  1. [Spatiotemporal changes of wetlands in Hangzhou Bay Industrial Belt].

    By using RS and GIS techniques, the spatiotemporal changes of wetlands in Hangzhou Bay Industrial Belt, one of the most developed zones in Zhejiang Province, from 1990 to 2005 were studied. There was a frequent conversion between the wetlands and other land use types and between the wetlands themselves, mainly manifested in the conversion between wetland and farmland, and from wetland to construction land and from tidal flat to aquiculture area. The comparative advantage of other land use types and the policy of cultivated land's requisition-compensation balance decided the inherent mechanisms of these spatiotemporal changes. Driven by the aquaculture's comparative advantage to traditional agriculture, large areas of inland farmland and of the tidal flat along the coast of Hangzhou Bay were reclaimed into aquiculture area, and the rapid expansion of construction land, limited land resources, and the implement of cultivated land's requisition-compensation balance policy induced the wetlands being occupied.

    Exposure to malachite green (MG) may pose great health risks to humans; thus, it is of prime importance to develop fast and robust methods to quantitatively screen the presence of malachite green in water. Herein the application of extractive electrospray ionization mass spectrometry (EESI-MS) has been extended to the trace detection of MG within lake water and aquiculture water, due to the intensive use of MG as a biocide in fisheries. This method has the advantage of obviating offline liquid-liquid extraction or tedious matrix separation prior to the measurement of malachite green in native aqueous medium. The experimental results indicate that the extrapolated detection limit for MG was ~3.8 μg·L−1 (S/N = 3) in lake water samples and ~0.5 μg·L−1 in ultrapure water under optimized experimental conditions. The signal intensity of MG showed good linearity over the concentration range of 10–1000 μg·L−1. Measurement of practical water samples fortified with MG at 0.01, 0.1 and 1.0 mg·L−1 gave a good validation of the established calibration curve. The average recoveries and relative standard deviation (RSD) of malachite green in lake water and Carassius carassius fish farm effluent water were 115% (6.64% RSD), 85.4% (9.17% RSD) and 96.0% (7.44% RSD), respectively. Overall, the established EESI-MS/MS method has been demonstrated suitable for sensitive and rapid (<2 min per sample) quantitative detection of malachite green in various aqueous media, indicating its potential for online real-time monitoring of real life samples.   5. Condensation of atmospheric moisture from tropical maritime air masses as a freshwater resource.

    A method is proposed whereby potable water may be obtained by condensing moisture from the atmosphere in suitable seashore or island areas. Deep, cold, offshore seawater is used as a source of cold and is pumped to condensers set up on shore to intercept the flow of highly humid, tropical, maritime air masses. This air, when cooled, condenses moisture, which is conducted away and stored for use as a water supply. Windmill-driven generators would supply low-cost power for the operation. Side benefits are derived by using the nutritious deep water to support aquiculture in nearby lagoons or to enhance the productivity of the outfall area. Additional benefits are derived from the condenser as an air-conditioning device for nearby residents. The islands of the Caribbean are used as an example of a location in the trade-winds belt where nearly optimum conditions for the operation of this system can be found.

  6. Water resources of the Swinomish Indian Reservation, Washington

    Average total inflow to and outflow from the hydrologic system of the Swinomish Indian Reservation, Wash., is 24 cfs (cubic feet per second). Recharge to the ground-water reservoir is 8 cfs. The study area is underlain by three types of unconsolidated deposits: the uppermost unit is till, the intermediate unit is sand and gravel, and the lowermost unit is clay and silt. During 1975 the average rate of water use was 0.19 cfs. About 70 percent was pumped from the ground-water reservoir and the rest was imported. Dissolved-solids concentrations are 10-20 mg/L (milligrams per liter). In the zone of saltwater, differsion concentrations up to 1,570 mg/L were measured. Human interaction with the hydrologic system has had little effect on water quality. Ground-water quality is generally well within acceptable limits for most uses. If 20 percent of ground-water can be intercepted then a net rate of withdrawal of 1.6 cfs can be attained. Aquiculture development is possible on the two largest streams in the reservation in the form of incubation stations handling 600,000 eggs each. (Woodard-USGS)

