Quantitative characterization of nanoparticle interactions with their surrounding environment is vital for safe nanotechnological development and standardization. A recent quantitative measure, the biological surface adsorption index (BSAI), has demonstrated promising applications in nanomaterial surface characterization and biological/environmental prediction. This paper further advances the approach beyond the application of five descriptors in the original BSAI to address the concentration dependence of the descriptors, enabling better prediction of the adsorption profile and more accurate categorization of nanomaterials based on their surface properties. Statistical analysis on the obtained adsorption data was performed based on three different models: the original BSAI, a concentration-dependent polynomial model, and an infinite dilution model. These advancements in BSAI modeling showed a promising development in the application of quantitative predictive modeling in biological applications, nanomedicine, and environmental safety assessment of nanomaterials.

Seabirds are thought to be reliable, real-time indicators of forage fish availability and the climatic and biotic factors affecting pelagic food webs in marine ecosystems. In this study, we tested the hypothesis that temporal trends and interannual variability in seabird indicators reflect simultaneously occurring bottom-up (climatic) and competitor (pink salmon) forcing of food webs. To test this hypothesis, we derived multivariate seabird indicators for the Bering Sea–Aleutian Island (BSAI) ecosystem and related them to physical and biological conditions known to affect pelagic food webs in the ecosystem. We examined covariance in the breeding biology of congeneric pelagic gulls (kittiwakes Rissa tridactyla and R. brevirostris) andauks (murres Uria aalge and U. lomvia), all of whichare abundant and well-studiedinthe BSAI. At the large ecosystem scale, kittiwake and murre breeding success and phenology (hatch dates) covaried among congeners, so data could be combined using multivariate techniques, but patterns of responsedifferedsubstantially betweenthe genera.Whiledata fromall sites (n = 5)inthe ecosystemcould be combined, the south eastern Bering Sea shelf colonies (St. George, St. Paul, and Cape Peirce) provided the strongest loadings on indicators, and hence had the strongest influence on modes of variability. The kittiwake breeding success mode of variability, dominated by biennial variation, was significantly related to both climatic factors and potential competitor interactions. The murre indicator mode was interannual and only weakly related to the climatic factors measured. The kittiwake phenology indicator mode of variability showed multi-year periods (“stanzas”) of late or early breeding, while the murre phenology indicator showed a trend towards earlier timing. Ocean climate relationships with the kittiwake breeding success indicator suggestthat early-season (winter–spring) environmental conditions and the abundance of pink salmon affect the pelagic food webs that support these seabirds in the BSAI ecosystem.

Surface of detonation nanodiamonds was functionalized for the covalent attachment of immunoglobulin, and simultaneously bovine serum albumin and Rabbit Anti-Mouse Antibody. The nanodiamond-IgGI125 and RAM-nanodiamond-BSAI125 complexes are stable in blood serum and the immobilized proteins retain their biological activity. It was shown that the RAM-nanodiamond-BSAI125 complex is able to bind to the target antigen immobilized on the Sepharose 6B matrix through antibody-antigen interaction. The idea can be extended to use nanodiamonds as carriers for delivery of bioactive substances (i.e., drugs) to various targets in vivo.

A complete characterization of the different physico-chemical properties of nanoparticles (NPs) is necessary for the evaluation of their impact on health and environment. Among these properties, the surface characterization of the nanomaterial is the least developed and in many cases limited to the measurement of surface composition and zetapotential. The biological surface adsorption index approach (BSAI) for characterization of surface adsorption properties of NPs has recently been introduced (Xia et al. Nat Nanotechnol 5:671-675, 2010; Xia et al. ACS Nano 5(11):9074-9081, 2011). The BSAI approach offers in principle the possibility to characterize the different interaction forces exerted between a NP's surface and an organic--and by extension biological--entity. The present work further develops the BSAI approach and optimizes a solid-phase microextraction gas chromatography-mass spectrometry (SPME/GC-MS) method which, as an outcome, gives a better-defined quantification of the adsorption properties on NPs. We investigated the various aspects of the SPME/GC-MS method, including kinetics of adsorption of probe compounds on SPME fiber, kinetic of adsorption of probe compounds on NP's surface, and optimization of NP's concentration. The optimized conditions were then tested on 33 probe compounds and on Au NPs (15 nm) and SiO2 NPs (50 nm). The procedure allowed the identification of three compounds adsorbed by silica NPs and nine compounds by Au NPs, with equilibrium times which varied between 30 min and 12 h. Adsorption coefficients of 4.66 ± 0.23 and 4.44 ± 0.26 were calculated for 1-methylnaphtalene and biphenyl, compared to literature values of 4.89 and 5.18, respectively. The results demonstrated that the detailed optimization of the SPME/GC-MS method under various conditions is a critical factor and a prerequisite to the application of the BSAI approach as a tool to characterize surface adsorption properties of NPs and therefore to draw any further conclusions on their potential impact on health.

