    Children can be lead poisoned when leaded paint is disturbed during home renovation or repair. We conducted a case-control study to assess the association between elevated blood lead levels (BLLs) in children younger than 5 years of age and renovation or repair of homes built before 1950 in New York City. In 1998, we interviewed parents of 106 case children (BLLs >/= 10 micro g/dL) and 159 control children (BLLs renovations or repairs in the 6 months before the blood lead test, and we adjusted for age and test month. Case children were only slightly more likely than control children to live in a house that had undergone any renovation (OR = 1.2, 95% confidence interval [95% CI] = 0.7, 2.1). Case children were more likely to (1) live in housing that had interior surfaces prepared for painting, especially by hand sanding (OR = 3.5, 95% CI = 1.1, 10.9; population attributable risk [PAR%] = 10.4%, 95% CI = 0.5%, 19.3%); and (2) have work-created dust throughout their housing unit (OR = 6.3, 95% CI = 1.2, 32.3; PAR% = 6.8%, 95% CI = 0.0%, 13.1%). The risk for excess lead exposure is increased by home renovation or repair work involving interior paint preparation or reported dispersal of dust beyond the work area. The proportion of cases related to this exposure is high enough to merit preventive measures. PMID:12468670

    Although blood lead levels (BLLs) >/=10 microg/dL are associated with adverse behavioral and developmental outcomes, and environmental and medical interventions are recommended at >/=20 microg/dL, no level is considered safe. A 1997 analysis conducted by the New York State Department of Health (NYSDOH) indicated that home renovation, repair, and painting (RRP) activities were important sources of lead exposure among children with BLLs >/=20 microg/dL in New York state (excluding New York City) during 1993--1994. Subsequently, local health departments in New York state began to routinely collect information about RRP activities when investigating children's home environments for lead sources. This report updates the 1997 analysis with data from environmental investigations conducted during 2006--2007 in New York state (excluding New York City) for 972 children with BLLs >/=20 microg/dL. RRP activities were identified as the probable source of lead exposure in 139 (14%) of the 972 children. Resident owners or tenants performed 66% of the RRP work, which often included sanding and scraping (42%), removal of painted materials or structures (29%), and other activities (29%) that can release particles of lead-based paint. RRP activities continued to be an important source of lead exposure during 2006--2007. Children living in housing built before 1978 (when lead-based paint was banned from residential use) that are undergoing RRP activities should be considered at high risk for elevated BLLs, and appropriate precautions should be taken to prevent exposure.   17. The contribution of housing renovation to children’s blood lead levels: a cohort study

    Background Routine renovation of older housing is a risk factor for childhood lead poisoning, but the contribution to children’s blood lead levels is poorly defined for children with lower exposure levels. Methods We examined a prospective cohort of 276 children followed from 6 to 24 months of age. We conducted surveys of renovation activities and residential lead hazards and obtained blood lead level (B-Pb) every six months. We analyzed B-Pb in a repeated measures design using a mixed effects linear model. Results Parent reported interior renovation ranged from 11 to 25% of housing units at the four, 6-month periods. In multivariable analysis, children whose housing underwent interior renovation had a 12% higher mean B-Pb by two years of age compared with children whose housing units were not renovated (p < 0.01). The time between renovation and the child blood lead sample was associated with higher B-Pb (p-value for trend <0.01); compared to children in non-renovated housing, children whose housing units underwent renovation in the prior month had a 17% higher mean B-Pb at two years of age, whereas children whose housing renovation occurred in the prior 2–6 months had an 8% higher mean B-Pb. We also found an association between higher paint lead loading, measured using an X-ray fluorescence (XRF) based paint lead index, and child B-Pb (p = 0.02); for every 10 mg/cm2 increase in paint lead loading index there was a 7.5% higher mean childhood B-Pb. Conclusions In an analysis of data collected before the recent changes to Environmental Protection Agency’s Lead, Renovation, Repair and Painting Rule, routine interior housing renovation was associated with a modest increase in children’s B-Pb. These results are important for the provision of clinical advice, for housing and public health professionals, and for policymakers. PMID:23981571

    In 1986, the ambient air for lead in radiator repair shops in the Minneapolis-St. Paul metropolitan area exceeded the Occupational Safety and Health Administration (OSHA) action level in nine of 12 shops sampled by Minnesota OSHA. We therefore sought to determine the prevalence of lead exposure/toxicity in this industry. Thirty-five radiator shops were identified, 30 were visited, and 53 workers were studied. The mean blood lead level was 1.53 (range 0.24-2.80). Seventeen individuals had blood lead levels greater than or equal to 1.93 mumol/L (40 micrograms/dl). The mean zinc protoporphyrin level (ZPP) was 0.55 mumol/L (range 0.16-1.43). No single worksite or personal characteristic was a strong determinant of either blood lead or ZPP level.

    In 1988, 83 automotive repair workers with blood lead levels (BLLs) greater than 25 micrograms/dL were reported to state health departments in the seven states that collaborated with CDC's National Institute for Occupational Safety and Health (NIOSH) in maintaining registries of elevated BLLs in adults. In 18 (22%) of these 83 persons, BLLs were greater than 50 micrograms/dL. Among automotive repair workers for whom a job category was specified, radiator repair work was the principal source of lead exposure. The major sources of exposure for radiator repair workers are lead fumes generated during soldering and lead dust produced during radiator cleaning. This report summarizes current BLL surveillance data for radiator repair workers and describes three control technologies that are effective in reducing lead exposures in radiator repair shops.

    In response to the Columbia Accident Investigation Board report, a plug repair kit has been developed to enable astronauts to repair the space shuttle's wing leading edge (WLE) during orbit. The plug repair kit consists of several 17.78- cm-diameter carbon/silicon carbide (C/SiC) cover plates of various curvatures that can be attached to the refractory carbon-carbon WLE panels using a TZM refractory metal attach mechanism. The attach mechanism is inserted through the damage in the WLE panel and, as it is tightened, the cover plate flexes to conform to the curvature of the WLE panel within 0.050 mm. An astronaut installs the repair during an extravehicular activity (EVA). After installing the plug repair, edge gaps are checked and the perimeter of the repair is sealed using a proprietary material, developed to fill cracks and small holes in the WLE.

    Compounds of lead and cadmium have been shown to be carcinogenic to humans and experimental animals. However, the underlying mechanisms are still not understood. In mammalian cells in culture, lead(II) is weakly mutagenic after long incubation times and generates DNA strand breaks only after treatment with high, toxic doses. Cadmium(II) induces DNA strand breaks and chromosomal aberrations, but its mutagenic potential is rather weak. However, both metals exert pronounced indirect genotoxic effects. Lead(II) is comutagenic towards UV and N-methyl-N-nitro-N-nitrosoguanidine (MNNG) and enhances the number of UV-induced sister chromatid exchanges in V79 Chinese hamster cells. With regard to DNA repair, lead(II) causes an accumulation of DNA strand breaks after UV-irradiation in HeLa cells, indicating an interference with the polymerization or ligation step in excision repair. Cadmium(II) enhances the mutagenicity of UV light in V79 Chinese hamster cells and an increased sensitivity toward UV light is observed in various rodent and human cell lines. Furthermore, an inhibition of unscheduled DNA synthesis after UV-irradiation and a partial inhibition of the removal of UV-induced DNA lesions has been shown. For both metals, the indirect genotoxic effects are observed at low, nontoxic concentrations, suggesting that an interference with DNA repair processes may be predominant at biologically relevant concentrations. This might also explain the conflicting results of epidemiological studies obtained for both metals. Possible mechanisms of repair inhibition are discussed. PMID:7843136

    A 1998 New York City Department of Transportation (NYCDOT) environmental impact statement (EIS) evaluated the proposed removal of deteriorated lead paint from NYCDOT-owned bridges. The EIS health risk assessment quantified the potential impact of particulate releases on blood lead levels among members of the public living and working near affected bridges. The risk assessment consisted of a fate and transport component and an exposure-dose component. The fate and transport component, modeled using the EPA's Industrial Source Complex (ISC3) model, calculated the impact of paint removal activities on ambient air lead concentrations and dust lead deposition rates. The exposure-dose component, modeled using EPA's Integrated Exposure Uptake Biokinetic (IEUBK) model, the Bowers et al. Adult Lead model, and the O'Flaherty lead model, calculated the impact of additional lead in air, street dust, interior house dust, and soil on blood lead levels, a conventional measure of body lead burden. The analysis was complicated because the ISC3 model provides a dust lead deposition rate ({micro}g/m{sup 2}-day), while the IEUBK, Bowers et al., and O'Flaherty models demand as input specification of dust lead concentrations ({micro}g lead per g dust). This paper describes a model developed for the EIS that quantifies long term average dust lead concentrations associated with typical bridge containment releases, and short term dust lead concentration spikes following worst case release events associated with bridge repair containment structure failures. The model reflects the influence of both lead and other debris associated with bridge repair activities, the contribution of background debris to street dust, and the impact of rainfall on removal of both lead and other material from the street dust reservoir.

  20. Flexible Metallic Overwrap Concept Developed for On-Orbit Repair of Space Shuttle Orbiter Leading Edges

    The Columbia accident has focused attention on the critical need for on-orbit repair concepts for leading edges in the event that damage is incurred during space shuttle orbiter flight. Damage that is considered as potentially catastrophic for orbiter leading edges ranges from simple cracks to holes as large as 16 in. in diameter. NASA is particularly interested in examining potential solutions for areas of larger damage since such a problem was identified as the cause for the Columbia disaster. One possible idea for the on-orbit repair of the reinforced carbon/carbon (RCC) leading edges is an overwrap concept that would use a metallic sheet flexible enough to conform to the contours of the orbiter and robust enough to protect any problem area from catastrophic failure during reentry. The simplified view of the application of a refractory metal sheet over a mockup of shuttle orbiter panel 9, which experiences the highest temperatures on the shuttle during reentry is shown. The metallic overwrap concept is attractive because of its versatility as well as the ease with which it can be included in an onboard repair kit. Reentry of the orbiter into Earth's atmosphere imposes extreme requirements on repair materials. Temperatures can exceed 1650 C for up to 15 min in the presence of an extremely oxidizing plasma environment. Several other factors are critical, including catalysity, emissivity, and vibrational and aerodynamic loads. Materials chosen for this application will need to be evaluated with respect to high-temperature capability, resistance to oxidation, strength, coefficient of thermal expansion, and thermal conductivity. The temperature profile across panel 9 during reentry as well as a schematic of the overwrap concept itself is shown.

    Renovation work for operation room A of the Chemical Processing Facility (CPF) was carried out. Cell renovation work involved disassembly, removal and installation of new equipment for the CA-3 cell of operation room A and the crane renovation work involved the repair of the in-cell crane for the CA-5 cell of operation room A. There were not many examples of renovation work performed on cells under high radiation environment and alpha contamination in Japan. Lessons learnt: With respect to the cell renovation work and crane repair work, a method that gave full consideration to safety was employed and the work was performed without accidents or disaster. Moreover, through improvement of the method, reduction of radioactive exposure of the workers was achieved and a melt reduction device was designed to deal with the radioactive waste material that was generated in the renovation work to achieve significant melt reduction of waste material.

