; while utrophins are dystrophin-homologous related family proteins with structural and functional similarities. We previously demonstrated the presence of Dp71 isoforms, utrophins, and various dystrophin-associated proteins and their participation in cytoskeleton re-organization, filopodia and lamellipodia extension, and in centralizing cytoplasmic granules during the adhesion process of human platelets. To evaluate the morphologic changes and actin-based structures of mdx(3cv) platelets during the adhesion process, we compared the topographic distribution of Dp71d/Dp71Delta110(m) and dystrophin-associated protein in adhered platelets from dystrophic mdx(3cv) mouse. By confocal microscopy, we showed that absence of Dp71 isoforms in platelets from this animal model disrupted dystrophin-associated protein expression and distribution without modifying the platelet morphology displayed during the glass-adhesion process. By immunoprecipitation assays, we proved that up-regulated utrophins were associated with dystrophin-associated proteins to conform the dystrophin-associated protein complex corresponding to utrophins, which might compensate for Dp71 absence in mdx(3cv) platelets.     Fructans are important vacuolar reserve carbohydrates with drought, cold, ROS and general abiotic stress mediating properties. They occur in 15% of all flowering plants and are believed to display health benefits as a prebiotic and dietary fiber. Fructans are synthesized by specific fructosyltransferases and classified based on the linkage type between fructosyl units. Inulins, one of these fructan types with β(2-1) linkages, are elongated by fructan:fructan 1-fructosyltransferases (1-FFT) using a fructosyl unit from a donor inulin to elongate the acceptor inulin molecule. The sequence identity of the 1-FFT of Viguiera discolor (Vd) and Helianthus tuberosus (Ht) is 91% although these enzymes produce distinct fructans. The Vd 1-FFT produces high degree of polymerization (DP) inulins by preferring the elongation of long chain inulins, in contrast to the Ht 1-FFT which prefers small molecules (DP3 or 4) as acceptor. Since higher DP inulins have interesting properties for industrial, food and medical applications, we report here on the influence of two amino acids on the high DP inulin production capacity of the Vd 1-FFT. Introducing the M19F and H308T mutations in the active site of the Vd 1-FFT greatly reduces its capacity to produce high DP inulin molecules.     This paper introduces a universal classification methodology for discretely sampled sealed bituminous road profile data for the study of shock and vibrations related to the road transportation process. Data representative of a wide variety of Victorian (Australia) road profiles were used to develop a universal classification methodology with special attention to their non-Gaussian and nonstationary properties. This resulted in the design of computer software to automatically detect and extract transient events from the road spatial acceleration data as well as to identify segments of the constant RMS level enabling transients to be analyzed separately from the underlying road process. Nine universal classification parameters are introduced to describe road profile spatial acceleration based on the statistical characteristics of the transient amplitude and stationary RMS segments. Results from this study are aimed at the areas of road transport simulation as well as road surface characterization.

  14. Biochemical and pharmacological characterization of AZD1981, an orally available selective DP2 antagonist in clinical development for asthma

    Background and Purpose The discovery of DP2 as a second receptor for PGD2 has prompted the search for antagonists as potential novel therapies based on the associations between PGD2 and disease. Here we describe the biochemical and pharmacological properties of 4-(acetylamino)-3-[(4-chlorophenyl)thio]-2-methyl-1H-indole-1-acetic acid (AZD1981), a novel DP2 receptor antagonist. Experimental Approach Binding to DP2, functional receptor pharmacology and selectivity were studied in both human and animal systems. Key Results AZD1981 displaced radio-labelled PGD2 from human recombinant DP2 with high potency (pIC50 = 8.4). Binding was reversible, non-competitive and highly selective against a panel of more than 340 other enzymes and receptors, including DP1 (>1000-fold selective). AZD1981 inhibited DP2-mediated shape change and CD11b up-regulation in human eosinophils, shape change in basophils and chemotaxis of human eosinophils and Th2 cells with similar potency. AZD1981 exhibited good cross-species binding activity against mouse, rat, guinea pig, rabbit and dog DP2. Evaluation in mouse, rat or rabbit cell systems was not possible as they did not respond to DP2 agonists. Agonist responses were seen in guinea pig and dog, and AZD1981 blocked DP2-mediated eosinophil shape change. Such responses were more robust in the guinea pig, where AZD1981 also blocked DP2-dependent eosinophil emigration from bone marrow. Conclusions and Implications AZD1981 is a DP2 antagonist that blocks functional responses in eosinophils, Th2 cells and basophils. It exhibited similar potency irrespective of the cell type, DP2 agonist or species used. This selective orally active agent is currently under clinical evaluation as a potential therapeutic agent in respiratory diseases including asthma. PMID:23146091

  18. Fine-Scale Road Stretch Forecasting along Main Danish Roads

    The DMI has in collaboration with the Danish Road Directorate (DRD) for almost two decades used a Road Condition Model (RCM) system (based on a dense road observations network and the numerical weather prediction model - HIgh Resolution Limited Area Model, HIRLAM) to provide operational forecasts of main road conditions at selected road stations of the Danish road network. As of Jan 2009, there are 357 road stations (equipped in total with 456 sensors), where measurements and forecasts of road surface temperature, air and dew point temperatures are conducted. Forecasts of other important meteorological parameters such as cloud cover and precipitations as well as radar and satellite images are also distributed to the users through the web-based interface vejvejr.dk and through DMI and DRD web-pages. For icing conditions, new technology has made it easy to vary the dose of spreaded salt, making it possible to use salt only on the parts of the road network where it is really needed. In our study measurements of road surface temperature from road stations and salt spreaders have additionally been used to examine both road stations and road stretches forecasts along the main roads of the Danish Road Network (accounting almost 23 thousand points located at distances of 250 m). These results showed critical importance of availability of detailed characteristics of the roads surroundings. To make local forecasts in a specific point all possible local detailed information is needed. Since high resolution models running at faster supercomputers as well as detailed physiographic datasets now are available, it is possible to improve the modelling and parameterization of significant physical processes influencing the formation of the slippery road conditions.     DP-2 type fluctuations caused by IMF southward/northward variations have important information about how the solar wind effects are transferred into the magnetosphere, ionosphere, and on the ground. Since dayside ground magnetic field variations are significantly enhanced during DP-2 events, dayside DP-2 fluctuations have been investigated for many years. However night-side DP-2 variations are not yet investigated sufficiently. In this study, we examined night-side magnetic and electric field variations when DP-2 was observed in dayside. Ground data from MAGDAS/CPMN (MAGnetic Data Acqisition System and Circum-pan Pacific Magnetometer Network) stations were analyzed. We also investigated the association of DP-2 with the ionospheric electric fields obtained by the FM-CW (Frequency Modulated Continuous Wave) radar at PTK (M.Lat. = 45.8 degree, M.Lon.= 221.6 degree) and solar wind parameters from the ACE satellite. The amplitudes of night-side DP-2 on 11 Oct., 2008 became greater with increasing of the latitude of 210 MM stations. While there were no Pi 2 pulsations, and signatures of substorms were not detected with the DP 2. At the time, westward electric field was observed by the FM-CW radar in the night-side sector simultaneously with the positive variation of night-side ground magnetic fields. Therefore the night-side DP-2 magnetic fluctuation is found not to be caused by the ionospheric currents.

    DP-b99 is a membrane-activated chelator of zinc and calcium ions, recently proposed as a therapeutic agent. Matrix metalloproteinases (MMPs) are zinc-dependent extracellularly operating proteases that might contribute to synaptic plasticity, learning and memory under physiological conditions. In excessive amounts these enzymes contribute to a number of neuronal pathologies ranging from the stroke to neurodegeneration and epileptogenesis. In the present study, we report that DP-b99 delays onset and severity of PTZ-induced seizures in mice, as well as displays neuroprotective effect on kainate excitotoxicity in hippocampal organotypic slices and furthermore blocks morphological reorganization of the dendritic spines evoked by a major neuronal MMP, MMP-9. Taken together, our findings suggest that DP-b99 may inhibit neuronal plasticity driven by MMPs, in particular MMP-9, and thus may be considered as a therapeutic agent under conditions of aberrant plasticity, such as those subserving epileptogenesis.   1. In vitro evaluation of the DP-4M PennCentury insufflator.