The primary goals of this dissertation were to determine the relationships between interpersonal teaching behaviors and student achievement and affective learning outcomes. The instrument used to collect student perceptions of teacher interpersonal teaching behaviors was the Questionnaire on Teacher Interactions (QTI). The instrument used to assess student affective learning outcomes was the Biology Student Affective Instrument (BSAI). The interpersonal teaching behavior data were collected using students as the observers. 111 students in an urban influenced, rural high school answered the QTI and BSAI in September 1997 and again in April 1998. At the same time students were pre and post tested using the Biology End of Course Examination (BECE). The QTI has been used primarily in European and Oceanic areas. The instrument was also primarily used in educational stratified environment. This was the first time the BSAI was used to assess student affective learning outcomes. The BECE is a Texas normed cognitive assessment test and it is used by Texas schools districts as the end of course examination in biology. The interpersonal teaching behaviors model was tested to ascertain if predictive power in the USA and in a non-stratified educational environment. Findings indicate that the QTI is an adequate predictor of student achievement in biology. The results were not congruent with the non-USA data and results, this indicates that the QTI is a society/culturally sensitive instrument and the instrument needs to be normed to a particular society/culture before it is used to affect teachers' and students' educational environments.

...NMFS proposes regulations to implement Amendment 102 to the Fishery Management Plan for Groundfish of the Bering Sea and Aleutian Islands Management Area (BSAI FMP), and amend the Individual Fishing Quota Program for the Fixed-Gear Commercial Fisheries for Pacific Halibut and Sablefish in Waters in and off Alaska (IFQ Program). Amendment 102 and its proposed implementing regulations would create a Community Quota Entity (CQE) Program in halibut IFQ regulatory area 4B (Area 4B) and the sablefish Aleutian Islands regulatory area that is similar to the existing CQE Program in the Gulf of Alaska (GOA). Amendment 102 would also allow an eligible community in Area 4B and in the Aleutian Islands to establish a non-profit organization as a CQE to purchase halibut catcher vessel quota share (QS) assigned to Area 4B and sablefish QS assigned to the Aleutian Islands. The CQE could assign the resulting annual halibut and sablefish IFQ to participants according to defined CQE Program elements. An additional proposed revision to the IFQ Program regulations would allow IFQ derived from D share halibut QS to be fished on Category C vessels in Area 4B. These actions are necessary to provide additional fishing opportunities for residents of fishery dependent communities and sustain participation in the halibut and sablefish IFQ fisheries. These actions are intended to promote the goals and objectives of the Magnuson-Stevens Fishery Conservation and Management Act, the Northern Pacific Halibut Act of 1982, the BSAI FMP, and other applicable law.

Proceedings of the National Academy of Sciences 100 (21), 12,223-12,228] hypothesized that great whales were an important prey resource for killer whales, and that the removal of fin and sperm whales by commercial whaling in the region of the Bering Sea/Aleutian Islands (BSAI) in the late 1960s and 1970s led to cascading trophic interactions that caused the sequential decline of populations of harbor seal, northern fur seal, Steller sea lion and northern sea otter. This hypothesis, referred to as the Sequential Megafaunal Collapse (SMC), has stirred considerable interest because of its implication for ecosystem-based management. The SMC has the following assumptions: (1) fin whales and sperm whales were important as prey species in the Bering Sea; (2) the biomass of all large whale species (i.e., North Pacific right, fin, humpback, gray, sperm, minke and bowhead whales) was in decline in the Bering Sea in the 1960s and early 1970s; and (3) pinniped declines in the 1970s and 1980s were sequential. We concluded that the available data are not consistent with the first two assumptions of the SMC. Statistical tests of the timing of the declines do not support the assumption that pinniped declines were sequential. We propose two alternative hypotheses for the declines that are more consistent with the available data. While it is plausible, from energetic arguments, for predation by killer whales to have been an important factor in the declines of one or more of the three populations of pinnipeds and the sea otter population in the BSAI region over the last 30 years, we hypothesize that the declines in pinniped populations in the BSAI can best be understood by invoking a multiple factor hypothesis that includes both bottom-up forcing (as indicated by evidence of nutritional stress in the western Steller sea lion population) and top-down forcing (e.g., predation by killer whales, mortality incidental to commercial fishing, directed harvests). Our second hypothesis is a modification of the top-down forcing mechanism (i.e., killer whale predation on one or more of the pinniped populations and the sea otter population is mediated via the recovery of the eastern North Pacific population of the gray whale). We remain skeptical about the proposed link between commercial whaling on fin and sperm whales, which ended in the mid-1960s, and the observed decline of populations of northern fur seal, harbor seal, and Steller sea lion some 15 years later.

