Breast cancer (BC) remains the primary cause of death from cancer among women worldwide. Cholesterol-5,6-epoxide (5,6-EC) metabolism is deregulated in BC but the molecular origin of this is unknown. Here, we have identified an oncometabolism downstream of 5,6-EC that promotes BC progression independently of estrogen receptor α expression. We show that cholesterol epoxide hydrolase (ChEH) metabolizes 5,6-EC into cholestane-3β,5α,6β-triol, which is transformed into the oncometabolite 6-oxo-cholestan-3β,5α-diol (OCDO) by 11β-hydroxysteroid-dehydrogenase-type-2 (11βHSD2). 11βHSD2 is known to regulate glucocorticoid metabolism by converting active cortisol into inactive cortisone. ChEH inhibition and 11βHSD2 silencing inhibited OCDO production and tumor growth. Patient BC samples showed significant increased OCDO levels and greater ChEH and 11βHSD2 protein expression compared with normal tissues. The analysis of several human BC mRNA databases indicated that 11βHSD2 and ChEH overexpression correlated with a higher risk of patient death, highlighting that the biosynthetic pathway producing OCDO is of major importance to BC pathology. OCDO stimulates BC cell growth by binding to the glucocorticoid receptor (GR), the nuclear receptor of endogenous cortisol. Interestingly, high GR expression or activation correlates with poor therapeutic response or prognosis in many solid tumors, including BC. 5,6α-epoxycholesterol (5,6α-EC) and 5,6β-epoxycholesterol (5,6β-EC) are oxysterols involved in the anticancer pharmacology of the widely used antitumor drug tamoxifen. They are both metabolized into cholestane-3β,5α,6β-triol (CT) by the cholesterol-5,6-epoxide hydrolase (ChEH) enzyme, and CT is metabolized by an as-yet uncharacterized enzyme into 6-oxo-cholestan-3β,5α-diol (OCDO). A recent feasibility study showed that the 5,6-ECs may represent surrogate markers of tamoxifen activity in breast cancer patients undergoing endocrine therapy, thus there is a growing interest in their accurate quantification. These oxysterols are usually quantified by gas-liquid chromatography coupled to mass spectrometry (GC/MS), using an isotope dilution methodology with the corresponding deuterated oxysterol. This method is considered to be relative quantitative since all of the standards used are deuterated oxysterols, however it is not known whether the preparation of each oxysterol is affected in the same way by the extraction, pre-purification by solid phase extraction (SPE) and trimethylsilylation steps, particularly when using biological samples that contain many other reactive compounds. Thus, in this study we investigated the yield of the 5,6-ECs, CT and OCDO recovery from patient serum samples at different stages of their work-up and trimethylsilylation prior to GC/MS analysis, using [ 14 C]-labeled analogs to follow these oxysterols at each step. We measured a 40 to 60% loss of material for the 5,6-ECs and OCDO, however we also describe the conditions that improved their recovery. Our data also show that the use of deuterated 5,6α-EC, 5,6β-EC, CT and OCDO is an absolute requirement for their accurate quantification. Copyright © 2017 Elsevier B.V. All rights reserved.
Materials for Advanced Ultra-supercritical (A-USC) Steam Turbines – A-USC Component Demonstration

The work by the United States Department of Energy (U.S. DOE)/Ohio Coal Development Office (OCDO) advanced ultra-supercritical (A-USC) Steam Boiler and Turbine Materials Consortia from 2001 through September 2015 was primarily focused on lab scale and pilot scale materials testing. This testing included air- or steam-cooled “loops” that were inserted into existing utility boilers to gain exposure of these materials to realistic conditions of high temperature and corrosion due to the constituents in the coal. Successful research and development resulted in metallic alloy materials and fabrication processes suited for power generation applications with metal temperatures up to approximately 1472°F (800°C).more » These materials or alloys have shown, in extensive laboratory tests and shop fabrication studies, to have excellent applicability for high-efficiency low CO 2 transformational power generation technologies previously mentioned. However, as valuable as these material loops have been for obtaining information, their scale is significantly below that required to minimize the risk associated with a power company building a multi-billion dollar A-USC power plant. To decrease the identified risk barriers to full-scale implementation of these advanced materials, the U.S. DOE/OCDO A-USC Steam Boiler and Turbine Materials Consortia identified the key areas of the technology that need to be tested at a larger scale. Based upon the recommendations and outcome of a Consortia-sponsored workshop with the U.S.’s leading utilities, a Component Test (ComTest) Program for A-USC was proposed. The A-USC ComTest program would define materials performance requirements, plan for overall advanced system integration, design critical component tests, fabricate components for testing from advanced materials, and carry out the tests. The AUSC Component Test was premised on the program occurring at multiple facilities, with the operating temperatures, pressure and/or size of these components determining the optimum test location. The first step of the ComTest, the steam turbine test, was determined best suited for a site in Youngstown, Ohio. Efforts were also undertaken to identify and evaluate other potential sites for high pressure testing.« less