    Dry powder formulations for inhalation have to be screened in animal studies for therapeutic efficacy and safety aspects and both are significantly affected by the dose and the particle size distribution (PSD) of the aerosol that is given. One of the most frequently used apparatus for pulmonary delivery of dry powder formulations in mice studies is the PennCentury DP-4M Dry Powder Insufflator. To make researchers of future preclinical animal studies with the DP-4M insufflator aware of the pitfalls regarding the conclusions to be drawn from their data, we investigated the dispersion behaviour by the DP-4M insufflator using two to three different powder preparation techniques for four different compounds. The primary PSDs of the different formulations were determined in duplicate by laser diffraction analysis. To measure the PSDs of the aerosols obtained with the DP-4M insufflator, the same diffractometer was used in combination with an in-house constructed adapter for the insufflator. The dispersion efficiency and delivered dose were highly affected by the amount of air available for dispersion; the 200 μL of air recommended for the type of insufflator used was insufficient for adequate dispersion. In contrast, the weighed dose did not have a profound effect on the dispersion behaviour and the delivered dose of the DP-4M insufflator. Also the physico-chemical powder properties and the applied particle preparation technique influenced the amount and PSD of the delivered aerosol only to a limited extend, with a few exceptions. We advise researchers to investigate the dispersion efficiency and delivered dose from the DP-4M insufflator with the formulation under investigation prior to in vivo studies and it may be necessary to optimise the formulation for administration to mice.   2. Crystal structure of HLA-DP2 and implications for chronic beryllium disease

    Chronic beryllium disease (CBD) is a fibrotic lung disorder caused by beryllium (Be) exposure and is characterized by granulomatous inflammation and the accumulation of Be-responsive CD4{sup +} T cells in the lung. Genetic susceptibility to CBD has been associated with certain alleles of the MHCII molecule HLA-DP, especially HLA-DPB1*0201 and other alleles that contain a glutamic acid residue at position 69 of the {beta}-chain ({beta}Glu69). The HLA-DP alleles that can present Be to T cells match those implicated in the genetic susceptibility, suggesting that the HLA contribution to disease is based on the ability of those molecules to bind and present Be to T cells. The structure of HLA-DP2 and its interaction with Be are unknown. Here, we present the HLA-DP2 structure with its antigen-binding groove occupied by a self-peptide derived from the HLA-DR {alpha}-chain. The most striking feature of the structure is an unusual solvent exposed acidic pocket formed between the peptide backbone and the HLA-DP2 {beta}-chain {alpha}-helix and containing three glutamic acids from the {beta}-chain, including {beta}Glu69. In the crystal packing, this pocket has been filled with the guanidinium group of an arginine from a neighboring molecule. This positively charged moiety forms an extensive H-bond/salt bridge network with the three glutamic acids, offering a plausible model for how Be-containing complexes might occupy this site. This idea is strengthened by the demonstration that mutation of any of the three glutamic acids in this pocket results in loss of the ability of DP2 to present Be to T cells.

  3. Relationship between Material Properties and Local Formability of DP980 Steels

    A noticeable degree of inconsistent forming behaviors has been observed for the 1st generation advanced high strength steels (AHSS) in production, and they appear to be associated with the inherent microstructural-level inhomogeneities for various AHSS. This indicates that the basic material property requirements and screening methods currently used for the mild steels and high strength low alloys (HSLA) are no longer sufficient for qualifying today’s AHSS. In order to establish more relevant material acceptance criteria for AHSS, the fundamental understandings on key mechanical properties and microstructural features influencing the local formability of AHSS need to be developed. For this purpose, in this study, DP980 was selected as model steels and eight different types of DP980 sheet steels were acquired from various steel suppliers. Various experiments were then performed on the eight different DP980 steels such as chemical composition analysis, static tensile test, hole expansion test, channel forming test. Scanning electron microscope (SEM) pictures of the DP980 steels were also obtained, and image processing tools were then adopted to those SEM pictures in order to quantify their various microstructural features. The results show that all DP980 steels show large discrepancy in their performance and that the tensile properties and hole expansion properties of these steels do not correlate with their local formability. According to the results up to date, it is not possible to correlate the microstructural features alone to the macroscopically measured deformation behaviors. In addition to image analysis, other experiments (i.e., nano-indentation test) are also planned to quantify the individual phase properties of the various DP steels.

    Staphylococcus lugdunensis is a known pathogen of infective endocarditis, but not of urinary tract infection. We report a previously healthy neonate without congenital anomalies of the kidney and urinary tract who developed urinary tract infection due to Staphylococcus lugdunensis, illustrating that Staphylococcus lugdunensis can cause urinary tract infection even in those with no urinary tract complications.     Rechargeable performance is realized in Mg batteries using a TiS3 cathode without the nanometer-scale downsizing of electrode particles. The specific capacity is about 80 mAh/g for the first 50 cycles at room temperature. This observed specific capacity is comparable to that of the prototype cathode for Mg batteries. First-principles calculation indicates that TiS3 is a semiconductor with d-p orbital hybridized electronic structures around the Fermi level. The reversible electrode performance is likely assisted by the delocalized electronic distribution over metal-ligand units through d-p orbital hybridization.

  6. Micro-Structural Response of DP 600 to High Strain Rate Deformation

    The object of this study was to investigate the micro-structural response of DP 600 subjected to high strain rate, ballistic impact tests. The ballistic tests were conducted using normal impact of a hardened steel penetrator into a 2 mm thick sheet of DP 600. The average strain rates produced from this test method are on the order of 10(exp 5)/s. Electron microscopy was used to investigate the microstructure before and after high strain rate deformation. A variation in material response was observed between tests conducted at 0.8 x 105 and 25 x 10(exp 5)/s.

    The detection of thermoelastic displacement by differential phase optical coherence tomography (DP-OCT) was analytically evaluated for identifying atherosclerotic plaques. Analytical solutions were developed to understand the dynamics of physical distribution of point hear sources during/after laser irradiation on thermoelastic responses of MION-injected tissue. Both analytical and experimental results demonstrated a delayed peak displacement along with slow decay after laser pulse due to heterogeneous distribution of the point heat sources. Detailed description of the heat sources in tissue as well as integration of a scanning mirror can improve computational accuracy as well as clinical applicability of DP-OCT for diagnosing vulnerable plaque.   8. Urinary Tract Endometriosis.

    Recently, occurrence of urinary tract endometriosis (UTE) is more frequently diagnosed. According to literature, it refers to approximately 0.3 to even 12% of all women with endometriosis. The pathogenesis of UTE has not been clearly explained so far. The actually proposed hypotheses include embryonic, migration, transplantation, and iatrogenic theory. Most frequently UTE affects bladder, less often ureters and kidneys. One-third of patients remains asymptomatic or exhibits only minor manifestations. In symptomatic patients main complaints include dysuria, urinary urgency, and/or frequency, painful micturition, and burning sensation in the urethra and discomfort in the retropubic area. Treatment of UTE is challenging and can be pharmacological, surgical or can be a combination of both methods.     Advances in cellular and molecular biology of extrahepatic cholangiocarcinoma and gallbladder adenocarcinoma are providing innovative means for the diagnosis and treatment of biliary tract cancer. Similarly, refinements in noninvasive studies--including helical computed tomography, magnetic resonance cholangiopancreatography, and endoscopic ultrasonography--are enabling more accurate diagnosis, staging, and treatment planning for these tumors. Complete resection remains the only means for cure, and recent reports from major hepatobiliary centers support aggressive wide resection for bile duct and gallbladder cancer. Palliation of malignant strictures has improved with advanced endoscopic techniques, newer polyurethane-covered stents, endoscopic microwave coagulation therapy, and radiofrequency intraluminal endohyperthermia. The preliminary data on such minimally invasive techniques suggest an improvement in quality of life and survival for selected patients. PMID:17031200

