  8. Subcritical Butane Extraction of Wheat Germ Oil and Its Deacidification by Molecular Distillation.

    Extraction and deacidification are important stages for wheat germ oil (WGO) production. Crude WGO was extracted using subcritical butane extraction (SBE) and compared with traditional solvent extraction (SE) and supercritical carbon dioxide extraction (SCE) based on the yield, chemical index and fatty acid profile. Furthermore, the effects of the molecular distillation temperature on the quality of WGO were also investigated in this study. Results indicated that WGO extracted by SBE has a higher yield of 9.10% and better quality; at the same time, its fatty acid composition has no significant difference compared with that of SE and SCE. The molecular distillation experiment showed that the acid value, peroxide value and p -anisidine value of WGO were reduced with the increase of the evaporation temperatures, and the contents of the active constituents of tocopherol, polyphenols and phytosterols are simultaneously decreased. Generally, the distillation temperature of 150 °C is an appropriate condition for WGO deacidification with the higher deacidification efficiency of 77.78% and the higher retention rate of active constituents.

  9. The Chemistry of Paper Preservation: Part 1. The Aging of Paper and Conservation Techniques

    This study provides an introduction to the problem of the aging of paper and the conservation techniques that are currently being employed in paper preservation. The chemical reactions that are responsible for the aging of paper are discussed with the conclusion that acid-catalyzed hydrolysis is the predominant mechanism for cellulose degradation and strength loss. A description and the chemistry of a number of mass deacidification methods are presented. The more viable deacidification methods include the DEZ, Wei T'o, FMC, Bookkeeper, Viennese, Book Preservation Associates, Sable and the Batelle processes. A summary of the literature on the evaluations of these processes is presented, and the benefits and limitations of the mass deacidification methods are discussed. Other conservation techniques such as paper strengthening, conservation bleaching, and pest control are briefly introduced, followed by an introduction to alkaline papermaking.

    The Library of Congress has patented a process to extend book life. It is called vapor phased deacidification, and involves the use of DEZ (diethyl zinc), a chemical vapor which neutralizes acid and deposits an alkaline reserve on book pages. As the process must be done in an airless environment, the library utilized Goddard Space Flight Center's vacuum chamber for deacidification. The chamber can treat 5,000 books at once, and a new facility is planned. The Library plans to license the technology to private companies; several universities are interested in the process.

  11. Changes of quality in the fruits of Prunus mume during deacidification by fermentation with Lactobacillus fermentium.

    The aim of this study was to investigate the changes of quality attributes of Prunus mume fruits during deacidification using the fermentation of Lactobacillus fermentium. Results of HPLC analysis showed that the sucrose and glucose were dominant sugars, and citric acid was dominant organic acids in P. mume fruits. The level of citric acid reaches 39.3 g/kg, and yet the sucrose and glucose content in the P. mume fruits was very lower, which were 2.16 and 0.66 g/L, respectively. After 8 d of fermentation, sugar and citric acid in the P. mume fruits was completely consumed, and the total phenolics, antioxidant activity (oxygen radical absorbance capacity value), and sarcocarp firmness retained 64.4%, 70.0%, 62.6%, respectively. Also, the viability counts of L. fermentium in fermentation broth increased slowly, which were near 8.0 lg CFU/mL after 8 d of fermentation at 30 °C. Overall, fermentation with L. fermentium can be applied in deacidification of P. mume fruits, and also the fermented P. mume fruits can meet the standard to be further processed into prune or sauces, and the fermentation broth of P. mume fruits with L. fermentium have a good prospect in the development of probiotic beverage. © 2015 Institute of Food Technologists®

  12. Optimization process condition for deacidification of palm oil by liquid-liquid extraction using NADES (Natural Deep Eutectic Solvent)

    One of many steps in palm oil refining process is deacidification which aims to separate free fatty acids and other compounds from the oil. The deacidification process was using a green solvent, known as NADES, that consisted of betaine monohydrate and propionic acid at molar ratio of 1:8. In this study, the process conditions were optimized using the response surface method (RSM) through central composite design in order to predict the maximum distribution coefficient of palmitic acid. The obtained regression equation of the basic model for optimization was: y = 0.717 + 0.003x1 + 0.043 x2 + 0.148x3 - 0.005 x1x1 - 0.030 x2x2 + 0.047 x3x3 - 0.008 x1x2 + 0.008 x1x3 + 0.033 x2x3. The independent variables are x1 ≡ temperature (40, 60, 80 °C), x2≡ amount of palmitic acid in the palm oil (2, 5, 8 %) and x3 ≡ mass ratios of oil to NADES (1:2, 1:1, 2:1). The optimum process condition found was temperature of 62.3°C, palmitic acid content of 8%, and NADES to palm oil mass ratio of 1:2, resulting in the maximum distribution coefficient of 0.96.