Currently, it is assumed that eastern Pacific halibut Hippoglossus stenolepis belong to a single, fully mixed population extending from California through the Bering Sea, in which adult halibut disperse randomly throughout their range during their lifetime. However, we hypothesize that hali but dispersal is more complex than currently assumed and is not spatially random. To test this hypo thesis, we studied the seasonal dispersal and behavior of Pacific halibut in the Bering Sea and Aleutian Islands (BSAI). Pop-up Archival Transmitting tags attached to halibut (82 to 154 cm fork length) during the summer provided no evidence that individuals moved out of the Bering Sea and Aleutian Islands region into the Gulf of Alaska during the mid-winter spawning season, supporting the concept that this region contains a separate spawning group of adult halibut. There was evidence for geographically localized groups of halibut along the Aleutian Island chain, as all of the individuals tagged there displayed residency, with their movements possibly impeded by tidal currents in the passes between islands. Mid-winter aggregation areas of halibut are assumed to be spawning grounds, of which 2 were previously unidentified and extend the species' presumed spawning range ~1000 km west and ~600 km north of the nearest documented spawning area. If there are indeed independent spawning groups of Pacific halibut in the BSAI, their dynamics may vary sufficiently from those of the Gulf of Alaska, so that specifically accounting for their relative segregation and unique dynamics within the larger population model will be necessary for correctly predicting how these components may respond to fishing pressure and changing environmental conditions.?? Quantitative analysis of the interactions between nanomaterials and their surrounding environment is crucial for safety evaluation in the application of nanotechnology as well as its development and standardization. In this chapter, we demonstrate the importance of the adsorption of surrounding molecules onto the surface of nanomaterials by forming biocorona and thus impact the bio-identity and fate of those materials. We illustrate the key factors including various physical forces in determining the interaction happening at bio-nano interfaces. We further discuss the mathematical endeavors in explaining and predicting the adsorption phenomena, and propose a new statistics-based surface adsorption model, the Biological Surface Adsorption Index (BSAI), to quantitatively analyze the interaction profile of surface adsorption of a large group of small organic molecules onto nanomaterials with varying surface physicochemical properties, first employing five descriptors representing the surface energy profile of the nanomaterials, then further incorporating traditional semi-empirical adsorption models to address concentration effects of solutes. These Advancements in surface adsorption modelling showed a promising development in the application of quantitative predictive models in biological applications, nanomedicine, and environmental safety assessment of nanomaterials.

EcoFlex: A Multifunctional MoClo Kit for E. coli Synthetic Biology.

Development of advanced synthetic biology tools is always in demand since they act as a platform technology to enable rapid prototyping of biological constructs in a high-throughput manner. EcoFlex is a modular cloning (MoClo) kit for Escherichia coli and is based on the Golden Gate principles, whereby Type IIS restriction enzymes (BsaI, BsmBI, BpiI) are used to construct modular genetic elements (biological parts) in a bottom-up approach. Here, we describe a collection of plasmids that stores various biological parts including promoters, RBSs, terminators, ORFs, and destination vectors, each encoding compatible overhangs allowing hierarchical assembly into single transcription units or a full-length polycistronic operon or biosynthetic pathway. A secondary module cloning site is also available for pathway optimization, in order to limit library size if necessary. Here, we show the utility of EcoFlex using the violacein biosynthesis pathway as an example.

Effect of Sildenafil on Pre-Eclampsia-Like Mouse Model Induced By L-Name.

