This report, Energy Policy Act Transportation Study: Interim Report on Natural Gas Flows and Rates, is the second in a series mandated by Title XIII, Section 1340, ``Establishment of Data Base and Study of Transportation Rates,`` of the Energy Policy Act of 1992 (P.L. 102--486). The first report Energy Policy Act Transportation Study: Availability of Data and Studies, was submitted to Congress in October 1993; it summarized data and studies that could be used to address the impact of legislative and regulatory actions on natural gas transportation rates and flow patterns. The current report presents an interim analysis of naturalmore » gas transportation rates and distribution patterns for the period from 1988 through 1994. A third and final report addressing the transportation rates and flows through 1997 is due to Congress in October 2000. This analysis relies on currently available data; no new data collection effort was undertaken. The need for the collection of additional data on transportation rates will be further addressed after this report, in consultation with the Congress, industry representatives, and in other public forums.« less

To develop a theoretical, functional model of community navigation for individuals with cognitive impairments: the Activities of Community Transportation (ACTs). Iterative design using qualitative methods (i.e. document review, focus groups and observations). Four agencies providing travel training to adults with cognitive impairments in the USA participated in the validation study. A thorough document review and series of focus groups led to the development of a comprehensive model (ACTs Wheels) delineating the requisite steps and skills for community navigation. The model was validated and updated based on observations of 395 actual trips by travellers with navigational challenges from the four participating agencies. Results revealed that the 'ACTs Wheel' models were complete and comprehensive. The 'ACTs Wheels' represent a comprehensive model of the steps needed to navigate to destinations using paratransit and fixed-route public transportation systems for travellers with cognitive impairments. Suggestions are made for future investigations of community transportation for this population.

The current status of the Active Controls Technology (ACT) for the advanced subsonic transport project is investigated through analysis of the systems technical data. Control systems technologies under examination include computerized reliability analysis, pitch axis fly by wire actuator, flaperon actuation system design trade study, control law synthesis and analysis, flutter mode control and gust load alleviation analysis, and implementation of alternative ACT systems. In the aftermath of disasters, planners and policymakers have to utilize scarce resources and work within legal frameworks to provide recovery for affected citizens and business. As seen with Hurricane Sandy, various observers noted the challenge with freight rerouting due to inoperable infrastructure and legal barriers involved with intermodal freight transportation. This paper focuses on how Geographic Information Systems (GIS); specifically WebTRAGIS (Transportation Routing Analysis GIS), a transportation routing platform developed at Oak Ridge National Laboratory, can be used to evaluate different routing options formore » freight transportation. In the case of freight transportation, the Merchant Marine Act of 1920 (also known as the Jones Act) restricts Short Sea Shipping (SSS) between coastwise points within U.S. territorial waters. This restriction leads to reliance on land-side modes for handling increased freight resulting from cargo diversion. Using Hurricane Sandy and the closure of the Port of New York/New Jersey as a case study, different modal studies were conducted; ultimately highlighting the various routes and provides insight into potential review of modal restrictions such as the Jones Act.« less

The performance and economic benefits of a constrained application of Active Controls Technology (ACT) are identified, and the approach to airplane design is established for subsequent steps leading to the development of a less constrained final ACT configuration. The active controls configurations are measured against a conventional baseline configuration, a state-of-the-art transport, to determine whether the performance and economic changes resulting from ACT merit proceeding with the project. The technology established by the conventional baseline configuration was held constant except for the addition of ACT. The wing, with the same planform, was moved forward on the initial ACT configuration to move the loading range aft relative to the wing mean aerodynamic chord. Wing trailing-edge surfaces and surface controls also were reconfigured for load alleviation and structural stabilization.

In 2006, the Agricultural Chemicals: Sources, Transport and Fate study team (Agricultural Chemicals Team, ACT) of the U.S. Geological Survey National Water-Quality Assessment Program began a study in northwestern Mississippi to evaluate the influence of surface-water recharge on the occurrence of agriculturally related nutrients and pesticides in the Mississippi River Valley alluvial aquifer. The ACT study was composed in the Bogue Phalia Basin, an indicator watershed within the National Water-Quality Assessment Program Mississippi Embayment Study Unit and utilized several small, subbasins within the Bogue Phalia to evaluate surface and groundwater interaction and chemical transport in the Basin. Data collected as part of this ACT study include water-quality data from routine and incident-driven water samples evaluated for major ions, nutrients, organic carbon, physical properties, and commonly used pesticides in the area; discharge, gage height and water-level data for surface-water sites, the shallow alluvial aquifer, and hyporheic zone; additionally, agricultural data and detailed management activities were reported by land managers for farms within two subbasins of the Bogue Phalia Basin—Tommie Bayou at Pace, MS, and an unnamed tributary to Clear Creek near Napanee, MS.

Neurological diseases caused by encephalitic flaviviruses are severe and associated with high levels of mortality. However, little is known about the detailed mechanisms of viral replication and pathogenicity in the brain. Previously, we reported that the genomic RNA of tick-borne encephalitis virus (TBEV), a member of the genus Flavivirus , is transported and replicated in the dendrites of neurons. In the present study, we analyzed the transport mechanism of the viral genome to dendrites. We identified specific sequences of the 5' untranslated region of TBEV genomic RNA that act as a cis -acting element for RNA transport. Mutated TBEV with impaired RNA transport in dendrites caused a reduction in neurological symptoms in infected mice. We show that neuronal granules, which regulate the transport and local translation of dendritic mRNAs, are involved in TBEV genomic RNA transport. TBEV genomic RNA bound an RNA-binding protein of neuronal granules and disturbed the transport of dendritic mRNAs. These results demonstrated a neuropathogenic virus hijacking the neuronal granule system for the transport of viral genomic RNA in dendrites, resulting in severe neurological disease.

