A hearing to reauthorize the Toxic Substances Control Act (TSCA) reviewed evidence that few new chemicals introduced into the consumer market are tested for toxicity and that no testing rules have been issued in the seven years of TSCA's existence in contrast to the testing record of some European countries. Plant managers in the CPI should proceed with caution in making some changes in chemical processes or waste disposal. A number of seemingly routine decisions, often made at the level of the plant manager and even below, can turn into costly compliance traps under the Toxic Substances Control Act (TSCA). TSCA is a federal statute designed to give the U.S. Environmental Protection Agency (EPA) information and control over commercial chemicals. Originally passed in 1976, the regulations it supports have been reworked at various times over the years. EPA is on the hunt for TSCA violators. In a December 1992 blitz,more » EPA assessed combined TSCA penalties of $9 million against 22 major chemical companies, including several of the top 20 international producers. According to a recent EPA press release, these actions are only the first in a series of planned enforcements initiatives. Nanomaterials exemplify a new class of emerging technologies that have significant economic and social value, pose uncertain health and environmental risks, and are entering the marketplace at a rapid pace. Effective regimes for regulating emerging technologies generate information about known or suspected hazards and draw on private sector expertise to guide managers' behavior toward risk reduction, even in the absence of clear evidence of harm. This paper considers the extent to which the federal Toxic Substances Control Act (TSCA) accomplishes those objectives. It offers the approach of the Massachusetts Toxics Use Reduction Act (TURA) as a possible supplement to TSCA, filling gaps in agency knowledge and private sector capacities. TURA is notable for its focus on chemicals use and hazard and its emphasis on strengthening firms' internal management systems. Given the current deadlock in Congressional efforts to modernize federal laws such as TSCA, the role of state laws like TURA merit attention. Absent definitive information about risk, a governance strategy that generates information and focuses management attention on reducing hazards is worth considering.

The Environmental Protection Agency is asserting regulatory control over the commercial production of genetically-engineered microorganisms, such as oil-eating bacteria, intended for release into the environment. EPA's legally-complex claim that such organisms are "new chemical substances," and thus subject to the Toxic Substances Control Act (TSCA), raises a challenge to the adequacy of regulation by the National Institute of Health's Recombinant DNA Advisory Committee (RAC) over the commercial and ecological implications of environmental release. Industry, which has been voluntarily adhering to the RAC guidelines, is reluctant to challenge application of the relatively lenient TSCA requirements but eager to see the legal uncertainties resolved.

A field study to evaluate the performance of three commercially available particulate matter (PM) continuous emission monitors (CEMs) was conducted in 1999-2000 at the US Department of Energy (DOE) Toxic Substances Control Act (TSCA) Incinerator. This study offers unique features that are believed to enhance the collective US experience with PM CEMs. The TSCA Incinerator is permitted to treat PCB-contaminated RCRA hazardous low-level radioactive wastes. The air pollution control system utilizes MACT control technology and is comprised of a rapid quench, venturi scrubber, packed bed scrubber, and two ionizing wet scrubbers in series, which create a saturated flue gas that must be conditioned by the CEMs prior to measurement. The incinerator routinely treats a wide variety of wastes including high and low BTU organic liquids, aqueous, and solid wastes. The various possible combinations for treating liquid and solid wastes may present a challenge in establishing a single, acceptable correlation relationship for individual CEMs. The effect of low-level radioactive material present in the waste is a unique site-specific factor not evaluated in previous tests. The three systems chosen for evaluation were two beta gauge devices and a light scattering device. The performance of the CEMs was evaluated using the requirements in draft Environmental Protection Agency (EPA) Performance Specification 11 (PS11) and Procedure 2. The results of Reference Method 5i stack tests for establishing statistical correlations between the reference method data and the CEMs responses are discussed.

