  1. Optimizing Medical Kits for Spaceflight

    The Integrated Medical Model (IMM) is a probabilistic model that estimates medical event occurrences and mission outcomes for different mission profiles. IMM simulation outcomes describing the impact of medical events on the mission may be used to optimize the allocation of resources in medical kits. Efficient allocation of medical resources, subject to certain mass and volume constraints, is crucial to ensuring the best outcomes of in-flight medical events. We implement a new approach to this medical kit optimization problem. METHODS We frame medical kit optimization as a modified knapsack problem and implement an algorithm utilizing a dynamic programming technique. Using this algorithm, optimized medical kits were generated for 3 different mission scenarios with the goal of minimizing the probability of evacuation and maximizing the Crew Health Index (CHI) for each mission subject to mass and volume constraints. Simulation outcomes using these kits were also compared to outcomes using kits optimized..RESULTS The optimized medical kits generated by the algorithm described here resulted in predicted mission outcomes more closely approached the unlimited-resource scenario for Crew Health Index (CHI) than the implementation in under all optimization priorities. Furthermore, the approach described here improves upon in reducing evacuation when the optimization priority is minimizing the probability of evacuation. CONCLUSIONS This algorithm provides an efficient, effective means to objectively allocate medical resources for spaceflight missions using the Integrated Medical Model.

  2. Mass and Volume Optimization of Space Flight Medical Kits

    Resource allocation is a critical aspect of space mission planning. All resources, including medical resources, are subject to a number of mission constraints such a maximum mass and volume. However, unlike many resources, there is often limited understanding in how to optimize medical resources for a mission. The Integrated Medical Model (IMM) is a probabilistic model that estimates medical event occurrences and mission outcomes for different mission profiles. IMM simulates outcomes and describes the impact of medical events in terms of lost crew time, medical resource usage, and the potential for medically required evacuation. Previously published work describes an approach that uses the IMM to generate optimized medical kits that maximize benefit to the crew subject to mass and volume constraints. We improve upon the results obtained previously and extend our approach to minimize mass and volume while meeting some benefit threshold. METHODS We frame the medical kit optimization problem as a modified knapsack problem and implement an algorithm utilizing dynamic programming. Using this algorithm, optimized medical kits were generated for 3 mission scenarios with the goal of minimizing the medical kit mass and volume for a specified likelihood of evacuation or Crew Health Index (CHI) threshold. The algorithm was expanded to generate medical kits that maximize likelihood of evacuation or CHI subject to mass and volume constraints. RESULTS AND CONCLUSIONS In maximizing benefit to crew health subject to certain constraints, our algorithm generates medical kits that more closely resemble the unlimited-resource scenario than previous approaches which leverage medical risk information generated by the IMM. Our work here demonstrates that this algorithm provides an efficient and effective means to objectively allocate medical resources for spaceflight missions and provides an effective means of addressing tradeoffs in medical resource allocations and crew mission success

  5. Development and Optimization of a Homemade ELISA Kit for Detection of Antibodies Against Haemophilus influenzae Type b

    Background: Haemophilus influenzae type b (Hib) infection has high morbidity and mortality rate, especially in children under 5 years of age. Enzyme-linked immunosorbent assay (ELISA) technique is the most used method to detect antibodies against H. influenzae. Available commercial ELISA kits are expensive and not always readily available, particularly for epidemiological studies. Objectives: This study was performed to develop and optimize a homemade ELISA kit for the detection of Hib anti-polyribosylribitol phosphate (PRP) antibodies in children. Materials and Methods: To develop and optimize an indirect ELISA method, pure PRP was prepared. The PRP was coupled to bovine serum albumin, using sodium periodate. Then optimal conditions for ELISA, including coating antigen concentration and peroxidase labeled conjugate concentrations, incubation temperature and incubation time, were determined. To confirm the efficacy of optimized kit, 83 serum samples from non-vaccinated children, aged less than 6 years were collected and analyzed, using homemade and commercial ELISA. Results: The optimal conditions were considered to perform ELISA. Comparison between results obtained from optimized ELISA kit and commercial ELISA kit showed a good agreement. Conclusions: Taking into account these data, we elaborated a homemade ELISA kit that is an efficacious and cost-effective substitute for commercial kit, in disease control and diagnosis.   6. Evaluation of two high-abundance protein depletion kits and optimization of downstream isoelectric focusing.

    Disease biomarkers for diagnostic and prognostic purposes are most likely within an extremely low concentration range and are thus masked by the presence of high‑abundance proteins. Therefore, removing high‑abundance proteins is the main challenge for identifying disease biomarkers. In addition, the solution obtained from high‑abundance protein depletion kits contains a rich array of compounds, which interfere with isoelectric focusing (IEF). In the present study, the effect of two commercial kits was evaluated and the downstream IEF protocol was optimized. High‑resolution results could be obtained according to the following conditions: The ProteoPrep Blue Albumin and IgG Depletion kit depleted albumin and IgG; immobilized pH gradient strips (typically 18 cm) were rehydrated with sample buffer containing 250 µg serum proteins at 30 v for 6 h, 60 v for 6 h, 200 v for 2 h, 500 v for 2 h, 1,000 v for 2 h, 5,000 v for 2 h, 10,000 v for 2 h and then focusing at 10,000 v up to 110 k vhs. In addition, the protein spots identified by matrix‑assisted laser desorption ionization time‑of‑flight mass spectrometry demonstrated that all proteins had a low abundance. The present study not only provides a definite and effective method for removing high‑abundance proteins, but also provides a proper protocol (protocol C) for downstream IEF.   7. Optimization of the RNeasy Mini Kit to obtain high-quality total RNA from sessile cells of Staphylococcus aureus

    Biofilm formed by Staphylococcus aureus is considered an important virulence trait in the pathogenesis of infections associated with implantable medical devices. Gene expression analyses are important strategies for determining the mechanisms involved in production and regulation of biofilm. Obtaining intact RNA preparations is the first and most critical step for these studies. In this article, we describe an optimized protocol for obtaining total RNA from sessile cells of S. aureus using the RNeasy Mini Kit. This method essentially consists of a few steps, as follows: 1) addition of acetone-ethanol to sessile cells, 2) lysis with lysostaphin at 37°C/10 min, 3) vigorous mixing, 4) three cycles of freezing and thawing, and 5) purification of the lysate in the RNeasy column. This simple pre-kit procedure yields high-quality total RNA from planktonic and sessile cells of S. aureus. PMID:26517334

  8. Optimization of pH values to formulate the bireagent kit for serum uric acid assay.

    A new formulation of the bireagent kit for serum uric acid assay was developed based on the effects of pH on enzyme stability. At 4 °C, half-lives of uricases from Bacillus fastidious and Arthrobacter globiforms were longer than 15 months at pH 9.2, but became shorter at pH below 8.0; half-lives of ascorbate oxidase and peroxidase were comparable at pH 6.5 and 7.0, but became much shorter at pH higher than 7.4. In the new formulation of the bireagent kit, Reagent A contained peroxidase, 4-aminoantipyrine, and ascorbate oxidase in 50 mM phosphate buffer at pH 6.5; Reagent B contained B. fastidious or A. globiforms uricase in 50 mM sodium borate buffer at pH 9.2; Reagents A and B were mixed at 4:1 to produce a final pH from 7.2 to 7.6 for developing a stable color. The new bireagent kit consumed smaller quantities of three enzymes for the same shelf life. With the new bireagent kit, there were linear responses of absorbance at 546 nm to uric acid up to 34 mM in reaction mixtures and a good correlation of uric acid levels in clinical sera with those by a commercial kit, but stronger resistance to ascorbate.     Collaborative teams of human operators and mobile ground robots are becoming popular in manufacturing plants to assist humans with a lot of the repetitive tasks such as the packing of related objects into different units, an operation known as kitting. In this paper, we present an ontology to provide a unified representation of all kitting-related tasks, which are decomposed into atomic actions that are either computational involving sensing, perception, planning, and control, or physical involving actuation and manipulation. The ontology is then used in a stochastic integer linear program for optimum partitioning of the atomic tasks between the robots and humans. Preliminary experiments on a single robot, single human case yield promising results where the kitting operations are completed with lower durations and manipulation failure rates using human-robot partnership versus just the human or only the robot. This success is achieved by the robot seeking human assistance for visual perception tasks while performing the other tasks primarily on its own.