Localization of mRNAs can involve multiple steps, each with its own cis -acting localization signals and transport factors. How is the transition between different steps orchestrated? We show that the initial step in localization of Drosophila oskar mRNA - transport from nurse cells to the oocyte - relies on multiple cis -acting signals. Some of these are binding sites for the translational control factor Bruno, suggesting that Bruno plays an additional role in mRNA transport. Although transport of oskar mRNA is essential and robust, the localization activity of individual transport signals is weak. Notably, increasing the strength of individual transport signals, or adding a strong transport signal, disrupts the later stages of oskar mRNA localization. We propose that the oskar transport signals are weak by necessity; their weakness facilitates transfer of the oskar mRNA from the oocyte transport machinery to the machinery for posterior localization. © 2017. Brain microvessel endothelial cells (BMEC) which form the blood-brain barrier (BBB) possess an amino acid carrier specific for large neutral amino acids (LNAA). The carrier is important for facilitating the delivery of nutrient LNAA's and centrally acting drugs that are LNAA's, to the brain. Bovine BMEC's were isolated and grown up to complete monolayers on regenerated cellulose-membranes in primary culture. To study the transendothelial transport of leucine, the monolayers were placed in a side-by-side diffusion cell, and transport across the monolayers followed with (/sup 3/H)-leucine. The transendothelial transport of leucine in this in vitro model was determined to be bidirectional,more » and time-, temperature-, and concentration-dependent. The transport of leucine was saturable and the apparent K/sub m/ and V/sub max/, 0.18 mM and 6.3 nmol/mg/min, respectively. Other LNAA's, including the centrally acting drugs, ..cap alpha..-methyldopa, L-DOPA, ..cap alpha..-methyl-tyrosine, and baclofen, inhibited leucine transport. The leucine carrier was also found to be stereospecific and not sensitive to inhibitors of active transport. These results are consistent with previous in vitro and in vivo studies. Primary cultures of BMEC's appear to be a potentially important tool for investigating at the cellular level, the transport mechanisms of the BBB.« less

Forests are one of the dominant vegetation types on Earth and are an important sink for carbon on our planet. Forests are special ecosystems due to their great canopy height und complex architecture consisting of a subcanopy and a canopy layer, which changes the mechanisms of turbulent exchange within the plant canopy. To date, the sinks and sources of turbulence in forest canopies are not completely understood, especially the role of the pressure transport remains unclear. The INTRAMIX experiment was conducted in a mountainous Norway spruce (Picea abies) forest at the Fluxnet Waldstein site (DE-Bay) in Bavaria, Germany, for a period of 10 weeks in order to experimentally evaluate the significance of the pressure transport to the TKE budget for the first time. The INTRAMIX data of the dense mountain forest was compared to observations from a sparse Ponderosa pine (Pinus ponderosa) stand in Oregon, USA, to study the influence of forest architecture. We hypothesized that the pressure transport is more important in dense forest canopies as the crown decouples the subcanopy from the buoyancy- and shear-driven flow above the canopy. It is also investigated how atmospheric stability influences the TKE budget. Based upon model results from literature we expect the pressure transport to act as a source for TKE especially under free convective and unstable dynamic stability. Results to date indicate that pressure transport is most important in the subcanopy with decreasing magnitude with increasing height. Nevertheless, pressure transport is a continuous source of TKE above the canopy, while in the canopy and subcanopy layer pressure transport acts both as a sink and source term for TKE. In the tree crown layer pressure transport is a source in the morning and afternoon hours and acts as a sink during the evening, while in the subcanopy pressure transport is a source around noon and during the night and acts as a sink in the early morning and afternoon hours. This complementary pattern suggests that the pressure transport is an important means for exchanging TKE across canopy layers.

The Indian government enacted 'The cigarettes and other tobacco products act, 2003' (COTPA), which prohibits smoking in public places. To validate the efficacy of the Act of 2003, enacted by the Government of India, to prevent secondhand smoking in public places. The study is based on a non-random sample survey of 2,600 bus passengers carried out in the premises of three mega public road transport organizations in Karnataka state, India, in June 2007. The information was gathered through administration of structured schedules. A sample of 1,000 each for the terminus of Bangalore Metropolitan Transport Corporation (BMTC) and Karnataka State Road Transport Corporation (KSRTC) in Bangalore and, 600 for North West Karnataka Road Transport Corporation (NWKRTC) in Hubli-Dharwad city was distributed proportionately according to the number of platforms in each terminus. Simple Averages. There is some reduction in smoking in general as perceived by 69% of the passengers as compared to the scenario a year before the enactment of COTPA. The observed smoking is lower in the bus premises of BMTC where there is strict regulation, and higher in the bus premises of NWKRTC, which has not taken any regulatory measures. Knowing smoking is banned in public places can itself create awareness depending on the coverage extended by media and implementing an agency to reach the public. The implementation of an act depends on the willingness of stakeholders to act upon it. The implementation of COTPA as done by BMTC could well become a role model for replication elsewhere, if BMTC can strive harder to accomplish a 100% smoke-free zone.

