    The EPA Office of Research and Development (ORD) identifies and prioritizes future research areas through a Waste Research Coordination Team. The team works together to plan the ORD Superfund research program, and it has members from each of the ORD laboratories as well as representatives from the Superfund office. Superfund scientists have identified several research topics in applied research areas to improve risk assessment methods and reduce uncertainty in site-specific risk assessments. Research areas include: dermal exposure models and toxicity values, improved methods for exposure factors, pharmaco-kinetic dose-response models, bioavailability and statistical methods. This paper presents ORD future research plans in response to these identified research areas.

    This paper presents major research needs for the Superfund program, and provides an overview of the EPA Office of Research and Development's (ORDs) current human health research program designed to fill some of those data gaps. Research is presented in terms of the risk paradigm and covers exposure, effects, and assessment activities directly funded by Superfund, as well as research not funded by Superfund but directly applicable to Superfund research needs. Research on risk management is not covered. Current research activities conducted by the Superfund program office are also included to provide a full picture of Superfund human health research activities being conducted by EPA.

    The scientific name Sorex niger Ord, 1815 (Mammalia, Soricidae) was originally applied to a North American species that George Ord called the “Black Shrew.” The origin of the name “Black Shrew,” however, was obscure, and Samuel Rhoads subsequently wrote that the species represented by this name could not be determined. The names Sorex niger Ord and Black Shrew have since been mostly forgotten. Two of Ord's contemporaries, however, noted that Ord's use of these names probably alluded to Benjamin Smith Barton's Black Shrew, whose discovery near Philadelphia was announced by Barton in 1806. Examination of two unpublished illustrations of the Black Shrew made by Barton indicates that the animal depicted is Blarina brevicauda (Say, 1822). Had the connection between Ord's and Barton's names been made more clearly, one of the most common mammals in eastern North America would bear a different scientific name today. This connection also would have affected the validity of Sorex niger Horsfield, 1851. While Sorex niger Ord remains a nomen nudum, the animal it referenced can now be identified.

    Research and development spending at the Environmental Protection Agency is slated to rise more than 5% in President Bill Clinton's fiscal 1994 budget. Congress is stepping in, however, and may have something to say not only about how much money is spent, but also how it is spent. For the first time in a decade, formal authorization of EPA's Office of Research and Development (ORD) is moving through Congress. The ORD authorization was approved May 20 in the House Science Committee's Subcommittee on Technology, Environment and Aviation. Introduced by subcommittee chairman Rep. Tim Valentine (D.-N.C.), the authorization bill (H.R. 1994) would provide $475 million in funding for fiscal 1994. This equals the amount proposed by Clinton, $536 million, if about $60 million earmarked for Superfund-related research is removed. The Valentine bill would set out programmatic guidelines for EPA research, requiring fundamental research in ecology, health, and risk reduction. It would also require the agency's Science Advisory Board to review these programs and submit progress reports to Congress every two years. Another part of the bill would require EPA to consolidate agency efforts to identify, compare, and assess risk to public health and the environment posed by pollution.

    The Clean Air Power Initiative (CAPI) is a multi-stakeholder project intended to improve air pollution control efforts involving the power generating industry. This paper documents the progress made in the first year of the initiative, which included a number of meetings with interested stakeholders and development and analysis of alternative approaches for more efficient and effective pollution control. The project`s goal is to develop an integrated regulatory strategy or three major pollutants emitted from electric power generators; namely, sulfur dioxide, nitrogen oxides, and, potentially, mercury. Major reductions in these pollutants are expected to be needed to reduce the detrimental health effects of ground-level ozone, fine particles, and hazardous air pollutants and reduce the environmental effects of acidification, eutrophication, ecosystem, crop, and materials damage, and regional haze. The Clean Air Power Initiative has considered, where feasible, new approaches to pollution control that recognize the long-range transport of many air pollutants and the economic benefits of emissions trading. The project was initiated by EPA`s Assistant Administrator for Air and Radiation in 1995. As individual companies develop and implement strategies to participate in more competitive power markets, they could benefit from greater certainty in being able to plan for and reduce costs of future environmental regulations. The EPA is interested in reinventing its regulatory approach to reduce the number, administrative complexity, and cost of its requirements while improving the likelihood of achieving environmental results.

    The Electrostatic Plasma Accelerator (EPA) is a thruster concept which promises specific impulse levels between low power arcjets and those of the ion engine while retaining the relative simplicity of the arcjet. The EPA thruster produces thrust through the electrostatic acceleration of a moderately dense plasma. No accelerating electrodes are used and the specific impulse is a direct function of the applied discharge voltage and the propellant atomic mass. The goal of the present program is to demonstrate feasibility of the EPA thruster concept through experimental and theoretical investigations of the EPA acceleration mechanism and discharge chamber performance. Experimental investigations will include operating the test bed ion (TBI) engine as an EPA thruster and parametrically varying the thruster geometry and operating conditions to quantify the electrostatic plasma acceleration effect. The theoretical investigations will include the development of a discharge chamber model which describes the relationships between the engine size, plasma properties, and overall performance. For the EPA thruster to be a viable propulsion concept, overall thruster efficiencies approaching 30% with specific impulses approaching 1000 s must be achieved.

