EAD Metallurgical, Inc., operated a facility in Tonawanda, New York, in which it utilized Americium 241 (Am-241) for the production of foil sources for use in smoke detectors. EAD was in operation between 1977 and 1983. By 1983, the company started losing money, and decided to relocate to Mexico. Before closing down its Tonawanda operation, however, it was required by the New York State Department of Labor (DOL) to decontaminate its facility to limits specified by DOL. No records of discharges to the sewer system were kept during this decontamination effort. Unsuccessful decontamination efforts by several EAD employees and contractorsmore » left the building contaminated, in particular the concrete floors and walls. To determine the scope of work for the decontamination project, staff from the New York State Departments of Health (DOH) and Environmental Conservation (DEC) conducted a Characterization Survey of the facility in 1993. This survey identified contamination levels of Am-241 in excess of release limits throughout the building, in the soil outside the facility, in pipes for sewage and interior drainage, and in an 8 x 8 x 11 foot sump pit in the building. DOH issued a request for proposals in early 1994 for the decontamination and subsequent decommissioning of the former EAD building, and NES/IES Inc. (NES) was awarded the contract to perform the remediation. DOH`s assignment was to provide an on-site presence to insure the completion of all agreed upon tasks, according to the terms of the contract and work plans submitted by NES. Additionally, the DOH staff acted as a liaison between NES, DOH, DEC and DOL central offices to review, comment and approve all changes or modifications to NES`s approach to the decontamination efforts. The assigned staff was also responsible for conducting confirmatory sampling and surveys of all areas deemed releasable to DOL and DEC criteria by NES.« less

To examine palliative care clinicians' level of knowledge of the law regarding the use of the Deprivation of Liberty Safeguards (DoLS). Regional postal survey of palliative care clinicians working in hospices in the East of England, undertaken in April 2015. Clinicians' level of knowledge was assessed by their response to 7 factual questions. Data regarding self-reported levels of confidence in applying the Safeguards was collected, alongside information regarding the number of times they had used DoLS in practice. A free-text section invited additional comments from participants. There were 47 responses from 14 different organisations; a response rate of 68%. Respondents included consultants, specialty and associate specialists, registrars, nurses and social workers. Higher self-reported confidence and training in the use of DoLS was associated with higher factual knowledge. Consultants had the highest level of knowledge, training and experience. Doctors of other grades, nurses and social workers recorded less knowledge and experience and scored lower in the knowledge sections. The free-text comments revealed difficulty applying the Safeguards in practice, particularly among the consultant responses, based around several themes: insufficient guidance on how to use the Safeguards, process after death, uncertainty as to relevance to palliative care and delays in assessments. Clinicians working in palliative care have good levels of knowledge of the DoLS. Despite this concerns were raised, particularly by consultants; uncertainty as to when they should be used and the relevance of the Safeguards in clinical practice. Further guidance should be given to clinicians working in this specialty to ensure that clinical practice is both lawful and in the patients' best interests. During protein N-glycosylation, dolichyl pyrophosphate (Dol-P-P) is discharged in the lumenal monolayer of the endoplasmic reticulum (ER). Dol-P-P is then cleaved to Dol-P by Dol-P-P phosphatase (DPPase). Studies with the yeast mutant cwh8Delta, lacking DPPase activity, indicate that recycling of Dol-P produced by DPPase contributes significantly to the pool of Dol-P utilized for lipid intermediate biosynthesis on the cytoplasmic leaflet. Whether Dol-P formed in the lumen diffuses directly back to the cytoplasmic leaflet or is first dephosphorylated to dolichol has not been determined. Incubation of sealed ER vesicles from calf brain with acetyl-Asn-Tyr-Thr-NH(2), an N-glycosylatable peptide, to generate Dol-P-P in the lumenal monolayer produced corresponding increases in the rates of Man-P-Dol, Glc-P-Dol, and GlcNAc-P-P-Dol synthesis in the absence of CTP. No changes in dolichol kinase activity were observed. When streptolysin-O permeabilized CHO cells were incubated with an acceptor peptide, N-glycopeptide synthesis, requiring multiple cycles of the dolichol pathway, occurred in the absence of CTP. The results obtained with sealed microsomes and CHO cells indicate that Dol-P, formed from Dol-P-P, returns to the cytoplasmic leaflet where it can be reutilized for lipid intermediate biosynthesis, and dolichol kinase is not required for recycling. It is possible that the flip-flopping of the carrier lipid is mediated by a flippase, which would provide a mechanism for the recycling of Dol-P derived from Man-P-Dol-mediated reactions in N-, O-, and C-mannosylation of proteins, GPI anchor assembly, and the three Glc-P-Dol-mediated reactions in Glc(3)Man(9)GlcNAc(2)-P-P-Dol (DLO) biosynthesis.
