    Jeanette Scissum joined NASA’s Marshall Space Flight Center in 1964 after earning bachelor's and master's degrees in mathematics from Alabama A&M University. Scissum published a NASA report in 1967, “Survey of Solar Cycle Prediction Models,” which put forward techniques for improved forecasting of the sunspot cycle. In the mid-1970s she worked as a space scientist in the Space Environment Branch of Marshall’s Space Sciences Laboratory and later led activities in Marshall’s Atmospheric, Magnetospheric, and Plasmas in Space project.In 1975, Scissum wrote an article for the National Technical Association, “Equal Employment Opportunity and the Supervisor – A Counselor’s View,” which argued that many discrimination complaints could be avoided “through adequate and meaningful communication.” Scissum later worked at NASA Headquarters as a computer systems analyst responsible for analyzing and directing NASA management information and technical support systems.

    The Danish amateur scholar Christian Jürgensen Thomsen has often been described as a founder of modern "scientific" archaeology. Thomsen's innovation, this essay argues, reflects developments within neighboring fields, such as philology and history. He reacted against historians who limited themselves to histories of texts and therefore abandoned the earliest human history. Instead, he proposed a new history of objects, which included the entire history of humankind. Thomsen's work as director of the Royal Museum of Nordic Antiquities in Copenhagen was especially important for this renewal. The arrangement of artifacts not only helped him formulate his theories, but also allowed him to present his arguments in a language of objects. At the same time, Thomsen's definition of archaeology as a museum science placed his branch of archaeology in a closer relationship with other museum sciences, such as geology and comparative anatomy. From the 1840s, Thomsen's museum became a model for how the study of human artifacts could deliver scientific insights into human nature and the laws of human development.

    In German-speaking countries it was Carl Westphal who in 1877 offered the first precise definition of obsessive ideas and distinguished obsessive compulsive disorder (OCD) as an independent disorder in its own right. The criteria mentioned by him for establishing OCD gave rise to a debate on the character and classification of OCD but were not fully acknowledged by his colleagues at the time. In 1895 Westphal's student Robert Thomsen tried to substantiate all points in his teacher's theory that had raised criticism. Thus the works by Westphal and Thomsen are most relevant for the current conceptualization and definition of OCD, for they laid the basis for the present phenomenology, definition and classification of OCD according to ICD-10 and DSM-5. Apart from phenomenologically differentiating between obsessions (i.e. obsessive thoughts and impulses) and compulsions (i.e. compulsive actions and inhibitions), Westphal and Thomsen also laid the basis for most of the current diagnostic criteria. Thomsen led the way to current classifications by subdifferentiating OCD as an illness on its own on the one hand and obsessions and compulsions as symptoms accompanying other conditions on the other.

    We describe a large Brazilian consanguineous kindred with 3 clinically affected patients with a Thomsen myotonia phenotype. They carry a novel homozygous nonsense mutation in the CLCN1 gene (K248X). None of the 6 heterozygote carriers show any sign of myotonia on clinical evaluation or electromyography. These findings confirm the autosomal recessive inheritance of the novel mutation in this family, as well as the occurrence of phenotypic variability in the autosomal recessive forms of myotonia.     Thomsen-Friedenreich antigen (TF; Galbeta1-3GalNAcalpha1-) is expressed on many human carcinomas. Evidence suggests that TF-carrying tumor cells specifically bind asialoglycoprotein receptors on hepatocytes resulting in metastasis formation in the liver. We used an animal model to examine the feasibility of preventing metastasis formation by an antibody to TF. Treatment of Colon 26 cells with neuraminidase led to the exposure of TF, and consequently to a higher frequency of liver metastases in syngeneic Balb/c mice. This could be prevented by an antibody to TF (A78-G/A7), but not by a control antibody. The results may open up a new strategy for the prophylaxis of metastatic spread to the liver.

    GNE myopathy is an adult-onset progressive myopathy, resulting from mutations in GNE, the key enzyme of sialic acid synthesis. The pathomechanism of GNE myopathy likely involves aberrant sialylation, since administration of sialic acid itself, or its precursor, N-acetylmannosamine (ManNAc), rescued hyposialylation of GNE myopathy mice. Recently, clinical trials for GNE myopathy patients were initiated. A robust, noninvasive biomarker is highly desirable for diagnosis of GNE myopathy and for evaluating response to therapy. Since muscle biopsies of patients with GNE myopathy demonstrated hyposialylation of predominantly O-linked glycans, we analyzed the O-linked glycome of patients’ plasma proteins using mass spectrometry. Most patients showed increased plasma levels of the core 1 O-linked glycan, Thomsen-Friedenreich (T)-antigen and/or decreased amounts of its sialylated form, ST-antigen. In addition, compared to unaffected individuals, all analyzed patients had a consistently increased ratio of T-antigen to ST-antigen. Importantly, the T/ST ratios were in the normal range in a GNE myopathy patient treated with intravenous immunoglobulins as a source of sialic acid, indicating response to therapy. Natural history and clinical trial data will reveal whether T/ST ratios can be correlated to muscle function. These findings not only highlight plasma T/ST ratios as a robust blood-based biomarker for GNE myopathy, but may also help explain the pathology and course of the disease. PMID:25123033

    Thomsen-Friedenreich (TF) antigen belongs to the mucin-type tumor-associated carbohydrate antigen. Notably, TF antigen is overexpressed in colorectal cancer (CRC) but is rarely expressed in normal colonic tissue. Increased TF antigen expression is associated with tumor invasion and metastasis. In this study, we sought to validate a novel nanobeacon for imaging TF-associated CRC in a preclinical animal model. We developed and characterized the nanobeacon for use with fluorescence colonoscopy. In vivo imaging was performed on an orthotopic rat model of CRC. Both white light and fluorescence colonoscopy methods were utilized to establish the ratio-imaging index for the probe. The nanobeacon exhibited specificity for TF-associated cancer. Fluorescence colonoscopy using the probe can detect lesions at the stage which is not readily confirmed by conventional visualization methods. Further, the probe can report the dynamic change of TF expression as tumor regresses during chemotherapy. Data from this study suggests that fluorescence colonoscopy can improve early CRC detection. Supplemented by the established ratio-imaging index, the probe can be used not only for early detection, but also for reporting tumor response during chemotherapy. Furthermore, since the data obtained through in vivo imaging confirmed that the probe was not absorbed by the colonic mucosa, no registered toxicity is associated with this nanobeacon. Taken together, these data demonstrate the potential of this novel probe for imaging TF antigen as a biomarker for the early detection and prediction of the progression of CRC at the molecular level. © 2014 UICC.

    Recurrent ovarian cancer (OvCa) is thought to result in part from the inability to eliminate rare quiescent cancer stem cells (CSCs) that survive cytotoxic chemotherapy and drive tumor resurgence. The Sialyl-Thomsen-nouveau antigen (STn) is a carbohydrate moiety present on protein markers of CSCs in pancreatic, colon, and gastric malignancies. We have demonstrated that human OvCa cell lines contain varying levels of cells that independently express either STn or the ovarian CSC marker CD133. Here we determine co-expression of STn and CD133 in a subset of human OvCa cell lines. Analyses of colony and sphere forming capacity and of response to standard-of-care cytotoxic therapy suggest a subset of OvCa STn + cells display some CSC features. The effect of the anti-STn antibody-drug conjugates (ADCs) S3F-CL-MMAE and 2G12-2B2-CL-MMAE on OvCa cell viability in vitro and in vivo was also assessed. Treatment with S3F-CL-MMAE reduced the viability of two of three OvCa cell lines in vitro and exposure to either S3F-CL-MMAE or 2G12-2B2-CL-MMAE reduced OVCAR3-derived xenograft volume in vivo , depleting STn + tumor cells. In summary, STn + cells demonstrate some stem-like properties and specific therapeutic targeting of STn in ovarian tumors may be an effective clinical strategy to eliminate both STn + CSC and STn + non-CSC populations.

    The Thomsen-Friedenreich antigen (TF), or more precisely epitope, has been known as a pancarcinoma antigen. It consists of galactose-beta1-3-N-acetylgalactose. We have already described the expression of TF in the normal placenta. TF is expressed by the syncytium and by extravillous trophoblast cells. In this study, we investigated the expression of TF in the abort placenta. Frozen samples of human abort placentas (12 placentas), obtained from the first and second trimesters of pregnancy and, for comparison, samples of normal placentas (17 placentas) from the first, second and third trimesters of pregnancy, were used. Expression of TF was investigated by immunohistochemical methods. For identification of TF-positive cells in abort placentas, immunofluorescence methods were used. Evaluation of simple and double immunofluorescence was performed on a laser scanning microscope. Furthermore, we isolated trophoblast cells from first and third trimester placentas and evaluated cytokeratin 7 and Muc1 expression by immunofluorescence methods. We observed expression of TF antigen in the syncytiotrophoblasts layer of the placenta in all three trimesters of pregnancy in normal and abort placentas evaluated by immunohistochemical methods. There was no expression of TF antigen in the decidua of abort placentas. Immunofluorescence double staining of TF antigen and cytokeratin 7 showed reduced expression of both antigens in the abort decidua and co-expression of both antigens in the syncytiotrophoblast layer of normal and abort placentas. TF expression in the syncytiotrophoblast was reduced in abort placentas. In the isolated trophoblast cells, no TF expression was found, however, Muc1 expression was visualized. Expression of TF antigen was reduced in the first and second trimester abort decidua compared to the normal decidua during the same time of pregnancy.   20. Waveform Tomography of Two-Dimensional Three-Component Seismic Data for HTI Anisotropic Media

    Reservoirs with vertically aligned fractures can be represented equivalently by horizontal transverse isotropy (HTI) media. But inverting for the anisotropic parameters of HTI media is a challenging inverse problem, because of difficulties inherent in a multiple parameter inversion. In this paper, when we invert for the anisotropic parameters, we consider for the first time the azimuthal rotation of a two-dimensional seismic survey line from the symmetry of HTI. The established wave equations for the HTI media with azimuthal rotation consist of nine elastic coefficients, expressed in terms of five modified Thomsen parameters. The latter are parallel to the Thomsen parameters for describing velocity characteristics of weak vertical transverse isotropy media. We analyze the sensitivity differences of the five modified Thomsen parameters from their radiation patterns, and attempt to balance the magnitude and sensitivity differences between the parameters through normalization and tuning factors which help to update the model parameters properly. We demonstrate an effective inversion strategy by inverting velocity parameters in the first stage and updates the five modified Thomsen parameters simultaneously in the second stage, for generating reliably reconstructed models.

  4. Response of Velocity Anisotropy of Shale Under Isotropic and Anisotropic Stress Fields

    We investigated the responses of P-wave velocity and associated anisotropy in terms of Thomsen's parameters to isotropic and anisotropic stress fields on Longmaxi shales cored along different directions. An array of piezoelectric ceramic transducers allows us to measure P-wave velocities along numerous different propagation directions. Anisotropic parameters, including the P-wave velocity α along a symmetry axis, Thomsen's parameters ɛ and δ, and the orientation of the symmetry axis, could then be extracted by fitting Thomsen's weak anisotropy model to the experimental data. The results indicate that Longmaxi shale displays weakly intrinsic velocity anisotropy with Thomsen's parameters ɛ and δ being approximately 0.05 and 0.15, respectively. The isotropic stress field has only a slight effect on velocity and associated anisotropy in terms of Thomsen's parameters. In contrast, both the magnitude and orientation of the anisotropic stress field with respect to the shale fabric are important in controlling the evolution of velocity and associated anisotropy in a changing stress field. For shale with bedding-parallel loading, velocity anisotropy is enhanced because velocities with smaller angles relative to the maximum stress increase significantly during the entire loading process, whereas those with larger angles increase slightly before the yield stress and afterwards decrease with the increasing differential stress. For shale with bedding-normal loading, anisotropy reversal is observed, and the anisotropy is progressively modified by the applied differential stress. Before reaching the yield stress, velocities with smaller angles relative to the maximum stress increase more significantly and even exceed the level of those with larger angles. After reaching the yield stress, velocities with larger angles decrease more significantly.   5. Postoperative change of anti-Thomsen-Friedenreich and Tn IgG level: the follow-up study of gastrointestinal cancer patients.

    To study the influence of tumor removal on the serum level of IgG antibodies to tumor-associated Thomsen-Friedenreich (TF), Tn carbohydrate epitopes and xenogeneic alphaGal, and to elucidate on the change of the level during the follow-up as well as its association with the stage and morphology of the tumor and the values of blood parameters in gastrointestinal cancer. Sixty patients with gastric cancer and 34 patients with colorectal cancer in stages I-IV without distant metastases were subjected to follow-up. The level of antibodies in serum was determined by the enzyme-linked immunosorbent assay (ELISA) using synthetic polyacrylamide (PAA) glycoconjugates. Biochemical and haematological analyses were performed using automated equipment. In gastrointestinal cancer, the TF antibody level was found to have elevated significantly after the removal of G3 tumors as compared with the preoperative level (u = 278.5, P < 0.05). After surgery, the TF and Tn antibody level was elevated in the majority of gastric cancer patients (sign test, 20 vs 8, P < 0.05, and 21 vs 8, P < 0.05, respectively). In gastrointestinal cancer, the elevated postoperative level of TF, Tn and alphaGal antibodies was noted in most patients with G3 tumors (sign test, 22 vs 5, P < 0.01; 19 vs 6, P < 0.05; 24 vs 8, P < 0.01, respectively), but the elevation was not significant in patients with G1 + G2 resected tumors. The postoperative follow-up showed that the percentage of patients with G3 resected tumors of the digestive tract, who had a mean level of anti-TF IgG above the cut-off value (1.53), was significantly higher than that of patients with G1 + G2 resected tumors (c2 = 3.89, all patients; c2 = 5.34, patients without regional lymph node metastases; P < 0.05). The percentage of patients with a tumor in stage I, whose mean anti-TF IgG level remained above the cut-off value (1.26), was significantly higher than that of patients with the cancer in stages III-IV (c2 = 4.71, gastric cancer; c2 = 4

    The carbohydrate molecules Sialyl Lewis X (SLeX), Sialyl Lewis A (SLeA), Lewis Y (LeY) and Thomsen-Friedenreich antigen (TF) are known to mediate the adhesion between tumor cells and endothelium. They are used as serum markers in diagnosis and treatment in a broad spectrum of human carcinomas, but their expression profile and role in the development of cervical cancer remains unclear. The aim of this study was to investigate the expression of SLeX, SLeA, LeY and TF in normal cervical squamous epithelium, cervical dysplasia and cervical cancer. Slides of paraffin-embedded tissue were fixed and incubated with monoclonal antibodies against SLeX, SLeA, LeY and TF. Immunohistochemical staining was evaluated by using a semi-quantitative score (IRS Score). We found a significant difference of SLeA expression in invasive cervical cancer compared to normal epithelium (p=0.006) and all grades of dysplasia (p=0.002). The expression of SLeX in normal epithelium was less intense than in carcinoma in situ (p=0.036). Staining for LeY showed the weakest results of the investigated markers. Significant differences were found when normal epithelium was compared to CIN I (p=0.011), to CIN II (p=0.013) and to invasive cervical cancer (p=0.005). For TF, significant differences were found in normal epithelium compared to CIN I (p=0.011), CIN II (p=0.013) and compared to invasive cervical cancer (p=0.005). This is the first study on the expression of SLeA, SLeX, LeY and TF in normal cervical endothelium, cervical dysplasia, carcinoma in situ and invasive cervical cancer. Further studies and higher numbers are desirable.