    We document an open-source toolbox for drawing large-scale undirected graphs. This toolbox is based on a previously implemented closed-source algorithm known as VxOrd. Our toolbox, which we call OpenOrd, extends the capabilities of VxOrd to large graph layout by incorporating edge-cutting, a multi-level approach, average-link clustering, and a parallel implementation. At each level, vertices are grouped using force-directed layout and average-link clustering. The clustered vertices are then re-drawn and the process is repeated. When a suitable drawing of the coarsened graph is obtained, the algorithm is reversed to obtain a drawing of the original graph. This approach results in layouts of large graphs which incorporate both local and global structure. A detailed description of the algorithm is provided in this paper. Examples using datasets with over 600K nodes are given. Code is available at www.cs.sandia.gov/~smartin.

    Nitrogen oxides (NO{sub x}) are highly reactive and play a major role in the formation of various gases and particles in the atmosphere which lead to harmful effects on human health and welfare. This paper briefly describes the multiple impacts on human health and welfare that result from emissions of NO{sub x} and describes EPA`s strategy to integrate NO{sub x} reductions from various mobile and stationary sources in a balanced manner to achieve environmental benefits. Based on modeling information, it is clear that substantial reductions in NO{sub x} emissions over large geographic areas are needed if many densely populated areas of the nation are to attain the national ambient air quality standard for ozone. In addition to attainment of the health standard for ozone, the reduction of NO{sub x} emissions will also likely improve the nation`s environment by reducing adverse impacts of acid deposition, eutrophication of waterbodies, global warming, nitrogen dioxide, particulate matter, stratospheric ozone depletion, toxics, and visibility.

Supported by advances in informatics, high-throughput screening (HTS) technologies, and systems biology, the U.S. Environmental Protection Agency EPA is developing robust and flexible computational tools that can be applied to the thousands of chemicals in commerce, and contaminant mixtures found in air, water, and hazardous-waste sites.     Ord's kangaroo rat is a kangaroo rat native to western North America. In this study, we first reported the complete mitochondrial genome of Dipodomys ordii that the first has the complete mitochondrial genome in the genus of Heteromyidae. The mitogenome is a circular molecule of 16 257 bp in length, containing 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs and a putative displacement loop region. All protein-coding genes started with a traditional ATN codon and terminated with the mitochondria stop codon (TAA/TAG/AGA) or a single T base. The gene order and composition of mitogenome was similar to that of most other Sciurognathi species and its GC content was 36.73%. Thirteen protein-coding genes of D. ordii together with eight other closely species were used to construct the species phylogenetic tree for verification of the accuracy of new determined mitogenome sequences.

The National Renewable Energy Laboratory (NREL) provided technical assistance for this project. The purpose of this report is to assess the site for a possible photovoltaic (PV) system installation and estimate the cost, performance, and site impacts of different PV options. In addition, the report recommends financing options that could assist in the implementation of a PV system at the site.

Supported by advances in informatics, high-throughput screening (HTS) technologies, and systems biology, EPA is developing robust and flexible computational tools that can be applied to the thousands of chemicals in commerce, and contaminant mixtures found in America’s air, water, and hazardous-waste sites. The ORD Computational Toxicology Research Program (CTRP) is composed of three main elements. The largest component is the National Center for Computational Toxicology (NCCT), which was established in 2005 to coordinate research on chemical screening and prioritization, informatics, and systems modeling. The second element consists of related activities in the National Health and Environmental Effects Research Laboratory (NHEERL) and the National Exposure Research Laboratory (NERL). The third and final component consists of academic centers working on various aspects of computational toxicology and funded by the EPA Science to Achieve Results (STAR) program.     The application of electric propulsion to communications satellites, however, has been limited to the use of hydrazine thrusters with electric heaters for thrust and specific impulse augmentation. These electrothermal thrusters operate at specific impulse levels of approximately 300 s with heater powers of about 500 W. Low power arcjets (1-3 kW) are currently being investigated as a way to increase specific impulse levels to approximately 500 s. Ion propulsion systems can easily produce specific impulses of 3000 s or greater, but have yet to be applied to communications satellites. The reasons most often given for not using ion propulsion systems are their high level of overall complexity, low thrust with long burn times, and the difficulty of integrating the propulsion system into existing commercial spacecraft busses. The Electrostatic Plasma Accelerator (EPA) is a thruster concept which promises specific impulse levels between low power arcjets and those of the ion engine while retaining the relative simplicity of the arcjet. The EPA thruster produces thrust through the electrostatic acceleration of a moderately dense plasma. No accelerating electrodes are used and the specific impulse is a direct function of the applied discharge voltage and the propellant atomic mass.

    The absolute configuration (AC) of the bioactive metabolites phyllostin (1) and scytolide (2), two hexahydro-1,4-benzodioxines produced by Phyllosticta cirsii, and oxysporone (3), a dihydrofuropyranone recently isolated from a strain of Diplodia africana, has been assigned by computational analysis of their optical rotatory dispersion (ORD), electronic circular dichroism (ECD), and vibrational circular dichroism (VCD) spectra. Computational prediction of ORD, ECD, and VCD allowed us to assign (3S,4aR,8S,8aR) AC to naturally occurring (-)-1, while (4aR,8S,8aR) AC was assigned to (-)-2 employing only ECD and VCD, because in this case ORD analysis turned out to be unsuitable for AC assignment. Theoretical prediction of both ORD and ECD spectra of 3 led to assignment of (4S,5R,6R) AC to (+)-3. In this case a satisfactory agreement between experimental and calculated VCD spectra was obtained only after taking into account solvent effects. This study shows that in the case of flexible and complex natural products only a concerted application of more than a single chiroptical technique permits unambiguous assignment of absolute configuration.