  2. Exposure Evaluation for Benzene, Lead and Noise in Vehicle and Equipment Repair Shops

    An exposure assessment was performed at the equipment and vehicle maintenance repair shops operating at the U. S. Department of Energy Hanford site, in Richland, Washington. The maintenance shops repair and maintain vehicles and equipment used in support of the Hanford cleanup mission. There are three general mechanic shops and one auto body repair shop. The mechanics work on heavy equipment used in construction, cranes, commercial motor vehicles, passenger-type vehicles in addition to air compressors, generators, and farm equipment. Services include part fabrication, installation of equipment, repair and maintenance work in the engine compartment, and tire and brake services. Work performed at the auto body shop includes painting and surface preparation which involves applying body filler and sanding. 8-hour time-weighted-average samples were collected for benzene and noise exposure and task-based samples were collected for lead dust work activities involving painted metal surfaces. Benzene samples were obtained using 3M™ 3520 sampling badges and were analyzed for additional volatile organic compounds. These compounds were selected based on material safety data sheet information for the aerosol products used by the mechanics for each day of sampling. The compounds included acetone, ethyl ether, toluene, xylene, VM&P naphtha, methyl ethyl ketone, and trichloroethylene. Laboratory data for benzene, VM&P naphtha, methyl ethyl ketone and trichloroethylene were all below the reporting detection limit. Airborne concentrations for acetone, ethyl ether, toluene and xylene were all less than 10% of their occupational exposure limit. The task-based samples obtained for lead dusts were submitted for a metal scan analysis to identify other metals that might be present. Laboratory results for lead dusts were all below the reporting detection limit and airborne concentration for the other metals observed in the samples were less than 10% of the occupational exposure limit

  3. Metallic Concepts for Repair of Reinforced Carbon-Carbon Space Shuttle Leading Edges

    The Columbia accident has focused attention on the critical need for on-orbit repair concepts for wing leading edges in the event that potentially catastrophic damage is incurred during Space Shuttle Orbiter flight. The leading edge of the space shuttle wings consists of a series of eleven panels on each side of the orbiter. These panels are fabricated from reinforced carbon-carbon (RCC) which is a light weight composite with attractive strength at very high temperatures. The damage that was responsible for the loss of the Colombia space shuttle was deemed due to formation of a large hole in one these RCC leading edge panels produced by the impact of a large piece of foam. However, even small cracks in the RCC are considered as potentially catastrophic because of the high temperature re-entry environment. After the Columbia accident, NASA has explored various means to perform on-orbit repairs in the event that damage is sustained in future shuttle flights. Although large areas of damage, such as that which doomed Columbia, are not anticipated to re-occur due to various improvements to the shuttle, especially the foam attachment, NASA has also explored various options for both small and large area repair. This paper reports one large area repair concept referred to as the "metallic over-wrap." Environmental conditions during re-entry of the orbiter impose extreme requirements on the RCC leading edges as well as on any repair concepts. These requirements include temperatures up to 3000 F (1650 C) for up to 15 minutes in the presence of an extremely oxidizing plasma environment. Figure 1 shows the temperature profile across one panel (#9) which is subject to the highest temperatures during re-entry. Although the RCC possesses adequate mechanical strength at these temperatures, it lacks oxidation resistance.     Progressive telomere attrition or deficiency of the protective shelterin complex elicits a DNA damage response as a result of a cell's inability to distinguish dysfunctional telomeric ends from DNA double-strand breaks. SNMIB/Apollo is a shelterin-associated protein and a member of the SMN1/PSO2 nuclease family that localizes to telomeres through its interaction with TRF2. Here, we generated SNMIB/Apollo knockout mouse embryo fibroblasts (MEFs) to probe the function of SNMIB/Apollo at mammalian telomeres. SNMIB/Apollo null MEFs exhibit an increased incidence of G2 chromatid-type fusions involving telomeres created by leading-strand DNA synthesis, reflective of a failure to protect these telomeres after DNA replication. Mutations within SNMIB/Apollo's conserved nuclease domain failed to suppress this phenotype, suggesting that its nuclease activity is required to protect leading-strand telomeres. SNMIB/Apollo(-/-)ATM(-/-) MEFs display robust telomere fusions when Trf2 is depleted, indicating that ATM is dispensable for repair of uncapped telomeres in this setting. Our data implicate the 5'-3' exonuclease function of SNM1B/Apollo in the generation of 3' single-stranded overhangs at newly replicated leading-strand telomeres to protect them from engaging the non-homologous end-joining pathway.     In southern Thailand, the wooden boat building and repair industry uses powdered lead oxide (Pb(3)O(4,) or red lead) in the caulking process. This had been shown to be a significant source of contamination in the workplace and the communities. This cross-sectional study was conducted to estimate the amount of lead oxide used in the region and to document the management of hazardous waste product in this industry. All boatyards in southern Thailand registered to the Ministry of Industry were visited with walk-through observation. After consent, questionnaire-based interviews were conducted among workers and boatyard owners who rented out the space for building and repairing the boats. Of the 63 active boatyards identified, approximately 26 tons of lead oxide was used to repair approximately 8,000 boats each year. Due to the casual employment system in this industry, no facility for the protection of workers and the environment from lead contamination exists. The findings indicate the need for further studies on the extent of contamination in the broader area. Intervention programmes are also urgently needed to reduce the release of this hazardous waste into the community. PMID:20886362

Through our experience over the last 15 years and in our planning for the facility's 12 GeV upgrade we reevaluated the control room environment to capitalize on emerging visualization and display technologies and improve workflow processes and ergonomic attributes. This effort also sets the foundation for the redevelopment of the accelerator's control system to deliver high reliability performance with improvements in beam specifications management and information flow. The complete renovation was performed over a three-week maintenance period with no interruption to beam operations. We present the results of this effort.

The new facility takes advantage of advances in display technology, analog and video signal management, server technology, ergonomic workspace design, lighting engineering, acoustic ceilings and raised flooring solutions to provide a marked improvement in the overall environment of machine operations.

    Chimneys are an integral part of a fossil fueled generating station and perhaps one of the most commonly neglected and misunderstood structures at the plant. Structural damage caused by wind, earthquake, corrosive operating conditions, inadequate design, ever changing operating conditions, or years of neglect can render the structure unsafe and result in the need to perform major renovations or replace the entire chimney. The decision to renovate or replace an existing chimney is complex and should consider the following factors: (1) Age and condition of shell; (2) Type and condition of lining; (3) New operating conditions; (4) Cost of renovation vs replacement; (5) Unit outage time required; and (6) Cost for replacement power during outage. Information needed to make an informed decision regarding renovation or replacement, which most often involves several million dollars, can be best obtained from experienced chimney contractors and engineering specialists. This paper addresses four projects that utilized unique renovation techniques in lieu of partial or complete replacement of the structure.

    Acute kidney injury is an increasingly common complication of hospital admission and is associated with high levels of morbidity and mortality. A hypotensive, septic, or toxic insult can initiate a cascade of events, resulting in impaired microcirculation, activation of inflammatory pathways and tubular cell injury or death. These processes ultimately result in acutely impaired kidney function and initiation of a repair response. This Review explores the various mechanisms responsible for the initiation and propagation of acute kidney injury, the prototypic mechanisms by which a substantially damaged kidney can regenerate its normal architecture, and how the adaptive processes of repair can become maladaptive. These mechanisms, which include G2/M cell-cycle arrest, cell senescence, profibrogenic cytokine production, and activation of pericytes and interstitial myofibroblasts, contribute to the development of progressive fibrotic kidney disease. The end result is a state that mimics accelerated kidney ageing. These mechanisms present important opportunities for the design of targeted therapeutic strategies to promote adaptive renal recovery and minimize progressive fibrosis and chronic kidney disease after acute insults.     The bioaccessibility of Pb in air particulate matter from two car battery repair workshops in Kumasi (Ghana) was measured (64 full shift personal aerosol samples). An artificial lung lining fluid simulant (Hatch solution) was applied for leaching the bioaccessible fraction in half of the samples, the other half was leached with synthetic gastric juice. At both locations, the Pb solubility (median) in gastric juice (89% and 92%) is substantially higher than in Hatch solution (4.9% and 5.6%). The high solubility of Pb in gastric juice may be related to the presence of Pb oxides. The low bioaccessibility of Pb in Hatch solution is in good agreement with previous work on mine tailings, urban aerosol, car exhaust, welding fumes and indoor dust. The high bioaccessibility of Pb in the gastrointestinal tract underpins the importance of improving the personal hygienic behavior at the workplace. It is recommended that air monitoring of Pb should include the extrathoracic aerosol fraction using inhalable aerosol samplers, as particles of this size fraction are most likely transferred to the gastrointestinal tract in addition to the non-lung-soluble particles transported from the lung by mucociliary and phagocytosis clearance.   8. DNA Methylation Leads to DNA Repair Gene Down-Regulation and Trinucleotide Repeat Expansion in Patient-Derived Huntington Disease Cells.

    Huntington disease (HD) is an autosomal dominantly inherited disease that exhibits genetic anticipation of affected progeny due to expansions of a trinucleotide repeat (TNR) region within the HTT gene. DNA repair machinery is a known effector of TNR instability; however, the specific defects in HD cells that lead to TNR expansion are unknown. We hypothesized that HD cells would be deficient in DNA repair gene expression. To test this hypothesis, we analyzed expression of select DNA repair genes involved in mismatch/loop-out repair (APEX1, BRCA1, RPA1, and RPA3) in patient-derived HD cells and found each was consistently down-regulated relative to wild-type samples taken from unaffected individuals in the same family. Rescue of DNA repair gene expression by 5-azacytidine treatment identified DNA methylation as a mediator of DNA repair gene expression deficiency. Bisulfite sequencing confirmed hypermethylation of the APEX1 promoter region in HD cells relative to control, as well as 5-azacytidine-induced hypomethylation. 5-Azacytidine treatments also resulted in stabilization of TNR expansion within the mutant HTT allele during long-term culture of HD cells. Our findings indicate that DNA methylation leads to DNA repair down-regulation and TNR instability in mitotically active HD cells and offer a proof of principle that epigenetic interventions can curb TNR expansions.     Lead, an environmental toxin is known to induce a broad range of physiological and biochemical dysfunctions in humans through a number of mechanisms including the deactivation of antioxidants thus leading to generation of reactive oxygen species (ROS) and subsequent DNA damage. Selenium on the other hand has been proven to play an important role in the protection of cells from free radical damage and oxidative stress, though its effects are thought to be form and dose dependent. As the liver is the primary organ required for metabolite detoxification, HepG2 cells were chosen to assess the protective effects of various selenium compounds following exposure to the genotoxic agent lead nitrate. Initially DNA damage was quantified using a comet assay, gene expression patterns associated with DNA damage and signalling were also examined using PCR arrays and the biological pathways which were most significantly affected by selenium were identified. Interestingly, the organic type selenium compounds (selenium yeast and selenomethionine) conferred protection against lead induced DNA damage in HepG2 cells; this is evident by reduction in the quantity of DNA present in the comet tail of cells cultured in their presence with lead. This trend also followed through the gene expression changes noted in DNA damage pathways analysed. These results were in contrast with those of inorganic sodium selenite which promoted lead induced DNA damage evident in both the comet assay results and the gene expression analysis. Over all this study provided valuable insights into the effects which various selenium compounds had on the DNA damage and signalling pathway indicating the potential for using organic forms of selenium such as selenium enriched yeast to protect against DNA damaging agents.   12. Fibrin glue repair leads to enhanced axonal elongation during early peripheral nerve regeneration in an in vivo mouse model