  7. Elastic anisotropy of Opalinus Clay under variable saturation and triaxial stress

    A novel experimental method is introduced to estimate the Thomsen's elastic anisotropy parameters ɛ and δ of a transversely isotropic shale under variable stress and saturation conditions. The method consists in recording P-wave velocities along numerous paths on a cylindrical specimen using miniature ultrasonic transducers. Such an overdetermined set of measurements is specifically designed to reduce the uncertainty associated with the determination of Thomsen's δ parameter compared to the classical method for which a single off-axis measurement is used (usually at 45° to the specimen's axis). This method is applied to a specimen of Opalinus Clay recovered from the Mont-Terri Underground Research Laboratory in Switzerland. The specimen is first saturated with brine at low effective pressure and then subjected to an effective pressure cycle up to 40 MPa, followed by a triaxial loading up to failure. During saturation and deformation, the evolution of P-wave velocities along a maximum of 240 ray paths is monitored and Thomsen's parameters α, ɛ and δ are computed by fitting Thomsen's weak anisotropy model to the data. The values of ɛ and δ obtained at the highest confining pressures reached during the experiment are comparable with those predicted from X-ray diffraction texture analysis and modelling for Opalinus Clay reported in the literature. These models neglect the effect of soft-porosity on elastic properties, but become relevant when soft porosity is closed at high effective pressure.

  18. Azimuthal Seismic Amplitude Variation with Offset and Azimuth Inversion in Weakly Anisotropic Media with Orthorhombic Symmetry

    Seismic amplitude variation with offset and azimuth (AVOaz) inversion is well known as a popular and pragmatic tool utilized to estimate fracture parameters. A single set of vertical fractures aligned along a preferred horizontal direction embedded in a horizontally layered medium can be considered as an effective long-wavelength orthorhombic medium. Estimation of Thomsen's weak-anisotropy (WA) parameters and fracture weaknesses plays an important role in characterizing the orthorhombic anisotropy in a weakly anisotropic medium. Our goal is to demonstrate an orthorhombic anisotropic AVOaz inversion approach to describe the orthorhombic anisotropy utilizing the observable wide-azimuth seismic reflection data in a fractured reservoir with the assumption of orthorhombic symmetry. Combining Thomsen's WA theory and linear-slip model, we first derive a perturbation in stiffness matrix of a weakly anisotropic medium with orthorhombic symmetry under the assumption of small WA parameters and fracture weaknesses. Using the perturbation matrix and scattering function, we then derive an expression for linearized PP-wave reflection coefficient in terms of P- and S-wave moduli, density, Thomsen's WA parameters, and fracture weaknesses in such an orthorhombic medium, which avoids the complicated nonlinear relationship between the orthorhombic anisotropy and azimuthal seismic reflection data. Incorporating azimuthal seismic data and Bayesian inversion theory, the maximum a posteriori solutions of Thomsen's WA parameters and fracture weaknesses in a weakly anisotropic medium with orthorhombic symmetry are reasonably estimated with the constraints of Cauchy a priori probability distribution and smooth initial models of model parameters to enhance the inversion resolution and the nonlinear iteratively reweighted least squares strategy.     Three mantras often guide species and ecosystem management: (i) for preventing invasions by harmful species, 'early detection and rapid response'; (ii) for conserving imperilled native species, 'protection of biodiversity hotspots'; and (iii) for assessing biosecurity risk, 'an ounce of prevention equals a pound of cure.' However, these and other management goals are elusive when traditional sampling tools (e.g. netting, traps, electrofishing, visual surveys) have poor detection limits, are too slow or are not feasible. One visionary solution is to use an organism's DNA in the environment (eDNA), rather than the organism itself, as the target of detection. In this issue of Molecular Ecology, Thomsen et al. (2012) provide new evidence demonstrating the feasibility of this approach, showing that eDNA is an accurate indicator of the presence of an impressively diverse set of six aquatic or amphibious taxa including invertebrates, amphibians, a fish and a mammal in a wide range of freshwater habitats. They are also the first to demonstrate that the abundance of eDNA, as measured by qPCR, correlates positively with population abundance estimated with traditional tools. Finally, Thomsen et al. (2012) demonstrate that next-generation sequencing of eDNA can quantify species richness. Overall, Thomsen et al. (2012) provide a revolutionary roadmap for using eDNA for detection of species, estimates of relative abundance and quantification of biodiversity. © 2012 Blackwell Publishing Ltd.

    Stereotomography is a slope tomographic method which gives good results for background velocity model estimation in 2-D isotropic media. We develop here the extension of the method to 3-D general anisotropic media for PP and PS events. We do not take into account the issue of shear wave degeneracy. As in isotropic media, the sensitivity matrix of the inversion can be computed by paraxial ray tracing. We introduce a `constant Z stereotomography' approach, which can reduce the size of the sensitivity matrix. Based on ray perturbation theory, we give all the derivatives of stereotomography data parameters with respect to model parameters in a 3-D general anisotropic medium. These general formulas for the derivatives can also be used in other applications that rely on anisotropic ray perturbation theory. In particular, we obtain derivatives of the phase velocity with respect to position, phase angle and elastic medium parameters, all for general anisotropic media. The derivatives are expressed using the Voigt notation for the elastic medium parameters. We include a Jacobian that allows to change the model parametrization from Voigt to Thomsen parameters. Explicit expressions for the derivatives of the data are given for the case of 2-D tilted transversely isotropic (TTI) media. We validate the method by single-parameter estimation of each Thomsen parameter field of a 2-D TTI synthetic model, where data are modelled by ray tracing. For each Thomsen parameter, the estimated velocity field fits well with the true velocity field.

    In this article, I will argue that Winterson's use of satire and the common tropes of science fiction in her 2007 novel The Stone Gods provides an effective and important critique of the gender discrepancies arising in the implementation of aesthetic medical biotechnologies under the logic of neoliberal consumerism. In particular, engaging with aspects of Winterson's fictional landscape in Part 1 of The Stone Gods, I will explore the themes of bodily normalization, the medicalization of youth and appearance, and the notion that biotechnologies such as cosmetic surgery can inculcate happiness through some sort of "psychological cure." Ultimately, I will argue that Winterson's aim in this novel is to raise important questions about where rising standards of enhancement and appearance, implemented through biotechnologies, will take us and, furthermore, to demonstrate that the problems of the human condition require more than the surface fixes offered by consumption, technological innovation, and narcissistic body projects.

    We formulate the derivatives of the phase and group velocities for each of the anisotropic parameters in a tilted transversely isotropic medium (TTI-medium). This is a common geological model in seismic exploration and has five elastic moduli or related Thomsen parameters and two orientation angles defining the axis of symmetry of the rock. We present two independent methods to compute the derivatives and examine the formulae with real anisotropic rocks. The formulations and numerical computations do not encounter any singularity problem when applied to the two quasi shear waves, which is a problem with other approaches. The two methods yield the same results, which show in a quantitative way the sensitivity behaviour of the phase and the group velocities to all of the elastic moduli or Thomsen's anisotropic parameters as well as the orientation angles in the 2D and 3D cases. One can recognize the dominant (strong effect) and weak (or 'dummy') parameters for the three seismic body-wave modes (qP, qSV, qSH) and their effective domains over the whole range of phase-slowness directions. These sensitivity patterns indicate the possibility of nonlinear kinematic inversion with the three wave modes for determining the anisotropic parameters and imaging an anisotropic medium.

    Galectin-1 (gal-1) belongs to the family of β-galactoside-binding proteins which primarily recognizes the Galβ1-4GlcNAc sequences of oligosaccharides associated with several cell surface glycoconjugates. The lectin recognizes correspondent glycoepitopes on human breast cancer cells. Galectin-1 is expressed both in normal and malignant tissues. Lymphatic organs naturally possessing high rates of apoptotic cells, express high levels of Galectin-1. Furthermore galectin-1 can initiate T cell apoptosis. Binding of galectin-1 to trophoblast tumor cells presenting the oncofetal Thomsen-Friedenreich (TF) carbohydrate antigen inhibits tumor cell proliferation. In this study we examined the impact galectin-1 has in vitro on cell proliferation, apoptotic potential and metabolic activity of MCF-7 and T-47D breast cancer cells in dependence to their expression of the Thomsen-Friedenreich (TF) tumor antigen. For proliferation and apoptosis assays cells were grown in presence of 10, 30 and 60 μg gal-1/ml medium. Cell proliferation was determined by a BrdU uptake ELISA. Detection of apoptotic cells was done by M30 cyto death staining, in situ nick translation and by a nucleosome ELISA method. Furthermore we studied the impact galectin-1 has on the metabolic activity of MCF-7 and T-47D cells in a homotypic three-dimensional spheroid cell culture model mimicking a micro tumour environment. Gal-1 inhibited proliferation of MCF-7 cells (strong expression of the TF epitope) but did not significantly change proliferation of T-47D cells (weak expression of the TF epitope). The incubation of MCF-7 cells with gal-1 raised number of apoptotic cells significantly. Treating the spheroids with 30 μg/ml galectin-1 in addition to standard chemotherapeutic regimes (FEC, TAC) resulted in further suppression of the metabolic activity in MCF-7 cells whereas T-47D cells were not affected.     In this report, we challenge a common perception that tumor embolism is a size-limited event of mechanical arrest, occurring in the first capillary bed encountered by blood-borne metastatic cells. We tested the hypothesis that mechanical entrapment alone, in the absence of tumor cell adhesion to blood vessel walls, is not sufficient for metastatic cell arrest in target organ microvasculature. The in vivo metastatic deposit formation assay was used to assess the number and location of fluorescently labeled tumor cells lodged in selected organs and tissues following intravenous inoculation. We report that a significant fraction of breast and prostate cancer cells escapes arrest in a lung capillary bed and lodges successfully in other organs and tissues. Monoclonal antibodies and carbohydrate-based compounds (anti-Thomsen-Friedenreich antigen antibody, anti-galectin-3 antibody, modified citrus pectin, and lactulosyl-l-leucine), targeting specifically beta-galactoside-mediated tumor-endothelial cell adhesive interactions, inhibited by >90% the in vivo formation of breast and prostate carcinoma metastatic deposits in mouse lung and bones. Our results indicate that metastatic cell arrest in target organ microvessels is not a consequence of mechanical trapping, but is supported predominantly by intercellular adhesive interactions mediated by cancer-associated Thomsen-Friedenreich glycoantigen and beta-galactoside-binding lectin galectin-3. Efficient blocking of beta-galactoside-mediated adhesion precludes malignant cell lodging in target organs.

  13. Seismic Imaging of VTI, HTI and TTI based on Adjoint Methods

    Recent studies show that isotropic seismic imaging based on adjoint method reduces low-frequency artifact caused by diving waves, which commonly occur in two-wave wave-equation migration, such as Reverse Time Migration (RTM). Here, we derive new expressions of sensitivity kernels for Vertical Transverse Isotropy (VTI) using the Thomsen parameters (ɛ, δ, γ) plus the P-, and S-wave speeds (α, β) as well as via the Chen & Tromp (GJI 2005) parameters (A, C, N, L, F). For Horizontal Transverse Isotropy (HTI), these parameters depend on an azimuthal angle φ, where the tilt angle θ is equivalent to 90°, and for Tilted Transverse Isotropy (TTI), these parameters depend on both the azimuth and tilt angles. We calculate sensitivity kernels for each of these two approaches. Individual kernels ("images") are numerically constructed based on the interaction between the regular and adjoint wavefields in smoothed models which are in practice estimated through Full-Waveform Inversion (FWI). The final image is obtained as a result of summing all shots, which are well distributed to sample the target model properly. The impedance kernel, which is a sum of sensitivity kernels of density and the Thomsen or Chen & Tromp parameters, looks crisp and promising for seismic imaging. The other kernels suffer from low-frequency artifacts, similar to traditional seismic imaging conditions. However, all sensitivity kernels are important for estimating the gradient of the misfit function, which, in combination with a standard gradient-based inversion algorithm, is used to minimize the objective function in FWI.

  15. Laboratory Experiments Enabling Electron Beam use in Tenuous Space Plasmas

    A mission concept is under development which involves firing a spacecraft-mounted electron beam from Earth's magnetosphere to connect distant magnetic field lines in real time. To prevent excessive spacecraft charging and consequent beam return, the spacecraft must be neutralized in the tenuous plasma environment of the magnetosphere. Particle-In-Cell (PIC) simulations suggest neutralization can be accomplished by emitting a neutral plasma with the electron beam. Interpretation of these simulations also led to an ion emission model in which ion current is emitted from a quasi-neutral plasma as defined by the space charge limit [1,2]. Experiments were performed at the University of Michigan's Plasmadynamics and Electric Propulsion Laboratory (PEPL) to help validate the ion emission model. A hollow cathode plasma contactor was used as a representative spacecraft and charged with respect to the chamber walls to examine the effect of spacecraft charging on ion emission. Retarding Potential Analyzer (RPA) measurements were performed to understand ion flow velocity as this parameter relates directly to the expected space charge limit. Planar probe measurements were also made to identify where ion emission primarily occurred and to determine emission current density levels. Evidence of collisions within the plasma (particularly charge exchange collisions) and a simple model predicting emitted ion velocities are presented. While a detailed validation of the ion emission model and of the simulation tools used in [1,2] is ongoing, these measurements add to the physical understanding of ion emission as it may occur in the magnetosphere. 1.     Optical region spectrophotometry at 3300-7850 A has been obtained for three helium rich stars, HR 1890, Sigma Ori E, and HD 37776, of the Orion OB1 Association. New uvby-beta photometry of HR 1890 and HD 37776 as well as published data are also used to investigate the variability of these stars. A new period of 1.53862 days was determined for HD 37776. For all three stars H-beta varies in antiphase with strong He I lines. The spectrophotometric bandpass containing the strong He I line at 4471 A varies in phase with the R index of Pedersen and Thomsen (1977). Evidence is found for weak absorption features which appear to be an extension of the 5200 A feature seen in cooler CP stars.

