The Analytical Chemistry Division of Oak Ridge National Laboratory (ORNL) is a large and diversified organization. As such, it serves a multitude of functions for a clientele that exists both in and outside of ORNL. These functions fall into the following general categories: Analytical Research, Development and Implementation; Programmatic Research, Development, and Utilization; and Technical Support. The Analytical Chemistry Division is organized into four major sections, each which may carry out any of the three types of work mentioned above. Each year, the annual meeting of the APS Division of Plasma Physics (DPP) brings together a broad representation of the many active subfields of plasma physics and enjoys an audience that is equally diverse. The meeting was well attended and largely went as planned despite the interventions of hurricane Sandy which caused the city of Providence to shut-down during the first day of the conference. The meeting began on Monday morning with a review of the physics of cosmic rays, 2012 being the 100th year since their discovery, which illustrated the central importance of plasma physics to astrophysical problems. Subsequent reviews covered the importance of tokamak plasma boundaries, progress towards ignition on the National Ignition Facility (NIF), and magnetized plasma turbulence. The Maxwell prize address, by Professor Liu Chen, covered the field of nonlinear Alfvén wave physics. Tutorial lectures were presented on the verification of gyrokinetics, new capabilities in laboratory astrophysics, magnetic flux compression, and tokamak plasma start-up.

Welcome to the Sixth Annual NASA Ames Research Center, Space Science and Astrobiology Jamboree at NASA Ames Research Center (ARC). The Space Science and Astrobiology Division consists of over 60 Civil Servants, with more than 120 Cooperative Agreement Research Scientists, Post-Doctoral Fellows, Science Support Contractors, Visiting Scientists, and many other Research Associates. Within the Division there is engagement in scientific investigations over a breadth of disciplines including Astrobiology, Astrophysics, Exobiology, Exoplanets, Planetary Systems Science, and many more. The Division's personnel support NASA spacecraft missions (current and planned), including SOFIA, K2, MSL, New Horizons, JWST, WFIRST, and others. Our top-notch science research staff is spread amongst three branches in five buildings at ARC. Naturally, it can thus be difficult to remain abreast of what fellow scientific researchers pursue actively, and then what may present and/or offer regarding inter-Branch, intra-Division future collaborative efforts. In organizing this annual jamboree, the goals are to offer a wholesome, one-venue opportunity to sense the active scientific research and spacecraft mission involvement within the Division; and to facilitate communication and collaboration amongst our research scientists. Annually, the Division honors one senior research scientist with a Pollack Lecture, and one early career research scientist with an Outstanding Early Career Space Scientist Lecture. For the Pollack Lecture, the honor is bestowed upon a senior researcher who has made significant contributions within any area of research aligned with space science and/or astrobiology. This year we are pleased to honor Linda Jahnke. With the Early Career Lecture, the honor is bestowed upon an early-career researcher who has substantially demonstrated great promise for significant contributions within space science, astrobiology, and/or, in support of spacecraft missions addressing such disciplines. This year we are pleased to honor Amanda Cook. We hope that you will make time to join us for the day in meeting fellow Division members, expanding knowledge of our activities, and creating new collaborations within the Space Science and Astrobiology Division.

This report covers work done during FY 1983 by the staff of the Energy Division and its subcontractors and by colleagues in other Oak Ridge National Laboratory divisions working on Energy Division projects. The work can be divided into four areas: (1) analysis and assessment, (2) models and data systems, (3) research to improve the efficiency of energy use and to improve electric power transmission and distribution, and (4) research utilization. Support came principally from the US Department of Energy (DOE), the US Nuclear Regulatory Commission, and the US Department of Defense, but also from a number of other agenciesmore » and organizations. Analysis and assessment included work on (a) environmental issues, including those deriving from the preparation of environmental impact statements; (b) energy and resource analysis; and (c) emergency preparedness. The models and data systems area involved research on evaluating and developing energy, environment, and engineering simulation models and on devising large data management systems, evaluating user data requirements, and compiling data bases. Research on improving the efficiency of energy use was focused primarily on the buildings and electricity sectors. A major effort on heat pump technology, which includes both heat-activated and electrically driven systems, continues. An important aspect of all the work was research utilization. Since the Energy Division is doing applied research, results are, by definition, intended to solve problems or answer questions of DOE and other sponsors. However, there are other users, and research utilization activities include technology transfer, commercialization efforts, outreach to state and regional organizations, and, of course, information dissemination.« less