Dr. Michael P. Wilson of UC Berkeley delivered his keynote address before the general assembly of the American Industrial Hygiene Conference and Exhibition (AIHce) in Portland, Oregon, in May 2011. Here, Dr. Wilson again discusses the political and economic drivers of occupational disease in the United States and proposes a role for AIHA in helping to highlight and resolve them. He proposes that until these underlying drivers are acknowledged and ameliorated, the toll of occupational disease will persist, despite the hard work of industrial hygienists in the workplace. Among these drivers, Dr. Wilson points to the decline of labor rights and unionization; economic inequality; economic insecurity; political resistance to public health protections for workers, notably the OSHA and NIOSH programs; and weaknesses in the Federal Toxic Substances Control Act of 1976 (TSCA). Of these, Dr. Wilson calls on the AIHA to participate in the historic effort to rewrite TSCA. He points to weaknesses in TSCA that have produced a chemicals market dominated by the function, price, and performance of chemicals, with little attention given to their health and environmental effects. Under these conditions, he argues, hazardous chemicals have remained economically competitive, and innovation in inherently safer chemicals-in green chemistry-has been held back by a lack of market transparency and public accountability in the industry. TSCA reform has the potential to shift the market toward green chemistry, with long-term implications for occupational disease prevention, industrial investment, and renewed energy in the industrial hygiene profession. Dr. Wilson proposes that, like previous legislative changes in the United States, TSCA reform is likely to occur in response to myriad social pressures, which include the emergence of the European Union's REACH regulation; recent chemicals policy actions in 18 U.S. states; growing support from downstream businesses; increasing public awareness; and a social movement that reaches across traditional boundaries. Dr. Wilson urges the AIHA to involve itself in this effort by building alliances with professional associations and other groups that share similar goals.

The use of quantitative structure-activity relationships (QSARs) for deriving the predicted no-effect concentration of discrete organic chemicals for the purposes of conducting a regulatory risk assessment in Europe and the United States is described. In the United States, under the Toxic Substances Control Act (TSCA), the TSCA Interagency Testing Committee and the U.S. Environmental Protection Agency (U.S. EPA) use SARs to estimate the hazards of existing and new chemicals. Within the Existing Substances Regulation in Europe, QSARs may be used for data evaluation, test strategy indications, and the identification and filling of data gaps. To illustrate where and when QSARs may be useful and when their use is more problematic, an example, methyl tertiary-butyl ether (MTBE), is given and the predicted and experimental data are compared. Improvements needed for new QSARs and tools for developing and using QSARs are discussed.

This study develops data on the acute aquatic toxicity of selected biodiesel fuels which may become subject to environmental effects test regulations under the US Toxic Substances Control Act (TSCA). The test substances are Rape Methyl Ester (RME), Rape Ethyl Ester (REE), Methyl Soyate (MS), a biodiesel mixture of 20% REE and 80% Diesel, a biodiesel mixture of 50% REE and diesel, and a reference substance of Phillips D-2 Reference Diesel. The test procedure follows the Daphnid Acute Toxicity Test outlined in 40 CFR {section} 797.1300 of the TSCA regulations. The ITC has been an independent advisory committee to the EPA Administrator since enactment of the Toxics Substances Control Act (TSCA) in 1976. The ITC identifies and coordinates U.S. Government data needs for TSCA-regulable chemicals, and makes recommendations to the Administrator for priority testing consideration. Chemicals recommended by the ITC are added to the TSCA Priority Testing List that is revised semi-annually in Reports to the Administrator. In recent Reports, the ITC added alkylphenols (APs) and ethoxylates to the Priority Testing List. About 500 million pounds are produced annually for industrial processing, cleaning and personal care products. APs have been detected in the tissues of fish from the Great Lakes, and one AP (nonylphenol) causes vitellogenin gene expression in trout hepatocytes. Numerous APs and ethoxylates were recommended by the ITC because data are needed on: (1) chemical composition, (2) environmental fate of parent chemicals and impurities, and (3) health and ecological effects (including toxicokinetics and endocrine-modulating effects). In response to the ITC's recommendations, two activities have ensued. First, the EPA promulgated rules requiring manufacturers, importers and processors of APs and ethoxylates to submit production and exposure reports, and unpublished health and safety studies, for review. Second, the Alkylphenols and Ethoxylates Panel of Chemical Manufacturers Association (CMA) and the ITC formed a Dialogue Group to discuss the data needs. Data needs and activities initiated by the Dialogue Group will be presented.