Inhaled medications prescribed for the hypersensitive airway typically combine corticosteroids and long-acting beta2 adrenergic agonists (LABAs). The phonatory side effects of these combination treatments are widely recognized. However, there is limited understanding of the physiological changes induced by these medications that underlie the phonatory side effects. The objective of this study was to investigate the distinct effects of corticosteroids and LABAs on vocal fold mucosal physiology. Understanding the physiological changes to the vocal folds after corticosteroid and LABA treatments is necessary to prevent the prevalent vocal decrement associated with these medications. Experimental in vitro design with treatment and control groups. Native porcine vocal fold mucosae (N = 38) were exposed to corticosteroid or LABA treatments. Ion transport was measured continuously at baseline and after treatment. To quantify the nature of ion transport, vocal folds were also treated with chloride and sodium channel inhibitors. Corticosteroid treatment did not alter ion transport. Conversely, exposure to LABAs significantly increased ion transport. This increase in ion transport was transient, observed immediately after treatment in all tissue and associated with increased chloride secretion. The distinct effects of corticosteroids and LABAs on vocal fold physiology have not been examined to date. This study demonstrates that short-term treatment with LABAs, but not corticosteroids, significantly increases ion transport. These findings suggest that one underlying physiological mechanism for phonatory changes associated with inhaled treatments may be related to acute alterations in vocal fold ion transport and surface hydration.

Fuel conservation that could be attained with two technology advancements, Q fan propulsion system and active control technology (ACT) was studied. Aircraft incorporating each technology were sized for a Federal Aviation Regulation (FAR) field length of 914 meters (3,000 feet), 148 passengers, and a 926 kilometer (500 nautical mile) mission. The cruise Mach number was .70 at 10100 meter (33,000 foot) altitude. The improvement resulting from application of the Q fan propulsion system was computed relative to an optimized fuel conservative transport design. The performance improvements resulting from application of ACT technology were relative to the optimized Q fan propulsion system configuration.

This report summarizes the Wing Planform Study Task and Final Configuration Selection of the Integrated Application of Active Controls (IAAC) Technology Project within the Energy Efficient Transport Program. Application of Active Controls Technology (ACT) in combination with increased wing span resulted in significant improvements over the Conventional Baseline Configuration (Baseline) and the Initial ACT Configuration previously established. The configurations use the same levels of technology (except for ACT), takeoff gross weight, and payload as the Baseline. The Final ACT Configuration (Model 768-107) incorporates pitch-augmented stability (which enabled an approximately 10% aft shift in cruise center of gravity and a 45% reduction in horizontal tail sizes), lateral/directional-augmented stability, an angle-of-attack limiter, and wing-load alleviation. Flutter-mode control was not beneficial for this configuration. This resulted in an 890 kg (1960 lb) reduction in airplane takeoff gross weight and a 9.8% improvement in cruise lift/drag. At the Baseline mission range (3590 km) (1938 nmi), this amounts to 10% block fuel reduction. Good takeoff performance at high-altitude airports on a hot day was also achieved. Results of this task strongly indicate that the IAAC Project should proceed with the Final ACT evaluation and begin the required control system development and testing.

Pharmaceutical and personal care products (PPCPs) and artificial sweeteners (ASs) are emerging organic contaminants (EOCs) in the aquatic environment. The presence of PPCPs and ASs in water bodies has an ecologic potential risk and health concern. Therefore, it is needed to detect the pollution sources by understanding the transport behavior of sewage molecular markers in a subsurface area. The aim of this study was to evaluate transport of nine selected molecular markers through saturated soil column experiments. The selected sewage molecular markers in this study were six PPCPs including acetaminophen (ACT), carbamazepine (CBZ), caffeine (CF), crotamiton (CTMT), diethyltoluamide (DEET), salicylic acid (SA) and three ASs including acesulfame (ACF), cyclamate (CYC), and saccharine (SAC). Results confirmed that ACF, CBZ, CTMT, CYC and SAC were suitable to be used as sewage molecular markers since they were almost stable against sorption and biodegradation process during soil column experiments. In contrast, transport of ACT, CF and DEET were limited by both sorption and biodegradation processes and 100% removal efficiency was achieved in the biotic column. Moreover, in this study the effect of different acetate concentration (0-100mg/L) as an easily biodegradable primary substrate on a removal of PPCPs and ASs was also studied. Results showed a negative correlation (r(2)>0.75) between the removal of some selected sewage chemical markers including ACF, CF, ACT, CYC, SAC and acetate concentration. CTMT also decreased with the addition of acetate, but increasing acetate concentration did not affect on its removal. CBZ and DEET removal were not dependent on the presence of acetate. The regulation of gene expression involves a multifarious regulatory system. Each gene contains a unique combination of cis-acting regulatory sequence elements in the 5' regulatory region that determines its temporal and spatial expression. Cis-acting regulatory elements are essential transcriptional gene regulatory units; they control many biological processes and stress responses. Thus a full understanding of the transcriptional gene regulation system will depend on successful functional analyses of cis-acting elements. Cis-acting regulatory elements present within the 5' regulatory region of the sucrose transporter gene families in rice (Oryza sativa Japonica cultivar-group) and Arabidopsis thaliana, were identified using a bioinformatics approach. The possible cis-acting regulatory elements were predicted by scanning 1.5kbp of 5' regulatory regions of the sucrose transporter genes translational start sites, using Plant CARE, PLACE and Genomatix Matinspector professional databases. Several cis-acting regulatory elements that are associated with plant development, plant hormonal regulation and stress response were identified, and were present in varying frequencies within the 1.5kbp of 5' regulatory region, among which are; A-box, RY, CAT, Pyrimidine-box, Sucrose-box, ABRE, ARF, ERE, GARE, Me-JA, ARE, DRE, GA-motif, GATA, GT-1, MYC, MYB, W-box, and I-box. This result reveals the probable cis-acting regulatory elements that possibly are involved in the expression and regulation of sucrose transporter gene families in rice and Arabidopsis thaliana during cellular development or environmental stress conditions. Acting as a new type of energy transportation mode, the coal pipeline hydraulic transmission can reduce the energy transportation cost and the fly ash pollution of the conventional coal transportation. In this study, the effect of average velocity, particle size and pumping time on particle composition of coal particles during hydraulic conveying was investigated by ring tube test. Meanwhile, the effects of particle composition change on slurry viscosity, transmission resistance and critical sedimentation velocity were studied based on the experimental data. The experimental and theoretical analysis indicate that the alter of slurry particle composition can lead to the change of viscosity, resistance and critical velocity of slurry. Moreover, based on the previous studies, the critical velocity calculation model of coal slurry is proposed.

