National dataset consisting of a snapshot of 303(d) Listed Impaired Waters combined with Impaired Waters with TMDLs whose parent cause is Nutrients, Sediments, and Pathogens. The snapshot constitutes the top water impairments. The Impaired Waters with TMDLs and the 303(d) Listed Impaired Waters are combined into three mapping service layers in MyWaters Mapper application to provide a comprehensive view of each of these impairments. These waters are linked to the 303(d) information stored in EPAs Assessment and TMDL Tracking and Implementation System (ATTAINS). Below is a brief description of the two programs.The 303(d) Listed Impaired Waters program system provides impaired water data and impaired water features reflecting river segments, lakes, and estuaries designated under Section 303(d) of the Clean Water Act. The Federal Clean Water Act (Section 303(d)) mandates that states develop Total Maximum Daily Load (TMDL) plans for water bodies that are on the Section 303(d) list. To be placed on the 303(d) list, a water body must be found to have water quality conditions that limit its ability to meet its designated beneficial uses. The TMDL for a water body is defined in 40 CFR 130 as the sum of waste load allocations from identified points sources and non-point sources within the water body's watershed. The TMDL plan for a listed water body should identify the current waste loads to the water body, the waste load capacity of the water body and then allocate the waste load capacity to the known point and non-point sources of pollution within the water body's watershed. The United States's Clean Water Act stipulates in section 303(d) that states must identify impaired water bodies for which total maximum daily loads (TMDLs) of pollution inputs into water bodies are developed. Decision-making procedures about how to list, or delist, water bodies as impaired, or not, per Clean Water Act 303(d) differ across states. In states such as California, whether or not a particular monitoring sample suggests that water quality is impaired can be regarded as a binary outcome variable, and California's current regulatory framework invokes a version of the exact binomial test to consolidate evidence across samples and assess whether the overall water body complies with the Clean Water Act. Here, we contrast the performance of California's exact binomial test with one potential alternative, the Sequential Probability Ratio Test (SPRT). The SPRT uses a sequential testing framework, testing samples as they become available and evaluating evidence as it emerges, rather than measuring all the samples and calculating a test statistic at the end of the data collection process. Through simulations and theoretical derivations, we demonstrate that the SPRT on average requires fewer samples to be measured to have comparable Type I and Type II error rates as the current fixed-sample binomial test. Policymakers might consider efficient alternatives such as SPRT to current procedure. Copyright © 2017 Elsevier Ltd. All rights reserved.

The Total Maximum Daily Load (TMDL) Tracking System contains information on waters that are Not Supporting their designated uses. These waters are listed by the state as impaired under Section 303(d) of the Clean Water Act. The status of TMDLs are also tracked. TMDLs are pollution control measures that reduce the discharge of pollutants into impaired waters. A TMDL or Total Maximum Daily Load is a calculation of the maximum amount of a pollutant that a waterbody can receive and still meet water quality standards, and an allocation of that amount to the pollutant's sources. What is a total maximum daily load (TMDL)? Water quality standards are set by States, Territories, and Tribes. They identify the uses for each waterbody, for example, drinking water supply, contact recreation (swimming), and aquatic life support (fishing), and the scientific criteria to support that use. A TMDL is the sum of the allowable loads of a single pollutant from all contributing point and nonpoint sources. The calculation must include a margin of safety to ensure that the waterbody can be used for the purposes the state has designated. The calculation must also account for seasonal variation in water quality. The Clean Water Act, section 303, establishes the water quality standards and TMDL programs.

The Mississippi Department of Environmental Quality is required to develop restoration and remediation plans for water bodies not meeting their designated uses, as stated in the U.S. Environmental Protection Agency's Clean Water Act section 303(d). The majority of streams in northwestern Mississippi are on the 303(d) list of water-quality limited waters. Agricultural effects on streams in northwestern Mississippi have reduced the number of unimpaired streams (reference streams) for water-quality comparisons. As part of an effort to develop an index to assess impairment, the U.S. Geological Survey collected water samples from 52 stream sites on the 303(d) list during May-June 2006, and analyzed the samples for nutrients and chlorophyll. The data were analyzed by trophic group as determined by total nitrogen concentrations. Seven constituents (nitrite plus nitrate, total Kjeldhal nitrogen, total phosphorus, orthophosphorus, total organic carbon, chlorophyll a, and pheophytina) and four physical property measurements (specific conductance, pH, turbidity, and dissolved oxygen) were determined to be significantly different (p < 0.05) between trophic groups. Total Kjeldhal nitrogen, turbidity, and dissolved oxygen were used as indicators of stream productivity with which to infer stream health. Streams having high total Kjeldhal nitrogen values and high turbidity values along with low dissolved oxygen concentrations were typically eutrophic abundant in nutrients), whereas streams having low total Kjeldhal nitrogen values and low turbidity values along with high dissolved oxygen concentrations were typically oligotrophic (deficient in nutrients).

