    MuB is an ATP-dependent DNA-binding protein that regulates the activity of MuA transposase and delivers the target DNA for transposition of phage Mu. Mechanistic insight into MuB function is limited to its AAA+ ATPase module, which upon ATP binding assembles into helical filaments around the DNA. However, the structure and function of the flexible N-terminal domain (NTD) appended to the AAA+ module remains uncharacterized. Here we report the solution structure of MuB NTD determined by NMR spectroscopy. The structure reveals a compact domain formed by four α-helices connected by short loops, and confirms the presence of a helix-turn-helix motif. High structural similarity and sequence homology with λ repressor-like DNA-binding domains suggest a possible role of MuB NTD in DNA binding. We also demonstrate that the NTD directly mediates the ability of MuB to establish filament-filament interactions. These findings lead us to a model in which the NTD interacts with the AAA+ spirals and perhaps also with the DNA bound within the filament, favoring MuB polymerization and filament clustering. We propose that the MuB NTD-dependent filament interactions might be an effective mechanism to bridge distant DNA regions during Mu transposition. PMID:26169224

    All possible MUB entanglement patterns for systems of N prime-state particles are obtained from standard ones by unitary transformations in the Hilbert space, thus preserving the relationships between the generalized Pauli operators, the phase point operators, and the MUB projectors. The transformations are described geometrically in discrete phase space. Illustrative examples show the invariance of the total entanglement content and the connection of entanglement with Galois fields. Different field representations for the same dimension may produce inequivalent MUB sets. This work provides alternative constructions and generalizes previous work on qubit systems [1,2].

  4. SIC-POVMS and MUBS: Geometrical Relationships in Prime Dimension

    The paper concerns Weyl-Heisenberg covariant SIC-POVMs (symmetric informationally complete positive operator valued measures) and full sets of MUBs (mutually unbiased bases) in prime dimension. When represented as vectors in generalized Bloch space a SIC-POVM forms a d{sup 2}-1 dimensional regular simplex (d being the Hilbert space dimension). By contrast, the generalized Bloch vectors representing a full set of MUBs form d+1 mutually orthogonal d-1 dimensional regular simplices. In this paper we show that, in the Weyl-Heisenberg case, there are some simple geometrical relationships between the single SIC-POVM simplex and the d+1 MUB simplices. We go on to give geometrical interpretations of the minimum uncertainty states introduced by Wootters and Sussman, and by Appleby, Dang and Fuchs, and of the fiduciality condition given by Appleby, Dang and Fuchs.

    We exhibit an infinite family of triplets of mutually unbiased bases (MUBs) in dimension 6. These triplets involve the Fourier family of Hadamard matrices, F(a, b). However, in the main result of this paper we also prove that for any values of the parameters (a, b), the standard basis and F(a, b) cannot be extended to a MUB-quartet. The main novelty lies in the method of proof which may successfully be applied in the future to prove that the maximal number of MUBs in dimension 6 is three.

  11. MuA-mediated in vitro cloning of circular DNA: transpositional autointegration and the effect of MuB.

    Transposons provide useful tools for genetics and genomics studies, as they can be modified easily for a variety of purposes. In this study, a strategy to clone circular DNA was developed on the basis of an efficient Mu in vitro transposition reaction catalyzed by MuA transposase. The transposon used contains a selectable marker as well as an origin of replication, and in vitro integration of the transposon into circular DNA generates a plasmid that can replicate in E. coli. We show that the substrate stoichiometry plays an important role in the profile of intermolecular versus intramolecular transposition reaction products. Increasing the relative amount of target DNA reduced the frequency of intramolecular products that are non-productive with regard to the developed cloning application. Such autointegration was also reduced in the reactions containing phage Mu-encoded MuB, indicating that this protein can be used for cloning in combination with MuA, and it is particularly useful with a limited amount of target DNA. The developed strategy can now be utilized to clone DNA circles regardless of their origin as long as their size is not prohibitive for transformation. PMID:26847688

  12. Mechanism of chromophore assisted laser inactivation employing fluorescent proteins.

    Chromophore assisted laser inactivation (CALI) is a technique that uses irradiation of chromophores proximate to a target protein to inactivate function. Previously, enhanced green fluorescent protein (EGFP) mediated CALI has been used to inactivate EGFP-fusion proteins in a spatio-temporally defined manner within cells, but the mechanism of inactivation is unknown. To help elucidate the mechanism of protein inactivation mediated by fluorescent protein CALI ([FP]-CALI), the activities of purified glutathione-S-transferase-FP (GST-EXFP) fusions were measured after laser irradiation in vitro. Singlet oxygen and free radical quenchers as well as the removal of oxygen inhibited CALI, indicating the involvement of a reactive oxygen species (ROS). At higher concentrations of protein, turbidity after CALI increased significantly indicating cross-linking of proximate fusion proteins suggesting that damage of residues on the surface of the protein, distant from the active site, results in inactivation. Control experiments removed sample heating as a possible cause of these effects. Different FP mutants fused to GST vary in their CALI efficiency in the order enhanced green fluorescent protein (EGFP) > enhanced yellow fluorescent protein (EYFP) > enhanced cyan fluorescent protein (ECFP), while a GST construct that binds fluorescein-based arsenical hairpin binder (FlAsH) results in significantly higher CALI efficiency than any of the fluorescent proteins (XFPs) tested. It is likely that the hierarchy of XFP effectiveness reflects the balance between ROS that are trapped within the XFP structure and cause fluorophore and chromophore bleaching and those that escape to effect CALI of proximate proteins.     The study explores the pathways and mechanisms of the relation between employment conditions and health inequalities. A significant amount of published research has proved that workers in several risky types of labor--precarious employment, unemployment, informal labor, child and bonded labor--are exposed to behavioral, psychosocial, and physio-pathological pathways leading to physical and mental health problems. Other pathways, linking employment to health inequalities, are closely connected to hazardous working conditions (material and social deprivation, lack of social protection, and job insecurity), excessive demands, and unattainable work effort, with little power and few rewards (in salaries, fringe benefits, or job stability). Differences across countries in the social contexts and types of jobs result in varying pathways, but the general conceptual model suggests that formal and informal power relations between employees and employers can determine health conditions. In addition, welfare state regimes (unionization and employment protection) can increase or decrease the risk of mortality, morbidity, and occupational injury. In a multilevel context, however, these micro- and macro-level pathways have yet to be fully studied, especially in middle- and low-income countries. The authors recommend some future areas of study on the pathways leading to employment-related health inequalities, using worldwide standard definitions of the different forms of labor, authentic data, and a theoretical framework.   17. Maternal employment and childhood obesity: a search for mechanisms in time use data.