    Deep sequencing of strand-specific cDNA libraries is now a ubiquitous tool for identifying and quantifying RNAs in diverse sample types. The accuracy of conclusions drawn from these analyses depends on precise and quantitative conversion of the RNA sample into a DNA library suitable for sequencing. Here, we describe an optimized method of preparing strand-specific RNA deep sequencing libraries from small RNAs and variably sized RNA fragments obtained from ribonucleoprotein particle footprinting experiments or fragmentation of long RNAs. Our approach works across a wide range of input amounts (400 pg to 200 ng), is easy to follow and produces a library in 2-3 days at relatively low reagent cost, all while giving the user complete control over every step. Because all enzymatic reactions were optimized and driven to apparent completion, sequence diversity and species abundance in the input sample are well preserved. PMID:25505164

  1. Development of 2MASS Catalog Server Kit

    We develop a software kit called "2MASS Catalog Server Kit" to easily construct a high-performance database server for the 2MASS Point Source Catalog (includes 470,992,970 objects) and several all-sky catalogs. Users can perform fast radial search and rectangular search using provided stored functions in SQL similar to SDSS SkyServer. Our software kit utilizes open-source RDBMS, and therefore any astronomers and developers can install our kit on their personal computers for research, observation, etc. Out kit is tuned for optimal coordinate search performance. We implement an effective radial search using an orthogonal coordinate system, which does not need any techniques that depend on HTM or HEALpix. Applying the xyz coordinate system to the database index, we can easily implement a system of fast radial search for relatively small (less than several million rows) catalogs. To enable high-speed search of huge catalogs on RDBMS, we apply three additional techniques: table partitioning, composite expression index, and optimization in stored functions. As a result, we obtain satisfactory performance of radial search for the 2MASS catalog. Our system can also perform fast rectangular search. It is implemented using techniques similar to those applied for radial search. Our way of implementation enables a compact system and will give important hints for a low-cost development of other huge catalog databases.

  2. Acute Phase Reactants as Novel Predictors of Cardiovascular Disease

    Acute phase reaction is a systemic response which usually follows a physiological condition that takes place in the beginning of an inflammatory process. This physiological change usually lasts 1-2 days. However, the systemic acute phase response usually lasts longer. The aim of this systemic response is to restore homeostasis. These events are accompanied by upregulation of some proteins (positive acute phase reactants) and downregulation of others (negative acute phase reactants) during inflammatory reactions. Cardiovascular diseases are accompanied by the elevation of several positive acute phase reactants such as C-reactive protein (CRP), serum amyloid A (SAA), fibrinogen, white blood cell count, secretory nonpancreatic phospholipase 2-II (sPLA2-II), ferritin, and ceruloplasmin. Cardiovascular disease is also accompanied by the reduction of negative acute phase reactants such as albumin, transferrin, transthyretin, retinol-binding protein, antithrombin, and transcortin. In this paper, we will be discussing the biological activity and diagnostic and prognostic values of acute phase reactants with cardiovascular importance.   3. Cryogenic reactant storage for lunar base regenerative fuel cells

    There are major advantages to be gained by integrating a cryogenic reactant storage system with a hydrogen-oxygen regenerative fuel cell (RFC) to provide on-site electrical power during the lunar night. Although applicable to any power system using hydrogen-oxygen RFC's for energy storage, cryogenic reactant storage offers a significant benefit whenever the sun/shade cycle and energy storage period approach hundreds of hours. For solar power installations on the moon, cryogenic reactant storage reduces overall specific mass and meteoroid vulnerability of the system. In addition, it offers synergistic benefits to on-site users, such as availability of primary fuel cell reactants for surface rover vehicles and cryogenic propellants for OTV's. The integration involves processing and storing the RFC reactant streams as cryogenic liquids rather than pressurized gases, so that reactant containment (tankage per unit mass of reactants) can be greatly reduced. Hydrogen-oxygen alkaline RFC's, GaAs photovoltaic (PV) arrays, and space cryogenic processing/refrigeration technologies are assumed to be available for the conceptual system design. Advantages are demonstrated by comparing the characteristics of two power system concepts: a conventional lunar surface PV/RFC power system using pressurized gas storage in SOA filament wound pressure vessels and, that same system with gas liquefaction and storage replacing the pressurized storage. Comparisons are made at 20 and 250 kWe. Although cryogenic storage adds a processing plant (drying and liquefaction) to the system plus 30 percent more solar array to provide processing power, the approximate order of magnitude reduction in tankage mass, confirmed by this analysis, results in a reduction in overall total system mass of approximately 50 percent.

    "The traveler's medical kit is an essential tool for both the novice and expert traveler. It is designed to treat travel-related illness and injury and to ensure preexisting medical conditions are managed appropriately. Travelers are at increased risk for common gastrointestinal issues during travel. Respiratory illnesses make up approximately 8% of the ailments present in returned international travelers. Approximately 12% of travelers experience a travel-related skin condition. First aid treatment for minor injuries is essential to all travel medical kits.   7. Fuel cell systems for first lunar outpost: Reactant storage options

    A Lunar Surface Power Working Group was formed to review candidate systems for providing power to the First Lunar Outpost habitat. The working group met for five days in the fall of 1992 and concluded that the most attractive candidate included a photovoltaic unit, a fuel cell, a regenerator to recycle the reactants, and storage of oxygen and hydrogen gases. Most of the volume (97%) and weight (64%) are taken up by the reactants and their storage tanks. The large volume is difficult to accommodate, and therefore, the working group explored ways of reducing the volume. An alternative approach to providing separate high pressure storage tanks is to use two of the descent stage propellant storage tanks, which would have to be wrapped with graphite fibers to increase their pressure capability. This saves 90% of the volume required for storage of fuel cell reactants. Another approach is to use the descent storage propellant tanks for storage of the fuel cell reactants as cryogenic liquids, but this requires a gas liquefaction system, increases the solar array by 40%, and increases the heat rejection rate by 170% compared with storage of reactants as high pressure gases. For a high power system (greater than 20 kW) the larger energy storage requirement would probably favor the cryogenic storage option.

    World resources, reserves, production, and costs of reactant materials, iron, chromium, titanium and bromine for proposed redox cell bulk energy storage systems are reviewed. Supplying required materials for multimegawatt hour systems appears to be feasible even at current production levels. Iron and chromium ores are the most abundant and lowest cost of four reactants. Chromium is not a domestic reserve, but redox system installations would represent a small fraction of U.S. imports. Vast quantities of bromine are available, but present production is low and therefore cost is high. Titanium is currently available at reasonable cost, with ample reserves available for the next fifty years.

    Great controversy surrounds the issue of United States (US) air carrier inflight medical kits. Although there are four medications mandated by the Federal Aviation Administration (FAA) that appeared to be adequate as determined by a 1988 survey, there is now a renewed call to review the medical kit contents with an eye toward making them more robust. This has been prompted by several well publicized inflight medical events and the fact that overseas airlines have a very wide array of pharmaceuticals and supplies. Consequently, the Aerospace Medical Association (AsMA) convened a Task Force of physicians across the major specialties to put forward recommendations regarding medication, medical supplies, and automatic external defibrillators (AEDs).     Surface-supported coupling reactions between 1,3,5-tris(4-formylphenyl)benzene and aromatic amines have been investigated on Au(111) using scanning tunneling microscopy under ultra-high-vacuum conditions. Upon annealing to moderate temperatures, various products, involving the discrete oligomers and the surface covalent organic frameworks, are obtained through thermal-triggered on-surface chemical reactions. We conclude from the systematic experiments that the stoichiometric composition of the reactants is vital to the surface reaction products, which is rarely reported so far. With this knowledge, we have successfully prepared two-dimensional covalently bonded networks by optimizing the stoichiometric proportions of the reaction precursors. PMID:27043277

    We have implemented an efficient, user-friendly biochemical reaction simulator called Web-based BEST-KIT (Biochemical Engineering System analyzing Tool-KIT) for analyzing large-scale nonlinear networks such as metabolic pathways. Users can easily design and analyze an arbitrary reaction scheme through the Internet and an efficient graphical user interface without considering the mathematical equations. The reaction scheme can include several reaction types, which are represented by both the mass action law (mass balance) and approximated velocity functions of enzyme kinetics at steady state, such as Michaelis-Menten, Hill cooperative, Competitive inhibition. However, since all modules in Web-based BEST-KIT have been developed in Java applet style, users cannot optionally make use of original mathematical equations in addition to the prepared equations. In the present study, we have developed a new version of BEST-KIT (for Microsoft Windows called WinBEST-KIT) to allow users to define original mathematical equations and to customize these equations very easily as user-defined reaction symbols. The following powerful system-analytical methods are prepared for system analysis: time-course calculation, parameter scanning, estimation of the values of unknown kinetic parameters based on experimentally observed time-course data of reactants, dynamic response of reactants against virtual external perturbations, and real-time simulation (Virtual Dry Lab).     The design and application of a supplementary power and heat rejection kit for the Spacelab are discussed. Two subsystems of electric power and thermal control were analyzed to define the requirements for the power and heat rejection kit (PHRK). Twelve exemplary experiments were defined and power timelines were developed. From these timeline, the experiment requirements for sustained power, peak power, and energy were determined. The electrical power subsystem of the PHRK will consist of two fuel cells, oxygen and hydrogen reactant tank assemblies, water storage tanks, plumbing, cabling, and inverters to convert the nominal 28 volt dc fuel cell output to ac power.

    The monomeric reactants approach was used to fabricate addition-type polyimide/graphite fiber composites with improved mechanical properties and thermal stability characteristics over those of composites derived from addition-type amide acid prepolymers. A screening study of 24 different monomer combinations was performed. The results of a more extensive investigation of a selected number of monomer combinations showed that the combination providing the best thermomechanical properties was 5-norbornene-2.3-dicarboxylic acid monomethyl ester/4,4(')-methylenedianiline/3,3(')4,4(')-benzophenonetetracarboxylic acid dimethyl ester at a molar ratio of 2/3.09/2.09.