    Among the enzymatic steps in the aflatoxin biosynthetic pathway, the conversion of O-methylsterigmatocystin to aflatoxin has been proposed to be catalyzed by an oxidoreductase. Transformants of Aspergillus flavus 649WAF2 containing a 3.3-kb genomic DNA fragment and the aflatoxin biosynthesis regulatory gene aflR converted exogenously supplied O-methylsterigmatocystin to aflatoxin B1. A gene, ord1, corresponding to a transcript of about 2 kb was identified within the 3.3-kb DNA fragment. The promoter region presented a putative AFLR binding site and a TATA sequence. The nucleotide sequence of the gene revealed an open reading frame encoding a protein of 528 amino acids with a deduced molecular mass of 60.2 kDa. The gene contained six introns and seven exons. Heterologous expression of the ord1 open reading frame under the transcriptional control of the Saccharomyces cerevisiae galactose-inducible gal1 promoter results in the ability to convert O-methylsterigmatocystin to aflatoxin B1. The data indicate that ord1 is sufficient to accomplish the last step of the aflatoxin biosynthetic pathway. A search of various databases for similarity indicated that ord1 encodes a cytochrome P-450-type monooxygenase, and the gene has been assigned to a new P-450 gene family named CYP64. PMID:9143099

    High densities of an aridland granivore, Ord's kangaroo rat (Dipodomys ordii), have been documented in floodplain habitats along the Yampa River in northwestern Colorado. Despite a high probability of inundation and attendant high mortality during the spring flood period, the habitat is consistently recolonized. To understand factors that potentially make riparian habitats attractive to D. ordii, we compared density and spatial pattern of seeds, density of a competitor (western harvester ant, Pogonomyrmex occidentalis), and digging energetics within floodplain habitats and between floodplain and adjacent upland habitats. Seed density within the floodplain was greatest in the topographically high (rarely flooded) floodplain and lowest immediately after a spring flood in the topographically low (frequently flooded) floodplain. Seed densities in adjacent upland habitat that never floods were higher than the lowest floodplain habitat. In the low floodplain prior to flooding, seeds had a clumped spatial pattern, which D. ordii is adept at exploiting; after spring flooding, a more random pattern resulted. Populations of the western harvester ant were low in the floodplain relative to the upland. Digging by D. ordii was energetically less expensive in floodplain areas than in upland areas. Despite the potential for mortality due to annual spring flooding, the combination of less competition from harvester ants and lower energetic costs of digging might promote the use of floodplain habitat by D. ordii.

  11. Dispersal and Germination Patterns of Monterey Spineflower at Fort Ord Natural Reserve.

    Some species are rare because they are restricted to certain habitats and/or have small population sizes. Monterey spineflower, a federally listed threatened annual plant, is found in open sandy regions of the California coast, in chaparral vegetation around the Monterey Bay. A model based on previous research suggests that the Monterey spineflower population at Fort Ord Natural Reserve should be rapidly increasing, but it is not. This suggests that the model may be using data that overestimates the percentage of spineflower seeds that successfully germinate. I tested three hypotheses to determine the cause of the difference in population sizes between the predicted model and the field results. First, I predicted that the spineflower seeds are blown by the wind into shrubs such as manzanita, and are unable to germinate due to the lack of a suitable environment. I tested this in two ways. A field experiment showed that seeds are easily blow by wind. Next, I took soil cores and found spineflower seeds within the manzanita shrubs. Secondly, I predicted that the germination rate used by the model (90%) was too high. However, my germination experiments did not support this hypothesis because 91% of new seeds successfully germinated. Lastly, I predicted that the newer seeds are more viable than older seeds and therefore have a higher chance of successfully germinating. After germinating seeds in a controlled environment I observed that the seeds from 2014 had a higher number of successfully germinated seeds compared to the number of successfully germinated seeds from 1995 (91% vs 33%). I conclude that the loss of seeds due to wind decreases germination expectancies and older seeds are less viable than new seeds. Therefore, Monterey spineflower is a rare plant because environmental barriers hinder seeds from dispersing to a suitable habitat and successfully germinating while seeds lose viability as they age.

    Aspergillus parasiticus produces the minor aflatoxins M(1) (AFM(1)), M(2) (AFM(2)), GM(1) (AFGM(1)), and GM(2) (AFGM(2)), as well as the major aflatoxins B(1) (AFB(1)), B(2) (AFB(2)), G(1) (AFG(1)), and G(2) (AFG(2)). Feeding of A. parasiticus with aspertoxin (12c-hydroxyOMST) caused AFM(1) and AFGM(1), and cell-free experiments using the microsomal fraction of A. parasiticus and aspertoxin caused production of AFM(1), indicating that aspertoxin is a precursor of AFM(1) and AFGM(1). Feeding of the same fungus with O-methylsterigmatocystin (OMST) caused AFM(1) and AFGM(1) together with AFB(1) and AFG(1); feeding with dihydroOMST (DHOMST) caused AFM(2) and AFGM(2) together with AFB(2) and AFG(2). Incubation of either the microsomal fraction or OrdA enzyme-expressing yeast with OMST caused production of aspertoxin together with AFM(1) and AFB(1). These results demonstrated that the OrdA enzyme catalyzes both 12c-hydroxylation reaction from OMST to aspertoxin and the successive reaction from aspertoxin to AFM(1). In contrast, feeding of the fungus with AFB(1) did not produce any AFM(1), demonstrating that M-/GM-aflatoxins are not produced from B-/G-aflatoxins. Furthermore, AFM(1) together with AFB(1) and AFG(1) was also produced from 11-hydroxyOMST (HOMST) in feeding experiment of A. parasiticus, whereas no aflatoxins were produced when used the ordA deletion mutant. These results demonstrated that OrdA enzyme can also catalyze 12c-hydroxylation of HOMST to produce 11-hydroxyaspertoxin, which serves as a precursor for the production of AFM(1) and AFGM(1). The same pathway may work for the production of AFM(2) and AFGM(2) from DHOMST and dihydroHOMST through the formation of dihydroaspertoxin and dihydro-11-hydroxyaspertoxin, respectively.