    Cells of the mature αβ T cell repertoire arise from the development in the thymus of bone marrow precursors (thymocytes). αβ T cell maturation is characterized by the expression of thousands of copies of identical αβ T cell receptors and the CD4 and/or CD8 co-receptors on the surface of thymocytes. The maturation stages of a thymocyte are: (1) double negative (DN) (TCR−, CD4− and CD8−), (2) double positive (DP) (TCR+, CD4+ and CD8+), and (3) single positive (SP) (TCR+, CD4+ or CD8+). Thymic antigen presenting cells provide the appropriate micro-architecture for the maturation of thymocytes, which “sense” the signaling environment via their randomly generated TCRs. Thymic development is characterized by (i) an extremely low success rate, and (ii) the selection of a functional and self-tolerant T cell repertoire. In this paper, we combine recent experimental data and mathematical modeling to study the selection events that take place in the thymus after the DN stage. The stable steady state of the model for the pre-DP, post-DP, and SP populations is identified with the experimentally measured cell counts from 5.5- to 17-week-old mice. We make use of residence times in the cortex and the medulla for the different populations, as well as recently reported asymmetric death rates for CD4 and CD8 SP thymocytes. We estimate that 65.8% of pre-DP thymocytes undergo death by neglect. In the post-DP compartment, 91.7% undergo death by negative selection, 4.7% become CD4 SP, and 3.6% become CD8 SP. Death by negative selection in the medulla removes 8.6% of CD4 SP and 32.1% of CD8 SP thymocytes. Approximately 46.3% of CD4 SP and 27% of CD8 SP thymocytes divide before dying or exiting the thymus.   13. Absence of Prenatal Forebrain Defects in the Dp(16)1Yey/+ Mouse Model of Down Syndrome

    Studies in humans with Down syndrome (DS) show that alterations in fetal brain development are followed by postnatal deficits in neuronal numbers, synaptic plasticity, and cognitive and motor function. This same progression is replicated in several mouse models of DS. Dp(16)1Yey/+ (hereafter called Dp16) is a recently developed mouse model of DS in which the entire region of mouse chromosome 16 that is homologous to human chromosome 21 has been triplicated. As such, Dp16 mice may more closely reproduce neurodevelopmental changes occurring in humans with DS. Here, we present the first comprehensive cellular and behavioral study of the Dp16 forebrain from embryonic to adult stages. Unexpectedly, our results demonstrate that Dp16 mice do not have prenatal brain defects previously reported in human fetal neocortex and in the developing forebrains of other mouse models, including microcephaly, reduced neurogenesis, and abnormal cell proliferation. Nevertheless, we found impairments in postnatal developmental milestones, fewer inhibitory forebrain neurons, and deficits in motor and cognitive performance in Dp16 mice. Therefore, although this new model does not express prenatal morphological phenotypes associated with DS, abnormalities in the postnatal period appear sufficient to produce significant cognitive deficits in Dp16. SIGNIFICANCE STATEMENT Down syndrome (DS) leads to intellectual disability. Several mouse models have increased our understanding of the neuropathology of DS and are currently being used to test therapeutic strategies. A new mouse model that contains an expanded number of DS-related genes, known as Dp(16)1Yey/+ (Dp16), has been generated recently. We sought to determine whether the extended triplication creates a better phenocopy of DS-related brain pathologies.   14. Is road safety management linked to road safety performance?

    This research aims to explore the relationship between road safety management and road safety performance at country level. For that purpose, an appropriate theoretical framework is selected, namely the 'SUNflower' pyramid, which describes road safety management systems in terms of a five-level hierarchy: (i) structure and culture, (ii) programmes and measures, (iii) 'intermediate' outcomes'--safety performance indicators (SPIs), (iv) final outcomes--fatalities and injuries, and (v) social costs. For each layer of the pyramid, a composite indicator is implemented, on the basis of data for 30 European countries. Especially as regards road safety management indicators, these are estimated on the basis of Categorical Principal Component Analysis upon the responses of a dedicated road safety management questionnaire, jointly created and dispatched by the ETSC/PIN group and the 'DaCoTA' research project. Then, quasi-Poisson models and Beta regression models are developed for linking road safety management indicators and other indicators (i.e. background characteristics, SPIs) with road safety performance. In this context, different indicators of road safety performance are explored: mortality and fatality rates, percentage reduction in fatalities over a given period, a composite indicator of road safety final outcomes, and a composite indicator of 'intermediate' outcomes (SPIs). The results of the analyses suggest that road safety management can be described on the basis of three composite indicators: "vision and strategy", "budget, evaluation and reporting", and "measurement of road user attitudes and behaviours". Moreover, no direct statistical relationship could be established between road safety management indicators and final outcomes. However, a statistical relationship was found between road safety management and 'intermediate' outcomes, which were in turn found to affect 'final' outcomes, confirming the SUNflower approach on the consecutive effect of each layer

    A Tunnel Traffic & Operation Simulator is a device of the Centre of Transport Research at the University of Žilina. The Simulator allows managing technological equipment of virtual two-tube highway tunnel, which is interconnected with simulation of vehicle traffic in tunnel. Changes of the traffic-operation states and other equipment are reflecting at the simulated traffic, as well as simulations of various emergency events in traffic initiate changes in tunnel detecting and measuring devices. It is thus possible to simulate emergency states, which can be affected by various faults of technology as well as by climatic conditions. The solutions can be found in irreplaceable experiences of Slovak road tunnel operators, changes of trafficoperation states, visualizations of operator technological display screens, technological devices labelling in order to increase operational safety of road tunnels.

    Road traffic is the most interfering noise source in developed countries. According to a publication of the European Union (EU) at the end of the twentieth century [1], about 40% of the population in 15 EU member states is exposed to road traffic noise at mean levels exceeding 55 dB(A). Nearly 80 million people, 20% of the population, are exposed to levels exceeding 65 dB(A) during daytime and more than 30% of the population is exposed to levels exceeding 55 dB(A) during night time. Such high noise levels cause health risks and social disorders (aggressiveness, protest, and helplessness), interference of communication and disturbance of sleep; the long- and short-term consequences cause adverse cardiovascular effects, detrimental hormonal responses (stress hormones), and possible disturbance of the human metabolism (nutrition) and the immune system. Even performance at work and school could be impaired.

    Granular surfaces to develop lateral ripples (so-called ``washboard" or ``corrugated" road) under the action of rolling wheels. Similar ripples are observed on railroad tracks and many other rolling, load bearing surfaces. Our aim was to investigate this instability of the flat road surface from the point of view of driven, dissipative granular dynamics. We report the results of both laboratory experiments and soft-particle direct numerical simulations. The experiment consisted of a rotating table 60 cm in radius with a thick layer of sand forming a roadbed around the circumference. A 6 cm radius hard rubber wheel, with a support stationary in the lab frame, rolled on the sand layer. We varied the speed of the table, the details of the grains and the suspension of the wheel. The ripple pattern appears as small patches of travelling waves which eventually spread to the entire circumference. The ripples drift slowly in the driving direction. Interesting secondary dynamics of the saturated ripples were observed, as well as various ripple creation and destruction events. The wavelength of the ripples can be quantized by the finite circumference of the road. All of these effects are captured qualitatively by 2D soft particle simulations in which a disk rolls over a 2D bed of polydisperse particles in a periodic box.

    The pervasiveness of mobile devices, which is increasing daily, is generating a vast amount of geo-located data allowing us to gain further insights into human behaviors. In particular, this new technology enables users to communicate through mobile social media applications, such as Twitter, anytime and anywhere. Thus, geo-located tweets offer the possibility to carry out in-depth studies on human mobility. In this paper, we study the use of Twitter in transportation by identifying tweets posted from roads and rails in Europe between September 2012 and November 2013. We compute the percentage of highway and railway segments covered by tweets in 39 countries. The coverages are very different from country to country and their variability can be partially explained by differences in Twitter penetration rates. Still, some of these differences might be related to cultural factors regarding mobility habits and interacting socially online. Analyzing particular road sectors, our results show a positive correlation between the number of tweets on the road and the Average Annual Daily Traffic on highways in France and in the UK. Transport modality can be studied with these data as well, for which we discover very heterogeneous usage patterns across the continent. PMID:25141161

