  1. An introduction to loricifera, cycliophora, and micrognathozoa.

    Loriciferans, cycliophorans and micrognathozoans are amongst the latest groups of animals to be discovered. Other than all being microscopic, they have very different body plans and are not closely related. Loriciferans were originally assigned to the Aschelminthes. However, both new molecular and ultrastructural researches have shown that Aschelminthes consist of two unrelated groups, Cycloneuralia and Gnathifera. Cycloneuralia may be included in the Ecdysozoa, including all molting invertebrates, and Gnathifera are more closely related to Platyhelminthes. The phylum Loricifera shares many apomorphic characters (e.g., scalids on the introvert) with both Priapulida and Kinorhyncha, and can be included in the taxon Scalidophora, a subgroup of Cycloneuralia. Cycliophora was originally allied to the Entoprocta and Ectoprocta (Bryozoa) based on ultrastructual research. Subsequent molecular data show they may be related to Rotifera and Acanthocephala, within the taxon Gnathifera. The phylogenetic position of Cycliophora is therefore not settled, and more ultrastructural and molecular data are needed. Micrognathozoa is the most recent major group of animals to be described. They show strong affinities with both Rotifera and Gnathostomulida (within the taxon Gnathifera), especially in the fine structure of the pharyngeal apparatus, where the jaw elements have cuticular rods with osmiophilic cores. Furthermore the micrognathozoans have two rows of multiciliated cells that form a locomotory organ, similar to that seen in some gastrotrichs and interstitial annelids. This character is never seen in Rotifera or in the monociliated Gnathostomulida. Rotifera and Acanthocephala always have a syncytial epidermis (Syndermata). Micrognathozoa lack this characteristic feature. Therefore, they are postulated to be placed basally in the Gnathifera, either as a sister-group to Gnathostomulida or as a sister-group to Rotifera + Acanthocephala.

  2. Colonization of maritime glacier ice by bdelloid Rotifera.

    Very few animal taxa are known to reside permanently in glacier ice/snow. Here we report the widespread colonization of Icelandic glaciers and ice fields by species of bdelloid Rotifera. Specimens were collected within the accumulation zones of Langjökull and Vatnajökull ice caps, among the largest European ice masses. Rotifers reached densities up to ∼100 individuals per liter-equivalent of glacier ice/snow, and were freeze-tolerant. Phylogenetic analyses indicate that glacier rotifers are polyphyletic, with independent ancestries occurring within the Pleistocene. Collectively, these data identify a previously undescribed environmental niche for bdelloid rotifers and suggest their presence in comparable habitats worldwide. Copyright © 2016 Elsevier Inc.   3. Phylogeny of Syndermata (syn. Rotifera): Mitochondrial gene order verifies epizoic Seisonidea as sister to endoparasitic Acanthocephala within monophyletic Hemirotifera.

    A monophyletic origin of endoparasitic thorny-headed worms (Acanthocephala) and wheel-animals (Rotifera) is widely accepted. However, the phylogeny inside the clade, be it called Syndermata or Rotifera, has lacked validation by mitochondrial (mt) data. Herein, we present the first mt genome of the key taxon Seison and report conflicting results of phylogenetic analyses: while mt sequence-based topologies showed monophyletic Lemniscea (Bdelloidea+Acanthocephala), gene order analyses supported monophyly of Pararotatoria (Seisonidea+Acanthocephala) and Hemirotifera (Bdelloidea+Pararotatoria). Sequence-based analyses obviously suffered from substitution saturation, compositional bias, and branch length heterogeneity; however, we observed no compromising effects in gene order analyses. Moreover, gene order-based topologies were robust to changes in coding (genes vs. gene pairs, two-state vs. multistate, aligned vs. non-aligned), tree reconstruction methods, and the treatment of the two monogonont mt genomes. Thus, mt gene order verifies seisonids as sister to acanthocephalans within monophyletic Hemirotifera, while deviating results of sequence-based analyses reflect artificial signal. This conclusion implies that the complex life cycle of extant acanthocephalans evolved from a free-living state, as retained by most monogononts and bdelloids, via an epizoic state with a simple life cycle, as shown by seisonids.   4. Biodiverse rotifer assemblage (Rotifera: Eurotatoria) of floodplain lakes of the Brahmaputra basin of lower Assam, northeast India: composition and ecosystem diversity

    The study aims to ascertain the hypothesis on the rich rotifer biodiversity of the floodplain lakes (beels) of the Brahmaputra river basin and to use these metazoans to assess trophic status or to characterize habitat variations of wetlands. The plankton samples collected from four beels of lower Assam revealed 160 Rotifera species belonging to 35 genera and 19 families. The richness is of biodiversity value as 38.0% and 57.0% of the rotifer species known till date from India and northeast India (NEI), respectively. One species each is new to the Oriental region and NEI, and three species are new to Assam; 23 species merit global biogeography interest and several exhibit distribution values in the Indian sub-region. The diverse Lecanidae > Brachionidae > Lepadellidae > Trichocercidae and speciose littoral-periphytic Lecane > Lepadella > Trichocerca, and richness of Brachionus spp. following removal of aquatic macrophytes are noteworthy. Overall rotifer composition showed homogeneity amongst beels while lower monthly richness and community similarities affirmed heterogeneity within individual beels. We propose L/B quotient based on Lecane: Brachionus species ratios to characterize habitat variations of the sampled wetlands. Sládeček's B/T quotient based on Brachionus: Trichocerca species ratios affirmed general' meso-trophic' status of different beels. Our results provided little insight on the influence of individual abiotic factors but the canonical correspondence analyses asserted higher cumulative influence of ten abiotic parameters on Rotifera richness in each beel.

  5. Psammon rotifers in Central Vietnam, with the descriptions of three new species (Rotifera: Monogononta).

    In order to address the dearth of information on Rotifera in Vietnam, and especially on rotifers inhabiting freshwater psammon in Southeast Asia, we collected and examined twenty-five hygropsammon samples from Bau Thiem Lake, Thua Thien Hue Province, central Vietnam. A total of eighty-nine species-level rotifer taxa were identified, belonging to 21 genera and 13 families. Of these, 48 taxa (54%) are new to Vietnam, including three species, Lecane climacois Harring & Myers, 1926, Notommata cerberus (Gosse, 1886) and Trichocerca intermedia (Stenroos, 1898) that are new to the Oriental region, and three species, Lecane phapi n. sp., Lecane dorysimilis n. sp. and Trichocerca bauthiemensis n. sp. that are new to science. These, and some additional rare species are commented upon and illustrated. We estimated that the α-diversity of psammon rotifers in Bau Thiem Lake may be as high as 99 (SD=8.4; Chao2) or 115 (Jacknife2) taxa. These results indicate a high potential of rotifer diversity in the hygropsammon at Bau Thiem Lake, central Vietnam.

  6. Modeling species richness and abundance of phytoplankton and zooplankton in radioactively contaminated water bodies.

    Water bodies polluted by the Mayak nuclear plant in Russia provide valuable information on multi-generation effects of radioactive contamination on freshwater organisms. For example, lake Karachay was probably the most radioactive lake in the world: its water contained ∼2 × 10 7 Bq/L of radionuclides and estimated dose rates to plankton exceeded 5 Gy/h. We performed quantitative modeling of radiation effects on phytoplankton and zooplankton species richness and abundance in Mayak-contaminated water bodies. Due to collinearity between radioactive contamination, water body size and salinity, we combined these variables into one (called HabitatFactors). We employed a customized machine learning approach, where synthetic noise variables acted as benchmarks of predictor performance. HabitatFactors was the only predictor that outperformed noise variables and, therefore, we used it for parametric modeling of plankton responses. Best-fit model predictions suggested 50% species richness reduction at HabitatFactors values corresponding to dose rates of 10 4 -10 5  μGy/h for phytoplankton, and 10 3 -10 4  μGy/h for zooplankton. Under conditions similar to those in lake Karachay, best-fit models predicted 81-98% species richness reductions for various taxa (Cyanobacteria, Bacillariophyta, Chlorophyta, Rotifera, Cladocera and Copepoda), ∼20-300-fold abundance reduction for total zooplankton, but no abundance reduction for phytoplankton. Rotifera was the only taxon whose fractional abundance increased with contamination level, reaching 100% in lake Karachay, but Rotifera species richness declined with contamination level, as in other taxa. Under severe radioactive and chemical contamination, one species of Cyanobacteria (Geitlerinema amphibium) dominated phytoplankton, and rotifers from the genus Brachionus dominated zooplankton. The modeling approaches proposed here are applicable to other radioecological data sets. The results provide quantitative information and easily interpretable model parameter estimates for the shapes and magnitudes of freshwater plankton responses to a wide range of radioactive contamination levels.   7. Speciation in ancient cryptic species complexes: evidence from the molecular phylogeny of Brachionus plicatilis (Rotifera).

    Continental lake-dwelling zooplanktonic organisms have long been considered cosmopolitan species with little geographic variation in spite of the isolation of their habitats. Evidence of morphological cohesiveness and high dispersal capabilities support this interpretation. However, this view has been challenged recently as many such species have been shown either to comprise cryptic species complexes or to exhibit marked population genetic differentiation and strong phylogeographic structuring at a regional scale. Here we investigate the molecular phylogeny of the cosmopolitan passively dispersing rotifer Brachionus plicatilis (Rotifera: Monogononta) species complex using nucleotide sequence variation from both nuclear (ribosomal internal transcribed spacer 1, ITS1) and mitochondrial (cytochrome c oxidase subunit I, COI) genes. Analysis of rotifer resting eggs from 27 salt lakes in the Iberian Peninsula plus lakes from four continents revealed nine genetically divergent lineages. The high level of sequence divergence, absence of hybridization, and extensive sympatry observed support the specific status of these lineages. Sequence divergence estimates indicate that the B. plicatilis complex began diversifying many millions of years ago, yet has showed relatively high levels of morphological stasis. We discuss these results in relation to the ecology and genetics of aquatic invertebrates possessing dispersive resting propagules and address the apparent contradiction between zooplanktonic population structure and their morphological stasis.

    The taxon Syndermata comprises the biologically interesting wheel animals ("Rotifera": Bdelloidea + Monogononta + Seisonidea) and thorny-headed worms (Acanthocephala), and is central for testing superordinate phylogenetic hypotheses (Platyzoa, Gnathifera) in the metazoan tree of life. Recent analyses of syndermatan phylogeny suggested paraphyly of Eurotatoria (free-living bdelloids and monogononts) with respect to endoparasitic acanthocephalans. Data of epizoic seisonids, however, were absent, which may have affected the branching order within the syndermatan clade. Moreover, the position of Seisonidea within Syndermata should help in understanding the evolution of acanthocephalan endoparasitism. Here, we report the first phylogenomic analysis that includes all four higher-ranked groups of Syndermata. The analyzed data sets comprise new transcriptome data for Seison spec. (Seisonidea), Brachionus manjavacas (Monogononta), Adineta vaga (Bdelloidea), and Paratenuisentis ambiguus (Acanthocephala). Maximum likelihood and Bayesian trees for a total of 19 metazoan species were reconstructed from up to 410 functionally diverse proteins. The results unanimously place Monogononta basally within Syndermata, and Bdelloidea appear as the sister group to a clade comprising epizoic Seisonidea and endoparasitic Acanthocephala. Our results support monophyly of Syndermata, Hemirotifera (Bdelloidea + Seisonidea + Acanthocephala), and Pararotatoria (Seisonidea + Acanthocephala), rejecting monophyly of traditional Rotifera and Eurotatoria. This serves as an indication that early acanthocephalans lived epizoically or as ectoparasites on arthropods, before their complex lifecycle with arthropod intermediate and vertebrate definite hosts evolved.

