  1. Three-dimensional reconstruction of brain-wide wiring networks in Drosophila at single-cell resolution.

    Animal behavior is governed by the activity of interconnected brain circuits. Comprehensive brain wiring maps are thus needed in order to formulate hypotheses about information flow and also to guide genetic manipulations aimed at understanding how genes and circuits orchestrate complex behaviors. To assemble this map, we deconstructed the adult Drosophila brain into approximately 16,000 single neurons and reconstructed them into a common standardized framework to produce a virtual fly brain. We have constructed a mesoscopic map and found that it consists of 41 local processing units (LPUs), six hubs, and 58 tracts covering the whole Drosophila brain. Despite individual local variation, the architecture of the Drosophila brain shows invariance for both the aggregation of local neurons (LNs) within specific LPUs and for the connectivity of projection neurons (PNs) between the same set of LPUs. An open-access image database, named FlyCircuit, has been constructed for online data archiving, mining, analysis, and three-dimensional visualization of all single neurons, brain-wide LPUs, their wiring diagrams, and neural tracts. We found that the Drosophila brain is assembled from families of multiple LPUs and their interconnections.     Ultrasound and microbubbles-mediated drug delivery has become a promising strategy to promote drug delivery and its therapeutic efficacy. The aim of this research was to assess the effects of microbubbles (MBs)-combined low-intensity pulsed ultrasound (LPUS) on the delivery and cytotoxicity of curcumin (Cur) to human breast cancer MDA-MB-231 cells. Under the experimental condition, MBs raised the level of acoustic cavitation and enhanced plasma membrane permeability; and cellular uptake of Cur was notably improved by LPUS-MBs treatment, aggravating Cur-induced MDA-MB-231 cells death. The combined treatment markedly caused more obvious changes of cell morphology, F-actin cytoskeleton damage and cell migration inhibition. Our results demonstrated that combination of MBs and LPUS may be an efficient strategy for improving anti-tumor effect of Cur, suggesting a potential effective method for antineoplastic therapy.   3. Mechanical properties of light-curing composites polymerized with different laboratory photo-curing units.

    This study aimed to analyze the microhardness (KHN) and diametral tensile strength (DTS) of two hybrid resin composites (TPH Spectrum and Filtek Z250). To this end, the composites were polymerized with six laboratory photo-curing units (LPUs) and the results compared with an alternative polymerization method using conventional halogen light source in conjunction with additional polymerization in an autoclave (15 minutes/100 degrees C). LPUs were used following the manufacturers' instructions. Diametral tensile strength and Knoop hardness tests were conducted for all groups (n=5). Data were statistically compared using ANOVA and Tukey's test (alpha = 0.05). Among the LPUs, the one that provided light curing in conjunction with heat and nitrogen pressure resulted in a significant increase in KHN and DTS of resin composites. Between the resin composites, Filtek Z250 showed higher hardness values than TPH Spectrum. It was concluded that the use of alternative polymerization with conventional light polymerization and autoclave was feasible with a wide implication for the general public in terms of reduced dental treatment cost.

  4. Finite element analysis of heat generation from different light-polymerization sources during cementation of all-ceramic crowns.

    Exothermic composite resin chemical reactions and visible light generators can produce heat during a restorative polymerization process. These thermal changes in restored teeth may cause pain and irreversible pulpitis. The purpose of this study was to analyze the temperature distribution and heat flow patterns of a crowned mandibular second premolar tooth model using 3 different light-polymerization technologies and a finite element technique. A 2-dimensional finite element model was used to simulate a clinical condition. Heat flow and thermal stress distribution in a tooth during cementation of an all-ceramic crown using 4 commercially available light-polymerization units (LPUs), each with different wavelengths (Elipar TriLight, Elipar Freelight, Apollo 95 E, and ADT 1000 PAC), were investigated. The temperature values were measured at 3, 10, 12, and 40 seconds for each light-polymerizing unit (LPU) at 6 different finite element nodes. Two-dimensional temporal and spatial distribution of the thermal stress within the tooth, including the thermal coefficients and boundary conditions of the dental materials, were obtained and evaluated. The temperature at the nodal points did not exceed 42 degrees C, which is a threshold value for tissue vitality within the recommended operating periods at the dentin and pulp surface for all LPUs, except for Elipar TriLight. In the case of Elipar TriLlight, the temperatures at the dentin and pulp surfaces were 47 degrees C and 42 degrees C, respectively. When the light-polymerization units were used according to the manufacturers' operating procedures and without prolonged operating periods, with the exception of Elipar TriLight, the investigated LPUs did not produce significant heat. However, when the operating periods were prolonged, unacceptable temperature increases were observed, especially with the high-intensity LPUs.

  5. Connectomics-based analysis of information flow in the Drosophila brain.

    Understanding the overall patterns of information flow within the brain has become a major goal of neuroscience. In the current study, we produced a first draft of the Drosophila connectome at the mesoscopic scale, reconstructed from 12,995 images of neuron projections collected in FlyCircuit (version 1.1). Neuron polarities were predicted according to morphological criteria, with nodes of the network corresponding to brain regions designated as local processing units (LPUs). The weight of each directed edge linking a pair of LPUs was determined by the number of neuron terminals that connected one LPU to the other. The resulting network showed hierarchical structure and small-world characteristics and consisted of five functional modules that corresponded to sensory modalities (olfactory, mechanoauditory, and two visual) and the pre-motor center. Rich-club organization was present in this network and involved LPUs in all sensory centers, and rich-club members formed a putative motor center of the brain. Major intra- and inter-modular loops were also identified that could play important roles for recurrent and reverberant information flow. The present analysis revealed whole-brain patterns of network structure and information flow. Additionally, we propose that the overall organizational scheme showed fundamental similarities to the network structure of the mammalian brain. Copyright © 2015 Elsevier Ltd. All rights reserved.

  6. The effects of short-cycle sprints on power, strength, and salivary hormones in elite rugby players.

    This study examined the effects of short-cycle sprints on power, strength, and salivary hormones in elite rugby players. Thirty male rugby players performed an upper-body power and lower-body strength (UPLS) and/or a lower-body power and upper-body strength (LPUS) workout using a crossover design (sprint vs. control). A 40-second upper-body or lower-body cycle sprint was performed before the UPLS and LPUS workouts, respectively, with the control sessions performed without the sprints. Bench throw (BT) power and box squat (BS) 1 repetition maximum (1RM) strength were assessed in the UPLS workout, and squat jump (SJ) power and bench press (BP) 1RM strength were assessed in the LPUS workout. Saliva was collected across each workout and assayed for testosterone (Sal-T) and cortisol (Sal-C). The cycle sprints improved BS (2.6 ± 1.2%) and BP (2.8 ± 1.0%) 1RM but did not affect BT and SJ power. The lower-body cycle sprint produced a favorable environment for the BS by elevating Sal-T concentrations. The upper-body cycle sprint had no hormonal effect, but the workout differences (%) in Sal-T (r = -0.59) and Sal-C (r = 0.42) concentrations correlated to the BP, along with the Sal-T/C ratio (r = -0.49 to -0.66). In conclusion, the cycle sprints improved the BP and BS 1RM strength of elite rugby players but not power output in the current format. The improvements noted may be explained, in part, by the changes in absolute or relative hormone concentrations. These findings have practical implications for prescribing warm-up and training exercises.