The U.S. Department of Energy (DOE) and the Ohio Coal Development Office (OCDO) have sponsored a project aimed at identifying, evaluating, and qualifying the materials needed for the construction of the critical components of coal-fired power plants capable of operating at much higher efficiencies than the current generation of supercritical plants. This increased efficiency is expected to be achieved principally through the use of advanced ultrasupercritical (A-USC) steam conditions. A limiting factor in this can be the materials of construction for boilers and for steam turbines. The overall project goal is to assess/develop materials technology that will enable achieving turbinemore » throttle steam conditions of 760°C (1400°F)/35MPa (5000 psi). This final technical report covers the research completed by the General Electric Company (GE) and Electric Power Research Institute (EPRI), with support from Oak Ridge National Laboratory (ORNL) and the National Energy Technology Laboratory (NETL) – Albany Research Center, to develop the A-USC steam turbine materials technology to meet the overall project goals. Specifically, this report summarizes the industrial scale-up and materials property database development for non-welded rotors (disc forgings), buckets (blades), bolting, castings (needed for casing and valve bodies), casting weld repair, and casting to pipe welding. Additionally, the report provides an engineering and economic assessment of an A-USC power plant without and with partial carbon capture and storage. This research project successfully demonstrated the materials technology at a sufficient scale and with corresponding materials property data to enable the design of an A-USC steam turbine. The key accomplishments included the development of a triple-melt and forged Haynes 282 disc for bolted rotor construction, long-term property development for Nimonic 105 for blading and bolting, successful scale-up of Haynes 282 and Nimonic 263 castings using traditional sand foundry practices, and a techno-economic study of an A-USC plant including cost estimates for an A-USC turbine which showed A-USC to be economically attractive for partial carbon and capture compared to today’s USC technology. Based on this successful materials research and a review with U.S. utility stakeholders, a new project to develop a component test facility (ComTest) including the world’s first A-USC turbine has been proposed to continue the technology development.« less

The U.S. Department of Energy (DOE) and the Ohio Coal Development Office (OCDO) have undertaken a project aimed at identifying, evaluating, and qualifying the materials needed for the construction of the critical components of coal-fired boilers capable of operating at much higher efficiencies than current generation of supercritical plants. This increased efficiency is expected to be achieved principally through the use of advanced ultrasupercritical (A-USC) steam conditions up to 760°C (1400°F) and 35 MPa (5000 psi). A limiting factor to achieving these higher temperatures and pressures for future A-USC plants are the materials of construction. The goal of this projectmore » is to assess/develop materials technology to build and operate an A-USC boiler capable of delivering steam with conditions up to 760°C (1400°F)/35 MPa (5000 psi). The project has successfully met this goal through a focused long-term public-private consortium partnership. The project was based on an R&D plan developed by the Electric Power Research Institute (EPRI) and an industry consortium that supplemented the recommendations of several DOE workshops on the subject of advanced materials. In view of the variety of skills and expertise required for the successful completion of the proposed work, a consortium led by the Energy Industries of Ohio (EIO) with cost-sharing participation of all the major domestic boiler manufacturers, ALSTOM Power (Alstom), Babcock and Wilcox Power Generation Group, Inc. (B&W), Foster Wheeler (FW), and Riley Power, Inc. (Riley), technical management by EPRI and research conducted by Oak Ridge National Laboratory (ORNL) has been developed. The project has clearly identified and tested materials that can withstand 760°C (1400°F) steam conditions and can also make a 700°C (1300°F) plant more economically attractive. In this project, the maximum temperature capabilities of these and other available high-temperature alloys have been assessed to provide a basis for materials selection and application under a range of conditions prevailing in the boiler. A major effort involving eight tasks was completed in Phase 1. In a subsequent Phase 2 extension, the earlier defined tasks were extended to finish and enhance the Phase 1 activities. This extension included efforts in improved weld/weldment performance, development of longer-term material property databases, additional field (in-plant) corrosion testing, improved understanding of long-term oxidation kinetics and exfoliation, cyclic operation, and fabrication methods for waterwalls. In addition, preliminary work was undertaken to model an oxyfuel boiler to define local environments expected to occur and to study corrosion behavior of alloys under these conditions. This final technical report provides a comprehensive summary of all the work undertaken by the consortium and the research findings from all eight (8) technical tasks including A-USC boiler design and economics (Task 1), long-term materials properties (Task 2), steam- side oxidation (Task 3), Fireside Corrosion (Task 4), Welding (Task 5), Fabricability (Task 6), Coatings (Task 7), and Design Data and Rules (Task 8).« less