    This report presents an intercomparison of three groundwater flow monitoring technologies at a trichloroethylene (TCE) groundwater plume at Operational Unit 1 (OU 1) adjacent to the former Fritzsche Army Airfield at the former Fort Ord Army Base, located on Monterey Bay in northern Monterey County, California. Soil and groundwater at this site became contaminated by fuels and solvents that were burned on a portion of OU 1 called the Fire Drill Area (FDA) as part of firefighter training from 1962 and 1985. Cont Contamination is believed to be restricted to the unconfined A-aquifer, where water is reached at a depth of approximately 60 to 80 feet below the ground surface; the aquifer is from 15 to 20 feet in thickness, and is bounded below by a dense clay layer, the Salinas Valley Aquitard. Soil excavation and bioremediation were initiated at the site of fire training activities in the late 1980s. Since that time a pump-and-treat operation has been operated close to the original area of contamination, and this system has been largely successful at reducing groundwater contamination in this source area. However, a trichloroethylene (TCE) groundwater plume extends approximately 3000 ft (900 m) to the northwest away from the FDA. In this report, we have augmented flow monitoring equipment permanently installed in an earlier project (Oldenburg et al., 2002) with two additional flow monitoring devices that could be deployed in existing monitoring wells, in an effort to better understand their performance in a nearly ideal, homogeneous sand aquifer, that we expected would exhibit laminar groundwater flow owing to the site's relatively simple hydrogeology. The three flow monitoring tools were the Hydrotechnics{reg_sign} In In-Situ Permeable Flow Sensor (ISPFS), the RAS Integrated Subsurface Evaluation Hydrophysical Logging tool (HPL), and the Lawrence Livermore National Laboratory Scanning Colloidal Borescope Flow Meter (SCBFM).     Under the sponsorship of the US EPA`s Office of Mobile Sources, SAI has developed a computer model for nonroad mobile sources. Although data and methodologies continue to be revisited, the alpha version of the model has been completed and is currently under review at EPA. The model contains modules for estimating equipment populations, activity, emission factors, and total emissions. It adjusts these estimates to predict total emissions for the calendar year and at a level of detail specified by the user (e.g., average annual estimates at a national level, hourly estimates for a specific county). The EPA`s nonroad model provides estimates for the following general classes of nonroad equipment: General Nonroad Sources: (1) Lawn and garden equipment; (2) Airport service equipment; (3) Recreational; (4) Light Commercial; (5) Industrial; (6) Construction; (7) Agricultural; and (8) Logging. Special Nonroad Sources: (1) Locomotives; (2) Aircraft; and (3) Marine Vessels. With the exception of locomotives, aircraft, and marine vessels, the methodologies used within the model are quite similar for all these source groupings. Information for specific equipment types (e.g., 15-25 horsepower, gasoline 2-stroke engine commercial turf equipment) will also be provided if the user requests this level of detail. Written in FORTRAN and designed for personal computers (though it can be ported to workstations), this new emissions tool includes an interface to improve its ease of use and flexibility. We present here some of the features of the model and provide an overview of the internal methods used for the equipment groups listed under general nonroad sources.

    The yeast COX5a and COX5b genes encode isoforms of subunit Va of the mitochondrial inner membrane protein complex cytochrome c oxidase. These genes have been shown to be inversely regulated at the level of transcription by oxygen, which functions through the metabolic coeffector heme. In earlier studies we identified several regulatory elements that control transcriptional activation and aerobic repression of one of these genes, COX5b. Here, we report the isolation of trans-acting mutants that are defective in the aerobic repression of COX5b transcription. The mutants fall into two complementation groups. One group specifies ROX1, which encodes a product reported to be involved in transcriptional repression. The other group identified the gene we have designated ORD1. Mutations in ORD1 cause overexpression of COX5b aerobically but do not affect the expression of the hypoxic genes CYC7, HEM13, and ANB1. ORD1 mutations also do not affect the expression of the aerobic genes COX5a, CYC1, ROX1, ROX3, and TIF51A. The yeast genome contains a single ORD1 gene that resides on chromosome XI. Strains carrying chromosomal deletions of the ORD1 locus are viable and exhibit phenotypes similar to, but less severe than, that of the original mutant. The nucleotide sequence of ORD1 revealed that it is identical to IXR1, a yeast gene whose product contains two high mobility group boxes, binds to platinated DNA, and confers sensitivity to the antitumor drug cisplatin. Consistent with the latter observations, we found that the ORD1 product could bind to both the upstream region of COX5b and to DNA modified with cisplatin. Images PMID:8041793