  1. New Insights on the Reaction Pathway Leading to Lactyl-ThDP: A Theoretical Approach.

    In all ThDP-dependent enzymes, the catalytic cycle is initiated with the attack of the C2 atom of the ylide intermediate on the Cα atom of a pyruvate molecule to form the lactyl-ThDP (L-ThDP) intermediate. In this study, the reaction between the ylide intermediate and pyruvate leading to the formation of L-ThDP is addressed from a theoretical point of view. The study includes molecular dynamics, exploration of the potential energy surface by means of QM/MM calculations, and reactivity analysis on key centers. The results show that the reaction occurs via a concerted mechanism in which the carboligation and the proton transfers occur synchronically. It is also observed that during the reaction the protonation state of the N1' atom changes: the reaction starts with the ylide having the N1' atom deprotonated and reaches a transition state showing the N1' atom protonated. This conversion leads to the reaction path of minimum energy, with an activation energy of about 20 kcal mol(-1). On the other hand, it is also observed that the approaching distance between the pyruvate and the ylide, i.e., the Cα-C2 distance, plays a fundamental role in the reaction mechanism since it determines the nucleophilic character of key atoms of the ylide, which in turn trigger the elemental reactions of the mechanism.     The chelator di-2-pyridylketone 4,4-dimethyl-3-thiosemicarbazone (Dp44mT) shows potent and selective anticancer and antimetastatic activity. However, the mechanism by which it is initially transported into cells to induce cytotoxicity is unknown. Hence, the current investigation examined the cellular uptake of ¹⁴C-Dp44mT relative to two structurally related ligands, namely the aroylhydrazone ¹⁴C-pyridoxal isonicotinoyl hydrazone (¹⁴C-PIH) and the thiosemicarbazone (¹⁴C-2-benzoylpyridine 4-ethyl-3-thiosemicarbazone (¹⁴C-Bp4eT). In marked contrast to the cellular uptake of ¹⁴C-PIH and ¹⁴C-Bp4eT, which were linear as a function of concentration, ¹⁴C-Dp44mT uptake was saturable using SK-N-MC neuroepithelioma cells (Bmax, 4.28 × 10⁷ molecules of chelator/cell; and Kd, 2.45 μM). Together with the fact that ¹⁴C-Dp44mT uptake was temperature-dependent and significantly (P < 0.01) decreased by competing unlabeled Dp44mT, these observations indicated a saturable transport mechanism consistent with carrier/receptor-mediated transport. Other unlabeled ligands that shared the saturated N4 structural moiety with Dp44mT significantly (P < 0.01) inhibited ¹⁴C-Dp44mT uptake, illustrating its importance for carrier/receptor recognition. Nevertheless, unlabeled Dp44mT most markedly decreased (¹⁴C-Dp44mT uptake, demonstrating that the putative carrier/receptor shows high selectivity for Dp44mT. Interestingly, in contrast to ¹⁴C-Dp44mT, uptake of its Fe complex [Fe(¹⁴C-Dp44mT)₂] was not saturable as a function of concentration and was much greater than the ligand alone, indicating an alternate mode of transport. Studies examining the tissue distribution of ¹⁴C-Dp44mT injected intravenously into a mouse tumor model demonstrated the ¹⁴C label was primarily identified in the excretory system.     Road traffic injuries affect the economy, health and quality of life of the people of Mozambique. Current road safety programmes are inadequate and inefficient given the magnitude of the problem. Data reported on road traffic crashes in the period 1990 to 2000 from the National Institute for Road Safety, the traffic police and the Central Hospital of Maputo were reviewed. The burden of road traffic injuries in Mozambique is rising, with at least three people killed daily. The age group most affected is 25-38 (39.35%), followed by 16-24 (20.79%). The main causes of crashes include reckless driving, drunken driving, roads with potholes, inadequate signs, lack of protection for pedestrians, and inadequate traffic law enforcement. However, the data are not adequate to reveal the true magnitude of the problem. Data collected by different sources are incomplete and not coordinated with other sources and databases. In urban areas, however, better response to crashes, treatment of the injured, reporting and data collection is attributable to a greater concentration of police and medical facilities. Road traffic safety programmes in Mozambique are inadequate and inefficient, starting with the data collection system. Improvement of injury surveillance systems is needed to help make road traffic safety a national development agenda priority and for developing and implementing road safety policies. For road safety programmes to be effective, government must facilitate stakeholders' involvement, and the clear definition of government activities, civil society activities and public-private partnerships need to be established.     Technological advances in the domain of digital signal processing adapted to cochlear implants (CI) are partially responsible for the ever-improving outcomes observed with this neural prosthesis. The goal of the present study was to evaluate audiometric outcomes with a new signal processing strategy implemented in Oticon Medical-Neurelec cochlear implant systems, the xDP strategy. The core of this approach is a preset-based back-end output compression system, modulating a multi-channel transfer function depending on the intensity and information content of input sounds. Twenty adult CI patients, matched for age and CI experience, were included in this study. Pure-tone thresholds and vocal audiometry scores were measured with their former signal processing strategy and with xDP. Speech perception was assessed using dissyllabic words presented in quiet, at different intensity levels: 40, 55, 70, and 85 dB SPL, and in a cocktail party noise at a signal-to-noise ratio of +10 dB. Results with the xDP strategy showed, as awaited, no major modification of pure-tone thresholds. A global increase of speech perception scores was observed after a 1-month habituation period, with significant improvements for speech perception in quiet for moderate (55 dB SPL), loud speech sounds (85 dB SPL), and speech-in-noise comprehension. Subjective signal quality assessment showed a preference for Crystalis(xDP) over the former strategy. These results allow the quantification of improvements provided by the xDP signal processing strategy. PMID:26476927

  4. Effects on road safety of new urban arterial roads.

    This paper presents an evaluation of the effects on road safety of new urban arterial roads in Oslo, Norway, and a synthesis of evidence from similar studies that have evaluated the safety effects of new urban arterial roads in other cities. A before-and-after study was made of four urban arterial road projects in Oslo. The study controlled for general accident trends in Oslo and for regression-to-the-mean. A statistically non-significant reduction of 9% in the number of injury accidents was found for all four projects combined. The effects on safety of new urban arterial roads were found to vary, depending on whether a new arterial road was built, or an existing arterial road upgraded by means of lane additions and reconstruction of junctions to interchanges. New arterial roads tend to induce more traffic, which tends to offset the benefits of a lower accident rate on the new roads. The results for other cities are very consistent with those for Oslo. For a total of seven cases in which new arterial roads were built, a statistically non-significant reduction of 1% in the number of injury accidents was found. Two cases that involved lane additions and converting at-grade junctions to interchanges resulted in a mean accident reduction of 51%, which was highly significant. On the average, the nine arterial road projects from which evidence was summarised resulted in a net induced traffic of 16%, and a net reduction in accident rate (accidents per million vehicle kilometres) of 18%. These effects almost cancel each other, leading to a very small net change in the expected number of accidents. PMID:14572833

  5. Road traffic injuries in Colombia.

    Road traffic injuries are a leading public health problem in Colombia. Pedestrians are the most vulnerable road users, especially in the main urban centers of Bogotá, Medellin and Cali. Data analyzed in this report include official statistics from the National Police and the National Institute of Legal Medicine and Forensic Sciences for 1996-2000, and results of a study conducted at the National University of Colombia in 2000. Methods from the Highway Capacity Manual were used for determining physical and technical variables, and a Geographical Information System tool was used for the location and spatial analysis of the road traffic crashes. Pedestrians accounted for close to 32% of injuries and 40% of the deaths from road traffic crashes. The problem of road traffic crashes existed predominately in urban areas. In the main urban centers, pedestrians constituted nearly 68% of road traffic crash victims. The high level of risky road use behaviors demonstrated by pedestrians and drivers, and inadequate infrastructure for safe mobility of pedestrians in some sections of the road network were the main contributing factors. Major improvements were achieved in Bogotá following enhancements to the municipal transport system and other policies introduced since 1995. In conclusion, policies and programs for improving road safety, in particular pedestrian safety, and strengthening urban planning are top priority. PMID:12772483