The challenges of regulating industrial chemicals remain unresolved in the United States. The Toxic Substances Control Act (TSCA) of 1976 was the first legislation to extend coverage to the regulation of industrial chemicals, both existing and newly registered. However, decisions related to both law and science that were made in passing this law inevitably rendered it ineffectual. Attempts to fix these shortcomings have not been successful. In light of the European Union's passage of innovative principles and requirements for chemical regulation, it is no longer possible to deny the opportunity and need for reform in US law and practice.

This request is submitted to seek interim approval to operate a Toxic Substances Control Act (TSCA) of 1976 chemical waste landfill for the disposal of polychlorinated biphenyl (PCB) waste. Operation of a chemical waste landfill for disposal of PCB waste is subject to the TSCA regulations of 40 CFR 761. Interim approval is requested for a period not to exceed 5 years from the date of approval. This request covers only the disposal of small 10 quantities of solid PCB waste contained in decommissioned, defueled submarine reactor compartments (SRC). In addition, the request applies only to disposal 12 of thismore » waste in Trench 94 of the 218-E-12B Burial Ground (Trench 94) in the 13 200 East Area of the US Department of Energy`s (DOE) Hanford Facility. Disposal of this waste will be conducted in accordance with the Compliance 15 Agreement (Appendix H) between the DOE Richland Operations Office (DOE-RL) and 16 the US Environmental Protection Agency (EPA), Region 10. During the 5-year interim approval period, the DOE-RL will submit an application seeking final 18 approval for operation of Trench 94 as a chemical waste landfill, including 19 any necessary waivers, and also will seek a final dangerous waste permit from 20 the Washington State Department of Ecology (Ecology) for disposal of lead 21 shielding contained in the SRCS.« less

This report summarizes the Interim Measure (IM) activities conducted at the Kennedy Space Center (KSC) Press Site ("the Press Site"). This facility has been designated as Solid Waste Management Unit 074 under KSC's Resource Conservation and Recovery Act Corrective Action program. The activities were completed as part of the Vehicle Assembly Building (VAB) Area Land Use Controls Implementation Plan (LUCIP) Elimination Project. The purpose of the VAB Area LUCIP Elimination Project was to delineate and remove soil affected with constituents of concern (COCs) that historically resulted in Land Use Controls (LUCs). The goal of the project was to eliminate the LUCs on soil. LUCs for groundwater were not addressed as part of the project and are not discussed in this report. This report is intended to meet the Florida Department of Environmental Protection (FDEP) Corrective Action Management Plan requirement as part of the KSC Hazardous and Solid Waste Amendments permit and the U.S. Environmental Protection Agency's (USEPA's) Toxic Substance Control Act (TSCA) self-implementing polychlorinated biphenyl (PCB) cleanup requirements of 40 Code of Federal Regulations (CFR) 761.61(a).

The registration of new chemicals under the Toxicological Substances Control Act (TSCA) and new pesticides under the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA) requires knowledge of the process science underlying the transformation of organic chemicals in natural ecosystems. The purpose of this presentation is to demonstrate how cheminformatics using chemical terms language in combination with the output of physicochemical property calculators can be employed to encode this knowledge and make it available to the appropriate decision makers. The encoded process science is realized through the execution of reaction libraries in simulators such as EPA’s Chemical Transformation Simulator (CTS). In support of the CTS, reaction libraries have or are currently being developed for a number of transformation processes including hydrolysis, abiotic reduction, photolysis and disinfection by-product formation. Examples of how the process science available in the peer-reviewed literature is being encoded will be presented. The registration of new chemicals under the Toxic Substances Control Act (TSCA) and new pesticides under the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA) requires knowledge of the process science underlying the transport and transformation of organic chemicals in natural ecosystems. The purpose of this presentation is to demonstrate how cheminformatics, using chemical terms language in combination with the output of physicochemical property calculators, can be employed to encode this knowledge and make it available to the appropriate decision makers. The encoded process science is realized through the execution of reaction libraries in simulators such as EPA’s Chemical Transformation Simulator (CTS). In support of the CTS, reaction libraries have, or are currently being developed for a number of transformation processes including hydrolysis, abiotic reduction, photolysis and disinfection by-product formation. Examples of how the process science in the peer-reviewed literature is being encoded will be presented. The purpose of this presentation is to demonstrate how cheminformatics, using chemical terms language in combination with the output of physicochemical property calculators, can be employed to encode this knowledge and make it available to the appropriate decision makers.