The National Research Program (NRP) of the US Geological Survey 's Water Resources Division (WRD) had its beginnings in the late 1950 's when ' core research ' was added as a line item to the Congressional budget. Since that time, the NRP has grown to encompass a broad spectrum of scientific investigations. The sciences of hydrology, mathematics, chemistry, physics, ecology, biology, geology, and engineering are used to gain a fundamental understanding of the processes that affect the availability, movement, and quality of the Nation 's water resources. The NRP is located principally in Reston, VA, Denver, CO, and Menlo Park , CA. The NRP is subdivided into six disciplines as follows: (1) Ecology; (2) Geomorphology and Sediment Transport; (3) Groundwater Chemistry; (4) Groundwater Hydrology; (5) Surface Water Chemistry; and (6) Surface Water Hydrology. It is our custom to publish a division progress report every 18 months that summarizes our programmatic progress and other measures of achievement over the reporting period. Since it is not feasible tomore » summarize in detail all of our work over the period covered by this report (October 1, 1988, to March 30, 1990), we intend this document to point the way to the expensive open literature that documents our findings. During the reporting period the Division continued to maintain strong programs in its traditional areas of R D, but also achieved noteworthy progress in other areas. Much of the Division's work on site characterization, development of new field instruments, compilation of data bases, and methodology development fits into this initiative. Other new work in tunneling microscopy in support of DOE's Human Genome Program and the comprehensive R D work related to surface-enhanced Raman spectroscopy have attained new and exciting results. These examples of our progress and numerous other activities are highlighted in this report.« less

The Nez Perce Tribe Department of Fisheries Resource Management, Watershed Division, approaches watershed restoration with a goal to protect, restore, and enhance a connected network of functioning habitat types capable of supporting all fish life stages. Its goal is also to re-establish normal patters of production, dispersal, and exchange of genetic information within the 1855 Treaty Area. The Nez Perce Tribe began watershed restoration projects within the Clearwater River Subbasin in 1996. Progress has been made in restoring the sub-basin by excluding cattle from critical riparian areas through fencing, stabilizing streambanks, decommissioning roads, and upgrading culverts. Coordination of these projectsmore » is critical to the success of the restoration of the sub-basin. The Nez Perce Tribe Department of Fisheries Resource Management, Watershed Division approaches watershed restoration with a ridge-top to ridge-top approach. Watershed restoration projects within the Mill Creek watershed are coordinated with the Nez Perce National Forest. The Nez Perce Tribe began watershed restoration projects within the Mill Creek watershed of the South Fork Clearwater River in 2000. Progress has been made in restoring the watershed through excluding cattle from critical riparian areas through fencing. During the FY 2002, trees were planted in riparian areas in the meadow of the upper watershed. In addition, a complete inventory of culverts at road-streammore » crossings was completed. Culverts have been prioritized for replacement to accommodate fish passage throughout the watershed. Maintenance to the previously built fence was also completed.« less

This fiscal year (FY) 1997 annual report describes key elements of the NASA Microgravity Research Program (MRP) as conducted by the Microgravity Research Division (MRD) within NASA's Office of Life and Microgravity, Sciences and Applications. The program's goals, approach taken to achieve those goals, and program resources are summarized. All snapshots of the program's status at the end of FY 1997 and a review of highlights and progress in grounds and flights based research are provided. Also described are major space missions that flew during FY 1997, plans for utilization of the research potential of the International Space Station, the Advanced Technology Development (ATD) Program, and various educational/outreach activities. The MRP supports investigators from academia, industry, and government research communities needing a space environment to study phenomena directly or indirectly affected by gravity.