    Road traffic injuries are already the leading cause of injury mortality and morbidity globally and by 2030 are predicted to be the fifth leading cause of mortality in the world. Australia has seen a dramatic reduction in road deaths and serious injuries since the 1970s and holds an international reputation for road traffic injury prevention due, in part, to its success in pioneering the multidisciplinary and intersectoral approach needed to address this significant issue and by applying an evidence-led approach to policy development. The paper will discuss Australia's early success in road traffic injury prevention (road safety), particularly the achievements following the implementation of targeted programs that focussed on road user behaviours for which health promotion played a role. The most successful of these programs was the introduction of comprehensive seat belt laws, random breath testing and more recently, strategic speed enforcement programs. Amid an array of significant challenges faced by the transport system in the future, the rapid development in information and communication technologies applied to transport is likely to provide the next generation of road safety benefits. The potential for a semi-autonomous transport system is likely to provide the next significant decline in road fatalities and serious injuries over the next 2 decades and the role of health promotion in relation to raising community engagement and building coalitions to increase uptake of new technologies will be discussed.     Dystrophin Dp71 is the most ubiquitous and highest expressed dystrophin isoform in brain, however, Dp71 function and those specific for its spliced d- and ab- isoforms remains undetermined. To study Dp71, utrophin and β-dystroglycan in cell-to-cell interactions, we first established a co-culture model using PC 12 cells and L6 myotubes. Confocal microscopy assays of these co-cultures, in which PC 12 cells are differentiated in the presence of L6 myotubes, showed that the Dp71d isoform accumulates in PC 12 nuclei, Golgi-complex- and endoplasmic reticulum-like structures, being depleted from neurites and cytoplasm, while Dp71ab concentrates at neurite tips and cytoplasm and colocalizes with β-dystroglycan, utrophin, synaptophysin and acetylcholine receptors. Evidences suggest Dp71ab isoform unlike Dp71d, may take part in neurite-related processes. This is the first work on the role of dystrophins as well as members of the DAP complex in a cell-line based co-culturing system, which may prove useful in determining protein associations in a more controlled environment than ex-vivo systems.     Although we previously demonstrated the contribution of the DP1receptor in nasal obstruction using animals sensitized with ovalbumin in the presence of adjuvant, the contribution of the DP1receptor in sneezing is unclear. Here, we developed a mouse model of Japanese cedar (JC:Cryptomeria japonica) pollinosis to evaluate the symptoms of sneezing. To achieve this, we used JC pollen crude extract in the absence of adjuvant to sensitize mice to develop a model closer to the pathophysiology of human JC pollinosis. The immunologic and pharmacologic features of this model are highly similar to those observed in JC pollinosis in humans. Using this model, we found that DP1receptor antagonists suppressed JC pollen extract-induced sneezing and that a DP1receptor agonist induced sneezing. Moreover, JC pollen extract-induced sneezing was diminished in DP1receptor knockout mice. In conclusion, we developed a novel mouse model of allergic rhinitis that closely mimics human JC pollinosis. A strong contribution of DP1receptor signaling to sneezing was demonstrated using this model, suggesting that DP1receptor antagonists could suppress sneezing and nasal obstruction, and therefore these agents could be a new therapeutic option for allergic rhinitis.   11. Clinical pharmacology of DP-b99 in healthy volunteers: First administration to humans

    Aims To investigate the safety, tolerability and pharmacokinetics of DP-b99 in healthy volunteers. DP-b99 is a newly developed lipophilic, cell permeable derivative of BAPTA (1,2-bis(2-aminophenoxy)ethane-N,N,N′,N′-tetraacetic acid), which selectively modulates the distribution of metal ions in hydrophobic milieu, and is in clinical development as a neuroprotectant for cerebral ischaemic stroke. To our knowledge no BAPTA derivative has ever been administered to man. Here we report the first human administration of DP-b99 in a phase I, two-part, double-blind, randomized placebo controlled study, with single IV doses of 0.003–1.0 mg kg−1 day−1 DP-b99 (part 1) or multiple ascending doses of 0.03–1.0 mg kg−1 day−1 DP-b99 over 4 days (part 2). Methods A double-blind, dose escalating tolerability study of DP-b99 in normal (young – aged between 18 and 40 years and elderly – aged between 65 and 85 years) healthy adult male volunteers was conducted. Part 1 of the study investigated single administration of ascending intravenous doses, and part 2 examined the effects of ascending doses given repeatedly over 4 days. Twenty-four young volunteers in part 1 received single dose administrations and 26 young volunteers in part 2 received repeated ascending dose administrations of either intravenous DP-b99 or placebo. Subsequently, 10 elderly volunteers received repeated intravenous DP-b99 (1 mg kg−1) or placebo in part 2 over 4 days. Adverse events were identified by either subject self reporting or based upon laboratory parameters (blood chemistry, complete blood cell count, prothrombin time (PT), activated partial thromboplastin (PTT), physical examination, vital signs (blood pressure, pulse rate, respiratory rate, body temperature) and urinalysis.     The cardiovascular complications reported to be associated with cyclooxygenase inhibitor use have shifted our focus toward prostaglandins and their respective receptors. Prostaglandin D2 and its DP1 receptor have been implicated in various normal and pathologic conditions, but their role in stroke is still poorly defined. Here, we tested whether DP1 deletion aggravates N-methyl-d-aspartic acid (NMDA)-induced acute toxicity and whether DP1 pharmacologic activation protects mice from acute excitotoxicity and transient cerebral ischemia. Moreover, since the elderly are more vulnerable to stroke-related damage than are younger patients, we tested the susceptibility of aged DP1 knockout (DP1−/−) mice to brain damage. We found that intrastriatal injection of 15 nmol NMDA caused significantly larger lesion volumes (27.2 ± 6.4%) in young adult DP1−/− mice than in their wild-type counterparts. Additionally, intracerebroventricular pretreatment of wild-type mice with 10, 25, and 50 nmol of the DP1-selective agonist BW245C significantly attenuated the NMDA-induced lesion size by 19.5 ± 5.0%, 39.6 ± 7.7%, and 28.9 ± 7.0%, respectively. The lowest tested dose of BW245C also was able to reduce middle cerebral artery occlusion-induced brain infarction size significantly (21.0 ± 5.7%). Interestingly, the aggravated NMDA-induced brain damage was persistent in older DP1−/− mice as well. We conclude that the DP1 receptor plays an important role in attenuating brain damage and that selective targeting of this receptor could be considered as an adjunct therapeutic tool to minimize stroke damage.   13. Correlations between Nanoindentation Hardness and Macroscopic Mechanical Properties in DP980 Steels

    Multiphase advanced high strength steels (AHSS) are being increasingly used in the automotive industry due to their low cost, good availability and excellent combination of strength and ductility. There is a keen interest from the automotive and steel industry for more fundamental understandings on the key microstructure features influencing the macroscopic properties, i.e., tensile properties, hole-expansion ratio and localized formability of AHSS. In this study, the micro- and macro-level properties for eight commercial DP980 steels are first characterized and quantified with various experimental methods. Correlations between macroscopic-level properties and relationships between various micro- and macro- properties for these steels are then established based on the experimental measurements. It is found that, despite their differences in their chemistry, processing parameters and sheet thickness, the eight DP980 steels do have common microstructural level properties governing their specific macroscopic properties in terms of strength, elongation and hole expansion performance.

  14. Phase Identification of Dual-Phase (DP980) Steels by Electron Backscatter Diffraction and Nanoindentation Techniques.

    Phase identification of multi-phase materials provides essential information relating the material to its mechanical properties. In this study we selected DP980, a type of dual-phase steel, to investigate the content of martensite and ferrite grains. A combination of advanced techniques was used to provide detailed and precise information of the microstructure. Scanning and transmission electron microscopy were used to provide observations of the sample surface at different scales. Martensite and ferrite phases of DP980 were further identified and characterized using electron backscatter diffraction and scanning probe microscopy. Results obtained with nanoindentation tests confirmed that the differences in nanohardness values in single-phase grains are martensite and ferrite with different surface heights shown by scanning probe microscopy. The similarity shown in the image quality map and scanning probe microscopy proves that a large fraction of martensite can be distinguished in this undeformed material using image quality parameters obtained during electron backscatter diffraction imaging.     Level energies and spectroscopic information for neutron-rich nuclei provide important input for r-process nucleosynthesis calculations; specifically, the location and strength of single-neutron l = 1 states when calculating neutron-capture rates. Surman and collaborators have performed sensitivity studies to show that varying neutron-capture rates can significantly alter final r-process abundances. However, there are many nuclei important to the r-process that cannot be studied. Extending studies to more neutron-rich nuclei will help constrain the nuclear shell-model in extrapolating to nuclei even further from stability. The (d,p) reaction has been measured with radioactive ion beams of 126Sn and 128Sn to complete the set of (d,p) studies on even mass tin isotopes from doubly-magic 132 to stable 124Sn.     The parent action method is utilized to the Born-Infeld and Dp-brane theories. Various new forms of Born-Infeld and Dp-brane actions are derived by using this systematic approach, in which both the already known 2-metric and newly proposed 3-metric prescriptions are considered. An auxiliary worldvolume tensor field, denoted by {omega}{sub {mu}}{sub {nu}}, is introduced and treated probably as an additional worldvolume metric because it plays a similar role to that of the auxiliary worldvolume (also called intrinsic) metric {gamma}{sub {mu}}{sub {nu}}. Some properties, such as duality, permutation and Weyl invariance as a local worldvolume symmetry of the new forms are analyzed. In particular, a new symmetry, i.e. the double Weyl invariance is discovered in 3-metric forms.