    Microsurgical suturing is the gold standard of nerve coaptation. Although literature on the usefulness of fibrin glue as an alternative is becoming increasingly available, it remains contradictory. Furthermore, no data exist on how both repair methods might influence the morphological aspects (arborization; branching) of early peripheral nerve regeneration. We used the sciatic nerve transplantation model in thy-1 yellow fluorescent protein mice (YFP; n = 10). Pieces of nerve (1cm) were grafted from YFP-negative mice (n = 10) into those expressing YFP. We performed microsuture coaptations on one side and used fibrin glue for repair on the contralateral side. Seven days after grafting, the regeneration distance, the percentage of regenerating and arborizing axons, the number of branches per axon, the coaptation failure rate, the gap size at the repair site and the time needed for surgical repair were all investigated. Fibrin glue repair resulted in regenerating axons travelling further into the distal nerve. It also increased the percentage of arborizing axons. No coaptation failure was detected. Gap sizes were comparable in both groups. Fibrin glue significantly reduced surgical repair time. The increase in regeneration distance, even after the short period of time, is in line with the results of others that showed faster axonal regeneration after fibrin glue repair. The increase in arborizing axons could be another explanation for better functional and electrophysiological results after fibrin glue repair. Fibrin glue nerve coaptation seems to be a promising alternative to microsuture repair. PMID:26330844

    Nonerythroid {alpha}-spectrin ({alpha}IISp) is a structural protein involved in repair of DNA interstrand cross-links and is deficient in cells from patients with Fanconi anemia (FA), which are defective in ability to repair cross-links. In order to further demonstrate the importance of the role that {alpha}IISp plays in normal human cells and in the repair defect in FA, {alpha}IISp was knocked down in normal cells using siRNA. Depletion of {alpha}IISp in normal cells by siRNA resulted in chromosomal instability and cellular hypersensitivity to DNA interstrand cross-linking agents. An increased number of chromosomal aberrations were observed and, following treatment with a DNA interstrand cross-linking agent, mitomycin C, cells showed decreased cell growth and survival and decreased formation of damage-induced {alpha}IISp and XPF nuclear foci. Thus depletion of {alpha}IISp in normal cells leads to a number of defects observed in FA cells, such as chromosome instability and a deficiency in cross-link repair.

    NADPH oxidases (NOX) mediate redox signaling by generating superoxide and/or hydrogen peroxide, which are involved in biosynthetic pathways, e.g. thyroid hormone generation, dityrosine crosslinking, as well as bacterial killing. Data investigating the role of NOX enzymes in cutaneous wound repair is limited and specifically their function in skin myofibroblast expression is unknown. The isoform NOX4 was recently shown to be a pre-requisite for the differentiation of cardiac and pulmonary myofibroblasts. In this study we investigate the role of NOX4 in wound repair using a wound model in NOX4 knockout mice (n=16) and wildtype mice (n=16). Wounds were photographed daily until complete wound closure. Mice were sacrificed at day 3, 7, 14; wound tissue was harvested. NOX4-deficient mice healed significantly slower (22 days, SD=1.9) than wild-type mice (17 days, SD=1.4, p<0.005). However, there was no difference in myofibroblast expression. Strong dityrosine formation was observed, but was significantly weaker in NOX4-/- mice (p<0.05). NOX2, HIF1α and CD31 expression was significantly weaker in NOX4-/- mice (p<0.05). In this study we show for the first time that NOX4 plays a role in cutaneous wound repair. Our data suggests that NOX4 mediates HIF1α expression and neoangiogenesis during wound repair. NOX4 deletion led to a decreased expression of NOX2, implying a role of NOX4 in phagocytic cell recruitment. NOX4 was required for effective wound contraction but not myofibroblast expression. We suggest that myofibroblast contraction in NOX4-deficient mice is less effective in contracting the wound because of insufficient dityrosine-crosslinking of the ECM, providing the first indication for a physiological function of dityrosine crosslinking in higher animals.   20. Conjunctival inclusion cyst following repair of tube erosion in a child with aphakic glaucoma, leading to endophthalmitis

    Introduction: Glaucoma in aphakia is a major long term complication following congenital cataract surgery. Implantation of glaucoma drainage device provides an effective approach to manage refractory paediatric glaucoma. However implant surgery in young individuals is not free of complications. The prompt detection and management of tube erosion is of utmost importance to prevent devastating sequel of endophthalmitis. Implantation cyst following repair of tube erosion has not been reported so far. This case illustrates the rare occurrence of inclusion cyst following repair of tube erosion, the possible causes and its consequences. Case description: A 2-year-old child with aphakia developed intractable glaucoma. Following a failed glaucoma filtering surgery he underwent sequential Ahmed Glaucoma Valve implantation in both the eyes. Six weeks following right eye surgery, the child presented with conjunctival erosion overlying the tube, which was treated with scleral patch graft and conjunctival advancement. One month after the repair of tube erosion, the child presented with implantation cyst under the scleral patch graft, which was treated by drainage with a 29G needle. The child presented with endophthalmitis of his right eye following an episode of bilateral conjunctivitis. This was managed by an emergency pars plana vitrectomy, intraocular antibiotics and tube excision. At the last follow up visit, the IOP was 20 mmHg with 2 topical antiglaucoma medications in the right eye following a trans scleral photocoagulation. Discussion: Lifelong careful follow-up of paediatric eyes with implant surgery is mandatory to look for complication such as tube erosion. It is important to place additional sutures to secure the patch graft during implantation of glaucoma drainage devices in children to prevent graft displacement and consequent tube erosion.     Originally opened in 1928 as the Capitol Theatre, the Oscar Mayer Theatre in Madison, WI underwent substantial renovation in 1980 to support its current performance program. The theatre is now home to the Madison Symphony and the Madison Opera, and is used for ballet, touring shows, and popular concerts as well. As part of the ongoing Overture Project which will transform the theatre's home, the Madison Civic Center, into the Overture Center, the Oscar Mayer Theatre will be further improved to support its new role. In this paper we will discuss the reasoning behind the upcoming renovation within the context of the Overture Project, we will describe the pending architectural modifications to the theatre, and we will discuss the intended changes in acoustics. Computer modeling results will also be presented for the existing and renovated conditions.

    Inhibition of DNA repair is a recognized mechanism for arsenic enhancement of ultraviolet radiation-induced DNA damage and carcinogenesis. Poly(ADP-ribose) polymerase-1 (PARP-1), a zinc finger DNA repair protein, has been identified as a sensitive molecular target for arsenic. The zinc finger domains of PARP-1 protein function as a critical structure in DNA recognition and binding. Since cellular poly(ADP-ribosyl)ation capacity has been positively correlated with zinc status in cells, we hypothesize that arsenite binding-induced zinc loss from PARP-1 is equivalent to zinc deficiency in reducing PARP-1 activity, leading to inhibition of DNA repair. To test this hypothesis, we compared the effects of arsenite exposure with zinc deficiency, created by using the membrane-permeable zinc chelator TPEN, on 8-OHdG formation, PARP-1 activity and zinc binding to PARP-1 in HaCat cells. Our results show that arsenite exposure and zinc deficiency had similar effects on PARP-1 protein, whereas supplemental zinc reversed these effects. To investigate the molecular mechanism of zinc loss induced by arsenite, ICP-AES, near UV spectroscopy, fluorescence, and circular dichroism spectroscopy were utilized to examine arsenite binding and occupation of a peptide representing the first zinc finger of PARP-1. We found that arsenite binding as well as zinc loss altered the conformation of zinc finger structure which functionally leads to PARP-1 inhibition. These findings suggest that arsenite binding to PARP-1 protein created similar adverse biological effects as zinc deficiency, which establishes the molecular mechanism for zinc supplementation as a potentially effective treatment to reverse the detrimental outcomes of arsenic exposure.   5. Dasatinib induces DNA damage and activates DNA repair pathways leading to senescence in non-small cell lung cancer cell lines with kinase-inactivating BRAF mutations

    Improved therapies are greatly needed for non-small cell lung cancer (NSCLC) that does not harbor targetable kinase mutations or translocations. We previously demonstrated that NSCLC cells that harbor kinase-inactivating BRAF mutations (KIBRAF) undergo senescence when treated with the multitargeted kinase inhibitor dasatinib. Similarly, treatment with dasatinib resulted in a profound and durable response in a patient with KIBRAF NSCLC. However, no canonical pathways explain dasatinib-induced senescence in KIBRAF NSCLC. To investigate the underlying mechanism, we used 2 approaches: gene expression and reverse phase protein arrays. Both approaches showed that DNA repair pathways were differentially modulated between KIBRAF NSCLC cells and those with wild-type (WT) BRAF. Consistent with these findings, dasatinib induced DNA damage and activated DNA repair pathways leading to senescence only in the KIBRAF cells. Moreover, dasatinib-induced senescence was dependent on Chk1 and p21, proteins known to mediate DNA damage-induced senescence. Dasatinib also led to a marked decrease in TAZ but not YAP protein levels. Overexpression of TAZ inhibited dasatinib-induced senescence. To investigate other vulnerabilities in KIBRAF NSCLC cells, we compared the sensitivity of these cells with that of WTBRAF NSCLC cells to 79 drugs and identified a pattern of sensitivity to EGFR and MEK inhibitors in the KIBRAF cells. Clinically approved EGFR and MEK inhibitors, which are better tolerated than dasatinib, could be used to treat KIBRAF NSCLC. Our novel finding that dasatinib induced DNA damage and subsequently activated DNA repair pathways leading to senescence in KIBRAF NSCLC cells represents a unique vulnerability with potential clinical applications.     Co-exposure to cadmium, cobalt, lead and other heavy metals occurs in many occupational settings, such as pigment and batteries production, galvanization and recycling of electric tools. However, little is known about interactions between several heavy metals. In the present study we determined DNA single strand break (DNA-SSB) induction and repair capacity for 8-oxoguanine in mononuclear blood cells of 78 individuals co-exposed to cadmium (range of concentrations in air: 0.05-138.00 micro g/m(3)), cobalt (range: 0-10 micro g/m(3)) and lead (range: 0-125 micro g/m(3)). Exposure to heavy metals was determined in air, blood and urine. Non-parametric correlation analysis showed a correlation between cadmium concentrations in air with DNA-SSB (P = 0.001, R = 0.371). Surprisingly, cobalt air concentrations correlated even better (P < 0.001, R = 0.401), whereas lead did not correlate with DNA-SSB. Logistic regression analysis including 11 possible parameters of influence resulted in a model showing that cobalt in air, cadmium in air, cadmium in blood and lead in blood influence the level of DNA-SSB. The positive result with cobalt was surprising, since exposure levels were much lower compared with the TRK-value of 100 micro g/m(3). To examine, whether the positive result with cobalt is stable, we applied several logistic regression models with two blocks, where all factors except cobalt were considered preferentially. All strategies resulted in the model described above. Logistic regression analysis considering also all possible interactions between the relevant parameters of influence finally resulted in the following model: Odds ratio = 1.286(Co in air) x 1.040(Cd in air) x 3.111(Cd in blood) x 0.861(Pb in air) x 1.023(Co in air x Pb in air). This model correctly predicts an increased level of DNA-SSB in 91% of the subjects in our study.     SET domain proteins are present in chromosomal proteins involved in epigenetic control of transcription. The yeast SET domain protein Set1p regulates chromatin structure, DNA repair, and telomeric functions. We investigated the mechanism by which the absence of Set1p increases DNA repair capacities of checkpoint mutants. We show that deletion of SET1 induces a response relayed by the signaling kinase Rad53p that leads to the MEC1/TEL1-independent hyperphosphorylation of replication protein A middle subunit (Rfa2p). Consequently, the binding of Rfa2p to upstream repressing sequences (URS) of repair genes is decreased, thereby leading to their derepression. Our results correlate the set1Delta-dependent phosphorylation of Rfa2p with the transcriptional induction of repair genes. Moreover, we show that the deletion of the amino-terminal region of Rfa2p suppresses the sensitivity to ultraviolet radiation of a mec3Delta checkpoint mutant, abolishes the URS-mediated repression, and increases the expression of repair genes. This work provides an additional link for the role of Rfa2p in the regulation of the repair capacity of the cell and reveals a role for the phosphorylation of Rfa2p and unveils unsuspected connections between chromatin, signaling pathways, telomeres, and DNA repair. A vessel broke loose from its moorings in a severe storm and collided with a wellhead platform. The collision severely damaged the platform buckling seven major support members and cracking joints throughout the structure. In view of the significant damage, there was an urgent need to repair the structure to avoid any further damage from potentially sever winter storm conditions. Various means of repair and their associated costs were evaluated: traditional dry hyperbaric welding, adjacent platforms, grouted clamped connections, and mechanical pipe connectors. The repair was completed using an innovative combination of clamps and wet welding to attach external braces to the structure.

