  1. Isolation and Identification of Sodium Fluoroacetate Degrading Bacteria from Caprine Rumen in Brazil

    The objective of this paper was to report the isolation of two fluoroacetate degrading bacteria from the rumen of goats. The animals were adult goats, males, crossbred, with rumen fistula, fed with hay, and native pasture. The rumen fluid was obtained through the rumen fistula and immediately was inoculated 100 μL in mineral medium added with 20 mmol L−1 sodium fluoroacetate (SF), incubated at 39°C in an orbital shaker. Pseudomonas fluorescens (strain DSM 8341) was used as positive control for fluoroacetate dehalogenase activity. Two isolates were identified by 16S rRNA gene sequencing as Pigmentiphaga kullae (ECPB08) and Ancylobacter dichloromethanicus (ECPB09). These bacteria degraded sodium fluoroacetate, releasing 20 mmol L−1 of fluoride ion after 32 hours of incubation in Brunner medium containing 20 mmol L−1 of SF. There are no previous reports of fluoroacetate dehalogenase activity for P. kullae and A. dichloromethanicus. Control measures to prevent plant intoxication, including use of fences, herbicides, or other methods of eliminating poisonous plants, have been unsuccessful to avoid poisoning by fluoroacetate containing plants in Brazil. In this way, P. kullae and A. dichloromethanicus may be used to colonize the rumen of susceptible animals to avoid intoxication by fluoroacetate containing plants. PMID:22919294

  4. Pigmentiphaga aceris sp. nov., isolated from tree sap.

    Two Gram-stain-negative bacterial strains, SAP-32T and SAP-36, were isolated from sap drawn from the Acer pictum from Mount Halla in Jeju, Republic of Korea. The organisms were strictly aerobic, non-sporulating, motile rods and showed growth at 10-30 °C, pH 7-8 and with 0-2 % NaCl. The major isoprenoid quinone was Q-8. The predominant fatty acids were C16 : 0, cyclo-C17 : 0, summed feature 3 and C18 : 0. The polar lipids contained phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, an unknown aminophosphoglycolipid, an unknown glycolipid, an unknown phospholipid and two unknown lipids. The DNA G+C content was 64.4 mol%. The results of phylogenetic analyses based on 16S rRNA gene sequences indicated that SAP-32T and SAP-36 formed a distinct cluster with members of the genus Pigmentiphaga within the family Alcaligenaceae. Both strains showed 16S rRNA gene sequence similarity of 100 % to each other. The closest relatives of the isolates were Pigmentiphaga daeguensis (97.08 % sequence similarity), Pigmentiphaga kullae (97.01 %) and Pigmentiphaga litoralis (96.73 %). On the basis of data from phenotypic, chemotaxonomic and phylogenetic analyses, SAP-32T (=KCTC 52619T=DSM 104039T) and SAP-36 (=KCTC 52620=DSM 104072) represent members of a novel species of the genus Pigmentiphaga, for which the name Pigmentiphaga aceris sp. nov. is proposed.

  5. Diversity of bacterial isolates from commercial and homemade composts.

    The diversity of heterotrophic bacterial isolates of three commercial and two homemade composts was studied. The commercial composts were produced from poultry litter (PC), sewage sludge (SC), municipal solid waste (MC), and homemade composts (thermal compost [DC] and vermicompost [VC]) from food wastes. The taxonomic and physiological diversity of the heterotrophic culturable bacteria was assessed using phenotypic and genotypic characterization and the analysis of the partial 16S rRNA gene sequence. Composts DC and SC presented the higher genotypic diversity, as could be inferred from the number of distinct genotypic patterns observed, 28 and 21, respectively. Gram-positive bacteria, mainly Firmicutes, were predominant in all the composts. Some organisms related with taxa rarely reported in composts, as Rhodanobacter spathiphylli, Moraxella osloensis, Lysobacter, Corynebacterium, Pigmentiphaga kullae, and new taxa were also isolated. The highest relative proportion of isolates able to degrade starch was found in compost SC (> 70%), to degrade gelatine in compost DC (> 70%), to degrade Tween 80 in compost PC (> 90%), and to degrade poly-epsilon-caprolactones in compost DC (> 80%). Compost MC presented the lowest relative proportions of isolates able to degrade starch (< 25%), gelatine (< 20%), and poly-epsilon-caprolactone (< 40%). When compared with the others, the homemade composts presented higher relative proportions of Gram-positive isolates able to inhibit the target organisms Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, or Pseudomonas aeruginosa. In compost MC, none of the Gram-positive isolates was able to inhibit those targets.