...The Agency is proposing to amend the Toxic Substances Control Act (TSCA) section 5(a) Significant New Use Rule (SNUR), for certain polybrominated diphenylethers (PBDEs) by: Designating processing of six PBDEs, or any combination of these chemical substances resulting from a chemical reaction, as a significant new use; designating manufacturing, importing, and processing of a seventh PBDE, decabromodiphenyl ether (decaBDE) for any use which is not ongoing after December 31, 2013, as a significant new use; and making inapplicable the article exemption for SNURs for this action. A person who intends to import or process any of the seven PBDEs included in the proposed SNUR, as part of an article for a significant new use would be required to notify EPA at least 90 days in advance to ensure that the Agency has an opportunity to review and, if necessary, restrict or prohibit a new use before it begins. EPA is also proposing a test rule under TSCA that would require any person who manufactures or processes commercial pentabromodiphenyl ether (c-pentaBDE), commercial octabromodiphenyl ether (c-octaBDE), or commercial decaBDE (c-decaBDE), including in articles, for any use after December 31, 2013, to conduct testing on their effects on health and the environment. EPA is proposing to designate all discontinued uses of PBDEs as significant new uses. Section 5 of the Toxic Substances Control Act (TSCA) requires that manufacturers and importers of new chemicals must submit a Premanufacture Notification (PMN) to the U.S. Environmental Protection Agency 90 days before they intend to commence manufacture or import. Certain information such as chemical identity, uses, etc., must be included in the notification. The submission of test data on the new substance, however, is not required, although any available health and environmental information must be provided. Nonetheless, over half of all PMNs submitted to the agency do not contain any test data; because PMN chemicals are new, no test data is generally available in the scientific literature. Given this situation, EPA has had to develop techniques for hazard assessment that can be used in the presence of limited test data. EPA's approach has been termed "structure-activity relationships" (SAR) and involves three major components: the first is critical evaluation and interpretation of available toxicity data on the chemical; the second component involves evaluation of test data available on analogous substances and/or potential metabolites; and the third component involves the use of mathematical expressions for biological activity known as "quantitative structure-activity relationships" (QSARs). At present, the use of QSARs is limited to estimating physical chemical properties, environmental toxicity, and bioconcentration factors. An important overarching element in EPA's approach is the experience and judgment of scientific assessors in interpreting and integrating the available data and information. Disposal costs for liquid PCB radioactive waste are among the highest of any category of regulated waste. The high cost is driven by the fact that disposal options are extremely limited. Toxic Substances Control Act (TSCA) regulations require most liquids with PCBs at concentration of {ge} 50 parts-per-million to be disposed by incineration or equivalent destructive treatment. Disposal fees can be as high as $200 per gallon. This figure does not include packaging and the cost to transport the waste to the disposal facility, or the waste generator's labor costs for managing the waste prior to shipment. Minimizing the generationmore » of liquid radioactive PCB waste is therefore a significant waste management challenge. PCB spill cleanups often generate large volumes of waste. That is because the removal of PCBs typically requires the liberal use of industrial solvents followed by a thorough rinsing process. In a nuclear facility, the cleanup process may be complicated by the presence of radiation and other occupational hazards. Building design and construction features, e.g., the presence of open grating or trenches, may also complicate cleanup. In addition to the technical challenges associated with spill cleanup, selection of the appropriate regulatory requirements and approach may be challenging. The TSCA regulations include three different sections relating to the cleanup of PCB contamination or spills. EPA has also promulgated a separate guidance policy for fresh PCB spills that is published as Subpart G of 40 CFR 761 although it is not an actual regulation. Applicability is based on the circumstances of each contamination event or situation. Other laws or regulations may also apply. Identification of the allowable regulatory options is important. Effective communication with stakeholders, particularly regulators, is just as important. Depending on the regulatory path that is taken, cleanup may necessitate the generation of large quantities of regulated waste. Allowable options must be evaluated carefully in order to reduce compliance risks, protect personnel, limit potential negative impacts on facility operations, and minimize the generation of wastes subject to TSCA. This paper will identify critical factors in selecting the appropriate TSCA regulatory path in order to minimize the generation of radioactive PCB waste and reduce negative impacts to facilities. The importance of communicating pertinent technical issues with facility staff, regulatory personnel, and subsequently, the public, will be discussed. Key points will be illustrated by examples from five former production reactors at the DOE Savannah River Site. In these reactors a polyurethane sealant was used to seal piping penetrations in the biological shield walls. During the intense neutron bombardment that occurred during reactor operation, the sealant broke down into a thick, viscous material that seeped out of the piping penetrations over adjacent equipment and walls. Some of the walls were painted with a PCB product. PCBs from the paint migrated into the degraded sealant, creating PCB 'spill areas' in some of these facilities. The regulatory cleanup approach selected for each facility was based on its operational status, e.g., active, inactive or undergoing decommissioning. The selected strategies served to greatly minimize the generation of radioactive liquid PCB waste. It is expected that this information would be useful to other DOE sites, DOD facilities, and commercial nuclear facilities constructed prior to the 1979 TSCA ban on most manufacturing and uses of PCBs.« less