    A substantial body of research documents that maternal employment is associated with childhood obesity. This paper explores possible mechanisms for that correlation in the American Time Use Survey (ATUS). We find that maternal employment is associated with working mothers spending, per day, 4 fewer minutes grocery shopping, 17 fewer minutes cooking, 10 fewer minutes eating with children, 12 fewer minutes playing with children, 4 fewer minutes supervising children, and 37 fewer minutes caring for children. The differences tend to be greatest for mothers with young children (age 0-5 years). We explore the extent to which these findings differ by day of the week, whether a partner or spouse is present in the household, whether the mother works non-standard hours, and socioeconomic status. Only a small percentage (about 15%) of the fewer minutes spent in these activities by working mothers appears to be offset by increases in time by husbands and partners. These findings suggest plausible mechanisms for the association between maternal employment and childhood obesity. PMID:22790446

  18. Grain boundary character modification employing thermo-mechanical processing in type 304L stainless steel

    Grain boundary engineering (GBE) approach has been employed to modify the boundaries character of a type 304L stainless steel through thermo-mechanical processing (TMP) route, which combined a low level of cold deformation (5, 10 and 15%) followed by annealing at 1173K and 1273K for 1hour. Employing Electron Back Scatter Diffraction based Orientation Imaging Microscopy, the fraction and distribution of low ∑ CSL boundaries (∑≤ 29) and its effect on random high-angle grain boundaries connectivity and triple junction distribution of as-received (AR) and GBE specimens were evaluated. It was possible to increase the fraction of low ∑ CSL boundaries up to 75% following GBE treatment (as compared to 50% in AR specimen). The GBE specimens also contained maximum number of percolation resistant triple junctions which could render better resistance against percolation related phenomena.

    The different possibilities of orientation of the systems of axes currently employed in flight mechanics are compiled and described. Of the three possible couplings between the wind and aircraft axes, the most suitable coupling is that in which the y axis is made the principal axis of rotation for one of the two coupling angles (angles of attack). In connection with this coupling, an experimental system of axes is introduced, whose axes x(sub e) and z(sub e) are situated in the plane of symmetry of the airplane and rotate about the airplane lateral axis y = y(sub e). This system of axes enables the utilization of the coefficients obtained in the wind tunnel in the flight-mechanic equations by a simple transformation, with the aid of the angle of attack, measured in the plane of symmetry of the airplane.

    Current international interest in finding alternative sources of energy to the diminishing supplies of fossil fuels has encouraged research efforts in improving biofuel production technologies. In countries which lack sufficient food, the use of sustainable lignocellulosic feedstocks, for the production of bioethanol, is an attractive option. In the pre-treatment of lignocellulosic feedstocks for ethanol production, various chemicals and/or enzymatic processes are employed. These methods generally result in a range of fermentable sugars, which are subjected to microbial fermentation and distillation to produce bioethanol. However, these methods also produce compounds that are inhibitory to the microbial fermentation process. These compounds include products of sugar dehydration and lignin depolymerisation, such as organic acids, derivatised furaldehydes and phenolic acids. These compounds are known to have a severe negative impact on the ethanologenic microorganisms involved in the fermentation process by compromising the integrity of their cell membranes, inhibiting essential enzymes and negatively interact with their DNA/RNA. It is therefore important to understand the molecular mechanisms of these inhibitions, and the mechanisms by which these microorganisms show increased adaptation to such inhibitors. Presented here is a concise overview of the molecular adaptation mechanisms of ethanologenic bacteria in response to lignocellulose-derived inhibitory compounds. These include general stress response and tolerance mechanisms, which are typically those that maintain intracellular pH homeostasis and cell membrane integrity, activation/regulation of global stress responses and inhibitor substrate-specific degradation pathways. We anticipate that understanding these adaptation responses will be essential in the design of 'intelligent' metabolic engineering strategies for the generation of hyper-tolerant fermentation bacteria strains.   2. Fuzzy clustering: critical analysis of the contextual mechanisms employed by three neural network models

    According to the following definition, taken from the literature, a fuzzy clustering mechanism allows the same input pattern to belong to multiple categories to different degrees. Many clustering neural network (NN) models claim to feature fuzzy properties, but several of them (like the Fuzzy ART model) do not satisfy this definition. Vice versa, we believe that Kohonen's Self-Organizing Map, SOM, satisfies the definition provided above, even though this NN model is well-known to (robustly) perform topologically ordered mapping rather than fuzzy clustering. This may sound as a paradox if we consider that several fuzzy NN models (such as the Fuzzy Learning Vector Quantization, FLVQ, which was first called Fuzzy Kohonen Clustering Network, FKCN) were originally developed to enhance Kohonen's models (such as SOM and the vector quantization model, VQ). The fuzziness of SOM indicates that a network of processing elements (PEs) can verify the fuzzy clustering definition when it exploits local rules which are biologically plausible (such as the Kohonen bubble strategy). This is equivalent to state that the exploitation of the fuzzy set theory in the development of complex systems (e.g., clustering NNs) may provide new mathematical tools (e.g., the definition of membership function) to simulate the behavior of those cooperative/competitive mechanisms already identified by neurophysiological studies. When a biologically plausible cooperative/competitive strategy is pursued effectively, neighboring PEs become mutually coupled to gain sensitivity to contextual effects. PEs which are mutually coupled are affected by vertical (inter-layer) as well as horizontal (intra-layer) connections. To summarize, we suggest to relate the study of fuzzy clustering mechanisms to the multi-disciplinary science of complex systems, with special regard to the investigation of the cooperative/competitive local rules employed by complex systems to gain sensitivity to contextual effects in