    A novel betaproteobacterium, designated strain Ch07(T), was isolated from granules from the wastewater-treatment bioreactor of an alcohol fermentation factory in South Korea. In order to determine its taxonomic position, the novel strain was characterized using a polyphasic approach. The new strain was Gram-negative, facultatively anaerobic, non-spore-forming, motile and short rod-shaped. 16S rRNA gene sequence analysis revealed that strain Ch07(T) belonged to the class Betaproteobacteria, being related to Pusillimonas noertemannii BN9(T) (gene sequence similarity 97.30 %), Achromobacter xylosoxidans subsp. xylosoxidans DSM 10346(T) (97.09 %), Bordetella pertussis DSM 5571(T) (97.01 %), Pigmentiphaga kullae DSM 13608(T) (96.68 %) and Castellaniella defragrans DSM 1214(T) (96.47 %). The results of DNA-DNA hybridization tests showed that reassociation values were less than 62 % with respect to these closely related type strains. Chemotaxonomic data showed that strain Ch07(T) possessed ubiquinone Q-8. The G+C content of the genomic DNA was 67.9+/-0.1 mol%. The major polyamine of strain Ch07(T) was putrescine. The major polar lipids of strain Ch07(T) were phosphatidylethanolamine, followed by diphosphatidylglycerol and phosphatidylglycerol. When strain Ch07(T) was incubated on tryptic soy agar, the major cellular fatty acids were C(16 : 0), C(17 : 0) cyclo, summed feature 3 (C(16 : 1)omega7c/iso C(15 : 0) 2-OH) and summed feature 5 (C(18 : 1)omega7c/omega9t/omega12t). The results of DNA-DNA hybridizations, in combination with the chemotaxonomic and physiological data, demonstrated that strain Ch07(T) represents a novel species of a new genus, for which the name Parapusillimonas granuli gen. nov., sp. nov. is proposed. The type strain of the type species is Ch07(T) (=KCTC 12668(T)=LMG 24012(T)).

  7. Chronic neck pain in young adults: perspectives on anatomic differences.

    Neck pain (NP) is a common musculoskeletal disorder, but little is known about the associated risk factors. We compared anatomic differences in the neck and trunk area of young adult patients with chronic NP and control subjects without NP to identify risk factors and predictors. This is an age-, sex-, and body mass index-matched retrospective case-control study of a consecutive sample. Patients with axial NP for longer than 6 months (23 males and 25 females) and pain-free volunteers (23 males and 25 females) were included. Outcome measures were linear and angular dimensions of the cervicothoracic junction. Midsagittal magnetic resonance imaging scans of the cervicothoracic spine were obtained. Four linear and four angular parameters were identified and measured. These parameters included depth of the T1-manubrium arch (T1AD), depth of the thoracic cage (TXD), tangential height of T1 (T1H1), relative height of T1 (T1H2), T1 slope (T1S), thoracic inlet inclination (TiI), T1-manubrium arch inclination (T1AI), and the angular difference between TiI and T1AI (TiI-T1AI). The measurements were taken by two neurosurgeons. Depth of the T1-manubrium arch and TiI were identified as predictors for NP in the binary logistic regression analysis. Each millimeter increase in T1AD lessened the probability of NP with an adjusted odds ratio (OR) of 0.823 (95% confidence interval [CI], 0.701-0.966) in females and 0.809 (95% CI, 0.681-0.959) in males. Each degree increase in TiI was associated with the probability of NP with an adjusted OR of 1.247 (95% CI, 1.060-1.466) in males. Measurement of cervicothoracic junctional structures is a reliable and feasible method of estimating potential predictor of chronic NP in young adults. Forward inclination of the thoracic inlet in males and a shallow thoracic cage in females were identified as important predictors. Copyright © 2014 Elsevier Inc. All rights reserved.