  18. Rotation Rate of Saturn's Magnetosphere using CAPS Plasma Measurements

    We present the present status of an investigation of the rotation rate of Saturn's magnetosphere using a 3D velocity moment technique being developed at Goddard which is similar to the 2D version used by Sittler et al. for SOI and similar to that used by Thomsen et al.. This technique allows one to nearly cover the full energy range of the Cassini Plasma Spectrometer (CAPS) IMS from 1 V . E/Q < 50 kV. Since our technique maps the observations into a local inertial frame, it does work during roll maneuvers. We make comparisons with the bi-Maxwellian fitting technique developed by Wilson et al. and the similar velocity moment technique by Thomsen et al. . We concentrate our analysis when ion composition data is available, which is used to weight the non-compositional data, referred to as singles data, to separate H+, H2+ and water group ions (W+) from each other. The chosen periods have high enough telemetry rates (4 kbps or higher) so that coincidence ion data, similar to that used by Sittler et al. for SOI is available. The ion data set is especially valuable for measuring flow velocities for protons, which are more difficult to derive using singles data within the inner magnetosphere, where the signal is dominated by heavy ions (i.e., proton peak merges with W+ peak as low energy shoulder). Our technique uses a flux function, which is zero in the proper plasma flow frame, to estimate fluid parameter uncertainties. The comparisons investigate the experimental errors and potential for systematic errors in the analyses, including ours. The rolls provide the best data set when it comes to getting 4PI coverage of the plasma but are more susceptible to time aliasing effects. In the future we will then make comparisons with magnetic field observations, Saturn ionosphere conductivities as presently known and the field aligned currents necessary for the planet to enforce corotation of the rotating plasma.

    Seismic wave propagation through the earth is often strongly affected by the presence of fractures. When these fractures are filled with fluids (oil, gas, water, CO{sub 2}, etc.), the type and state of the fluid (liquid or gas) can make a large difference in the response of the seismic waves. This paper will summarize some early work of the author on methods of deconstructing the effects of fractures, and any fluids within these fractures, on seismic wave propagation as observed in reflection seismic data. Methods to be explored here include Thomsen's anisotropy parameters for wave moveout (since fractures often inducemore » elastic anisotropy), and some very convenient fracture parameters introduced by Sayers and Kachanov that permit a relatively simple deconstruction of the elastic behavior in terms of fracture parameters (whenever this is appropriate).« less

  20. Studying Spacecraft Charging via Numerical Simulations

    The electrical charging of spacecraft due to bombarding charged particles can affect their performance and operation. We study this charging using CPIC; a particle-in-cell code specifically designed for studying plasma-material interactions [1]. CPIC is based on multi-block curvilinear meshes, resulting in near-optimal computational performance while maintaining geometric accuracy. Relevant plasma parameters are imported from the SHIELDS framework (currently under development at LANL), which simulates geomagnetic storms and substorms in the Earth's magnetosphere.     An axiom of Thomas Hobbes states that "people are never more helpless than when the force meant to protect their rights turns against them." Hobbes' axiom holds true today, with Amnesty International reporting that hundreds of thousands have been murdered by their governments. This article examines the medicolegal aspects of an investigation into the deaths of two Salvadoran peasants who were reportedly tortured and executed by soldiers in February 1988. One of the authors, Thomsen, participated in the investigation as a court-ordered expert, and as a representative of the American Association for the Advancement of Science and of a Salvadoran legal aid organization. His necropsy findings are reported with observations and comments. The article concludes with suggestions for initiatives that might be undertaken by individual physicians and institutions to improve the quality and impartiality of medicolegal investigations into political killings.

    Dr. Louis J. Battan died on October 29, 1986, after a short illness. Although he had been a professor of atmospheric sciences at the University of Arizon a since 1958 and director of the Institute of Atmospheric Physics there from 1973 to 1982 (and associate director from 1958 to 1973), his activities on the national and international scenes were so varied that he was known in different ways by the numerous communities with which he was involved. However, all who knew him shared his uncommonly good humor, took pride in his friendship, and respected his wisdom and good judgment. Lou is survived by his wife Jeanette, daughter Suzette, and son Paul.Battan was born in New York City on February 9, 1923. He was the second of four sons born to Annibale and Luisa Battan, immigrants to the United States from the village of Vigo in the Tyrolean Alps of Austria. He grew up in Brooklyn, N.Y., in a family environment that encouraged the highest moral standards and a devotion to hard work and self-reliance, qualities that remained with him throughout his life.

    We present the present status of an investigation of the rotation rate of Saturn 's magnetosphere using a 3D velocity moment technique being developed at Goddard which is similar to the 2D version used by Sittler et al. (2005) [1] for SOI and similar to that used by Thomsen et al. (2010). This technique allows one to nearly cover the full energy range of the CAPS IMS from 1 V less than or equal to E/Q less than 50 kV. Since our technique maps the observations into a local inertial frame, it does work during roll manoeuvres. We have made comparisons with Wilson et al. (2008) [2] (2005-358 and 2005-284) who performs a bi-Maxwellian fit to the ion singles data and our results are nearly identical. We will also make comparisons with results by Thomsen et al. (2010) [3]. Our analysis uses ion composition data to weight the non-compositional data, referred to as singles data, to separate H+, H2+ and water group ions (W+) from each other. The ion data set is especially valuable for measuring flow velocities for protons, which are more difficult to derive using singles data within the inner magnetosphere, where the signal is dominated by heavy ions (i.e., proton peak merges with W+ peak as low energy shoulder). Our technique uses a flux function, which is zero in the proper plasma flow frame, to estimate fluid parameter uncertainties. The comparisons investigate the experimental errors and potential for systematic errors in the analyses, including ours. The rolls provide the best data set when it comes to getting 4PI coverage of the plasma but are more susceptible to time aliasing effects. Since our analysis is a velocity moments technique it will work within the inner magnetosphere where pickup ions are important and velocity distributions are non-Maxwellian. So, we will present results inside Enceladus' L shell and determine if mass loading is important.   4. Reactive transport simulation via combination of a multiphase-capable transport code for unstructured meshes with a Gibbs energy minimization solver of geochemical equilibria

    We apply a new, C++-based computational model for hydrothermal fluid-rock interaction and scale formation in geothermal reservoirs. The model couples the Complex System Modelling Platform (CSMP++) code for fluid flow in porous and fractured media (Matthai et al., 2007) with the Gibbs energy minimization numerical kernel GEMS3K of the GEM-Selektor (GEMS3) geochemical modelling package (Kulik et al., 2010) in a modular fashion. CSMP++ includes interfaces to commercial file formats, accommodating complex geometry construction using CAD (Rhinoceros) and meshing (ANSYS) software. The CSMP++ approach employs finite element-finite volume spatial discretization, implicit or explicit time discretization, and operator splitting. GEMS3K can calculate complex fluid-mineral equilibria based on a variety of equation of state and activity models. A selection of multi-electrolyte aqueous solution models, such as extended Debye-Huckel, Pitzer (Harvie et al., 1984), EUNIQUAC (Thomsen et al., 1996), and the new ELVIS model (Hingerl et al., this conference), makes it well-suited for application to a wide range of geothermal conditions. An advantage of the GEMS3K solver is simultaneous consideration of complex solid solutions (e.g., clay minerals), gases, fluids, and aqueous solutions. Each coupled simulation results in a thermodynamically-based description of the geochemical and physical state of a hydrothermal system evolving along a complex P-T-X path. The code design allows efficient, flexible incorporation of numerical and thermodynamic database improvements. We demonstrate the coupled code workflow and applicability to compositionally and physically complex natural systems relevant to enhanced geothermal systems, where temporally and spatially varying chemical interactions may take place within diverse lithologies of varying geometry.   5. Calculational investigation of impact cratering dynamics - Material motions during the crater growth period

    The considered investigation was conducted in connection with studies which are to provide a better understanding of the detailed dynamics of impact cratering processes. Such an understanding is vital for a comprehension of planetary surfaces. The investigation is the continuation of a study of impact dynamics in a uniform, nongeologic material at impact velocities achievable in laboratory-scale experiments conducted by Thomsen et al. (1979). A calculation of a 6 km/sec impact of a 0.3 g spherical 2024 aluminum projectile into low strength (50 kPa) homogeneous plasticene clay has been continued from 18 microseconds to past 600 microseconds.     IBC's Drug Discovery Technology Series is a group of conferences highlighting technological advances and applications in niche areas of the drug discovery pipeline. This 2-day meeting focused on G-protein-coupled receptors (GPCRs), probably the most important and certainly the most valuable class of targets for drug discovery. The meeting was chaired by J Beesley (Vice President, European Business Development for LifeSpan Biosciences, Seattle, USA) and included 17 presentations on various aspects of GPCR activity, drug screens and therapeutic analyses. Keynote Addresses covered two of the emerging areas in GPCR regulation; receptor dimerisation (G Milligan, Professor of Molecular Pharmacology and Biochemistry, University of Glasgow, UK) and proteins that interact with GPCRs (J Bockaert, Laboratory of Functional Genomics, CNRS Montpellier, France). A third Keynote Address from W Thomsen (Director of GPCR Drug Screening, Arena Pharmaceuticals, USA) discussed Arena's general approach to drug discovery and illustrated this with reference to the development of an agonist with potential efficacy in Type II diabetes.

  7. Born scattering and inversion sensitivities in viscoelastic transversely isotropic media

    We analyse the scattering of seismic waves from anisotropic-viscoelastic inclusions using the Born approximation. We consider the specific case of Vertical Transverse Isotropic (VTI) media with low-loss attenuation and weak anisotropy such that second- and higher-order contributions from quality factors and Thomsen parameters are negligible. To accommodate the volume scattering approach, the viscoelastic VTI media is broken into a homogeneous viscoelastic reference medium with distributed inclusions in both viscoelastic and anisotropic properties. In viscoelastic reference media in which all propagations take place, wave modes are of P-wave type, SI-wave type and SII-wave type, all with complex slowness and polarization vectors. We generate expressions for P-to-P, P-to-SI, SI-to-SI and SII-to-SII scattering potentials, and demonstrate that they reduce to previously derived isotropic results. These scattering potential expressions are sensitivity kernels related to the Fréchet derivatives which provide the weights for multiparameter full waveform inversion updates.

    Picosecond laser ultrasonics uses femtosecond laser pulses for the generation and detection of acoustic pulses with a typical duration between few picoseconds and few hundreds of pico seconds. The shorter the duration of the acoustic pulse is, the more precisely could be made the measurements of the film thickness [C. Thomsen et al., Phys. Rev. B 34, 4129 (1986)] and the elastic modulus by pulse-echo method or through Brillouin scattering detection. In this short communication we report the results of the evaluation of the properties of nanoporous silicon oxide thin films which present potential low-k and thermal barrier properties and are also of great interest for the microelectronic industry to replace the traditional silicate glass films in order to decrease the resistance-capacitance transition delay in the VLSI circuits. Most of the studies that have been carried so far have treated the optical properties of such structures. We report the results of the evaluation of acoustic properties of nanoporous thin films.

    The grant "Taus at ATLAS" supported the group of Sarah Demers at Yale University over a period of 8.5 months, bridging the time between her Early Career Award and her inclusion on Yale's grant cycle within the Department of Energy's Office of Science. The work supported the functioning of the ATLAS Experiment at CERN's Large Hadron Collider and the analysis of ATLAS data. The work included searching for the Higgs Boson in a particular mode of its production (with a W or Z boson) and decay (to a pair of tau leptons.) This was part of a broad program ofmore » characterizing the Higgs boson as we try to understand this recently discovered particle, and whether or not it matches our expectations within the current standard model of particle physics. In addition, group members worked with simulation to understand the physics reach of planned upgrades to the ATLAS experiment.     The development of antibodies against specific glycan epitopes poses a significant challenge due to difficulties obtaining desired glycans at sufficient quantity and purity, and the fact that glycans are usually weakly immunogenic. To address this challenge, we leveraged the potent immunostimulatory activity of bacterial outer membrane vesicles (OMVs) to deliver designer glycan epitopes to the immune system. This approach involved heterologous expression of two clinically important glycans, namely polysialic acid (PSA) and Thomsen-Friedenreich antigen (T antigen) in hypervesiculating strains of non-pathogenic Escherichia coli. The resulting glycOMVs displayed structural mimics of PSA or T antigen on their surfaces, and induced highmore » titers of glycan-specific IgG antibodies following immunization in mice. In the case of PSA glycOMVs, serum antibodies potently killed Neisseria meningitidis serogroup B (MenB), whose outer capsule is PSA, in a serum bactericidal assay.   11. Novel anti-Sialyl-Tn monoclonal antibodies and antibody-drug conjugates demonstrate tumor specificity and anti-tumor activity.

    Targeted therapeutics that can differentiate between normal and malignant tumor cells represent the ideal standard for the development of a successful anti-cancer strategy. The Sialyl-Thomsen-nouveau antigen (STn or Sialyl-Tn, also known as CD175s) is rarely seen in normal adult tissues, but it is abundantly expressed in many types of human epithelial cancers. We have identified novel antibodies that specifically target with high affinity the STn glycan independent of its carrier protein, affording the potential to recognize a wider array of cancer-specific sialylated proteins. A panel of murine monoclonal anti-STn therapeutic antibodies were generated and their binding specificity and efficacy were characterized in vitro and in in vivo murine cancer models. A subset of these antibodies were conjugated to monomethyl auristatin E (MMAE) to generate antibody-drug conjugates (ADCs). These ADCs demonstrated in vitro efficacy in STn-expressing cell lines and significant tumor growth inhibition in STn-expressing tumor xenograft cancer models with no evidence of overt toxicity.

  12. Seismic anisotropy in gas-hydrate- and gas-bearing sediments on the Blake Ridge, from a walkaway vertical seismic profile

    We present results from an analysis of anisotropy in marine sediments using walkaway vertical seismic profiles from the Blake Ridge, offshore South Carolina. We encountered transverse isotropy (TI) with a vertical symmetry axis in a gas-hydrate-bearing unit of clay and claystone with Thomsen parameters ?? = 0.05 ?? 0.02 and ?? = 0.04 ?? 0.06. TI increased to ?? = 0.16 ?? 0.04 and ?? = 0.19 ?? 0.12 in the underlying gas zone. Rock physics modeling suggests that the observed TI is caused by a partial alignment of clay particles rather than high-velocity gas-hydrate veins. Similarly, the increase of TI in the gas zone is not caused by thin low-velocity gas layers but rather, we speculate, by the sharp contrast between seismic properties of an anisotropic sediment frame and elongated gas-bearing pore voids. Our results underscore the significance of anisotropy for integrating near-vertical and wide-angle seismic data.

    Microsolvated bimolecular nucleophilic substitution (SN2) reaction of monohydrated hydrogen peroxide anion [HOO-(H2O)] with methyl chloride (CH3Cl) has been investigated with direct chemical dynamics simulations at the M06-2X/6-31+G(d,p) level of theory. Dynamic exit-channel pathways and corresponding reaction mechanisms at the atomic level are revealed in detail. Accordingly, a product distribution of 0.85:0.15 is obtained for Cl-:Cl-(H2O), which is consistent with a previous experiment [D. L. Thomsen et al. J. Am. Chem. Soc. 135, 15508 (2013)]. Compared with the HOO- + CH3Cl SN2 reaction, indirect dynamic reaction mechanisms are enhanced by microsolvation for the HOO-(H2O) + CH3Cl SN2 reaction. On the basis of our simulations, further crossed molecular beam imaging experiments are highly suggested for the SN2 reactions of HOO- + CH3Cl and HOO-(H2O) + CH3Cl.