    Mental healthcare domain highlights the significance of trustworthiness between patient and psychiatrist for treatment process. In this paper, the issue of assessing psychiatrist trustworthiness from patient perspective, within a Ubiquitous Healthcare (UH) environment, is addressed. To meet that challenge, a Trust Assessment mechanism mimicking human cognitive judgment, is proposed. The exploitation of innovative fuzzy-probabilistic transformation model, denoted as cloud, for mechanism deployment enables fuzziness as well as adhered randomness of cognitive perception and assessment to be captured.     Owing to their high thermoelectric (TE) figure-of-merit, nanostructured Si{sub 80}Ge{sub 20} alloys are evolving as a potential replacement for their bulk counterparts in designing efficient radio-isotope TE generators. However, as the mechanical properties of these alloys are equally important in order to avoid in-service catastrophic failure of their TE modules, we report the strength, hardness, fracture toughness, and thermal shock resistance of nanostructured n-type Si{sub 80}Ge{sub 20} alloys synthesized employing spark plasma sintering of mechanically alloyed nanopowders of its constituent elements. These mechanical properties show a significant enhancement, which has been correlated with the microstructural features at nano-scale, delineated by transmission electron microscopy.

    Over the past few decades, the use of portable and wearable electronics has grown steadily. These devices are becoming increasingly more powerful, however, the gains that have been made in the device performance has resulted in the need for significantly higher power to operate the electronics. This issue has been further complicated due to the stagnate growth of battery technology over the past decade. In order to increase the life of these electronics, researchers have begun investigating methods of generating energy from ambient sources such that the life of the electronics can be prolonged. Recent developments in the field have led to the design of a number of mechanisms that can be used to generate electrical energy, from a variety of sources including thermal, solar, strain, inertia, etc. Many of these energy sources are available for use with humans, but their use must be carefully considered such that parasitic effects that could disrupt the user's gait or endurance are avoided. This study develops a novel energy harvesting backpack that can generate electrical energy from the differential forces between the wearer and the pack. The goal of this system is to make the energy harvesting device transparent to the wearer such that his or her endurance and dexterity is not compromised. This will be accomplished by replacing the strap buckle with a mechanically amplified piezoelectric stack actuator. Piezoelectric stack actuators have found little use in energy harvesting applications due to their high stiffness which makes straining the material difficult. This issue will be alleviated using a mechanically amplified stack which allows the relatively low forces generated by the pack to be transformed to high forces on the piezoelectric stack. This paper will develop a theoretical model of the piezoelectric buckle and perform experimental testing to validate the model accuracy and energy harvesting performance.

    CRISPR-Cas9 and CRISPR-Cpf1 systems have been successfully harnessed for genome editing. In the CRISPR-Cas9 system, the preordered A-form RNA seed sequence and preformed protein PAM-interacting cleft are essential for Cas9 to form a DNA recognition-competent structure. Whether the CRISPR-Cpf1 system employs a similar mechanism for target DNA recognition remains unclear. Here, we have determined the crystal structure of Acidaminococcus sp. Cpf1 (AsCpf1) in complex with crRNA and target DNA. Structural comparison between the AsCpf1-crRNA-DNA ternary complex and the recently reported Lachnospiraceae bacterium Cpf1 (LbCpf1)-crRNA binary complex identifies a unique mechanism employed by Cpf1 for target recognition. The seed sequence required for initial DNA interrogation is disordered in the Cpf1-cRNA binary complex, but becomes ordered upon ternary complex formation. Further, the PAM interacting cleft of Cpf1 undergoes an "open-to-closed" conformational change upon target DNA binding, which in turn induces structural changes within Cpf1 to accommodate the ordered A-form seed RNA segment. This unique mechanism of target recognition by Cpf1 is distinct from that reported previously for Cas9. PMID:27444870

    This invention relates to novel reciprocating shuttle inlet valves, effective with every type of two-cycle engine, from small high-speed single cylinder model engines, to large low-speed multiple cylinder engines, employing spark or compression ignition. Also permitting the elimination of out-of-phase piston arrangements to control scavenging and supercharging of opposed-piston engines. The reciprocating shuttle inlet valve (32) and its operating mechanism (34) is constructed as a single and simple uncomplicated member, in combination with the lost-motion abutments, (46) and (48), formed in a piston skirt, obviating the need for any complex mechanisms or auxiliary drives, unaffected by heat, friction, wear or inertial forces. The reciprocating shuttle inlet valve retains the simplicity and advantages of two-cycle engines, while permitting an increase in volumetric efficiency and performance, thereby increasing the range of usefulness of two-cycle engines into many areas that are now dominated by the four-cycle engine.

    The mechanism of hydrogenated microcrystalline silicon (µc-Si:H) film deposition by magnetron sputtering employing a Si target and H2/Ar gas mixture has been investigated by measuring Si and H atom densities in the gas phase by laser-induced fluorescence spectroscopy. The crystalline volume fraction of the film correlated positively with H atom density. The variation in Si atom density indicated the increase in sputtering yield from the Si target in the H2/Ar discharge. The surface of the Si target immersed in the H2/Ar discharge was hydrogenated. Therefore, it is reasonable to expect the production of SiHx molecules (typically SiH4) from the hydrogenated Si target via reactive ion etching. Since SiHx molecules produced from the target may function as a deposition precursor, the mechanism of µc-Si:H film deposition is considered to be similar to that of plasma-enhanced chemical vapor deposition (PECVD) employing a SiH4/H2 gas mixture. The advantage of magnetron sputtering deposition over PECVD is the production of SiHx molecules without using toxic, explosive SiH4.

    Recently, China has frequently experienced large-scale, severe and persistent haze pollution due to surging urbanization and industrialization and a rapid growth in the number of motor vehicles and energy consumption. The vehicle emission due to the consumption of a large number of fossil fuels is no doubt a critical factor of the haze pollution. This work is focused on the causation mechanism of haze pollution related to the vehicle emission for Guangzhou city by employing the Fault Tree Analysis (FTA) method for the first time. With the establishment of the fault tree system of "Haze weather-Vehicle exhausts explosive emission", all of the important risk factors are discussed and identified by using this deductive FTA method. The qualitative and quantitative assessments of the fault tree system are carried out based on the structure, probability and critical importance degree analysis of the risk factors. The study may provide a new simple and effective tool/strategy for the causation mechanism analysis and risk management of haze pollution in China. PMID:26923237

Here, we provide novel evidence demonstrating that mindfulness meditation produces greater pain relief and employs distinct neural mechanisms than placebo cream and sham mindfulness meditation. Specifically, mindfulness meditation-induced pain relief activated higher-order brain regions, including the orbitofrontal and cingulate cortices. In contrast, placebo analgesia was associated with decreased pain-related brain activation. These findings demonstrate that mindfulness meditation reduces pain through unique mechanisms and may foster greater acceptance of meditation as an adjunct pain therapy. PMID:26586819

  14. Resolving the mechanism of bacterial inhibition by plant secondary metabolites employing a combination of whole-cell biosensors.