Cells employing a molten alkali metal electrode, e.g., sodium, in contact with a ceramic electrolyte, which is a conductor of the ions of the alkali metal forming the electrode, exhibit a lower resistance when a reactive metal, e.g., vanadium, is allowed to react with and reduce the alkali metal oxide. Such cells exhibit less degradation of the electrolyte and of the glass seals often used to joining the electrolyte to the other components of the cell under cycling conditions.

    In this paper we report on the features of the Java Commodity Grid Kit. The Java CoG Kit provides middleware for accessing Grid functionality from the Java framework. Java CoG Kit middleware is general enough to design a variety of advanced Grid applications with quite different user requirements. Access to the Grid is established via Globus protocols, allowing the Java CoG Kit to communicate also with the C Globus reference implementation. Thus, the Java CoG Kit provides Grid developers with the ability to utilize the Grid, as well as numerous additional libraries and frameworks developed by the Java community to enable network, Internet, enterprise, and peer-to peer computing. A variety of projects have successfully used the client libraries of the Java CoG Kit to access Grids driven by the C Globus software. In this paper we also report on the efforts to develop server side Java CoG Kit components. As part of this research we have implemented a prototype pure Java resource management system that enables one to run Globus jobs on platforms on which a Java virtual machine is supported, including Windows NT machines.

    3BP2 is a cytoplasmic adaptor protein that acts as a positive regulator in mast cell FcεRI-dependent signaling. The KIT receptor whose ligand is the stem cell factor (SCF) is necessary for mast cell development, proliferation and survival as well as for optimal IgE-dependent signal. Activating mutations in KIT have been associated with several diseases including mastocytosis. In the present work, we found that 3BP2 silencing impairs KIT signaling pathways, thus affecting PI3K and MAP kinase pathways in human mast cells from HMC-1, LAD2 (human mast cell lines) and CD34+-derived mast cells. Unexpectedly, silencing of 3BP2 reduces KIT expression in normal human mast cells as well as in HMC-1 cells where KIT is mutated, thus increasing cellular apoptosis and caspase 3/7 activity. 3BP2 silencing reduces KIT transcription expression levels. Interestingly, 3BP2 silencing decreased MITF expression, a transcription factor involved in KIT expression. Reconstitution of 3BP2 in knockdown cells leads to reversal of KIT expression as well as survival phenotype. Accordingly MITF reconstitution enhances KIT expression levels in 3BP2 silenced cells. Moreover, downregulation of KIT expression by miRNA221 overexpression or the proteasome inhibitor bortezomib also reduced 3BP2 and MITF expression. Furthermore, KIT tyrosine activity inhibition reduced 3BP2 and MITF expression, demonstrating again a tight and reciprocal relationship between these molecules. Taken together, our results show that 3BP2 regulates human mast cell survival and participates in KIT-mediated signal transduction by directly controlling KIT receptor expression, suggesting its potential as a therapeutic target in mast cell-mediated inflammatory diseases and deregulated KIT disorders.     In the last decade, melatonin has been found to be highly protective against damage to macromolecules resulting from oxygen and nitrogen-based reactants. Considering this, numerous studies have examined the mechanisms whereby this indoleamine directly detoxifies these damaging agents. The evidence is compelling that melatonin scavenges several oxygen-derived reactive agents including the hydroxyl radical (OH), hydrogen peroxide (H2O2), singlet oxygen (1O2) and hypochlorous acid (HOCl). Additionally, melatonin reportedly reacts with nitric oxide (NO), the peroxynitrite anion (ONOO-) and/or peroxynitrous acid (ONOOH) to detoxify them. In some cases the products that are formed as a consequence of melatonin's scavenging actions have been identified. Whereas the ability of melatonin to neutralize these toxic agents likely accounts, in part, for the antioxidant activity of melatonin, it is not the only means by which melatonin serves to protect molecules from oxygen and nitrogen-based reactive metabolites.   1. Calculating fusion neutron energy spectra from arbitrary reactant distributions

    The Directional Relativistic Spectrum Simulator (DRESS) code can perform Monte-Carlo calculations of reaction product spectra from arbitrary reactant distributions, using fully relativistic kinematics. The code is set up to calculate energy spectra from neutrons and alpha particles produced in the D(d, n)3He and T(d, n)4He fusion reactions, but any two-body reaction can be simulated by including the corresponding cross section. The code has been thoroughly tested. The kinematics calculations have been benchmarked against the kinematics module of the ROOT Data Analysis Framework. Calculated neutron energy spectra have been validated against tabulated fusion reactivities and against an exact analytical expression for the thermonuclear fusion neutron spectrum, with good agreement. The DRESS code will be used as the core of a detailed synthetic diagnostic framework for neutron measurements at the JET and MAST tokamaks.

    Optimal application of zero-valent iron (Fe{sup 0}) in contaminant remediation requires an understanding of the mechanism by which the process occurs. An important first step towards developing such an understanding is determining the dependence of reaction rate on the concentration of each reactant. Previous efforts by Matheson and Tratnyek (1994) found that the first-order rate constant for reduction of CCl{sub 4} by Fe{sup 0} increased linearly with iron surface area. However, heterogeneous systems often exhibit more complex behavior, suggesting that the simple linear relationships previously reported may not fully reflect all of the potentially contributing processes. To investigate this possibility, further study of the effect of initial CCl{sub 4} concentration and iron surface area has been undertaken. Our findings suggest that site availability arguments can be applied to the effect of CCl{sub 4} concentration but that the effect of Fe{sup 0} surface area is more complex.

  4. Fever and acute phase reactants in the rat.

    In rats synthesis of some acute phase reactants can be induced by a combination of corticosteroids and adrenaline. During fever both hormones show high plasma levels. We studied the effect of fever induced by intra-cerebroventricular (i.c.v.) injection of PGE2 on the acute phase response. Fever was continuously recorded and 24 h after induction acute phase reactant (APR) response was measured as indicated by the rise of alpha-macrofetoprotein (alpha M FP, alpha 2 macroglobulin of the rat). Controls received 0.9% saline i.c.v. Controls did not develop fever (dTmax less than or equal to 1 degree C) nor did they show significant APR response. The maximal rise in body temperature after PGE2 (2.6 +/- 0.7 degrees C) correlated significantly with the rise in alpha M FP concentration 24 h later. Adrenalectomy prevented the APR response completely but the magnitude of the fever reaction remained the same (2.1 +/- 0.3 degrees C). alpha-Blockade gave a smaller fever response but had no effect on the APR response. In alpha- and beta-blockade, fever response was normal but no APR response was obtained. Destroying the sympathetic nerve supply to the liver with 6-OH dopamine retarded the fever response but again APR response was not impeded. In order to differentiate between the role of fever as such and the effect of PGE2 on APR synthesis, we used heat exposure to induce hyperthermia in normal rats who showed an APR response comparable with that after i.c.v. PGE2. Pretreatment with sodium salicylate before inducing hyperthermia led to a variable rise in alpha M FP. Fever as such, without tissue injury, induces an APR response. The pathway to this effect probably involves circulating corticosterone and adrenaline, possibly via a beta-receptor mediated stimulation. PMID:2460123

  5. Comparison of DNA Extraction Kits for Detection of Burkholderia pseudomallei in Spiked Human Whole Blood Using Real-Time PCR

    Burkholderia pseudomallei, the etiologic agent of melioidosis, is endemic in northern Australia and Southeast Asia and can cause severe septicemia that may lead to death in 20% to 50% of cases. Rapid detection of B. pseudomallei infection is crucial for timely treatment of septic patients. This study evaluated seven commercially available DNA extraction kits to determine the relative recovery of B. pseudomallei DNA from spiked EDTA-containing human whole blood. The evaluation included three manual kits: the QIAamp DNA Mini kit, the QIAamp DNA Blood Mini kit, and the High Pure PCR Template Preparation kit; and four automated systems: the MagNAPure LC using the DNA Isolation Kit I, the MagNAPure Compact using the Nucleic Acid Isolation Kit I, and the QIAcube using the QIAamp DNA Mini kit and the QIAamp DNA Blood Mini kit. Detection of B. pseudomallei DNA extracted by each kit was performed using the B. pseudomallei specific type III secretion real-time PCR (TTS1) assay. Crossing threshold (CT) values were used to compare the limit of detection and reproducibility of each kit. This study also compared the DNA concentrations and DNA purity yielded for each kit. The following kits consistently yielded DNA that produced a detectable signal from blood spiked with 5.5×104 colony forming units per mL: the High Pure PCR Template Preparation, QIAamp DNA Mini, MagNA Pure Compact, and the QIAcube running the QIAamp DNA Mini and QIAamp DNA Blood Mini kits. The High Pure PCR Template Preparation kit yielded the lowest limit of detection with spiked blood, but when this kit was used with blood from patients with confirmed cases of melioidosis, the bacteria was not reliably detected indicating blood may not be an optimal specimen.   6. Influence of the reactant carbon-hydrogen-oxygen composition on the key products of the direct gasification of dewatered sewage sludge in supercritical water.