  16. Plasma Transport and Magnetic Flux Circulation in Saturn's Magnetosphere

    Radial transport of plasma in the rapidly rotating magnetospheres is an important dynamical process. Radial transport is due to the centrifugally driven interchange instability and magnetodisc reconnection, allowing net mass to be transported outward while conserving magnetic flux. Using Cassini Plasma Spectrometer instrument (CAPS) data products (e.g., Thomsen et al., [2010]; Wilson et al., [2017]) we estimate plasma mass and magnetic flux transport rates as functions of radial distance and local time. The physical requirement for zero net magnetic flux transport provides a key benchmark for assessing the validity of our mass transport estimate. We also evaluate magnetodisc stability using a two-dimensional axisymmetric equilibrium model [Caudal, 1986]. Observed local properties (e.g., specific entropy and estimates of flux tube mass and entropy content) are compared with modeled equilibrium conditions such that departures from equilibrium can be correlated with radial flows and local magnetic field structure. Finally, observations of specific entropy indicate that plasma is non-adiabatic heated during transport. However, the values of specific entropy are well organized in inner magnetosphere (i.e. L<10), and become widely scattered in the middle magnetosphere, suggesting that the transport dynamics of the inner and middle magnetosphere are different.

    Myotonia congenita (MC) is a non-dystrophic myotonia inherited either in dominant (Thomsen) or recessive (Becker) form. MC is due to an abnormal functioning of skeletal muscle voltage-gated chloride channel (CLCN1), but the genotype/phenotype correlation remains unclear. A 48-year-old man, from consanguineous parents, presented with a fixed muscle weakness, muscle atrophy, and a cognitive impairment. Notably, his brother presented the same mutation but with a different phenotype, mainly involving cognitive function. The patient was submitted to cognitive assessment, needle electromyography, brain and muscle MRI, and genetic analysis. The Milan Overall Dementia Assessment showed short-term memory, verbal fluency and verbal intelligence impairment. His genetic analysis showed a recessive splice-site mutation in the CLCN1 gene (IVS19+2T>A). Muscle MRI revealed a symmetric and bilateral fat infiltration of the tensor of fascia lata, gluteus medius, and gluteus maximus muscles, associated to mild atrophy. Recessive myotonia congenita was diagnosed. Further studies should establish if and to which extent the CLCN1 mutation is responsible for this c MC phenotype, taking into account a gene-gene and /or a gene-environment.

  18. A determination of the L dependence of the radial diffusion coefficient for protons in Jupiter's inner magnetosphere

    In a previous paper (Thomsen et al., 1977), a technique was proposed for estimating the radial diffusion coefficient (n) in the inner magnetosphere of Jupiter from the observations of the sweeping effect of the inner Jovian satellites on the fluxes of the energetic charged particles. The present paper extends this technique to permit the unique identification of the parameters D sub O and n, where the diffusion coefficient is assumed to be of the form D = D sub O L to the nth. The derived value of D sub O depends directly on assumptions regarding the nature and efficiency of the loss mechanism operating on the particles, while the value of n depends only on the assumed width of the loss region. The extended technique is applied to the University of Iowa Pioneer 11 proton data, leading to values of n of about O and D(6) of about 3 x 10 to the -8th (R sub J)-squared/sec, when satellite sweepup losses are assumed to be the only loss operating on the protons. The small value of n is strong evidence that the radial diffusion is driven by ionospheric winds.

    Members of the copepod genus Caligus Müller, 1785 (Siphonostomatoida: Caligidae) are commonly referred to as sea lice. Virtually all of the more than 450 species utilize marine fishes as hosts, however, an increasing number of records from marine plankton samples shows that at least some species reside in the water column during their adult phase. Members of three different genera, Caligus, Lepeophtheirus von Nordmann, 1832, and Metacaligus Thomsen, 1949 have been reported from plankton samples off eastern Asia and in the north-western Atlantic. Thirteen species have so far been consistently reported from the plankton only with no information on their hosts. Here we review the various hypotheses previously proposed to explain the presence of caligids in the water column, i.e. accidental occurrence, behavioral detachment from the host during mate location, and host-switching. The discovery of adults of two species of Caligus with no other developmental stages on their teleost hosts, suggests an ontogenetic host-switching after the final molt since both species also occurred in plankton samples. A checklist of all caligid records from the marine plankton, including known host data, is presented.

    Jacalin, the major protein from the jackfruit (Artocarpus heterophyllus) seeds, is a tetrameric two-chain lectin (molecular mass 65 kDa) combining a heavy alpha chain of 133 amino acid residues with a light beta chain of 20-21 amino acid residues. It is highly specific for the alpha-O-glycoside of the disaccharide Thomsen-Friedenreich antigen (Gal beta1-3GalNAc), even in its sialylated form. This property has made jacalin suitable for studying various O-linked glycoproteins, particularly human IgA1. Jacalin's uniqueness in being strongly mitogenic for human CD4+ T lymphocytes has made it a useful tool for the evaluation of the immune status of patients infected with human immunodeficiency virus (HIV)-1. The abundance of source material for the production of jacalin, its ease of purification, yield and stability have made it an attractive cost-effective lectin. It has found applications in diverse areas such as the isolation of human plasma glycoproteins (IgA1, C1-inhibitor, hemopexin, alpha2-HSG), the investigation of IgA-nephropathy, the analysis of O-linked glycoproteins and the detection of tumours.

    For successful carbohydrate based anti-cancer vaccines, it is critical that B cells are activated to secret antibodies targeting the tumor associated carbohydrate antigens (TACAs). Despite the availability of many TACA based constructs, systematic understanding of the effects of structural features on anti-glycan antibody responses is lacking. In this study, a series of defined synthetic glyco-polymers bearing a representative TACA, i.e., the Thomsen-nouveau (Tn) antigen, have been prepared to probe the induction of early B cell activation and antibody production via a T cell independent mechanism. Valency and density of the antigen in the polymers turned out to be critical. An average of greater than 6 Tn per chain was needed to induce antibody production. Glycopolymers with 40 antigens per chain and backbone molecular weight of 450 kDa gave the strongest stimulation to B cells in vitro, which correlated well with its in vivo activity. Deviations from the desired valency and density led to decreased antibody production or even antigen specific B cell non-responsiveness. These findings provide important insights on how to modulate anti-TACA immune responses facilitating the development of TACA based anti-cancer vaccines using glycopolymers.     The goal of this prospective study was to determine (a) concentrations of the carbohydrate biomarkers Thomsen Friedenreich (TF) antigen and its precursor, Tn antigen, in nipple discharge (ND) collected from women requiring biopsy because of a suspicious breast lesion; and (b) if concentration levels predicted pathologic diagnosis. Adult women requiring biopsy to exclude breast cancer were enrolled and ND obtained. The samples from 124 women were analyzed using an anti-TF and anti-Tn monoclonal antibodies in direct immunoassay. The highest median concentration in ND for TF and Tn was in women with ductal carcinoma in situ (DCIS). TF was higher in women with 1) cancer (DCIS or invasive) vs. either no cancer (atypia or benign pathology, p = .048), or benign pathology (p = .018); and 2) abnormal (atypia or cancer) versus benign pathology (p = .016); and was more predictive of atypia or cancer in post- compared to premenopausal women. Tn was not predictive of disease. High TF concentration and age were independent predictors of disease, correctly classifying either cancer or abnormal vs. benign pathology 83% of the time in postmenopausal women. TF concentrations in ND were higher in women with precancer and cancer compared to women with benign disease, and TF was an independent predictor of breast atypia and cancer. TF may prove useful in early breast cancer detection.

  3. Crystallization and preliminary X-ray study of the common edible mushroom (Agaricus bisporus) lectin.

    The lectin from the common edible mushroom Agaricus bisporus (ABL) belongs to the group of proteins that have the property of binding the Thomsen-Friedenreich antigen (T-antigen) selectively and with high affinity, but does not show any sequence similarity to the other proteins that share this property. The ABL sequence is instead similar to those of members of the saline-soluble fungal lectins, a protein family with pesticidal properties. The presence of different isoforms has been reported. It has been found that in order to be able to grow diffraction-quality crystals of the lectin, it is essential to separate the isoforms, which was performed by preparative isoelectric focusing. Using standard procedures, it was possible to crystallize the most basic of the forms by either vapour diffusion or equilibrium dialysis, but attempts to grow crystals of the other more acidic forms were unsuccessful. The ABL crystals belong to the orthorhombic space group C222(1), with unit-cell parameters a = 93.06, b = 98.16, c = 76.38 A, and diffract to a resolution of 2.2 A on a conventional source at room temperature. It is expected that the solution of this structure will yield further valuable information on the differences in the T-antigen-binding folds and will perhaps help to clarify the details of the ligand binding to the protein.

  4. Shear wave anisotropy from aligned inclusions: ultrasonic frequency dependence of velocity and attenuation

    To understand their influence on elastic wave propagation, anisotropic cracked media have been widely investigated in many theoretical and experimental studies. In this work, we report on laboratory ultrasound measurements carried out to investigate the effect of source frequency on the elastic parameters (wave velocities and the Thomsen parameter γ) and shear wave attenuation) of fractured anisotropic media. Under controlled conditions, we prepared anisotropic model samples containing penny-shaped rubber inclusions in a solid epoxy resin matrix with crack densities ranging from 0 to 6.2 per cent. Two of the three cracked samples have 10 layers and one has 17 layers. The number of uniform rubber inclusions per layer ranges from 0 to 100. S-wave splitting measurements have shown that scattering effects are more prominent in samples where the seismic wavelength to crack aperture ratio ranges from 1.6 to 1.64 than in others where the ratio varied from 2.72 to 2.85. The sample with the largest cracks showed a magnitude of scattering attenuation three times higher compared with another sample that had small inclusions. Our S-wave ultrasound results demonstrate that elastic scattering, scattering and anelastic attenuation, velocity dispersion and crack size interfere directly in shear wave splitting in a source-frequency dependent manner, resulting in an increase of scattering attenuation and a reduction of shear wave anisotropy with increasing frequency.

    Seismic wave propagation through the earth is often stronglyaffected by the presence of fractures. When these fractures are filledwith fluids (oil, gas, water, CO2, etc.), the type and state of the fluid(liquid or gas) can make a large difference in the response of theseismic waves. This paper summarizes recent work on methods ofdeconstructing the effects of fractures, and any fluids within thesefractures, on seismic wave propagation as observed in reflection seismicdata. One method explored here is Thomsen's weak anisotropy approximationfor wave moveout (since fractures often induce elastic anisotropy due tononuniform crack-orientation statistics). Another method makes use ofsome very convenient fracturemore » parameters introduced previously thatpermit a relatively simple deconstruction of the elastic and wavepropagation behavior in terms of a small number of fracture parameters(whenever this is appropriate, as is certainly the case for small crackdensities). Then, the quantitative effects of fluids on thesecrack-influence parameters are shown to be directly related to Skempton scoefficient B of undrained poroelasticity (where B typically ranges from0 to 1). In particular, the rigorous result obtained for the low crackdensity limit is that the crack-influence parameters are multiplied by afactor (1 ? B) for undrained systems. It is also shown how fractureanisotropy affects Rayleigh wave speed, and how measured Rayleigh wavespeeds can be used to infer shear wave speed of the fractured medium.Higher crack density results are also presented by incorporating recentsimulation data on such cracked systems.« less

  6. [My hybrid carrier of clinical pathologist].

    In this review, I showed a brief summary of my carrier in multiple special fields (clinical pathologist, anatomical pathologist of lung, respiratory physician and infection control doctor), my studies and my own view of laboratory medicine. We chiefly study pathology of the lung, especially about type II pneumocytes. Type II pneumocytes had abundant surface coat on the apical surface containing a specific carbohydrate structure of Thomsen-Friedenreich (TF) antigen. TF antigen is a marker of type II pneumocytes beyond animal species, and can be used for evaluating activity of various interstitial pneumonia as type II pneumocyte index (number/lmm alveolar length). Three dimensional views generated from thick sections of ordinary processed paraffin blocks showed new information of normal and abnormal lung morphology. Type II pneumocytes linearly located along the elastic fibers forming framework of polygonal alveoli, and in usual interstitial pneumonia, destruction of these elastic fibers were observed. In Japan, roles of a clinical pathologist are not definite as a radiologist, and clinical laboratory in a hospital is recognized as a section only performing blood and chemical tests. Evaluation of the data and participation in diagnosis were not requested. In future, medical doctors devote themselves to treat patients, and clinical pathologists and laboratory technicians have to help the doctors in diagnostic process. Routine tests (blood and urine) are most frequently performed in clinical medicine, but the data are not adequately used. Therefore, a system is necessary for interpreting routine tests and reporting them to other medical staffs.

  7. Mass loss at Saturn: The contribution of plasmoids

    Jia and Kivelson (2012) showed that an MHD simulation of Saturn's magnetosphere including rotating field-aligned currents generates a plasmoid on each rotation period. For southern summer conditions, plasmoids form through reconnection in the tail near midnight at a rotation phase for which the core magnetic field perturbation points towards the planet in the LT sector of the release. After release, they move northward and outward in the post midnight sector. Mass loss is found to be dominated by plasmoid release, and to compare closely to the mass source rate; much of the outward mass flux is found in the 0300-0900 LT sector. However, studies of plasmoid signatures in Cassini magnetometer data conclude that releases do not occur each rotation period and infer mass loss rates far smaller than Saturn's mass source rate (Jackman et al., 2014). Studies based on plasma data also infer loss rates smaller than the source rate from Enceladus (Thomsen, 2013). The simulation suggests that plasmoid occurrence frequency depends strongly on local time and latitude. Indeed, in regions near the equator and near midnight, where plasmoids are present in the simulation, the occurrence frequency of plasmoid releases inferred from magnetometer data is higher than elsewhere (Jackman et al., 2016; Smith et al., 2016), with occurrence probability peaking at the phase predicted by the simulation. We consider how the observational limitations should be modified to establish mass loss rates and plasmoid occurrence frequency.

  8. Carcinoma autoantigens T and Tn and their cleavage products interact with Gal/GalNAc-specific receptors on rat Kupffer cells and hepatocytes.