    Tightening regulations regarding the use of biocides have stimulated interest in investigating alternatives to current antimicrobial strategies. Plant essential oils and their constituent compounds are promising candidates as novel antimicrobial agents because of their excellent ability in killing microbes while being non-toxic to humans at antimicrobially-active concentrations. Allyl isothiocyanate (AIT), carvacrol, cinnamaldehyde (CNAD), citral, and thymol were investigated for their antibacterial activity against Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. The five compounds were screened via disc diffusion assay and broth microdilution method, by which inhibition zone diameters, minimum inhibitory concentrations (MICs), and minimum bactericidal concentrations (MBCs) were determined. AIT and CNAD displayed the greatest inhibitory effects against all species tested, with AIT yielding MICs of 156.25mg/L and MBCs of 156.25 to 312.5mg/L, and CNAD yielding MICs of 78.125 to 156.25mg/L and MBCs of 78.125 to 312.5mg/L. Based on these results, AIT and CNAD were selected for closer examination of their toxic effects. Two complementary bioluminescence-based bacterial biosensors, E. coli HB101_pUCD607_lux and Acinetobacter baylyi ADP1_recA_lux, were employed to examine the dose-response relationships and mechanism of action of AIT and CNAD. This is the first reported study to employ a lux-based biosensor assay coupled with parallel plate count experiments to demonstrate that AIT and CNAD not only damaged cell membranes, but also disrupted cellular metabolism and energy production in bacteria. It is also the first to use genotoxicity-sensing whole-cell bioreporters to demonstrate that neither AIT nor CNAD induced expression of the universal DNA repair gene, recA.     Evasion of complement-mediated opsonophagocytosis enables group A Streptococcus pyogenes (GAS) to establish infection. Different strain-dependent mechanisms are employed by the host to accomplish this goal. In general, GAS inhibits the amplification of the complement cascade on its cell surface by facilitating the degradation of C3b, an opsonin, to an inactive product, inactivated C3b (iC3b), in a step catalyzed by factor I (FI) and its cofactor, factor H (FH), with or without the participation of human host plasmin (hPm). GAS recruits FH to its cell surface via FH receptors, which are transcriptionally controlled by the two-component cluster of virulence responder-sensor system. The manner in which FI-FH and hPm function together on GAS cells is unknown. Using GAS strain AP53, which strongly binds host human plasminogen/plasmin (hPg/hPm) directly via an hPg/hPm surface receptor (PAM), we show that both FI-FH and hPm sequentially cleave C3b. Whereas FI-FH proteolytically cleaves C3b into iC3b, PAM-bound hPm catalyzes cleavage of iC3b into multiple smaller peptides. Unlike AP53, GAS strain M23ND weakly binds FH and recruits hPg/hPm to its cell surface indirectly via fibrinogen bound to M-protein, M23. In this case, FH-FI cleaves C3b into iC3b, with negligible degradation of iC3b by hPm that is bound to fibrinogen on the cells. AP53 and M23ND display similar resistance to human neutrophil-mediated phagocytosis, which results in a corresponding high lethality in mice after injection of these cells. These results suggest that GAS utilizes diverse mechanisms to degrade C3b and thus to protect bacterial cells from the complement response of the host. PMID:26945067

  19. A novel approach to explain the inactivation mechanism of Escherichia coli employing a commercially available peracetic acid.

    The chemical inactivation of Escherichia coli employing a commercial mixture of peracetic acid (PAA) was studied. For this purpose, experiments were carried out using dilutions of the unmodified mixture, and also the same mixture but altered with hydrogen peroxide (HP) previously inhibited. Also, these results were compared to those obtained before employing HP alone. It was found that the mixture is much more efficient than HP and PAA acting separately. Furthermore, it was found that PAA without HP is much more efficient than HP alone. A plausible explanation is presented. The homolysis of PAA would give rise to a chain reaction that generates a significant number of highly oxidizing radicals. An attacking scheme to bacteria in two stages is proposed, where the initial step, mainly caused by PAA, is very fast and eliminates some specific components of the bacteria that would otherwise inhibit the parallel action of HP. Thereafter, the emergence of a potentiating synergetic action of the second oxidant seems to be immediately unveiled. PMID:24473306

    To eliminate the effects of motion coupling for measuring cylindrical work pieces, a novel alignment mechanism integrating functions of both leveling and centering is designed and fabricated by introducing multi-layered orthogonal connected flexible hinges as the key supporting and joining elements. Different from traditional leveling mechanisms with many separate parts fabricated together, all of the flexible hinges were integrated in one three-dimensioned machining part without assembling process, and thus synchronously simplifying the structure and reducing assembly errors. Based on the screw theory, the mathematic model of the proposed alignment mechanism is established for any resolution requirements depending on screw characteristics. A millimeter-sized device is fabricated with the alignment precision of 1.0 μm for centering within the range of ±1 mm and 1 in. for leveling within ±1°. The experiment results are in very close agreement to those obtained by simulation, which validate the feasibility of introducing multi-layered orthogonal flexible hinges in the centering and leveling mechanisms.   2. A wavelet Galerkin method employing B-spline bases for solid mechanics problems without the use of a fictitious domain

    This study develops a wavelet Galerkin method (WGM) that uses B-spline wavelet bases for application to solid mechanics problems. A fictitious domain is often adopted to treat general boundaries in WGMs. In the analysis, the body is extended to its exterior but very low stiffness is applied to the exterior region. The stiffness matrix in the WGM becomes singular without the use of a fictitious domain. The problem arises from the lack of linear independence of the basis functions. A technique to remove basis functions that can be represented by the superposition of the other basis functions is proposed. The basis functions are automatically eliminated in the pre conditioning step. An adaptive strategy is developed using the proposed technique. The solution is refined by superposing finer wavelet functions. Numerical examples of solid mechanics problems are presented to demonstrate the multiresolution properties of the WGM.