Transport of a fullerene-like nanoparticle across a lipid bilayer is investigated by coarse-grained molecular dynamics (MD) simulations. Potentials of mean force (PMF) acting on the nanoparticle in a flexible bilayer suspended in water and a bilayer restrained to a flat surface are computed by constrained MD simulations. The rate of the nanoparticle transport into the bilayer interior is predicted using one-dimensional Langevin models based on these PMFs. The predictions are compared with the transport rates obtained from a series of direct (unconstrained) MD simulations of the solute transport into the flexible bilayer. It is observed that the PMF acting on the solute in the flexible membrane underestimates the transport rate by more than an order of magnitude while the PMF acting on the solute in the restrained membrane yields an accurate estimate of the activation energy for transport into the flexible membrane. This paradox is explained by a coexistence of metastable membrane configurations for a range of the solute positions inside and near the flexible membrane. This leads to a significant reduction of the contribution of the transition state to the mean force acting on the solute. Restraining the membrane shape ensures that there is only one stable membrane configuration corresponding to each solute position and thus the transition state is adequately represented in the PMF. This mechanism is quite general and thus this phenomenon is expected to occur in a wide range of interfacial systems. A simple model for the free energy landscape of the coupled solute-membrane system is proposed and validated. This model explicitly accounts for effects of the membrane deformations on the solute transport and yields an accurate prediction of the activation energy for the solute transport.

Nanotechnology permits broad advances in agriculture. However, as it is still at a relatively early stage of development, the potential risks remain unclear. Herein, for the first time, we reveal the following: 1) the impact of multi-walled carbon nanotubes (MWCNTs) on the accumulation/depuration behaviors of contaminants in crop, mustard (Brassica juncea), and 2) the permeability and transportability of MWCNTs in intact mature mustard plants. Using an in vivo sampling technique, the kinetic accumulation/depuration processes of several contaminants in mustard plans exposed to MWCNTs were traced, and an enhancement of contaminant accumulation in living plants was observed. Meanwhile, we observed that the MWCNTs permeated into the roots of intact living plants (three months old) and were then transported to the upper organs under the force of transpiration steam. This study demonstrated that MWCNTs can act as contaminant carriers and be transported to the edible parts of crops.

Four compounds were isolated from roots and aerial parts of Pluchea sagittalis (Asteraceae), 3, 5-dihydroxy-6, 7, 3', 4'-tetramethoxiflavunol (1), 5-hydroxymethylfurfural (2), 3, 4-dimethoxybenzaldehyde (3) and 2, 3, 4-trihydroxybenzaldeyde (4). Their herbicidal potential was detected by polarographic techniques. All of them inhibited the non-cyclic electron transport on basal, phosphorylating and uncoupled conditions from H 2 O to methylviologen (MV); thus, they act as Hill reaction inhibitors. Studies on fluorescence of chlorophyll a (ChL a) indicated they have different modes of interaction and inhibition sites on the photosystem II electron transport chain; 1-3 have interacted with the acceptor side while 4 has interacted at the donor side.

The initial ACT configuration design task of the integrated application of active controls (IAAC) technology project within the Energy Efficient Transport Program is summarized. A constrained application of active controls technology (ACT) resulted in significant improvements over a conventional baseline configuration previously established. The configuration uses the same levels of technology, takeoff gross weight, payload, and design requirements/objectives as the baseline, except for flying qualities, flutter, and ACT. The baseline wing is moved forward 1.68 m. The configuration incorporates pitch-augmented stability (which enabled an approximately 10% aft shift in cruise center of gravity and a 45% reduction in horizontal tail size), lateral/directional-augmented stability, an angle of attack limiter, wing load alleviation, and flutter mode control. This resulted in a 930 kg reduction in airplane operating empty weight and a 3.6% improvement in cruise efficiency, yielding a 13% range increase. Adjusted to the 3590 km baseline mission range, this amounts to 6% block fuel reduction and a 15.7% higher incremental return on investment, using 1978 dollars and fuel cost.

Flame propagation in both the optically thin and the optically thick regime of radiative transport was studied experimentally using particle-laden gas mixtures. Data on flame shapes, propagation rates, peak pressure, maximum rate of pressure rise, and thermal decay in the burned gases are consistent with the hypothesis that, at low particle loadings, the particles act to increase the radiative loss from the gases, whereas at higher loadings, reabsorption of emitted radiation becomes significant. The reabsorption acts to decrease the net radiative loss and augment conductive heat transport. It is speculated that, in sufficiently large systems, in which the absorption length is much smaller than the system size, flammability limits might not exist at microgravity conditions because emitted radiation would not constitute a loss mechanism.

Paracellular transport of solutes and water accompanies transcellular transport across epithelial barriers and together they serve to maintain internal body composition. However, whether paracellular transport is necessary and why it evolved is unknown. In this commentary I discuss our recent studies to address this question in the proximal tubule of the kidney. Paracellular reabsorption of sodium occurs in the proximal tubule and is mediated by claudin-2. However, deletion of claudin-2 in mice does not affect whole kidney sodium excretion because it can be completely compensated by downtream transcellular transport mechanisms. This occurs at the expense of increased oxygen consumption, tissue hypoxia and increased susceptibility to ischemic injury. It is concluded that paracellular transport acts as an energy saving mechanism to increase transport without consuming additional oxygen. It is speculated that this might be why paracellular transport evolved in leaky epithelia with high transport needs.