  7. An Operational Coastal Forecasting System in Galicia (NW Spain)

    The Galician coast (NW Iberian Peninsula coast) and mainly the Rias Baixas (southern Galician rias) are one of the most productive ecosystems in the world, supporting a very active fishing and aquiculture industry. This high productivity lives together with a high human pressure and an intense maritime traffic, which means an important environmental risk. Besides that, Harmful Algae Blooms (HAB) are common in this area, producing important economical losses in aquiculture. In this context, the development of an Operational Hydrodynamic Ocean Forecast System is the first step to the development of a more sophisticated Ocean Integrated Decision Support Tool. A regional oceanographic forecasting system in the Galician Coast has been developed by MeteoGalicia (the Galician regional meteorological agency) inside ESEOO project to provide forecasts on currents, sea level, water temperature and salinity. This system is based on hydrodynamic model MOHID, forced with the operational meteorological model WRF, supported daily at MeteoGalicia . Two grid meshes are running nested at different scales, one of ~2km at the shelf scale and the other one with a resolution of 500 m at the rias scale. ESEOAT (Puertos del Estado) model provide salinity and temperature fields which are relaxed at all depth along the open boundary of the regional model (~6km). Temperature and salinity initial fields are also obtained from this application. Freshwater input from main rivers are included as forcing in MOHID model. Monthly mean discharge data from gauge station have been provided by Aguas de Galicia. Nowadays a coupling between an hydrological model (SWAT) and the hydrodynamic one are in development with the aim to verify the impact of the rivers discharges. The system runs operationally daily, providing two days of forecast. First model verifications had been performed against Puertos del Estado buoys and Xunta de Galicia buoys network along the Galician coast. High resolution model results were validated against a CTDs profiles campaign carried out during an oil spill exercise in the Ria de Vigo in April 2007. During EROCIPS INTERREG IIIB and EASY INTERREG IVB projects, a Galician oceanographic observation network were built. Three stations located inside the Rias Baixas allow to collect meteorological and oceanographic data at different depths to calibrate and validate the modelization of the rias. To complete this network and to create a common data platform a new project emerged (RAIA INTERREG IVA). It will provide MeteoGalicia more scientific data to improve the study of the rias. Furthermore, MeteoGalicia is also involved in DRIFTER AMPERA project which allows to improve the capability of modelling and monitoring the trajectory of hazardous substances and inerts.

  8. Detection of adenoviruses in shellfish by means of conventional-PCR, nested-PCR, and integrated cell culture PCR (ICC/PCR).

    We tested three PCR based methodologies to detect adenoviruses associated with cultivated oysters. Conventional-PCR, nested-PCR, and integrated cell culture-PCR (ICC/PCR) were first optimized using oysters seeded with know amounts of Adenovirus serotype 5 (Ad5). The maximum sensitivity for Ad5 detection was determined for each method, and then used to detect natural adenovirus contamination in oysters from three aquiculture farms in Florianopolis, Santa Catarina State, Brazil, over a period of 6 months. The results showed that the nested-PCR was more sensitive (limit of detection: 1.2 PFU/g of tissue) than conventional-PCR and ICC-PCR (limit of detection for both: 1.2 x 10(2)PFU/g of tissue) for detection of Ad5 in oyster extracts. Nested-PCR was able to detect 90% of Ad5 contamination in harvested oyster samples, while conventional-PCR was unable to detect Ad5 in any of the samples. The present work suggests that detection of human adenoviruses can be used as a tool to monitor the presence of human viruses in marine environments where shellfish grow, and that nested-PCR is the method of choice.

    Mactra veneriformis has been used as sea food and traditional Chinese medicine (TCM) for thousands of years in China. In the present study, a high performance liquid chromatograph coupled with photodiode array detector and electrospray ionisation-mass spectrometer (HPLC-DAD-ESI-MS) method was established for detection of the nucleosides and nucleobases in M. veneriformis from four aquaticultural area of Jiangsu during different harvest time of one year. The validated method was successfully applied to identifying 10 nucleosides and nucleobases in 48 M. veneriformis samples. Quantitative analysis showed that nucleosides and nucleobases are rich in all M. veneriformis samples. However, their contents vary in different areas and harvest times. Principal component analysis (PCA) was used to classify the 48 samples based on the contents of the nucleosides and nucleobases. As a result, the samples could be mainly clustered into four groups, which was similar as aquaticultural areas classification. Based on the results, present method might be applicable for the quality control of M. veneriformis, or even other marine shellfish aquiculture and their products, and the quality of M. veneriformis might be more related with aquaticultural areas.   10. Nutrient and chlorophyll a anomaly in red-tide periods of 2003-2008 in Sishili Bay, China