  9. A decade of predatory control of zooplankton species composition of Lake Michigan

    From 1983 to 1992, 71 species representing 38 genera from the Calanoida, Cladocera, Cyclopoida, Mysidacea, Rotifera, Mollusca and Harpacticoida comprised the offshore zooplankton community of Lake Michigan. Our data demonstrate that the composition and abundance of the calanoid community after 1983 is not unlike that of 1960s and that species diversity of the calanoid community is more diverse than the cladoceran community in the 1990s as compared to the early 1980s. Even though the relative biomass of the cladocerans has remained similar over the 1983-1993 period, the species diversity and evenness of the Cladocera community in the early 1990s is unlike anything that has been previously reported for Lake Michigan. Cladocera dominance is centered in one species, Daphnia galeata mendotae, and only three species of Cladocera were observed in the pelagic region of the lake in 1991 and 1992. Nutrient levels, phytoplankton biomass, and the abundance of planktivorous alewife and bloater chub and Bythotrephes are examined as possible causes of these changes in zooplankton species composition. The increase in Rotifera biomass, but not Crustacea, was correlated with an increase in relative biomass of unicellular algae. Food web models suggest Bythotrephes will cause Lake Michigan's plankton to return to a community similar to that of the 1970s; that is Diaptomus dominated. Such a change has occurred. However, correlational analysis suggest that alewife and bloater chubs (especially juveniles) are affecting size and biomass of larger species of zooplankton as well as Bythotrephes.

  10. Postembryonic development of hard jaws (trophi) in a species belonging to the Brachionus plicatilis complex (Rotifera, Monogononta): a morphometric analysis.

    The presence of hard jaws (trophi), with species-specific shape and size, is a taxonomic feature of Rotifera, a group of microscopic metazoans. Since trophi are used to discriminate among species, it is important to know whether these structures change in taxonomically important ways during postembryonic development. Using both SEM and optical images, we analyzed more than 100 individuals of a single clonal lineage of a monogonont rotifer, Brachionus plicatilis, in order to describe body and trophi development after hatching. Body size, expressed as lorica width and length, was isometrically related to age of the animals only during preadult stages. Trophi size, expressed as length of the different parts, was unrelated to either age or body size. Therefore, trophi elements do not grow after hatching in B. plicatilis. Despite the dimensional invariance with age, some differences in trophi size among individuals of the same clone were recorded. No difference in left-right asymmetry of the trophi was shown; the asymmetric elements of the trophi named rami consistently had the right ramus longer than the left. This constancy is in contrast to the reported trophi asymmetries in bdelloid rotifers, in which left-right asymmetries are not constant within clonal lineages. In conclusion, we suggest that also trophi size, constant within the analyzed clone, may be used as an additional taxonomic feature to help in the discrimination of taxa within the B. plicatilis complex of cryptic species.     The Aschelminthes is a collection of at least eight animal phyla, historically grouped together because the absence of a true body cavity was perceived as a pseudocoelom. Analyses of 18S rRNA sequences from six Aschelminth phyla (including four previously unpublished sequences) support polyphyly for the Aschelminthes. At least three distinct groups of Aschelminthes were detected: the Priapulida among the protostomes, the Rotifera-Acanthocephala as a sister group to the protostomes, and the Nematoda as a basal group to the triploblastic Eumetazoa.

  12. Sensitivity of plankton indices to lake trophic conditions.

    Herein, we report the response of indices based on phytoplankton and zooplankton and their combination to different nutrient concentrations in lakes. The study was carried out in ten lakes in northeastern Poland. Integrated samples were collected from the epilimnion during the summer of 2012-2013. Secchi disk visibility (SD), total phosphorus (TP), total nitrogen (TN), and chlorophyll a were used as proxies for eutrophication. We calculated 16 plankton indices: two phytoplankton indices, six crustacean indices, five rotiferan indices, two zooplankton diversity indices, and one combined phytoplankton and zooplankton index. Among them, nine indices with the strongest correlations with TP were selected: percentage share of Crustacean species indicative of high trophy in the indicative group's numbers (IHT CRU ), percentage share of Rotifera species indicative of high trophy in the indicative group's numbers IHT ROT , Crustacean ratio of biomass to numbers B/N CRU , phytoplankton trophic index (TI TP+TN ), Margalef's index, percentage share of cyclopoid biomass in total biomass of Crustacea (CB), Rotifera numbers (N ROT ), biomass of Cyclopoida (B CY ), and ratio of the cyclopoid biomass to the biomass of Cladocera (CY/CL). The sensitivity of the normalized values of these indices to proxies of eutrophication was tested. IHT CRU , IHT ROT , and B/N CRU were the most sensitive and gave the strongest responses at lower TP concentrations (<35 μg/L). The phytoplankton trophic index, TI TP+TN , together with the zooplankton-based Margalef's index and CB were very sensitive in both low (<35 μg/L) and high (>60 μg/L) TP conditions. On the other hand, N ROT , B CY , and CY/CL were slightly sensitive at low TP concentrations while their reaction was notable at high TP concentrations. A similar pattern of response was observed for TN concentration and SD visibility.

    Animal cells are protected from oxidative damage by an antioxidant network operating as a coordinated system, with strong synergistic interactions. Lifespan studies with whole animals are expensive and laborious, so there has been little investigation of which antioxidant interactions might be useful for life extension. Animals in the phylum Rotifera are particularly promising models for aging studies because they are small (0.1–1 mm), have short, two-week lifespan, display typical patterns of animal aging, and have well characterized, easy to measure phenotypes of aging and senescence. One class of interventions that has consistently produced significant rotifer life extension is antioxidants. Although the mechanism of antioxidant effects on animal aging remains controversial, the ability of some antioxidant supplements to extend rotifer lifespan was unequivocal. We found that exposing rotifers to certain combinations of antioxidant supplements can produce up to about 20% longer lifespan, but that most antioxidants have no effect. We performed life table tests with 20 single antioxidants and none yielded significant rotifer life extension. We tested 60 two-way combinations of selected antioxidants and only seven (12%) produced significant rotifer life extension. None of the 20 three- and four-way antioxidant combinations tested yielded significant rotifer life extension. These observations suggest that dietary exposure of antioxidants can extend rotifer lifespan, but most antioxidants do not. We observed significant rotifer life extension only when antioxidants were paired with trolox, N-acetyl cysteine, l-carnosine, or EUK-8. This illustrates that antioxidant treatments capable of rotifer life extension are patchily distributed in the parameter space, so large regions must be searched to find them. It furthermore underscores the value of the rotifer model to conduct rapid, facile life table experiments with many treatments, which makes such a search feasible.     In order to broaden the comparative scope of evolutionary developmental biology and to refine our picture of animal macroevolution, it is necessary to establish new model organisms, especially from previously underrepresented groups, like the Lophotrochozoa. We have established the culture and protocols for molecular developmental biology in the rotifer species Brachionus plicatilis Müller (Rotifera, Monogononta). Rotifers are nonsegmented animals with enigmatic basal position within the lophotrochozoans and marked by several evolutionary novelties like the wheel organ (corona), the median eye, and the nonpaired posterior foot. The expression of Bp-Pax-6 is shown using whole-mount in situ hybridization. The inexpensive easy culture and experimental tractability of Brachionus as well as the range of interesting questions to which it holds the key make it a promising addition to the "zoo" of evo-devo model organisms.

  16. Ecological factors affecting gene flow in the Brachionus plicatilis complex (Rotifera).

    We investigated how adaptation to salinity and temperature acts as reproductive barriers in three sympatric species from the Brachionus plicatilis species complex. These species co-occur in a salt marsh in Spain, and a previous electrophoretic study of variation revealed no hybrids between them. A factorial experiment was designed to test for differences in population growth rates and patterns of bisexual reproduction. The design combined representative strains from each species in different salinity and temperature conditions, representing the range over which these rotifers are found in their natural environment. We found differences in the growth response of the three species to both factors and in the pattern of bisexual reproduction. These differences help to explain patterns of succession observed in the field. We conclude that these ecological factors, together with mate recognition systems, account for the absence of gene flow in these sympatric species.

The enzyme glutathione S-transferase (GST), a critical element in xenobiotic metabolism, was isolated from the marine rotifer Brachionus plicatilis and its freshwater congener B. calyciflorus. 2. In B. plicatilis, GST comprised 4.2% of cytosolic protein and was present as three separate isozymes with mol. wts 30,000, 31,400 and 33,700. Specific activity of crude homogenates was 56 nmol min-1 mg-1 protein, while that of affinity chromatography purified GST was 1850. 3. In B. calyciflorus, GST was present as two isozymes with mol. wts of 26,300 and 28,500, representing 1.0% of cytosolic protein. Crude GST specific activity was 1750 nmol min-1 mg-1 protein and purified was 72,400. 4. Rotifer GSTs are unusual because they are monomers whereas all other animals thus far investigated posses dimeric GSTs.

    The complete mitochondrial genome was obtained from the assembled genome data sequenced by next generation sequencing (NGS) technology from the monogonont rotifer Brachionus koreanus. The mitochondrial genome of B. koreanus was composed of two circular chromosomes designated as mtDNA-I (10,421 bp) and mtDNA-II (11,923 bp). The gene contents of B. koreanus were identical with previously reported B. plicatilis mitochondrial genomes. However, gene orders of B. koreanus showed one rearrangement between the two species. Of 12 protein-coding genes (PCGs), 3 genes (ATP6, ND1, and ND3) had an incomplete stop codon. The A + T base composition of B. koreanus mitochondrial genome was high (68.81%). They also showed anti-G bias (12.03% and 10.97%) on the second and third position of PCGs as well as slight anti-C bias (15.96% and 14.31%) on the first and third position of PCGs.

  1. Phylogeny of the Acanthocephala based on morphological characters.

    Only four previous studies of relationships among acanthocephalans have included cladistic analyses, and knowledge of the phylogeny of the group has not kept pace with that of other taxa. The purpose of this study is to provide a more comprehensive analysis of the phylogenetic relationships among members of the phylum Acanthocephala using morphological characters. The most appropriate outgroups are those that share a common early cell-cleavage pattern (polar placement of centrioles), such as the Rotifera, rather than the Priapulida (meridional placement of centrioles) to provide character polarity based on common ancestry rather than a general similarity likely due to convergence of body shapes. The phylogeny of 22 species of the Acanthocephala was evaluated based on 138 binary and multistate characters derived from comparative morphological and ontogenetic studies. Three assumptions of cement gland structure were tested: (i) the plesiomorphic type of cement glands in the Rotifera, as the sister group, is undetermined; (ii) non-syncytial cement glands are plesiomorphic; and (iii) syncytial cement glands are plesiomorphic. The results were used to test an early move of Tegorhynchus pectinarius to Koronacantha and to evaluate the relationship between Tegorhynchus and Illiosentis. Analysis of the data-set for each of these assumptions of cement gland structure produced the same single most parsimonious tree topology. Using Assumptions i and ii for the cement glands, the trees were the same length (length = 404 steps, CI = 0.545, CIX = 0.517, HI = 0.455, HIX = 0.483, RI = 0.670, RC = 0.365). Using Assumption iii, the tree was three steps longer (length = 408 steps, CI = 0.539, CIX = 0.512, HI = 0.461, HIX = 0.488, RI = 0.665, RC = 0.359). The tree indicates that the Palaeacanthocephala and Eoacanthocephala both are monophyletic and are sister taxa. The members of the Archiacanthocephala are basal to the other two clades, but do not themselves form a clade. The results provide strong support for the Palaeacanthocephala and the Eoacanthocephala and the hypothesis that the Eoacanthocephala is the most primitive group is not supported. Little support for the Archiacanthocephala as a monophyletic group was provided by the analysis. Support is provided for the recognition of Tegorhynchus and Illiosentis as distinct taxa, as well as the transfer of T. pectinarius to Koronacantha.