Identifying new persistent and bioaccumulative organics among chemicals in commerce.

The goal of this study was to identify commercial chemicals that might be persistent and bioaccumulative (P&B) and that were not being considered in current Great Lakes, North American, and Arctic contaminant measurement programs. We combined the Canadian Domestic Substance List (DSL), a list of 3059 substances of "unknown or variable composition complex reaction products and biological materials" (UVCBs), and the U.S. Environmental Protection Agency (U.S. EPA) Toxic Substances Control Act (TSCA) Inventory Update Rule (IUR) database for years 1986, 1990, 1994, 1998, 2002, and 2006 yielding a database of 22263 commercial chemicals. From that list, 610 chemicals were identified by estimates from U.S EPA EPISuite software and using expert judgment. This study has yielded some interesting and probable P&B chemicals that should be considered for further study. Recent studies, following up our initial reports and presentations on this work, have confirmed the presence of many of these chemicals in the environment.

Under the Toxic Substances Control Act (TSCA), the Environmental Protection Agency (EPA) is required to perform new chemical reviews of engineered nanomaterials (ENMs) identified in pre-manufacture notices. However, environmental fate models developed for traditional contaminants are limited in their ability to simulate the environmental behavior of nanomaterials due to incomplete understanding and representation of the processes governing nanomaterial distribution in the environment and by scarce empirical data quantifying the interaction of nanomaterials with environmental surfaces. We have updated the Water Quality Analysis Simulation Program (WASP), version S, to incorporate nanomaterials as an explicitly simulated state variable. WASPS now has the capability to simulate nanomaterial fate and transport in surface waters and sediments using heteroaggregation, the kinetic process governing the attachment of nanomaterials to particles and subsequently ENM distribution in the aqueous and sediment phases. Unlike dissolved chemicals which use equilibrium partition coefficients, heteroaggregation consists of a particle collision rate and an attachment efficiency ( lXhet) that generally acts as a one direction process. Toxicologists are often called upon to assist in environmental, industrial, occupational and public health assessments. Accordingly, medical toxicologists may find it prudent to be aware of applicable federal toxicological regulations and reporting requirements and of the roles of relevant federal agencies. These regulations are numerous, complex, and have evolved and expanded over time, making it difficult for toxicologists to sustain a current knowledge base. This article reviews the pertinent federal toxicological reporting requirements with regard to the Toxic Substances Control Act (TSCA), the Atomic Energy Act (AEA), the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA), the Resource Conservation and Recovery Act (RCRA), the Clean Air Act, the Clean Water Act, the Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA), the Emergency Planning and Community Right to Know Act (EPCRA), the Occupational Safety and Health Act, the Department of Transportation, and information about the National Response Center. We reference internet-based government resources and offer direct links to applicable websites in an attempt to offer rapid and current sources of practical information. The format of the article is a series of hypothetical scenarios followed by commentary. Discussions of the Safe Drinking Water Act, the Food, Drug, and Cosmetic Act, and the Dietary Supplement Health and Education Act are beyond the scope of this paper. Three grab samples 6AP-98-1, 6AP-98-2 and 6AP-98-3 were taken from riser 1 of tank 241-AP-106 on May 28, 1998 and received by the 222-S Laboratory on May 28, 1998. Analyses were performed in accordance with the ''Compatability Grab Sampling and Analysis Plan'' (TSAP) (Sasaki, 1998) and the ''Data Quality Objectives for Tank Farms Waste Compatability Program (DQO). The analytical results are presented in the data summary report. No notification limits were exceeded. In this Final Rule, EPA is amending both the Asbestos Worker Protection Rule (WPR) and the Asbestos-in-Schools Rule. The WPR amendment protects State and local government employees from the health risks of exposure to asbestos to the same extent as private sector workers by adopting for these employees the Asbestos Standards of the Occupational Safety and Health Administration (OSHA). The WPR's coverage is extended to State and local government employees who are performing construction work, custodial work, and automotive brake and clutch repair work. This final rule cross-references the OSHA Asbestos Standards for Construction and for General Industry, so that future amendments to these OSHA standards are directly and equally effective for employees covered by the WPR. EPA also amends the Asbestos-in-Schools Rule to provide coverage under the WPR for employees of public local education agencies who perform operations, maintenance, and repair activities. EPA is issuing this final rule under section 6 of the Toxic Substances Control Act (TSCA).

