  1. Characterization of a novel MIIA domain-containing protein (MdcE) in Bradyrhizobium spp.

    Several genes coding for proteins with metal ion-inducible autocleavage (MIIA) domains were identified in type III secretion system tts gene clusters from draft genomes of recently isolated Bradyrhizobium spp. MIIA domains have been first described in the effectors NopE1 and NopE2 of Bradyrhizobium diazoefficiens USDA 110. All identified genes are preceded by tts box promoter motifs. The identified proteins contain one or two MIIA domains. A phylogenetic analysis of 35 MIIA domain sequences from 16 Bradyrhizobium strains revealed four groups. The protein from Bradyrhizobium sp. LmjC strain contains a single MIIA domain and was designated MdcE (MdcELmjC). It was expressed as a fusion to maltose-binding protein (MalE) in Escherichia coli and subsequently purified by affinity chromatography. Recombinant MalE-MdcELmjC-Strep protein exhibited autocleavage in the presence of Ca2+, Cu2+, Cd2+ and Mn2+, but not in the presence of Mg2+, Ni2+ or Co2+. Site-directed mutagenesis at the predicted cleavage site abolished autocleavage activity of MdcELmjC.     Many surface virulence factors of bacterial pathogens show mosaicism and confounding phylogenetic origin. The Streptococcus gordonii platelet-binding GspB protein, the Streptococcus sanguinis SrpA adhesin and the Streptococcus pneumoniae DiiA protein, share an imperfect 27-residue motif. Given the disparate domain architectures of these proteins and its association to invasive disease, this motif was named MiiA from Multiarchitecture invasion-involved motif A. MiiA is predicted to adopt a beta-sheet folding, probably related to the Ig-like fold, with a symmetrical positioning of two conserved aspartic residues. A specific hidden Markov model profiling MiiA was built, which specifically detected the motif in proteins from 58 species, mainly in cell-wall proteins from Gram-positive bacteria. These proteins contained one to ten MiiA motifs, which were embedded within larger repeat units of 70-82 residues. MiiA motifs combined to other domains and elements such as coiled-coils and low-complexity regions. The species carrying MiiA-proteins included commensals from the urogenital tract and the oral cavity, which can cause opportunistic endocarditis and sepsis. Intra-protein MiiA repeats showed a complex mixture of orthologal, paralogal and inter-species relationships, suggestive of a multistep origin. Presence of these repeats in proteins involved in oligosaccharide recognition and lifestyle of species suggest a putative function for MiiA repeats in sugars binding, probably those present in receptors of epithelial and blood cells. MiiA modules appear to have been transferred horizontally between species co-habiting in the same niche to create their own MiiA-containing determinants.   3. The darker side of fatherhood: clinical and developmental ramifications of the "Laius motif".

    "The Darker Side of Fatherhood" is intended as a clinical sequel to an earlier article, "Oedipus Revisited: Laius and the 'Laius Complex'". After briefly summarizing the allegorical implications of the various forgotten Oedipus myths and the father's fateful role within the Theban tragedy, this paper elaborates on those pederastic and filicidal inclinations that I believe to be universal among fathers. When such wishes are subject to primal repression, they enhance adaptation, fueling a father's aggressive dialogue with sons and daughters from infancy through the oedipal era into adolescence. Maladaptations arise when fathers give free rein to a more or less unalloyed destructive aggressivity toward offspring, a manifest hostility that can further serve to defend against ambisexual amibitions or identifications. Another pathogenic situation occurs when a father succumbs to an excessive neurotic inhibition in a desperate effort to repress such urges, thereby depriving his children of age- and state-appropriated aggressive stimulation in the form of play, active providing, and necessary discipline. Observational, analytic, and clinical case examples are presented to illustrate the implications for observation, developmental theory, and technique of an emphasis on a father's gender- and generation-specific aggression toward his offspring and its abiding intrapsychic reverberations.