    In order to broaden the comparative scope of evolutionary developmental biology and to refine our picture of animal macroevolution, it is necessary to establish new model organisms, especially from previously underrepresented groups, like the Lophotrochozoa. We have established the culture and protocols for molecular developmental biology in the rotifer species Brachionus plicatilis Müller (Rotifera, Monogononta). Rotifers are nonsegmented animals with enigmatic basal position within the lophotrochozoans and marked by several evolutionary novelties like the wheel organ (corona), the median eye, and the nonpaired posterior foot. The expression of Bp-Pax-6 is shown using whole-mount in situ hybridization. The inexpensive easy culture and experimental tractability of Brachionus as well as the range of interesting questions to which it holds the key make it a promising addition to the “zoo” of evo-devo model organisms.     1. Tests for glycosidases were performed in homogenates of Brachionus plicatilis. 2. Hydrolytic activity was detected with the following substrates: (a) with synthetic substrates (NP = 4-nitrophenyl): NP-alpha- and NP-beta-D-glucopyranoside, NP-alpha- and NP-beta-D-galactopyranoside, NP-N-acetyl-beta-D-glucosaminide, NP-N-acetyl-beta-D-galactosaminide, NP-alpha- and NP-beta-D-mannopyranoside and NP-alpha-L-fucopyranoside; (b) with disaccharides: sucrose, maltose, trehalose, isomaltose, cellobiose, gentiobiose and lactose; (c) with polysaccharides: laminarine, carboxymethyl-cellulose, avicel, Micrococcus luteus (for lysozyme) and 4-nitrophenyl-alpha-D-maltoheptaoside (for amylase). 3. The pH dependence of the glycosidase activities was determined. 4. The distribution of enzyme activities within fractions from the homogenate was studied in order to localize them within the cell. 5.     Zooplankton was successfully used for the biological control of unicellular algal contaminants in Spirulina mass cultures even under conditions adverse to the growth of Spirulina (maximal winter daily temperature of approximately 10 degrees C and very low bicarbonate concentration). Brachionus plicatilis (Rotifera) was the most successful species of zooplankton used. The interrelationships between Spirulina, green unicellular contaminant, and B. plicatilis were studied under various conditions. Two species of unicellular contaminant were used; Monoraphidium minutum was isolated from local cultures and Chlorella vulgaris, obtained from contaminated Spirulina cultures in Israel. The rotifer B. plicatilis successfully controlled the population size of bothmore » contaminants whether they were introduced in a single addition or as a daily dose. The biological control of the unicellular contaminants allows Spirulina to be cultured in a medium low in bicarbonate, thereby reducing the cost of the medium and increasing the quantity of CO2 that may be freely absorbed from the atmosphere at the optimal pH for Spirulina cultivation.     To survive exposure to space conditions, organisms should have certain characteristics including a high tolerance for freezing, radiation and desiccation. The organisms with the best chance for survival under such conditions are extremophiles, like some species of Bacteria and Archea, Rotifera, several species of Nematoda, some of the arthropods and Tardigrada (water bears). There is no denying that tardigrades are one of the toughest animals on our planet and are the most unique in the extremophiles group. Tardigrada are very small animals (50 to 2,100 μm in length), and they inhabit great number of Earth environments. Ever since it was proven that tardigrades have high resistance to the different kinds of stress factors associated with cosmic journeys, combined with their relatively complex structure and their relative ease of observation, they have become a perfect model organism for space research. This taxon is now the focus of astrobiologists from around the world. Therefore, this paper presents a short review of the space research performed on tardigrades as well as some considerations for further studies.

  6. Evaluation of abyssal meiobenthos in the eastern central Pacific (Clarion-Clipperton fracture zone)

    Meiobenthos were sampled from 17 stations in the abyssal deep-sea system of the central Pacific centered around 14°N, 130°W at depths 4960-5154m, during the Nixo 47 R/V Jean Charcot cruise. Meiofaunal density range from 45-89 ind. 10cm 2. Predominant taxa are nematodes (84-100%) and copepods (0-10%). Rotifera, Polychaeta, and Acarina also occur. Nematodes are uniformly distributed spatially with 45 species or so; Monhysteridae is the dominant taxon, and Syringolaimus sp. (Ironidae) co-occurs faithfully. Low biomass (0.4-70.6μg 10cm 2) are attributed to supposed dwarfism of metazoan meiofauna and very high proportion (60-80%) of juveniles and pre-adult forms. The majority of protozoans and metazoans are detritus- or deposit-feeders; in addition symbiotic associations, coprophagy and gardening activities are frequent. In such an oligotrophic environment, low food supply may limit meiofaunal abundance, biomass and maturation, and to a lesser extent species richness.

    In western Lake Erie in the summer and fall of 1975–1976, food eaten by seven forage fishes—emerald shiner (Notropis atherinoides), spottail shiner (Notropis hudsonius), trout-perch (Percopsis omiscomaycus), andyoung-of-the-year (YOY) of alewife (Alosa pseudoharengus), gizzard shad (Dorosoma cepedianum), white bass (Morone chrysops), and freshwater drum (Aplodi-notus grunniens)—was divided among six major taxa: Cladocera, Copepoda, Diptera, Ostracoda, Amphipoda, and Algae. In addition, fish were eaten by YOY white bass, and Rotifera were consumed by YOY gizzard shad. Interspecies diet overlap indices, calculated to compare the food of the different species and to evaluate diet similarities, were usually highest for YOY white bass and YOY freshwater drum when compared with the other species and usually lowest between emerald shiners and all other forage fishes. Understanding the feeding interactions among fishes that could influence production at the forage-food level of the food web could provide insight into how cascading trophic interactions influence the production of piscivorous predators.

  9. Synergistic toxicity of Macondo crude oil and dispersant Corexit 9500A(®) to the Brachionus plicatilis species complex (Rotifera).

    Using the marine rotifer Brachionus plicatilis acute toxicity tests, we estimated the toxicity of Corexit 9500A(®), propylene glycol, and Macondo oil. Ratios of 1:10, 1:50 and 1:130 for Corexit 9500A(®):Macondo oil mixture represent: maximum exposure concentrations, recommended ratios for deploying Corexit (1:10-1:50), 1:130 the actual dispersant:oil ratio used in the Deep Water Horizon spill. Corexit 9500A(®) and oil are similar in their toxicity. However, when Corexit 9500A(®) and oil are mixed, toxicity to B. manjavacas increases up to 52-fold. Extrapolating these results to the oil released by the Macondo well, suggests underestimation of increased toxicity from Corexit application. We found small differences in sensitivity among species of the B. plicatilis species complex, likely reflecting phylogenetic similarity. Just 2.6% of the water-accommodated fraction of oil inhibited rotifer cyst hatching by 50%, an ecologically significant result because rotifer cyst in sediments are critical resources for the recolonization of populations each Spring.     We measured life span and fecundity of three reproductive modes in a clone of the monogonont rotifer Brachionus manjavacas subjected to chronic caloric restriction (CCR) over a range of food concentrations or to intermittent fasting (IF). IF increased life span 50%–70% for all three modes, whereas CCR increased life span of asexual females derived from sexually or asexually produced eggs, but not that of sexual females. The main effect of CR on both asexual modes was to delay death at young ages, rather than to prevent death at middle ages or to greatly extend maximum life span; in contrast CR in sexual females greatly increased the life span of a few long-lived individuals. Lifetime fecundity did not decrease with CCR, suggesting a lack of resource allocation trade-off between somatic maintenance and reproduction. Multiple outcomes for a clonal lineage indicate that different responses are established through epigenetic programming, whereas differences in life-span allocations suggest that multiple genetic mechanisms mediate life-span extension.     In recent years, the keeping of ornamental freshwater animals and plants in garden ponds has been growing in popularity. Water hyacinth (Eichhornia crassipes) is one of the preferred macrophytes seasonally imported mainly from South-eastern Asia throughout the world. This constitutes a secondary introduction inasmuch as the species is native to South America. Although many assemblages of aquatic invertebrates have been described as associated with this plant in the wild, there has been no research focused on their potential introduction via the international plant trade. We examined 216 specimens of water hyacinths imported for ornamental purposes from Indonesia into the Czech Republic. Numerous meio- and macroinvertebrates belonging to at least 39 species were captured. On the total number of individuals, the highest prevalence was of Tubulinea and Rotifera. Most of these were still alive and vital, including a caterpillar of the Indo-Australian invasive moth Spodoptera litura. Water hyacinths are usually placed into outdoor ponds immediately after import, which facilitates the release of non-target alien species. The present paper aims to draw attention to "hitchhikers" associated with the ornamental trade.

    The potential impact of environmental change on terrestrial Antarctic ecosystems can be explored by inspecting biodiversity patterns across large-scale gradients. Unfortunately, morphology-based surveys of Antarctic invertebrates are time-consuming and limited by the cryptic nature of many taxa. We used biodiversity information derived from high-throughput sequencing (HTS) to elucidate the relationship between soil properties and invertebrate biodiversity in the Prince Charles Mountains, East Antarctica. Across 136 analysed soil samples collected from Mount Menzies, Mawson Escarpment and Lake Terrasovoje, we found invertebrate distribution in the Prince Charles Mountains significantly influenced by soil salinity and/or sulfur content. Phyla Tardigrada and Arachnida occurred predominantly in low-salinity substrates with abundant nutrients, whereas Bdelloidea (Rotifera) and Chromadorea (Nematoda) were more common in highly saline substrates. A significant correlation between invertebrate occurrence, soil salinity and time since deglaciation indicates that terrain age indirectly influences Antarctic terrestrial biodiversity, with more recently deglaciated areas supporting greater diversity. Our study demonstrates the value of HTS metabarcoding to investigate environmental constraints on inconspicuous soil biodiversity across large spatial scales.

  15. The influence of physical and chemical parameters on the extinction of the god fish (Tor duoronensis) species: case study in Balong Dalem Kuningan district

    Tor douronensis (God Fish) species are endemic freshwater fish that only live in Kuningan District. Originally these fish scattered in the pool Cigugur, Cibulan, Balong Kambang, Darma Loka and Balong Dalem. However, since 2006 the god fish that existed in Balong Dalem extinct. This study aims to analyze the possible factors causing extinction of the god fish in situ Balong Dalem. The study was conducted during May 2017. The results showed that there were 11 phytoplankton and 13 genera of Gomphonema, Stigeoclonium, Desmidium, Closterium, Navicula, Spirulina, Phormidium, Melosira, Gloeotrichia, Oedogonium, Hyalodiscus, Moscocrocis and Cladophora. While zooplankton found 3 genus of Cyclops, Rotifera and Tanypus. The results showed that the abundance, diversity and uniformity of zooplankton correlated positively with pH, temperature, DO and water flow but not with others. Condition of water quality (physical-chemical parameter) Balong Dalem in bad condition, low productivity and labile. Condition of substrate base there Balong Dalem in the form of sand and fine mud. All of these conditions caused the extinction of Tor douronensis in situ Balong Dalem.