    The use of steel bracings is possible in all four stages of engineering activities in the renovation and modernization of historic buildings, i. e.: protecting, repairing, strengthening and rebuilding. Steel bracings are auxiliary structural systems, which, due to their relatively small cross-section dimensions and low weight, provide an efficient way to achieve the required resistance and stability of the original historic building structural systems. The steel structure in most cases is the tissue that differs in style and material of genuine tissue which is present in a renewed historic building, but is also often a rational solution acceptable to the conservation and structural point of view.

    The Plutonium Experimental Facility (PEF) will be used to develop a new fuel pellet fabrication process and to evaluate equipment upgrades. The facility was used from 1978 until 1982 to optimize the parameters for fuel pellet production using a process which was developed at Los Alamos National Laboratory. The PEF was shutdown and essentially abandoned until mid-1987 when the facility renovations were initiated by the Actinide Technology Section (ATS) of SRL. A major portion of the renovation work was related to the restart of the hot press system. This report describes the renovations and modifications which were required to restart the PEF hot press. The primary purpose of documenting this work is to help provide a basis for Separations to determine the best method of renovating the hot press in the Plutonium Fuel Fabrication (PuFF) facility. This report also includes several SRL recommendations concerning the renovation and modification of the PuFF hot press. 4 refs.

    In 1895, Percival Lowell hired eminent telescope maker Alvan G. Clark to build a 24-inch refractor. Lowell intended the telescope intitally for observing Mars in support of his controversial theories about life on that planet. Clark finished the telescope within a year and at a cost of $20,000. Lowell and his staff of assistants and astronomers began observing through it on July 23, 1896, setting off a long and productive career for the telescope.While Lowell's Mars studies dominated early work with the Clark, V.M. Slipher by the 1910s was using it to observe planetary rotations and atmospheric compositions. He soon revolutionized spectroscopic studies, gathering excruciatingly long spectra - some in excess of 40 hours - of the so-called white nebula and determining startling radial velocities, evidence of an expanding universe. In the 1960s, scientists and artists teamed up on the Clark and created detailed lunar maps in support of the Apollo program.In recent decades, the Clark has played a central role in the education programs at Lowell, with general public audiences, students, and private groups all taking advantage of this unique resource.With this nearly 120 years of contant use, the Clark had been wearing down in recent years. The telescope was becoming more difficult to move, old electrical wiring in the dome was a fire hazard, and many of the telescope's parts needed to be repaired or replaced.In 2013, Lowell Observatory began a fundraising campaign, collecting $291,000 to cover the cost of dome and telescope renovation. Workers removed the entire telescope mount and tube assembly from the dome, examining every part from tube sections to individuals screws. They also stabilized the dome, adding a water vapor barrier and new outer wall while reinforcing the upper dome. The project lasted from January, 2014 through August, 2015. The facility reopened for daytime tours in September, 2015 and evening viewing the following month.

    The installation or renovation of a cleanroom is an increasing priority among compounders who, to offer sterile preparations, must comply with the most recent version of United States Pharmacopeia revised General Chapter 797 guidelines. To upgrade their pharmacy facilites, many compounders have worked successfully with a large-scale mechanical engineering firm or a specialized construction company dedicated to cleanroom installation. However, a qualified local mechanical contractor with design capability is a valuable and often overlooked resource for the construction of customized stae-of-the-art cleanrooms. In this report, we describe the installation of such a facility in a busy pharmacy and suggest ways in which compounding pharmacists can identify competent local contractors and control the cost of construction and upgrades. The addition of a cleanroom has enabled The Compounding Pharmacy in Hickory, North Carolina, to achieve accreditation from the Pharmacy Compounding Accreditation Board. PMID:23970002

  9. Acoustical components of the Orpheum Theatre renovation

    The before and after acoustically-important listening conditions and measurements are described for a $14.2 million restoration and renovation of this 1,400-seat grand 1929 Movie Palace-at one time the tallest building in Phoenix. Great care was taken to restore all the acoustically good parts of the original design and to subtly modify the acoustically-troublesome parts (including severely focusing dome and sidewalls) so they looked the same but performed properly. A beautiful playhouse was achieved in 1997 with fine, fixed acoustics and conventional audio reinforcement, a surrounding Italian village, plenty of gilt, moving clouds and sunsets overhead. Today, the venue successfully hosts shows from performance art to ballet to jazz. It was not intended (acoustically) to serve classical music but does so on occasion.

    This study deals with the occurrence of volatile organic compounds (VOCs) in the indoor environment of a new and a renovated apartment. Qualitative determination of VOCs was carried out with a gas chromatograph with surface acoustic wave detector (GC/SAW). Concentrations of total volatile organic compounds (TVOC) were determined by a photoionization detector with UV lamp. Simultaneously, temperature and relative humidity were monitored with a data logger. The aim of this study was to determine of TVOC concentrations, to use of GC/SAW for determination of individual VOCs in indoor air as well as to predict possible sources of VOCs in these apartments. Measurements were performed after each construction work for better resolution of the contributions of individual materials to the levels of VOC. Mean concentrations of TVOC were 624 μg/m3 in the renovated apartment and 1,686 μg/m3 in the new apartment after completion of all works. The results from the renovated apartment showed that the use of new materials can lead to lower levels of organic compounds in indoor air compared to old materials that were less environmentally friendly. Many types of VOCs were found in both apartments. After reviewing the possible sources, it seems that the main sources of these substances were applied coatings and flooring materials.

    Summary The development of cell replacement strategies to repair the injured brain has gained considerable attention, with a particular interest in mobilizing endogenous neural stem and progenitor cells (known as neural precursor cells [NPCs]) to promote brain repair. Recent work demonstrated metformin, a drug used to manage type II diabetes, promotes neurogenesis. We sought to determine its role in neural repair following brain injury. We find that metformin administration activates endogenous NPCs, expanding the size of the NPC pool and promoting NPC migration and differentiation in the injured neonatal brain in a hypoxia-ischemia (H/I) injury model. Importantly, metformin treatment following H/I restores sensory-motor function. Lineage tracking reveals that metformin treatment following H/I causes an increase in the absolute number of subependyma-derived NPCs relative to untreated H/I controls in areas associated with sensory-motor function. Hence, activation of endogenous NPCs is a promising target for therapeutic intervention in childhood brain injury models. PMID:26235894

  18. Downregulation of the cancer susceptibility protein WRAP53β in epithelial ovarian cancer leads to defective DNA repair and poor clinical outcome

    Alterations in the scaffold protein WRAP53β have previously been linked to carcinogenesis and, in particular, associated with an increased risk for epithelial ovarian cancer. Here, we investigated the pathogenic impact and prognostic significance of WRAP53β in connection with epithelial ovarian cancer and examined the underlying mechanisms. We find that reduced expression of WRAP53β in ovarian tumors correlated with attenuated DNA damage response and poor patient survival. Furthermore, in ovarian cancer cell lines, WRAP53β was rapidly recruited to DNA double-strand breaks, where it orchestrated the recruitment of repair factors involved in homologous recombination and non-homologous end joining, including RNF168, 53BP1, BRCA1 and RAD51. Mechanistically, WRAP53β accomplishes this by facilitating the necessary ubiquitinylation at DNA breaks. Finally, we demonstrate that loss of WRAP53β significantly impairs the repair of DNA double-strand breaks, resulting in their accumulation. Our findings establish WRAP53β as a regulator of homologous recombination and non-homologous end joining repair in ovarian cancer cells, suggesting that loss of this protein contributes to the development and/or progression of ovarian tumors. Moreover, our current observations identify the nuclear levels of WRAP53β as a promising biomarker for the survival of patients with ovarian cancer. PMID:26426684

    Oligonucleotides can be used to direct the alteration of single nucleotides in chromosomal genes in yeast. Rad51 protein appears to play a central role in catalyzing the reaction, most likely through its DNA pairing function. Here, we re-engineer the RAD51 gene in order to produce proteins bearing altered levels of known activities. Overexpression of wild-type ScRAD51 elevates the correction of an integrated, mutant hygromycin resistance gene ∼3-fold. Overexpression of an altered RAD51 gene, which encodes a protein that has a higher affinity for ScRad54, enhances the targeting frequency nearly 100-fold. Another mutation which increases the affinity of Rad51 for DNA was also found to increase gene repair when overexpressed in the cell. Other mutations in the Rad51 protein, such as one that reduces interaction with Rad52, has little or no effect on the frequency of gene repair. These data provide the first evidence that the Rad51 protein can be modified so as to increase the frequency of gene repair in yeast. PMID:15087488

    Cellular DNA is subjected to continual attack, both by reactive species inside cells and by environmental agents. Toxic and mutagenic consequences are minimized by distinct pathways of repair, and 130 known human DNA repair genes are described here. Notable features presently include four enzymes that can remove uracil from DNA, seven recombination genes related to RAD51, and many recently discovered DNA polymerases that bypass damage, but only one system to remove the main DNA lesions induced by ultraviolet light. More human DNA repair genes will be found by comparison with model organisms and as common folds in three-dimensional protein structures are determined. Modulation of DNA repair should lead to clinical applications including improvement of radiotherapy and treatment with anticancer drugs and an advanced understanding of the cellular aging process. Accumulation of unrepaired CPD lesions causes inflammation, premature ageing and skin cancer. Photolyases are DNA repair enzymes that can rapidly restore DNA integrity in a light-dependent process called photoreactivation, but these enzymes are absent in humans. Here, we present a novel mRNA-based gene therapy method that directs synthesis of a marsupial, Potorous tridactylus, CPD-photolyase in cultured human keratinocytes. Pseudouridine was incorporated during in vitro transcription to make the mRNA non-immunogenic and highly translatable. Keratinocytes transfected with lipofectamine-complexed mRNA expressed photolyase in the nuclei for at least 2 days. Exposing photolyase mRNA-transfected cells to UVB irradiation resulted in significantly less CPD in those cells that were also treated with photoreactivating light, which is required for photolyase activity. The functional photolyase also diminished other UVB-mediated effects, including induction of IL-6 and inhibition of cell proliferation. These results demonstrate that pseudouridine-containing photolyase mRNA is a powerful tool to repair UVB-induced DNA lesions. The pseudouridine-modified mRNA approach has a strong potential to discern cellular effects of CPD in UV-related cell biological studies.   4. Construction/renovation influence on indoor air quality

    Much of the current construction activity in the US is renovation of existing buildings. So IAQ issues related to renovation are becoming more important. Indoor air contaminants that originate from nearby construction projects are also receiving increased attention. In some critical applications such as hospitals, IAQ control protocols have been developed and are being implemented. Extensive abatement methods for a few activities such as asbestos removal and fire damage cleanup have been developed that provide guidance for more general contamination control efforts. ASHRAE sponsored a research project to review the published literature and document current practice on indoor air quality issues related to construction and renovation activities (804-RP). This article outlines the major results from the study.