  4. Single step purification of recombinant proteins using the metal ion-inducible autocleavage (MIIA) domain as linker for tag removal.

    For fast and easy purification, proteins are typically fused with an affinity tag, which often needs to be removed after purification. Here, we present a method for the removal of the affinity tag from the target protein in a single step protocol. The protein VIC_001052 of the coral pathogen Vibrio coralliilyticus ATCC BAA-450 contains a metal ion-inducible autocatalytic cleavage (MIIA) domain. Its coding sequence was inserted into an expression vector for the production of recombinant fusion proteins. Following, the target proteins MalE and mCherry were produced as MIIA-Strep fusion proteins in Escherichia coli. The target proteins could be separated from the MIIA-Strep part simply by the addition of calcium or manganese(II) ions within minutes. The cleavage is not affected in the pH range from 5.0 to 9.0 or at low temperatures (6°C). Autocleavage was also observed with immobilized protein on an affinity column.   5. Who killed Laius?: On Sophocles' enigmatic message.

    Using Laplanche's basic conceptualisation of the role of the other in unconscious processes, the author proposes a reading of Sophocles' tragedy, Oedipus the King, according to basic principles of dream interpretation. This reading corroborates contemporary literary perspectives suggesting that Sophocles' tragedy may not only convey the myth but also provide a critical analysis of how myths work. Important textual inconsistencies and incoherence, which have been noted through the centuries, suggest the existence of another, repressed story. Moreover, the action of the play points to enigmatic parental messages of infanticide and the silencing of Oedipus's story, as well as their translation into primordial guilt, as the origins of the tragic denouement. Oedipus's self-condemnation of parricide follows these enigmatic codes and is unrelated to, and may even contradict, the evidence offered in the tragedy as to the identity of Laius's murderers. Moreover, Sophocles' text provides a complex intertwining of hermeneutic and deterministic perspectives. Through the use of the mythical deterministic content, the formal characteristics of Sophocles' text, mainly its complex time perspective and extensive use of double meaning, dramatise in the act of reading an acute awareness of interpretation. This reading underscores the fundamental role of the other in the constitution of unconscious processes.

  6. Crawling from soft to stiff matrix polarizes the cytoskeleton and phosphoregulates myosin-II heavy chain

    On rigid surfaces, the cytoskeleton of migrating cells is polarized, but tissue matrix is normally soft. We show that nonmuscle MIIB (myosin-IIB) is unpolarized in cells on soft matrix in 2D and also within soft 3D collagen, with rearward polarization of MIIB emerging only as cells migrate from soft to stiff matrix. Durotaxis is the tendency of cells to crawl from soft to stiff matrix, and durotaxis of primary mesenchymal stem cells (MSCs) proved more sensitive to MIIB than to the more abundant and persistently unpolarized nonmuscle MIIA (myosin-IIA). However, MIIA has a key upstream role: in cells on soft matrix, MIIA appeared diffuse and mobile, whereas on stiff matrix, MIIA was strongly assembled in oriented stress fibers that MIIB then polarized. The difference was caused in part by elevated phospho-S1943–MIIA in MSCs on soft matrix, with site-specific mutants revealing the importance of phosphomoderated assembly of MIIA. Polarization is thus shown to be a highly regulated compass for mechanosensitive migration. PMID:23128239

  7. Myosin IIA/IIB restrict adhesive and protrusive signaling to generate front-back polarity in migrating cells.

    Migratory front-back polarity emerges from the cooperative effect of myosin IIA (MIIA) and IIB (MIIB) on adhesive signaling. We demonstrate here that, during polarization, MIIA and MIIB coordinately promote localized actomyosin bundling, which generates large, stable adhesions that do not signal to Rac and thereby form the cell rear. MIIA formed dynamic actomyosin proto-bundles that mark the cell rear during spreading; it also bound to actin filament bundles associated with initial adhesion maturation in protrusions. Subsequent incorporation of MIIB stabilized the adhesions and actomyosin filaments with which it associated and formed a stable, extended rear. These adhesions did not turn over and no longer signal to Rac. Microtubules fine-tuned the polarity by positioning the front opposite the MIIA/MIIB-specified rear. Decreased Rac signaling in the vicinity of the MIIA/MIIB-stabilized proto-bundles and adhesions was accompanied by the loss of Rac guanine nucleotide exchange factor (GEFs), like βPIX and DOCK180, and by inhibited phosphorylation of key residues on adhesion proteins that recruit and activate Rac GEFs. These observations lead to a model for front-back polarity through local GEF depletion.