    Fish farming in net cages causes changes in environmental conditions. We evaluated the resilience of zooplankton concerning this activity in Rosana Reservoir (Paranapanema River, PR-SP). Samples were taken near the net cages installed at distances upstream and downstream, before and after net cage installation. The resilience was estimated by the decrease in the groups' abundance after installing the net cages. The zooplankton community was represented by 106 species. The most abundant species were Synchaeta pectinata, S. oblonga, Conochilus coenobasis, Polyarthra dolichoptera and C. unicornis (Rotifera), Ceriodaphnia cornuta, Moina minuta, Bosmina hagmanni and C. silvestrii (Cladocera) and Notodiaptomus amazonicus (Copepoda). The resilience of microcrustaceans was affected in the growing points as this activity left the production environment for longer, delaying the natural ability of community responses. Microcrustaceans groups, mainly calanoid and cyclopoid copepods, had a different return rate. The net cage installation acted as a stress factor on the zooplankton community. Management strategies that cause fewer risks to the organisms and maximize energy flow may help in maintaining system stability.

  18. Effect of periodical starvation on the life history of Brachionus plicatilis O.F. Müller (Rotifera): a possible strategy for population stability.

    To estimate the changes in the life history of the rotifer Brachionus plicatilis O.F. Müller under starvation, we carried out an individual culture and determined the effects of periodical food deprivation on its asexual reproductive characteristics such as lifespan, reproductive period, age at first egg and offspring production, and lifetime fecundity (total number of offspring produced in her lifetime). Rotifers were fed for 1-3 h daily, and were then starved until the next day. Control animals were fed throughout their lifespan. Starved rotifers matured and produced their first offspring at an older age than the control animals. The periodical starvation resulted in a decrease in the lifetime fecundity to less than half that of the non-starved control. The reproductive period and lifespan were 2-3 times longer in the starved animals than in the control animals. The negative relationship between lifespan and lifetime fecundity is interpreted as a trade-off in an alternative life-history strategy of rotifers under starved conditions. The great decrease in fecundity and extension of lifespan enables rotifers to compensate to keep the population in equilibrium.

  19. Morphological alterations in the freshwater rotifer Brachionus calyciflorus Pallas 1766 (Rotifera: Monogononta) caused by vinclozolin chronic exposure.

    Vinclozolin (VZ) is a dicarboximide fungicide widely used on fruits, vegetables and wines, effective against fungi plagues. In this study we characterized the effects of VZ using a 4-day reproductive chronic assay with the freshwater rotifer Brachionus calyciflorus. The assay included observations of several features of asexual and sexual reproduction. Our results indicate that VZ: (a) increased asexual and sexual reproduction, (b) caused severe abnormality in females and (c) these abnormalities were inherited by sexual and asexual reproduction. At 1.2 mg/L three abnormal females were found out of 457 total females (0.66 %). This low percentage is consistent and reproducible according to further analysis, where we increased the number of replicates and total females exposed to 1.2 mg/L of VZ, and found 18 abnormal females out of 2868 total females (0.63 % abnormality). Interestingly, abnormal females found at 5.6 mg/L VZ exposure, were able to show mating behavior. Our results suggest that VZ behaves as a strong endocrine disruptor whose effects show the characteristic inverted-U-shape exposure concentration response curve regarding the intrinsic population increase and the percentage of abnormalities as endpoints.

    Glaciers and ice sheets are a peculiar biome with characteristic abiotic and biotic components. Mountain glaciers are predicted to decrease their volume and even to melt away within a few decades. Despite the threat of a disappearing biome, the diversity and the role of microscopic animals as consumers at higher trophic levels in the glacial biome still remain largely unknown. In this study, we report data on tardigrades and rotifers found in glacial mosses on Mount Stanley, Uganda, and describe a new tardigrade species. Adropion afroglacialis sp. nov. differs from the most similar species by having granulation on the cuticle, absence of cuticular bars under the claws, and a different macroplacoid length sequence. We also provide a morphological diagnosis for another unknown tardigrade species of the genus Hypsibius. The rotifers belonged to the families Philodinidae and Habrotrochidae. In addition, we discuss the diversity of microinvertebrates and potential role of tardigrades and rotifers on mountain glaciers as top consumers. As for any organism living apparently exclusively in glacial habitats on tropical glaciers, their extinction in the near future is inevitable, possibly before we can even discover their existence.

    Due to the importance of water variables conditions and available food in the development and survival of fish larvae, the current research evaluates the effects of two different food treatments (ration + zooplankton and only zooplankton) and water quality in tanks with Brycon orbignyanus larvae. Total water transparency (45 cm) has been mainly associated with short residence time, continuous water flow and shallowness. Dissolved oxygen ranged between 1.32 and 7.00 mg.L(-1) in tanks with ration + zooplankton and between 1.82 and 7.60 mg.L(-1) in tanks with only zooplankton treatments. Nutrients were directly affected by the addition of ration in water, with the exception of nitrite. Ten Rotifera species were found represented by high densities, ranging between 8.7 x 10(5) and 1.3 x 10(6) org.m(-3), throughout the experimental period (January to March/1996). Cladocera had the lowest density in the four tanks under analysis and ranged between 4.7 x 10(4) and 2.1 x 10(5) org.m(-3) for the six species. Diaphanosoma birgei has been classified as the most frequent species. Since ration + zooplankton produced better larvae yield, this treatment is recommended for Brycon orbignyanus larvae.

    The distribution and diet of larval and juvenile Arctic cod ( Boreogadus saida) were studied during summer 2005 in the coastal Canadian Beaufort Sea. A total of 275 individuals were captured and the highest abundance was observed at station depths of 20-30 m. This corresponds well with the location of the frontal zone where the Mackenzie River plume water and open sea water meet. Diet examinations were performed on 220 Arctic cod, which were found undamaged from sampling. We observed a gradual decrease in prey number per fish and increase in prey size as larvae grew which corresponded to a shift from Rotifera and nauplii towards larger copepodid stages. However, at all sizes, the larvae remain generalists and feed on a broad range of organisms. Environmental changes due to climate warming could have a two-fold impact on fish larvae feeding in the studied region. First, the potential for increased primary production may lead to increased zooplankton production that may impact the feeding and nutrition positively. On the other hand, greater discharge of turbid water from the Mackenzie River may reduce light penetration in the water column that may negatively influence the ability of visual predators to successively forage.

  3. Plankton community and the relationship with the environment in saline lakes of Onon-Torey plain, Northeastern Mongolia.

    The plankton community of sixteen saline lakes located on Onon-Torey plain (Northeastern Mongolia) during the filling phase and the raising of the water level was investigated in July 2011. Thirty-five taxa of phytoplankton and thirty-one species of zooplankton were found. For phytoplankton, blue-green algae ( Merismopedia elegans , Anabaenopsis elenkinii , Arthrospora fusiformis , Spirulina major , Lyngbya sp., Oscillatoria sp.) and green algae ( Monoraphidium minutum , Tetrastrum komarekii , Ankyra ocellata , Oocystis sp.) were dominant. For zooplankton, Filinia longiseta, Brachionus plicatilis , B. variabilis , Hexarthra mira (Rotifera), Daphnia magna , Moina brachiata , M. mongolica (Cladocera), Arctodiaptomus bacillifer , Mixodiaptomus incrassatus , Metadiaptomus asiaticus (Copepoda) dominated. Mineralization, active hydrogen ratio, dissolved oxygen and water temperature were the main factors influencing the diversity, structure and distribution of plankton organisms in the steppe lakes during low water level. The RDA analysis for phytoplankton and zooplankton from different lakes was carried out for selected two groups which included lakes and a subset related species. The first group is of oligohaline and mesohaline lakes in which mostly green algae, rotifers and copepods inhabit. The second group is of mesohaline and polyhaline lakes with mainly blue-green algae , some crustaceans and rotifers inhabiting. High abundance and biomass of Spirulina major , Oscillatoria sp. and Brachionus variabilis were observed in lakes with high mineralization, pH and temperature.

    The bacterial and microeukaryotic biodiversity were studied using pyrosequencing analysis on a 454 GS FLX+ platform of partial SSU rRNA genes in terrestrial and aquatic habitats of the Sør Rondane Mountains, including soils, on mosses, endolithic communities, cryoconite holes and supraglacial and subglacial meltwater lenses. This inventory was complemented with Denaturing Gradient Gel Electrophoresis targeting Chlorophyta and Cyanobacteria. OTUs belonging to the Rotifera, Chlorophyta, Tardigrada, Ciliophora, Cercozoa, Fungi, Bryophyta, Bacillariophyta, Collembola and Nematoda were present with a relative abundance of at least 0.1% in the eukaryotic communities. Cyanobacteria, Proteobacteria, Bacteroidetes, Acidobacteria, FBP and Actinobacteria were the most abundant bacterial phyla. Multivariate analyses of the pyrosequencing data revealed a general lack of differentiation of both eukaryotes and prokaryotes according to habitat type. However, the bacterial community structure in the aquatic habitats was dominated by the filamentous cyanobacteria Leptolyngbya and appeared to be significantly different compared with those in dry soils, on mosses, and in endolithic habitats. A striking feature in all datasets was the detection of a relatively large amount of sequences new to science, which underscores the need for additional biodiversity assessments in Antarctic inland locations. © FEMS 2016.   5. Divergent gene copies in the asexual class Bdelloidea (Rotifera) separated before the bdelloid radiation or within bdelloid families.

    Rotifers of the asexual class Bdelloidea are unusual in possessing two or more divergent copies of every gene that has been examined. Phylogenetic analysis of the heat-shock gene hsp82 and the TATA-box-binding protein gene tbp in multiple bdelloid species suggested that for each gene, each copy belonged to one of two lineages that began to diverge before the bdelloid radiation. Such gene trees are consistent with the two lineages having descended from former alleles that began to diverge after meiotic segregation ceased or from subgenomes of an alloploid ancestor of the bdelloids. However, the original analyses of bdelloid gene-copy divergence used only a single outgroup species and were based on parsimony and neighbor joining. We have now used maximum likelihood and Bayesian inference methods and, for hsp82, multiple outgroups in an attempt to produce more robust gene trees. Here we report that the available data do not unambiguously discriminate between gene trees that root the origin of hsp82 and tbp copy divergence before the bdelloid radiation and those which indicate that the gene copies began to diverge within bdelloid families. The remarkable presence of multiple diverged gene copies in individual genomes is nevertheless consistent with the loss of sex in an ancient ancestor of bdelloids.