    During field study of Ord's kangaroo rat (Dipodomys ordii) at Suffield National Wildlife Area, Alberta, Canada, a high prevalence of parasitism by botfly (Diptera: Oestridae) larvae was observed. Botflies have not previously been documented as parasites of kangaroo rats. Botfly parasitism could have a significant impact on the growth, survival, and reproduction of Ord's kangaroo rat, which is considered a vulnerable species in Canada. Therefore, it is important to investigate how botfly parasitism varies with season and with gender or age of host. In 1995, 525 individual kangaroo rats were caught by nightlighting and live trapping for a total of 952 capture records. Upon capture, each kangaroo rat was ear-tagged and thoroughly examined for parasites and wounds. Third-instar botfly (Cuterebra polita) larvae were observed in kangaroo rats between 16 June and 23 August. Prevalence was 34% based on 454 kangaroo rats sampled during that time, whereas the mean intensity was 2.3 larvae per infested host (n = 156, range = 1-11). In contrast to some other studies of botfly parasitism of rodents, there were no gender or age biases in either prevalence or intensity of infestation. The index of dispersion was 2.8, indicating that the parasites were aggregated in hosts. Botfly parasitism could be an important factor affecting northern populations of kangaroo rats; future investigations into the potential effects of botfly larvae on host fitness are warranted.

    The Remote Sensing Information Gateway was developed by the U.S. Environmental Protection Agency (EPA) to assist researchers in easily obtaining and combining a variety of environmental datasets related to air quality research. Current datasets available include, but are not limited to surface PM2.5 and O3 data, satellite derived aerosol optical depth , and 3-dimensional output from U.S. EPA's Models 3/Community Multi-scale Air Quality (CMAQ) modeling system. The presentation will include a demonstration that illustrates several scenarios of how researchers use the tool to help them visualize and obtain data for their work; with a particular focus on episode analysis related to biomass burning impacts on air quality. The presentation will provide an overview on how RSIG works and how the code has been—and can be—adapted for other projects. One example is the Virtual Estuary, which focuses on automating the retrieval and pre-processing of a variety of data needed for estuarine research. RSIG’s source codes are freely available to researchers with permission from the EPA principal investigator, Dr. Jim Szykman. RSIG is available to the community and can be accessed online at http://www.epa.gov/rsig. Once the JAVA policy file is configured on your computer you can run the RSIG applet on your computer and connect to the RSIG server to visualize and retrieve available data sets. The applet allows the user to specify the temporal/spatial areas of interest, and the types of data to retrieve. The applet then communicates with RSIG subsetter codes located on the data owners’ remote servers; the subsetter codes assemble and transfer via ordinary Internet protocols only the specified data to the researcher’s computer. This is much faster than the usual method of transferring large files via FTP and greatly reduces network traffic. The RfD, which is part of the EPA risk assessment guidelines, is an estimation of a level that is likely to be without any health risk to sensitive individuals.     The overall objective of this project is the continued development, installation, and testing of continuous water sampling and analysis technologies for application to on-site monitoring of groundwater treatment systems and remediation sites. In a previous project, an on-line analytical system (OLAS) for multistream water sampling was installed at the Fort Ord Operable Unit 2 Groundwater Treatment System, with the objective of developing a simplified analytical method for detection of Compounds of Concern at that plant, and continuous sampling of up to twelve locations in the treatment system, from raw influent waters to treated effluent. Earlier implementations of the water sampling and processing system (Analytical Sampling and Analysis Platform, A A+RT, Milpitas, CA) depended on off-line integrators that produced paper plots of chromatograms, and sent summary tables to a host computer for archiving. We developed a basic LabVIEW (National Instruments, Inc., Austin, TX) based gas chromatography control and data acquisition system that was the foundation for further development and integration with the ASAP system. Advantages of this integration include electronic archiving of all raw chromatographic data, and a flexible programming environment to support development of improved ASAP operation and automated reporting. The initial goals of integrating the preexisting LabVIEW chromatography control system with the ASAP, and demonstration of a simplified, site-specific analytical method were successfully achieved. However, although the principal objective of this system was assembly of an analytical system that would allow plant operators an up-to-the-minute view of the plant's performance, several obstacles remained. Data reduction with the base LabVIEW system was limited to peak detection and simple tabular output, patterned after commercial chromatography integrators, with compound retention times and peak areas.     Strain FYK2218(T) was isolated from a specimen of the chiton Acanthopleura japonica, which had been collected from a beach on the Boso peninsula in Japan. Phylogenetic analyses based on 16S rRNA gene sequences revealed that the strain belonged to the phylum 'Acidobacteria'. The most closely related type strains to strain FYK2218(T) were Holophaga foetida TMBS4(T) (83.6 % 16S rRNA gene sequence similarity) and Geothrix fermentans H-5(T) (83.6 %) in subdivision 8 of the 'Acidobacteria'. Cells of FYK2218(T) were motile, rod-shaped, Gram-negative, mesophilic and strictly aerobic. The G+C content of the strain was 56.7 mol%. The strain had isoprenoid quinones MK-6 and MK-7 as major components. Major fatty acids of the strain were iso-C(15 : 0), iso-C(17 : 0), C(16 : 0) and C(20 : 5)omega3c (cis-5,8,11,14,17-eicosapentaenoic acid). From the taxonomic data obtained in this study, it is proposed that the new marine isolate be placed into a novel genus and species named Acanthopleuribacter pedis gen. nov., sp. nov. within the new family, order and class Acanthopleuribacteraceae fam. nov., Acanthopleuribacterales ord. nov. and Holophagae classis nov. The family Holophagaceae fam. nov. is also described. The type strain of Acanthopleuribacter pedis is FYK2218(T) (=NBRC 101209(T) =KCTC 12899(T)).