An investigation of the potential for using the ACTS to provide the data distribution network for a distributed set of users of an astronomical observatory has been conducted. The investigation consisted of gathering the data and interface standards for the ACTS network and the observatory instrumentation and telecommunications devices. A simulation based on COMNET was then developed to test data transport configurations for real-time suitability. The investigation showed that the ACTS network should support the real-time requirements and allow for growth in the observatory needs for data transport.

Charge transport in the mesoscale bulk heterojunctions (BHJs) of organic photovoltaic devices (OPVs) is studied using multiscale simulations in combination with molecular dynamics, the density functional theory, the molecular-level kinetic Monte Carlo (kMC) method, and the coarse-grained kMC method, which was developed to estimate mesoscale carrier mobility. The effects of the degree of crystallinity and the anisotropy of the conductivity of donors on hole mobility are studied for BHJ structures that consist of crystalline and amorphous pentacene grains that act as donors and amorphous C60 grains that act as acceptors. We find that the hole mobility varies dramatically with the degree of crystallinity of pentacene because it is largely restricted by a low-mobility amorphous region that occurs in the hole transport network. It was also found that the percolation threshold of crystalline pentacene is relatively high at approximately 0.6. This high percolation threshold is attributed to the 2D-like conductivity of crystalline pentacene, and the threshold is greatly improved to a value of approximately 0.3 using 3D-like conductive donors. We propose essential guidelines to show that it is critical to increase the degree of crystallinity and develop 3D conductive donors for efficient hole transport through percolative networks in the BHJs of OPVs.

Peritoneal dialysis involves diffusive and convective transports and osmosis through the highly vascularized peritoneal membrane. Several lines of evidence have demonstrated that the water channel aquaporin-1 (AQP1) corresponds to the ultrasmall pore predicted by the modelization of peritoneal transport. Proof-of-principle studies have shown that upregulation of the expression of AQP1 in peritoneal capillaries is reflected by increased water permeability and ultrafiltration, without affecting the osmotic gradient and the permeability for small solutes. Inversely, studies in Aqp1 mice have shown that haplo-insufficiency in AQP1 is reflected by significant attenuation of water transport. Recent studies have identified lead compounds that could act as agonists of aquaporins, as well as putative binding sites and potential mechanisms of gating the water channel. By modulating water transport, these pharmacological agents could have clinically relevant effects in targeting specific tissues or disease states. These studies on the peritoneal membrane also provide an experimental framework to investigate the role of water channels in the endothelium and various cell types.

Transmembrane drug export mediated by the ATP-binding cassette (ABC) transporter P-glycoprotein contributes to clinical resistance to antineoplastics. In this study, we identified the substituted quinoline HG-829 as a novel, noncompetitive, and potent P-glycoprotein inhibitor that overcomes in vitro and in vivo drug resistance. We found that nontoxic concentrations of HG-829 restored sensitivity to P-glycoprotein oncolytic substrates. In ABCB1-overexpressing cell lines, HG-829 significantly enhanced cytotoxicity to daunorubicin, paclitaxel, vinblastine, vincristine, and etoposide. Coadministration of HG-829 fully restored in vivo antitumor activity of daunorubicin in mice without added toxicity. Functional assays showed that HG-829 is not a Pgp substrate or competitive inhibitor of Pgp-mediated drug efflux but rather acts as a noncompetitive modulator of P-glycoprotein transport function. Taken together, our findings indicate that HG-829 is a potent, long-acting, and noncompetitive modulator of P-glycoprotein export function that may offer therapeutic promise for multidrugresistant malignancies. Synthetic cathinones are similar in chemical structure to amphetamines, and their behavioral effects are associated with enhanced dopaminergic signaling. The past ten years of research on the common constituent of bath salts, MDPV (the synthetic cathinone 3,4-methylenedioxypyrovalerone), has aided the understanding of how synthetic cathinones act at the dopamine (DA) transporter (DAT). Several groups have described the ability of MDPV to block the DAT with high-affinity. In this study, we demonstrate for the first time a new mode of action of MDPV, namely its ability to promote DAT-mediated DA efflux. Using single cell amperometric assays, we determined that low concentrations of MDPV (1nM) can cause reverse transport of DA via DAT. Notably, administration of MDPV leads to hyperlocomotion in Drosophila melanogaster. These data describe further how MDPV acts at the DAT, possibly paving the way for novel treatment strategies for individuals who abuse bath salts. Copyright © 2017 Elsevier B.V. All rights reserved.

The aqueous ethanolic extract from leaves of the marine plant Thalassia testudinum has shown antioxidant, cytoprotective, and neuroprotective properties. The chemical composition of this extract, rich in polyphenols, could interfere with active transport of drugs out of the cell and circumvent the phenomenon of multidrug resistance (MDR). The extract can act as an MDR modulator through its interaction with efflux transporters. The ABCG2/BCRP has been shown to confer MDR acting in tumor cells. To evaluate the interaction of ABCG2/BCRP with the extract, studies in cells overexpressing human BCRP transporter and its murine ortholog Bcrp1 were performed. T. testudinum extract could be included as MDR modulator, as interaction with ABCG2/BCRP has been shown through flow cytometry and MTT assays. The cells overexpressing ABCG2/BCRP in the presence of the extract (25-150 μg/mL) decreased the survival rates of the anti-tumoral mitoxantrone. Our results support its inclusion as a possible MDR modulator against tumor cells that overexpress ABCG2/BCRP.