    The quantum yield of photosynthetic O2 exchange was measured in eight species of leaf succulents representative of both malic enzyme type and phosphoenolpyruvate carboxykinase type CAM plants. Measurements were made at 25°C and CO2 saturation using a leaf disc O2 electrode system, either during or after deacidification. The mean quantum yield was 0.095 ± 0.012 (sd) moles O2 per mole quanta, which compared with 0.094 ± 0.006 (sd) moles O2 per mole quanta for spinach leaf discs measured under the same conditions. There were no consistent differences in quantum yield between decarboxylation types or during different phases of CAM metabolism. On the basis of current notions of compartmentation of CAM biochemistry, our observations are interpreted to indicate that CO2 refixation is energetically independent of gluconeogenesis during deacidification.     Serendipity and experiment have been a frequent approach for the development of materials and methodologies used for a long time for either cleaning or consolidation of works of art. Recently, new perspectives have been opened by the application of materials science, colloid science, and interface science frameworks to conservation, generating a breakthrough in the development of innovative tools for the conservation and preservation of cultural heritage. This Article is an overview of the most recent contributions of colloid and materials science to the art conservation field, mainly focusing on the use of amphiphile-based fluids, gels, and alkaline earth metal hydroxide nanoparticles dispersions for the cleaning of pictorial surfaces, the consolidation of artistic substrates, and the deacidification of paper, canvas, and wood. Future possible directions for solving several conservation issues that still need to be faced are also highlighted.

    Alkaline earth metal hydroxide nanoparticles dispersions have demonstrated to be efficient for the preservation of cellulose-based artifacts, providing a stable neutral environment and, if in excess, turning into mild alkaline species. New formulations tailored for specific conservation issues have been recently obtained via a solvothermal reaction, starting from bulk metal, and short chain alcohols. Using this synthetic procedure, stable, and high concentrated calcium hydroxide nanoparticles dispersions can be obtained. The characterization of nanoparticles was carried out by dynamic light scattering, transmission electron microscopy and X-ray powder diffraction and showed that the dispersed systems are particularly suitable for the application on porous substrates. In a direct application of this technology, acidic paper and canvas samples were artificially aged after deacidification using calcium hydroxide nanoparticles dispersed in short chain alcohols. Cellulose viscosimetric polymerization degree (DPv), cellulose pyrolysis temperature, and samples' pH were evaluated upon the aging and in terms of protective action arising from the applied treatment. In particular, determinations of DPv clearly showed that the degradation of acidic paper and canvas samples proceeds at higher rates with respect to deacidified samples. These evidences were also confirmed by the thermogravimetric analysis of samples, in which the benefits due to the deacidification treatments are measured in terms of pyrolysis temperature of cellulose. These new formulations of nanoparticles dispersions expand the palette of available tools for the conservation of cellulose-based works of art, such as easel paintings, and manuscripts, potentially opening the way for the intervention on parchment and leather, whose preservation is a particularly challenging task.

    Herein, we report the influence of different combinations of initial concentration of acetic acid and ethanol on the removal of acetic acid from acidic wines by two commercial Saccharomyces cerevisiae strains S26 and S29. Both strains reduced the volatile acidity of an acidic wine (1.0 gl(-1) acetic acid and 11% (v/v) ethanol) by 78% and 48%, respectively. Acetic acid removal by strains S26 and S29 was associated with a decrease in ethanol concentration of 0.7 and 1.2% (v/v), respectively. Strain S26 revealed better removal efficiency due to its higher tolerance to stress factors imposed by acidic wines. Sulfur dioxide (SO(2)) in the concentration range 95-170 mg l(-1)inhibits the ability of both strains to reduce the volatile acidity of the acidic wine used under our experimental conditions. Therefore, deacidification should be carried out either in wines stabilized by filtration or in wines with SO(2)concentrations up to 70 mg l(-1). Deacidification of wines with the better performing strain S26 was associated with changes in the concentration of volatile compounds. The most pronounced increase was observed for isoamyl acetate (banana) and ethyl hexanoate (apple, pineapple), with an 18- and 25-fold increment, respectively, to values above the detection threshold. The acetaldehyde concentration of the deacidified wine was 2.3 times higher, and may have a detrimental effect on the wine aroma. Moreover, deacidification led to increased fatty acids concentration, but still within the range of values described for spontaneous fermentations, and with apparently no negative impact on the organoleptical properties.

    The food oil refinery process for deacidification is ususally conducted by the neutralization after degumming. In this study, commercialized resins will be used as adsorbents to remove the free fatty acid (FFA) in food oil without using any solvent. Applying this environmental friendly green process, the energy efficiency will be increased and the waste water will be reduced compared to the traditional process. The selected adsorbent can be reused which may reduce the process cost. Instead of using alkali neutralization, the proposed process may reduce the concern of food oil security. The commercial resins A26OH and IRA900Cl were compared as adsorbents to remove the FFA in deacidification for refinery of food oil without adding any alkali chemicals. This process will be conducted to remove the FFA form peanut oil in this study. Besides, this study will get the adsorption isotherms for one of the better sorbents of A26OH or IRA900Cl to remove FFA from peanut oil under 25, 35, and 45°C. The Langmuir and Freundlich isotherm models were compared to fit the experimental data. The obtained isotherm data is important for the adsorption system design.

    Acid catalysts were prepared by sulfonation of carbon materials obtained from the pyrolysis of sugar beet molasses, a cheap, viscous byproduct in the processing of sugar beets into sugar. Conditions for the pyrolysis of molasses (temperature and time) influenced catalyst performance; the best combination came from pyrolysis at low temperature (420 °C) for a relatively long time (8-15 h), which ensured better stability of the final material. The most effective molasses catalyst was highly active in the esterification of fatty acids with methanol (100 % yield after 3 h) and more active than common solid acidic catalysts in the transesterification of vegetable oils with 25-75 wt % of acid content (55-96 % yield after 8 h). A tandem process using a solid acid molasses catalyst and potassium hydroxide in methanol was developed to de-acidificate and transesterificate algal oils from Chlamydomonas reinhardtii, Nannochloropsis gaditana, and Phaeodactylum tricornutum, which contain high amounts of free fatty acids. The amount of catalyst required for the de-acidification step was influenced by the chemical composition of the algal oil, thus operational conditions were determined not only in relation to free fatty acids content in the oil, but according to the composition of the lipid extract of each algal species.   9. Degradation products from naturally aged paper leaves of a 16th-century-printed book: a spectrochemical study.