    The Thacher School is in the process of renovating the campus observatory with a new meter class telescope and full automation capabilities for the purpose of scientific research and education. New equipment on site has provided a preliminary site characterization including seeing and V-band sky brightness measurements. These data, along with commissioning data from the MINERVA project (which uses comparable hardware) are used to estimate the capabilities of the observatory once renovation is complete. Our V-band limiting magnitude is expected to be better than 21.3 for a one minute integration time, and we estimate that milli-magnitude precision photometry will be possible for a V=14.5 point source over approximately 5 min timescales. The quick response, autonomous operation, and multi-band photometric capabilities of the renovated observatory will make it a powerful follow-up science facility for exoplanets, eclipsing binaries, near-Earth objects, stellar variability, and supernovae.

    DOE funding was used to renovate 4,500 square feet of aging laboratories and classrooms that support science, engineering, and mathematics disciplines (specifically environmental science, and computer engineering). The expansion of the labs was needed to support robotics and environmental science research, and to better accommodate a wide variety of teaching situations. The renovated space includes a robotics laboratory, two multi-use labs, safe spaces for the storage of instrumentation, modern ventilation equipment, and other “smart” learning venues. The renovated areas feature technologies that are environmentally friendly with reduced energy costs. A campus showcase, the laboratories are a reflection of the University’s commitment to the environment and research as a tool for teaching. As anticipated, the labs facilitate the exploration of emerging technologies that are compatible with local and regional economic plans.

  7. EUVL Mask Blank Repair

    EUV mask blanks are fabricated by depositing a reflective Mo/Si multilayer film onto super-polished substrates. Small defects in this thin film coating can significantly alter the reflected field and introduce defects in the printed image. Ideally one would want to produce defect-free mask blanks; however, this may be very difficult to achieve in practice. One practical way to increase the yield of mask blanks is to effectively repair multilayer defects, and to this effect they present two complementary defect repair strategies for use on multilayer-coated EUVL mask blanks. A defect is any area on the mask which causes unwanted variations in EUV dose in the aerial image obtained in a printing tool, and defect repair is correspondingly defined as any strategy that renders a defect unprintable during exposure. The term defect mitigation can be adopted to describe any strategy which renders a critical defect non-critical when printed, and in this regard a non-critical defect is one that does not adversely affect device function. Defects in the patterned absorber layer consist of regions where metal, typically chrome, is unintentionally added or removed from the pattern leading to errors in the reflected field. There currently exists a mature technology based on ion beam milling and ion beam assisted deposition for repairing defects in the absorber layer of transmission lithography masks, and it is reasonable to expect that this technology will be extended to the repair of absorber defects in EUVL masks. However, techniques designed for the repair of absorber layers can not be directly applied to the repair of defects in the mask blank, and in particular the multilayer film. In this paper they present for the first time a new technique for the repair of amplitude defects as well as recent results on the repair of phase defects.

  8. Rapid new methods for paint collection and lead extraction.

    Chronic exposure of children to lead can result in permanent physiological impairment. In adults, it can cause irritability, poor muscle coordination, and nerve damage to the sense organs and nerves controlling the body. Surfaces coated with lead-containing paints are potential sources of exposure to lead. In April 2008, the U.S. Environmental Protection Agency (EPA) finalized new requirements that would reduce exposure to lead hazards created by renovation, repair, and painting activities, which disturb lead-based paint. On-site, inexpensive identification of lead-based paint is required. Two steps have been taken to meet this challenge. First, this paper presents a new, highly efficient method for paint collection that is based on the use of a modified wood drill bit. Second, this paper presents a novel, one-step approach for quantitatively grinding and extracting lead from paint samples for subsequent lead determination. This latter method is based on the use of a high-revolutions per minute rotor with stator to break up the paint into approximately 50 micron-size particles. Nitric acid (25%, v/v) is used to extract the lead in <3 minutes. Recoveries are consistently >95% for real-world paints, National Institute of Standards and Technology's standard reference materials, and audit samples from the American Industrial Hygiene Association's Environmental Lead Proficiency Analytical Testing Program. This quantitative extraction procedure, when paired with quantitative paint sample collection and lead determination, may enable the development of a lead paint test kit that will meet the specifications of the final EPA rule.     Funding from this grant has allowed Morehouse School of Medicine to complete its first professionally developed, comprehensive campus master plan that is in alignment with the recently completed strategic plan. In addition to master planning activities, funds were used for programming and designing research renovations, and also to supplement other research facility upgrades by providing lighting and equipment. The activities funded by this grant will provide the catalyst for substantial improvement in the School's overall facilities for biomedical education and research, and will also provide much of the information needed to conduct a successful campaign to raise funds for proposed buildings and renovations.

    Epidemiological and experimental studies have shown that cadmium is carcinogenic to human and experimental animals, however, the mechanism of cadmium-induced carcinogenesis is not clear. The aberrant expression of cell cycle and DNA repair genes resulting in increased cell proliferation and genomic instability are the characteristic features of cancer cells. The purpose of this study was to determine if exposure to cadmium can perturb cell proliferation/survival and causes genomic instability in TM3 cells, a mouse testicular Leydig cell line. The results of this study revealed that short-duration exposure to lower doses of cadmium significantly increase the growth of TM3 cells, whereas, higher doses are toxic and cause cell death. The long duration exposure to higher doses of cadmium, however, results in increased cell survival and acquisition of apoptotic resistance. Gene expression analysis by real-time PCR revealed increased expression of the anti-apoptotic gene Bcl-2, whereas decreased expression of pro-apoptotic gene Bax. Decreased expression of genes for maintenance of DNA methylation, DNMT1, and DNA repair, OGG1 and MYH, was also observed in cells exposed to cadmium for 24h. The random amplified polymorphic DNA (RAPD) assay revealed genomic instability in cells with chronic exposure to cadmium. The findings of this study indicate that mouse testicular Leydig cells adapt to chronic cadmium exposure by increasing cell survival through increased expression of Bcl-2, and decreased expression of Bax. The increased proliferation of cells with genomic instability may result in malignant transformation, and therefore, could be a viable mechanism for cadmium-induced cancers. PMID:19232459

    CPF (Chemical Processing Facility) was constructed at Nuclear Fuel Cycle Engineering Laboratories of JAEA (Japan Atomic Energy Agency) in 1980 as a basic research field where spent fuel pins from fast reactor (FR) and high level liquid waste can be dealt with. The renovation consists of remodeling of the CA-3 cell and the laboratory A, installation of globe boxes, hoods and analytical equipments to the laboratory C and the analytical laboratory. Also maintenance equipments in the CA-5 cell which had been out of order were repaired. The CA-3 cell is the main cell in which important equipments such as a dissolver, a clarifier and extractors are installed for carrying out the hot test using the irradiated FR fuel. Since the CPF had specialized originally in the research function for the Purex process, it was desired to execute the research and development of such new, various reprocessing processes. Formerly, equipments were arranged in wide space and connected with not only each other but also with utility supply system mainly by fixed stainless steel pipes. It caused shortage of operation space in flexibility for basic experimental study. Old equipments in the CA-3 cell including vessels and pipes were removed after successful decontamination, and new equipments were installed conformably to the new design. For the purpose of easy installation and rearranging the experimental equipments, equipments are basically connected by flexible pipes. Since dissolver is able to be easily replaced, various dissolution experiments is conducted. Insoluble residue generated by dissolution of spent fuel is clarified by centrifugal. This small apparatus is effective to space-saving. Mini mixer settlers or centrifugal contactors are put on to the prescribed limited space in front of the backside wall. Fresh reagents such as solvent, scrubbing and stripping solution are continuously fed from the laboratory A to the extractor by the reagent supply system with semi-automatic observation

    Critical to the Hospital's rebuilding efforts have been its public partners at the federal, state, and local government levels who have made a major financial commitment to renovating the Hospital's research infrastructure. To date, the Hospital for Special Surgery (HSS) has been awarded a total of nearly $8.5 million to create and equip new, state-of-the-art laboratories for scientific investigations. The modernization of the Hospital's research facilities was jump-started in 1998 with a $950,000 seed grant from the National Institutes of Health (NIH) to renovate laboratories for immunology research in the Caspary Research Building. Coupled with a matching $5.5 million commitment from HSS, this infusion of NIH funding laid the groundwork for an overhaul of all of the Hospital's research space.