Pseudo-constitutivity of nitrate-responsive genes in nitrate reductase mutants

In fungi, transcriptional activation of genes involved in NO3- assimilation requires the presence of an inducer (nitrate or nitrite) and low intracellular concentrations of the pathway products ammonium or glutamine. In Aspergillus nidulans, the two transcription factors NirA and AreA act synergistically to mediate nitrate/nitrite induction and nitrogen metabolite derepression, respectively. In all studied fungi and in plants, mutants lacking nitrate reductase (NR) activity express nitrate-metabolizing enzymes constitutively without the addition of inducer molecules. Based on their work in A. nidulans, Cove and Pateman proposed an “autoregulation control” model for the synthesis of nitrate metabolizing enzymes in which the functional nitrate reductase molecule would act as co-repressor in the absence and as co-inducer in the presence of nitrate. However, NR mutants could simply show “pseudo-constitutivity” due to induction by nitrate which accumulates over time in NR-deficient strains. Here we examined this possibility using strains which lack flavohemoglobins (fhbs), and are thus unable to generate nitrate internally, in combination with nitrate transporter mutations (nrtA, nrtB) and a GFP-labeled NirA protein. Using different combinations of genotypes we demonstrate that nitrate transporters are functional also in NR null mutants and show that the constitutive phenotype of NR mutants is not due to nitrate accumulation from intracellular sources but depends on the activity of nitrate transporters. However, these transporters are not required for nitrate signaling because addition of external nitrate (10 mM) leads to standard induction of nitrate assimilatory genes in the nitrate transporter double mutants. We finally show that NR does not regulate NirA localization and activity, and thus the autoregulation model, in which NR would act as a co-repressor of NirA in the absence of nitrate, is unlikely to be correct. Results from this study instead suggest that transporter-mediated NO3- accumulation in NR deficient mutants, originating from traces of nitrate in the media, is responsible for the constitutive expression of NirA-regulated genes, and the associated phenotype is thus termed “pseudo-constitutive”. Polycyclic Aromatic Hydrocarbons (PAHs) transported from contaminated soils by surface runoff pose significant risk for aquatic ecosystems. Based on a rainfall-runoff simulation experiment, this study investigated the impact of carbonaceous materials (CMs) in soil, identified by organic petrology analysis, on the transport of soil-bound PAHs under rainfall conditions. The hypothesis that composition of soil organic matter significantly impacts the enrichment and transport of PAHs was proved. CMs in soil, varying significantly in content, mobility and adsorption capacity, act differently on the transport of PAHs. Anthropogenic CMs like black carbon (BC) largely control the transport, as PAHs may be preferentially attached to them. Eventually, this study led to a rethink of the traditional enrichment theory. An important implication is that CMs in soil have to be explicitly considered to appropriately model the nonpoint source pollution of PAHs (possibly other hydrophobic chemicals as well) and assess its environmental risk. This dissertation reports the findings from the Salt Lake Valley flux study. The Salt Lake Valley flux study was designed to improve our understanding of the complex land-atmosphere interactions in urban areas. The flux study used the eddy covariance technique to quantify carbon dioxide and surface energy budget in the semiarid Salt Lake Valley. Apart from quantifying fluxes, the study has also added new insight into the nature of turbulent scalar transport in urban areas and has addressed some of the complications in using Eddy Covariance technique in urban areas. As part of this experiment, eddy fluxes of CO2 and surface energy fluxes were measured at two sites, with distinct urban landforms; One site was located in a suburban neighborhood with substantial vegetative cover, prototypical of many residential neighborhoods in the valley. The other CO2 site was in a preurban surrounding that resembled the Salt Lake Valley before it was urbanized. The two sites were intentionally chosen to illustrate the impact of urbanization on CO 2 and surface energy flux cycles. Results indicate that the suburban site acted as a sink of CO2 during the midday period due to photosynthesis and acted as a source of CO2 during the evening and nighttime periods. The vegetative cover around the suburban site also had a significant impact on the surface energy fluxes. Contribution from latent heat flux was substantially high at the suburban site during the summer months compared to sensible heat. The turbulence investigation found that the general behavior of turbulence was very much influenced by local factors and the statistics did not always obey Monin-Obukhov Similarity parameters. This investigation also found that the scalar (co)spectra observed at the suburban site were characterized by multiple peaks and were different compared to (co)spectra reported over forest and crop canopies. The study also observed multiscale CO2 transport at the suburban site during the convective period. While the suburban site acted as a net sink of CO2 during the midday periods, CO2 was transported positively at lower frequencies. Also, during the evening transition period, when the site switched from a sink of CO2 to a source, the net upward transport of CO2 was cancelled by the net downward transport of CO2. This balance caused the total net transport reduce to zero. Overall this research, apart from characterizing the behavior of scalar fluxes at a suburban site in a semiarid desert city has also added some valuable insight on the complex nature of urban scalar transport.

The Final ACT Configuration Evaluation Task of the Integrated Application of Active Controls (IAAC) technology project within the energy efficient transport program is summarized. The Final ACT Configuration, through application of Active Controls Technology (ACT) in combination with increased wing span, exhibits significant performance improvements over the conventional baseline configuration. At the design range for these configurations, 3590 km, the block fuel used is 10% less for the Final ACT Configuration, with significant reductions in fuel usage at all operational ranges. Results of this improved fuel usage and additional system and airframe costs and the complexity required to achieve it were analyzed to determine its economic effects. For a 926 km mission, the incremental return on investment is nearly 25% at 1980 fuel prices. For longer range missions or increased fuel prices, the return is greater. The technical risks encountered in the Final ACT Configuration design and the research and development effort required to reduce these risks to levels acceptable for commercial airplane design are identified.