    In this work, we present a wide-range spectrochemical analysis of the degradation products from naturally aged paper. The samples obtained from wash waters used during the de-acidification treatment of leaves from a 16th-century-printed book were analysed through NMR, IR, Raman UV/Vis, EPR and X-ray fluorescence (XRF) spectroscopy and HPLC-MS and inductively coupled plasma (ICP) analysis. By these methods we also studied some of the previous samples treated by acidification (sample AP) and catalytic hydrogenation (sample HP). Crossing all the data, we obtained precise indications about the main functional groups occurring on the degraded, water-soluble cellulose oligomers. These results point out that the chromophores responsible for browning are conjugated carbonyl and carboxyl compounds. As a whole, we show that the analysis of wash waters, used in the usual conservation treatments of paper de-acidification, gives much valuable information about both the conservation state of the book and the degradation reactions occurring on the leaves, due to the huge amount of cellulose by-products contained in the samples. We propose therefore this procedure as a new very convenient general method to obtain precious and normally unavailable information on the cellulose degradation by-products from naturally aged paper.   10. A New Strategy to Refine Crude Indian Sardine Oil.

    Current work aims to develop a refining process for removing phospholipids, free fatty acids (FFA), and metal ions without affecting n-3 polyunsaturated fatty acid (n-3 PUFA) esters present in the crude Indian sardine oil. Sardine oil was subjected to degumming with various acids (orthophosphoric acid, acetic acid, and lactic acid), conventional and membrane assisted deacidification using various solvents (methanol, ethanol, propanol and butanol) and bleaching with bleaching agents (GAC, activated earth and bentonite) and all the process parameters were further optimized. Degumming with 5%(w/w) ortho phosphoric acid, two stage solvent extraction with methanol at 1:1 (w/w) in each stage and bleaching with 3% (w/w) activated charcoal loading, at 80ºC for 10 minutes resulted in the reduction of phospholipid content to 5.66 ppm from 612.66 ppm, FFA to 0.56% from 5.64% with the complete removal of iron and mercury. Under these conditions, the obtained bleached oil showed an enhancement of n-3 PUFA from 16.39 % (11.19 Eicosapentaenoic acid (EPA) + 5.20 Docosahexaenoic acid (DHA)) to 17.91 % (11.81 EPA + 6.1 DHA). Replacing conventional solvent extraction with membrane deacidification using microporous, hydrophobic polytetrafluoroethylene membrane (PTFE), resulted in a lesser solvent residue (0.25% (w/w)) in the deacidified oil. In view of lack of reports on refining of n-3 PUFA rich marine oils without concomitant loss of n-3 PUFA, this report is significant.

    The effects of temperature, O(2), and CO(2) on titratable acid content and on CO(2) exchange were measured in detached pineapple (Ananas comosus) leaves during the daily 15-hour light period. Comparative measurements were made in air and in CO(2)-free air. Increasing the leaf temperature from 20 to 35 C decreased the total CO(2) uptake in air and slightly increased the total CO(2) released into CO(2)-free air. Between 25 and 35 C, the activation energy for daily acid loss was near 12 kcal mol(-1), but at lower temperatures the activation energy was much greater.Increasing O(2) or decreasing the CO(2) concentration decreased the total CO(2) fixation in air, whereas the total CO(2) released in CO(2)-free air was increased. The total acid content remained constant at 20 C, but it decreased progressively with increasing temperature both in air and in CO(2)-free air. The total acid content at 30 C remained constant in 2% O(2) irrespective of CO(2) concentration. The total acid content decreased in 21 and 50% O(2) as the CO(2) increased from 0 to 300, and 540 mul/l of CO(2). The data indicate that photorespiration is present in pineapple. The lack of acid loss in 2% O(2) suggests that light deacidification is dependent upon respiration and that higher O(2) concentrations are required to saturate deacidification.