    As living beings that encounter every kind of traumatic event from paper cut to myocardial infarction, we must possess ways to heal damaged tissues. While some animals are able to regrow complete body parts following injury (such as the earthworm who grows a new head following bisection), humans are sadly incapable of such feats. Our means of recovery following tissue damage consists largely of repair rather than pure regeneration. Thousands of times in our lives, a meticulously scripted but unseen wound healing drama plays, with cells serving as actors, extracellular matrix as the setting and growth factors as the means of communication. This article briefly reviews the cells involved in tissue repair, their signaling and proliferation mechanisms and the function of the extracellular matrix, then presents the actors and script for the three acts of the tissue repair drama.   11. Health Outcomes and Green Renovation of Affordable Housing

    Objective This study sought to determine whether renovating low-income housing using “green” and healthy principles improved resident health and building performance. Methods We investigated resident health and building performance outcomes at baseline and one year after the rehabilitation of low-income housing using Enterprise Green Communities green specifications, which improve ventilation; reduce moisture, mold, pests, and radon; and use sustainable building products and other healthy housing features. We assessed participant health via questionnaire, provided Healthy Homes training to all participants, and measured ventilation, carbon dioxide, and radon. Results Adults reported statistically significant improvements in overall health, asthma, and non-asthma respiratory problems. Adults also reported that their children's overall health improved, with significant improvements in non-asthma respiratory problems. Post-renovation building performance testing indicated that the building envelope was tightened and local exhaust fans performed well. New mechanical ventilation was installed (compared with no ventilation previously), with fresh air being supplied at 70% of the American Society of Heating, Refrigerating, and Air-Conditioning Engineers standard. Radon was <2 picocuries per liter of air following mitigation, and the annual average indoor carbon dioxide level was 982 parts per million. Energy use was reduced by 45% over the one-year post-renovation period. Conclusions We found significant health improvements following low-income housing renovation that complied with green standards. All green building standards should include health requirements. Collaboration of housing, public health, and environmental health professionals through integrated design holds promise for improved health, quality of life, building operation, and energy conservation.   12. Subjective and objective assessments in classrooms following acoustical renovation

    The effectiveness of an expensive acoustical intervention in an old Italian high school building has been assessed in this work. The school building has fifty classrooms, the majority of which were acoustically renovated. A subjective survey and measurements were performed in both the renovated and non renovated classrooms. With the competence of some psychologists from Turin University, a questionnaire was set up for the subjective analysis. The questionnaire, validated after numerous pilot tests, was submitted to all the students and the teachers in two periods of the year. The questions on acoustical features included questions on annoyance from room noise, reverberation, speech comprehension, overall acoustical satisfaction and the consequences of bad acoustical conditions. Apart from the acoustics, other aspects of environmental quality, such as the thermal and visual environmental features and IAQ were investigated. The statistical analysis of the subjective answers allowed aggregated information to be obtained on the users and different data to be correlated. The aim of the statistical correlation was to determine any significant relationships between the objective and subjective data, and between the overall satisfaction scores and the different environmental factors. The effects of bad environmental conditions and their influence on learning capacity were also examined.

    Phytoplankton inhabiting oligotrophic ocean gyres actively reduce their phosphorus demand by replacing polar membrane phospholipids with those lacking phosphorus. Although the synthesis of nonphosphorus lipids is well documented in some heterotrophic bacterial lineages, phosphorus-free lipid synthesis in oligotrophic marine chemoheterotrophs has not been directly demonstrated, implying they are disadvantaged in phosphate-deplete ecosystems, relative to phytoplankton. Here, we show the SAR11 clade chemoheterotroph Pelagibacter sp. str. HTCC7211 renovates membrane lipids when phosphate starved by replacing a portion of its phospholipids with monoglucosyl- and glucuronosyl-diacylglycerols and by synthesizing new ornithine lipids. Lipid profiles of cells grown with excess phosphate consisted entirely of phospholipids. Conversely, up to 40% of the total lipids were converted to nonphosphorus lipids when cells were starved for phosphate, or when growing on methylphosphonate. Cells sequentially limited by phosphate and methylphosphonate transformed >75% of their lipids to phosphorus-free analogs. During phosphate starvation, a four-gene cluster was significantly up-regulated that likely encodes the enzymes responsible for lipid renovation. These genes were found in Pelagibacterales strains isolated from a phosphate-deficient ocean gyre, but not in other strains from coastal environments, suggesting alternate lipid synthesis is a specific adaptation to phosphate scarcity. Similar gene clusters are found in the genomes of other marine α-proteobacteria, implying lipid renovation is a common strategy used by heterotrophic cells to reduce their requirement for phosphorus in oligotrophic habitats.   14. Usefulness of vascular stenting with and without transvenous pacing leads for vena caval obstruction among children and adults with repaired congenital heart disease.

    Vena caval obstruction (VCO) is a common complication after vascular manipulation for congenital heart disease. Long-term efficacy of stent therapy for relief of VCO and long-term stent patency with and without intrastent transvenous pacing leads (TPLs) is not well described. This was a retrospective review of patients treated for VCO, including those who received intrastent TPLs, between 1995 and 2012. Patient demographics, diagnoses, vascular pressure gradients, and vessel diameters were analyzed. Forty-one patients (mean age 23.5 ± 10.3 years) with and without congenital heart disease underwent stent implantation, 26 of whom also received intrastent TPLs. Short-term stent implantation success in relieving obstructions was 93%. Poststent vascular pressure gradients and percentage vascular narrowing significantly improved (from 6.2 ± 4.5 to 1.1 ± 1.6 mm Hg and from 63.1 ± 19.5% to 18.0 ± 17.1%, respectively, p <0.05). On follow-up in 38 of 41 patients from 0.2 to 18 years (median 6.0), all survived; 6 (14%) required stent reintervention. Freedom from reintervention was 87% at 15 years. Patients with short-term procedural failure were at higher risk for stent reintervention. Among 27 patients with intrastent TPLs, freedom from reintervention was 96%. In 26 patients with follow-up catheterization, intrastent intimal proliferation was not significantly associated with TPL but was higher in the superior vena cava-innominate vein junction compared with other stent locations (p <0.05). In conclusion, stent therapy for VCO can be successfully and safely performed with good long-term results. Pre-pacing lead stent placement for VCO is effective in allowing TPL placement with encouraging long-term patency. PMID:25910527

  1. The Datacon Master -- Renovation of a Datacon field bus communications system for accelerator control

    The Datacon system is a serial coaxial transformer isolated communication field bus system used to control and monitor accelerator remote devices. The Datacon field bus has been a BNL accelerator standard since its initial use in 1965. A single Datacon field bus supports up to 256 devices on a multidrop RG62A/U coaxial cable with up to 33 devices or 2,000 feet between repeaters or buffered branches. The forcing factor to renovate was the inability to repair the aging PDP-8E and PDP10 computers. The maintenance on this aging system was costly and the large number of accelerator devices dependent on the Datacon system could not be converted in a reasonable period of time to a new modern field bus. A commercial VMEbus host CPU mated with a custom designed VMEbus SBC event driven serial communications engine featuring a superscaler RISC 32-bit Intel i960 CPU met the design challenge. The commercial VMEbus host runs the VxWorks real-time operating system and connects to UNIX workstations over a Ethernet LAN. The V110 Datacon Master is the custom designed front end computer that integrates an accelerator event time line system with accelerator devices for up to 8 ppm users adding new capabilities.

    Window replacement is a key method of reducing childhood lead exposure, but the long-term effectiveness has not been previously evaluated. Windows have the highest levels of interior lead paint and dust compared to other building components. Our objective was to conduct a follow-up study of residential window replacement and lead hazard control 12 years after homes were enrolled in an evaluation of the HUD Lead Hazard Control Grant Program, sampling settled lead dust in housing in four cities (n=189 homes). Previous work evaluated lead hazard controls up to 6 years after intervention using dust lead measurements and two years after intervention using both dust and blood lead data. But the earlier work could not examine the effect of window replacement over the longer time period examined here: 12 years. The individual homes were assigned to one of three categories, based on how many windows had been replaced: all replacement, some replacement, or non-replacement. Windows that were not replaced were repaired. We controlled for covariates such as site, housing condition, presence of lead paint, and season using longitudinal regression modeling. Adjusted floor and sill dust lead geometric mean dust lead loadings declined at least 85% from pre-intervention to 12 years after the intervention for homes with all replacement windows, some windows replaced and no windows replaced. Twelve years after intervention, homes with all replacement windows had 41% lower interior floor dust lead, compared to non-replacement homes (1.4 versus 2.4 μg/ft2, p<0.001), and window sill dust lead was 51% lower (25 versus 52 μg/ft2, p=0.006) while controlling for covariates. Homes with some windows replaced had interior floor and window sill dust lead loadings that were 28% (1.7 versus 2.4 μg/ft2, p=0.19) and 37% (33 versus 52 μg/ft2, p=0.07) lower, respectively, compared to non-replacement homes.     The two broad categories of deposited weld metal repair and fiber-reinforced composite repair technologies were reviewed for potential application for internal repair of gas transmission pipelines. Both are used to some extent for other applications and could be further developed for internal, local, structural repair of gas transmission pipelines. Preliminary test programs were developed for both deposited weld metal repairs and for fiber-reinforced composite repair. To date, all of the experimental work pertaining to the evaluation of potential repair methods has focused on fiber-reinforced composite repairs. Hydrostatic testing was also conducted on four pipeline sections with simulated corrosion damage: two with composite liners and two without.

    The renovation of an existing 1800 square foot commercial greenhouse to incorporate passive solar reliant and energy conserving features is detailed. The Aquatic-Agriculture Institute for Research, a non-profit group, sponsored the project to develop efficient production methods to raise vegetables and fish at the community level. The performance of the remodeled greenhouse will be compared to the performance of the same greenhouse as it was originally designed. The restored greenhouse began operation in September 1979. Accurate fuel and temperature records maintained through-out the past winter, show the cost of back-up heating under operating conditions to be approximately $150.00. Old fuel receipts dating back into the 1940's show an average use of 2000 gallons of heating fuel each winter prior to remodeling. This would indicate a yearly fuel savings of better than 90% through the use of passive solar techniques.

During the blood stage of infection, the parasite is a master renovator of its erythrocyte host cell, and the changes in cell morphology and function that are induced by the parasite promote survival and contribute to the pathogenesis of severe malaria. In this Review, we discuss how Plasmodium parasites use the protein trafficking motif Plasmodium export element (PEXEL), protease-mediated polypeptide processing, a novel translocon termed the Plasmodium translocon of exported proteins (PTEX) and exomembranous structures to export hundreds of proteins to discrete subcellular locations in the host erythrocytes, which enables the parasite to gain access to vital nutrients and to evade the immune defence mechanisms of the host. Here, we aimed at investigating the complete Nucleotide Excision Repair (NER), a DNA repair pathway involved in the removal of bulky DNA adducts, status in an Alzheimer-like cell line. The level of DNA damage was quantified using mass spectrometry, NER gene expression was assessed by qPCR, and the NER protein activity was analysed through a modified version of the COMET assay. Interestingly, we found that in the presence of the Amyloid β peptide (Aβ), NER factors were upregulated at the mRNA level and that NER capacities were also specifically increased following oxidative stress. Surprisingly, NER capacities were not differentially improved following a typical NER-triggering of ultraviolet C (UVC) stress. Oxidative stress generates a differential and specific DNA damage response in the presence of Aβ. We hypothesized that the release of NER components such as DNA damage binding protein 2 (DDB2) and Xeroderma Pigmentosum complementation group C protein (XPC) following oxidative stress might putatively involve their apoptotic role rather than DNA repair function.     To assess lead exposure in the Jamaican lead-acid battery industry, we surveyed three battery manufacturers (including 46 production workers) and 10 battery repair shops (including 23 battery repair workers). Engineering controls and respiratory protection were judged to be inadequate at battery manufacturers and battery repair shops. At manufacturers, 38 of 42 air samples for lead exceeded a work-shift time-weighted average concentration of 0.050 mg/m3 (range 0.030-5.3 mg/m3), and nine samples exceeded 0.50 mg/m3. Only one of seven air samples at repair shops exceeded 0.050 mg/m3 (range 0.003-0.066 mg/m3). Repair shop workers, however, had higher blood lead levels than manufacturing workers (65% vs. 28% with blood lead levels above 60 micrograms/dl, respectively). Manufacturing workers had a higher prevalence of safe hygienic practices and a recent interval of minimal production had occurred at one of the battery manufacturers. Workers with blood lead levels above 60 micrograms/dl tended to have higher prevalences of most symptoms of lead toxicity than did workers with lower blood lead levels, but this finding was not consistent or statistically significant. The relationship between zinc protoporphyrin concentrations and increasing blood lead concentrations was consistent with that described among workers in developed countries. The high risk of lead toxicity among Jamaican battery workers is consistent with studies of battery workers in other developing countries. PMID:2773946