    This paper summarizes the isolation of arboviruses from mosquitoes collected in the Ord Valley between 1972 and 1976. A total of one hundred and ninety five strains of at least fifteen antigenically distinct viruses have been isolated. Seven of these isolates appear to be "new' antigenic types, and several are undergoing further testing. These are three new rhabdoviruses (Kununurra [OR194], a virus provisionally named Kimberley [OR250] and OR189 [provisionally named Parry's Creek]), three ungrouped, non-haemagglutinating viruses (OR379, OR512, OR869) and a virus (OR540) which reacts to Poly Anopheles A world grouping fluid. The remaining viruses have been previously identified in Australia. These include Murray Valley encephalitis (MVE), Kunjin, Kokobera, Sindbis, Koongol, Wongal, Wongorr and a virus in the Corriparta serological group. The most important finding of these studies is that MVE displays as annually recurrent pattern of activity with a peak seasonal transmission rate at the end of the wet monsoon. This is the first definition of a probable endemic focus of MVE activity in Australia. The major vector for the majority of the viruses isolated was, by inference, Culex annulirostris. However, Aedeomyia catasticta was implicated as a major vector of the Corriparta group virus.

  2. Groundwater interaction with surface drains in the Ord River Irrigation Area, northern Australia: investigation by multiple methods

    Following 35 years of persistent groundwater rise beneath northern Ivanhoe Plain in the Ord River Irrigation Area, northern Australia, the water table appears to have stabilized near the base of the irrigation surface-drain network. Hydrometric evidence indicates that intersection of the deepest surface drains by the rising water table simultaneously reduced aquifer recharge from surface-water infiltration and increased aquifer discharge by groundwater exfiltration. Water-table analysis supports the working hypothesis that the largest irrigation drain D4 on north Ivanhoe Plain has been receiving a significant amount of groundwater discharge since the mid-1990s. The rate of groundwater discharge to surface drains on north Ivanhoe Plain was estimated to be around 15-20 million (M)L/day based on groundwater-flow modelling. Groundwater tracing using radon and electrical conductivity indicated that groundwater discharge to drain D4 was ˜6-12 ML/day in August 2007. The rate of groundwater discharge was significantly larger where the drain traverses a very-permeable sand and gravel palaeochannel. Relatively modest exfiltration rates of order of magnitude tens to hundreds of mm/day into the drain were estimated to mitigate 0.5 m/year groundwater accretion for a land area of order of magnitude hundreds to thousands of ha.

    This report presents a continuation from Oldenburg et al. (2002) of analysis of the hydrogeology, In-Situ Permeable Flow Sensor (ISPFS) results, aquifer response, and changes in the trichloroethylene (TCE) groundwater plume at Operational Unit 1 (OU 1) adjacent to the former Fritzsche Army Airfield at the former Fort Ord Army Base, located on Monterey Bay in northern Monterey County. Fuels and solvents were burned on a portion of OU 1 called the Fire Drill Area (FDA) during airport fire suppression training between 1962 and 1985. This activity resulted in soil and groundwater contamination in the unconfined A-aquifer. In the late 1980's, soil excavation and bioremediation were successful in remediating soil contamination at the site. Shortly thereafter, a groundwater pump, treat, and recharge system commenced operation. This system has been largely successful at remediating groundwater contamination at the head of the groundwater plume. However, a trichloroethylene (TCE) groundwater plume extends approximately 3000 ft (900 m) to the northwest away from the FDA. In the analyses presented here, we augment our prior work (Oldenburg et al., 2002) with new information including treatment-system totalizer data, recent water-level and chemistry data, and data collected from new wells to discern trends in contaminant migration and groundwater flow that may be useful for ongoing remediation efforts. Some conclusions from the prior study have been modified based on these new analyses, and these are pointed out clearly in this report.

    The clinical efficacy of fish oil and high-purity eicosapentaenoic acid ethyl ester (hp-EPA-E) for treating cardiovascular disease (CVD) has been reported. Fish oil contains saturated and monounsaturated fatty acids that have pharmacological effects opposite to those of ω3 fatty acids (ω3). Moreover, ω3, such as EPA and docosahexaenoic acid (DHA), do not necessarily have the same metabolic and biological actions. This has obscured the clinical efficacy of ω3. Recently, the Japan EPA Lipid Intervention Study (JELIS) of hp-EPA-E established the clinical efficacy of EPA for CVD, and higher levels of blood EPA, not DHA, were found to be associated with a lower incidence of major coronary events. A significant reduction in the risk of coronary events was observed when the ratio of EPA to arachidonic acid (AA) (EPA/AA) was > 0.75. Furthermore, the ratio of prostaglandin (PG) I3 and PGI2 to thromboxane A2 (TXA2) ([PGI2 + PGI3]/TXA2) was determined to have a linear relationship with the EPA/AA ratio as follows: (PGI2 + PGI3)/TXA2 =λ + π* (EPA/AA). Like PGI2, PGI3 not only inhibits platelet aggregation and vasoconstriction, but also is assumed to reduce cardiac ischemic injury and arteriosclerosis and promote angiogenesis. Thus, the effects of EPA in reducing the risk of CVD could be mediated by biological action of PGI3 in addition to hypotriglyceridemic action of EPA. Compared with DHA, EPA administration increases the EPA/AA ratio and the (PGI2 + PGI3)/TXA2 balance to a state that inhibits the onset and/or progression of CVD.