Intestinal divisions in Caenorhabditis elegans take place in 3 stages: (1) cell divisions during embryogenesis, (2) binucleations at the L1 stage, and (3) endoreduplications at the end of each larval stage. Here, we report that CDC-25.2, a C. elegans ortholog of Cdc25, is required for these specialized division cycles between the 16E cell stage and the onset of endoreduplication. Results of our genetic analyses suggest that CDC-25.2 regulates intestinal cell divisions and binucleations by counteracting WEE-1.3 and by activating the CDK-1/CYB-1 complex. CDC-25.2 activity is then repressed by LIN-23 E3 ubiquitin ligase before the onset of intestinal endoreduplication, and this repression is maintained by LIN-35, the C. elegans ortholog of Retinoblastoma (Rb). These findings indicate that timely regulation of CDC-25.2 activity is essential for the progression of specialized division cycles and development of the C. elegans intestine.

Phosphorylation of histone H3 on Ser-10 (H3S10ph) is involved in regulating mitotic chromosome condensation and decondensation, which plays an important regulatory role during mitotic cell cycle progression in mammalian cells. However, whether H3S10ph plays a similar role in early porcine embryos during the first mitotic division remains uncertain. In this study, the subcellular localization and possible roles of H3S10ph were evaluated in the first mitotic cell cycle progression of porcine embryos using western blot, indirect immunofluorescence and barasertib (H3S10ph upstream regulator Aurora-B inhibitor) treatments. H3S10ph exhibited a dynamic localization pattern and was localized to chromosomes from prometaphase to anaphase stages. Treatment of porcine embryos with barasertib inhibited mitotic division at the prophase stage and was associated with a defect in chromosome condensation accompanied by the reduction of H3S10ph. These results indicated that H3S10ph is involved in the first mitotic division in porcine embryos through its regulatory function in chromosome condensation, which further affects porcine embryo cell cycle progression during mitotic division.

Watershed restoration projects within the Meadow Creek watershed are coordinated with the Nez Perce National Forest. The Nez Perce Tribe began watershed restoration projects within the Meadow Creek watershed of the South Fork Clearwater River in 1996. Progress has been made in restoring the watershed by excluding cattle from critical riparian areas through fencing. During years 2000-2003, trees were planted in riparian areas within the meadow and its tributaries. Culverts have been prioritized for replacement to accommodate fish passage throughoutmore » the watershed. Designs for replacement are being coordinated with the Nez Perce National Forest. This progress report presents the results of 11 projects funded ($500K) in FY01 by the Technology Development, Evaluation, and Application (TDEA) Committee of the Environment, Safety, and Health Division (ESH). Five projects fit into the Health Physics discipline, 5 projects are environmental science and one is industrial hygiene/safety. As a result of their TDEA-funded projects, investigators have published sixteen papers in professional journals, proceedings, or Los Alamos reports and presented their work at professional meetings. We have made substantial progress toward our objectives outlined in our BPA supported proposal entitled 'Columbia River Basin Juvenile Salmonids: Survival and Growth in the Columbia River Plume and northern California Current' which we report on herein. During 2008, we were able to successfully conduct 3 mesoscale cruises. We also were able to conduct 7 biweekly predator cruises, along with substantial shore-based visual observations of seabirds. Detailed results of the mesoscale cruises are available in the Cruise Reports and summarized in the next section. We have taken a proactive approach to getting the results of our research to fisheries managersmore » and the general public. We have begun to make annual predictions based on ocean conditions of the relative survival of juvenile coho and Chinook salmon well before they return as adults. This is based on both biological and physical indicators that we measure during our surveys or collect from outside data sources. This Annual Report presents summaries of selected representative research activities grouped according to the principal disciplines of the Earth Sciences Division: Reservoir Engineering and Hydrogeology, Geology and Geochemistry, and Geophysics and Geomechanics. Much of the Division`s research deals with the physical and chemical properties and processes in the earth`s crust, from the partially saturated, low-temperature near-surface environment to the high-temperature environments characteristic of regions where magmatic-hydrothermal processes are active. Strengths in laboratory and field instrumentation, numerical modeling, and in situ measurement allow study of the transport of mass and heat through geologic media -- studies that now include the appropriatemore » chemical reactions and the hydraulic-mechanical complexities of fractured rock systems. Of particular note are three major Division efforts addressing problems in the discovery and recovery of petroleum, the application of isotope geochemistry to the study of geodynamic processes and earth history, and the development of borehole methods for high-resolution imaging of the subsurface using seismic and electromagnetic waves. In 1989 a major DOE-wide effort was launched in the areas of Environmental Restoration and Waste Management. Many of the methods previously developed for and applied to deeper regions of the earth will in the coming years be turned toward process definition and characterization of the very shallow subsurface, where man-induced contaminants now intrude and where remedial action is required.« less