    The supercritical water gasification of ten different types of dewatered sewage sludges was investigated to understand the relationship between sludge properties and gasification products. Experiments were performed in a high-pressure autoclave at 400°C for 60 min. Results showed that gasification of sewage sludge in supercritical water consists mainly of a gasification reaction, a carbonization reaction and a persistent organic pollutants synthesis reaction. Changes in the reactant C/H/O composition have significant effects on the key gasification products. Total gas production increased with increasing C/H2O of the reactant. The char/coke content increased with increasing C/H ratio of the reactant. A decrease in the C/O ratio of the reactant led to a reduction in polycyclic aromatic hydrocarbon formation. This means that we can adjust the reactant C/H/O composition by adding carbon-, hydrogen-, and oxygen-containing substances such as coal, algae and H2O2 to optimize hydrogen production and to inhibit an undesired by-product formation.   7. Microfoams as Reactant Transport Media for In-Situ Immobilization of Radionuclide and Metallic Contaminants in Deep Vadose Zone

    The U.S. Department of Energy (DOE) is currently addressing issues related to remediation of Cr, U and Tc contamination in the deep vadose zone at the Hanford Site in Washington State. One of the transformational technology alternatives being considered by the DOE Office of Environmental Management, is the use of Reactant Carrier Microfoams (RCM) for in-situ immobilization of contaminants. Foam injection technology for Enhance Oil Recovery (EOR) has well-established pedigree. Use of surfactant foams have also been explored for mobilizing DNAPL from sediments. However, the novel concept of using RCM for in situ immobilization contaminants in the deep vadose zone has not been explored, therefore, presents many daunting challenges for successful implementation. Scienists at Pacific Northwest National Laboratory (PNNL), leveraged previous EMSP-funded studies on microfoams conducted at LBNL with the goal to formulate robust stable microfoams for delivering reductive and/or precipitating reactants to the deep subsurface. Following an extensive literature review, a protocol was deisnged to select appropriate surfactant blends, and tested three different methods of foam generation namely, Venturi foam generato , high-speed gas entrainment and porous plate method. The resulting RCMs were characterized as to their quality, stability, bubble size distribution, surface tension and viscosity. The foam stabilities as a function of reactant (polyphosphate and polysulfides) concentrations and entrained polyatomic gases were also examined. Based on these experiments, optimal carrier foam compositions were identified for each Hanford deep vadose zone Contaminant of Concern (COC) namely U(VI) and Cr(VI). Finally, MSE Technology Applications, Inc (MSE) in collaboration with PNNL, conducted a series of scale-up reactant carrier foam injection tests to evaluate the efficacy of this technology for potential deep vadose zone remediation.

    Coral microbiology is an expanding field, yet there is no standard DNA extraction protocol. Although many researchers depend on commercial extraction kits, no specific kit has been optimized for use with coral samples. Both soil and plant DNA extraction kits from MO BIO Laboratories, Inc., have been used by many research groups for this purpose. MO BIO recently replaced their PowerPlant® kit with an improved PowerPlantPro kit, but it was unclear how these changes would affect the kit’s use with coral samples. In order to determine which kit produced the best results, we conducted a comparison between the original PowerPlant kit, the new PowerPlantPro kit, and an alternative kit, PowerSoil, using samples from several different coral genera. The PowerPlantPro kit had the highest DNA yields, but the lack of 16S rRNA gene amplification in many samples suggests that much of the yield may be coral DNA rather than microbial DNA. The most consistent positive amplifications came from the PowerSoil kit.

  11. Numerical Simulation of Turbulent Propane-Air Combustion with Non-Homogeneous Reactants

    Two-dimensional numerical simulations of turbulent propane-air combustion have been performed including complex chemistry and realistic molecular transport. The aerothermochemical conditions simulated (reactant temperature and pressure, turbulence rms velocity and integral length scale) correspond to conditions at the time of ignition in an automotive gasoline direct-injection reciprocating IC engine at low speed and light load. Both stoichiometric homogeneous reactants and non-homogeneous reactants with fuel-based equivalence ratios ranging from zero to four have been simulated. In the case of non-homogeneous reactants, a primary premixed flame (defined based on disappearance of the propane fuel) is followed by a secondary heat-release zone that is dominated by CO kinetics and turbulent mixing. Beyond a few flame thicknesses behind the primary flame, any remaining fuel has been broken down into carbon monoxide and hydrogen. Quantitative information relevant for modeling turbulent flame propagation through nonhomogeneous reactants has been extracted.

    General matrix approach to the consideration of multistage geminate reactions of isolated pairs of reactants depending on reactant mobility is formulated on the basis of the concept of “effective” particles. Various elementary reactions (stages of multistage reaction including physicochemical processes of internal quantum state changes) proceeding with the participation of isolated pairs of reactants (or isolated reactants) are taken into account. Investigation has been made in terms of kinetic approach implying the derivation of general (matrix) kinetic equations for local and mean probabilities of finding any of the reaction species in the sample under study (or for local and mean concentrations). The recipes for the calculation of kinetic coefficients of the equations for mean quantities in terms of relative coordinates of reactants have been formulated in the general case of inhomogeneous reacting systems. Important specific case of homogeneous reacting systems is considered.

StochKit2 provides highly efficient implementations of several variants of Gillespie's stochastic simulation algorithm (SSA), and tau-leaping with automatic step size selection. StochKit2 features include automatic selection of the optimal SSA method based on model properties, event handling, and automatic parallelism on multicore architectures.   14. KIT mutations are common in testicular seminomas.

    Expression of KIT tyrosine kinase is critical for normal germ cell development and is observed in the majority of seminomas. Activating mutations in KIT are common in gastrointestinal stromal tumors and mastocytosis. In this study we examined the frequency and spectrum of KIT mutations in 54 testicular seminomas, 1 ovarian dysgerminoma and 37 non-seminomatous germ cell tumors (NSGCT). Fourteen seminomas (25.9%) contained exon 17 point mutations including D816V (6 cases), D816H (3 cases), Y823D (2 cases), and single examples of Y823C, N822K, and T801I. No KIT mutations were found in the ovarian dysgerminoma or the NSGCTs. In transient transfection assays, mutant isoforms D816V, D816H, Y823D, and N822K were constitutively phosphorylated in the absence of the natural ligand for KIT, stem cell factor (SCF). In contrast, activation of T801I and wild-type KIT required SCF. Mutants N822K and Y823D were inhibited by imatinib mesylate (Gleevec, previously STI571) whereas D816V and D816H were both resistant to imatinib mesylate. Biochemical evidence of KIT activation, as assessed by KIT phosphorylation and KIT association with phosphatidylinositol (PI) 3-kinase in tumor cell lysates, was largely confined to seminomas with a genomic KIT mutation. These findings suggest that activating KIT mutations may contribute to tumorigenesis in a subset of seminomas, but are not involved in NSGCT. PMID:14695343

  15. Differential Use of Elementary Science Kits

    The use of kits in elementary science classes is a growing trend in some countries. Kits provide materials and inquiry lessons in a ready-to-teach format for teachers to use in their science instruction. This study examined elementary teachers' instructional strategies, classroom practices, and assessment types in relation to the frequency of science kit use. A total of 503 elementary teachers from an urban school district received professional development, implemented kits in their classrooms for a year, and then completed a survey about science kit use and teaching practices. Despite similarities in demographic characteristics (gender, ethnicity, certification/educational level), there were significant differences in teachers' use of inquiry-based teaching and assessment practices by kit use. Teachers who reported using kits the most often were significantly more likely to report that their students designed and implemented laboratory investigations as well recorded, represented, and analyzed data. In addition, the high kit users indicated that they were more likely to use student groups, require students to use evidence to support claims, and use alternative assessments of student work including portfolios, notebooks, and long-term projects than those teachers who used kits less frequently. Those teachers who reported using kits the least often were significantly more likely to report having students practice for standardized tests. The role of kits in promoting reform-based teaching practices is discussed.

    Flight operations and the preparation for it has become increasingly complex as mission complexities increase. Further, the mission model dictates that a significant increase in flight operations activities is upon us. Finally, there is a need for process improvement and economy in the operations arena. It is therefore time that we recognize flight operations as a complex process requiring a defined, structured, and life cycle approach vitally linked to space segment, ground segment, and science operations processes. With this recognition, an FOT Tool Kit consisting of six major components designed to provide tools to guide flight operations activities throughout the mission life cycle was developed. The major components of the FOT Tool Kit and the concepts behind the flight operations life cycle process as developed at NASA's GSFC for GSFC-based missions are addressed. The Tool Kit is therefore intended to increase productivity, quality, cost, and schedule performance of the flight operations tasks through the use of documented, structured methodologies; knowledge of past lessons learned and upcoming new technology; and through reuse and sharing of key products and special application programs made possible through the development of standardized key products and special program directories.

    In the presence of CO2, the negative reactant ions of ion mobility spectrometry (IMS) are ion clusters of CO4(-) and CO3(-). Methyl salicylate is ionized by the CO4(-)(H2O(n))(N2(m)) reactant ions, but not by the CO3(-)(H2O(n))(N2(m)) reactant ions. While the CO4(-) ions are formed by direct association, the CO3(-) ions require additional energy to be formed. The additional energy is provided by either excited neutral gas molecules in a metastable state or UV (ultraviolet) radiation.