In 1998, a 5 river-mile reach of the Jacks Fork was included on Missouri's list of impaired waters as required by Section 303(d) of the Federal Clean Water Act. The identified pollutant on the Jacks Fork was fecal coliform bacteria. The length of the impaired reach was changed to 7 miles on the Missouri 2002 303(d) list because of data indicating the fecal coliform bacteria problem existed over a broader area. The U.S. Geological Survey, in cooperation with the National Park Service, conducted a study to better understand the extent and sources of microbiological contamination within the Jacks Fork from Alley Spring to the mouth, which includes the 7-mile 303(d) reach. Ten sites were sampled from June 2003 through October 2003 and from June 2004 through October 2004. Water-column and streambed sediment samples were collected from main-stem and tributary sites mostly during base-flow conditions during a variety of recreational season river uses and analyzed for fecal coliform and Escherichia coli bacteria. Isolates of Escherichia coli obtained from water samples collected at five sites were submitted for rep-PCR analysis to identify presumptive sources of fecal indicator bacteria in the Jacks Fork. Results indicate that recreational users (including boaters and swimmers) are not the primary source of fecal coliform bacteria in the Jacks Fork; rather, the presence of fecal coliform bacteria is associated with other animals, of which horses are the primary source. Increases in fecal coliform bacteria densities in the Jacks Fork are associated with cross-country horseback trail-riding events.

The homologous glycosyltransferases α-1,3-N-acetylgalactosaminyltransferase (GTA) and α-1,3-galactosyltransferase (GTB) carry out the final synthetic step of the closely related human ABO(H) blood group A and B antigens. The catalytic mechanism of these model retaining enzymes remains under debate, where Glu303 has been suggested to act as a putative nucleophile in a double displacement mechanism, a local dipole stabilizing the intermediate in an orthogonal associative mechanism or a general base to stabilize the reactive oxocarbenium ion-like intermediate in an SNi-like mechanism. Kinetic analysis of GTA and GTB point mutants E303C, E303D, E303Q and E303A shows that despite the enzymes having nearly identical sequences, the corresponding mutants of GTA/GTB have up to a 13-fold difference in their residual activities relative to wild type. High-resolution single crystal X-ray diffraction studies reveal, surprisingly, that the mutated Cys, Asp and Gln functional groups are no more than 0.8 Å further from the anomeric carbon of donor substrate compared to wild type. However, complicating the analysis is the observation that Glu303 itself plays a critical role in maintaining the stability of a strained "double-turn" in the active site through several hydrogen bonds, and any mutation other than E303Q leads to significantly higher thermal motion or even disorder in the substrate recognition pockets. Thus, there is a remarkable juxtaposition of the mutants E303C and E303D, which retain significant activity despite disrupted active site architecture, with GTB/E303Q, which maintains active site architecture but exhibits zero activity. These findings indicate that nucleophilicity at position 303 is more catalytically valuable than active site stability and highlight the mechanistic elasticity of these enzymes. © The Author 2016. Watershed-based sampling design and assessment tools help serve the multiple goals for water quality monitoring required under the Clean Water Act, including assessment of regional conditions to meet Section 305(b), identification of impaired water bodies or watersheds to meet Section 303(d), and development of empirical relationships between causes or sources of impairment and biological responses. Creation of GIS databases for hydrography, hydrologically corrected digital elevation models, and hydrologic derivatives such as watershed boundaries and upstream–downstream topology of subcatchments would provide a consistent seamless nationwide framework for these designs. The elements of a watershed-based sample framework can be represented either as a continuous infinite set defined by points along a linear stream network, or as a discrete set of watershed polygons. Watershed-based designs can be developed with existing probabilistic survey methods, including the use of unequal probability weighting, stratification, and two-stage frames for sampling. Case studies for monitoring of Atlantic Coastal Plain streams, West Virginia wadeable streams, and coastal Oregon streams illustrate three different approaches for selecting sites for watershed-based survey designs.

Section 303(d) of the Clean Water Act requires States and Tribes to list waters not meeting water quality standards. A total maximum daily load must be prepared for waters identified as impaired with respect to water quality standards. Historically, the management of pollution in Minnesota has been focused on point-source regulation. Regulatory effort in Minnesota has improved water quality over the last three decades. Non-point source pollution has become the largest driver of conventional 303(d) listings in the 21st century. Conventional pollutants, i.e., organic, sediment and nutrient imbalances can be identified with poor land use management practices. However, the cause and effect relationship can be elusive because of natural watershed-system influences that vary with scale. Elucidation is complex because the current water quality standards in Minnesota were designed to work best with water quality permits to control point sources of pollution. This paper presents a sentinel watershed-systems approach (SWSA) to the monitoring and assessment of Minnesota waterbodies. SWSA integrates physical, chemical, and biological data over space and time using advanced technologies at selected small watersheds across Minnesota to potentially improve understanding of natural and anthropogenic watershed processes and the management of point and non-point sources of pollution. Long-term, state-of-the-art monitoring and assessment is needed to advance and improve water quality standards. Advanced water quality or ecologically-based standards that integrate physical, chemical, and biological numeric criteria offer the potential to better understand, manage, protect, and restore Minnesota's waterbodies.