Usability Considerations in Developing a Graphic Interface for Intra Office Communications

This paper outlines the basis of incorporating functional features in a new GUI based software under development for addressing comprehensive communication and interaction needs within an office environment. Bench marking of features in existing communication software products such as Microsoft Outlook, IBM Lotusnotes, Office Communicator, Mozilla Thunderbird etc. was done by asking a set of questions related to the usage of these existing softwares. Usability issues were identified through a user survey involving 30 subjects of varied profiles (domain, designation, age etc.) in a corporate office. It is posited that existing software products that have been developed for a universal market may be highly underutilized or have redundant features especially for use as an intra office (within the same office) communication medium. Simultaneously they may not cater to some very contextual requirements of intra office communications. Based on the findings of the survey of feature preferences & usability of existing products, a simple 'person to person' communicating medium for intra office situations was visualized with a new interactive GUI. Usability issues that need to be considered for a new intra-office product have been brought out.

The US Department of Energy (DOE) Office of Environmental Management, formerly the Office of Environmental Restoration and Waste Management (EM), was established in November 1989 as the first step toward correcting contamination problems resulting from nearly 50 years of nuclear weapons production and fuel processing activities. EM consolidates several DOE organizations previously responsible for the handling, treatment, and disposition of radioactive and hazardous waste. To determine how U.S. MD-granting medical schools manage, fund, and evaluate faculty affairs/development functions and to determine the evolution of these offices between 2000 and 2010. In December 2010, the authors invited faculty affairs designees at 131 U.S. MD-granting medical schools to complete a questionnaire developed by the Association of American Medical Colleges Group on Faculty Affairs, based on a 2000 survey. Schools were asked about core functions, budget, staffing, and performance metrics. The authors analyzed the data using descriptive statistics. A total of 111 schools (84.7%) responded. Fifty percent of the offices were established since 2000. Seventy-eight percent reported their top core function as administrative support for appointments, promotions, and tenure, as in 2000. Faculty policies, appointments, databases, governance support, grievance proceedings, management issues, and annual trend analyses continued as major functions. All 11 core functions identified in 2000 remain predominantly provided by central offices of faculty affairs, except support of major leadership searches. Web site communication emerged as a new core function. Similar to 2000, several other offices were responsible for some faculty development functions. Office size and budget correlated positively with size of the faculty and age of the office (P < .05 for all). Thirty-five schools (31.5%) reported formally evaluating their faculty affairs office. The number of faculty affairs offices and their responsibilities have substantially increased since 2000. Most major core functions have not changed. These offices are now an established part of the central administration of most medical schools.

As part of efforts to develop training and career development experiences to enhance leadership skills among public health officials, the Public Health Foundation, Association of State and Territorial Health Officials, National Association of County Health Officials, United States Conference of Local Health Officers, and Public Health Practice Program Office, Centers for Disease Control and Prevention, conducted a training needs assessment survey in 1988. Fifty-five State and territorial health officers were asked about potential knowledge, skills, and abilities (KSAs) that a prospective or new health officer might require in performing his or her job. Thirty-eight health officers returned completed questionnaires, a 69 percent response rate. For each KSA, respondents assigned scores from 1 (low) to 5 (high) to three different variables: the KSA's importance to job, as an initial ability of a new health officer, and as a desired ability for someone in that job. Of 78 KSAs, those scoring in the top 25 percent for importance to job were identified, and individual composite scores were calculated using the formula: (importance to job) x (desired ability minus initial ability). The top 10 mean composite scores ranged from 7.55 to 10.40 and were in five competence areas: public image (working with the community) (3 KSAs); policy development and program planning (3 KSAs); interpersonal skills (2 KSAs); agency management (1 KSA); and legal issues (1 KSA). These skills are not commonly acquired in schools of medicine or public health. Public health agencies should develop programs to assure that persons with leadership potential are identified early and given guided experiences and mentors, as well as specific training and education. Additional studies of public health officers are needed to develop and strengthen leadership KSAs among new health officers. PMID:8434086