    Excitation of reactant rotational degrees of freedom is known to influence reactivity in bimolecular reactions. In this work, this effect is examined for several prototypical activated atom-diatom and atom-triatom reactions through exact quantum scattering calculations on accurate ab initio potential energy surfaces. To rationalize these mode-specific effects, the recently proposed sudden vector overlap model is extended to include rotational motions of reactants. Specifically, the enhancement of reactivity is attributed to their coupling with the reaction coordinate at the transition state, as quantified by the alignment between the corresponding normal mode vectors. In addition, a Franck-Condon model is introduced to predict the effect of reactant rotational excitation for reactions in which the reactant rotations are decoupled from the reaction coordinate.

    Deuterium fusion reactant energy spectra have been measured using a diagnostic that records the Doppler shift imparted to charged particle fusion products of the D(d,p)T reaction by the center-of-mass velocity of the deuterium reactants. This diagnostic, known as the fusion ion Doppler shift diagnostic (FIDO) measures fast deuterium energy spectra in the inertial electrostatic confinement (IEC) experiment at the University of Wisconsin-Madison {l_brace}Santarius et al. [Fusion Sci. Technol. 47, 1238 (2005)]{r_brace}, a device to confine high energy light ions in a spherically symmetric, electrostatic potential well. This article details the first measurements of the fusion reactant energy spectra in an IEC device as well as the design and principles of operation of the FIDO diagnostic. Scaling of reactant energy spectra with a variety of experimental parameters have been explored.

  20. Liquid Water Transport in the Reactant Channels of Proton Exchange Membrane Fuel Cells

    Water management has been identified as a critical issue in the development of PEM fuel cells for automotive applications. Water is present inside the PEM fuel cell in three phases, i.e. liquid phase, vapor phase and mist phase. Liquid water in the reactant channels causes flooding of the cell and blocks the transport of reactants to the reaction sites at the catalyst layer. Understanding the behavior of liquid water in the reactant channels would allow us to devise improved strategies for removing liquid water from the reactant channels. In situ fuel cell tests have been performed to identify and diagnose operating conditions which result in the flooding of the fuel cell. A relationship has been identified between the liquid water present in the reactant channels and the cell performance. A novel diagnostic technique has been established which utilizes the pressure drop multiplier in the reactant channels to predict the flooding of the cell or the drying-out of the membrane. An ex-situ study has been undertaken to quantify the liquid water present in the reactant channels. A new parameter, the Area Coverage Ratio (ACR), has been defined to identify the interfacial area of the reactant channel which is blocked for reactant transport by the presence of liquid water. A parametric study has been conducted to study the effect of changing temperature and the inlet relative humidity on the ACR. The ACR decreases with increase in current density as the gas flow rates increase, removing water more efficiently. With increase in temperature, the ACR decreases rapidly, such that by 60°C, there is no significant ACR to be reported. Inlet relative humidity of the gases does change the saturation of the gases in the channel, but did not show any significant effect on the ACR.     Ribonuclease enzymes (RNases) play key roles in the maturation and metabolism of all RNA molecules. Computational simulations of the processes involved can help to elucidate the underlying enzymatic mechanism and is often employed in a synergistic approach together with biochemical experiments. Theoretical calculations require atomistic details regarding the starting geometries of the molecules involved, which, in the absence of crystallographic data, can only be achieved from computational docking studies. Fortunately, docking algorithms have improved tremendously in recent years, so that reliable structures of enzyme-ligand complexes can now be successfully obtained from computation. However, most docking programs are not particularly optimized for nucleotide docking. In order to assist our studies on the cleavage of RNA by the two most important ribonuclease enzymes, RNase A and RNase H, we evaluated four docking tools - MOE2009, Glide 5.5, QXP-Flo+0802, and Autodock 4.0 - for their ability to simulate complexes between these enzymes and RNA oligomers. To validate our results, we analyzed the docking results with respect to the known key interactions between the protein and the nucleotide. In addition, we compared the predicted complexes with X-ray structures of the mutated enzyme as well as with structures obtained from previous calculations. In this manner, we were able to prepare the desired reaction state complex so that it could be used as the starting structure for further DFT/B3LYP QM/MM reaction mechanism studies.

    In this paper we report on the features of the Java Commodity Grid Kit (Java CoG Kit). The Java CoG Kit provides middleware for accessing Grid functionality from the Java framework. Java CoG Kit middleware is general enough to design a variety of advanced Grid applications with quite different user requirements. Access to the Grid is established via Globus Toolkit protocols, allowing the Java CoG Kit to also communicate with the services distributed as part of the C Globus Toolkit reference implementation. Thus, the Java CoG Kit provides Grid developers with the ability to utilize the Grid, as well as numerous additional libraries and frameworks developed by the Java community to enable network, Internet, enterprise and peer-to-peer computing. A variety of projects have successfully used the client libraries of the Java CoG Kit to access Grids driven by the C Globus Toolkit software. In this paper we also report on the efforts to develop serverside Java CoG Kit components. As part of this research we have implemented a prototype pure Java resource management system that enables one to run Grid jobs on platforms on which a Java virtual machine is supported, including Windows NT machines.

    During the investigation of the degradation products of 2,4,6-trinitrotoluene (TNT) using ion mobility spectrometry (IMS), 2,4-dichlorophenoxyacetic acid (2,4-D) and 2,4-dichlorophenol (DCP) were found to have IMS responses which overlapped those of the TNT degradation products. It was observed that the Cl- reactant ion chemistry, often used for explosives analysis, was not always successful in resolving peak overlap of analytes and interferents. It is shown here that resolution of the analytes and interferences can sometimes be achieved using only air for the formation of reactant ions, at other times through the use of Br- as an alternative to Cl- for producing reactant ions, and also through the promotion of adduct stability by lowering the IMS temperature.

  15. Control of the reactant ion chemistry for the analysis of explosives by ion mobility spectroscopy

    Changes in the reactant ion composition in the ion mobility spectrometer (IMS) can result in a change in the ionization processes occurring in the ionization region, ultimately leading to an altered instrumental response for the analyte, and exacerbating the problem of qualitative and quantitative analysis. Some species are very susceptible to changes in reactant ions, while other species are relatively unaffected. These types of behavior are observed for two common explosives, namely, hexahydro-1,3,5-trinitrol,3,5-triazine (RDX) and 1,3,5-trinitrotoluene (TNT), respectively. To control the reactant ion composition, and hence the gas phase chemistry, it is necessary to control the composition of gases present in the ionization region of the IMS. A series of modifications are described for the PCP Phemto-Chem 100 IMS that afford the requisite control. The effectiveness of these modifications for analysis of RDX and TNT are described and contrasted with that observed for the unmodified system.

Therefore, in my work for the Group, and in this report, I have concentrated on finding ways to reduce these volumes and weights. Three options were considered: (1) the baseline case considered in the preliminary system design, that of separate high pressure (200 bar) storage tanks; (2) the use of two of the descent storage propellant tanks wrapped with graphite fibers to increase the pressure capability; and (3) the use of cryogenic storage of reactants in the propellant tanks. The first option results in high storage tank mass and volume. The second option saves 90 percent of the volume by making use of the propellant tanks, but it has little if any weight advantages; the weight saved by not providing extra tanks for reactant storage is nearly entirely added back by the weight of the additional material (graphite fibers) to strengthen the propellant tanks. Use of the descent storage propellant tanks for storage of the fuel cell reactants as cryogenic liquids requires a gas liquefaction system. The weight of this system is expected to be less than that of the storage tanks but it would require development and testing to prove its reliability. The solar array would have to be 40 percent larger and the heat projection range would be 170 percent larger than for storage of reactants as high pressure gases. For a high power system (greater than 20 kW) the larger energy storage requirement would probably favor the cryogenic storage option.

    The paper presents the results of studies of acute phase reactants in the 60 treated patients with rheumatoid arthritis. Patients were divided into two groups, depending on the applied treatment: group I (n = 30) was treated with methotrexate, sulfasalazine and hydroxychloroquine, and group II (n = 30) with methotrexate. The results of our study shows that there is a statistically significant reduction in the value of acute phase reactants and clinical parameters after treatment in both investigated groups of patients, and also a significant statistical difference between the first and second group of treated patients.     The Steinbuch Centre for Computing (SCC) at Karlsruhe Institute of Technology (KIT) has been involved fairly early in 100GE network technology. Initiated by DFN1 (the German NREN), a first 100GE wide area network testbed over a distance of approx. 450 km was deployed between the national research organizations KIT and FZ-Jülich in 2010. Three years later in 2013. KIT joined the Caltech SuperComputing 2013 (SC132) 100GE "show floor" initiative using the transatlantic ANA-100GE link to transfer LHC data from a storage at DE-KIT (GridKa) in Europe to hard disks at the show floor of SC13 in Denver (USA). The network infrastructure of KIT as well as of the German Tier-1 installation DE-KIT (GridKa). however. is still based on 10Gbps. As highlighted in the contribution "Status and Trends in Networking at LHC Tier1 Facilities" to CHEP 2012. proactive investment is required at the Tier-1 sites. Bandwidth requirements will grow beyond current capacity and the required upgrades are expected in 2015. In close cooperation with DFN. KIT drives the upgrade from 10GE to 100GE. The process is divided into several phases. due to upgrade costs and differing requirements in different parts of the network infrastructure. The requirements of the different phases as well as the planned topology will be described. Some of the obstacles we discovered during the deployment will be discussed and solutions or workarounds presented.