N(omega)-nitro-L-arginine methyl ester (L-NAME) decreases the vasodilator effect of nitric oxide (NO) and induces pre-eclampsia in mouse. Sildenafil inhibits the degradation of nitric oxide and increases vasodilation. This study aimed to determine the effects of sildenafil citrate on angiogenesis and oxidative stress at the maternal foetal interface on pre-eclampsia-like mouse model induced by L-NAME. Twenty pregnant mice were divided into four groups: (i) vehicle control; (ii) L-NAME; (iii) sildenafil; (4) L-NAME+sildenafil. L-NAME was administered from day 7 of pregnancy and sildenafil from day 8 until day 16; animals were euthanized on day 17. Placental and foetal sizes and weights were measured; lipid peroxide levels and catalase activity in placental homogenates were determined, and placental vascular endothelia were identified by lectin-histochemistry using BSA-I lectin. Western blot analysis was used to determine VEGF expression in placental homogenates. No changes were seen in placental and foetal development in mice with normal pregnancies treated with sildenafil. Treatments with L-NAME reduced significantly the placental weight and average height and decreased the percentage of the endothelial surface. These alterations may be mediated by the reduction of NO levels in trophoblastic cells, due to the inhibitory effect of L-NAME on nitric oxide synthase (NOS) synthesis. This effect was offset by the treatment with sildenafil, with an increase in the percentage of the endothelial surface. In conclusion, our results indicate that treatment with sildenafil on pre-eclampsia mouse model can be used without adverse effects on the concept and its use in the treatment of pre-eclampsia is promising. © 2015 Blackwell Verlag GmbH.

To examine the effects of a family-based teen dating abuse prevention program, Families for Safe Dates, primarily on outcomes related to testing the conceptual underpinnings of the program including (1) factors motivating and facilitating caregiver engagement in teen dating abuse prevention activities, and 2) risk factors for teen dating abuse, and secondarily on dating abuse behaviors. Families were recruited nationwide using listed telephone numbers. Caregivers and teens completed baseline and 3-month follow-up telephone interviews (n = 324). Families randomly allocated to treatment condition received the Families for Safe Dates program including six mailed activity booklets followed-up by health educator telephone calls. There were significant (<.05) treatment effects in hypothesized directions on most of the factors motivating and facilitating caregiver engagement in teen dating abuse prevention activities including caregiver perceived severity of dating abuse, response efficacy for preventing dating abuse, self-efficacy for talking about dating abuse, knowledge of dating abuse, acceptance of dating abuse, communication skills with the teen, and belief in the importance of involvement in their male (but not female) teen's dating. The latter effect was the only one moderated by sex of the teen. The targeted risk factor affected by the program was teen acceptance of dating abuse. Treatment was also significantly associated with less physical dating abuse victimization. Modifications to the program are warranted, but overall, the findings are very favorable for the first family-based teen dating abuse prevention program to be evaluated. Over a quarter of young women have experienced some form of violence within a dating relationship. The experience of dating violence is associated with problems in psychological functioning, including symptoms of anxiety and depression. However, not all women who experience dating violence exhibit anxious or depressive symptoms. One factor that may influence symptom expression is interpersonal style. In this study, we examined the main and moderating effects of dimensions of interpersonal style (dominance and warmth) on the association between dating violence and symptoms of anxiety and depression. Warmth exhibited a main effect on anxious and depressive symptoms over and above the effects of dating violence and other life stressors. Dominance moderated the association between dating violence and anxious and depressive symptoms. When levels of dating violence were high, women with higher levels of dominance reported fewer symptoms of anxiety and depression than women with lower dominance. These results indicated that whereas high warmth was associated with fewer symptoms of psychopathology generally, high dominance was a buffer against the effect of dating violence on symptoms more specifically. Directions for future research are discussed.

The purpose of the present study was to examine whether cognitive distortions (e.g., cognitive errors; negative views of self, world, and future) influence the association between dating violence and problematic substance use behaviors in a sample of psychiatrically hospitalized adolescents. Participants included 155 adolescents, aged 13-17 years, who had initiated dating. Adolescents completed measures of dating violence, substance-related problems (alcohol and marijuana), and cognitive distortions. Logistic regressions were conducted to examine the direct and interactive effects of dating violence exposure and cognitive distortions on likelihood of recent problematic substance use. Results suggested a main effect of dating violence on problematic alcohol and other drug use as well as an interactive effect of dating violence and cognitive distortions. Specifically, the relationship between dating violence and odds of substance-related problems was higher among those with greater (vs. fewer) cognitive distortions.