    We studied interactions of isolated Thomsen-Friedenreich (T)- and Tn-specific glycoproteins with the Gal/GalNAc-specific receptors on rat Kupffer cells and compared them to those with rat hepatocytes. Immunoreactive T and Tn are specific pancarcinoma epitopes. Electron microscopy of gold-labelled T and Tn antigens revealed their specific binding to Kupffer cells, followed by their uptake via the coated pit/vesicle pathway of receptor-mediated endocytosis. Preincubation of Kupffer cells with GalNAc and GalNAc-BSA, but not GlcNAc or GlcNAc-BSA specifically inhibited binding of the T and Tn glycoproteins. Desialylated, isologous erythrocytes (T RBC) are known to bind to the Gal/GalNAc receptors of rat Kupffer cells and hepatocytes. This attachment was specifically inhibited by T and Tn in a concentration-dependent manner: 50% T RBC-Kupffer cell contacts were inhibited at 8.5.10(-6) mM T and 8.5.10(-5) mM Tn antigen concentrations, respectively. The corresponding figures for hepatocytes were 6.10(-6) mM T and 1.2.10(-6) mM Tn antigen. Amino-terminal cleavage products of the T glycoprotein, possessing clusters terminating in non-reducing Gal/GalNAc, inhibited T RBC binding to Kupffer cells and hepatocytes usually at 10(-2) to 10(-5) mM concentrations, whereas GalNAc, galactose and galactose glycosides inhibited at millimolar concentrations. Galactose-unrelated carbohydrates were inactive at concentrations greater than or equal to 50 mM.

    Red blood cell (RBC) Thomsen-Friedenreich antigen exposure (T activation) in infants with necrotizing enterocolitis (NEC) has occasionally been associated with posttransfusional intravascular hemolysis thought to be due to anti-T antibodies in the donor plasma. We describe an infant with NEC and Clostridium perfringens infection complicated by severe hemolysis after plasma transfusion. After this case, infants with confirmed NEC were prospectively evaluated for T activation. We checked for hemolysis in patients with T activation receiving plasma-containing blood products. The infant had received 80 mL of fresh-frozen plasma (FFP). His RBCs displayed strong T activation, and agglutination was observed with four of six ABO-compatible FFP units. A direct antiglobulin test was negative. IgM-class anti-T antibodies were present in small amounts (titer of 8) in the transfused FFP. Anti-T antibodies from the blood donor were not hemolytic in vitro. In the prospective study, T activation was observed in three of 28 infants with NEC (11%). One infant presented moderate T activation and two infants presented very strong T activation but only moderate decreases in sialic acid expression on the RBC membrane. These three infants presented no signs of hemolysis after transfusion with unwashed blood products or FFP. Anti-T antibodies are unlikely to be the etiologic factor for the hemolytic reactions observed in infants with NEC and T activation. Massive RBC desialylation and the direct action of bacterial toxins are more probable causes. Strict avoidance of plasma-containing blood products does not seem justified in these infants. © 2017 AABB.

  10. Evolution of petrophysical properties of across natural faults: a study on cores from the Tournemire underground research laboratory (France)

    Faults in general, and in clay materials in particular, have complex structures that can be linked to both a polyphased tectonic history and the anisotropic nature of the material. Drilling through faults in shaly materials allows one to measure properties such as the structure, the mineralogical composition, the stress orientation or physical properties. These relations can be investigated in the laboratory in order to have a better understanding on in-situ mechanisms. In this study we used shales of Toarcian age from the Tournemire underground research laboratory (France). We decided to couple different petrophysical measurements on core samples retrieved from a borehole drilled perpendicularly to a fault plane, and the fault size is of the order of tens of meters. This 25m long borehole was sampled in order to perform several types of measurements: density, porosity, saturation directly in the field, and velocity of elastic waves and magnetic susceptibility anisotropy in the laboratory. For all these measurements, special protocols were developed in order to preserve as much as possible the saturation state of the samples. All these measurements were carried out in three zones that intersects the borehole: the intact zone , the damaged zone and the fault core zone. From our measurements, we were able to associate specific properties to each zone of the fault. We then calculated Thomsen's parameters in order to quantify the elastic anisotropy across the fault. Our results show strong variations of the elastic anisotropy with the distance to the fault core as well as the occurrence of anisotropy reversal.

    The combining site of the nontoxic carbohydrate binding protein (Abrus precatorius agglutinin, APA) purified from the needs of Abrus precatorius (Jequirity bean), was studied by quantitative precipitin and precipitin-inhibition assays. Of 26 glycoproteins and polysaccharides tested, all, except sialic acid-containing glycoproteins and desialized ovine salivary glycoproteins, reacted strongly with the lectin, and precipitated over 70% of the lectin added, indicating that APA has a broad range of affinity and recognizes (internal) Gal beta 1----sequences of carbohydrate chains. The strong reaction with desialized porcine and rat salivary glycoproteins as well as pneumococcus type XIV polysaccharide suggests that APA has affinity for one or more of the following carbohydrate sequences: Thomsen-Friedenreich (T, Gal beta 1----3GalNAc), blood group precursor type I and/or type II (Gal beta 1----3/4GlcNAc) disaccharide determinants of complex carbohydrates. Among the oligosaccharides tested, the T structure was the best inhibitor; it was 2.4 and 3.2 times more active than type II and type I sequences, respectively. The blood group I Ma-active trisaccharide, Gal beta 1----4GlcNAc beta 1----6Gal, was about as active as the corresponding disaccharide (II). From the above results, we conclude that the size of the combining site of the A. precatorius agglutinin is probably as large as a disaccharide and most strongly complementary to the Gal beta 1----3GalNAc (T determinant) sequence. The carbohydrate specificities of this lectin will be further investigated once the related oligosaccharide structures become available.

  12. Design and synthesis of multifunctional gold nanoparticles bearing tumor-associated glycopeptide antigens as potential cancer vaccines.

    The development of vaccines against specific types of cancers will offer new modalities for therapeutic intervention. Here, we describe the synthesis of a novel vaccine construction prepared from spherical gold nanoparticles of 3-5 nm core diameters. The particles were coated with both the tumor-associated glycopeptides antigens containing the cell-surface mucin MUC4 with Thomsen Friedenreich (TF) antigen attached at different sites and a 28-residue peptide from the complement derived protein C3d to act as a B-cell activating "molecular adjuvant". The synthesis entailed solid-phase glycopeptide synthesis, design of appropriate linkers, and attachment chemistry of the various molecules to the particles. Attachment to the gold surface was mediated by a novel thiol-containing 33 atom linker which was further modified to be included as a third "spacer" component in the synthesis of several three-component vaccine platforms. Groups of mice were vaccinated either with one of the nanoplatform constructs or with control particles without antigen coating. Evaluation of sera from the immunized animals in enzyme immunoassays (EIA) against each glycopeptide antigen showed a small but statistically significant immune response with production of both IgM and IgG isotypes. Vaccines with one carbohydrate antigen (B, C, and E) gave more robust responses than the one with two contiguous disaccharides (D), and vaccine E with a TF antigen attached to threonine at the 10th position of the peptide was selected for IgG over IgM suggesting isotype switching. The data suggested that this platform may be a viable delivery system for tumor-associated glycopeptide antigens.

  13. Lewis x is highly expressed in normal tissues: a comparative immunohistochemical study and literature revision.

    An immunohistochemical analysis was employed to determine the expression of carbohydrate antigens associated to mucins in normal epithelia. Tissue samples were obtained as biopsies from normal breast (18), colon (35) and oral cavity mucosa (8). The following carbohydrate epitopes were studied: sialyl-Lewis x, Lewis x, Lewis y, Tn hapten, sialyl-Tn and Thomsen-Friedenreich antigen. Mucins were also studied employing antibodies against MUC1, MUC2, MUC4, MUC5AC, MUC6 and also normal colonic glycolipid. Statistical analysis was performed and Kendall correlations were obtained. Lewis x showed an apical pattern mainly at plasma membrane, although cytoplasmic staining was also found in most samples. TF, Tn and sTn haptens were detected in few specimens, while sLewis x was found in oral mucosa and breast tissue. Also, normal breast expressed MUC1 at a high percentage, whereas MUC4 was observed in a small number of samples. Colon specimens mainly expressed MUC2 and MUC1, while most oral mucosa samples expressed MUC4 and MUC1. A positive correlation between MUC1VNTR and TF epitope (r=0.396) was found in breast samples, while in colon specimens MUC2 and colonic glycolipid versus Lewis x were statistically significantly correlated (r=0.28 and r=0.29, respectively). As a conclusion, a defined carbohydrate epitope expression is not exclusive of normal tissue or a determined localization, and it is possible to assume that different glycoproteins and glycolipids may be carriers of carbohydrate antigens depending on the tissue localization considered.

  14. Saturn's periodicities: New results from an MHD simulation of magnetospheric response to rotating ionospheric vortices

    In previous work we demonstrated that a magnetohydrodynamic (MHD) simulation of Saturn's magnetosphere in which periodicity is imposed by rotating vortical flows in the ionosphere reproduces many reported periodically varying properties of the system. Here we shall show that previously unreported features of the MHD simulation of Saturn's magnetosphere illuminate additional measured properties of the system. By averaging over a rotation period, we identify a global electric field whose magnitude is a few tenths of a mV/m (see Figure 1). The electric field intensity decreases with radial distance in the middle magnetosphere, consistent with drift speeds v=E/B of a few km/s towards the morning side and relatively independent of radial distance. The electric field within 10 RS in the equatorial plane is oriented from post-noon to post-midnight, in excellent agreement with observations [e.g., Thomsen et al., 2012; Andriopoulou et al., 2012, 2013; Wilson et al., 2013]. By following the electric field over a full rotation phase we identify oscillatory behavior whose magnitude is consistent with the reported fluctuations of measured electric fields. Of particular interest is the nature of the fast mode perturbations that produce periodic displacement of the magnetopause and flapping of the current sheet. Figure (2) shows the total perturbation pressure (the sum of magnetic and thermal pressure) in the equatorial plane at a rotation phase for which the ionospheric flow near noon is equatorward. By following the perturbations over a full rotation period, we demonstrate properties of the fast mode wave launched by the rotating flow structures and thereby characterize the 'cam' signal originally proposed by Espinosa et al. [2003].

  15. A novel mutation of the CLCN1 gene associated with myotonia hereditaria in an Australian cattle dog.

    Heritable myotonia is a genetic muscle disorder characterized by slow relaxation of skeletal muscles. The main clinical signs are skeletal muscle stiffness, especially after vigorous contraction, and muscle hypertrophy. Muscle stiffness may be enhanced by inactivity, and often is relieved by exercise. Myotonia can be inherited in an autosomal dominant or recessive manner (Thomsen- or Becker-type myotonia, respectively). In mice, goats, Miniature Schnauzer dogs, and most affected humans, the disorder is caused by mutations in CLCN1, which encodes the skeletal muscle voltage-gated chloride channel, Cl1C-1. We hypothesized that an Australian Cattle Dog with generalized muscle stiffness and hypertrophy examined at the Ontario Veterinary College would have a mutation in the CLCN1 gene. A pure-bred Australian Cattle Dog from Ontario, Canada, was used. Based on clinical signs and electromyographic test results, a diagnosis of myotonia hereditaria was made, and a muscle biopsy was collected for genetic analysis. Sequence data obtained from the affected dog confirmed that it was homozygous for a single base insertion in the CLCN1 coding sequence. This mutation would result in a truncated ClC-1 protein being expressed, which, based on molecular evidence from other studies, would result in functionally compromised chloride conduction in the skeletal muscles of the animal. To the authors' knowledge, this report describes the Ist case of myotonia in an Australian Cattle Dog and represents the 1st non-Schnauzer canine myotonia to be genetically characterized. In addition, we developed a polymerase chain reaction-based genetic screen to detect heterozygotes with this mutation in the at-large Australian Cattle Dog population.

  16. Nonhyperbolic reflection moveout for horizontal transverse isotropy

    The transversely isotropic model with a horizontal axis of symmetry (HTI) has been used extensively in studies of shear-wave splitting to describe fractured formations with a single system of parallel vertical penny-shaped cracks. Here, the authors present an analytic description of long-spread reflection moveout in horizontally layered HTI media with arbitrary strength of anisotropy. To account for nonhyperbolic moveout, the authors have derived an exact expression for the azimuthally dependent quartic term of the Taylor series traveltime expansion valid for any pure mode in an HTI layer. The quartic moveout coefficient and the NMO velocity are then substituted into themore » nonhyperbolic moveout equation of Tsvankin and Thomsen, originally designed for vertical transverse isotropy (VTI). In multilayered HTI media, the NMO velocity and the quartic moveout coefficient reflect the influence of layering as well as azimuthal anisotropy. The authors show that the conventional Dix equation for NMO velocity remains entirely valid for any azimuth in HTI media if the group-velocity vectors (rays) for data in a common-midpoint (CMP) gather do not deviate from the vertical incidence plane. Although this condition is not exactly satisfied in the presence of azimuthal velocity variations, rms averaging of the interval NMO velocities represents a good approximation for models with moderate azimuthal anisotropy. Furthermore, the quartic moveout coefficient for multilayered HTI media can also be calculated with acceptable accuracy using the known averaging equations for vertical transverse isotropy. This allows one to extend the nonhyperbolic moveout equation to horizontally stratified media composed of any combination of isotropic, VTI, and HTI layers.« less

  17. Electron temperature and de Hoffmann-Teller potential change across the Earth's bow shock: New results from ISEE 1

    We present a survey of the trends between the electron temperature increase ΔTe and the de Hoffmann-Teller frame (HTF) electrostatic potential jump ΔΦHT and their correlation with other parameters that characterize the shock transition using a new ISEE 1 database of 129 Earth bow shock crossings. A fundamental understanding of the HTF potential is central to distinguishing the reversible and irreversible changes to electron temperature across collisionless shocks. The HTF potential is estimated using three different techniques: (1) integrating the steady state, electron fluid momentum equation across the shock layer using high time resolution plasma and field data from ISEE 1, (2) using the steady state, electron fluid energy equation, and (3) using an electron polytrope approximation. We find that ΔΦHT and ΔTe are strongly and positively correlated with |Δ(mpUn2/2)|, which is in good qualitative agreement with earlier experimental surveys [Thomsen et al., 1987b; Schwartz et al., 1988] that used bow shock model normals and used the flow in the spacecraft frame. There is a strong linear organization of the ΔTe with ΔΦHT, which suggests an average effective electron polytropic index of <γe>~2. In addition, ΔTe and ΔΦHT are organized by βe, although our results may be biased by our limited sampling of shock conditions. Comparisons indicate that the differentials in the HTF potential δΦHT are proportional to the differentials in the magnetic field intensity δB across the shock, with a proportionality constant κ that is a fixed constant for a given shock crossing.