    Here we discuss results obtained as part of a three-year investigation at Pacific Northwest National Laboratory of ultrasound processing to effect selectivity and activity in the hydrogenation of water-soluble olefins on transition metal catalysts. We have shown previously that of the two regimes for ultrasound processing, high-power cavitating and high-power non-cavitating, only the former can effect product selectivity dramatically (> 1000%) whereas the selectivity of the latter was comparable with those obtained in stirred/silent control experiments [R.S. Disselkamp, Y.-H. Chin, C.H.F. Peden, J. Catal., 227, 552 (2005)]. As a means of ensuring the benefits of cavitating ultrasound processing, we introduced the concept of employing inert dopants into the reacting solution. These inert dopants do not partake in solution chemistry but enable a more facile transition from high-power non-cavitating to cavitating conditions during sonication treatment. With cavitation processing conditions ensured, we discuss here results of isotopic H/D substitution for a variety of substrates and illustrate how such isotope dependent chemistries during substrate hydrogenation elucidate detailed mechanistic information about these reaction systems.

    The correlation between the composition, aging heat treatments, microstructural features and mechanical properties of β Ti alloys is of primary significance because it is the foundation for developing and improving new Ti alloys for orthopedic biomaterials. However, in the case of Ti-Mo alloys, this correlation is not fully described in the literature. Therefore, the purpose of this study was to experimentally investigate the effect of composition and aging heat treatments on the microstructure, Vickers hardness and elastic modulus of Ti-Mo alloys. These alloys were solution heat-treated and water-quenched, after which their response to aging heat treatments was investigated. Their microstructure, Vickers hardness and elastic modulus were evaluated, and the results allow us to conclude that stabilization of the β phase is achieved with nearly 10% Mo when a very high cooling rate is applied. Young's modulus was found to be more sensitive to phase variations than hardness. In all of the compositions, the highest hardness values were achieved by aging at 723K, which was attributed to the precipitation of α and ω phases. All of the compositions aged at 573K, 623K and 723K showed overaging within 80h. PMID:24394773

  6. A novel procedure employing laser ultrasound technique and simplex algorism for the characterization of mechanical and geometrical properties in Zircaloy tubes with different levels of hydrogen charging

    In this research, a procedure employing a laser ultrasound technique (LUT) and an inversion algorism is reported for nondestructive characterization of mechanical and geometrical properties in Zircaloy tubes with different levels of hydrogen charging. With the LUT, guided acoustic waves are generated to propagate in the Zircaloy tubes and are detected remotely by optical means. By measuring the dispersive wavespeeds followed by the inversion algorism, mechanical properties such as elastic moduli and geometrical property such as wall-thickness of Zircaloy tubes are characterized for different levels of hydrogen charging. Having the advantages of remote, non-contact and point-wise generation/detection, the reported procedure serves as a competitive candidate for the characterization of Zircaloy tubes generally operated in irradiative and temperature-elevated environments.

  11. A Potential of Rail Vehicle Having Bolster with Side Bearers for Improving Curving Performance on Sharp Curves Employing Link-Type Forced Steering Mechanism

    The air spring of bolsterless bogie trucks, which have been widely employed in railway vehicles in recent years, undergoes a large distortion when the vehicles negotiate sharp curves in lines such as subway lines, and this can deteriorate the durability of air springs. Furthermore, bolsterless trucks tend to suffer from increased wheel lateral force around sharp curves with a radius of 100 m or less. In this paper we discuss the application of a link-type forced steering mechanism to bogie trucks with a bolster as a countermeasure against the above-mentioned situation. A numerical simulation is carried out using a MBS software, SIMPACK. As a result, under the condition of reduced longitudinal stiffness in the primary suspension, a bolster truck with the link-type steering mechanism exhibits the potential to suppress the wheel lateral force occurring around sharp curves. Also, the deterioration in running stability due to the application of the steering mechanism can be recovered by adding moderate lateral damping in the secondary suspension. In addition, the obtained wear index shows that the forced steering truck has decreased flange wear resulting from passing through sharp curves.