Hydrologic Drought of Water Year 2006 Compared with Four Major Drought Periods of the 20th Century in Oklahoma

Water Year 2006 (October 1, 2005, to September 30, 2006) was a year of extreme hydrologic drought and the driest year in the recent 2002-2006 drought in Oklahoma. The severity of this recent drought can be evaluated by comparing it with four previous major hydrologic droughts, water years 1929-41, 1952-56, 1961-72, and 1976-81. The U.S. Geological Survey, in cooperation with the Oklahoma Water Resources Board, completed an investigation to summarize the Water Year 2006 hydrologic drought and compare it to the four previous major hydrologic droughts in the 20th century. The period of water years 1925-2006 was selected as the period of record because before 1925 few continuous record streamflow-gaging sites existed and gaps existed where no streamflow-gaging sites were operated. Statewide annual precipitation in Water Year 2006 was second driest and statewide annual runoff in Water Year 2006 was sixth driest in the 82 years of record. Annual area-averaged precipitation totals by the nine National Weather Service Climate Divisions from Water Year 2006 are compared to those during four previous major hydrologic droughts to show how rainfall deficits in Oklahoma varied by region. Only two of the nine climate divisions, Climate Division 1 Panhandle and Climate Division 4 West Central, had minor rainfall deficits, while the rest of the climate divisions had severe rainfall deficits in Water Year 2006 ranging from only 65 to 73 percent of normal annual precipitation. Regional streamflow patterns for Water Year 2006 indicate that Oklahoma was part of the regionwide below-normal streamflow conditions for Arkansas-White-Red River Basin, the sixth driest since 1930. The percentage of long-term stations in Oklahoma (with at least 30 years of record) having below-normal streamflow reached 80 to 85 percent for some days in August and November 2006. Twelve long-term streamflow-gaging sites with periods of record ranging from 62 to 78 years were selected to show how streamflow deficits varied by region. The hydrologic drought worsened going from north to south in Oklahoma, ranging from 45 percent in the north, to just 14 percent in east-central Oklahoma, and 20 percent of normal annual streamflow in the southwest. The low streamflows resulted in only 86.3 percent of the statewide conservation storage available at the end of the water year in major reservoirs, and 7 to 47 percent of hydroelectric power generation at sites in Oklahoma in Calendar Year 2005.

This report summarizes the research performed in 2000 in the Physics Division of Argonne National Laboratory. The Division's programs include operation of ATLAS as a national user facility, nuclear structure and reaction research, nuclear theory and medium energy physics research, and accelerator research and development. As the Nuclear Science Advisory Committee and the nuclear science community create a new long range plan for the field in 2001, it is clear that the research of the Division is closely aligned with and continues to help define the national goals of our field. The NSAC 2001 Long Range Plan recommends as themore » highest priority for major new construction the Rare Isotope Accelerator (RIA), a bold step forward for nuclear structure and nuclear astrophysics. The accelerator R&D in the Physics Division has made major contributions to almost all aspects of the RIA design concept and the community was convinced that this project is ready to move forward. 2000 saw the end of the first Gammasphere epoch at ATLAS, One hundred Gammasphere experiments were completed between January 1998 and March 2000, 60% of which used the Fragment Mass Analyzer to provide mass identification in the reaction. The experimental program at ATLAS then shifted to other important research avenues including proton radioactivity, mass measurements with the Canadian Penning Trap and measurements of high energy gamma-rays in nuclear reactions with the MSU/ORNL/Texas A&M BaF{sub 2} array. ATLAS provided 5460 beam-research hours for user experiments and maintained an operational reliability of 95%. Radioactive beams accounted for 7% of the beam time. ATLAS also provided a crucial test of a key RIA concept, the ability to accelerate multiple charge states in a superconducting heavy-ion linac. This new capability was immediately used to increase the performance for a scheduled experiment. The medium energy program continued to make strides in examining how the quark-gluon structure of matter impacts the structure of nuclei and extended the exquisite sensitivity of the Atom-Trap-Trace-Analysis technique to new species and applications. All of this progress was built on advances in nuclear theory, which the Division pursues at the quark, hadron, and nuclear collective degrees of freedom levels. This paper reviews the recent progress in the development of holographic neural networks using surface-emitting laser diode arrays (SELDAs). Since the previous work on ultrafast holographic memory readout system and a robust incoherent correlator, progress has been made in several areas: the use of an array of monolithic `neurons' to reconstruct holographic memories; two-dimensional (2-D) wavelength-division multiplexing (WDM) for image transmission through a single-mode fiber; and finally, an associative memory using time- division multiplexing (TDM). Experimental demonstrations on these are presented.