    Medical education is moving toward assessment of educational outcomes rather than educational processes. The American Board of Internal Medicine and American Board of Pediatrics milestones and the concept of entrustable professional activities (EPA)--skills essential to the practice of medicine that educators progressively entrust learners to perform--provide new approaches to assessing outcomes. Although some defined EPAs exist for internal medicine and pediatrics, the continued development and implementation of EPAs remains challenging. As residency programs are expected to begin reporting milestone-based performance, however, they will need examples of how to overcome these challenges. The authors describe a model for the development and implementation of an EPA using the resident handoff as an example. The model includes nine steps: selecting the EPA, determining where skills are practiced and assessed, addressing barriers to assessment, determining components of the EPA, determining needed assessment tools, developing new assessments if needed, determining criteria for advancement through entrustment levels, mapping milestones to the EPA, and faculty development. Following implementation, 78% of interns at the University of Minnesota Medical School were observed giving handoffs and provided feedback. The authors suggest that this model of EPA development--which includes engaging stakeholders, an iterative process to describing the behavioral characteristics of each domain at each level of entrustment, and the development of specific assessment tools that support both formative feedback and summative decisions about entrustment--can serve as a model for EPA development for other clinical skills and specialty areas.

    Modeled blood lead concentrations derived from EPA`s Integrated Exposure Uptake Biokinetic Model LEAD99 (Model) were compared to measured blood lead concentrations for two populations of children living in Colorado mining communities. The study areas were Clear Creek and California Gulch. Both studies presented paired blood lead level measurements and environmental media lead concentrations. The data for this paper were obtained from the Colorado Department of Health and publicly-available submissions made to EPA Region VIII. EPA has used the Model to identify soil lead concentrations that are protective of individual children.

This project activity, which was an essential part of the program to enhance the collaborative ongoing research between the NSHE and ORD, was intended to support all quality-affecting activities funded during the five-year period of the cooperative agreement. However, the cooperative agreement was down-graded to non quality-affecting after 4 years.

    Recent studies have shown that EPA enriched PLs have beneficial effects on lipid metabolism. Our previous study has demonstrated that the anti-obesity and hypolipidemic effects of EPA-PL were superior to DHA-PL. In the present study, we comparatively evaluated the effects of EPA-enriched phosphatidylcholine (EPA-PC) and EPA-enriched phosphatidylserine (EPA-PS) on lipid metabolism in mice. Both 2% dietary EPA-PC and EPA-PS significantly improved serum and hepatic lipid levels in mice. The HDL-c level in mice on EPA-PC diet was significantly higher than the other two groups. The level of DHA in hepatic TG and PL were significantly increased in both EPA-PC and EPA-PS fed groups (98.3 and 117.8%, respectively; p < 0.05). Notably, the proportion of DHA in EPA-PS group was significantly higher than the EPA-PC group. EPA-PC and EPA-PS suppressed hepatic SREBP-1c mediated lipogenesis and activated PPARα mediated fatty acid β-oxidation in the liver. These data are the first to indicate that EPA-PS has beneficial effects on lipid metabolism.

It provides the foundation for what we know about our planet, how it has changed, and how it could be altered in the future. The National Center for Environmental Research (NCER) in the U.S. Environmental Protection Agency's (EPA) Office of Research and Development (ORD) supports high-quality, extramural research by the nation's leading scientists and engineers to strengthen the basis for decisions about local and national environmental issues. NCER works with academia, state and local governments, other federal agencies, and scientists in EPA to increase human knowledge of how to protect our health and natural resources through its three major programs: · Science to Achieve Results (STAR) Grants · Small Business Innovative Research (SBIR) · Science to Achieve Results (STAR) Fellowships STAR, NCER's primary program, funds research grants and graduate fellowships in environmental science and engineering. Developing the next generation of environmental scientists and engineers is one of NCER's most important objectives. Each year, NCER helps between 80 and 160 students achieve Master's or Ph.D. degrees in environmental science and engineering through its STAR and Greater Research Opportunities (GRO) fellowships. Some of these students have moved on to careers in government while others are now full-time professors and researchers. Still others are working for state environmental agencies or furthering their studies through postdoctoral positions at universities. Since the inception of the NCER program, STAR fellowships (along with grants and SBIR projects) have been awarded in every state in the country. With the help of STAR, current and future scientists and engineers have been able to explore ways to preserve and protect human health and our precious resources.

    As a result of on-going internal and external assessment of the Acid Rain Program monitoring and reporting requirements, EPA, state environmental agencies, and utilities have identified areas of the Part 75 CEM regulations which would benefit from revision or clarification. Many of the suggested revisions will add increased flexibility to the utility industry in implementing and complying with the requirements of Part 75. Other revisions will clarify existing provisions in an effort to make the regulation more understandable. Still other revisions will provide increased quality assurance of the Acid Rain Program CEM data. The panel will present EPA`s current plans for a proposed rulemaking to incorporate the technical revisions to Part 75. The panel will briefly discuss EPA`s proposed revisions to some of the major issues and will respond to questions.