    Sishili Bay is the most important aquiculture and tourism area for the city of Yantai, China; however, red tides occurred frequently and have caused huge economic losses in this bay in recent years. To gain a better understanding of the local ecological environments in the bay, we conducted this research between 2003 and 2008 to analyze variations in nutrients and chlorophyll (chl- a) during high frequency red tide period (May to September). The results show that the chl- a concentration increased from 2.70 in 2003 to 7.26 mg/m3 in 2008, while the concentration of total inorganic nitrogen (TIN) and silicate (SiO3-Si) increased lineally from 5.18 and 1.45 μmol/L in 2003 to 18.57 and 9.52 μmol/L in 2008, respectively, and the annual phosphate (PO4-P) varied between 0.15 and 0.46 μmol/L. Special attention was given to a red tide in August 2007 occurred when water temperature was high and nutrient concentrations increased sharply because of a heavy rainfall. Overall, the results show the P limitation in Sishili Bay, and reveal that red tides were caused by eutrophication from terrestrial inputs and local warm weather, particularly during rainy periods. Therefore, to control red tide, greater efforts should be made to reduce sewage discharges into Sishili Bay, particularly during rainfall seasons.

  11. Arsenic methylation by an arsenite S-adenosylmethionine methyltransferase from Spirulina platensis.

    Arsenic-contaminated water is a serious hazard for human health. Plankton plays a critical role in the fate and toxicity of arsenic in water by accumulation and biotransformation. Spirulina platensis (S. platensis), a typical plankton, is often used as a supplement or feed for pharmacy and aquiculture, and may introduce arsenic into the food chain, resulting in a risk to human health. However, there are few studies about how S. platensis biotransforms arsenic. In this study, we investigated arsenic biotransformation by S. platensis. When exposed to arsenite (As(III)), S. platensis accumulated arsenic up to 4.1mg/kg dry weight. After exposure to As(III), arsenate (As(V)) was the predominant species making up 64% to 86% of the total arsenic. Monomethylarsenate (MMA(V)) and dimethylarsenate (DMA(V)) were also detected. An arsenite S-adenosylmethionine methyltransferase from S. platensis (SpArsM) was identified and characterized. SpArsM showed low identity with other reported ArsM enzymes. The Escherichia coli AW3110 bearing SparsM gene resulted in As(III) methylation and conferring resistance to As(III). The in vitro assay showed that SpArsM exhibited As(III) methylation activity. DMA(V) and a small amount of MMA(V) were detected in the reaction system within 0.5hr. A truncated SpArsM derivative lacking the last 34 residues still had the ability to methylate As(III). The three single mutants of SpArsM (C59S, C186S, and C238S) abolished the capability of As(III) methylation, suggesting the three cysteine residues are involved in catalysis. We propose that SpArsM is responsible for As methylation and detoxification of As(III) and may contribute to As biogeochemistry. Copyright © 2016. Published by Elsevier B.V.

  12. De-confounding of Relations Between Land-Level and Sea-Level Change, Humboldt Bay, Northern California: Uncertain Predictions of Magnitude and Timing of Tectonic and Eustatic Processes

    Humboldt Bay in Northern California provides a unique opportunity to investigate the effects of relative sea level change on both native flora and maritime aquiculture as influenced by both tectonic and eustatic sea-level changes. This combination of superposed influences makes quantitatively predicting relative sea-level more uncertain and consumption of the results for public planning purposes exceedingly difficult. Public digestion for practical purposes is confounded by the fact that the uncertainty for eustatic sea-level changes is a magnitude issue while the uncertainty associated with the tectonic land level changes is both a magnitude and timing problem. Secondly, the public is less well informed regarding how crustal deformation contributes to relative sea-level change. We model the superposed effects of eustatic sea-level rise and tectonically driven land-level changes on the spatial distribution of habitats suitable to native eelgrass (Zostera marina) and oyster mariculture operations in Humboldt Bay. While these intertidal organisms were chosen primarily because they have vertically restricted spatial distributions that can be successfully modeled, the public awareness of their ecologic and economic importance is also well developed. We employ easy to understand graphics depicting conceptual ideas along with maps generated from the modeling results to develop locally relevant estimates of future sea level rise over the next 100 years, a time frame consistent with local planning. We bracket these estimates based on the range of possible vertical deformation changes. These graphic displays can be used as a starting point to propose local outcomes from global and regional relative sea-level changes with respect to changes in the distribution of suitable habitat for ecologically and economically valuable species. Currently the largest sources of uncertainty for changes in relative sea-level in the Humboldt Bay area are 1) the rate and magnitude of tectonic deformation throughout the earthquake cycle and 2) the stability and reliability of the tide gauges and other benchmarks assumed to be stable in the Humboldt Bay region.