    Current pharmacological treatments for lung cancer show very poor clinical outcomes, therefore, the development of novel anticancer agents with innovative mechanisms of action is urgently needed. Cancer cells have a reversed pH gradient compared to normal cells, which favours cancer progression by promoting proliferation, metabolic adaptation and evasion of apoptosis. In this regard, the use of ionophores to modulate intracellular pH appears as a promising new therapeutic strategy. Indeed, there is a growing body of evidence supporting ionophores as novel antitumour drugs. Despite this, little is known about the implications of pH deregulation and homeostasis imbalance triggered by ionophores at the cellular level. In this work, we deeply analyse for the first time the anticancer effects of tambjamine analogues, a group of highly effective anion selective ionophores, at the cellular and molecular levels. First, their effects on cell viability were determined in several lung cancer cell lines and patient-derived cancer stem cells, demonstrating their potent cytotoxic effects. Then, we have characterized the induced lysosomal deacidification, as well as, the massive cytoplasmic vacuolization observed after treatment with these compounds, which is consistent with mitochondrial swelling. Finally, the activation of several proteins involved in stress response, autophagy and apoptosis was also detected, although they were not significantly responsible for the cell death induced. Altogether, these evidences suggest that tambjamine analogues provoke an imbalance in cellular ion homeostasis that triggers mitochondrial dysfunction and lysosomal deacidification leading to a potent cytotoxic effect through necrosis in lung cancer cell lines and cancer stem cells.     The preservation of manuscripts and archive materials is a serious problem for librarians and restorers. Paper manuscript is subjected to numerous degradation factors affecting their conservation state. This research represents an attempt to evaluate the conservation restoration process applied in Moroccan libraries, especially the alkaline treatment for strengthening weakened paper. In this study, we focused on six samples of degraded and restored paper taken from three different Moroccan manuscripts aged 150, 200 and 800 years. In addition, the Japanese paper used in restoration has been characterized. A modern paper was also analyzed as reference. A three-step analytical methodology based on infrared spectroscopy (ATR-FTIR), X-ray diffraction (XRD) and scanning electron microscopy coupled to energy dispersive spectrometry (SEM-EDS) analysis was developed before and after restoration in order to determine the effect of the consolidation treatment on the paper structure. The results obtained by XRD and ATR-FTIR disclosed the presence of barium sulfate (BaSO4) in all restored paper manuscripts. The presence of calcium carbonate (CaCO3) in all considered samples was confirmed by FTIR spectroscopy. The application of de-acidification treatment causes significant changes connected with the increase of intensity mostly in the region 1426 cm(-1), assigned to the asymmetric and symmetric CO stretching mode of calcite, indicating the effectiveness of de-acidification procedure proved by the rise of the alkaline reserve content allowing the long term preservation of paper. Observations performed by SEM magnify the typical paper morphology and the structure of fibbers, highlighting the effect of the restoration process, manifested by the reduction of impurities.   14. Reduction of volatile acidity of acidic wines by immobilized Saccharomyces cerevisiae cells.

    Excessive volatile acidity in wines is a major problem and is still prevalent because available solutions are nevertheless unsatisfactory, namely, blending the filter-sterilized acidic wine with other wines of lower volatile acidity or using reverse osmosis. We have previously explored the use of an empirical biological deacidification procedure to lower the acetic acid content of wines. This winemaker's enological practice, which consists in refermentation associated with acetic acid consumption by yeasts, is performed by mixing the acidic wine with freshly crushed grapes, musts, or marc from a finished wine fermentation. We have shown that the commercial strain Saccharomyces cerevisiae S26 is able to decrease the volatile acidity of acidic wines with a volatile acidity higher than 1.44 g L(-1) acetic acid, with no detrimental impact on wine aroma. In this study, we aimed to optimize the immobilization of S26 cells in alginate beads for the bioreduction of volatile acidity of acidic wines. We found that S26 cells immobilized in double-layer alginate-chitosan beads could reduce the volatile acidity of an acidic wine (1.1 g L(-1) acetic acid, 12.5 % (v/v) ethanol, pH 3.12) by 28 and 62 % within 72 and 168 h, respectively, associated with a slight decrease in ethanol concentration (0.7 %). Similar volatile acidity removal efficiencies were obtained in medium with high glucose concentration (20 % w/v), indicating that this process may also be useful in the deacidification of grape musts. We, therefore, show that immobilized S. cerevisiae S26 cells in double-layer beads are an efficient alternative to improve the quality of wines with excessive volatile acidity.

  15. A Multi-Analytical Approach for the Evaluation of the Efficiency of the Conservation-Restoration Treatment of Moroccan Historical Manuscripts Dating to the 16th, 17th, and 18th Centuries.

    The most critical steps during the conservation-restoration treatment applied in Moroccan libraries are the deacidification using immersion in a saturated aqueous calcium hydroxide (Ca(OH)2) solution and the consolidation of degraded manuscripts using Japanese paper. The present study aims to assess the efficiency of this restoration method using a multi-analytical approach. For this purpose, three ancient Arabic Moroccan manuscript papers dating back to the 16th, 17th, and 18th centuries were investigated to characterize the paper support and make a comparative study between pre-restoration and post-restoration states. Three structural and molecular characterization techniques including solid-state nuclear magnetic resonance spectroscopy on (13)C with cross-polarization and magic-angle spinning nuclear magnetic resonance ((13)C CP-MAS NMR), attenuated total reflectance Fourier transform infrared spectroscopy (ATR FT-IR), and X-ray diffraction (XRD) were used to elucidate the cellulose main features, to identify the inorganic composition of the papers, and to study the crystallinity of the samples. Inductively coupled plasma atomic emission spectrometry (ICP-AES) allowed us to obtain a qualitative and quantitative characterization of the mineral fillers used in the manufacturing of the papers. Scanning electron microscopy coupled to energy dispersive spectrometry (SEM-EDS) ascertained the state of conservation of the different papers and helped us to study the elemental composition of the samples. After restoration, it was shown that the deacidification improved the stability of papers by providing an important alkaline buffer, as demonstrated using FT-IR and energy dispersive spectrometry (EDS) results. However, XRD and ICP-AES did not confirm the pertinence of the treatment for all samples because of the unequal distribution of Ca on the paper surface during the restoration. The consolidation process was studied using SEM analysis; its effectiveness in restoring torn areas was found to be significant.