  12. Field turbidity method for the determination of lead in acid extracts of dried paint.

    Lead, which can be found in old paint, soil, and dust, has been clearly shown to have adverse health effects on the neurological systems of both children and adults. As part of an ongoing effort to reduce childhood lead poisoning, the US Environmental Protection Agency promulgated the Lead Renovation, Repair, and Painting Program (RRP) rule requiring that paint in target housing built prior to 1978 be tested for lead before any renovation, repair, or painting activities are initiated. This rule has led to a need for a rapid, relatively easy, and an inexpensive method for measuring lead in paint. This paper presents a new method for measuring lead extracted from paint that is based on turbidimetry. This method is applicable to paint that has been collected from a surface and extracted into 25% (v/v) of nitric acid. An aliquot of the filtered extract is mixed with an aliquot of solid potassium molybdate in 1 M ammonium acetate to form a turbid suspension of lead molybdate. The lead concentration is determined using a portable turbidity meter. This turbidimetric method has a response of approximately 0.9 NTU per microg lead per mL extract, with a range of 1-1000 Nephelometric Turbidity Units (NTUs). Precision at a concentration corresponding to the EPA-mandated decision point of 1 mg of lead per cm(2) is <2%. This method is insensitive to the presence of other metals common to paint, including Ba(2+), Ca(2+), Mg(2+), Fe(3+), Co(2+), Cu(2+), and Cd(2+), at concentrations of 10 mg mL(-1) or to Zn(2+) at 50 mg mL(-1). Analysis of 14 samples from six reference materials with lead concentrations near 1 mg cm(-2) yielded a correlation to inductively coupled plasma-atomic emission spectroscopy (ICP-AES) analysis of 0.97, with an average bias of 2.8%. Twenty-four sets of either 6 or 10 paint samples each were collected from different locations in old houses, a hospital, tobacco factory, and power station.     The rationale for cartilage repair is to prevent precocious osteoarthritis in untreated focal cartilage injuries in the young and middle-aged population. The gamut of surgical techniques, normal postoperative radiological appearances, and possible complications have been described. An objective method of recording the quality of repair tissue is with the magnetic resonance observation of cartilage repair tissue (MOCART) score. This scoring system evaluates nine parameters that include the extent of defect filling, border zone integration, signal intensity, quality of structure and surface, subchondral bone, subchondral lamina, and records presence or absence of synovitis and adhesions. The five common techniques of cartilage repair currently offered include bone marrow stimulation (microfracture or drilling), mosaicplasty, synthetic resorbable scaffold grafts, osteochondral allograft transplants, and autologous chondrocyte implantation (ACI). Complications of cartilage repair procedures that may be demonstrated on magnetic resonance imaging (MRI) include plug loosening, graft protuberance, graft depression, and collapse in mosaicplasty, graft hypertrophy in ACI, and immune response leading to graft rejection, which is more common with synthetic grafts and cadaveric allografts.   18. Evaluation of cleaning methods applied in home environments after renovation and remodeling activities.

    We conducted a cleaning trial in 40 northern New Jersey homes where home renovation and remodeling (R&R) activities were undertaken. Two cleaning protocols were used in the study: a specific method recommended by the US Department of Housing and Urban Development (HUD), in the 1995 "Guidelines for the Evaluation and Control of Lead-Based Paint Hazards in Housing," using a high-efficiency particulate air (HEPA)-filtered vacuum cleaner and a tri-sodium phosphate solution (TSP); and an alternative method using a household vacuum cleaner and a household detergent. Eligible homes were built before the 1970s with potential lead-based paint and had recent R&R activities without thorough cleaning. The two cleaning protocols were randomly assigned to the participants' homes and followed the HUD-recommended three-step procedure: vacuuming, wet washing, and repeat vacuuming. Wipe sampling was conducted on floor surfaces or windowsills before and after cleaning to evaluate the efficacy. All floor and windowsill data indicated that both methods (TSP/HEPA and non-TSP/non-HEPA) were effective in reducing lead loading on the surfaces (P < 0.001). When cleaning was applied to surfaces with initial lead loading above the clearance standards, the reductions were even greater, above 95% for either cleaning method. The mixed-effect model analysis showed no significant difference between the two methods. Baseline lead loading was found to be associated with lead loading reduction significantly on floors (P < 0.001) and marginally on windowsills (P = 0.077). Such relations were different between the two cleaning methods significantly on floors (P < 0.001) and marginally on windowsills (P = 0.066), with the TSP/HEPA method being favored for higher baseline levels and the non-TSP/non-HEPA method for lower baseline levels.   2. Renovation of a pear orchard site with sludge compost

    A lime-stabilized sewage sludge compost was used as a surface amendment to improve the soil and nutritional status of a number of established pear cultivars grown on an acidic, low fertility soil site. Leaf Ca status was significantly increased while trace metals were not elevated and in most cases decreased by sludge compost over the course of the study. Soil chemical properties were modified in a manner similar to liming. Addition of sludge composts, particularly low metal containing materials, appears to represent an acceptable aid in renovating established pear orchards located on poor soil sites. Leaf, fruit flesh or peel Cd were not significantly effected by the compost addition. The compost added twice the recommended level of available N the first year and sub-optimum N the following two years. Leaf N, although significantly increased in the composted versus non-composted controls, was below sufficiency levels by the third year after addition. This indicates that compost cannot fully supply required N from a one time application even over the short term.

DNA modifications can lead to permanent genetic changes that promote human disease. To combat the deleterious effects of DNA damage, organisms are equipped with DNA repair systems. The focus of our investigations has been to elucidate the details of mammalian DNA repair. These studies have provided important insights into the relationship of DNA repair to human disease and the genetic factors that contribute to individual susceptibility to the harmful effects of environmental mutagens (e.g. ionizing radiation), and have established a framework for designing more effective anti-cancer treatment schemes. To investigate questions related to DNA repair, its mechanism(s) and linkage to disease development, the authors are employing an array of molecular, cellular, biochemical and structural approaches. These complementary experimental techniques have permitted a detailed analysis into various processes of human DNA repair.

    The Composite Cryotank Technologies and Demonstration Project is working to advance the technologies for composite cryogenic propellant tanks at diameters suitable for future heavy lift vehicles and other in-space applications. The main goals of the project are to reduce weight and cost. One aspect of this project has focused on damage evaluation and repair development. Test panels have been impacted, repaired, and tested. Several repair methods were used to compare their effectiveness at restoring the integrity of the composite. Panels were evaluated by nondestructive evaluations at several points during the process to assess the damage and repair. The testing performed and the results and conclusions from the nondestructive evaluations and the destructive testing will be discussed. These results will lead to further development of inspection techniques and repair methods.

    Among DNA damages, double-strand breaks (DSBs) are one of the most harmful lesions to a cell. Failure in DSB repair could lead to genomic instability and cancer. Homologous recombination (HR) and nonhomologous end joining (NHEJ) are major DSB repair pathways in higher eukaryotes. It is known that expression of DSB repair genes is altered in various cancers. Activation of DSB repair genes is one of the reasons for chemo- and radioresistance. Therefore, targeting DSB repair is an attractive strategy to eliminate cancer. Besides, therapeutic agents introduce breaks in the genome as an intermediate. Therefore, blocking the residual repair using inhibitors can potentiate the efficacy of cancer treatment. In this review, we discuss the importance of targeting DSB repair pathways for the treatment of cancer.   12. Effects of Full-Scale Beach Renovation on Fecal Indicator Levels in Shoreline Sand and Water

    Recolonization of enterococci, at a non-point source beach known to contain high background levels of bacteria, was studied after a full-scale beach renovation project. The renovation involved importation of new exogenous sand, in addition to infrastructure improvements. The study's objectives were to document changes in sand and water quality and to evaluate the relative contribution of different renovation activities towards these changes. These objectives were addressed: by measuring enterococci levels in the sand and fecal indicator bacteria levels (enterococci and fecal coliform) in the water, by documenting sediment characteristics (mineralogy and biofilm levels), and by estimating changes in observable enterococci loads. Analysis of enterococci levels on surface sand and within sediment depth cores were significantly higher prior to beach renovation (6.3 to 72 CFU/g for each sampling day) when compared to levels during and after beach renovation (0.8 CFU/g to 12 CFU/g) (p<0.01). During the renovation process, sand enterococci levels were frequently below detection limits (<0.1 CFU/g). For water, exceedances in the regulatory thresholds that would trigger a beach advisory decreased by 40% for enterococci and by 90% for fecal coliform. Factors that did not change significantly between pre- and post- renovation included the enterococci loads from animals (approx. 3 × 1011 CFU per month). Factors that were observed to change between pre- and post- renovation activities included: the composition of the beach sand (64% versus 98% quartz, and a significant decrease in biofilm levels) and loads from direct stormwater inputs (reduction of 3 × 1011 CFU per month). Overall, this study supports that beach renovation activities contributed to improved sand and water quality resulting in a 50% decrease of observable enterococci loads due to upgrades to the stormwater infrastructure.     Heading for possible use for clinical trial, THOR (Tsing Hua Open-pool Reactor) at Taiwan was shutdown for renovation of a new epithermal neutron beam in January 2003. In November 2003, concrete cutting was finished for closer distance from core and larger treatment room. This article presents the design base that the construction of the new beam is based on. The filter/moderator design along the beam is Cd(0.1cm)+Al(10 cm)+FLUENTAL (16 cm)+Al(10 cm)+FLUENTAL(24 cm)+Void(18 cm)+Cd(0.1cm)+Bi(10 cm) with 6 cm Pb as reflector. Following the filter/moderator is an 88 cm long, 6 cm thick Bi-lined collimator with Li(2)CO(3)-PE at the end. The collimator is surrounded by Li(2)CO(3)-PE and Pb. The calculated beam parameters under 2 MW at the beam exit is phi(epi) = 3.4 x 10(9) n/cm(2)/s, Df/phi(epi) = 2.8 x 10(-11) cGy cm(2)/n, Dgamma/phi(epi) = 1.3 x 10(-11) cGy cm(2)/n, and J+/phi = 0.8. For a phantom placed 10 cm from beam exit, MCNP calculation shows that the advantage depth is 8.9 cm, and advantage ratio is 5.6 if boron concentration in tumor and normal tissue are assumed to be 65 and 18 ppm. The maximum dose rate for normal tissue is 50 cGy/min. The maximum therapeutic ratio is 6. The construction of the beam is scheduled to be finished by the end of April 2004. PMID:15308189