  18. A Study for Anisotropic Wavefield Analysis with Elastic Layered Models

    Subsurface materials are generally anisotropic due to complicated geological conditions, for example, sedimentary materials, fractures reflecting various stress conditions in the past and present in the subsurface. There are many studies on seismic wave propagation in TI (transversely isotropic) and orthorhombic media (e.g., Thomsen, 1986; Alkhalifah, 2000; Bansal and Sen, 2008). In most of those studies, the magnitude of anisotropy is assumed to be weak. Therefore, it may be not appropriate to apply their theories directly to strongly anisotropic subsurface media in seismic exploration. It is necessary to understand the effects of the anisotropy on the behavior of seismic wave propagation in strongly anisotropic media in the seismic exploration. In this study, we investigate the influence of strong anisotropy on received seismic waveforms using three-dimensional numerical models, and verified capability of detecting subsurface anisotropy. Our numerical models contain an isotropic and an anisotropic (VTI, transversely isotropic media with vertical symmetry axis) layer, respectively, in the isotropic background subsurface. Since the difference between the two models is only the anisotropy in the vertical propagation velocity, we could look at the influence of anisotropy in the residual wavefield that is the difference in the observed wavefields of two models. We analyzed the orbital motions of the residual wavefield to see what kind of wave motions the waveforms show. We found that the residual waveforms generated by the anisotropic layer include the orbital motions of shear waves right after the first arrival, i.e., mode conversion from the compressional waves due to the anisotropy. The residual waveforms could be exploited to estimate both the order of anisotropy and the thickness of anisotropic layer in subsurface.

  19. Gravity-dependent signal path variation in a large VLBI telescope modelled with a combination of surveying methods

    The very long baseline interferometry (VLBI) antenna in Medicina (Italy) is a 32-m AZ-EL mount that was surveyed several times, adopting an indirect method, for the purpose of estimating the eccentricity vector between the co-located VLBI and Global Positioning System instruments. In order to fulfill this task, targets were located in different parts of the telescope’s structure. Triangulation and trilateration on the targets highlight a consistent amount of deformation that biases the estimate of the instrument’s reference point up to 1 cm, depending on the targets’ locations. Therefore, whenever the estimation of accurate local ties is needed, it is critical to take into consideration the action of gravity on the structure. Furthermore, deformations induced by gravity on VLBI telescopes may modify the length of the path travelled by the incoming radio signal to a non-negligible extent. As a consequence, differently from what it is usually assumed, the relative distance of the feed horn’s phase centre with respect to the elevation axis may vary, depending on the telescope’s pointing elevation. The Medicina telescope’s signal path variation Δ L increases by a magnitude of approximately 2 cm, as the pointing elevation changes from horizon to zenith; it is described by an elevation-dependent second-order polynomial function computed as, according to Clark and Thomsen (Techical report, 100696, NASA, Greenbelt, 1988), a linear combination of three terms: receiver displacement Δ R, primary reflector’s vertex displacement Δ V and focal length variations Δ F. Δ L was investigated with a combination of terrestrial triangulation and trilateration, laser scanning and a finite element model of the antenna. The antenna gain (or auto-focus curve) Δ G is routinely determined through astronomical observations. A surprisingly accurate reproduction of Δ G can be obtained with a combination of Δ V, Δ F and Δ R.

    The oncofoetal Thomsen-Friedenreich (TF1, CD176) epitope is a carbohydrate cancer stem cell (CSC) antigen, and TF1-mediated cancer progression can be widely reversed by anti-TF1 antibodies. Particularly, CSC-like cells are regarded to be tumorigenic and chemoresistant. Aberrant p53 is probably the factor most closely associated with chemoresistance and tumour aggressiveness in ovarian tumours. We thus questioned whether TF1 in combination with p53 or as a single marker may be related to clinico-pathological features and survival of ovarian cancer patients. Both markers were quantified in ovarian cancer tissue (n = 151) by immunohistochemistry. p53 staining was subdivided into three subgroups [n (completely negative) = 57, n (moderately stained) = 28, n (overexpressing) = 66]. TF1 was scored as positive (n = 30) versus negative (n = 121). Only in those cancers classified with moderate p53 staining-and thus most likely displaying with wild-type TP53-TF1 positivity turned out to be a predictor for shortened overall survival (univariate: p < 0.001, multivariate: p = 0.001). By screening 17 different protein markers for correlation with TF1, only mucin-1 emerged as a potential TF1 carrier protein. It is hypothesized that TF1 may confer tumour-promoting features, especially in a TP53 wild-type genetic background. In addition, TF1 is an attractive immunotherapeutic target. Whether those cases classified as TF1 positive and at the same time as moderately stained for p53 might particularly benefit from a future anti-TF1 antibody treatment or from TF1 vaccination therapy remains to be determined.

  1. Inversion of azimuthally dependent NMO velocity in transversely isotropic media with a tilted axis of symmetry

    Just as the transversely isotropic model with a vertical symmetry axis (VTI media) is typical for describing horizontally layered sediments, transverse isotropy with a tilted symmetry axis (TTI) describes dipping TI layers (such as tilted shale beds near salt domes) or crack systems. P-wave kinematic signatures in TTI media are controlled by the velocity V{sub PO} in the symmetry direction, Thomsen's anisotropic coefficients {xi} and {delta}, and the orientation (tilt {nu} and azimuth {beta}) of the symmetry axis. Here, the authors show that all five parameters can be obtained from azimuthally varying P-wave NMO velocities measured for two reflectors withmore » different dips and/or azimuths (one of the reflectors can be horizontal). The shear-wave velocity V{sub SO} in the symmetry direction, which has negligible influence on P-wave kinematic signatures, can be found only from the moveout of shear waves. Using the exact NMO equation, the authors examine the propagation of errors in observed moveout velocities into estimated values of the anisotropic parameters and establish the necessary conditions for a stable inversion procedure. Since the azimuthal variation of the NMO velocity is elliptical, each reflection event provides them with up to three constraints on the model parameters. Generally, the five parameters responsible for P-wave velocity can be obtained from two P-wave ellipses, but the feasibility of the moveout inversion strongly depends on the tilt {nu}. While most of the analysis is carried out for a single layer, the authors also extend the inversion algorithm to vertically heterogeneous TTI media above a dipping reflector using the generalized Dix equation. A synthetic example for a strongly anisotropic, stratified TTI medium demonstrates a high accuracy of the inversion.« less

  2. Computing the Sensitivity Kernels for 2.5-D Seismic Waveform Inversion in Heterogeneous, Anisotropic Media

    2.5-D modeling and inversion techniques are much closer to reality than the simple and traditional 2-D seismic wave modeling and inversion. The sensitivity kernels required in full waveform seismic tomographic inversion are the Fréchet derivatives of the displacement vector with respect to the independent anisotropic model parameters of the subsurface. They give the sensitivity of the seismograms to changes in the model parameters. This paper applies two methods, called `the perturbation method' and `the matrix method', to derive the sensitivity kernels for 2.5-D seismic waveform inversion. We show that the two methods yield the same explicit expressions for the Fréchet derivatives using a constant-block model parameterization, and are available for both the line-source (2-D) and the point-source (2.5-D) cases. The method involves two Green's function vectors and their gradients, as well as the derivatives of the elastic modulus tensor with respect to the independent model parameters. The two Green's function vectors are the responses of the displacement vector to the two directed unit vectors located at the source and geophone positions, respectively; they can be generally obtained by numerical methods. The gradients of the Green's function vectors may be approximated in the same manner as the differential computations in the forward modeling. The derivatives of the elastic modulus tensor with respect to the independent model parameters can be obtained analytically, dependent on the class of medium anisotropy. Explicit expressions are given for two special cases—isotropic and tilted transversely isotropic (TTI) media. Numerical examples are given for the latter case, which involves five independent elastic moduli (or Thomsen parameters) plus one angle defining the symmetry axis.

    Comment is presented on the results of measurements, reported by Gosling et al. (1991), that were made on ISEE in the vicinity of the high-latitude dusk magnetopause near the terminator plane, at a time when the local magnetosheath and tail lobe magnetic fields were nearly oppositely directed. The character of the observed plasma flowing both tailward and sunward within the high-latitude magnetopause current layer presented real evidence for the local reconnection process. Gosling et al. argued that this process may be a manifestation of different global magnetospheric topology structures. In the comment, a global magnetospheric convection pattern is constructed for the northward IMF and for the case of a large azimuthal component of the IMF with small Bz, irrespective of its sign. The suggested scheme provides a simple explanation for the observed sunward convection in the polar caps both for the northward and for strong By with small Bz. According to the present model, for the magnetosheath field at 2300 UT on June 11, 1978, the reconnection between the open field lines appears at the northern neutral point.

    Herein we present the results of two related investigations. The first study determined if concentrations in breast nipple discharge (ND) of two proteins (urinary plasminogen activator, uPA and its inhibitor, PAI-1) predicted the presence of breast atypia and cancer in pre- and/or postmenopausal women requiring surgery because of a suspicious breast lesion. The second study assessed if these proteins increased the predictive ability of a carbohydrate (Thomsen Friedenreich, TF) which we previously demonstrated predicted the presence of disease in postmenopausal women requiring surgery. In the first study we prospectively enrolled 79 participants from whom we collected ND, measured uPA and PAI-1 and correlated expression with pathologic findings. In the second study we analyzed 35 (uPA and PAI-1 in 24, uPA in an additional 11) ND samples collected from different participants requiring breast surgery, all of whom also had TF results. uPA expression was higher in pre- and PAI-1 in postmenopausal women with 1) cancer (DCIS or invasive) vs. either no cancer (atypia or benign pathology, p = .018 and .025, respectively), or benign pathology (p = .017 and .033, respectively); and 2) abnormal (atypia or cancer) versus benign pathology (p = .018 and .052, respectively). High uPA and PAI-1 concentrations and age were independent predictors of disease in premenopausal women, with an area under the curve (AUC) of 83-87% when comparing diseased vs. benign pathology. uPA, TF, and age correctly classified 35 pre- and postmenopausal women as having disease or not 84-91% of the time, whereas combining uPA+PAI-1+TF correctly classified 24 women 97-100% of the time. uPA and PAI-1 concentrations in ND were higher in women with atypia and cancer compared to women with benign disease. Combining uPA, PAI-1 and TF in the assessment of women requiring diagnostic breast surgery maximized disease prediction. The assessment of these markers may prove useful in early breast cancer detection.

  8. SAMI3 Simulations of the Persistent May 1994 Plasmasphere Plume

    We use the Naval Research Laboratory SAMI3 ionosphere/plasmasphere model[1] to explore the physics of a long-lived plasmasphere plume. A plasmasphere plume is a storm feature that extends the cold plasma that is normally trapped by the geomagnetic field (the plasmasphere) outward towards the bow shock. In the case of the May 1994 storm, the storm and the plume continued for 12 days. For the model storm, we imposed a Kp-driven Volland/Stern-Maynard/Chen potential [2-4]. Results are compared to measurements of the cold ion density from the 1989-046 spacecraft in geosynchronous orbit [5]. We find that many details of the observed plume are reproduced by SAMI3, but only if a background magnetosphere density is included as a boundary condition.   9. The derivation of an anisotropic velocity model from a combined surface and borehole seismic survey in crystalline environment at the COSC-1 borehole, central Sweden

    The Scandinavian Caledonides provide a well-preserved example of a Palaeozoic continent-continent collision, where surface geology in combination with geophysical data provides information about the geometry of parts of the Caledonian structure. The project COSC (Collisional Orogeny in the Scandinavian Caledonides) investigates the structure and physical conditions of the orogen units and the underlying basement with two approximately 2.5 km deep cored boreholes in western Jämtland, central Sweden. In 2014, the COSC-1 borehole was successfully drilled through a thick section of the Seve Nappe Complex. This tectonostratigraphic unit, mainly consisting of gneisses, belongs to the so-called Middle Allochthons and has been ductilely deformed and transported during the collisional orogeny. After the drilling, a major seismic survey was conducted in and around the COSC-1 borehole with the aim to recover findings on the structure around the borehole from core analysis and downhole logging. The survey comprised both seismic reflection and transmission experiments, and included zero-offset and multiazimuthal walkaway Vertical Seismic Profile (VSP) measurements, three long offset surface lines centred on the borehole, and a limited 3-D seismic survey. In this study, the data from the multiazimuthal walkaway VSP and the surface lines were used to derive detailed velocity models around the COSC-1 borehole by inverting the first-arrival traveltimes. The comparison of velocities from these tomography results with a velocity function calculated directly from the zero-offset VSP revealed clear differences in velocities for horizontally and vertically travelling waves. Therefore, an anisotropic VTI (transversely isotropic with vertical axis of symmetry) model was found that explains first-arrival traveltimes from both the surface and borehole seismic data. The model is described by a vertical P-wave velocity function derived from zero-offset VSP and the Thomsen parameters ε = 0

  11. Time-lapse ultrasonic imaging of elastic anisotropy in saturated sandstone under polyaxial stress state

    Although true-triaxial test (TTT) of rocks is now more extensive worldwide, stress-induced heterogeneity is not accounted for and usually simplified anisotropic models are used. Data from a TTT on a cubic sample of Fontainebleau sandstone is used in this study to evaluate our velocity imaging methodology. An anisotropic P wave velocity tomography method was developed using a geometrical approach based on an ellipsoidal P wavefront surface. During the two non-damaging phases of the experiment, saturation of the rock sample with water resulted in inaccurate tomographic images; however, during the final elasto-plastic phase of the experiment comprising major AE activities, tomographic images demonstrated reasonable anomalies. Thus, the P-S1-S2 velocity survey was utilized to obtain an accurate and reliable velocity image of the sample during the two non-damaging phases. This was accomplished using a numerical investigation by FLAC3D on the non-uniform distribution of stress over the sample to estimate the compaction pseudo-boundary surfaces within the rock. Thus, the problem of breakdown in the expected symmetry of shear wave velocities was resolved. It was discovered that a homogeneous anisotropic core in the center of the sample is formed under the standard polyaxial setup where elastic parameters could be computed. Off-diagonal elastic tensor parameters were obtained by a combination of various velocity survey data and justified the ellipsoidal model as being the most appropriate and facilitated the calculation of Thomsen parameters. The ellipsoidal heterogeneous velocity model was also verified by AE event location of transducer shots through the cubic rock specimen especially at the final phase of the experiment consisting lower-velocity zones bearing partially saturated fractures. AE of the rock during the whole experiment recorded by the surrounding transducers were investigated by location methods developed for anisotropic heterogeneous medium. AE events

  12. In situ seismic anisotropy around deep earthquakes in Japan subduction slabs using Japan Meteorological Agency moment tensors