    Books produced since 1850, held in the Library of Congress, have a very high acid content, hence a shorter life expectancy than prior-published volumes. A means of "deacidifying" books on a large, economically-viable scale is of great interest, particularly to the research library community, which stores millions of valuable, often irreplaceable documents. The General Electric vacuum chamber used in deacidification tests serves a dual purpose. It creates an environment in which DEZ can do its job without presence of oxygen and also dries the books. After the drying phase, DEZ is introduced to the chamber as a paper-penetrating vapor. The complete process typically takes eight days, four days for vacuum drying and four days of book exposure to DEZ. Accelerated aging tests showed that the process can extend paper life almost fourfold, even on color illustrations.

    A series of 12 N -substituted sophoridinamine derivatives were synthesized and evaluated for their cytotoxic activities in human HepG2 hepatoma cells. Structure-activity relationship revealed that introduction of a suitable arylidene or arylethyl at the N '-end could greatly enhance antiproliferation potency. Among them, compound 6b possessing a N '-trimethoxyphenyl methylene exhibited potent antiproliferation effect against three human tumor cell lines including HepG2, leukemia (K562), and breast cancer (HMLE), with IC 50 between 0.55 and 1.7 μM. The underlying mechanism of 6b against tumor cells is to block autophagic flux, mainly through neutralizing lysosomal acidity. Our results indicated that compound 6b is a potent lysosomal deacidification agent and is accordingly able to block autophagic flux and inhibit tumor cell growth.

    Membrane processing technology (MPT) is increasingly used nowadays in a wide range of applications (demineralization, desalination, stabilization, separation, deacidification, reduction of microbial load, purification, etc.) in food industries. The most frequently applied techniques are electrodialysis (ED), reverse osmosis (RO), nanofiltration (NF), ultrafiltration (UF), and microfiltration (MF). Several membrane characteristics, such as pore size, flow properties, and the applied hydraulic pressure mainly determine membranes' potential uses. In this review paper the basic membrane techniques, their potential applications in a large number of fields and products towards the food industry, the main advantages and disadvantages of these methods, fouling phenomena as well as their effects on the organoleptic, qualitative, and nutritional value of foods are synoptically described. Some representative examples of traditional and modern membrane applications both in tabular and figural form are also provided.

  19. Selected Schizosaccharomyces pombe Strains Have Characteristics That Are Beneficial for Winemaking

    At present, wine is generally produced using Saccharomyces yeast followed by Oenococus bacteria to complete malolactic fermentation. This method has some unsolved problems, such as the management of highly acidic musts and the production of potentially toxic products including biogenic amines and ethyl carbamate. Here we explore the potential of the fission yeast Schizosaccharomyces pombe to solve these problems. We characterise an extensive worldwide collection of S. pombe strains according to classic biochemical parameters of oenological interest. We identify three genetically different S. pombe strains that appear suitable for winemaking. These strains compare favourably to standard Saccharomyces cerevisiae winemaking strains, in that they perform effective malic acid deacidification and significantly reduce levels of biogenic amines and ethyl carbamate precursors without the need for any secondary bacterial malolactic fermentation. These findings indicate that the use of certain S. pombe strains could be advantageous for winemaking in regions where malic acid is problematic, and these strains also show superior performance with respect to food safety.     As part of a model-data investigation project, results of several water treatment studies were compared with hydrochemical models. The models used comparable hydrogeological reaction types that occur within aquifers and water treatment processes. They were tested on 3 different examples of water softening, de-nitrification and iron removal. Comparison of simulated and measured water chemical dynamics showed good agreement. In addition to mixing and formation of complexes, de-acidification and de-carbonisation processes were reproduced in the first example. The second example investigated de-nitrification in a straw filter and in a water plant filter with subsequent aeration. The third example showed iron removal where reactions with partially combusted dolomite were simulated with a computer model. All simulations showed good agreement with the observed data. The models have the advantage of yielding parameter results that are difficult to measure. This includes nitrogen gas release and the content of reacted and degradable organic substances. These tools may help to provide better insights into water treatment reactions.

    Vesicle fusion governs many important biological processes, and imbalances in the regulation of membrane fusion can lead to a variety of diseases such as diabetes and neurological disorders. Here we show that the Vibrio parahaemolyticus effector protein VopQ is a potent inhibitor of membrane fusion based on an in vitro yeast vacuole fusion model. Previously, we demonstrated that VopQ binds to the Vo domain of the conserved V-type H+-ATPase (V-ATPase) found on acidic compartments such as the yeast vacuole. VopQ forms a nonspecific, voltage-gated membrane channel of 18 Å resulting in neutralization of these compartments. We now present data showing that VopQ inhibits yeast vacuole fusion. Furthermore, we identified a unique mutation in VopQ that delineates its two functions, deacidification and inhibition of membrane fusion. The use of VopQ as a membrane fusion inhibitor in this manner now provides convincing evidence that vacuole fusion occurs independently of luminal acidification in vitro. PMID:25453092

  3. Mango kernel fat fractions as potential healthy food ingredients: A review.

    Mango kernel fat (MKF) has been reported to have high functional and nutritional potential. However, its application in food industry has not been fully explored or developed. In this review, the chemical compositions, physical properties and potential health benefits of MKF are described. MKF is a unique fat consisting of 28.9-65.0% of 1,3-distearoyl-2-oleoyl-glycerol with excellent oxidative stability index (58.8-85.2 h at 110 °C), making the fat and its fractions suitable for use as high-value added food ingredients such as cocoa butter alternatives, trans-free shortenings, and a source of natural antioxidants (e.g., sterol, tocopherol and squalene). Unfortunately, the long period of dehydration of mango kernels at hot temperature results in the hydrolysis of triacylglycerols. The high levels of hydrolysates (mainly free fatty acids and diacylglycerols) limit the application of MKF in manufacturing these food ingredients. It is suggested that the physico-chemical and functional properties of MKF could be further improved through moderated refining (e.g., degumming and physical deacidification), fractionation, and interesterification.