    Constructed soil filter (CSF) also known as Soil Biotechnology (SBT) is a process for water renovation which makes use of formulated media with culture of soil macro- and microorganisms. CSF combines sedimentation, infiltration and biodegradation processes to remove oxidizable organics and inorganics of wastewater in a single facility. Operating experience shows hydraulic loading in the range of 0.05-0.25 m(3)/m(2) h and organic loading up to 200-680 g/m(2) d. The results show increase in dissolved oxygen levels, COD removal (from 352 mg/l to 20 mg/l); BOD removal (from 211 mg/l to 7.0 mg/l); suspended solids removal (from 293 mg/l to 16 mg/l); turbidity reduction (from 145 NTU to 5.3 NTU); iron (from 5 mg/l to 0.3 mg/l); arsenic (from 500 microg/l to 10 microg/l); total coliform and fecal coliform removal (from 145 x 10(5) to 55 CFU/100 mL and 150 x 10(8) to 110 CFU/100 mL respectively), with desired pathogen levels as per WHO standards, i.e. < or =10(3) CFU/100 mL. CSF reveals advantages such as low HRT (0.5-2.0 h), low energy requirement (0.04 kWh/m(3)), no pre-treatment, high dissolved oxygen levels in the effluent, no biosludge production, no mechanical aeration and no odor, fish compatible water quality and evergreen ambience.     Temperate grasslands constitute over 30% of the Earth's naturally-occurring biomes and make an important contribution towards the partial mitigation of anthropogenic greenhouse gas emissions by terrestrial ecosystems. In permanent temperate grasslands, biomass production and sward quality can deteriorate over time and periodic renovation activities, involving soil tillage and reseeding, are commonly carried out to halt this decline. Long-term cultivation of agricultural land has been associated with soil aggregate degradation and reduced soil carbon storage. However, the impact of these single tillage disturbances on C cycling in grasslands is less clear. This study evaluated gaseous and dissolved organic carbon (DOC) losses following a single tillage event by subjecting grassland lysimeters with contrasting soil drainage characteristics to simulated conventional inversion or minimum tillage. Field-scale CO2 emissions after conventional tillage were also quantified and empirically modelled over short- and medium-term timeframes to delineate the ecosystem response to environmental variables. Soil moisture was the limiting determinant of ecosystem carbon release following conventional tillage. Freshly-tilled soils were associated with reduced water retention and increased sensitivity to soil moisture, which was particularly pronounced following rewetting events. Significantly elevated but ephemeral CO2 effluxes were detected in the hours following inversion ploughing, however tillage disturbance did not generate significantly enhanced C emission rates in the medium term. Equally, DOC losses were not significantly amplified by conventional tillage compared with conservative minimum tillage and were predominantly controlled by soil drainage across tillage regimes. Our results suggest that a net ecosystem source of 120 to 210 g C m-2 over an approximately two-month period was most likely a consequence of reduced productivity and C input rather than enhanced soil CO2

    DNA double-strand breaks (DSBs) are among the most deleterious DNA lesions, which if unrepaired or repaired incorrectly can cause cell death or genome instability that may lead to cancer. To counteract these adverse consequences, eukaryotes have evolved a highly orchestrated mechanism to repair DSBs, namely DNA-damage-response (DDR). DDR, as defined specifically in relation to DSBs, consists of multi-layered regulatory modes including DNA damage sensors, transducers and effectors, through which DSBs are sensed and then repaired via DNAprotein interactions. Unexpectedly, recent studies have revealed a direct role of RNA in the repair of DSBs, including DSB-induced small RNA (diRNA)-directed and RNA-templated DNA repair. Here, we summarize the recent discoveries of RNA-mediated regulation of DSB repair and discuss the potential impact of these novel RNA components of the DSB repair pathway on genomic stability and plasticity.   6. Repair material properties for effective structural application

    Strength and engineering properties of three generic repair materials which are likely to influence long-term performance of repaired concrete structures were studied. Measured properties include strength, stiffness, shrinkage and creep deformations, together with the complete compressive stress-strain characteristics including post-cracking behavior. The repair materials considered in this investigation are commercially available and widely used. These included a high performance non-shrinkable concrete, a mineral based cementitious material with no additives or coarse aggregate size particles, and a cementitious mortar containing styrene acrylic copolymer with fiber additives. Performance comparisons are also made between these materials and plain concrete mixes of similar strength and stiffness, suitable for repair applications. The results show that shrinkage of the repair materials was significantly greater than the shrinkage of normal concrete. Moreover, the shrinkage of those modified with a polymer admixture was found to be very sensitive to the relative humidity of the exposure compared to normal concrete. The post-peak strain capacity of the material modified with a polymer admixture was markedly improved leading to a more pronounced falling branch of stress-strain curve. The ultimate stress level (at a maximum load) of specially formulated repair materials varies significantly, the lowest ultimate stress being recorded for the porous mineral-based material. The inclusion of aggregates improves the mechanical properties and dimensional stability of repair materials.

  7. Failure of distal biceps repair by gapping

    Background We describe the clinical, radiological and surgical findings of failed distal biceps repair by gapping and report the functional outcomes following revision repair. Methods A retrospective review of five consecutive patients was conducted. Patients presented with radial-sided forearm pain after their distal biceps fixation. All patients had less than 5 cm of retraction of the biceps muscle belly, a palpable tendon although the manoeuvre was painful with weakness on resisted supination. Flexed abducted supinated magnetic resonance imaging (FABS MRI) showed a gap between the distal end of the tendon and the footprint on the radial tuberosity. Results Mean FEA score at presentation was 44/100 (35 to 49). Mean time to re-operation was 18 months (range 4 months to 36 months). At revision, the distal end of the tendon was retracted and not making contact with the bone. All cases were revised to an in-bone endobutton repair. Mean postoperative Functional Elbow Assessment (FEA) scores undertaken at a mean of 14 months (range 5 months to 22 months) after revision improved to 95/100 (90 to 100). Conclusions Patients presenting with persistent radial sided forearm pain and weakness on provocative testing after distal biceps repair with a seemingly intact repair should be investigated with FABS MRI to look for evidence of failure of repair by gapping. Revision repair with an anatomic in-bone technique can lead to good results. PMID:27583018

    Arthroscopic rotator cuff repair is being performed by an increasing number of orthopaedic surgeons. The principles, techniques, and instrumentation have evolved to the extent that all patterns and sizes of rotator cuff tear, including massive tears, can now be repaired arthroscopically. Achieving a biomechanically stable construct is critical to biologic healing. The ideal repair construct must optimize suture-to-bone fixation, suture-to-tendon fixation, abrasion resistance of suture, suture strength, knot security, loop security, and restoration of the anatomic rotator cuff footprint (the surface area of bone to which the cuff tendons attach). By achieving optimized repair constructs, experienced arthroscopic surgeons are reporting results equal to those of open rotator cuff repair. As surgeons' arthroscopic skill levels increase through attendance at surgical skills courses and greater experience gained in the operating room, there will be an increasing trend toward arthroscopic repair of most rotator cuff pathology.   10. Shuttle Repair Tools Automate Vehicle Maintenance

    Successfully building, flying, and maintaining the space shuttles was an immensely complex job that required a high level of detailed, precise engineering. After each shuttle landed, it entered a maintenance, repair, and overhaul (MRO) phase. Each system was thoroughly checked and tested, and worn or damaged parts replaced, before the shuttle was rolled out for its next mission. During the MRO period, workers needed to record exactly what needed replacing and why, as well as follow precise guidelines and procedures in making their repairs. That meant traceability, and with it lots of paperwork. In 2007, the number of reports generated during electrical system repairs was getting out of hand-placing among the top three systems in terms of paperwork volume. Repair specialists at Kennedy Space Center were unhappy spending so much time at a desk and so little time actually working on the shuttle. "Engineers weren't spending their time doing technical work," says Joseph Schuh, an electrical engineer at Kennedy. "Instead, they were busy with repetitive, time-consuming processes that, while important in their own right, provided a low return on time invested." The strain of such inefficiency was bad enough that slow electrical repairs jeopardized rollout on several occasions. Knowing there had to be a way to streamline operations, Kennedy asked Martin Belson, a project manager with 30 years experience as an aerospace contractor, to co-lead a team in developing software that would reduce the effort required to document shuttle repairs. The result was System Maintenance Automated Repair Tasks (SMART) software. SMART is a tool for aggregating and applying information on every aspect of repairs, from procedures and instructions to a vehicle s troubleshooting history. Drawing on that data, SMART largely automates the processes of generating repair instructions and post-repair paperwork.     This paper documents Fluor Hanford's use of Leading Indicators, management leadership, and statistical methodology in order to improve safe performance of work. By applying these methods, Fluor Hanford achieved a significant reduction in injury rates in 2003 and 2004, and the improvement continues today. The integration of data, leadership, and teamwork pays off with improved safety performance and credibility with the customer. The use of Statistical Process Control, Pareto Charts, and Systems Thinking and their effect on management decisions and employee involvement are discussed. Included are practical examples of choosing leading indicators. A statistically based color coded dashboard presentation system methodology is provided. These tools, management theories and methods, coupled with involved leadership and employee efforts, directly led to significant improvements in worker safety and health, and environmental protection and restoration at one of the nation's largest nuclear cleanup sites.

    Renovation of long-term grassland is associated with a peak in soil organic N mineralisation which, coupled with diminished plant N uptake can lead to large gaseous and leaching N losses. This study reports on the effect of ploughing and subsequent N fertilisation on the N2O emissions and DON/NO3(-) leaching, and evaluates the impact of ploughing technique on the magnitude and profile of N losses. This study was carried out on isolated grassland lysimeters of three Irish soils representing contrasting drainage properties (well-drained Clonakilty, moderately-drained Elton and poorly-drained Rathangan). Lysimeters were manually ploughed simulating conventional (CT) and minimum tillage (MT) as two treatments. Renovation of grassland increased N2O flux to a maximum of 0.9kgN2O-Nha(-1) from poorly-drained soil over four days after treatment. Although there was no difference between CT and MT in the post-ploughing period, the treatment influenced subsequent N2O after fertiliser applications. Fertilisation remained the major driver of N losses therefore reducing fertilisation rate post-planting to account for N mineralised through grassland renovation could reduce the losses in medium to longer term. Leaching was a significant loss pathway, with the cumulative drainage volume and N leached highly influenced by soil type. Overall, the total N losses (N2O+N leached) were lowest from poorly and moderately draining soil and highest for the well draining soil, reflecting the dominance of leaching on total N losses and the paramount importance of soil properties.     Repair on damaged composite panels was conducted. To better understand adhesively bonded repair, the study investigates the effect of design parameters on the joint strength. The design parameters include bondline length, thickness of adherend and type of adhesive. Adhesives considered in this study were tested to measure their tensile material properties. Three types of adhesively bonded joints, single strap, double strap, and single lap joint were considered under changing bondline lengths, thickness of adherend and type of adhesive. Based on lessons learned from bonded joints, a one-sided patch repair method for composite structures was conducted. The composite patch was bonded to the damaged panel by either film adhesive FM-73M or paste adhesive EA-9394 and the residual strengths of the repaired specimens were compared under varying patch sizes. A new repair method using attachments has been suggested to enhance the residual strength. Results obtained through experiments were analyzed using finite element analysis to provide a better repair design and explain the experimental results. It was observed that the residual strength of the repaired specimen was affected by patch length. Method for rapid repairs of damaged composite structures was investigated. The damage was represented by a circular hole in a composite laminated plate. Pre-cured composite patches were bonded with a quick-curing commercial adhesive near (rather than over) the hole. Tensile tests were conducted on specimens repaired with various patch geometries. The test results showed that, among the methods investigated, the best repair method restored over 90% of the original strength of an undamaged panel. The interfacial stresses in the adhesive zone for different patches were calculated in order to understand the efficiencies of the designs of these patch repairs.