  6. Transcriptome Data Reveal Syndermatan Relationships and Suggest the Evolution of Endoparasitism in Acanthocephala via an Epizoic Stage

    The taxon Syndermata comprises the biologically interesting wheel animals (“Rotifera”: Bdelloidea + Monogononta + Seisonidea) and thorny-headed worms (Acanthocephala), and is central for testing superordinate phylogenetic hypotheses (Platyzoa, Gnathifera) in the metazoan tree of life. Recent analyses of syndermatan phylogeny suggested paraphyly of Eurotatoria (free-living bdelloids and monogononts) with respect to endoparasitic acanthocephalans. Data of epizoic seisonids, however, were absent, which may have affected the branching order within the syndermatan clade. Moreover, the position of Seisonidea within Syndermata should help in understanding the evolution of acanthocephalan endoparasitism. Here, we report the first phylogenomic analysis that includes all four higher-ranked groups of Syndermata. The analyzed data sets comprise new transcriptome data for Seison spec. (Seisonidea), Brachionus manjavacas (Monogononta), Adineta vaga (Bdelloidea), and Paratenuisentis ambiguus (Acanthocephala). Maximum likelihood and Bayesian trees for a total of 19 metazoan species were reconstructed from up to 410 functionally diverse proteins. The results unanimously place Monogononta basally within Syndermata, and Bdelloidea appear as the sister group to a clade comprising epizoic Seisonidea and endoparasitic Acanthocephala. Our results support monophyly of Syndermata, Hemirotifera (Bdelloidea + Seisonidea + Acanthocephala), and Pararotatoria (Seisonidea + Acanthocephala), rejecting monophyly of traditional Rotifera and Eurotatoria. This serves as an indication that early acanthocephalans lived epizoically or as ectoparasites on arthropods, before their complex lifecycle with arthropod intermediate and vertebrate definite hosts evolved.   7. Rapid toxicity assessment using an in vivo enzyme test for Brachionus plicatilis (Rotifera).

    A 1-hr in vivo enzyme inhibition assay based on esterase activity has good potential for marine toxicity assessment. A test was developed for the rotifer Brachionus plicatilis based on the nonfluorescent substrate fluorescein diacetate (FDA), which is metabolized by esterases to a fluorescent product. Enzyme inhibition, as determined by reduced fluorescence, can be scored visually or quantified using a fluorometer. Quantification of fluorescence permits the calculation of NOEC, LOEC, chronic value, and IC20. The 1-hr esterase inhibition test has sensitivity comparable to that of 24-hr rotifer acute tests for several compounds. The toxicity of six compounds was examined using the quantified assay. The resulting IC20s were within a factor of 3 of the 24-hour LC50s. IC20 values ranged from 0.017 mg/l for tributyltin to 3.1 mg/l for zinc, with an average coefficient of variation of 17.8%. Electrophoretic analysis of rotifer homogenates suggested that a single C esterase (acetylesterase) was responsible for FDA metabolism in B. plicatilis. Several other aquatic species are capable of metabolizing FDA, including Brachionus calyciflorus, Mysidopsis bahia, Menidia beryllina, Pimephales promelas, Ceriodaphnia dubia, Daphnia pulex, Artemia salina, and Ophryotrocha sp. The esterase inhibition test is an attractive tool for assessing aquatic toxicity because of its speed, simplicity, sensitivity, and applicability to a broad range of aquatic species.

  8. A comparison of the fate and effects of two pyrethroid insecticides (lambda-cyhalothrin and cypermethrin) in pond mesocosms.

    : The fate and effects of two pyrethroid insecticides (lambda-cyhalothrin and cypermethrin) were investigated in replicated 25 m(3) pond mesocosms. Three pesticide treatments which simulated spray drift deposition were examined: 0.7 g a.i. ha(-1) cypermethrin and 0.17 and 1.7 g a.i. ha(-1) lambda-cyhalothrin. Based on the use rate and pesticidal activity of the chemicals, the cypermethrin and lower lambda-cyhalothrin rates were approximately equivalent. After applications, pyrethroid residues in the water column declined rapidly. Treatment-related effects were observed on some macroinvertebrate taxa, most notably the Asellidae and Gammaridae. Surfacedwelling insects also suffered initial knock-down, particularly in the 1.7 g a.i. ha(-1) lambda-cyhalothrin treatment, but there was recovery after the spray period. No adverse effects occurred on algae, macrophytes or zooplankton, but there were occasional enhancements (e.g. algal biomass and abundances of copepod nauplii and Rotifera) which may have been indirect effects. An overall comparison of the treatments indicated that the higher lambda-cyhalothrin rate had the greatest effects, whilst the cypermethrin application had a somewhat greater impact than the lower lambda-cyhalothrin treatment rate (due to effects on peracarid crustaceans). The study indicated that should spray drift occur at the levels expected for either pyrethroid's normal use patterns, potential impacts on natural aquatic ecosystems would be minor and transient.

  9. Tolerance to X-rays and Heavy Ions (Fe, He) in the Tardigrade Richtersius coronifer and the Bdelloid Rotifer Mniobia russeola.

    The aim of this study was to analyze tolerance to heavy ions in desiccated animals of the eutardigrade Richtersius coronifer and the bdelloid rotifer Mniobia russeola within the STARLIFE project. Both species were exposed to iron (Fe) and helium (He) ions at the Heavy Ion Medical Accelerator in Chiba (HIMAC) in Chiba, Japan, and to X-rays at the German Aerospace Center (DLR) in Cologne, Germany. Results show no effect of Fe and He on viability up to 7 days post-rehydration in both R. coronifer and M. russeola, while X-rays tended to reduce viability in R. coronifer at the highest doses. Mean egg production rate tended to decline with higher doses in R. coronifer for all radiation types, but the pattern was not statistically confirmed. In M. russeola, there was no such tendency for a dose response in egg production rate. These results confirm the previously reported high tolerance to high linear energy transfer (LET) radiation in tardigrades and show for the first time that bdelloid rotifers are also very tolerant to high-LET radiation. These animal phyla represent the most desiccation- and radiation-tolerant animals on Earth and provide excellent eukaryotic models for astrobiological research.   10. Applicability of cryoconite consortia of microorganisms and glacier-dwelling animals in astrobiological studies

    For several years it has been of interest to astrobiologists to focus on Earth's glaciers as a habitat that can be similar to glaciers on other moons and planets. Microorganisms on glaciers form consortia - cryoconite granules (cryoconites). They are granular/spherical mineral particles connected with archaea, cyanobacteria, heterotrophic bacteria, algae, fungi, and micro animals (mainly Tardigrada and Rotifera). Cryophilic organisms inhabiting glaciers have been studied in different aspects: from taxonomy, ecology and biogeography, to searching of biotechnological potentials and physiological strategies to survive in extreme glacial habitats. However, they have never been used in astrobiological experiments. The main aim of this paper is brief review of literature and supporting assumptions that cryoconite granules and microinvertebrates on glaciers, are promising models in astrobiology for looking for analogies and survival strategies in terms of icy planets and moons. So far, astrobiological research have been conducted on single strains of prokaryotes or microinvertebrates but never on a consortium of them. Due to the hypothetical similarity of glaciers on the Earth to those on other planets these cryoconites consortia of microorganisms and glacier microinvertebrates may be applied in astrobiological experiments instead of the limno-terrestrial ones used currently. Those consortia and animals have qualities to use them in such studies and they may be the key to understanding how organisms are able to survive, reproduce and remain active at low temperatures.

  11. Comparison of biofilm ecology supporting growth of individual Naegleria species in a drinking water distribution system.

    Free-living amoebae (FLA) are common components of microbial communities in drinking water distribution systems (DWDS). FLA are of clinical importance both as pathogens and as reservoirs for bacterial pathogens, so identifying the conditions promoting amoebae colonisation of DWDSs is an important public health concern for water utilities. We used high-throughput amplicon sequencing to compare eukaryotic and bacterial communities associated with DWDS biofilms supporting distinct FLA species (Naegleria fowleri, N. lovaniensis or Vermamoeba sp.) at sites with similar physical/chemical conditions. Eukaryote and bacterial communities were characteristics of different FLA species presence, and biofilms supporting Naegleria growth had higher bacterial richness and higher abundance of Proteobacteria, Bacteroidetes (bacteria), Nematoda and Rotifera (eukaryota). The eukaryotic community in the biofilms had the greatest difference in relation to the presence of N. fowleri, while the bacterial community identified individual bacterial families associated with the presence of different Naegleria species. Our results demonstrate that ecogenomics data provide a powerful tool for studying the microbial and meiobiotal content of biofilms, and, in these samples can effectively discriminate biofilm communities supporting pathogenic N. fowleri.   12. Environmental DNA metabarcoding reveals primary chemical contaminants in freshwater sediments from different land-use types.

    Land-use intensification threatens freshwater biodiversity. Freshwater eukaryotic communities are affected by multiple chemical contaminants with a land-use specific manner. However, biodiversities of eukaryotes and their associations with multiple chemical contaminants are largely unknown. This study characterized in situ eukaryotic communities in sediments exposed to mixtures of chemical contaminants and assessed relationships between various environmental variables and eukaryotic communities in sediments from the Nanfei River. Eukaryotic communities in the sediment samples were dominated by Annelida, Arthropoda, Rotifera, Ochrophyta, Chlorophyta and Ciliophora. Alpha-diversities (Shannon entropy) and structures of eukaryotic communities were significantly different between land-use types. According to the results of multiple statistical tests (PCoA, distLM, Mantel and network analysis), dissimilarity of eukaryotic community structures revealed the key effects of pyrethroid insecticides, manganese, zinc, lead, chromium and polycyclic aromatic hydrocarbons (PAHs) on eukaryotic communities in the sediment samples from the Nanfei River. Furthermore, taxa associated with land-use types were identified and several sensitive eukaryotic taxa to some of the primary contaminants were identified as potential indicators to monitor effects of the primary chemical contaminants. Overall, environmental DNA metabarcoding on in situ eukaryotic communities provided a powerful tool for biomonitoring and identifying primary contaminants and their complex effects on benthic eukaryotic communities in freshwater sediments.     Currently, Viral Nervous Necrotic (VNN) is not only attacking the marine fish but also the freshwater fish like tilapia (Oreochromis niloticus). The aims of study to identify the type of plankton, water quality status, blood cell status, also histology of VNN infected tilapia obtained in culture ponds. The methods included plankton identification and water quality analysis from the infected fish pond in the Krakal, Blitar. The quality of blood cells and the histology of tilapia infected by VNN observed using a microscope with Hematoxylin-Eosin staining. The result show plankton in a fish pond of infected tilapia includes 3 divisions: Chlorophyta, Cyanophyta, and Bacillariophyta and 2 phyla: Arthropoda, and Rotifera. The values of erythrocyte, hematocrit, and hemoglobin were smaller than normal tilapia, however, the leukocyte and macronucleus values of VNN-infected fish were higher than normal fish.   14. Do small animals have a biogeography?

    It has been stated that small organisms do not have barriers for distribution and will not show biogeographic discreteness. General models for size-mediated biogeographies establish a transition region between ubiquitous dispersal and restricted biogeography at about 1-10 mm. We tested patterns of distribution versus size with water mites, a group of freshwater organisms with sizes between 300 microm and 10 mm. We compiled a list of all known water mite species for Sierra del Guadarrama (a mountain range in the centre of the Iberian Peninsula) from different authors and our own studies in the area. Recorded habitats include lotic, lentic and interstitial environments. Species body size and world distribution were drawn from our work and published specialized taxonomic literature. The null hypothesis was that distribution is size-independent. The relationship between distribution and size was approached via analysis of variance and between size and habitat via logistic regression. Contrary to expectations, there is no special relationship between water mite size and area size distribution. On the other hand, water mite size is differentially distributed among habitats, although this ecological sorting is very weak. Larger water mites are more common in lentic habitats and smaller water mites in lotic habitats. Size-dependent distribution in which small organisms tend to be cosmopolitan breaks down when the particular biology comes into play. Water mites do not fit a previously proposed size-dependent biogeographical distribution, and are in accordance with similar data published on Tardigrada, Rotifera, Gastrotricha and the like.