To evaluate the participatory ergonomic method on the development of upper extremity musculoskeletal disorders and disability in office employees. This study is a randomized controlled intervention study. It comprised 116 office workers using computers. Those in the intervention group were taught office ergonomics and the risk assessment method. Cox proportional hazards model and generalized estimating equations (GEEs) were used. In the 10-month postintervention follow-up, the possibility of developing symptoms was 50.9%. According to multivariate analysis results, the possibility of developing symptoms on the right side of the neck and in the right wrist and hand was significantly less in the intervention group than in the control group (p<0.05). Neck disability/symptom scores over time were significantly lower in the intervention group compared with the control group (p<0.05). The participatory ergonomic intervention decreases the possibility of musculoskeletal complaints and disability/symptom level in office workers.

An effective office ergonomics program can predict positive results in reducing musculoskeletal injury rates, enhancing productivity, and improving staff well-being and job satisfaction. Its objective is to provide a systematic solution to manage the potential risk of musculoskeletal disorders among computer users in an office setting. A FITS Model office ergonomics program is developed. The FITS Model Office Ergonomics Program has been developed which draws on the legislative requirements for promoting the health and safety of workers using computers for extended periods as well as previous research findings. The Model is developed according to the practical industrial knowledge in ergonomics, occupational health and safety management, and human resources management in Hong Kong and overseas. This paper proposes a comprehensive office ergonomics program, the FITS Model, which considers (1) Furniture Evaluation and Selection; (2) Individual Workstation Assessment; (3) Training and Education; (4) Stretching Exercises and Rest Break as elements of an effective program. An experienced ergonomics practitioner should be included in the program design and implementation. Through the FITS Model Office Ergonomics Program, the risk of musculoskeletal disorders among computer users can be eliminated or minimized, and workplace health and safety and employees' wellness enhanced.

The Office of Environmental Restoration and Waste Management (EM) was established by the Department of Energy (DOE) to direct and coordinate waste management and site remediation programs/activities throughout the DOE complex. In order to successfully achieve the goal of properly managing waste and the cleanup of the DOE sites, the EM was divided into five organizations: the Office of Planning and Resource Management (EM-10); the Office of Environmental Quality Assurance and Resource Management (EM-20); the Office of Waste Operations (EM-30); the Office of Environmental Restoration (EM-40); and the Office of Technology and Development (EM-50). The mission of the Office ofmore » Technology Development (OTD) is to develop treatment technologies for DOE`s operational and environmental restoration wastes where current treatment technologies are inadequate or not available. The Mixed Waste Integrated Program (MWIP) was created by OTD to assist in the development of treatment technologies for the DOE mixed low-level wastes (MLLW). The MWIP has established five Technical Support Groups (TSGs) whose purpose is to identify, evaluate, and develop treatment technologies within five general technical areas representing waste treatment functions from initial waste handling through generation of final waste forms. These TSGs are: (1) Front-End Waste Handling, (2) Physical/Chemical Treatment, (3) Waste Destruction and Stabilization, (4) Second-Stage Destruction and Offgas Treatment, and (5) Final Waste Forms. This report describes the functions of the Waste Destruction and Stabilization (WDS) group. Drawing on developmental and policy research, this study examined whether 3 dimensions of caseworker-recipient interaction in welfare offices functioned as critical ecological contexts for recipient families. The sample consisted of 1,098 families from 10 welfare offices in National Evaluation of Welfare to Work Strategies (NEWWS). In multilevel analyses, caseworker support, caseload size, and emphasis on employment predicted 5-year quarterly trajectories of earnings, income, and welfare receipt. Recipients in offices characterized by high support had steeper increases in earnings and income; those in offices with high caseload size had steeper decreases in income and welfare receipt; and those in offices with high emphasis on employment had steeper decreases in welfare receipt. These economic trajectories were associated with children's reading and math achievement and internalizing behavior at ages 8-10. © 2011 The Authors. PURPOSE: It is essential to adjust the responsibilities and function of medical education offices (MEOs) in regard to the current societal requirements. Therefore, it is a good idea to learn lessons from the experiences about the establishment and function of these offices around the world. The aim of the present study was to carry out a comparative study to investigate the function and structure of MEOs at some of the medical universities from America, Europe, and Asia. SUBJECTS AND METHODS: This is a comparative, descriptive study that was conducted in 2015. Eleven offices around the world (in America, Europe, and Asia) were selected for the study. Expert group discussion and literature review were used in order to select research sample. The data were gathered using self-constructed checklists. Content and face validity of the checklist was assessed by gathering feedback from experts. The Kappa coefficient was used to determine the inter-rater reliability. RESULTS: All the 11 offices in our study (100%) dealt with the issues of faculty development and research and scholarship activities. Only one out of the 11 offices (27%) dealt with the issues of society and patient education. Five out of the 11 offices (36%) dealt with the continuing medical education and continuing professional development. Consultation services are provided at seven of the 11 offices (64%). CONCLUSIONS: This study revealed both commonalities and differences in the function and structure of MEO among the 11 offices we examined. Although uncommon, medical emergencies do occur in the dental office setting. This article describes the development and implementation of an office-based emergencies course for third-year dental students. The course reviews the basic management of selected medical emergencies. Background information is provided that further highlights the importance of proper training to manage medical emergencies in the dental office. Details regarding course development, implementation, logistics, and teaching points are highlighted. The article provides a starting point from which dental educators can modify and adapt this course and its objectives to fit their needs or resources. This is a timely topic that should benefit both dental students and dental educators.
Understanding how police officers think about mental/emotional disturbance calls