    The 2MASS Catalog Server Kit is open source software for use in easily constructing a high performance search server for important astronomical catalogs. This software utilizes the open source RDBMS PostgreSQL, therefore, any users can setup the database on their local computers by following step-by-step installation guide. The kit provides highly optimized stored functions for positional searchs similar to SDSS SkyServer. Together with these, the powerful SQL environment of PostgreSQL will meet various user's demands. We released 2MASS Catalog Server Kit version 2.1 in 2012 May, which supports the latest WISE All-Sky catalog (563,921,584 rows) and 9 major all-sky catalogs. Local databases are often indispensable for observatories with unstable or narrow-band networks or severe use, such as retrieving large numbers of records within a small period of time. Herein, we report a simple lyophilized kit formulation for labeling peptides with (18)F, based on the aluminum-fluoride procedure. The prototype kit contains IMP485, a NODA (1,4,7-triazacyclononane-1,4-diacetate)-MPAA (methyl phenylacetic acid)-di-HSG (histamine-succinyl-glycine) hapten-peptide, [NODA-MPAA-D-Lys(HSG)-D-Tyr-D-Lys(HSG)-NH(2)], used for pretargeting, but we also examined a similar kit formulation for a somatostatin-binding peptide [IMP466, NOTA-D-Phe-Cys-Phe-D-Trp-Lys-Thr-Cys-Throl] bearing a NOTA ligand to determine if the benefits of using a kit can be extended to other AlF-binding peptides. The NODA-MPAA ligand forms a single stable complex with (AlF)(2+) in high yields. In order to establish suitable conditions for a facile kit, the formulation was optimized for pH, peptide to Al(3+) ratio, bulking agent, radioprotectant, and the buffer. For optimal labeling, the kit was reconstituted with an aqueous solution of (18)F(-) and ethanol (1:1), heated at 100-110 °C for 15 min, and then simply and rapidly purified using one of two equally effective solid-phase extraction (SPE) methods. Al(18)F-IMP485 was isolated as a single isomer complex, in high yield (45-97%) and high specific activity (up to 223 GBq/μmol), within 20 min. The labeled product was stable in human serum at 37 °C for 4 h and in vivo, urine samples showed the intact product was eliminated. Tumor targeting of the Al(18)F-IMP485 in nude mice bearing human colon cancer xenografts pretargeted with an anti-CEACAM5 bispecific antibody showed very low uptake (0.06% ± 0.02 ID/g) in bone, further illustrating its stability.     A novel effective co-reactant for electrogenerated chemiluminescence (ECL) of Ru(bpy)(3)(2+) has been found. Alpha-position-dialkylated thiophene derivatives such as 2,5-dimethylthiophene (DMT) could be used as a co-reactant for Ru(bpy)(3)(2+) ECL. The reaction mechanism of the Ru(bpy)(3)(2+)/DMT system was proposed on the basis of the identification of the reaction product, the relationship between the molecular structure and the chemiluminescent intensity, and the electrochemical study. The obtained reaction mechanism was similar to that of the Ru(bpy)(3)(2+)/aliphatic tertiary amine system. Based on these results, the preliminary studies of the Ru(bpy)(3)(2+) ECL detection system using DMT as a co-reactant were performed. Under the optimal ECL conditions, the plot of ECL intensity versus the concentration of Ru(bpy)(3)(2+) was linear over the concentration range 1.0x10(-8) to 1.5x10(-7) M (determination coefficient=0.9996).   4. The SCRAM tool-kit

    This paper proposes a new series of on-orbit capabilities to support the near-term Hubble Space Telescope, Extended Duration Orbiter, Long Duration Orbiter, Space Station Freedom, other orbital platforms, and even the future manned Lunar/Mars missions. These proposed capabilities form a toolkit termed Space Construction, Repair, and Maintenance (SCRAM). SCRAM addresses both intra-Vehicular Activity (IVA) and Extra-Vehicular Activity (EVA) needs. SCRAM provides a variety of tools which enable welding, brazing, cutting, coating, heating, and cleaning, as well as corresponding nondestructive examination. Near-term IVA-SCRAM applications include repair and modification to fluid lines, structure, and laboratory equipment inside a shirt-sleeve environment (i.e. inside Spacelab or Space Station). Near-term EVA-SCRAM applications include construction of fluid lines and structural members, repair of punctures by orbital debris, refurbishment of surfaces eroded by contaminants. The SCRAM tool-kit also promises future EVA applications involving mass production tasks automated by robotics and artificial intelligence, for construction of large truss, aerobrake, and nuclear reactor shadow shields structures. The leading candidate tool processes for SCRAM, currently undergoing research and development, include Electron Beam, Gas Tungsten Arc, Plasma Arc, and Laser Beam. A series of strategic space flight experiments would make SCRAM available to help conquer the space frontier.

    The challenge of a global operations capability led to the Telescience Resource Kit (TReK) project, an in-house software development project of the Mission Operations Laboratory (MOL) at NASA's Marshall Space Flight Center (MSFC). The TReK system is being developed as an inexpensive comprehensive personal computer- (PC-) based ground support system that can be used by payload users from their home sites to interact with their payloads on board the International Space Station (ISS). The TReK project is currently using a combination of the spiral lifecycle model and the incremental lifecycle model. As with any software development project, there are four activities that can be very time consuming: Software design and development, project documentation, testing, and umbrella activities, such as quality assurance and configuration management. In order to produce a quality product, it is critical that each of these activities receive the appropriate amount of attention. For TReK, the challenge was to lay out a lifecycle and project plan that provides full support for these activities, is flexible, provides a way to deal with changing risks, can accommodate unknowns, and can respond to changes in the environment quickly. This paper will provide an overview of the TReK lifecycle, a description of the project's environment, and a general overview of project activities.

  11. Genericity of confined chemical garden patterns with regard to changes in the reactants.

    The growth of chemical gardens is studied experimentally in a horizontal confined geometry when a solution of metallic salt is injected into an alkaline solution at a fixed flow rate. Various precipitate patterns are observed-spirals, flowers, worms or filaments-depending on the reactant concentrations. In order to determine the relative importance of the chemical nature of the reactants and physical processes in the pattern selection, we compare the structures obtained by performing the same experiment using different pairs of reactants of varying concentrations with cations of calcium, cobalt, copper, and nickel, and anions of silicate and carbonate. We show that although the transition zones between different patterns are not sharply defined, the morphological phase diagrams are similar in the various cases. We deduce that the nature of the chemical reactants is not a key factor for the pattern selection in the confined chemical gardens studied here and that the observed morphologies are generic patterns for precipitates possessing a given level of cohesiveness when grown under certain flow conditions.     A glider kit consists of a new truck without the power train, with the customer supplying the engine, transmission, clutch, driveline and rear axles for mounting in a new frame and cab complete with steering axle, wiring harnesses, exhaust system and cooling system. Glider kits have long been regarded as a cost effective means of replacing accident or fire damaged late model vehicles. The financial advantages, assembly and remanufacturing are discussed. Points out that at a basic cost about one third of a new truck, a glider kit offers lower cost of borrowed capital, tax savings, and elimination of high maintenance and downtime costs. The paper concludes that use of glider kits as an alternative to major repair or reconditioning or outright replacement of coal haulers presents some interesting options.

    The overall goal of this project was to evaluate the ability of novel adsorbent/reactants to remove specific toxic target chemicals from ash and scrubber pond effluents while producing stable residuals for ultimate disposal. The target chemicals studied were arsenic (As(III) and As(V)), mercury (Hg(II)) and selenium (Se(IV) and Se(VI)). The adsorbent/reactants that were evaluated are iron sulfide (FeS) and pyrite (FeS{sub 2}). Procedures for measuring concentrations of target compounds and characterizing the surfaces of adsorbent-reactants were developed. Effects of contact time, pH (7, 8, 9, 10) and sulfate concentration (0, 1, 10 mM) on removal of all target compounds on both adsorbent-reactants were determined. Stability tests were conducted to evaluate the extent to which target compounds were released from the adsorbent-reactants when pH changed. Surface characterization was conducted with x-ray photoelectron spectroscopy (XPS) to identify reactions occurring on the surface between the target compounds and surface iron and sulfur. Results indicated that target compounds could be removed by FeS{sub 2} and FeS and that removal was affected by time, pH and surface reactions. Stability of residuals was generally good and appeared to be affected by the extent of surface reactions. Synthesized pyrite and mackinawite appear to have the required characteristics for removing the target compounds from wastewaters from ash ponds and scrubber ponds and producing stable residuals.