    Infant rats switch randomly between the sleeping and waking states; during early infancy (up to postnatal day 8), sleep and wake bouts are random, brief (with means on the order of several seconds) and exponentially distributed, with the length of a particular bout independent of the length of prior bouts. As the rat ages during this early period, mean sleep and wake bout lengths gradually increase, though sleep and wake bouts remain exponentially distributed. Additionally, sleep and wake bouts are regulated independently of each other - alterations in the development of sleep (wake) bouts has no impact on the regulation wake (sleep) bouts. Sleep and wake bout behavior is associated with the activity of mutually inhibitory sleep-active and wake-active brainstem populations. In this work, I employ a simplified biophysical model of two mutually inhibitory populations consisting of ten integrate-and-fire neurons each and a noise-based switching mechanism. I show that such a noise-based switching mechanism naturally accounts for the experimentally observed features of sleep-wake switching during early infancy - random alternating activity bouts occur as a consequence of noise (provided inhibition is strong relative to excitation), bout durations are exponential (due to a lack of memory within the system), and cross-population inhibition or intrapopulation excitatory coupling provide mechanisms for changing and independently regulated sleep and wake bout means. Reaction dynamics calculations were carried out for the testosterone 6β-hydrogen abstraction and the 6β-d1-testosterone 6β-duterium abstraction employing a model that consists of the substrate and the active oxidant compound I. The calculations were performed at the level of canonical variational transition state theory with multidimensional tunneling and were based on a semiglobal full-dimensional potential energy surface generated by the multiconfiguration molecular mechanics technique. The tunneling coefficients were found to be around 3, indicating substantial contributions by quantum tunneling. However, the tunneling made only modest contributions to the kinetic isotope effects.     This study aimed at elucidating an optimal synergistic polymer composite for achieving a desirable molecular bioadhesivity and Matrix Erosion of a bioactive-loaded Intravaginal Bioadhesive Polymeric Device (IBPD) employing Molecular Mechanic Simulations and Artificial Neural Networks (ANN). Fifteen lead caplet-shaped devices were formulated by direct compression with the model bioactives zidovudine and polystyrene sulfonate. The Matrix Erosion was analyzed in simulated vaginal fluid to assess the critical integrity. Blueprinting the molecular mechanics of bioadhesion between vaginal epithelial glycoprotein (EGP), mucin (MUC) and the IBPD were performed on HyperChem 8.0.8 software (MM+ and AMBER force fields) for the quantification and characterization of correlative molecular interactions during molecular bioadhesion. Results proved that the IBPD bioadhesivity was pivoted on the conformation, orientation, and poly(acrylic acid) (PAA) composition that interacted with EGP and MUC present on the vaginal epithelium due to heterogeneous surface residue distributions (free energy= -46.33 kcalmol(-1)). ANN sensitivity testing as a connectionist model enabled strategic polymer selection for developing an IBPD with an optimally prolonged Matrix Erosion and superior molecular bioadhesivity (ME = 1.21-7.68%; BHN = 2.687-4.981 N/mm(2)). Molecular modeling aptly supported the EGP-MUC-PAA molecular interaction at the vaginal epithelium confirming the role of PAA in bioadhesion of the IBPD once inserted into the posterior fornix of the vagina.     Cerebral venous sinus thrombosis (CVST) can be life threatening. A previously healthy woman in her early forties on oral contraceptives presented with global CVST and rapid clinical deterioration. A novel 'Triaxial Angiojet technique' (KSAW Shuttle [Cook Inc., Bloomington, IN, USA], 5 MAX ACE [Penumbra Inc., Alameda, CA, USA], and Angiojet [Boston Scientific, Marlborough, MA, USA]) was employed to gain access into the superior sagittal sinus. The 5 MAX ACE reperfusion catheter was shortened prior to placing a 4 Fr Angiojet catheter through it. This resulted in markedly improved recanalization with good anterograde flow. The patient was extubated on postoperative day 2 and discharged neurologically intact on postoperative day 10. We report the first case of placing an Angiojet catheter through a larger Penumbra reperfusion catheter when access through a tortuous sigmoid and transverse sinus could not be obtained with a 6 Fr support catheter.     Due to risk of environmental and biological accumulation of Cadmium (Cd), improved methods of early detection and monitoring must be explored as a preventative measure. Listed as one of the top three toxic heavy metals by the Environmental Protection Agency (EPA), the effects on ecological and human systems have well documented side-effects of physical mutation, reproductive sterility, kidney failure, liver disease, bone loss, and death. Found in batteries, metal plating, pigments, plastics, and cigarettes, Cd is also used as a neutron absorber in the nuclear industry as well as having 3 known radioactive isotopes. Urine Cd levels, which have been widely used to predict whole body levels, increase when kidney damage occurs, thus increasing the importance to monitor and detect as early as possible. Although several methods of detection and monitoring are currently in use, they are insufficient for reasons including massive expense, weak specificity causing false readings, and/or a lack of portability. By exploiting naturally occurring mechanisms known to micro-/nanofluidics, a novel approach to Cd detection, measurement, and preconcentration was explored using the finite element computational software COMSOL. An open flow system of a nanochannel was explored through manipulation of the surface charge density. With a dominant negatively charged density on the walls, positive surface charge densities were adjacently placed at the center of the nanochannel causing a constriction of flow and allowing preconcentration of the analytes. When the open flow system was scaled up to a microchannel, the mechanism was found to have little effect on constriction of the flow. A preconcentration effect was discovered in a closed flow system when the adjacent patches were modeled as being impermeable to charge, causing the molecules to migrate to and remain at the central region of the microchannel once the dynamic process reached steady state.     Using the Early Childhood Longitudinal Survey-Kindergarten Class of 1998-1999 (ECLS-K) data from kindergarten through eighth grade, this paper investigate the relationships among maternal employment, family routines and obesity. More hours worked by the mother tend to be negatively related to positive routines like eating meals as a family or at regular times, or having family rules about hours of television watched. Many of these same routines are significantly related to the probability of being obese, implying that family routines may be a mechanism by which maternal employment intensity affects children's obesity. However, inclusion of family routines in the obesity regression does not appreciably change the estimated effect of maternal employment hours. Thus, the commonly estimated deleterious effect of maternal employment on children's obesity cannot be explained by family routines, leaving the exact mechanisms an open question for further exploration.     This paper reviews various published reports from surveys on employer opinion, perception of needs, and trends with regard to healthcare benefits; the consumer perspective regarding healthcare is also discussed. Surveys indicate that businesses want continuous evidence that high-quality healthcare can positively impact company profits. Employers and labor unions are demanding more cost-effective healthcare. At both employer and consumer levels, greater patient education is needed, as well as traditional educational media. Direct-to-consumer advertising and use of the World Wide Web are increasingly important in enabling consumers to participate more fully in their own lipid-related decision-making. Finally, the transition of lipid-lowering drugs to over-the-counter accessibility has great implications with respect to issues of patient preferences and willingness to pay in the evolving healthcare environment. Groups in the United States, such as the National Committee on Quality Assurance and the Foundation for Accountability, are setting standards and beginning to assess both process and outcomes in patient care. Further collaborative efforts are needed that raise standards of care and stimulate more cost-effective healthcare. The pharmacoeconomics and outcomes data gathered will, one hopes, also demonstrate to global businesses the positive financial impact of high-quality healthcare and appropriate lipid therapy.     Employment outcomes of individuals participating in 17 Massachusetts Clubhouses certified by the International Center for Clubhouse Development were examined through an annual survey. Major components of employment programs in contemporary clubhouses are identified and individual employment outcomes are described. Within contemporary practice in ICCD clubhouses in this sample, clubhouses provided a three-pronged approach to employment. Between 1998-2001, 1702 individuals worked in 2714 separate job placements, employed in Transitional (TE), Supported (SE), and Independent Employment (IE). Forty percent of members with more than one job (N = 385) participated in at least one TE. Individuals with longer memberships tended to work longer and had higher job earnings. PMID:16075694