    Hollow halloysite nanotubes have been used as nanocontainers for loading and for the triggered release of calcium hydroxide for paper preservation. A strategy for placing end-stoppers into the tubular nanocontainer is proposed and the sustained release from the cavity is reported. The incorporation of Ca(OH)₂ into the nanotube lumen, as demonstrated using transmission electron microscopy (TEM) imaging and Energy Dispersive X-ray (EDX) mapping, retards the carbonatation, delaying the reaction with CO₂ gas. This effect can be further controlled by placing the end-stoppers. The obtained material is tested for paper deacidification. We prove that adding halloysite filled with Ca(OH)₂ to paper can reduce the impact of acid exposure on both the mechanical performance and pH alteration. The end-stoppers have a double effect: they preserve the calcium hydroxide from carbonation, and they prevent from the formation of highly basic pH and trigger the response to acid exposure minimizing the pH drop-down. These features are promising for a composite nanoadditive in the smart protection of cellulose-based materials.

  7. Optimizing anti-coking abilities of zeolites by ethylene diamine tetraacetie acid modification on catalytic fast pyrolysis of corn stalk

    In order to minimize coke yield during biomass catalytic fast pyrolysis (CFP) process, ethylene diamine tetraacetie acid (EDTA) chemical modification method is carried out to selectively remove the external framework aluminum of HZSM-5 catalyst. X-ray diffraction (XRD), nitrogen (N2)-adsorption and ammonia-temperature programmed desorption (NH3-TPD) techniques are employed to investigate the porosity and acidity characteristics of original and modified HZSM-5 samples. Py-GC/MS and thermo-gravimetric analyzer (TGA) experiments are further conducted to explore the catalytic effect of modified HZSM-5 samples on biomass CFP and to verify the positive effect on coke reduction. Results show that EDTA treatment does not damage the crystal structure of HZSM-5 zeolites, but leads to a slight increase of pore volume and pore size. Meanwhile, the elimination of the strong acid peak indicates the dealumination of outer surface of HZSM-5 zeolites. Treatment time of 2 h (labeled EDTA-2H) is optimal for acid removal and hydrocarbon formation. Among all modified catalysts, EDTA-2H performs the best for deacidification and can obviously increase the yields of positive chemical compositions in pyrolysis products. Besides, EDTA modification can improve the anti-coking properties of HZSM-5 zeolites, and EDTA-2H gives rise to the lowest coke yield.

  8. Physicochemical, Antioxidant and Sensory Quality of Brazilian Blueberry Wine.

    Currently, Rio Grande do Sul state is the main producer of blueberry in Brazil. Practically all production is commercialized in fresh state and only a small portion is subject to processing. The blueberry wine making process is an alternative to expand the beverage industry and offers to the consumer a value-added product as well as a new market for Brazilian blueberry producers. The objectives of this study were to produce wines from blueberries and to evaluate the effect of deacidification (with calcium carbonate) and chaptalization (with glucose syrup or sucrose) on physicochemical characteristics, antioxidant content, and sensory parameters. Samples were analyzed for total soluble solids, pH, total titratable acidity, total sugar content, alcohol content, monomeric and total anthocyanin, total flavonols, total phenolic compounds and antioxidant activity by DPPH and FRAP methods. The use of calcium carbonate caused a reduction in total titratable acidity, while the use of glucose syrup resulted in wines with low alcohol content. The blueberries wine from Climax and Aliceblue cultivars had higher content of anthocyanin when produced with glucose syrup. The use of calcium carbonate and glucose syrup also provided wines more appreciated by tasters in relation to color. With regard to flavor, George and Aliceblue were the cultivars with lower preference under the control treatments (without carbonate and sugar). The presence of phenolic compounds may have provided a positive influence on wine flavor, once the more preferred wines presented the greater phenolic content.

  9. Operating and environmental performances of commercial-scale waste gasification and melting technology.

    Gasification technologies for waste processing are receiving increased interest. A lot of gasification technologies, including gasification and melting, have been developed in Japan and Europe. However, the flue gas and heavy metal behaviors have not been widely reported, even though those of grate furnaces have been reported. This article reports flue gas components of gasification and melting technology in different flue gas treatment systems. Hydrogen chloride concentrations at the inlet of the bag filter ranged between 171 and 180 mg Nm(-3) owing to de-acidification by limestone injection to the gasifier. More than 97.8% of hydrogen chlorides were removed by a bag filter in both of the flue gas treatment systems investigated. Sulfur dioxide concentrations at the inlet of the baghouse were 4.8 mg Nm(-3) and 12.7 mg Nm(-3), respectively. Nitrogen oxides are highly decomposed by a selective catalytic reduction system. Owing to the low regenerations of polychlorinated dibenzo-p-dioxins and furans, and the selective catalytic reduction system, the concentrations of polychlorinated dibenzo-p-dioxins and furans at the stacks were significantly lower without activated carbon injection. More than 99% of chlorine is distributed in fly ash. Low-boiling-point heavy metals, such as lead and zinc, are distributed in fly ash at rates of 97.6% and 96.5%, respectively. Most high-boiling-point heavy metals, such as iron and copper, are distributed in metal. It is also clarified that the slag is stable and contains few harmful heavy metals, such as lead. The heavy metal distribution behaviors are almost the same regardless of the compositions of the processed waste. These results indicate that the gasification of municipal solid waste constitutes an ideal approach to environmental conservation and resource recycling.