  15. Diversity and distribution of heterotrophic dinoflagellates from the coastal waters of Port Blair, South Andaman.

    The interaction between the environment and heterotrophic dinoflagellates inhabiting coastal waters of South Andaman was studied based on year round collections made during September 2012-August 2013 in the bay, eastern, and western region of South Andaman. The distribution pattern of microzooplankton in South Andaman showed high abundance in eutrophic waters (bay region) and gradually decreased towards the off shore region. Microzooplankton community comprised of six different taxa, viz. tintinnids, heterotrophic dinoflagellates, non-loricate ciliates, Foraminifera, Rotifera, and Copepoda (nauplii). Tintinnids were the major component of the microzooplankton (43.8 ± 7%) followed by heterotrophic dinoflagellates (34 ± 12%) and copepod nauplii (18.8 ± 4.0%). This study focused on heterotrophic dinoflagellates which ranked next to tintinnids in overall abundance and contributed 38-42% in the bay, 22-37% in the eastern, and 15-29% in the western region to the microzooplankton community. Dinoflagellates showed a positive correlation with salinity and a negative correlation with dissolved oxygen and chlorophyll a (r = - 0.3). Abundance of heterotrophic dinoflagellates in this area may be due to their diverse and advantageous mode of nutrition. A total of 35 species belonging to 8 genera of heterotrophic dinoflagellates were recorded during the study period. Heterotrophic dinoflagellates showed a great potential to thrive in low oxygenated and low productive area (p < 0.001, Mann-Whitney test). Relatively higher diversity (H') in the dinoflagellates population was found in the bay region (avg. H' = 3.46).

Yet, such variation might provide important clues on the tempo and mode of genome size evolution. In this study we used the flow-cytometry method to study the evolution of genome size in the rotifer Brachionus plicatilis, a cryptic species complex consisting of at least 14 closely related species. We found an unexpectedly high variation in this species complex, with genome sizes ranging approximately seven-fold (haploid '1C' genome sizes: 0.056-0.416 pg). Most of this variation (67%) could be ascribed to the major clades of the species complex, i.e. clades that are well separated according to most species definitions. However, we also found substantial variation (32%) at lower taxonomic levels--within and among genealogical species--and, interestingly, among species pairs that are not completely reproductively isolated. In one genealogical species, called B. 'Austria', we found greatly enlarged genome sizes that could roughly be approximated as multiples of the genomes of its closest relatives, which suggests that whole-genome duplications have occurred early during separation of this lineage. Overall, genome size was significantly correlated to egg size and body size, even though the latter became non-significant after controlling for phylogenetic non-independence. Our study suggests that substantial genome size variation can build up early during speciation, potentially even among isolated populations. An alternative, but not mutually exclusive interpretation might be that reproductive isolation tends to build up unusually slow in this species complex.

  18. Variability of invertebrate abundance in drinking water distribution systems in the Netherlands in relation to biostability and sediment volumes.

    A survey of invertebrates in drinking water from treatment works, internal taps and hydrants on mains was carried out by almost all water companies in the Netherlands from September 1993 to August 1995. Aquatic sow bugs (Asellidae, 1-12 mm) and oligochaeta worms (Oligochaeta, 1-100 mm), both known to have caused rare though embarrassing consumer complaints, were found to form 98% of the mean biomass in water flushed from mains. Their numbers in the mains water ranged up to 1500 (mean 37) Asellidae m(-3) and up to 9900 (mean 135) Oligochaeta m(-3). Smaller crustaceans (0.5-2 mm) dominated the numbers in water from mains. e.g. water fleas (Cladocera and Copepoda up to 14,000 m(-3)). Common invertebrates in treated water and in tap water were Rotifera (<1 mm) and nematode worms (Nematoda, <2 mm). No Asellidae, large Oligochaeta (>5 mm) or other large invertebrates were found in 1560 samples of 200 l treated water or tap water. Large variations in invertebrate abundance were found within and between distribution systems. Of the variability of mean biomass in mains per system, 55%, 60% and 63% could statistically be explained by differences in the Biofilm Formation Rate, non-particulate organic matter and the permanganate index of the treated water of the treatment works respectively.   19. Multigenerational effects of triclosan on the demography of Plationus patulus and Brachionus havanaensis (ROTIFERA).

    Triclosan is a personal care product widely used in North America, Europe and Asia as antimicrobial ingredient in many consumer chemical products. In Mexico concentrations of triclosan have been reported in aquatic systems. However, there is no law regulating the presence of chemicals such as triclosan, in aquatic systems. The scarce data about this chemical has increased concern among ecotoxicologists regarding possible effects on aquatic organisms. Moreover, multigenerational studies are rarely studied and the results vary depending on the contaminant. Rotifers, are a dominant group of zooplankton, and have been used in aquatic risk assessments of personal care products due to their sensitivity and high reproductive rates. Plationus patulus and Brachionus havanaensis are common rotifers distributed in aquatic ecosystems of Mexico and have been used in ecotoxicological bioassays. In this study, the median lethal concentration (LC50, 24h) of P. patulus and B. havanaensis exposed to triclosan was determined. Based on the LC50, we tested three sublethal concentrations of triclosan to quantify the demographic responses of both rotifers for two successive generations (F0, and F1). The 24h LC50 of triclosan for P. patulus and B. havanaensis were 300 and 500µgL -1 respectively. Despite the concentration, triclosan had an adverse effect on both Plationus patulus and Brachionus havanaensis in both generations exposed. Experiments show that P. patulus was more sensitive than B. havanaensis when exposed to triclosan. When exposed to triclosan the parental generation (F0) of P. patulus was far more affected than F1.   20. Food web effects of titanium dioxide nanoparticles in an outdoor freshwater mesocosm experiment.

    Over the course of 78 days, nine outdoor mesocosms, each with 1350 L capacity, were situated on a pontoon platform in the middle of a lake and exposed to 0 μg L(-1) TiO2, 25 μg L(-1) TiO2 or 250 μg L(-1) TiO2 nanoparticles in the form of E171 TiO2 human food additive five times a week. Mesocosms were inoculated with sediment, phytoplankton, zooplankton, macroinvertebrates, macrophytes and fish before exposure, ensuring a complete food web. Physicochemical parameters of the water, nutrient concentrations, and biomass of the taxa were monitored. Concentrations of 25 μg L(-1) TiO2 and 250 μg L(-1) TiO2 caused a reduction in available soluble reactive phosphorus in the mesocosms by 15 and 23%, respectively, but not in the amount of total phosphorus. The biomass of Rotifera was significantly reduced by 32 and 57% in the TiO2 25 μg L(-1) and TiO2 250 μg L(-1) treatments, respectively, when compared to the control; however, the biomass of the other monitored groups-Cladocera, Copepoda, phytoplankton, macrophytes, chironomids and fish-remained unaffected. In conclusion, environmentally relevant concentrations of TiO2 nanoparticles may negatively affect certain parameters and taxa of the freshwater lentic aquatic ecosystem. However, these negative effects are not significant enough to affect the overall function of the ecosystem, as there were no cascade effects leading to a major change in its trophic state or primary production.

  1. Spiralian phylogeny informs the evolution of microscopic lineages.

    Despite rapid advances in the study of metazoan evolutionary history [1], phylogenomic analyses have so far neglected a number of microscopic lineages that possess a unique combination of characters and are thus informative for our understanding of morphological evolution. Chief among these lineages are the recently described animal groups Micrognathozoa and Loricifera, as well as the two interstitial "Problematica" Diurodrilus and Lobatocerebrum [2]. These genera show a certain resemblance to Annelida in their cuticle and gut [3, 4]; however, both lack primary annelid characters such as segmentation and chaetae [5]. Moreover, they show unique features such as an inverted body-wall musculature or a novel pharyngeal organ. This and their ciliated epidermis have led some to propose relationships with other microscopic spiralians, namely Platyhelminthes, Gastrotricha, and in the case of Diurodrilus, with Micrognathozoa [6, 7]-lineages that are grouped by some analyses into "Platyzoa," a clade whose status remains uncertain [1, 8-11]. Here, we assess the interrelationships among the meiofaunal and macrofaunal members of Spiralia using 402 orthologs mined from genome and transcriptome assemblies of 90 taxa. Lobatocerebrum and Diurodrilus are found to be deeply nested members of Annelida, and unequivocal support is found for Micrognathozoa as the sister group of Rotifera. Analyses using site-heterogeneous substitution models further recover a lophophorate clade and position Loricifera + Priapulida as sister group to the remaining Ecdysozoa. Finally, with several meiofaunal lineages branching off early in the diversification of Spiralia, the emerging concept of a microscopic, acoelomate, direct-developing ancestor of Spiralia is reviewed.   2. Accumulation of atmospheric radionuclides and heavy metals in cryoconite holes on an Arctic glacier.

    Surface of glaciers is covered by mineral and organic dust, together with microorganisms forming cryoconite granules. Despite fact that glaciers and ice sheets constitute significance part of land surface, reservoir of freshwater, and sites of high biological production, the knowledge on the cryoconite granules still remain unsatisfactory. This study presents information on radionuclide and heavy metal contents in cryoconites. Cryoconites collected from the Hans Glacier in SW Spitsbergen reveal high activity concentrations of anthropogenic ((238,239,240)Pu, (137)Cs, (90)Sr) and natural ((210)Pb) radionuclides. The (238)Pu/(239+240)Pu activity ratios in these cryoconites significantly exceed the mean global fallout ratio (0.025). The (238)Pu/(239+240)Pu ranged from 0.064 to 0.118. The (239+240)Pu/(137)Cs varied from 0.011 ± 0.003 to 0.030 ± 0.007. Such activity ratios as observed in these cryoconites were significantly higher than the values characterizing global fallout, pointing to possible contributions of these radionuclides from other sources. Heavy metals (Pb, Cd, Cu, Zn, Fe, and Mn) in cryoconites exceed both UCC concentrations and local rocks' concentrations, particularly for cadmium. The concentration ratios of stable lead isotopes ((206)Pb/(207)Pb, (208)Pb/(206)Pb) were determined to discriminate between the natural and anthropogenic sources of Pb in cryoconites and to confirm the strong anthropogenic contribution to heavy metal deposition in the Arctic. In investigated cryoconite holes, two groups of invertebrates, both extremophiles, Tardigrada and Rotifera were detected. Our study indicate that cryoconites are aggregates of mineral and organic substances on surfaces of glaciers are able to accumulate large amounts of airborne pollutants bound to extracellular polymeric substances secreted by microorganisms.   3. A novel adhering junction in the apical ciliary apparatus of the rotifer Brachionus plicatilis (Rotifera, Monogononta).