  9. Perception of the upright and susceptibility to motion sickness as functions of angle of tilt and angular velocity in off-vertical rotation. [human tolerance to angular accelerations

    Motion sickness susceptibility of four normal subjects was measured in terms of duration of exposure necessary to evoke moderate malaise (MIIA) as a function of velocity in a chair rotated about a central axis tilted 10 deg with respect to gravitational upright. The subjects had little or no susceptibility to this type of rotation at 2.5 and 5.0 rpm, but with further increases in rate, the MIIA endpoint was always reached and with ever shorter test durations. Minimal provocative periods for all subjects were found at 15 or 20 rpm. Higher rotational rates dramatically reversed the vestibular stressor effect, and the subjects as a group tended to reach a plateau of relatively low susceptibility at 40 and 45 rpm. At these higher velocities, furthermore, the subjects essentially lost their sensation of being tilted off vertical. In the second half of the study, the effect of tilt angle was varied while the rotation rate was maintained at a constant 17.5 rpm. Two subjects were completely resistant to symptoms of motion sickness when rotated at 2.5 deg off vertical; with greater off-vertical angles, the susceptibility of all subjects increased sharply at first, then tapered off in a manner reflecting a Fechnerian function.

    Magnetic properties of three families of metal-organic coordinated networks which have the general form of M(II)A(4,4'-bipyridine), where M=Fe, Ni, Co, and Cu and A=Cl2, (ox) and (N3)2, are studied in this dissertation. Novel Ni(N3)2(4,4'-bipyridine), Co(N3)2(4,4'-bipyridine) and Cu(N 3)2(4,4'-bipyridine) have been synthesized. We applied different synthesis procedures and produced Ni, Co, and Cu azide compounds for the first time, thus leaving the hydrothermal route procedure. Powder x-ray diffraction at room temperature was done in order to establish the crystal structure of the members of these three families. It was found that all of them crystallize in orthorhombic structure, where transitional metals have an octahedral coordination. Since all three families have identical crystal structure we got opportunity to examine how ligands facilitate magnetic interaction between metallic centers and also to test existing magnetic theoretical models. Since 4,4'-bipyridine is much longer than other ligands, our systems can be considered as 1-D magnetic systems. Their interchain magnetic interactions are very weak, and they order magnetically at very low temperatures of the order of few K. Measurements of M(H) at temperatures T=1.9K and T=2K and chi(T) in different external magnetic fields in zero field and field cooled modes have been made. In the case of MCl2(4,4'-bipyridine) family of compounds, we observed ferromagnetic interactions between metal ions within the chains and antiferromagnetic interactions between adjacent chains. M(ox)(4,4'-bipyridine) family of metal-organic compounds has antiferromagnetic interactions between the transitional metal ions within the chain, while weak ferromagnetic interaction exists between the chains. All members in the M(N3)2(4,4'-bipyridine) family except in the case of the copper compound were found to have ferromagnetic interactions between metal ions within the chains and then antiferromagnetic interactions between adjacent