    Amblyopia ("lazy eye") is a major public health problem, caused by misalignment of the eyes (strabismus) or defocus. If detected early in childhood, there is an excellent response to therapy, yet most children are detected too late to be treated effectively. Commercially available vision screening devices that test for amblyopia's primary causes can detect strabismus only indirectly and inaccurately via assessment of the positions of external light reflections from the cornea, but they cannot detect the anatomical feature of the eyes where fixation actually occurs (the fovea). Our laboratory has been developing technology to detect true foveal fixation, by exploiting the birefringence of the uniquely arranged Henle fibers delineating the fovea using retinal birefringence scanning (RBS), and we recently described a polarization-modulated approach to RBS that enables entirely direct and reliable detection of true foveal fixation, with greatly enhanced signal-to-noise ratio and essentially independent of corneal birefringence (a confounding variable with all polarization-sensitive ophthalmic technology). Here, we describe the design and operation of a new pediatric vision screener that employs polarization-modulated, RBS-based strabismus detection and bull's eye focus detection with an improved target system, and demonstrate the feasibility of this new approach.

    In recent years, the idea of preconception care-education, counseling, and interventions delivered to women before they become pregnant--has gained traction as a critically important health promotion opportunity for women and their families. Employers, as purchasers of health care and as providers of wellness services, have an important role to play in the promotion of preconception care. Large, self-insured employers can craft their medical benefit plans to include evidence-informed preventive health benefits such as preconception care. Employers can also design and implement worksite health promotion programs that address preconception, pregnancy, and postpartum health. And employers of all sizes can educate women and their partners on pregnancy health through tailored communication. This article provides an overview of the business case for preconception care and concrete steps employers can take to support and incent preconception care among their beneficiaries. The article also includes suggestions on ways providers and health professionals support employers in these efforts. PMID:18977153

  14. Deterministic phase retrieval employing spherical illumination

    Deterministic Phase Retrieval techniques (DPRTs) employ a series of paraxial beam intensities in order to recover the phase of a complex field. These paraxial intensities are usually generated in systems that employ plane-wave illumination. This type of illumination allows a direct processing of the captured intensities with DPRTs for recovering the phase. Furthermore, it has been shown that intensities for DPRTs can be acquired from systems that use spherical illumination as well. However, this type of illumination presents a major setback for DPRTs: the captured intensities change their size for each position of the detector on the propagation axis. In order to apply the DPRTs, reescalation of the captured intensities has to be applied. This condition can increase the error sensitivity of the final phase result if it is not carried out properly. In this work, we introduce a novel system based on a Phase Light Modulator (PLM) for capturing the intensities when employing spherical illumination. The proposed optical system enables us to capture the diffraction pattern of under, in, and over-focus intensities. The employment of the PLM allows capturing the corresponding intensities without displacing the detector. Moreover, with the proposed optical system we can control accurately the magnification of the captured intensities. Thus, the stack of captured intensities can be used in DPRTs, overcoming the problems related with the resizing of the images. In order to prove our claims, the corresponding numerical experiments will be carried out. These simulations will show that the retrieved phases with spherical illumination are accurate and can be compared with those that employ plane wave illumination. We demonstrate that with the employment of the PLM, the proposed optical system has several advantages as: the optical system is compact, the beam size on the detector plane is controlled accurately, and the errors coming from mechanical motion can be suppressed easily.