    Knowing the in situ seismic anisotropy around deep earthquakes in slabs is important in understanding deep-earthquake mechanism as it may provide critically needed information about the rock fabric where deep earthquakes occur. It has been recognized for about 50 years that many deep earthquakes are not double-couple (DC) events. Previously we showed that in situ anisotropy around deep earthquakes could explain such observed non-DC events. Traditionally, the shear wave splitting method has been used to infer such anisotropy around deep earthquakes but this is challenging because it will need many crossing ray paths for the method to localize the anisotropic region (Long 2013). In this abstract, we adopt the same procedure to obtain anisotropy in the Pacific slab under Japan using moment tensors provided by the Japan Meteorological Agency using the F-net data. We directly probe the in situ anisotropy within the subducting slabs using the radiation patterns (represented by the moment tensors) of deep earthquakes (with depth greater than 60 km). By assuming a group of shear dislocation events embedded in a common tilted transversely isotropic (TTI) medium, we used the moment tensors as our input data to invert for the anisotropy in Mariana-Japan-Kuril subducting zone. The TTI medium is characterized by the P and S wave velocities along the symmetry axis (described by two free angles) and three Thomsen parameters. We divided the deep earthquake events into 9 groups by their spatial proximity using the k-means clustering method (Hartigan and Wong 1979). These 9 groups include 2 intermediate-depth groups (depth from 60 km to 300 km) and 7 deep-focus groups (depth greater than 300 km). Our inversion results show that the inverted TTI symmetry axes are perpendicular to the slab interface for two intermediate-depth groups (consistent with dehydration metamorphic reactions) and parallel to the slab interface for 7 deep-focus group.     Singh et al. (2016) published a geochemical record of sediment compositions from the flood plain of the Ghaggar River in western India and use the changing provenance, particularly as traced by Nd isotope composition, to reconstruct how erosion patterns have changed over the past 100 k.y. In doing so they propose a link between climate change and erosion, and they argue for more erosion from the Higher Himalaya during warmer interglacial periods and more from the Lesser Himalaya during glacial intervals. While we support the concept of erosion patterns being climatically modulated we here take the opportunity to compare the data presented by Singh et al.   14. Analysis of the Type IV Fimbrial-Subunit Gene fimA of Xanthomonas hyacinthi: Application in PCR-Mediated Detection of Yellow Disease in Hyacinths

    A sensitive and specific detection method was developed for Xanthomonas hyacinthi; this method was based on amplification of a subsequence of the type IV fimbrial-subunit gene fimA from strain S148. The fimA gene was amplified by PCR with degenerate DNA primers designed by using the N-terminal and C-terminal amino acid sequences of trypsin fragments of FimA. The nucleotide sequence of fimA was determined and compared with the nucleotide sequences coding for the fimbrial subunits in other type IV fimbria-producing bacteria, such as Xanthomonas campestris pv. vesicatoria, Neisseria gonorrhoeae, and Moraxella bovis. In a PCR internal primers JAAN and JARA, designed by using the nucleotide sequences of the variable central and C-terminal region of fimA, amplified a 226-bp DNA fragment in all X. hyacinthi isolates. This PCR was shown to be pathovar specific, as assessed by testing 71 Xanthomonas pathovars and bacterial isolates belonging to other genera, such as Erwinia and Pseudomonas. Southern hybridization experiments performed with the labelled 226-bp DNA amplicon as a probe suggested that there is only one structural type IV fimbrial-gene cluster in X. hyacinthi. Only two Xanthomonas translucens pathovars cross-reacted weakly in PCR. Primers amplifying a subsequence of the fimA gene of X. campestris pv. vesicatoria (T. Ojanen-Reuhs, N. Kalkkinen, B. Westerlund-Wikström, J. van Doorn, K. Haahtela, E.-L. Nurmiaho-Lassila, K. Wengelink, U. Bonas, and T. K. Korhonen, J. Bacteriol. 179: 1280–1290, 1997) were shown to be pathovar specific, indicating that the fimbrial-subunit sequences are more generally applicable in xanthomonads for detection purposes. Under laboratory conditions, approximately 1,000 CFU of X. hyacinthi per ml could be detected. In inoculated leaves of hyacinths the threshold was 5,000 CFU/ml. The results indicated that infected hyacinths with early symptoms could be successfully screened for X. hyacinthi with PCR. PMID:11157222

  15. Soil Biota and Litter Decay in High Arctic Ecosystems

    Frost heave action contributes to the formation of non-sorted circles in the High Arctic. Non-sorted circles tend to heave more than the surrounding tundra due to deeper thaw and the formation of ice lenses. Thus, the geomorphology, soils and vegetation on the centers of the patterned-ground feature (non-sorted circles) as compared to the surrounding soils (inter-circles) can be different. We established a decomposition experiment to look at in situ decay rates of the most dominant graminoid species on non-sorted circles and adjacent inter-circle soils along a climatic gradient in the Canadian High Arctic as a component of a larger study looking at the biocomplexity of small-featured patterned ground ecosystems. Additionally, we investigated variation in soil chemical properties and biota, including soil microarthropods and microbial composition and biomass, as they relate to climate, topographic position, and litter decay rates. Our three sites locations, from coldest to warmest, are Isachsen, Ellef Ringnes Island (ER), NU (bioclimatic subzone A); Mould Bay (MB), Prince Patrick Island, NT (bioclimatic subzone B), and Green Cabin (GC), Aulavik National Park, Thomsen River, Banks Island, NT (bioclimatic subzone C). Our sample design included the selection of 15 non-sorted circles and adjacent inter-circle areas within the zonal vegetation at each site (a total of 90 sites), and a second set of 3 non-sorted circles and adjacent inter-circle areas in dry, mesic and wet tundra at each of the sites. Soil invertebrates were sampled at each site using both pitfall traps, soil microbial biomass was determined using substrate induced respiration and bacterial populations were determined using the most probable number method. Decomposition rates were measured using litterbags and as the percent of mass remaining of Carex misandra, Luzula nivalis and Alopecuris alpinus in GC, MB and ER, respectively.     In unconventional tough gas reservoirs (e.g. tight sandstones or shales) the presence of fractures, either naturally formed or hydraulically induced, is almost always a prerequisite for hydrocarbon productivity to be economically viable. One of the formations classified so far as a potential interesting formation for shale gas exploration in the Netherlands is the Lower Jurassic Posidonia Shale Formation (PSF). However data of the Posidonia Shale Formation is scarce so far and samples are hard to come by, especially on the variability and heterogeneity of the petrophysical parameters of this shale little is known. Therefore research and sample collection is conducted on a time and depositional analogue of the PSF: the Whitby Mudstone Formation (WMF) in the United Kingdom. A large number of samples along a ~7m stratigraphic section of the Whitby Mudstone Formation have been collected and analysed. Standard petrophysical properties such as porosity and matrix densities are quantified for a number of samples throughout the section, as well as mineral composition analysis based on XRD/XRF and SEM analyses. Seismic velocity measurements are also conducted at multiple heights in the section and in multiple directions to elaborate on anisotropy of the material. Attenuation anisotropy is incorporated as well as Thomsen's parameters combined with elastic parameters, e.g. Young's modulus and Poisson's ratio, to quantify the elastic anisotropy. Furthermore rock mechanical experiments are conducted to determine the elastic constants, rock strength, fracture characteristics, brittleness index, fraccability and rock mechanical anisotropy across the stratigraphic section of the Whitby mudstone formation. Results show that the WMF is highly anisotropic and it exhibits an anisotropy on the large limit of anisotropy reported for US gas shales.     HTI media with constant anisotropy orientation as recovered in the first step. It leads to an effective horizontal orthorhombic elastic model. From this model Thomsen-style anisotropy parameters are calculated to derive azimuth-dependent normal move out (NMO) velocities (see Grechka & Tsvankin 1998). In our presentation we will show results of our approach from sonic well logs in the Surat Basin to investigate the potential of reconstructing S-wave velocity anisotropy and fracture density from azimuth dependent NMO velocities profiles.

  19. The Synthesis of N-Acetyllactosamine Functionalized Dendrimers, and the Functionalization of Silica Surfaces Using Tunable Dendrons and beta-Cyclodextrins

    Galectin-3 is beta-galactoside binding protein which is found in many healthy cells. In cancer, the galectin-3/tumor-associated Thomsen-Friedenreich antigen (TF antigen) interaction has been implicated in heterotypic and homotypic cellular adhesion and apoptotic signaling pathways. However, a stronger mechanistic understanding of the role of galectin-3 in these processes is needed. N-acetyllactosamine (LacNAc) is a non-native ligand for galectin-3 which binds with comparable affinity to the TF antigen and therefore an important ligand to study galectin-3 mediated processes. To study galectin-3 mediated homotypic cellular aggregation, four generations of polyamidoamine (PAMAM) dendrimers were functionalized with N-acetyllactosamine using a four-step chemoenzymatic route. The enzymatic step controlled the regiochemistry of the galactose addition to N-acetylglucosamine functionalized dendrimers using a recombinant beta-1,4-Galactosyltransferase-/UDP-4'-Gal Epimerase Fusion Protein (lgtB-galE). Homotypic cellular aggregation, which is promoted by the presence of galectin-3 as it binds to glycosides at the cell surface, was studied using HT-1080 fibrosarcoma, A549 lung, and DU-145 prostate cancer cell lines. In the presence of small LacNAc functionalized PAMAM dendrimers, galectin-3 induced cancer cellular aggregation was inhibited. However, the larger glycodendrimers induced homotypic cellular aggregation. Additionally, novel poly(aryl ether) dendronized silica surfaces designed for reversible adsorbtion of targeted analytes were synthesized, and characterization using X-ray Photoelectron Spectroscopy (XPS) was performed. Using a Cu(I) mediated cycloaddition "click" reaction, beta-cyclodextrin was appended to dendronized surfaces via triazole formation and also to a non-dendronized surface for comparison purposes.   1. Common reflection point migration and velocity analysis for anisotropic media

    An efficient Kirchhoff-style prestack depth migration, called 'parsimonious' migration was developed a decade ago for isotropic 2D and 3D media. The common-reflection point (CRP) migration velocity analysis (MVA) was developed later for isotropic media. The isotropic parsimonious migration produces incorrect images when the media is actually anisotropic. Similarly, isotropic CRP MVA produces incorrect inversions when the medium is anisotropic. In this study both parsimonious depth migration and common-reflection point migration velocity analysis are extended for application to 2D tilted transversely isotropic (TTI) media and illustrated with synthetic P-wave data. While the framework of isotropic parsimonious migration may be retained, the extension to TTI media requires redevelopment of each of the numerical components, including calculation of the phase and group velocity for TTI media, development of a new two-point anisotropic ray tracer, and substitution of an initial-angle and anisotropic shooting ray-trace algorithm to replace the isotropic one. The 2D model parameterization consists of Thomsen's parameters (Vpo, epsilon, delta) and the tilt angle of the symmetry axis of the TI medium. The parsimonious anisotropic migration algorithm is successfully applied to synthetic data from a TTI version of the Marmousi-2 model. The quality of the image improves by weighting the impulse response by the calculation of the anisotropic Fresnel radius. The accuracy and speed of this migration makes it useful for anisotropic velocity model building. The common-reflection point migration velocity analysis for TTI media for P-waves includes (and inverts for) Vpo, epsilon, and delta. The orientation of the anisotropic symmetry axis have to be constrained. If it constrained orthogonal to the layer bottom (as it conventionally is), it is estimated at each CRP and updated at each iteration without intermediate picking.   2. Luminescence and Radiocarbon dating of roman time harbour sediments from Cologne - A comparison of different methods

    Optically stimulated luminescence (OSL) dating was applied to fluvial and colluvial deposits which were taken from a roman time harbour basin in Cologne. Hence, independent age control was given by roman artefacts (coins, bricks) of known age and by three radiocarbon data. For one sample (well sorted fluvial sand), a single aliquot regenerative (SAR) protocol was applied to coarse grain quartz. A large number of equivalent doses (80) were collected and after De-measurements, different statistical approaches were tested. The results show, that calculating the age by using the Mean or Median of all accepted De-values yields OSL ages of 3.5 ± 0.3 ka and 2.8 ± 0.3 ka, respectively, what would be significantly too old. The application of the Leading Edge Method (Lepper & McKeever 2002), the statistical approach after Fuchs & Lang (2001) or the Minimum Age Method (MAM3) after Galbraith et al. (1999) yield OSL age estimates of 2.0 ± 0.1 ka, 2.1 ± 0.1 ka and 1.7 ± 0.6 ka, respectively. Those ages fit very well to the 14C-ages and are in better agreement with the known archaeological background. For the colluvial sample, the quartz OSL signal was effected by feldspar impurities. For this sample, different methodological approaches were tested to see which one has the best potential to minimize the feldspar contribution. Therefore the post-IR blue stimulated quartz luminescence was recorded. The quartz signal was measured using continuous wave (CW) and pulsed OSL (POSL). For the CW measurements, it was tested if a prior IR-bleach @ 50°C or @ 225°C has better potential for minimizing the feldspar signal. In this case, an IR-bleach @ 225°C (Buylaert et al. in press) was the most effective way to obtain a higher purity of the quartz OSL signal.     In Quaternary sciences, luminescence dating (OSL, TL, RF) yielded paramount importance due to its capability to trace the geomorphological process itself. However, every method has its own limitation and, e.g., the choice of a mineral depends on its local availability and mineral specific characteristic. Since quartz has been proved as reliable dosimeter, but its usual dose saturation level of ca. 150 Gy might be not sufficient in particular cases, feldspar grains are preferred for dating events where higher saturation limits are desired or quartz is not available. However, feldspar suffers from anomalous fading and the efforts undertaken during the last decades to overcome this challenge varied in their success. By contrast, infrared radiofluorescence (IR-RF) of K-feldspar (Trautmann et al., 1999) seems to offer a promising alternative, but until date it has been rarely applied. Likely reasons for this lack of attention are a shortage of commercially available measurement equipment until the recent past and serious methodological doubts that had been raised in the literature (e.g., Buyleart et al., 2012). At the IRAMAT-CRP2A the formerly proposed IR-RF single aliquot regenerative dose (SAR) protocol approach (IRSAR, Erfurt et al., 2003) for K-feldspar grains have been adapted and enhanced (Frouin et al., 2015; Huot et al., 2015; Frouin et al., 2017) using (a) sample adapted bleaching settings and (b) a stimulation at higher temperatures. Together with more recent methodological findings, we present the advantage and the limitations of using the IR-RF dating approach for establishing reliable chronologies on believed Middle Pleistocene sediments.   4. Biocides in hydraulic fracturing: hazard and vulnerability with respect to potential groundwater pollution

    Biocides are one possible chemical additive to frack fluids and their role is to control bacterial growth. Bacterial growth might lead to biofilm build up; and acid sulfide species and hydrogen sulfide (H2S) production: biofilm build up may reduce formation permeability and hinder gas extraction. Kahrilas et al. (2014) published a review of common biocides used in fracking in the USA. The biocides assessed in the review were the sixteen most commonly used in the USA, based on the hydraulic fracturing chemical registry Frac Focus (Frac Focus, 2015). However, the review of Kahrilas et al. (2014) contained no data or observations and so the objective of this study was to consider whether biocides proposed for use in hydrofacturing could be a threat to English groundwater. The study considered all groundwater samples analysed for biocides in English groundwater between 2005 and 2014. The monitoring records were compared to: records of application (both amount and area); and chemical and molecular data for the biocides. The study did not use traditional adsorption and degradation data as these parameters are to prone to variability and are not pure molecular parameters. The study then used the approach of Worrall and Thomsen (2004) to consider the hazard represented by proposed frack biocides and the approach of Worrall and Kolpin (2003) to consider the vulnerability of the areas of potential shale gas exploitation. The study showed that of the 113 biocides tested for in English groundwaters in the decade 2005 - 2014 that 95 were detected above 0.1 g/l . Of these 95, 41 were compounds that were not recorded as being applied during the period of record and the detection of these 41 compounds did not decline over the 10 year period which implies very long residence times and that once compounds do pollute an aquifer then they will be a persistent problem.   5. Insulin aspart pharmacokinetics: an assessment of its variability and underlying mechanisms.