    Cultures of the rotifer Brachionus plicatilis were examined with regard to their interepithelial junctions after infiltration with the extracellular tracer lanthanum, freeze-fracturing or quick-freeze deep-etching. The lateral borders between ciliated cells have an unusual apical adhering junction. This apical part of their intercellular cleft looks desmosome-like, but it is characterized by unusual intramembranous E-face clusters of particles. Deep-etching reveals that these are packed together in short rows which lie parallel to one another in orderly arrays. The true membrane surface in these areas features filaments in the form of short ribbons; these are produced by projections, possibly part of the glycocalyx, emerging from the membranes, between which the electron-dense tracer lanthanum permeates. These projections appear to overlap with each other in the centre of the intercellular cleft; this would provide a particularly flexible adaptation to maintain cell-cell contact and coordination as a consequence. The filamentous ribbons may be held in position by the intramembranous particle arrays since both have a similar size and distribution. These contacts are quite different from desmosomes and appear to represent a distinct new category of adhesive cell-cell junction. Beneath these novel structures, conventional pleated septate junctions are found, exhibiting the undulating intercellular ribbons typical of this junctional type, as well as the usual parallel alignments of intramembranous rows of EF grooves and PF particles. Below these are found gap junctions as close-packed plaques of intramembranous particles on either the P-face or E-face. After freeze-fracturing, the complementary fracture face to the particles shows pits, usually on the P-face, arrayed with a very precise hexagonal pattern.

  4. Fate of thiamethoxam in mesocosms and response of the zooplankton community.

    Thiamethoxam is a neonicotinoid insecticide that can reach wetlands in agro-ecosystems through runoff. The fate and effects of thiamethoxam on non-target organisms in shallow wetland ecosystems have not been well characterized. To this end, a mesocosm study was conducted with a focus on characterizing zooplankton community responses. A single pulse application of thiamethoxam (0, 25, 50, 100, 250, and 500 μg/L; n = 3) was applied to experimental systems and monitored for 8 weeks. The mean half-life of thiamethoxam among the different treatments was 3.7 days in the water column with concentrations of <0.8 μg/L in the majority of mesocosms by 56 days. Principal response curve analysis did not show any significant concentration-dependent differences in the zooplankton community among treatments over the course of the study. The minimum detectable difference (MDD%) values for abundance of potentially sensitive arthropod taxa (nauplius larvae, cyclopoid copepods) allowed the detections from controls as low as 42 and 59% effect, respectively. The MDD% values for total abundance of zooplankton (including the potentially less sensitive taxonomic group of Rotifera) allowed the detection from controls as low as 41% effect. There were no statistically significant differences in zooplankton abundance or diversity between control and treated mesocosms at the end of the study. There were also no statistically significant differences for individual taxa that were sustained between sampling points, or manifested as a concentration-response. We conclude that acute exposure to thiamethoxam at environmentally relevant concentrations (typically ng/L) likely does not represent a significant adverse ecological risk to wetland zooplankton community abundance and structure.     The magnitude of community-wide dispersal is central to metacommunity models, yet dispersal is notoriously difficult to quantify in passive and cryptic dispersers such as many freshwater invertebrates. By overcoming the problem of quantifying dispersal rates, colonization rates into new habitats can provide a useful estimate of the magnitude of effective dispersal. Here we study the influence of spatial and local processes on colonization rates into new ponds that indicate differential dispersal limitation of major zooplankton taxa, with important implications for metacommunity dynamics. We identify regional and local factors that affect zooplankton colonization rates and spatial patterns in a large-scale experimental system. Our study differs from others in the unique setup of the experimental pond area by which we were able to test spatial and environmental variables at a large spatial scale. We quantified colonization rates separately for the Copepoda, Cladocera and Rotifera from samples collected over a period of 21 months in 48 newly constructed temporary ponds of 0.18–2.95 ha distributed in a restored wetland area of 2,700 ha in Doñana National Park, Southern Spain. Species richness upon initial sampling of new ponds was about one third of that in reference ponds, although the rate of detection of new species from thereon were not significantly different, probably owing to high turnover in the dynamic, temporary reference ponds. Environmental heterogeneity had no detectable effect on colonization rates in new ponds. In contrast, connectivity, space (based on latitude and longitude) and surface area were key determinants of colonization rates for copepods and cladocerans. This suggests dispersal limitation in cladocerans and copepods, but not in rotifers, possibly due to differences in propagule size and abundance. PMID:22792241

  7. Multitasking of the piRNA Silencing Machinery: Targeting Transposable Elements and Foreign Genes in the Bdelloid Rotifer Adineta vaga.

    RNA-mediated silencing processes play a key role in silencing of transposable elements, especially in the germ line, where piwi-interacting RNAs (piRNAs) are responsible for suppressing transposon mobility and maintaining genome integrity. We previously reported that the genome of Adineta vaga, the first sequenced representative of the phylum Rotifera (class Bdelloidea), is characterized by massive levels of horizontal gene transfer, by unusually low transposon content, and by highly diversified RNA-mediated silencing machinery. Here, we investigate genome-wide distribution of pi-like small RNAs, which in A. vaga are 25-31 nucleotides in length and have a strong 5'-uridine bias, while lacking ping-pong amplification signatures. In agreement with expectations, 71% of mapped reads corresponded to annotated transposons, with 93% of these reads being in the antisense orientation. Unexpectedly, a significant fraction of piRNAs originate from predicted coding regions corresponding to genes of putatively foreign origin. The distribution of piRNAs across foreign genes is not biased toward 3'-UTRs, instead resembling transposons in uniform distribution pattern throughout the gene body, and in predominantly antisense orientation. We also find that genes with small RNA coverage, including a number of genes of metazoan origin, are characterized by higher occurrence of telomeric repeats in the surrounding genomic regions, and by higher density of transposons in the vicinity, which have the potential to promote antisense transcription. Our findings highlight the complex interplay between RNA-based silencing processes and acquisition of genes at the genome periphery, which can result either in their loss or eventual domestication and integration into the host genome.   8. Combined effects of temperature and salinity on the demographic response of Proales similis (Beauchamp, 1907) and Brachionus plicatilis (Müller, 1786) (Rotifera) to mercury.

    The demographic response of the brackish-water rotifers Proales similis and Brachionus plicatilis to mercury (0.5, 2, 8 and 32 μg L -1 of HgCl 2 ) at different salinity levels (10 and 20‰) and two temperature (25 °C and 32 °C) regimes were evaluated. Median lethal concentration (LC 50 ) for P. similis and B. plicatilis was 10 and 16 μg L -1 , respectively, showing that Proales similis was more sensitive to mercury than B. plicatilis. The rate of population increase (r) for both species was greater at 10‰ salinity and 32 °C (ranged from 0.6 to 0.95 d -1 ). The r-value decreased as the concentration of mercury in the medium increased. Regardless of the temperature, at lower salinity and higher mercury concentration (32 μg L -1 ), P. similis died within six days. The survivorship of P. similis and B. plicatilis was higher at 25 °C than at 32 °C (ranged from 5 to 8 and 7-13 d, respectively). Fecundity was higher at 32 °C than at 25 °C for both rotifers species. There was a significant effect of the interaction among salinity, temperature, and mercury in both species on the reproductive variables such as net and gross reproductive rates, generation time and the rate of population increase.   9. Patterns of diversity in soft-bodied meiofauna: dispersal ability and body size matter.

    Biogeographical and macroecological principles are derived from patterns of distribution in large organisms, whereas microscopic ones have often been considered uninteresting, because of their supposed wide distribution. Here, after reporting the results of an intensive faunistic survey of marine microscopic animals (meiofauna) in Northern Sardinia, we test for the effect of body size, dispersal ability, and habitat features on the patterns of distribution of several groups. As a dataset we use the results of a workshop held at La Maddalena (Sardinia, Italy) in September 2010, aimed at studying selected taxa of soft-bodied meiofauna (Acoela, Annelida, Gastrotricha, Nemertodermatida, Platyhelminthes and Rotifera), in conjunction with data on the same taxa obtained during a previous workshop hosted at Tjärnö (Western Sweden) in September 2007. Using linear mixed effects models and model averaging while accounting for sampling bias and potential pseudoreplication, we found evidence that: (1) meiofaunal groups with more restricted distribution are the ones with low dispersal potential; (2) meiofaunal groups with higher probability of finding new species for science are the ones with low dispersal potential; (3) the proportion of the global species pool of each meiofaunal group present in each area at the regional scale is negatively related to body size, and positively related to their occurrence in the endobenthic habitat. Our macroecological analysis of meiofauna, in the framework of the ubiquity hypothesis for microscopic organisms, indicates that not only body size but mostly dispersal ability and also occurrence in the endobenthic habitat are important correlates of diversity for these understudied animals, with different importance at different spatial scales. Furthermore, since the Western Mediterranean is one of the best-studied areas in the world, the large number of undescribed species (37%) highlights that the census of marine meiofauna is still very far from being complete.

  10. Epiphytic invertebrate patterns in coastal lakes along a gradient of salinity and water exchange with the sea

    The species composition and abundance of epiphytic fauna inhabiting common reed (Phragmites australis (Cav.) Trin. ex Steud.) was performed in five coastal lakes in Słowiński National Park (southern Baltic coast in northern Poland). The lakes represent a salinity gradient (from freshwater to β-oligohaline waters) and four types of coastal lakes: (1) lagoon, L (Lake Łebsko, seawater enters it permanently); (2) coastal lake with periodically brackish water, CLB (Lake Gardno); (3) freshwater costal lake, CLF (Lake Smołdzińskie); and (4) coastal dune lakes, CLD (Dołgie Wielkie and Dołgie Małe). Using statistical ordination techniques, we found that the structure of epiphytic fauna (microinvertebrates and macroinvertebrates) is determined primarily by hydrological connectivity (water exchange) with the sea. Canonical Correspondence Analysis, coupled with variance partitioning, showed that hydrological connectivity accounted for 24% of the variation in the invertebrate community, followed by physico-chemical (19%) and trophic (8%) factors. Our results indicate that the assemblages of Ciliata-libera and Cnidaria are characteristic for L (β-oligohaline), Rotifera, Suctoria, Chaetogaster sp., Gastropoda and Trichoptera are characteristic for CLB (limnetic/β-oligohaline), but no taxonomic groups are characteristic for CLF and CLD (both limnetic). The index of multivariate dispersion showed a decreasing trend with the increasing lake isolation from the open sea, except for CLD. However, in terms of the structure of epiphytic fauna, Multi-Response Permutation Procedures showed that CLD significantly differed only from CLB. Our results suggest that the identified characteristic taxonomic groups of plant-associated macroinvertebrates have a high potential to be used as bioindicators of salinity and water exchange with the sea, due to their sensitivity to environmental stress.

