    Photometric data for 44 asteroids were collected over 54 nights of observing during 2009 October thru 2010 April at the Oakley Southern Sky Observatory.   3. Enantioselectivity and Thermostability of a Novel Hyperhermotolerant Lipase from Geobacillus Thermodenitrificans nr68 (Lip.nr-68) on Secondary Racemic Alcohols Acetylation

    In our previous work, a new lipase enzyme has been purified from a species identified as a Gram negative Geobacillus thermodenitrificans nr68, isolated from a hot spring in Malaysia with growth temperature of 48°C. This new lipase, called Lip.nr-68 has been characterized as a hyperthermotolerant protein with high stability at 65°C and has been showing excellent characteristics that are very much comparable yet better than some of those of well-known industrially-used lipases. It shows high activity against long-chain triglycerides with molecular weight of the purified enzyme estimated to be 33.5 kDa using SDS-PAGE analysis. This paper is focusing on hyperthermotolerant Lip.nr-68 performance in promoting for enantioselectivity activities towards three secondary racemic alcohols namely 1-phenylethanol, 1-cyclohexilethanol and 1-(naft-2-il) ethanol by acetylation with vinyl acetate. Lip.nr-68 has been confirmed to show high and usual enantioselectivitiy according to the Kazlauskas Rule towards all secondary racemic alcohols and has significantly approved as an enantiomer selective biocatalyst towards 1-phenylethanol and 1-cyclohexylethanol at 65°C. Lip.nr-68 has showed a reduction of (R) and (S) enantiomers as well as the production of 68-98% ee and almost 94% yield of 3-4 mg/ml for 1-cyclohexilethanol.

  4. Est16, a New Esterase Isolated from a Metagenomic Library of a Microbial Consortium Specializing in Diesel Oil Degradation.

    Lipolytic enzymes have attracted attention from a global market because they show enormous biotechnological potential for applications such as detergent production, leather processing, cosmetics production, and use in perfumes and biodiesel. Due to the intense demand for biocatalysts, a metagenomic approach provides methods of identifying new enzymes. In this study, an esterase designated as Est16 was selected from 4224 clones of a fosmid metagenomic library, revealing an 87% amino acid identity with an esterase/lipase (accession number ADM63076.1) from an uncultured bacterium. Phylogenetic studies showed that the enzyme belongs to family V of bacterial lipolytic enzymes and has sequence and structural similarities with an aryl-esterase from Pseudomonas fluorescens and a patented Anti-Kazlauskas lipase (patent number US20050153404). The protein was expressed and purified as a highly soluble, thermally stable enzyme that showed a preference for basic pH. Est16 exhibited activity toward a wide range of substrates and the highest catalytic efficiency against p-nitrophenyl butyrate and p-nitrophenyl valerate. Est16 also showed tolerance to the presence of organic solvents, detergents and metals. Based on molecular modeling, we showed that the large alpha-beta domain is conserved in the patented enzymes but not the substrate pocket. Here, it was demonstrated that a metagenomic approach is suitable for discovering the lipolytic enzyme diversity and that Est16 has the biotechnological potential for use in industrial processes.

  5. COLOR OF THE STARS: Oh Be A Fine Girl, Kiss Me!

    Classification of stars by color is important in stellar studies because from it we are able to attain essential information about stars like: temperature, composition, age and mass; from these we can also derive its history, and future evolution!. This classification can be done by photometry or spectroscopy. Photometry provides information from more stars in a given field of view, magnitude and approximate size. The Strömvil photometric system, developed by V. Straizys (Vilnius Observatory, Lithuania), allows more precise photometry using 7 filters, ranging from 330-700 nm. Since the color of a star is associated with the wavelength of the electromagnetic radiation of light emitted by it, each filter allows only certain wavelengths to go through into the CCD camera; then, each neighboring wavelength band can be compared against the others and the color relationship can be converted to magnitude. Our Milky Way galaxy has billions of stars, of which we only have information from a small set. We obtained images of the NGC6811 and NGC6819 Open clusters, and the M56 Globular cluster at the Vatican Advanced Technology Telescope in Mt Graham AZ. During an 8 night observing run, images were taken in each filter with 3 different pointings overlapping by 2 arc-min. Calibration by known standards from A. Kazlauskas (e.i. Baltic Astronomy Vol II) that fall in the observed regions will be done. From this photometry other star information; such as luminosity, distance, metallicity, surface gravity, and spectral class will be determined.