    With the design and development of the Orbital Maneuvering Vehicle (OMV) progressing toward an early 1990 initial operating capability (IOC), a new era in remote space operations will evolve. The logical progression to OMV front end kits would make available in situ satellite servicing, repair, and consummables resupply to the satellite community. Several conceptual design study efforts are defining representative kits (propellant tanks, debris recovery, module servicers); additional focus must also be placed on an efficient combination module servicer and consummables resupply kit. A remote servicer kit of this type would be designed to perform many of the early maintenance/resupply tasks in both nominal and high inclination orbits. The kit would have the capability to exchange Orbital Replacement Units (ORUs), exchange propellant tanks, and/or connect fluid transfer umbilicals. Necessary transportation system functions/support could be provided by interfaces with the OMV, Shuttle (STS), or Expendable Launch Vehicle (ELV). Specific remote servicer kit designs, as well as ground and flight demonstrations of servicer technology are necessary to prepare for the potential overwhelming need. Ground test plans should adhere to the component/system/breadboard test philosophy to assure maximum capability of one-g testing. The flight demonstration(s) would most likely be a short duration, Shuttle-bay experiment to validate servicer components requiring a micro-g environment.

A variant of instantaneous reactions is considered that explains the proposed approach simply. In particular, the turbulent mixing events according to this approach are uniquely related to the acts of chemical interaction, which makes it possible to exclude from consideration the mixing of inert impurities–the most difficult point of the theory formulated using classical notions. The obtained system of equations is closed without introducing arbitrarily adopted correlations, by naturally introducing the concept of effective reaction and writing the equations of conservation for both the concentrations of reactants and their volumes.

    Heterogeneous catalysis occurs at the interface between a solid catalyst and the reactants. The structure of metal catalyst nanoparticles at the metal-gas interface is a key factor that determines catalytic selectivity and activity. Here we report that second-generation nanoclusters are formed on the initial catalyst nanoparticles as a result of interaction with the reactant molecules when the nanoparticles are in a gas phase at Torr pressure or higher. The formation of the second-generation nanoclusters is manifested by a decrease of the average coordination number of the metal atoms and a shift of their core level energies in the presence of gases. The formation of second-generation nanoclusters increases the number of undercoordinated sites, which are the most active for catalysis in many cases. PMID:27328034

    In mammals, females are born with finite numbers of oocytes stockpiled as primordial follicles. Oocytes are "reawakened" via an ovarian-intrinsic process that initiates their growth. The forkhead transcription factor Foxo3 controls reawakening downstream of PI3K-AKT signaling. However, the identity of the presumptive upstream cell surface receptor controlling the PI3K-AKT-Foxo3 axis has been questioned. Here we show that the receptor tyrosine kinase Kit controls reawakening. Oocyte-specific expression of a novel constitutively-active KitD818V allele resulted in female sterility and ovarian failure due to global oocyte reawakening. To confirm this result, we engineered a novel loss-of-function allele, KitL. Kit inactivation within oocytes also led to premature ovarian failure, albeit via a contrasting phenotype. Despite normal initial complements of primordial follicles, oocytes remained dormant with arrested oocyte maturation. Foxo3 protein localization in the nucleus versus cytoplasm explained both mutant phenotypes. These genetic studies provide formal genetic proof that Kit controls oocyte reawakening, focusing future investigations into the causes of primary ovarian insufficiency and ovarian aging.     The goal of this project is to develop a fluid dynamics demonstration/experiment kit that can be used by professors and graduate students at high school outreach events. The demonstrations in the kit will be easy to use and true crowd pleasers in order to inspire understanding and pique curiosity about the physics of flow. The kits will be inexpensive, containing readily available materials so that teachers can duplicate the demonstrations and experiments. The kits will be left with the teachers as a gift from the American Physics Society. The experiments and demonstrations cover the concepts of conservation of mass, momentum, and energy, Bernoulli's equation, frictional losses and the ideal gas law. For each experiment, the teachers will receive presentation material, access to instructional videos, plus a worksheet that can be used in a high school physics classroom. This kit has been developed through the efforts of the APS-DFD Mentoring and Outreach Committee and has received funding from the APS-DFD. Work funded by the APS-DFD.

    The deterioration of stannous ion (Sn++) in inhouse-prepared and commercial radiopharmaceutical kits was studied. Sn++ content of three types of nonlyophilized, deoxygenated, aqueous inhouse-prepared kits [diethylenetriamine pentaacetic acid (DTPA), pyrophosphate and glucoheptonate] and of three commercially prepared kits (two lyophilized pyrophosphate kits and one diphosphonate in sealed glass ampul kit) was measured by differential pulse polarography. Inhouse-prepared kits were assayed initially and after storage for 6, 12, 24 and 48 days at 24, 5 and -18 C. Commercial kits were assayed initially and after storage for 12, 24 and 48 days at 5 and 24 C. Of the inhouse-prepared kits, Sn++ stability when stored for 48 days at 5 and 24 C. Freezer storage should be used, when possible, to insure maximum stability of Sn++ in inhouse-prepared, nonlyophilized ratiopharmaceutical kits. The commercial procedures of lyophilization and of sealing the reagent in a sealed glass ampul prolong Sn++ stability. PMID:507080

  3. Effects of self-propagating synthesis reactant compact character on ignition, propagation, and microstructure

    Studies of reactions involving Ti to produce TiC, TiB{sub 2}, TiC + TiB{sub 2}, or 3TiB{sub 2} + 5Al{sub 2}O{sub 3} ignited at one end of test plates showed that reactant powder compact densities were a major factor in the reaction propagation rate along the plate, i.e., a maximum in propagation rate was observed at {approx}60 {plus minus} 10% of theoretical density. At higher densities, propagation rates not only decreased but terminated due to self extinction in some cases or failed to ignite and propagate, typically at {ge}90% of theoretical density. Both reactant particle size and shape can also affect results, i.e., compacts of large (200 {mu}m in diameter) Ti particles, Ti flakes or foil, or wires failed to ignite or had slower propagation rates. Also, the ignition and propagation rates of carbon fiber tows infiltrated with titanium metal powders depended significantly on the local thermal conductivity. However, overall propagation rates for a given range of reactant compact microstructures increased with the heat of the reaction involved.

  4. Shuttle Kit Freezer Refrigeration Unit Conceptual Design

    The refrigerated food/medical sample storage compartment as a kit to the space shuttle orbiter is examined. To maintain the -10 F in the freezer kit, an active refrigeration unit is required, and an air cooled Stirling Cycle refrigerator was selected. The freezer kit contains two subsystems, the refrigeration unit, and the storage volume. The freezer must provide two basic capabilities in one unit. One requirement is to store 215 lbs of food which is consumed in a 30-day period by 7 people. The other requirement is to store 128.3 lbs of medical samples consisting of both urine and feces. The unit can be mounted on the lower deck of the shuttle cabin, and will occupy four standard payload module compartments on the forward bulkhead. The freezer contains four storage compartments.

    One of the major needs of the law enforcement field is a product that quickly, accurately, and inexpensively identifies whether a person has recently fired a gun--even if the suspect has attempted to wash the traces of gunpowder off. The Field Test Kit for Gunshot Residue Identification based on Sandia National Laboratories technology works with a wide variety of handguns and other weaponry using gunpowder. There are several organic chemicals in small arms propellants such as nitrocellulose, nitroglycerine, dinitrotoluene, and nitrites left behind after the firing of a gun that result from the incomplete combustion of the gunpowder. Sandia has developed a colorimetric shooter identification kit for in situ detection of gunshot residue (GSR) from a suspect. The test kit is the first of its kind and is small, inexpensive, and easily transported by individual law enforcement personnel requiring minimal training for effective use. It will provide immediate information identifying gunshot residue.

    FES is a cytoplasmic tyrosine kinase activated by several membrane receptors, originally identified as a viral oncogene product. We have recently identified FES as a crucial effector of oncogenic KIT mutant receptor. However, FES implication in wild-type KIT receptor function was not addressed. We report here that FES interacts with KIT and is phosphorylated following activation by its ligand SCF. Unlike in the context of oncogenic KIT mutant, FES is not involved in wild-type KIT proliferation signal, or in cell adhesion. Instead, FES is required for SCF-induced chemotaxis. In conclusion, FES kinase is a mediator of wild-type KIT signalling implicated in cell migration.

    Western Research Institute (WRI) is commercializing Diesel Dog Portable Soil Test Kits for performing analysis of fuel-contaminated soils in the field. The technology consists of a method developed by WRI (U.S. Patents 5,561,065 and 5,976,883) and hardware developed by WRI that allows the method to be performed in the field (patent pending). The method is very simple and does not require the use of highly toxic reagents. The aromatic components in a soil extract are measured by absorption at 254 nm with a field-portable photometer. WRI added significant value to the technology by taking the method through the American Society for Testing and Materials (ASTM) approval and validation processes. The method is designated ASTM Method D-5831-96, Standard Test Method for Screening Fuels in Soils. This ASTM designation allows the method to be used for federal compliance activities. In FY 99, twenty-five preproduction kits were successfully constructed in cooperation with CF Electronics, Inc., of Laramie, Wyoming. The kit components work well and the kits are fully operational. In the calendar year 2000, kits were provided to the following entities who agreed to participate as FY 99 and FY 00 JSR (Jointly Sponsored Research) cosponsors and use the kits as opportunities arose for field site work: Wyoming Department of Environmental Quality (DEQ) (3 units), F.E. Warren Air Force Base, Gradient Corporation, The Johnson Company (2 units), IT Corporation (2 units), TRC Environmental Corporation, Stone Environmental, ENSR, Action Environmental, Laco Associates, Barenco, Brown and Caldwell, Dames and Moore Lebron LLP, Phillips Petroleum, GeoSyntek, and the State of New Mexico. By early 2001, ten kits had been returned to WRI following the six-month evaluation period. On return, the components of all ten kits were fully functional. The kits were upgraded with circuit modifications, new polyethylene foam inserts, and updated instruction manuals.