The Nez Perce Tribe Department of Fisheries Resource Management, Watershed Division, approaches watershed restoration with a goal to protect, restore, and enhance a connected network of functioning habitat types capable of supporting all fish life stages. Its goal is also to re-establish normal patterns of production, dispersal, and exchange of genetic information within the 1855 Treaty Area. The Nez Perce Tribe began watershed restoration projects within the Clearwater River Subbasin in 1996. Progress has been made in restoring the sub-basin by excluding cattle from critical riparian areas through fencing, stabilizing stream banks, decommissioning roads, and upgrading culverts. The Nez Perce Tribe Department of Fisheries Resource Management, Watershed Division approaches watershed restoration with a ridge-top to ridge-top approach. Watershed restoration projects within the Lolo Creek watershed are coordinated with the Clearwater National Forest and Potlatch Corporation. The Nez Perce Tribe began watershed restoration projects within the Lolo Creek watershed of the Clearwater River in 1996. Progress has been made in restoring the watershed by excluding cattle from critical riparian areas through fencing, stabilizing streambanks, decommissioning roads, and upgrading culverts. During the years 2000-2003, trees were planted in riparian areas of headwater streams to Lolo Creek. Inventory of culvertsmore » is an on-going practice, being completed by sub-drainage, and are being prioritized for replacement to accommodate fish passage and 100-year flow events throughout the watershed. Tribal crews completed maintenance to the previously built fence.« less

To address the questions and needs of local resource managers, a multi-agency research project was initiated in 2005 to study the ecology, forage relations, and habitat relations of elk and bison in the Great Sand Dunes–Sangre de Cristo–Baca complex of lands. Meetings and discussions of what this research should include were started in 2001 with representatives from NPS, USFWS, TNC, the Colorado Division of Wildlife (CDOW), and USDA-FS/BLM. The final study plan was successfully funded in 2004 with research scheduled to start in 2005. The research was designed to encompass three major study elements: (1) animal movements and population dynamics, (2) vegetation and nutrient effects from ungulate herbivory, and (3) development of ecological models, using empirical data collected from the first two components, that will include estimates of elk carrying capacity and management scenarios for resource managers.

In this article the core components of a comprehensive quality assurance and improvement plan are outlined. Quality anatomic pathology work comes with focus on accurate, timely, and complete reports. A commitment to continuous quality improvement and a systems approach with a persistent effort helps to achieve this end. Departments should have a quality assurance and improvement plan that includes a risk assessment of real and potential problems facing the laboratory. The plan should also list the individuals responsible for carrying out the program with adequate resources, a defined timetable, and annual assessment for progress and future directions. Quality assurance monitors should address regulatory requirements and be organized by laboratory division (surgical pathology, cytology, etc) as well as 5 segments (preanalytic, analytic, postanalytic phases of the test cycle, turn-around-time, and customer satisfaction). Quality assurance data can also be used to evaluate individual pathologists using multiple parameters with peer group comparison.