    This paper focuses on the research of a semi-submersible platform equipped with a DP-assisted mooring system. Based on the working principles of the DP-assisted mooring system and the model of the platform motion, a time domain simulation program is applied to analyze the impact, in the case of one line failure, on the platform motion, power consumption of the thrusters and the tension of the mooring lines. The results show that, under the 10-year wind dominant, a one line failure will have little impact on the tension of the mooring lines. When the failure line is windward, the power consumption will increase greatly with a weakened position of accuracy. However when the failure line is leeward, the power consumption will be reduced with a partly strengthened position of accuracy.

    The purpose of this effort was to advance the selection, characterization, and modeling of a propulsion electric grid for a Turboelectric Distributed Propulsion (TeDP) system for transport aircraft. The TeDP aircraft would constitute a miniature electric grid with 50 MW or more of total power, two or more generators, redundant transmission lines, and multiple electric motors driving propulsion fans. The study proposed power system architectures, investigated electromechanical and solid state circuit breakers, estimated the impact of the system voltage on system mass, and recommended DC bus voltage range. The study assumed an all cryogenic power system. Detailed assumptions within the study include hybrid circuit breakers, a two cryogen system, and supercritical cyrogens. A dynamic model was developed to investigate control and parameter selection.

    Purpose Dp71 is the main product of the Duchenne muscular dystrophy (DMD) gene in the central nervous system. While studying the impact of its absence on retinal functions, we discovered that mice lacking Dp71 also developed a progressive opacification of the crystalline lens. The purpose of this study was to perform a detailed characterization of the cataract formation in Dp71 knockout (KO-Dp71) mice. Methods Cataract formations in KO-Dp71 mice and wild-type (wt) littermates were assessed in vivo by slit-lamp examination and ex vivo by histological analysis as a function of aging. The expression and cellular localization of the DMD gene products were monitored by western blot and immunohistochemical analysis. Fiber cell integrity was assessed by analyzing the actin cytoskeleton as well as the expression of aquaporin-0 (AQP0). Results As expected, a slit-lamp examination revealed that only one of the 20 tested wt animals presented with a mild opacification of the lens and only at the most advanced age. However, a lack of Dp71 was associated with a 40% incidence of cataracts as early as 2 months of age, which progressively increased to full penetrance by 7 months. A subsequent histological analysis revealed an alteration in the structures of the lenses of KO-Dp71 mice that correlated with the severity of the lens opacity. An analysis of the expression of the different dystrophin gene products revealed that Dp71 was the major DMD gene product expressed in the lens, especially in fiber cells. The role of Dp71 in fiber cells was also suggested by the progressive disorganization of the lens fibers, which was observed in the absence of Dp71 and demonstrated by irregular staining of the actin network and the aqueous channel AQP0. Conclusions While its role in the retina has been well characterized, this study demonstrates for the first time the role played by Dp71 in a different ocular tissue: the crystalline lens.     We show how to estimate accurately the Jones matrix of the transmission line by analyzing the Stokes vectors of DP-QPSK signals. This method can be used to perform in-situ PMD measurement in dual-polarization QPSK systems, and in addition to the constant modulus algorithm (CMA) to mitigate polarization-induced impairments. The applicability of this method to other modulation formats is discussed.

  8. EnzDP: improved enzyme annotation for metabolic network reconstruction based on domain composition profiles.

    Determining the entire complement of enzymes and their enzymatic functions is a fundamental step for reconstructing the metabolic network of cells. High quality enzyme annotation helps in enhancing metabolic networks reconstructed from the genome, especially by reducing gaps and increasing the enzyme coverage. Currently, structure-based and network-based approaches can only cover a limited number of enzyme families, and the accuracy of homology-based approaches can be further improved. Bottom-up homology-based approach improves the coverage by rebuilding Hidden Markov Model (HMM) profiles for all known enzymes. However, its clustering procedure relies firmly on BLAST similarity score, ignoring protein domains/patterns, and is sensitive to changes in cut-off thresholds. Here, we use functional domain architecture to score the association between domain families and enzyme families (Domain-Enzyme Association Scoring, DEAS). The DEAS score is used to calculate the similarity between proteins, which is then used in clustering procedure, instead of using sequence similarity score. We improve the enzyme annotation protocol using a stringent classification procedure, and by choosing optimal threshold settings and checking for active sites. Our analysis shows that our stringent protocol EnzDP can cover up to 90% of enzyme families available in Swiss-Prot. It achieves a high accuracy of 94.5% based on five-fold cross-validation. EnzDP outperforms existing methods across several testing scenarios. Thus, EnzDP serves as a reliable automated tool for enzyme annotation and metabolic network reconstruction.   9. Scale invariance in road networks

    We study the topological and geographic structure of the national road networks of the United States, England, and Denmark. By transforming these networks into their dual representation, where roads are vertices and an edge connects two vertices if the corresponding roads ever intersect, we show that they exhibit both topological and geographic scale invariance. That is, we show that for sufficiently large geographic areas, the dual degree distribution follows a power law with exponent 2.2⩽α⩽2.4 , and that journeys, regardless of their length, have a largely identical structure. To explain these properties, we introduce and analyze a simple fractal model of road placement that reproduces the observed structure, and suggests a testable connection between the scaling exponent α and the fractal dimensions governing the placement of roads and intersections.

    Purpose: We carried out this study to find out the relevance between rs2281388 T/C polymorphism of human leukocyte antigen (HLA) gene and hepatocellular carcinoma (HCC) risk in Chinese Han population. Methods: The method of polymerase chain reaction (PCR) was applied to amplify the genomic DNA. Then the PCR products were sequenced to test the HLA-DP gene rs2281388T/C polymorphism of the case and control groups. Odds ratios (ORs) and 95% confidence interval (95% CIs) were utilized to evaluate the potential correlation between rs2281388 variants and HCC risk. Results: We analyzed the rs2281388 polymorphism distribution among the clinical pathological features. The results showed that there existed a significant statistic correlation between rs2281388T/C polymorphism of HLA-DP gene and HBsAg feature, and no significant correlation was found between rs2281388 and other clinical features. Further analysis showed that the TT genotype of rs2281388 was significantly correlated with HCC risk, and the same to T allele, but there was no significant difference of CT genotype distribution in case and control groups. Conclusion: TT genotype and T allele of HLA-DP gene rs2281388 polymorphism may increase the risk of HCC. PMID:26261648

  11. Road safety performance indicators for the interurban road network.

    Various road safety performance indicators (SPIs) have been proposed for different road safety research areas, mainly as regards driver behaviour (e.g. seat belt use, alcohol, drugs, etc.) and vehicles (e.g. passive safety); however, no SPIs for the road network and design have been developed. The objective of this research is the development of an SPI for the road network, to be used as a benchmark for cross-region comparisons. The developed SPI essentially makes a comparison of the existing road network to the theoretically required one, defined as one which meets some minimum requirements with respect to road safety. This paper presents a theoretical concept for the determination of this SPI as well as a translation of this theory into a practical method. Also, the method is applied in a number of pilot countries namely the Netherlands, Portugal, Greece and Israel. The results show that the SPI could be efficiently calculated in all countries, despite some differences in the data sources. In general, the calculated overall SPI scores were realistic and ranged from 81 to 94%, with the exception of Greece where the SPI was relatively lower (67%). However, the SPI should be considered as a first attempt to determine the safety level of the road network. The proposed method has some limitations and could be further improved.   12. Potentiating the cellular targeting and anti-tumor activity of Dp44mT via binding to human serum albumin: two saturable mechanisms of Dp44mT uptake by cells