    Insulin aspart (IAsp) is used by many diabetics as a meal-time insulin to control post-prandial glucose levels. As is the case with many other insulin types, the pharmacokinetics (PK), and consequently the pharmacodynamics (PD), is associated with clinical variability, both between and within individuals. The present article identifies the main physiological mechanisms that govern the PK of IAsp following subcutaneous administration and quantifies them in terms of their contribution to the overall variability. CT scanning data from Thomsen et al. (2012) are used to investigate and quantify the properties of the subcutaneous depot. Data from Brange et al. (1990) are used to determine the effects of insulin chemistry in subcutis on the absorption rate. Intravenous (i.v.) bolus and infusion PK data for human insulin are used to understand and quantify the systemic distribution and elimination (Pørksen et al., 1997; Sjöstrand et al., 2002). PK and PD profiles for type 1 diabetics from Chen et al. (2005) are analyzed to demonstrate the effects of IAsp antibodies in terms of bound and unbound insulin. PK profiles from Thorisdottir et al. (2009) and Ma et al. (2012b) are analyzed in the nonlinear mixed effects software Monolix® to determine the presence and effects of the mechanisms described in this article. The distribution of IAsp in the subcutaneous depot show an initial dilution of approximately a factor of two in a single experiment. Injected insulin hexamers exist in a chemical equilibrium with monomers and dimers, which depends strongly on the degree of dilution in subcutis, the presence of auxiliary substances, and a variety of other factors. Sensitivity to the initial dilution in subcutis can thus be a cause of some of the variability. Temporal variations in the PK are explained by variations in the subcutaneous blood flow.     Diabetes Self-care Activities Measure scores, clinical measures, comorbid conditions, health services resource consumption, and technology system usage statistics. We have completed phase 1 data collection. Formal analysis of phase 1 data has not been completed. We have obtained institutional review board approval and began phase 1 research in late fall 2016. The study hypotheses suggest that patients can, and will, improve their activation in chronic care management. Improved activation should translate into improved diabetes self-care. Expected benefits of this research to the scientific community and health care services include improved understanding of how to leverage mHealth technology to activate patients living with type 2 diabetes in self-management behaviors. The research will shed light on implementation strategies in integrating mHealth into the clinical workflow of the PCMH setting.     m scale). For each of the above properties, leading-edge experimental techniques have been associated with novel interpretation tools. In this contribution, these experimental and interpretation methods are described. Relationships between the measured properties and the corresponding micro-/meso-structural features are discussed. For example, P-wave velocity was measured along 100 different propagation paths on a single cylindrical shale specimen using miniature ultrasonic transducers. Assuming that (i) the elastic tensor of this shale is transversely isotropic; and (i) the sample has been cored perfectly perpendicular to the bedding plane (symmetry plane is horizontal), Thomsen's anisotropy parameters inverted from the measured velocities are: - P-wave velocity along the symmetry axis (perpendicular to the bedding plane) αo=3.45km/s; - P-wave anisotropy ɛ=0.12; - Parameter controlling the wave front geometry δ=0.058. A novel inversion algorithm allows for recovering these parameters without assuming a priori a horizontal bedding (symmetry) plane. The inversion of the same data set using this algorithm yields (i) αo=3.23km/s, ɛ=0.25 and δ=0.18, and (ii) the elastic symmetry axis is inclined of ω=30° with respect to the specimen's axis. Such difference can have strong impact on field applications (AVO, ray tracing, tomography).

  9. Brittle Creep of Tournemire Shale: Orientation, Temperature and Pressure Dependences

    Time and temperature dependent rock deformation has both scientific and socio-economic implications for natural hazards, the oil and gas industry and nuclear waste disposal. During the past decades, most studies on brittle creep have focused on igneous rocks and porous sedimentary rocks. To our knowledge, only few studies have been carried out on the brittle creep behavior of shale. Here, we conducted a series of creep experiments on shale specimens coming from the French Institute for Nuclear Safety (IRSN) underground research laboratory located in Tournemire, France. Conventional tri-axial experiments were carried under two different temperatures (26˚ C, 75˚ C) and confining pressures (10 MPa, 80 MPa), for three orientations (σ1 along, perpendicular and 45˚ to bedding). Following the methodology developed by Heap et al. [2008], differential stress was first increased to ˜ 60% of the short term peak strength (10-7/s, Bonnelye et al. 2016), and then in steps of 5 to 10 MPa every 24 hours until brittle failure was achieved. In these long-term experiments (approximately 10 days), stress and strains were recorded continuously, while ultrasonic acoustic velocities were recorded every 1˜15 minutes, enabling us to monitor the evolution of elastic wave speed anisotropy. Temporal evolution of anisotropy was illustrated by inverting acoustic velocities to Thomsen parameters. Finally, samples were investigated post-mortem using scanning electron microscopy. Our results seem to contradict our traditional understanding of loading rate dependent brittle failure. Indeed, the brittle creep failure stress of our Tournemire shale samples was systematically observed ˜50% higher than its short-term peak strength, with larger final axial strain accumulated. At higher temperatures, the creep failure strength of our samples was slightly reduced and deformation was characterized with faster 'steady-state' creep axial strain rates at each steps, and larger final axial strain

    singularity point may even vanish. The Thomsen parameters are helpful in fluid type indication in shale. An indicator of gas-filled aligned cracks is ?? > ??. If aligned cracks in illite-rich shale are brine-filled, ?? < ??. Negative value of ?? indicates brine-filled cracks in illite-rich shale. A shale with brine-filled cracks exhibits higher Vp/Vs ratio in the vertical direction as compared to the gas-filled shale. A disorientation of clay platelets and brine-filled cracks may lead to that the singularity point is absent for brine-saturated shale as well. In this case one can also observe ?? > ?? and decreased values of Vp/Vs in the vertical direction as in the case of gas-filled cracks. In the presence of vertically aligned cracks, shales exhibit distinctly revealed features of orthorhombic symmetry.     our results, in terms of the P-T locations of the solidus, phengite- and carbonate-out boundaries, to the thermal structures of the slab-surface in cold-intermediate subduction zones indicates that most of the phengite-bound H2O and carbonate-bound CO2 are recycled into the deep upper mantle (~200 km depth). On the other hand, substantial amounts of C-O-H volatiles, in the form of either hydrous silicate melt or K-rich calcio-carbonatitic melt, are likely to be released from relatively hot subducting slabs. The observation of carbonate melt inclusion in cpx and garnet in deeply subducted carbonate-rich sediments [3] might be explained by our experimental results that carbonatite is the stable near-solidus sediment melt at deep sub-arc depths.     demonstrate that previous isotopic results on graphite deficient Isua rocks obtained by single step combustion are unreliable. The proposed biologic significance of graphite occurring as inclusions in apatite [4] in Isua rocks is not supported by our findings because such graphite-apatite association can only be found in biologically irrelevant metacarbonate rocks. The isotopic systematics of the epigenetic processes responsible for formation of isotopically light graphite enclosed in apatite crystals [4] will be discussed, integrating new ion microprobe isotope data on graphite in apatite and other mineral phases occurring in Isua metacarbonates.     The first popular lecture on the Einstein theory of relativity was given in Estonia already in 1914 by Jaan Sarv (1877-1954)[1],afterwards a professor of mathematics at the Tartu University. The first student courses on special relativity were delivered by Professor of Mathematics Juri Nuut (1892-1952): non-Euclidean geometry (1930), the mathematical theory of relativity (1932/1933),the Lorenz transformations (1937). His own research work concerned the Lobachevsky geometry [7] and its application to cosmology [6]. Harald Keres qraguated from the Tartu University in 1936. He gave the first student course on general relativity (based on books [11-14]in 1940.In 1942,he got the dr.phil.nat degree form the Tartu University for his theses "Raum und Zeit in der allgemeinen Relativitatstheorie". The degree of the doctor of mathematical and physical sciences was confirmed by VAK (the All-Union Higher Attestation Commission) in 1949.In this period, he got aquainted with the leading Soviet scientists working on General Relativity, prof.V.A.Fock,Prof.D.D.Ivanenko,Prof.A.Z.Petrov,and Prof.M.F.Shirokov. After World War two all-union university courses were introduced in Tartu State University. According to the curriculum of the course the special theory of relativity is a part of electrodynamics obligatory for all students of the department of Physics. From 1947 till 1985 this course was delivered by Prof.PaulKard(1914-1985).He also published a number of text-books on the subject [15-19]. The general theory of relativity was read by Prof.H.Keres in 1951-1960 and later by his pupils R.Lias and A.Koppel [20-23] as a special course for students specializing in theoretical Physics. The first PHD-s in general relativity were made by R.Lias [27](1954) and I.Piir [28] (1955). In 1961, Prof.H.Keres was elected a member of the Academy of Sciences of the Estonian S.S.R.     the WDC-A for STP and the STP Division Chief for NOAA NGDC. She was passionate about the World Data Center system and maintaining data archives for future generations. She would introduce herself as "I am the World Data Center for Solar-Terrestrial Physics." Attending many foreign and U.S. meetings, she constantly searched for new data sets to add to the STP collection. She retired in 1980 after 38 years of federal service. When she was inducted into the Colorado Women's Hall of Fame in March 2000, she said: "My work with the World Data Centers introduced me to colleagues worldwide that became a source of much enjoyment, seeing them periodically at the international scientific meetings in Europe, Asia, and Australia." She was a past chair of the Denver Section of the Society of Women Engineers and very active in encouraging girls to study math and science. A member of the Association of Federal Professional and Administrative Women (AFPAW) and the Federally Employed Women (FEW), she supported improving the status of women. Virginia categorized herself as a joiner. She was active in many organizations, achieving life membership in the American Association of University Women (AAUW) and the Young Women's Christian Association (YWCA). In her younger days she was a figure skater and she enjoyed square dancing, playing golf, and traveling. She also enjoyed the arts and held season tickets to the University of Colorado Artist Series, the Denver Center for the Performing Arts, the opera, and the Colorado Music Festival. She was preceded in death by her parents and her brother Rush. Survivors include a nephew, Rush B. Lincoln III, a niece Deborah Lincoln Niekras, four great nieces and a great nephew. Her memoirs, "My Busy Life: How I Never Stopped Enjoying It" by Jeanette Virginia Lincoln, are available at the Carnegie Library in Boulder, Colorado. Also available are her history of her father "Rush Blodget Lincoln, My Father - the General" and a history of her mother's family

Bacterial microtextures in all of the above mentioned deposits are as a rule associated with oxysulphides i.e. compounds with mixed and intermediate sulphur valences (Kucha et al., 1989). The origin of oxysulphides is probably related either directly to incomplete bacterial reduction of the sulphatic sulphur, or reaction of bacterial H2S with sulphatic S present in the fluids. Some of peloids are composed of oxysulphides (Kucha & Stumpfl, 1992; Kucha, 2003). Therefore, an interpretation of the S isotopic signature of bacterial textures should consider not only microbial community structure, but also the oxidative part of the sulphur cycle proceeding through compounds with mixed sulphur valences. We performed a comprehensive transcriptional analysis of 460 early-stage urothelial carcinomas and showed that NMIBC can be subgrouped into 3 major classes with basal- and luminal-like characteristics and different clinical outcomes. Large differences in biological processes such as the cell cycle, epithelial-mesenchymal transition, and differentiation were observed. Analysis of transcript variants revealed frequent mutations in genes encoding proteins involved in chromatin organization and cytoskeletal functions. Furthermore, mutations in well-known cancer driver genes (e.g., TP53 and ERBB2) were primarily found in high-risk tumors, together with APOBEC-related mutational signatures. The identification of subclasses in NMIBC may offer better prognostication and treatment selection based on subclass assignment. Copyright © 2017 Elsevier Inc. All rights reserved.

[3]: The photometric redshift method makes it possible to judge the distance to a remote celestial object (a galaxy, a quasar, a gamma-ray burst afterglow) from its measured colours. It is based on the proportionality between the distance and the velocity along the line of sight (Hubble's law) that reflects the expansion of the Universe. The larger the distance of an object is, the larger is its velocity and, due to the Doppler effect, the spectral shift of its emission towards longer (redder) wavelengths. Thus, the measured colour provides a rough indication of the distance. Examples of this method are shown in ESO PR 20/98 (Photos 48a/00 and 48e/00). [4]: In fact, the object was so faint that the positioning of the spectrograph slit had to be done in "blind" offset, i.e. without actually seeing the object on the slit during the observation. This very difficult observational feat was possible because of excellent preparations by the team of astronomers and the very good precision of the telescope and instrument.     important ingredient in the cosmological models is the dark matter that is believed to contribute about 95% of the mass of the universe. The present confirmation of the predictions of the models therefore also indirectly confirms that it is the dark matter that controls the formation of structures in the universe. However, there is still a long way to go before it will be possible to make a more detailed comparison between observations and predictions, e.g., from PR Photo 19e/01 to PR Photo 19a/01 ! Asked about what they consider the most important consequence of their observations, the team responds: " We have shown that we now have an observational method with which we may study the cosmic web in the early universe, and the VLT is a great tool for such studies. The way forward is now pretty clear - we just have to find those faint and distant LEGOs and then do the spectral observations from which we may determine how they are distributed in space ". [00:03:36] Natacha Chough: We do take some samples and stuff in flight on return, but, you know, the crew can be pretty symptomatic in terms of like returning to a 1G environment, and so we kind of mitigate a lot of the symptoms that they're having, motion sickness, that type of thing, in the early hours, post-landing. [00:03:54] Host: So now you're a -- you're a flight surgeon now, right? Who's your crew members that you're working with? [00:03:59] Natacha Chough: So currently I'm assigned to Jeanette Epps, and she's launching next spring. [00:04:03] Host: Okay. Okay. So you -- what's some of the stuff you have to do this early ahead of time? [00:04:08] Natacha Chough: So right now, we just did her L minus 6 months physical, make sure that, you know, she's still within standards for long duration spaceflight. She's actually out of the country right now because in this part of the pre-launch timeframe, she and Alex [inaudible], he's a crew member, and then Sergei [inaudible], the Russian crew member, they're all serving as the backup crew to the prime crew that's launching this December. [00:04:33] Host: Oh, okay. So they're out there with Scott Tingle and [inaudible] and those guys? Okay, cool. Very cool. So you're -- you don't have to follow them for that then? You get to stay here? [00:04:43] Natacha Chough: Yeah. In the meantime, you know, there's a lot of just like pre-travel prep, making sure all of us, including the docs, are up on our immunizations for, you know, upcoming travel. In the next few months before launch, we'll also get together with her in our pharmacy, and make sure that she's got any prescription meds she takes on a regular basis put in these ISS medical accessory packs, people take, you know, nutritional supplements or daily vitamins or whatever, we make sure that all that is packed for them and any motion sickness meds they might need on the