Neuronal differentiation and cell-cycle exit are tightly coordinated, even in pathological situations. When pathological neurons re-enter the cell cycle and progress through the S phase, they undergo cell death instead of division. However, the mechanisms underlying mitotic resistance are mostly unknown. Here, we have found that acute inactivation of retinoblastoma (Rb) family proteins (Rb, p107 and p130) in mouse postmitotic neurons leads to cell death after S-phase progression. Checkpoint kinase 1 (Chk1) pathway activation during the S phase prevented the cell death, and allowed the division of cortical neurons that had undergone acute Rb family inactivation, oxygen-glucose deprivation (OGD) or in vivo hypoxia-ischemia. During neurogenesis, cortical neurons became protected from S-phase Chk1 pathway activation by the DNA methyltransferase Dnmt1, and underwent cell death after S-phase progression. Our results indicate that Chk1 pathway activation overrides mitotic safeguards and uncouples neuronal differentiation from mitotic resistance. © 2017. Published by The Company of Biologists Ltd.

The Accelerators and Class II Facilities Division (ACFD) of the Canadian Nuclear Safety Commission (CNSC), is responsible for the oversight of radiotherapy facilities containing Class II prescribed equipment in Canada. This poster will highlight a number of new initiatives that the CNSC has implemented recently that have an impact on radiotherapy facility licensees. The presentation will discuss the recent policy decision to regulate particle accelerators of above 1 MeV. Challenges and progress with respect to the implementation of the policy will be presented. Other initiatives which will be described include: • The new ACFD webspace on the CNSC website, with direct links to relevant information on licensing, compliance and Class II prescribed equipment • The improved structure of the Appendix of Licence Documents that is part of every Class II licence • Updated licence application guides • Changes to Annual Compliance reporting requirements and progress on the ACR-Online initiative • Changes to some regulatory expectations related to medical accelerator facilities • Consolidation of Class II facility licences The poster will also include other initiatives that may be of particular interest to COMP membership. © 2012 American Association of Physicists in Medicine.

To estimate the average annual rates of reduction of stunting, underweight and wasting for the period 1996 to 2011, and to evaluate whether Bangladesh will be expected to achieve the target of Millennium Development Goal 1C of reducing the prevalence of underweight by half by 2015. We used five nationwide, cross-sectional, Demographic and Health Survey data sets to estimate prevalence of undernutrition defined by stunting, underweight and wasting among children under 5 years of age using the WHO child growth standards. We then computed the average annual rates of reduction of prevalence of undernutrition using the formula derived by UNICEF. Finally, we projected the prevalence of undernutrition for the year 2015 using the estimated average annual rates of reduction. Nationwide covering Bangladesh. Children under 5 years of age (n 28,941). The prevalence of stunting decreased by 18.8% (from 60.0% to 41.2%), underweight by 16.0% (from 52.2% to 36.2%) and wasting by 5.1% (from 20.6% to 15.5%) during 1996 to 2011. The overall average annual rates of reduction were 2.84%, 2.69 % and 2.47%, respectively, for stunting, underweight and wasting. We forecast that in 2015, the prevalence of stunting, underweight and wasting will be 36.7%, 32.5% and 14.0%, respectively, at the national level. The prevalence of undernutrition is likely to remain high in rural areas, in the Sylhet division and in the poorest wealth quintile. Bangladesh is likely to achieve the Millennium Development Goal 1C target of reducing the prevalence of underweight by half by 2015. However, it is falling behind in reducing stunting and further investment is needed to reduce individual, household and environmental determinants of stunting in Bangladesh.