Organophosphorus flame retardants (OPFRs) are widely used as additives in industrial and consumer products such as electrical and electronic products, furniture, plastics, textiles, and building/construction materials. Due to human exposure and potential health effects, OPFRs including tris(2-chloroethyl) phosphate (TCEP), tris(1-chloro-2-propyl) phosphate (TCPP), and tris(1,3-dichloro-2-propyl) phosphate (TDCPP) are EPA Action Plan chemicals for chemical assessments under the Toxic Substances Control Act (TSCA). This work investigated the sorption of these three compounds from the air to settled Arizona Test Dust (ATD) and house dust (HD) in a dual small chamber system. The OPFR exposed dust was analyzed to determine the sorption concentration and sorption rate of OPFRs on the dust. The effect of the composition of the dust on OPFR sorption was evaluated. The results showed that ATD and HD have varied sorption capacity for OPFRs from air. This work explores the relationship between OPFR concentrations in settled dust and air. The data can be used to determine partitioning of OPFRs between the gas phase and settled dust indoors and to inform strategies to reduce exposure and risk.

The Significant New Use Rule (SNUR) would require people to notify EPA at least 90 days before commencing the manufacture, import or processing of lead and lead compounds for new uses. Sludge from tank bottoms containing PCB`s, radioactivity and hazardous constituents are present in several tanks at one of the National Laboratories. Disposal of the material can proceed if the material is removed from TSCA regulations by decreasing the concentration of the PCB`s to {le}2 ppm. ON the bench scale, this sludge was treated by the DECHLOR/KGME{sup {trademark}} chemical dechlorination process. The levels of PCB`s were reduced to below 2 ppm, allowing the material to be managed outside the TSCA regulations. RUST believes that this is the first successful chemical dechlorination of a radioactive, RCRA listed, PCB bearing waste. A pilotmore » scale unit is available to provide on-site treatment of the remaining waste. Because of the small amounts of waste, treatment costs are high on a per unit volume. As a result of these high costs and other concerns the client is investigating potential non-treatment options of delisting the waste of obtaining a waiver. In the event that this particular waste cannot be delisted or a waiver is not granted, then dechlorination of the waste to remove it from TSCA regulations remains a viable option to allow the material to be disposed.« less