Police officers frequently respond to calls involving persons with mental illnesses and in doing so, they are key gatekeepers of access to mental health treatment as well as entry into the criminal justice system. Programs such as Crisis Intervention Teams (CIT) are being implemented across the United States and elsewhere to train officers to respond more effectively and facilitate access to mental health services when appropriate. These programs would benefit from a thorough understanding of these encounters from the perspective of police officers. We take as a premise that officers develop frames of reference or “schema” for understanding and responding to these encounters that are shaped by socialization, training, and their experience as police officers. In this study, we examine police officer schema of mental/emotional disturbance (M/EDP) calls. Qualitative interviews provided the foundation to develop the Needs on the Street Interview (NOSI) to tap officer schema of four types of M/EDP scenarios. The NOSI was administered to 147 officers in Chicago and Philadelphia. Latent Class Analysis (LCA) was conducted separately for each scenario to examine groups of officers with different schema as well as predictors of schema group. For three of the four scenarios, officers were classified into a two category or schema model, for the fourth (crime reported) a three category model was supported. Schema groups tended to be differentiated by ratings of level of resistance/threat and substance use. Contrary to our expectations, CIT and law enforcement experience did not predict officer schema group. While the CIT model emphasizes de-escalation skills to reduce resistance and the need for officers to use force, CIT and other training programs may want to consider increasing content related to factors such as co-occurring substance use and managing resistance. The IAU established its Office of Astronomy for Development in 2011 in Cape Town, South Africa, in partnership with the South African National Research Foundation, and hosted at the South African Astronomical Observatory. The primary purpose of the office has been to implement the IAU's decadal strategic plan which aims to use astronomy to stimulate education and development globally. Since its inception there have been nine regional offices established, including one in the Latin American region which focuses on Andean countries. Following a very positive external review in 2015, as well as a supportive resolution by the 2015 IAU General Assembly to continue the work of the office, the OAD is now in a position to move beyond the setting up of structures to a more intensive effort aimed at measuring and delivering programmes with broader global impact.

The IAU Office of Astronomy for Development (OAD)†, established in March 2011 as part of the implementation of the IAU Strategic Plan, is currently located in South Africa and serves as a global coordinating centre for astronomy-for-development activities. In terms of structure the OAD is required to establish regional nodes (similar offices in different parts of the world which focus on a particular geographic or cultural region) and three task forces: (i) Astronomy for Universities and Research, (ii) Astronomy for Children and Schools, and (iii) Astronomy for the Public. This paper will describe the progress of the OAD towards the realisation of the vision `Astronomy for a better world'.

The purpose of this exploratory study was to explore correctional officers' perceptions and experiences during a solution-focused training program and to initiate development of a modified pattern for correctional officers to use in jails. The study uses grounded theory procedures combined with a follow-up survey. The findings identified six emergent themes: obstacles to doing counseling work in prisons, offenders' amenability to change, correctional officers' self-image, advantages of a solution-focused approach (SFA), potential advantages of applying SFA to offenders, and the need for the consolidation of learning and transformation. Participants perceived the use of solution-focused techniques as appropriate, important, functional, and of only moderate difficulty in interacting with offenders. Finally, a modified pattern was developed for officers to use when working with offenders in jails. Suggestions and recommendations are made for correctional interventions and future studies.