It offers a variety of major and minor academic programs designed to meet the mission and objectives of the college. The division`s pursuit to achieve excellence in science education is adversely impacted by the poor academic preparation of entering students and the lack of equipment, facilities and research participation, required to impart adequate academic training and laboratory skills to the students. The Astrophysics Science Division (ASD) at Goddard Space Flight Center (GSFC) is one of the largest and most diverse astrophysical organizations in the world, with activities spanning a broad range of topics in theory, observation, and mission and technology development. Scientific research is carried out over the entire electromagnetic spectrum from gamma rays to radio wavelengths as well as particle physics and gravitational radiation. Members of ASD also provide the scientific operations for three orbiting astrophysics missions WMAP, RXTE, and Swift, as well as the Science Support Center for the Fermi Gamma-ray Space Telescope. A number of key technologies for future missions are also under development in the Division, including X-ray mirrors, and new detectors operating at gamma-ray, X-ray, ultraviolet, infrared, and radio wavelengths. Cell cycle control is modified at meiosis compared to mitosis, because two divisions follow a single DNA replication event. Cyclin-dependent kinases (CDKs) promote progression through both meiosis and mitosis, and a central regulator of their activity is the APC/C (Anaphase Promoting Complex/Cyclosome) that is especially required for exit from mitosis. We have shown previously that OSD1 is involved in entry into both meiosis I and meiosis II in Arabidopsis thaliana; however, the molecular mechanism by which OSD1 controls these transitions has remained unclear. Here we show that OSD1 promotes meiotic progression through APC/C inhibition. Next, we explored the functional relationships between OSD1 and the genes known to control meiotic cell cycle transitions in Arabidopsis. Like osd1, cyca1;2/tam mutation leads to a premature exit from meiosis after the first division, while tdm mutants perform an aberrant third meiotic division after normal meiosis I and II. Remarkably, while tdm is epistatic to tam, osd1 is epistatic to tdm. We further show that the expression of a non-destructible CYCA1;2/TAM provokes, like tdm, the entry into a third meiotic division. Finally, we show that CYCA1;2/TAM forms an active complex with CDKA;1 that can phosphorylate OSD1 in vitro. We thus propose that a functional network composed of OSD1, CYCA1;2/TAM, and TDM controls three key steps of meiotic progression, in which OSD1 is a meiotic APC/C inhibitor.

Cell division cycle 45 promotes papillary thyroid cancer progression via regulating cell cycle.

Cell division cycle 45 was reported to be overexpressed in some cancer-derived cell lines and was predicted to be a candidate oncogene in cervical cancer. However, the clinical and biological significance of cell division cycle 45 in papillary thyroid cancer has never been investigated. We determined the expression level and clinical significance of cell division cycle 45 using The Cancer Genome Atlas, quantitative real-time polymerase chain reaction, and immunohistochemistry. A great upregulation of cell division cycle 45 was observed in papillary thyroid cancer tissues compared with adjacent normal tissues. Furthermore, overexpression of cell division cycle 45 positively correlates with more advanced clinical characteristics. Silence of cell division cycle 45 suppressed proliferation of papillary thyroid cancer cells via G1-phase arrest and inducing apoptosis. The oncogenic activity of cell division cycle 45 was also confirmed in vivo. In conclusion, cell division cycle 45 may serve as a novel biomarker and a potential therapeutic target for papillary thyroid cancer.

Mutations in TOO MANY MOUTHS (TMM), which encodes a receptor-like protein, cause stomatal patterning defects in Arabidopsis leaves but eliminate stomatal formation in stems. Stomatal development in wild-type and tmm stems was analyzed to define TMM function. Epidermal cells in young tmm stems underwent many asymmetric divisions characteristic of entry into the stomatal pathway. The resulting precursor cells, meristemoids, appropriately expressed cell fate markers such as pTMM:GFP. However, instead of progressing developmentally by forming a guard mother cell, the meristemoids arrested, dedifferentiated, and enlarged. Thus asymmetric divisions are necessary but not sufficient for stomatal formation in stems, and TMM promotes the fate and developmental progression of early precursor cells. Comparable developmental and mature stomatal phenotypes were also found in tmm hypocotyls and in the proximal flower stalk. TMM is also a positive regulator of meristemoid division in leaves suggesting that TMM generally promotes meristemoid activity. Our results are consistent with a model in which TMM interacts with other proteins to modulate precursor cell fate and progression in an organ and domain-specific manner. Finally, the consistent presence of a small number of dedifferentiated meristemoids in mature wild-type stems suggests that precursor cell arrest is a normal feature of Arabidopsis stem development.