    Although the bone marrow mononuclear cell (BMMNC) is known as an ideal cell type for cell-based therapy for MI treatment, the effective subpopulation still remains unknown. Our study aimed at identifying the optimal subset of BMMNCs suited for cardiac regeneration. In this study, we observed that MI led to (i) a significant increase of the c-kit+AT2R+ BMMNC subpopulation in mice and (ii) a modest increase of AT2R+ BMMNCs in humans. c-kit+AT2R+ and c-kit+AT2R− BMMNC subpopulations were obtained from mice after MI. Then, we cocultured cardiac H9C2 cells with c-kit+AT2R+, c-kit+AT2R−, and unfractionated BMMNCs; finally, we found that the c-kit+AT2R+ subset is superior to the c-kit+AT2R− subset in improving cardiomyocyte protection in vitro. Of note, c-kit+AT2R+ BMMNCs showed a more robust migration capacity than c-kit+AT2R− and unfractionated BMMNCs in vitro and in vivo. Additionally, compared to c-kit+AT2R− and unfractionated BMMNCs, intravenous transplantation of c-kit+AT2R+ BMMNC resulted in smaller infarct size and lower levels of inflammatory reactions in heart tissue, leading to a higher global heart function improvement. In conclusion, our results indicate that the c-kit+AT2R+ BMMNC subpopulation exerts a protective effect against MI and shows promising therapeutic possibilities with regard to the treatment of ischemic heart disease.     Optimal means of characterizing the distribution of product energy states resulting from reactive collisions of molecules with restricted distributions of initial states are considered, along with those for characterizing the particular reactant state distribution which yields a given set of product states at a specified total energy. It is suggested to represent the energy-dependence of global-type results in the form of square-faced bar plots, and of data for specific-type experiments as triangular-faced prismatic plots. The essential parameters defining the internal state distribution are isolated, and the information content of such a distribution is put on a quantitative basis. The relationship between the information content, the surprisal, and the entropy of the continuous distribution is established. The concept of an entropy deficiency, which characterizes the specificity of product state formation, is suggested as a useful measure of the deviance from statistical behavior. The degradation of information by experimental averaging is considered, leading to bounds on the entropy deficiency.

    We determined c-kit expression in the stroma and epithelia of benign, borderline, and malignant phyllodes tumors (PTs), respectively, as well as the relationship between c-kit expression in stromal elements and KIT gene copy number variations (CNVs). To assess c-kit expression and KIT CNVs, 348 PT cases were studied: 120 (34.4 %) benign cases, 115 (33.1 %) borderline cases, and 113 (32.5 %) malignant cases. All of these cases were evaluated for c-kit (CD117) expression using immunohistochemistry. Forty-two cases (29 c-kit-positive in the stromal cells cases and 13 negative cases) were investigated for KIT gene CNVs via genomic polymerase chain reaction (PCR). The overall rate of c-kit positivity in the stroma was 46.8 %, as well as 24.2, 53.1, and 64.6 %, respectively, in PTs of three different grades. However, in the majority of cases, the epithelia were c-kit positive (98.2 %), and the positivity was 100, 99.1, and 95 % in PTs of three different grades, respectively. There was a significant change in the expression of c-kit in the stroma and epithelia according to grade (P < 0.001, P = 0.014). From the genomic PCR results, we can confirm that c-kit positivity in the stroma is directly correlated with KIT gene copy numbers increases (P = 0.003, P = 0.041). We demonstrated that c-kit expression in the stroma of PTs is positively associated with malignancy. c-Kit epithelial positivity was inversely correlated with PTs malignancy. c-Kit overexpression in the stroma was related to KIT gene copy numbers increases.     Closed form analytical expressions are obtained for predicting the limited rate of reactant conversion in a binary reaction of the type F + rO yields (1 + r) Product in unpremixed homogeneous turbulence. These relations are obtained by means of a single point Probability Density Function (PDF) method based on the Amplitude Mapping Closure. It is demonstrated that with this model, the maximum rate of the reactants' decay can be conveniently expressed in terms of definite integrals of the Parabolic Cylinder Functions. For the cases with complete initial segregation, it is shown that the results agree very closely with those predicted by employing a Beta density of the first kind for an appropriately defined Shvab-Zeldovich scalar variable. With this assumption, the final results can also be expressed in terms of closed form analytical expressions which are based on the Incomplete Beta Functions. With both models, the dependence of the results on the stoichiometric coefficient and the equivalence ratio can be expressed in an explicit manner. For a stoichiometric mixture, the analytical results simplify significantly. In the mapping closure, these results are expressed in terms of simple trigonometric functions. For the Beta density model, they are in the form of Gamma Functions. In all the cases considered, the results are shown to agree well with data generated by Direct Numerical Simulations (DNS). Due to the simplicity of these expressions and because of nice mathematical features of the Parabolic Cylinder and the Incomplete Beta Functions, these models are recommended for estimating the limiting rate of reactant conversion in homogeneous reacting flows. These results also provide useful insights in assessing the extent of validity of turbulence closures in the modeling of unpremixed reacting flows.     A 42-kDa protein appeared at a much higher concentration in plasma from Trypanosoma brucei-resistant (C57BL/6) mice after infection than in plasma from trypanosome-susceptible (C3H/He) mice. This protein was purified by sequential steps of gel filtration, protein A-Sepharose affinity chromatography, isoelectric focusing, and ammonium sulfate precipitation.     Merging the chemistry of transfer hydrogenation and carbonyl or imine addition, a broad new family of redox-neutral or reductive hydrohydroxyalkylations and hydroaminomethylations have been developed. In these processes, hydrogen redistribution between alcohols and π-unsaturated reactants is accompanied by C-C bond formation, enabling direct conversion of lower alcohols to higher alcohols. Similarly, hydrogen redistribution between amines to π-unsaturated reactants results in direct conversion of lower amines to higher amines. Alternatively, equivalent products of hydrohydroxyalkylation and hydroaminomethylation may be generated through the reaction of carbonyl compounds or imines with π-unsaturated reactants under the conditions of 2-propanol-mediated reductive coupling. Finally, using vicinally dioxygenated reactants, that is, diol, ketols, or diones, successive transfer hydrogenative coupling occurs to generate 2 C-C bonds, resulting in products of formal [4+2] cycloaddition.   1. Single cell performance studies on the Fe/Cr Redox Energy Storage System using mixed reactant solutions at elevated temperature

    Experimental studies in a 14.5 sq cm single cell system using mixed reactant solutions at 65 C are described. Systems were tested under isothermal conditions i.e., reactants and the cell were at the same temperature. Charging and discharging performance were evaluted by measuring watt-hour and coulombic efficiencies, voltage-current relationships, hydrogen evolution and membrane resistivity. Watt-hour efficiencies ranged from 86% at 43 ma/sq cm to 75% at 129 ma/sq cm with corresponding coulombic efficiencies of 92% and 97%, respectively. Hydrogen evolution was less than 1% of the charge coulombic capacity during charge-discharge cycling. Bismuth and bismuth-lead catalyzed chromium electrodes maintained reversible performance and low hydrogen evolution under normal and adverse cycling conditions. Reblending of the anode and cathode solutions was successfully demonstrated to compensate for osmotic volume changes. Improved performance was obtained with mixed reactant systems in comparison to the unmixed reactant systems.

    In addition to its physiologic role as central regulator of the hematopoietic and reproductive systems, the Kit receptor tyrosine kinase (RTK) is pathologically overexpressed in some forms of leukemia and constitutively activated by oncogenic mutations in mast-cell proliferations and gastrointestinal stromal tumors. To gain insight into the general activation and signaling mechanisms of RTKs, we investigated the activation-dependent dynamic membrane distributions of wild-type and oncogenic forms of Kit in hematopoietic cells. Ligand-induced recruitment of wild-type Kit to lipid rafts after stimulation by Kit ligand (KL) and the constitutive localization of oncogenic Kit in lipid rafts are necessary for Kit-mediated proliferation and survival signals. KL-dependent and oncogenic Kit kinase activity resulted in recruitment of the regulatory phosphatidylinositol 3-kinase (PI3-K) subunit p85 to rafts where the catalytical PI3-K subunit p110 constitutively resides. Cholesterol depletion by methyl-β-cyclodextrin prevented Kit-mediated activation of the PI3-K downstream target Akt and inhibited cellular proliferation by KL-activated or oncogenic Kit, including mutants resistant to the Kit inhibitor imatinib-mesylate. Our data are consistent with the notion that Kit recruitment to lipid rafts is required for efficient activation of the PI3-K/Akt pathway and Kit-mediated proliferation. PMID:17554062