    As stated at the beginning of this paper conclusions reached thus far cannot be discussed in this paper. However, a great deal of information is available for examination.EPA displayed its ability to coordinate widely separated laboratories, both Federal and private, into a smooth working team in a very short period of time. A very comprehensive study plan was also developed and implemented quickly. EPA was fortunate to have already had GCA under contract when the emergency arose. In no small part the success of the field effort was due to the managerial and technical abilities of the GCA team.Within a period of 6 weeks a plan was developed, a prime contractor retained, subcontractors hired, and field activities begun. Within a period of 3 months in excess of 8600 field samples were collected and over 12,000 field and QC samples were analyzed. During this same period 2 major data systems were developed, debugged, and placed into operation.In short this EPA project was probably the most comprehensive multimedia field project ever attempted by EPA and certainly the data is being subjected to the most strenuous quality control measures ever imposed by this Agency. The entire program is presently under peer review and the results are being prepared for publication by EPA Headquarters.

    Organochlorine pesticide concentrations, particularly those of the DDT family and of toxaphene, were measured by gas chromatography in samples of liver and body fat taken from Australian freshwater crocodiles Crocodylus johnstoni at three locations along the Ord River in Western Australia. The three sampling sites were the irrigation area, downstream of the irrigation area, and well upstream of the irrigation area; the last site serving as the control. DDT and toxaphene were applied in large and known quantities to cotton grown in the Ord Irrigation Area from 1964 to 1974. Thus the residues in the crocodile tissues are representative of the situation almost thirty years after the use of DDT and toxaphene ceased in the area. Very high concentrations of p,p'-DDE and toxaphene were found in the lipid-rich tissues that were examined. Livers and body fat from estuarine crocodiles Crocodylus porosus from the downstream site were also analysed. As p,p'-DDE and toxaphene are both known to be disruptive of endocrine systems, a range of blood parameters, including estradiol and testesterone concentrations, were also measured for all the animals studied. The ovaries and testes of the freshwater crocodiles were also examined histologically. There were no obvious effects on blood chemistry or gonad histology of the large burden of pesticides and their metabolites carried by exposed animals, although the limited number of samples and the variability of the breeding state of the animals examined may have masked possible effects. The isolation of the area, the accurately known applications of DDT and toxaphene, and the simplicity of the drainage system make the lower Ord River a unique natural laboratory for studying the long term breakdown and effects of pesticides applied in a tropical environment.

However, the agency's scientific practices and performance at times have been criticized by the U.S. General Accounting Office, the National Academies of Science (NAS), Congress, and EPA's own science advisory board, as well as in a number of lawsuits.New legislation introduced by Rep. Vernon Ehlers (R-Mich.), chair of the House of Representatives' Science Subcommittee on Environment, Technology, and Standards, includes measures meant to improve the agency's science component. The legislation, H.R. 64, would require the president to appoint an EPA deputy administrator for science and technology This deputy administrator, who would rank higher than existing assistant administrators (AAs), would be responsible for the overall scientific and technical foundation of the agency's decisions, including ensuring that the agency's scientific endeavors use the best possible peer review and research planning practices.

    An important goal of toxicology research is the development of robust methods that use in vitro and chemical structure information to predict in vivo toxicity endpoints. The US EPA ToxCast program is addressing this goal using ~600 in vitro assays to create bioactivity profiles on a set of 320 compounds, mostly pesticide actives, that have well characterized in vivo toxicity. These 320 compounds (EPA-320 set evaluated in Phase I of ToxCast) are a subset of a much larger set of ~10,000 candidates that are of interest to the EPA (called here EPA-10K). Predictive models of in vivo toxicity are being constructed from the in vitro assay data on the EPA-320 chemical set. These models require validation on additional chemicals prior to wide acceptance, and this will be carried out by evaluating compounds from EPA-10K in Phase II of ToxCast. We have used cheminformatics approaches including clustering, data visualization, and QSAR to develop models for EPA-320 that could help prioritizing EPA-10K validation chemicals. Both chemical descriptors, as well as calculated physicochemical properties have been used. Compounds from EPA-10K are prioritized based on their similarity to EPA-320 using different similarity metrics, with similarity thresholds defining the domain of applicability for the predictive models built for EPA-320 set.     A morphometric analysis of river network, basins and relief using geomorphic indices and geostatistical analyses of Digital Elevation Model (DEM) are useful tools for discussing the morphometric evolution of the basin area. In this study, three different indices including valley floor width to height ratio (Vf), stream gradient (SL), and stream sinuosity were applied to Afşin-Elbistan lignite basin to test the imprints of tectonic activity. Perturbations of these indices are usually indicative of differences in the resistance of outcropping lithological units to erosion and active faulting. To map the clusters of high and low indices values, the Kernel density estimation (K) and the Getis-Ord Gi∗ statistics were applied to the DEM-derived indices. The K method and Gi∗ statistic highlighting hot spots and cold spots of the SL index, the stream sinuosity and the Vf index values helped to identify the relative tectonic activity of the basin area. The results indicated that the estimation by the K and Gi∗ including three conceptualization of spatial relationships (CSR) for hot spots (percent volume contours 50 and 95 categorized as high and low respectively) yielded almost similar results in regions of high tectonic activity and low tectonic activity. According to the K and Getis-Ord Gi∗ statistics, the northern, northwestern and southern parts of the basin indicates a high tectonic activity. On the other hand, low elevation plain in the central part of the basin area shows a relatively low tectonic activity.