    Di-2-pyridylketone 4,4-dimethyl-3-thiosemicarbazone (Dp44mT) demonstrates potent anti-cancer activity. We previously demonstrated that 14C-Dp44mT enters and targets cells through a carrier/receptor-mediated uptake process. Despite structural similarity, 2-benzoylpyridine 4-ethyl-3-thiosemicarbazone (Bp4eT) and pyridoxal isonicotinoyl hydrazone (PIH) enter cells via passive diffusion. Considering albumin alters the uptake of many drugs, we examined the effect of human serum albumin (HSA) on the cellular uptake of Dp44mT, Bp4eT and PIH. Chelator-HSA binding studies demonstrated the following order of relative affinity: Bp4eT≈PIH>Dp44mT. Interestingly, HSA decreased Bp4eT and PIH uptake, potentially due to its high affinity for the ligands. In contrast, HSA markedly stimulated Dp44mT uptake by cells, with two saturable uptake mechanisms identified. The first mechanism saturated at 5-10 μM (Bmax:1.20±0.04 × 107 molecules/cell; Kd:33±3 μM) and was consistent with a previously identified Dp44mT receptor/carrier. The second mechanism was of lower affinity, but higher capacity (Bmax:2.90±0.12 × 107 molecules/cell; Kd:65±6 μM), becoming saturated at 100 μM and was only evident in the presence of HSA. This second saturable Dp44mT uptake process was inhibited by excess HSA and had characteristics suggesting it was mediated by a specific binding site. Significantly, the HSA-mediated increase in the targeting of Dp44mT to cancer cells potentiated apoptosis and could be important for enhancing efficacy. PMID:25848850

    The transmission performance of coherent dual-polarization multi-band OFDM (DP-MB-OFDM) and QPSK (DP-QPSK) are experimentally compared for 100 Gb/s long-haul transport over legacy infrastructure combining G.652 fiber and 10 Gb/s WDM system. It is shown that DP-MB-OFDM and DP-QPSK have nearly the same performance at 100 Gb/s after transmission over a 10 × 100-km fiber line. Furthermore, the origin of performance degradations and limitations of the DP-MB-OFDM is explored numerically, as well as the impact of transmission distance and sub-band spacing. PMID:23938547

    Background Fast food restaurants reportedly target specific populations by locating in lower-income and in minority neighborhoods. Physical proximity to fast food restaurants has been associated with higher obesity rates. Objective To examine possible associations, at the census tract level, between area demographics, arterial road density, and fast food restaurant density in King County, WA, USA. Methods Data on median household incomes, property values, and race/ethnicity were obtained from King County and from US Census data. Fast food restaurant addresses were obtained from Public Health-Seattle & King County and were geocoded. Fast food density was expressed per tract unit area and per capita. Arterial road density was a measure of vehicular and pedestrian access. Multivariate logistic regression models containing both socioeconomic status and road density were used in data analyses. Results Over one half (53.1%) of King County census tracts had at least one fast food restaurant. Mean network distance from dwelling units to a fast food restaurant countywide was 1.40 km, and 1.07 km for census tracts containing at least one fast food restaurant. Fast food restaurant density was significantly associated in regression models with low median household income (p < 0.001) and high arterial road density (p < 0.001) but not with percent of residents who were nonwhite. Conclusion No significant association was observed between census tract minority status and fast food density in King County. Although restaurant density was linked to low household incomes, that effect was attenuated by arterial road density. Fast food restaurants in King County are more likely to be located in lower income neighborhoods and higher traffic areas. PMID:19630979

    A program to map the locations and frequencies of DNA tracts composed of only two bases ('Binary DNA') is described. The program, TRACTS (URL http://bioportal.weizmann.ac.il/tracts/tracts.html and/or http://bip.weizmann.ac.il/miwbin/servers/tracts) is of interest because long tracts composed of only two bases are highly over-represented in most genomes. In eukaryotes, oligopurine.oligopyrimidine tracts ('R.Y tracts') are found in the highest excess. In prokaryotes, W tracts predominate (A,T 'rich'). A pre-program, ANEX, parses database annotation files of GenBank and EMBL, to produce a convenient one-line list of every gene (exon, intron) in a genome. The main unit lists and analyzes tracts of the three possible binary pairs (R.Y, K.M and S;W). As an example, the results of R.Y tract mapping of mammalian gene p53 is described. PMID:12824393

    On average, there are over 5.8 M vehicle crashes each year of which 23% are weather-related. Weather-related crashes are defined as those crashes that occur in adverse weather or on slick pavement. The vast majority of weather-related crashes happen on wet pavement (74%) and during rainfall (46%). Connected vehicle technologies hold the promise to transform road-weather management by providing improved road weather data in real time with greater temporal and geographic accuracy. This will dramatically expand the amount of data that can be used to assess, forecast, and address the impacts that weather has on roads, vehicles, and travelers. The use of vehicle-based measurements of the road and surrounding atmosphere with other, more traditional weather data sources, and create road and atmospheric hazard products for a variety of users. The broad availability of road weather data from mobile sources will vastly improve the ability to detect and forecast weather and road conditions, and will provide the capability to manage road-weather response on specific roadway links. The RWMP is currently demonstrating how weather, road conditions, and related vehicle data can be used for decision making through an innovative Integrated Mobile Observations project. FHWA is partnering with 3 DOTs (MN, MI, & NV) to pilot these applications. One is a mobile alerts application called the Motorists Advisories and Warnings (MAW) and a maintenance decision support application. These applications blend traditional weather information (e.g., radar, surface stations) with mobile vehicle data (e.g., temperature, brake status, wiper status) to determine current weather conditions. These weather conditions, and other road-travel-relevant information, are provided to users via web and phone applications.     Traditional inspections of road surfaces for the condition assessment and for locating cracks are time-consuming, expensive and can prove to be dangerous. What is ideally required would be a fully equipped automated inspecting vehicle capable of high precision location and characterization of road surface cracks over the width of the road (single pass). We propose an automatic crack monitoring system (akin to HARRIS - UK) with the video-based subsystem substituted by Global Positioning Systems for more accurate positioning. Besides, our technique avoids the storage of large volumes of scanned images of 'acceptable' road surface conditions. A pulse coupled neural network (PCNN) is used as a preprocessor for each scanned image to detect cracks while another PCNN segments this image to characterize identified defects. The latter image is then stored as binary image along with the GPS data. The type of cracks is later identified (offline) from the recorded binary images. This mode of data collection leads to a more accurate, less costly and faster automated system. Our results for road surface (concrete and bituminous) images reveal the suitability of this novel technique for a fully automated road inspection system for crack identification and characterization.

    Duchenne muscular dystrophy (DMD) causes cognitive impairment in one third of the patients, although the underlying mechanisms remain to be elucidated. Recent studies showed that mutations in the distal part of the dystrophin gene correlate well with the cognitive impairment in DMD patients, which is attributed to Dp71. The study on the expression of the shortest isoform, Dp40, has not been possible due to the lack of an isoform specific antibody. Dp40 has the same promoter as that found in Dp71 and lacks the normal C-terminal end of Dp427. In the present study, we have raised polyclonal antibody against the N-terminal sequence common to short isoforms of dystrophin, including Dp40, and investigated the expression pattern of Dp40 in the mouse brain. Affinity chromatography with this antibody and the consecutive LC-MS/MS analysis on the interacting proteins revealed that Dp40 was abundantly expressed in synaptic vesicles and interacted with a group of presynaptic proteins, including syntaxin1A and SNAP25, which are involved in exocytosis of synaptic vesicles in neurons. We thus suggest that Dp40 may form a novel protein complex and play a crucial role in presynaptic function. Further studies on these aspects of Dp40 function might provide more insight into the molecular mechanisms of cognitive impairment found in patients with DMD. PMID:22258561

  1. Analysis of the Interaction of Dp44mT with Human Serum Albumin and Calf Thymus DNA Using Molecular Docking and Spectroscopic Techniques

    Di-2-pyridylketone-4,4,-dimethyl-3-thiosemicarbazone (Dp44mT) exhibits significant antitumor activity. However, the mechanism of its pharmacological interaction with human serum albumin (HSA) and DNA remains poorly understood. Here, we aimed to elucidate the interactions of Dp44mT with HSA and DNA using MTT assays, spectroscopic methods, and molecular docking analysis. Our results indicated that addition of HSA at a ratio of 1:1 did not alter the cytotoxicity of Dp44mT, but did affect the cytotoxicity of the Dp44mT-Cu complex. Data from fluorescence quenching and UV-VIS absorbance measurements demonstrated that Dp44mT could bind to HSA with a moderate affinity (Ka = approximately 104 M−1). CD spectra revealed that Dp44mT could slightly disrupt the secondary structure of HSA. Dp44mT could also interact with Ct-DNA, but had a moderate binding constant (KEB = approximately 104 M−1). Docking studies indicated that the IB site of HSA, but not the IIA and IIIA sites, could be favorable for Dp44mT and that binding of Dp44mT to HSA involved hydrogen bonds and hydrophobic force, consistent with thermodynamic results from spectral investigations. Thus, the moderate binding affinity of Dp44mT with HSA and DNA partially contributed to its antitumor activity and may be preferable in drug design approaches. PMID:27376275