  11. Orthognathic Surgery in Craniofacial Microsomia: Treatment Algorithm

    Summary: Craniofacial microsomia is a broad term that covers a variety of craniofacial malformation conditions that are caused by alterations in the derivatives of the first and second pharyngeal arches. In general terms, diverse therapeutic alternatives are proposed according to the growth stage and the severity of the alteration. When craniofacial growth has concluded, conventional orthognathic surgery (Le Fort I osteotomy, bilateral sagittal split osteotomy, and genioplasty) provides good alternatives for MI and MIIA type cases. Reconstruction of the mandibular ramus and temporomandibular joint before orthognathic surgery is the indicated treatment for cases MIIB and MIII. The goal of this article is to establish a surgical treatment algorithm for orthognathic surgery on patients with craniofacial microsomia, analyzing the points that allow the ideal treatment for each patient to be chosen.   12. A Countermeasure for Space Motion Sickness

    the possibility of producing functionally useful adaptation during space flight without the penalty of disabling motion sickness by controlling the rate of the adaptive process by means of an appropriate stroboscopically presented environment. After several recent meetings with Professor Melvill Jones, we were encouraged to repeat the motion sickness portions of his and Mandl's 1981 stroboscopic experiment. In conducting this experiment we used a randomized cross-over design where subjects were randomly assigned to either a stroboscopic flash or no strobe for their first exposure in the experimental design. Twenty subjects (19 subjects completed the study) read a short passage from Treasure Island mounted on the wall approximately 1 m from their eyes while wearing left-right reversing prisms. The strobe on time of 3 microseconds and flash frequency of 4 Hz was set to equal that used in the original study. Motion sickness was scored using a modified Miller and Graybiel scale that we constructed to include symptoms that may be elicited under conditions where reversing prisms are worn. On this scale a score of 5 represented Malaise IIa (mild motion sickness) and a score of 8 or above is approaching frank sickness. Symptoms were tracked and recorded every 5 min during the task. Testing was limited to 30 min unless the subject had reached the MIIa score, at which time the test was terminated. Performance under stroboscopic illumination was significantly better than when the subjects read under normal room illumination while wearing the left-right reversing prisms. Based on these results we developed a goggle system using LCD material that can be strobed. To evaluate the effectiveness of stroboscopic goggles we tested an additional 9 subjects in addition to retesting 10 used in the stroboscopic pilot study described above.   13. Oedipus and the sphinx: triumph and catastrophe.

    The downfall of Oedipus in Sophocles' most famous play, Oedipus Tyrannus, is the reversal of his triumphant entry into the city of Thebes 17 years earlier. The oracle at Delphi has told the Thebans that the plague will end only when the murderer of Laius is identified and punished. The unconscious collusion between Oedipus and the Thebans to ignore the unsolved murder breaks down. Told by Teiresias that he is the culprit, Oedipus begins a desperate effort to find evidence to refute this accusation. When other witnesses come forward, his cause is lost. The effect of Oedipus's collusive interaction with the Thebans is balanced in Sophocles' account by the story of the riddle of the Sphinx. The Sphinx is a goddess of discontinuity. She represents the unconscious forces at work both in Oedipus's personal fantasies and in the collective fantasies of the Thebans. The tragedy results from the interaction of these fantasies in the government of the polis. Sophocles' play thus exemplifies the intricate intertwining of the child's innate wishes and the reaction they evoke from the environment during the oedipal period. The neurotic patient's illusion of sexual triumph over his or her father or mother as a child can be best understood through the interaction of these factors.

  14. The role of pre-oedipal and oedipal factors in psychic life.

    The Oedipus complex, a basic concept in Freudian theory, is an essential factor in the constitution of the human subject. It plays a key role in the structuring of the personality and in the orientation of desire. It is the oedipal triangular structure that precedes the pre-oedipal situation (in a logical, not chronological, order), and not vice versa. The oedipal structure exists before the infant's biological birth. It is present in the parents' desires and identifications, which inexorably fall upon each subject. That is why the author believes that it is necessary to leave behind a solipsistic reading of the nuclear complex of neuroses--a reading that is based solely on Oedipus's drive nucleus--and take a joint and comprehensive view of Laius's and Jocasta's histories and traumatic experiences, which were invested in their son. Among these three vertices, a dynamic set of forces emerges whereby a basic, original unconscious field phantasy is created that bears a unique narrative and an invisible and hermetic web made of passions and beliefs, scandals and secrets. This phantasy gives shape to an unrepeatable oedipal structure in each subject, a structure that articulates with the effects of the narcissistic and fraternal dynamic and may determine the subject's fate. This paper develops the following issues: 1) Oedipus, victimizer or victim?; 2) the generational confrontation as dynamic field; and 3) neuroses with a preponderance of dualistic relationships.