Black-Right-Pointing-Pointer Aggregates indicate tight spatial relationship between minerals and microorganisms. - Abstract: Microbial community diversity in two thermophilic laboratory-scale and three full-scale anaerobic co-digesters was analysed by genetic profiling based on PCR-amplified partial 16S rRNA genes. In parallel operated laboratory reactors a stepwise increase of the organic loading rate (OLR) resulted in a decrease of methane production and an accumulation of volatile fatty acids (VFAs). However, almost threefold different OLRs were necessary to inhibit themore » gas production in the reactors. During stable reactor performance, no significant differences in the bacterial community structures were detected, except for in the archaeal communities. Sequencing of archaeal PCR products revealed a dominance of the acetoclastic methanogen Methanosarcina thermophila, while hydrogenotrophic methanogens were of minor importance and differed additionally in their abundance between reactors. As a consequence of the perturbation, changes in bacterial and archaeal populations were observed. After organic overload, hydrogenotrophic methanogens (Methanospirillum hungatei and Methanoculleus receptaculi) became more dominant, especially in the reactor attributed by a higher OLR capacity. In addition, aggregates composed of mineral and organic layers formed during organic overload and indicated tight spatial relationships between minerals and microbial processes that may support de-acidification processes in over-acidified sludge. Comparative analyses of mesophilic stationary phase full-scale reactors additionally indicated a correlation between the diversity of methanogens and the VFA concentration combined with the methane yield. This study demonstrates that the coexistence of two types of methanogens, i.e. hydrogenotrophic and acetoclastic methanogens is necessary to respond successfully to perturbation and leads to stable process performance.« less

  11. Lactic acid bacteria in the quality improvement and depreciation of wine.

    The winemaking process includes two main steps: lactic acid bacteria are responsible for the malolactic fermentation which follows the alcoholic fermentation by yeasts. Both types of microorganisms are present on grapes and on cellar equipment. Yeasts are better adapted to growth in grape must than lactic acid bacteria, so the alcoholic fermentation starts quickly. In must, up to ten lactic acid bacteria species can be identified. They belong to the Lactobacillus, Pediococcus, Leuconostoc and Oenococcus genera. Throughout alcoholic fermentation, a natural selection occurs and finally the dominant species is O. oeni, due to interactions between yeasts and bacteria and between bacteria themselves. After bacterial growth, when the population is over 10(6) CFU/ml, malolactic transformation is the obvious change in wine composition. However, many other substrates can be metabolized. Some like remaining sugars and citric acid are always assimilated by lactic acid bacteria, thus providing them with energy and carbon. Other substrates such as some amino acids may be used following pathways restricted to strains carrying the adequate enzymes. Some strains can also produce exopolysaccharides. All these transformations greatly influence the sensory and hygienic quality of wine. Malic acid transformation is encouraged because it induces deacidification. Diacetyl produced from citric acid is also helpful to some extent. Sensory analyses show that many other reactions change the aromas and make malolactic fermentation beneficial, but they are as yet unknown. On the contrary, an excess of acetic acid, the synthesis of glucane, biogenic amines and precursors of ethylcarbamate are undesirable. Fortunately, lactic acid bacteria normally multiply in dry wines; moreover some of these activities are not widespread. Moreover, the most striking trait of wine lactic acid bacteria is their capacity to adapt to a hostile environment. The mechanisms for this are not yet completely elucidated. Molecular biology has provided some explanations for the behaviour and the metabolism of bacteria in wine. New tools are now available to detect the presence of desirable and undesirable strains. Even if much remains unknown, winemakers and oenologists can nowadays better control the process. By acting upon the diverse microflora and grape musts, they are more able to produce healthy and pleasant wines.

  12. Characterization of Extreme Deposition of Air Pollutants in MT. Mitchell State Park: Potential for Forest Decline and Opportunity for Cloud Deacidification