This article considers all 87 attacks worldwide against air and rail transport systems that killed at least two passengers over the 30-year period of 1982-2011. The data offer strong and statistically significant evidence that successful acts of terror have "gone to ground" in recent years: attacks against aviation were concentrated early in the three decades studied whereas those against rail were concentrated later. Recent data are used to make estimates of absolute and comparative risk for frequent flyers and subway/rail commuters. Point estimates in the "status quo" case imply that mortality risk from successful acts of terror was very low on both modes of transportation and that, whereas risk per trip is higher for air travelers than subway/rail commuters, the rail commuters experience greater risk per year than the frequent flyers. © 2015 Society for Risk Analysis.

Jones Act ships must be used to move crude and petroleum products between US ports. However, if no suitable Jones Act tankers or barges are available, shippers may apply, on a case-by-case basis, for a wavier to use substantially cheaper foreign capacity instead. Case-by-case determination of these waivers creates a potential problem for an emergency drawdown of the Strategic Petroleum Reserve (SPR): potential buyers of crude would have to submit bids while facing considerable uncertainty about their ultimate costs of transporting the SPR oil. This report examines an alternative to the case-by-case waiver process, which we call a ''composite waivermore » procedure.'' The composite waiver procedure has two elements: a general waiver of the Jones Act requirement for moving SPR crude; and a rebate, equal to the price differential between foreign ships and Jones Act ships, to be paid to any buyer of SPR crude who uses US bottoms for transport. The composite waiver is designed to reduce the uncertainty surrounding transportation costs in the crude bidding process, and to keep Jones Act tankers price competitive so these vessels are not eliminated from use during an SPR emergency drawdown. 17 figs.« less

Long-distance axonal transport is critical to the maintenance and function of neurons. Robust transport is ensured by the coordinated activities of multiple molecular motors acting in a team. Conventional live-cell imaging techniques used in axonal transport studies detect this activity by visualizing the translational dynamics of a cargo. However, translational measurements are insensitive to torques induced by motor activities. By using gold nanorods and multichannel polarization microscopy, we simultaneously measure the rotational and translational dynamics for thousands of axonally transported endosomes. We find that the rotational dynamics of an endosome provide complementary information regarding molecular motor activities to the conventionally tracked translational dynamics. Rotational dynamics correlate with translational dynamics, particularly in cases of increased rotation after switches between kinesin- and dynein-mediated transport. Furthermore, unambiguous measurement of nanorod angle shows that endosome-contained nanorods align with the orientation of microtubules, suggesting a direct mechanical linkage between the ligand-receptor complex and the microtubule motors.

For the first time, we have directly measured the transport of a vector magnetic field by isotropic turbulence in a high Reynolds number liquid metal flow. In analogy with direct measurements of the turbulent Reynolds stress (turbulent viscosity) that governs momentum transport, we have measured the turbulent electromotive force (emf) by simultaneously measuring three components of velocity and magnetic fields, and computed the correlations that lead to mean-field current generation. Furthermore, we show that this turbulent emf tends to oppose and cancel out the local current, acting to increase the effective resistivity of the medium, i.e., it acts as anmore » enhanced magnetic diffusivity. Current estimates of biogenic carbon fluxes over North America based on top-down atmospheric inversions are subject to considerable uncertainty. This uncertainty stems to a large part from the uncertain prior fluxes estimates with the associated error covariances and approximations in the atmospheric transport models that link observed carbon dioxide mixing ratios with surface fluxes. Specifically, approximations in the representation of vertical mixing associated with atmospheric turbulence or convective transport and largely under-determined prior fluxes and their error structures significantly hamper our capacity to reliably estimate regional carbon fluxes. The Atmospheric Carbon and Transport - America (ACT-America) mission aims at reducing the uncertainties in inverse fluxes at the regional-scale by deploying airborne and ground-based platforms to characterize atmospheric GHG mixing ratios and the concurrent atmospheric dynamics. Two aircraft measure the 3-dimensional distribution of greenhouse gases at synoptic scales, focusing on the atmospheric boundary layer and the free troposphere during both fair and stormy weather conditions. Here we analyze two main questions: (i) What level of information can we expect from the currently planned observations? (ii) How might ACT-America reduce the hindcast and predictive uncertainty of carbon estimates over North America?

This report summarizes the Integrated Application of Active Controls (IAAC) Technology to an Advanced Subsonic Transport Project, established as one element of the NASA/Boeing Energy Efficient Transport Technology Program. The performance assessment showed that incorporating ACT into an airplane designed to fly approximately 200 passengers approximately 2,000 nmi could yield block fuel savings from 6 to 10 percent at the design range. The principal risks associated with incorporating these active control functions into a commercial airplane are those involved with the ACT system implementation. The Test and Evaluation phase of the IAAC Project focused on the design, fabrication, and test of a system that implemented pitch axis fly-by-wire, pitch axis augmentation, and wing load alleviation. The system was built to be flight worthy, and was planned to be experimentally flown on the 757. The system was installed in the Boeing Digital Avionics Flight Controls Laboratory (DAFCL), where open loop hardware and software tests, and a brief examination of a direct drive valve (DDV) actuation concept were accomplished. The IAAC Project has shown that ACT can be beneficially incorporated into a commercial transport airplane. Based on the results achieved during the testing phase, there appears to be no fundamental reason(s) that would preclude the commercial application of ACT, assuming an appropriate development effort is included.