  15. Legacies and Memorializations in the Intergenerational Transmission of Psychic Life: Psychoanalytic Notes on Some of the Ways in Which Sons Assume Their Father's Mantle.

    The intergenerational transmission of psychic life has been difficult to explore psychoanalytically given that a treatment of both parent and child is rarely, if ever, feasible. Here the issue is explored in terms of father-son relationships. Three legendary fathers (Abraham, Laius, Shiva) are discussed as illustrating the filicidal legacy they imposed psychically and corporeally on their sons. Three contemporary case vignettes are then presented, indicating ways in which sons ambivalently memorialize their fathers. This material provides a model as to some of the ways in which sons assume the mantle of the father's conscious and unconscious dynamics in the course of their personal history. Paternal legacies generate "memorializations" that express profound and poignant conflicts animating the son's psychic life. In the context of this investigation, the significance of the distinction between actual fathers or father figures (who are mortal, who possess some sort of gendered qualities, and who may be present or absent, emotionally and physically) and the "symbolic father" (who is the locus of the phallus that organizes the possibility of representational discourse) is discussed. It is suggested that boys and men typically develop in terms of confusions around this distinction. The oedipal conundrum and challenge of the boy's passage into manhood is then formulated as follows: Masculinity involves both the integration of patricidal desire (in response to the paternal legacy) and the mourning of actual fathers or father figures. It also involves the acceptance of the human condition, which is both that of mortality and of our "castratedness" in relation to the "symbolic father" as the locus of the phallus (the only one who knows whereof "He" speaks and acts, and who can neither be mourned nor surpassed).

  16. Rhodopsin Photointermediates in 2D Crystals at Physiological Temperatures

    Bovine rhodopsin photointermediates formed in 2D rhodopsin crystal suspensions were studied by measuring the time dependent absorbance changes produced after excitation with 7 nanosecond laser pulses at 15, 25 and 35 °C. The crystalline environment favored the Meta I480 photointermediate, with its formation from Lumi beginning faster than it does in rhodopsin membrane suspensions at 35 °C and its decay to a 380 nm absorbing species being less complete than it is in the native membrane at all temperatures. Measurements performed at pH 5.5 in 2D crystals showed that the 380 nm absorbing product of Meta I480 decay did not display the anomalous pH dependence characteristic of classical Meta II in the native disk membrane. Crystal suspensions bleached at 35 °C and quenched to 19 °C showed that a rapid equilibrium existed on the ∼1 second time scale which suggests that the unprotonated predecessor of Meta II in the native membrane environment (sometimes called MIIa), forms in 2D rhodopsin crystals, but that the non-Schiff base proton uptake completing classical Meta II formation is blocked there. Thus, the 380 nm absorbance arises from an on-pathway intermediate in GPCR activation and does not result from early Schiff base hydrolysis. Kinetic modeling of the time-resolved absorbance data of the 2D crystals was generally consistent with such a mechanism, but details of kinetic spectral changes and the fact that the residuals of exponential fits were not as good as are obtained for rhodopsin in the native membrane suggested the photoexcited samples were heterogeneous. Variable fractional bleach due to the random orientation of linearly dichroic crystals relative to the linearly polarized laser was explored as a cause of heterogeneity but was found unlikely to fully account for it. The fact that the 380 nm product of photoexcitation of rhodopsin 2D crystals is on the physiological pathway of receptor activation suggests that determination of its structure would be of