    The decline of forests has long been attributed to various natural (e.g. drought), man-made (e.g. logging), and perhaps, combinations of these (eg. fires caused by loggers) causes. A new type of forest decline (attributed to the deposition of air pollutants and other natural causes) has become apparent at high elevation sites in western Europe and North America; especially for above cloudbase forests like those in the Mt. Mitchell State Park. Investigations of air pollutant deposition are plentiful and laboratory studies have shown extreme deposition of these pollutants to be potentially harmful to forests. However, no field study has concentrated on these events. The primary objective of this study is to characterize (i.e., meterologically, microphysically, chemically) extreme episodes of air pollutant deposition. This study defines extreme aqueous events as having a pH < 3.1. pH's of this order are known to reduce laboratory tree growth depending on their age and species. On the average, one out of three aqueous events, sampled in the park during the 1986-1988 growing seasons (mid-May through mid-September), was extreme. Their occurrence over time may lead to the death of infant and 'old' trees, and to the reduced vigor of trees in their prime, as a result of triggering the decline mechanisms of these trees. These events usually last ~ 4.0 h, form during extended periods of high atmospheric pressure, have a liquid water content of ~ 0.10 gm^{-3}, and near typical cloud droplet sizes (~ 8.0 μm). Extreme aqueous events deposit most of their acid at their end. The deposition from the infrequent occurrences of very high ozone ( >=q100 ppb) and sulfur dioxide (>=q 5 ppb) concentrations in conjunction with these cloud events may be even more detrimental to the canopy, then that by extreme aqueous events alone. The physical characteristics of these combined events appear to include those of mature, precipitating clouds. Their occurrence may provide a clue as to how very low pH clouds might be deacidified. That is, base gases (eg. ammonia) locally introduced into such clouds at the proper time may render them harmless upon impact with the forest canopy, and beneficial to regional water supply users.

  13. High temperature abatement of acid gases from waste incineration. Part II: Comparative life cycle assessment study.

    The performances of a new dolomitic sorbent, named Depurcal®MG, to be directly injected at high temperature in the combustion chamber of Waste-To-Energy (WTE) plants as a preliminary stage of deacidification, were experimentally tested during full-scale commercial operation. Results of the experimentations were promising, and have been extensively described in Biganzoli et al. (2014). This paper reports the Life Cycle Assessment (LCA) study performed to compare the traditional operation of the plants, based on the sole sodium bicarbonate feeding at low temperature, with the new one, where the dolomitic sorbent is injected at high temperature. In the latter the sodium bicarbonate is still used, but at lower rate because of the decreased load of acid gases entering the flue gas treatment line. The major goal of the LCA was to make sure that a burden shifting was not taking place somewhere in the life cycle stages, as it might be the case when a new material is used in substitution of another one. According to the comparative approach, only the processes which differ between the two operational modes were included in the system boundaries. They are the production of the two reactants and the treatment of the corresponding solid residues arising from the neutralisation of acid gases. The additional CO2 emission at the stack of the WTE plant due to the activation of the sodium bicarbonate was also included in the calculation. Data used in the modelling of the foreground system are primary, derived from the experimental tests described in Biganzoli et al. (2014) and from the dolomitic sorbent production plant. The results of the LCA show minor changes in the potential impacts between the two operational modes of the plants. These differences are for 8 impact categories in favour of the new operational mode based on the addition of the dolomitic sorbent, and for 7 impact categories in favour of the traditional operation. A final evaluation was conducted on the potential role of the dolomitic sorbent in enhancing the electric energy production efficiency of the plant, thanks to the better cleaning of the heat exchange surface that can be achieved. If such improvement is accounted for, all the potential impacts are considerably decreased (e.g. the Climate change by 28%), and in the comparison with the traditional operation 17 impact categories out of 19 are reduced.   14. Non-catalytic alcoholysis process for production of biodiesel fuel by using bubble column reactor

    Biodiesel fuel is a replacement for diesel as a fuel produced from biomass resources. It is usually defined as a fatty acid methyl ester (FAME) derived from vegetable oil or animal fat. In European countries, such as Germany and France, biodiesel fuel is commercially produced mainly from rapeseed oil, whereas in the United States and Argentina, soybean oil is more frequently used. In many other countries such as Japan and countries in Southeast Asia, lipids that cannot be used as a food source could be more suitable materials for the production of biodiesel fuel because its production from edible oils could result in an increase in the price of edible oils, thereby increasing the cost of some foodstuffs. Therefore, used edible oil, lipids contained in waste effluent from the oil milling process, byproducts from oil refining process and crude oils from industrial crops such as jatropha could be more promising materials in these countries. The materials available in Japan and Southeast Asia for the production of biodiesel fuel have common characteristics; they contain considerable amount of impurities and are high in free fatty acids (FFA). Superheated methanol vapor (SMV) reactor might be a promising method for biodiesel fuel production utilizing oil feedstock containing FFA such as waste vegetable oil and crude vegetable oil. In the conventional method using alkaline catalyst, FFA contained in waste vegetable oil is known to react with alkaline catalyst such as NaOH and KOH generating saponification products and to inactivate it. Therefore, the FFA needs to be removed from the feedstock prior to the reaction. Removal of the alkaline catalyst after the reaction is also required. In the case of the SMV reactor, the processes for removing FFA prior to the reaction and catalyst after the reaction can be omitted because it requires no catalyst. Nevertheless, detailed study on the productivity of biodiesel fuel produced from waste vegetable oils and other non-edible lipids by use of the SMV reactor has not been examined yet. Therefore, this study aims to investigate the productivity of biodiesel produced from waste vegetable oils using the SMV reactor. Biodiesel fuel is a replacement for diesel as a fuel produced from biomass resources. It is generally produced as a FAME derived from vegetable oil by using alkaline catalyzed alcoholysis process. This alkaline method requires deacidification process prior to the reaction process and the alkaline catalyst removal process after the reaction. Those process increases the total cost of biodiesel fuel production. In order to solve the problems in the conventional alkaline catalyzed alcoholysis process, the authors proposed a non-catalytic alcoholysis process called the Superheated Methanol Vapor (SMV) method with bubble column reactor. So, this study aims to investigate the productivity of biodiesel produced from vegetable oils and other lipids using the SMV method with bubble column reactor.