Under Section 5 of the Toxic Substances Control Act (TSCA), manufacturers must notify the US Environmental Protection Agency (EPA) 90 days before manufacturing, processing, or importing a new chemical substance. This is referred to as a premanufacture notice (PMN). The PMN must contain certain information including chemical identity, production volume, proposed uses, estimates of exposure and release, and any health or environmental test data that are available to the submitter. Because there is no explicit statutory authority that requires testing of new chemicals prior to their entry into the market, most PMNs are submitted with little or no data. Asmore » a result, EPA has developed special techniques for hazard assessment of PMN chemicals. These include (1) evaluation of available data on the chemical itself, (2) evaluation of data on analogues of the PMN, or evaluation of data on metabolites or analogues of metabolites of the PMN, (3) use of quantitative structure activity relationships (QSARs), and (4) knowledge and judgement of scientific assessors in the interpretation and integration of the information developed in the course of the assessment. This approach to evaluating potential hazards of new chemicals is used to identify those that are most in need of addition review of further testing. NaKnowBaseTM: The EPA Nanomaterials Research ...
The ability to predict the environmental and health implications of engineered nanomaterials is an important research priority due to the exponential rate at which nanotechnology is being incorporated into consumer, industrial and biomedical applications. To address this need and develop predictive capability, we have created the NaKnowbaseTM, which provides a platform for the curation and dissemination of EPA nanomaterials data to support functional assay development, hazard risk models and informatic analyses. To date, we have combined relevant physicochemical parameters from other organizations (e.g., OECD, NIST), with those requested for nanomaterial data submitted to EPA under the Toxic Substances Control Act (TSCA). Physiochemical characterization data were collated from >400 unique nanomaterials including metals, metal oxides, carbon-based and hybrid materials evaluated or synthesized by EPA researchers. We constructed parameter requirements and table structures for encoding research metadata, including experimental factors and measured response variables. As a proof of concept, we illustrate how SQL-based queries facilitate a range of interrogations including, for example, relationships between nanoparticle characteristics and environmental or toxicological endpoints. Toxicologists conducting safety assessments for either human or ecological health are responsible for generating data for possible adverse effects of a rapidly increasing number of substances. For example, the Registration, Evaluation, Authorisation and Restriction of Chemicals (REACH) program in Europe, and the recent reauthorization of the Toxic Substances Control Act (TSCA) in the United States (US) both explicitly mandate consideration of the potential toxicity of many thousands of chemicals that, in the past, would not have been assessed. There also is an increasing emphasis on understanding the effects of existing chemical mixtures on human health and the environment; for example in North America, the Great Lakes Restoration Initiative, sponsored by the Office of the President of the US, has identified complex mixtures of chemicals of emerging concern as one of the highest priority stressors in the lakes. Cheminformatics based tools, such as the Chemical Transformation Simulator under development in EPA’s Office of Research and Development, are being increasingly used to evaluate chemicals for their potential to degrade in the environment or be transformed through metabolism. Hydrolysis represents a major environmental degradation pathway; unfortunately, only a small fraction of hydrolysis rates for about 85,000 chemicals on the Toxic Substances Control Act (TSCA) inventory are in public domain, making it critical to develop in silico approaches to estimate hydrolysis rate constants. In this presentation, we compare three complementary approaches to estimate hydrolysis rates for carbamates, an important chemical class widely used in agriculture as pesticides, herbicides and fungicides. Fragment-based Quantitative Structure Activity Relationships (QSARs) using Hammett-Taft sigma constants are widely published and implemented for relatively simple functional groups such as carboxylic acid esters, phthalate esters, and organophosphate esters, and we extend these to carbamates. We also develop a pKa based model and a quantitative structure property relationship (QSPR) model, and evaluate them against measured rate constants using R square and root mean square (RMS) error. Chemical substances other than pesticides, drugs, and food additives are regulated by the U.S. Environmental Protection Agency (U.S. EPA) under the Toxic Substances Control Act (TSCA), but the United States does not require that new substances be tested automatically for such critical properties as biodegradability. The resulting lack of submitted data has fostered the development of estimation methods, and the BioWIN models for predicting biodegradability from chemical structure have played a prominent role in premanufacture notice (PMN) review. Until now, validation efforts have used only the Japanese Ministry of International Trade and Industry (MITI) test data and have not included all models. To assess BioWIN performance with PMN substances, we assembled a database of PMNs for which ready biodegradation data had been submitted over the period 1995 through 2001. The 305 PMN structures are highly varied and pose major challenges to chemical property estimation. Despite the variability of ready biodegradation tests, the use of at least six different test methods, and widely varying quality of submitted data, accuracy of four of six BioWIN models (MITI linear, MITI nonlinear, survey ultimate, survey primary) was in the 80+% range for predicting ready biodegradability. Greater accuracy (>90%) can be achieved by using model estimates only when the four models agree (true for 3/4 of the PMNs). The BioWIN linear and nonlinear probability models did not perform as well even when classification criteria were optimized. The results suggest that the MITI and survey BioWIN models are suitable for use in screening-level applications.