  17. Tuning Genetic Clocks Employing DNA Binding Sites

    Periodic oscillations play a key role in cell physiology from the cell cycle to circadian clocks. The interplay of positive and negative feedback loops among genes and proteins is ubiquitous in these networks. Often, delays in a negative feedback loop and/or degradation rates are a crucial mechanism to obtain sustained oscillations. How does nature control delays and kinetic rates in feedback networks? Known mechanisms include proper selection of the number of steps composing a feedback loop and alteration of protease activity, respectively. Here, we show that a remarkably simple means to control both delays and effective kinetic rates is the employment of DNA binding sites. We illustrate this design principle on a widely studied activator-repressor clock motif, which is ubiquitous in natural systems. By suitably employing DNA target sites for the activator and/or the repressor, one can switch the clock “on” and “off” and precisely tune its period to a desired value. Our study reveals a design principle to engineer dynamic behavior in biomolecular networks, which may be largely exploited by natural systems and employed for the rational design of synthetic circuits.     Mucus-binding proteins (MUBs) have been revealed as one of the effector molecules involved in mechanisms of the adherence of lactobacilli to the host; mub, or mub-like, genes are found in all of the six genomes of Lactobacillus reuteri that are available. We recently reported the crystal structure of a Mub repeat from L. reuteri ATCC 53608 (also designated strain 1063), revealing an unexpected recognition of immunoglobulins. In the current study, we explored the diversity of the ATCC 53608 mub gene, and MUB expression levels in a large collection of L. reuteri strains isolated from a range of vertebrate hosts. This analysis revealed that the MUB was only detectable on the cell surface of two highly related isolates when using antibodies that were raised against the protein. There was considerable variation in quantitative mucus adhesion in vitro among L. reuteri strains, and mucus binding showed excellent correlation with the presence of cell-surface ATCC 53608 MUB. ATCC 53608 MUB presence was further highly associated with the autoaggregation of L. reuteri strains in washed cell suspensions, suggesting a novel role of this surface protein in cell aggregation. We also characterized MUB expression in representative L. reuteri strains. This analysis revealed that one derivative of strain 1063 was a spontaneous mutant that expressed a C-terminally truncated version of MUB. This frameshift mutation was caused by the insertion of a duplicated 13 nt sequence at position 4867 nt in the mub gene, producing a truncated MUB also lacking the C-terminal LPxTG region, and thus unable to anchor to the cell wall. This mutant, designated 1063N (mub-4867(i)), displayed low mucus-binding and aggregation capacities, further providing evidence for the contribution of cell-wall-anchored MUB to such phenotypes.     Consumer and rehabilitation provider factors that might limit employment opportunities for 154 individuals with six slowly progressive neuromuscular diseases (NMD) were investigated. The NMDs were spinal muscular atrophy (SMA), hereditary motor sensory neuropathy (HMSN), Becker's muscular dystrophy (BMD), facioscapulohumeral muscular dystrophy (FSHD), myotonic muscular dystrophy (MMD), and limb-girdle syndrome (LGS). Forty percent were employed in the competitive labor market at the time of the study, 50% had been employed in the past, and 10% had never been employed. The major consumer barrier to employment was education. Other important factors were type of occupation, intellectual capacity, psychosocial adjustment, and the belief by most individuals that their physical disability was the only or major barrier to obtaining a job. Psychological characteristics were associated with level of unemployment. However, physical impairment and disability were not associated with level of unemployment. There also were differences among the types of NMDs. Compared with the SMA, HMSN, BMD, and FSHD groups, the MMD and LGS groups had significantly higher levels of unemployment, lower educational levels, and fewer employed professional, management, and technical workers. Nonphysical impairment factors such as a low percentage of college graduates, impaired intellectual function in some individuals, and poor psychological adjustment were correlated with higher unemployment levels in the MMD group. Unemployment in the LGS group was correlated with a failure to complete high school. Major provider barriers to employment were the low level of referrals to Department of Rehabilitation by physicians and the low percentage of acceptance into the State Department of Rehabilitation.     Background There is evidence from North American trials that supported employment using the individual placement and support (IPS) model is effective in helping individuals with severe mental illness gain competitive employment. There have been few trials in other parts of the world. Aims To investigate the effectiveness and cost-effectiveness of IPS in the UK. Method Individuals with severe mental illness in South London were randomised to IPS or local traditional vocational services (treatment as usual) (ISRCTN96677673). Results Two hundred and nineteen participants were randomised, and 90% assessed 1 year later. There were no significant differences between the treatment as usual and intervention groups in obtaining competitive employment (13% in the intervention group and 7% in controls; risk ratio 1.35, 95% CI 0.95–1.93, P = 0.15), nor in secondary outcomes. Conclusions There was no evidence that IPS was of significant benefit in achieving competitive employment for individuals in South London at 1-year follow-up, which may reflect suboptimal implementation. Implementation of IPS can be challenging in the UK context where IPS is not structurally integrated with mental health services, and economic disincentives may lead to lower levels of motivation in individuals with severe mental illness and psychiatric professionals.     This article draws on data from qualitative interviews with ethnic enclave and ethnic economy business entrepreneurs from Chinese, Bangladeshi and Turkish-speaking communities in London. Routes into business and worker recruitment practices are explored, demonstrating the centrality of social capital in the form of family and other social networks within these processes. The article investigates what employers consider the desirable characteristics of workers: trust, kinship, gender, social networks, language compatibility and the needs of the business intersect with racialised notions of workers’ strengths and characteristics. Finally, we consider changing practices in relation to the employment of undocumented migrants, in the context of an increasingly punitive legislative regime. The complex and variable impact of policy alongside the ways in which other obligations and positions outweigh the fear and risks of sanctions associated with non-compliance is revealed.     In quantum information, complementarity of quantum mechanical observables plays a key role. The eigenstates of two complementary observables form a pair of mutually unbiased bases (MUBs). More generally, a set of MUBs consists of bases that are all pairwise unbiased. Except for specific dimensions of the Hilbert space, the maximal sets of MUBs are unknown in general. Even for a dimension as low as six, the identification of a maximal set of MUBs remains an open problem, although there is strong numerical evidence that no more than three simultaneous MUBs do exist. Here, by exploiting a newly developed holographic technique, we implement and test different sets of three MUBs for a single photon six-dimensional quantum state (a “qusix”), encoded exploiting polarization and orbital angular momentum of photons. A close agreement is observed between theory and experiments. Our results can find applications in state tomography, quantitative wave-particle duality, quantum key distribution. PMID:24067548

    Iron nanoparticles were employed to induce the synthesis of diamond on molybdenum, silicon, and quartz substrates. Diamond films were grown using conventional conditions for diamond synthesis by hot filament chemical vapor deposition, except that dispersed iron oxide nanoparticles replaced the seeding. This approach to diamond induction can be combined with dip pen nanolithography for the selective deposition of diamond and diamond patterning while avoiding surface damage associated to diamond-seeding methods.

    Statistical science provides important techniques for decision making in a large variety of fields of human endeavor mainly through statistical practice. According to Deming (1965), ``Statistical practice is mostly a collaborative venture between (professional) statisticians and experts in subject-matter (area).`` The professional statistician must be properly trained in areas of statistical science most suited to the discipline in which the applications are made. Due to a large variety of applications. The education of a professional statistician by universities can only be very general. Higher education provides the theoretical basis of statistical science whereas the specific expertise needed by the professional statistician is given by the application areas. In this paper, the authors review the approach of academic programs which are geared towards applications and discuss some of the innovative strategies for preparing statisticians as consultants in industry and business. A survey (census) of statisticians at the national laboratories of the US Department of Energy was made to assess their training by universities and to get a broad view of additional training which has brought them to their present professional status. A particular interest is the extent to which employers have contributed to the training of statisticians. A few recommendations are made which may serve to improve the future training of statisticians for applied collaborative work by universities and employers.

    There has been a conventionally held nostrum that fission system specific power and energy content is insufficient to provide the requisite high accelerations and velocities to enable interstellar rendezvous missions within a reasonable fraction of a human lifetime. As a consequence, all forms of alternative mechanisms that are not yet, and may never be technologically feasible, have been proposed, including laser light sails, fusion and antimatter propulsion systems. In previous efforts, [Lenard and Lipinski, 1999] the authors developed an architecture that employs fission power to propel two different concepts: one, an unmanned probe, the other a crewed vehicle to Alpha Centauri within mission times of 47 to 60 years. The first portion of this paper discusses employing a variant of the ``Forward Resupply Runway'' utilizing fission systems to enable both high accelerations and high final velocities necessary for this type of travel. The authors argue that such an architecture, while expensive, is considerably less expensive and technologically risky than other technologically advanced concepts, and, further, provides the ability to explore near-Earth stellar systems out to distances of 8 light years or so. This enables the ability to establish independent human societies which can later expand the domain of human exploration in roughly eight light-year increments even presuming that no further physics or technology breakthroughs or advances occur. In the second portion of the paper, a technology requirement assessment is performed. The authors argue that reasonable to extensive extensions to known technology could enable this revolutionary capability. .