Statistics from the French Employee National Health Insurance Fund indicate high accident levels in the transport sector. This study represents initial thinking on a new approach to transport sector prevention based on the assumption that a work situation could be improved by acting on another interconnected work situation. Ergonomic analysis of two connected work situations, involving the road haulage drivers and cross-docking platform employees, was performed to test this assumption. Our results show that drivers are exposed to a number of identified risks, but their multiple tasks raise the question of activity intensification. The conditions, under which the drivers will perform their work and take to the road, are partly determined by the quality and organisation of the platform with which they interact. We make a number of recommendations (e.g. changing handling equipment, re-appraising certain jobs) to improve platform organisation and employee working conditions with the aim of also improving driver conditions. These initial steps in this prevention approach appear promising, but more detailed investigation is required. Copyright © 2013 Elsevier Ltd. All rights reserved.

Isotonic transport by the Na+-glucose cotransporter SGLT1 from humans and rabbit

In order to study its role in steady state water transport, the Na+-glucose cotransporter (SGLT1) was expressed in Xenopus laevis oocytes; both the human and the rabbit clones were tested. The transport activity was monitored as a clamp current and the flux of water followed optically as the change in oocyte volume. SGLT1 has two modes of water transport. First, it acts as a molecular water pump: for each 2 Na+ and 1 sugar molecule 264 water molecules were cotransported in the human SGLT1 (hSGLT1), 424 for the rabbit SGLT1 (rSGLT1). Second, it acts as a water channel. The cotransport of water was tightly coupled to the sugar-induced clamp current. Instantaneous changes in clamp current induced by changes in clamp voltage were accompanied by instantaneous changes in the rate of water transport. The cotransported solution was predicted to be hypertonic, and an osmotic gradient built up across the oocyte membrane with continued transport; this resulted in an additional osmotic influx of water. After 5-10 min a steady state was achieved in which the total influx was predicted to be isotonic with the intracellular solution. With the given expression levels, the steady state water transport was divided about equally between cotransport, osmosis across the SGLT1 and osmosis across the native oocyte membrane. Coexpression of AQP1 with the SGLT1 increased the water permeability more than 10-fold and steady state isotonic transport was achieved after less than 2 s of sugar activation. One-third of the water was cotransported, and the remainder was osmotically driven through the AQP1. The data suggest that SGLT1 has three roles in isotonic water transport: it cotransports water directly, it supplies a passive pathway for osmotic water transport, and it generates an osmotic driving force that can be employed by other pathways, for example aquaporins. The Washington, DC, Youth Curfew: effect on transports of injured youth and homicides.

Curfews are implemented to curtail youth violence. Trauma systems and emergency medical services (EMS) may need to prepare for changes in patient volume resulting from local ordinances. This study evaluated the impact of the 1995 Washington, DC, Juvenile Curfew Act on EMS transports of injured youth and on youth homicides. A retrospective, comparative cohort study was performed. Transports of injured youth and youth homicides were counted in corresponding months of 1994 and 1995. Cohorts were formed by year, time of day, age group, and mechanism of injury. Year-to-year statistical comparisons of injury proportions were performed using the chi square and Fisher's exact tests. One thousand forty-eight transports were included. No significant difference was observed in transports with curfew implementation. Most assaults on youth occurred outside the curfew time. No effect of the curfew on homicides was detected. Of 67 homicides, only two victims were under the curfew. No effect of the curfew on transports for injuries or on homicides was demonstrated. The curfew was not in effect during the period of highest risk.

Pre-hospital airway management represents the intervention most likely to impact outcomes in critically injured patients. As such, airway management issues dominate quality improvement (QI) reviews of aero-medical programs. The purpose of this study was to evaluate current practice patterns of airway management in trauma among U.S. aero-medical service (AMS) programs. The Association of Air Medical Services (AAMS) Resource Guide from 2005 to 2006 was utilized to identify the e-mail addresses of all directors of U.S. aero-medical transport programs. Program directors from 182 U.S. aero-medical programs were asked to participate in an anonymous, web-based survey of emergency airway management protocols and practices. Non-responders to the initial request were contacted a second time by e-mail. 89 programs responded. 98.9% have rapid sequence intubation (RSI) protocols. 90% use succinylcholine, 70% use long-acting neuromuscular blockers (NMB) within their RSI protocol. 77% have protocols for mandatory in-flight sedation but only 13% have similar protocols for maintenance paralytics. 60% administer long-acting NMB immediately after RSI, 13% after confirmation of neurological activity. Given clinical scenarios, however, 97% administer long-acting NMB to patients with scene and in-flight Glasgow Coma Scale (GCS) of 3, even for brief transport times. The majority of AMS programs have well defined RSI and in-flight sedation protocols, while protocols for in-flight NMB are uncommon. Despite this, nearly all programs administer long-acting NMB following RSI, irrespective of GCS or flight time. Given the impact of in-flight NMB on initial assessment, early intervention, and injury severity scoring, a critical appraisal of current AMS airway management practices appears warranted.

Motivated by discrepancies between Eulerian transport estimates and the behavior of Lagrangian surface drifters, near-surface transport pathways and processes in the North Atlantic are studied using a combination of data, altimetric surface heights, statistical analysis of trajectories, and dynamical systems techniques. Particular attention is paid to the issue of the subtropical-to-subpolar intergyre fluid exchange. The velocity field used in this study is composed of a steady drifter-derived background flow, upon which a time-dependent altimeter-based perturbation is superimposed. This analysis suggests that most of the fluid entering the subpolar gyre from the subtropical gyre within two years comes from a narrow region lying inshore of the Gulf Stream core, whereas fluid on the offshore side of the Gulf Stream is largely prevented from doing so by the Gulf Stream core, which acts as a strong transport barrier, in agreement with past studies. The transport barrier near the Gulf Stream core is robust and persistent from 1992 until 2008. The qualitative behavior is found to be largely independent of the Ekman drift.