    Dipeptidyl peptidase IV (DP-IV; dipeptidyl-peptide hydrolase, EC 3.4.14.5) is a serine protease with a specificity for cleaving Xaa-Pro dipeptides from polypeptides and proteins. It is found in a variety of mammalian cells and tissues, including those of lymphoid origin where it is found specifically on the surface of CD4+ T cells. Although the functional significance of this enzyme has not been established, a role in T-cell activation and immune regulation has been proposed. Here we report that Ala-boroPro and Pro-boroPro, where boroPro is the alpha-amino boronic acid analog of proline, are potent and specific inhibitors of DP-IV, having Ki values in the nanomolar range. Blocking the N terminus of Ala-boroPro abolishes the affinity of this inhibitor for DP-IV, while removal of the N-terminal residue, to give boroPro, reduces the affinity for DP-IV by 5 orders of magnitude. The dipeptide boronic acids exhibit slow-binding kinetics, while boroPro does not. We also report here that low concentrations of Pro-boroPro inhibit antigen-induced proliferation and interleukin 2 production in murine T-cell lines but do not inhibit the response of these T cells to the mitogen concanavalin A. These results indicate that DP-IV plays a role in antigen-induced, but not mitogen-induced, activation of T lymphocytes. PMID:1671716

    Objectives: To identify the changing trends and crucial preventive approaches to road traffic accidents (RTAs) adopted in the Kingdom of Saudi Arabia (KSA) over the last 2.5 decades, and to analyze aspects previously overlooked. Methods: This systematic review was based on evidence of RTAs in KSA. All articles published during the last 25 years on road traffic accident in KSA were analyzed. This study was carried out from December 2013 to May 2014 in the Department of Family and Community Medicine, Taibah University, Al-Madinah Al-Munawwarah, KSA. Results: Road traffic accidents accounted for 83.4% of all trauma admissions in 1984-1989, and no such overall trend was studied thereafter. The most frequently injured body regions as reported in the latest studies were head and neck, followed by upper and lower extremities, which was found to be opposite to that of the studies reported earlier. Hospital data showed an 8% non-significant increase in road accident mortalities in contrast to police records of a 27% significant reduction during the years 2005-2010. Excessive speeding was the most common cause reported in all recent and past studies. Conclusion: Disparity was common in the type of reporting of RTAs, outcome measures, and possible causes over a period of 2.5 decade. All research exclusively looked into the drivers’ faults. A sentinel surveillance of road crashes should be kept in place in the secondary and tertiary care hospitals for all regions of KSA. PMID:25828277

  6. Measurement of (d,p) reactions near the doubly-magic ^132Sn

    Measurements of (d,p) reactions on n-rich fission fragments yield important information on nuclear structure away from stability, and are of astrophysical interest due to the proximity to suggested r-process paths. The energies, spins and spectroscopic information of single-particle states near to shell closures are of particular importance, since they provide both an important constraint on nuclear structure models and are directly relevant to direct neutron-capture cross sections. The development of re-accelerated fission fragment beams at the HRIBF at ORNL has, for the first time, made possible the measurement of (d,p) reactions at the Coulomb barrier on nuclei around the doubly-magic ^132Sn nucleus. The ^130Sn(d,p)^131Sn, ^132Sn(d,p)^133Sn and ^ 134Te(d,p)^135Te reactions have been measured at the HRIBF at around 4.5 MeV/A utilizing deuterated plastic targets. Proton ejectiles were detected forward and backwards of θlab = 90^o using an early implementation of the Oak Ridge Rutgers University Barrel Array (ORRUBA) in conjunction with the Silicon Detector Array (SIDAR). Details of the experiments and the data analysis, including excitation energies and angular distributions for the states populated, will be presented. *This work is supported in part by the U.S. Department of Energy and the National Science Foundation.

    As the exploration and exploitation of oil and gas proliferate throughout deepwater area, the requirements on the reliability of dynamic positioning system become increasingly stringent. The control objective ensuring safety operation at deep water will not be met by a single controller for dynamic positioning. In order to increase the availability and reliability of dynamic positioning control system, the triple redundancy hardware and software control architectures were designed and developed according to the safe specifications of DP-3 classification notation for dynamically positioned ships and rigs. The hardware redundant configuration takes the form of triple-redundant hot standby configuration including three identical operator stations and three real-time control computers which connect each other through dual networks. The function of motion control and redundancy management of control computers were implemented by software on the real-time operating system VxWorks. The software realization of task loose synchronization, majority voting and fault detection were presented in details. A hierarchical software architecture was planed during the development of software, consisting of application layer, real-time layer and physical layer. The behavior of the DP-3 dynamic positioning control system was modeled by a Markov model to analyze its reliability. The effects of variation in parameters on the reliability measures were investigated. The time domain dynamic simulation was carried out on a deepwater drilling rig to prove the feasibility of the proposed control architecture.

    The roentgenographic identification of femoral component loosening after hip surface arthroplasty is often impossible because the metallic femoral component obscures the bone-cement interface. The use of combined technetium sulfur colloid and technetium methylene diphosphonate radionuclide imaging has been especially useful in the diagnosis of loosening. In 40 patients, follow-up combined TcSC and TcmDP scans at an average of three, nine, and 27 months postoperation revealed significant differences in the isotope uptakes in patients who had loose prostheses compared with those without complications. Scans were evaluated by first dividing them into eight anatomical regions and then rating the uptake in each region or 'zone' on a five-point scale. Results were compared using the Student's t-test and differences were noted between normal controls and patients who had femoral component loosening. Combining both TcSC and TcmDP studies increased the statistical significance obtained when comparing patients who had complications to those in the control group.

  9. Quantitative prediction of peptide binding to HLA-DP1 protein.

    The exogenous proteins are processed by the host antigen-processing cells. Peptidic fragments of them are presented on the cell surface bound to the major hystocompatibility complex (MHC) molecules class II and recognized by the CD4+ T lymphocytes. The MHC binding is considered as the crucial prerequisite for T-cell recognition. Only peptides able to form stable complexes with the MHC proteins are recognized by the T-cells. These peptides are known as T-cell epitopes. All T-cell epitopes are MHC binders, but not all MHC binders are T-cell epitopes. The T-cell epitope prediction is one of the main priorities of immunoinformatics. In the present study, three chemometric techniques are combined to derive a model for in silico prediction of peptide binding to the human MHC class II protein HLA-DP1. The structures of a set of known peptide binders are described by amino acid z-descriptors. Data are processed by an iterative self-consisted algorithm using the method of partial least squares, and a quantitative matrix (QM) for peptide binding prediction to HLA-DP1 is derived. The QM is validated by two sets of proteins and showed an average accuracy of 86 percent. PMID:24091413

  10. Effects of Strain Rates on Mechanical Properties and Fracture Mechanism of DP780 Dual Phase Steel

    The mechanical properties of DP780 dual phase steel were measured by quasi-static and high-speed tensile tests at strain rates between 0.001 and 1000 s-1 at room temperature. The deformation and fracture mechanisms were analyzed by observation of the tensile fracture and microstructure near the fracture. Dynamic factor and feret ratio quantitative methods were applied to study the effect of strain rate on the microstructure and properties of DP780 steel. The constitutive relation was described by a modified Johnson-Cook and Zerilli-Armstrong model. The results showed that the strain rate sensitivity of yield strength is bigger than that of ultimate tensile strength; as strain rate increased, the formation of microcracks and voids at the ferrite/martensite interface can be alleviated; the strain rate effect is unevenly distributed in the plastic deformation region. Moreover, both models can effectively describe the experimental results, while the modified Zerilli-Armstrong model is more accurate because the strain-hardening rate of this model is independent of strain rate.