  11. Multitasking of the piRNA Silencing Machinery: Targeting Transposable Elements and Foreign Genes in the Bdelloid Rotifer Adineta vaga

    RNA-mediated silencing processes play a key role in silencing of transposable elements, especially in the germ line, where piwi-interacting RNAs (piRNAs) are responsible for suppressing transposon mobility and maintaining genome integrity. We previously reported that the genome of Adineta vaga, the first sequenced representative of the phylum Rotifera (class Bdelloidea), is characterized by massive levels of horizontal gene transfer, by unusually low transposon content, and by highly diversified RNA-mediated silencing machinery. Here, we investigate genome-wide distribution of pi-like small RNAs, which in A. vaga are 25–31 nucleotides in length and have a strong 5′-uridine bias, while lacking ping-pong amplification signatures. In agreement with expectations, 71% of mapped reads corresponded to annotated transposons, with 93% of these reads being in the antisense orientation. Unexpectedly, a significant fraction of piRNAs originate from predicted coding regions corresponding to genes of putatively foreign origin. The distribution of piRNAs across foreign genes is not biased toward 3′-UTRs, instead resembling transposons in uniform distribution pattern throughout the gene body, and in predominantly antisense orientation. We also find that genes with small RNA coverage, including a number of genes of metazoan origin, are characterized by higher occurrence of telomeric repeats in the surrounding genomic regions, and by higher density of transposons in the vicinity, which have the potential to promote antisense transcription. Our findings highlight the complex interplay between RNA-based silencing processes and acquisition of genes at the genome periphery, which can result either in their loss or eventual domestication and integration into the host genome.   12. Effect of crude extracts from cyanobacterial blooms in Lake Texcoco (Mexico) on the population growth of Brachionus calyciflorus (Rotifera).

    Unlike temperate regions, tropical ecosystems are characterized by high temperatures (>18 °C) all year, promoting blooms of cyanobacteria which often produce secondary metabolites toxic to zooplankton. Nabor Carillo and the Recreational Lake are part of the saline, Lake Texcoco, in Central Mexico which is filled nowadays with treated waste water. Both water bodies are dominated by Planktothrix, Anabaenopsis, Spirulina and Microcystis. In this study we present the concentration of microcystins in these waterbodies over an annual cycle. We also evaluated the chronic effects of cyanobacterial crude extracts from both lakes on two clones of the rotifer Brachionus calyciflorus, one from Nabor Carrillo Lake and the other from a canal in the shallow, Lake Xochimilco. The experiments on population growth were performed, beginning with 10 individuals per container for each of the following treatments: control (no crude extract), concentrated crude extract, and diluted crude extract (50:50) with moderately hard water and Chlorella vulgaris in a concentration of 0.5 × 10 6  cells ml -1 . The cyanotoxin levels were measured using an ELISA test and ranged between 0.20 and 2.4 μg L -1 in the lake water. The results showed that the Recreational Lake extracts were more toxic, killing the rotifers in less than five days. The r values ranged from -1.74 to 0.48 in the presence of the crude extracts and 0.16 and 0.24 in the controls. The results have been discussed with emphasis on the importance of conducting regular studies to test ecotoxicological impacts of cyanobacterial blooms in tropical waters.   13. Changes of concentrations and possibility of accumulation of bisphenol A and alkylphenols, depending on biomass and composition, in zooplankton of the Southern Baltic (Gulf of Gdansk).

    The focus of the present study was to find the relationship between concentrations of bisphenol A (BPA), 4-tert-octylphenol (OP) and 4-nonylphenol (NP) in zooplankton and seasonal changes in the composition and biomass of particular zooplankton taxa in the Gulf of Gdansk (Southern Baltic) in the years 2011-2012. Assays of BPA, OP and NP in water and zooplankton samples were performed using the HPLC/FL system. High mean concentrations of the studied compounds, determined in spring (405.9 (BPA); 25.7 (OP); 111.2 (NP) ng g(-1) dw), can be linked to the high proportion of meroplankton in that season. Rotifera also had an influence on the rise in concentrations of the studied compounds but to a lesser degree, while the lowest concentrations (determined in summer) can be associated with the high participation of Copepoda and Cladocera in zooplankton biomass. It was also observed that juvenile forms can be more susceptible to accumulating endocrine disrupting compounds (EDCs). This is indicated by the positive correlation between BPA concentration in zooplankton and the proportion of Copepoda nauplii biomass in spring (r = 0.90; p < 0.05). In most cases, greater zooplankton biomass accumulated higher concentrations and loads of the studied compounds. With biomass growth (to 123.32 μg m(-3)), the bioconcentration factor also rose (to max 46.1·10(3)), demonstrating that unlike typical hydrophobic compounds the studied EDCs do not become "diluted" in zooplankton biomass.   14. Global isolation by distance despite strong regional phylogeography in a small metazoan

    Background Small vagile eukaryotic organisms, which comprise a large proportion of the Earth's biodiversity, have traditionally been thought to lack the extent of population structuring and geographic speciation observed in larger taxa. Here we investigate the patterns of genetic diversity, amongst populations of the salt lake microscopic metazoan Brachionus plicatilis s. s. (sensu stricto) (Rotifera: Monogononta) on a global scale. We examine the phylogenetic relationships of geographic isolates from four continents using a 603 bp fragment of the mitochondrial COI gene to investigate patterns of phylogeographic subdivision in this species. In addition we investigate the relationship between genetic and geographic distances on a global scale to try and reconcile the paradox between the high vagility of this species and the previously reported patterns of restricted gene flow, even over local spatial scales. Results Analysis of global sequence diversity of B. plicatilis s. s. reveals the presence of four allopatric genetic lineages: North American-Far East Asian, Western Mediterranean, Australian, and an Eastern Mediterranean lineage represented by a single isolate. Geographically orientated substructure is also apparent within the three best sampled lineages. Surprisingly, given this strong phylogeographic structure, B. plicatilis s. s. shows a significant correlation between geographic and genetic distance on a global scale ('isolation by distance' – IBD). Conclusion Despite its cosmopolitan distribution and potential for high gene flow, B. plicatilis s. s. is strongly structured at a global scale. IBD patterns have traditionally been interpreted to indicate migration-drift equilibrium, although in this system equilibrium conditions are incompatible with the observed genetic structure. Instead, we suggest the pattern may have arisen through persistent founder effects, acting in a similar fashion to geographic barriers for larger organisms. Our data indicates that geographic speciation, contrary to historical views, is likely to be very important in microorganisms. By presenting compelling evidence for geographic speciation in a small eukaryote we add to the growing body of evidence that is forcing us to rethink our views of global biodiversity. PMID:17999774

  15. Effects of the fungicide metiram in outdoor freshwater microcosms: responses of invertebrates, primary producers and microbes.

    The ecological impact of the dithiocarbamate fungicide metiram was studied in outdoor freshwater microcosms, consisting of 14 enclosures placed in an experimental ditch. The microcosms were treated three times (interval 7 days) with the formulated product BAS 222 28F (Polyram®). Intended metiram concentrations in the overlying water were 0, 4, 12, 36, 108 and 324 μg a.i./L. Responses of zooplankton, macroinvertebrates, phytoplankton, macrophytes, microbes and community metabolism endpoints were investigated. Dissipation half-life (DT₅₀) of metiram was approximately 1-6 h in the water column of the microcosm test system and the metabolites formed were not persistent. Multivariate analysis indicated treatment-related effects on the zooplankton (NOEC(community) = 36 μg a.i./L). Consistent treatment-related effects on the phytoplankton and macroinvertebrate communities and on the sediment microbial community could not be demonstrated or were minor. There was no evidence that metiram affected the biomass, abundance or functioning of aquatic hyphomycetes on decomposing alder leaves. The most sensitive populations in the microcosms comprised representatives of Rotifera with a NOEC of 12 μg a.i./L on isolated sampling days and a NOEC of 36 μg a.i./L on consecutive samplings. At the highest treatment-level populations of Copepoda (zooplankton) and the blue-green alga Anabaena (phytoplankton) also showed a short-term decline on consecutive sampling days (NOEC = 108 μg a.i./L). Indirect effects in the form of short-term increases in the abundance of a few macroinvertebrate and several phytoplankton taxa were also observed. The overall community and population level no-observed-effect concentration (NOEC(microcosm)) was 12-36 μg a.i./L. At higher treatment levels, including the test systems that received the highest dose, ecological recovery of affected measurement endpoints was fast (effect period < 8 weeks).

  17. Temporal and spatial distribution of the meiobenthic community in Daya Bay, South China Sea

    Spatial and temporal biodiversity patterns of the meiobenthos were studied for the first time in Daya Bay, which is a tropical semi-enclosed basin located in the South China Sea. The abundance, biomass, and composition of the meiobenthos and the basic environmental factors in the bay were investigated. The following 19 taxonomic groups were represented in the meiofauna: Nematoda, Copepoda, Polychaeta, Oligochaeta, Kinorhyncha, Gastrotricha, Ostracoda, Bivalvia, Turbellaria, Nemertinea, Sipuncula, Hydroida, Amphipoda, Cumacea, Halacaroidea, Priapulida, Echinodermata, Tanaidacea, and Rotifera. Total abundance and biomass of the meiobenthos showed great spatial and temporal variation, with mean values of 993.57 ± 455.36 ind cm-2 and 690.51 ± 210.64 μg 10 cm-2, respectively. Nematodes constituted 95.60 % of the total abundance and thus had the greatest effect on meiofauna quantity and distribution, followed by copepods (1.55 %) and polychaetes (1.39 %). Meiobenthos abundance was significantly negatively correlated with water depth at stations (r=-0.747, P<0.05) and significantly negatively correlated with silt-clay content (r=-0.516, P<0.01) and medium diameter (r=-0.499, P<0.01) of the sediment. Similar results were found for correlations of biomass and abundance of nematodes with environmental parameters. Polychaete abundance was positively correlated with the bottom water temperature (r=0.456, P<0.01). Meiobenthos abundance differed significantly among seasons (P<0.05), although no significant difference among stations and the interaction of station × season was detected by two-way ANOVA. In terms of vertical distribution, most of the meiobenthos was found in the surface layer of sediment. This pattern was apparent for nematodes and copepods, but a vertical distribution pattern for polychaetes was not as obvious. Based on the biotic indices and analyses of their correlations and variance, the diversity of this community was likely to be influenced by environmental variations.

    Rotifers are an important live feed for first feeding larvae of many fish species. The use of concentrated algae cells in the mass culture of the rotifer Brachionus plicatilis (Brachionidae) has opened new horizons for research on this organism. Pastes of Rhodomonas salina (Pyrenomonadaceae) obtained either by centrifugation or flocculation with chitosan were preserved, with or without vitamin C, at -20 degrees C for four weeks and were evaluated biochemically (proteins, lipids, pigments and fatty acids contents) and subsequently, were used to feed the rotifer Brachionus plicatilis at a ratio of 25 mg/L/day. Four different microalgae pastes were prepared: (1) centrifuged and preserved with vitamin C (CV), (2) centrifuged and preserved without vitamin C (C), (3) flocculated and with vitamin C (FV) and (4) flocculated without vitamin C (F). All treatments showed similar contents of proteins and total lipids with respect to control culture (a fresh culture of R. salina), with mean values of 40.0 +/- 2.32% and 12.0 +/- 1.45%, respectively. The pheophytin a/chlorophyll a ratio, a general indicator of the chemical status of microalgal concentrates, was similar (0.09-0.11) between centrifuged pastes and control culture, but was found to be higher in flocculated pastes (1.28-1.48). The fatty acid profile varied with respect to the control culture, mainly in the proportion of the essential polyunsaturated fatty acids (PUFAs): eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). Total PUFAs, EPA and DHA contents were statistically similar between centrifuged pastes and control culture (PUFAs: 47%, EPA: 4% and DHA: 4.7%), whereas values obtained for flocculated pastes were significantly lower. The rotifers grew equally well when fed with centrifuged pastes or control culture (maximum density: 320 rotifers/mL; instantaneous growth rate: 0.23 rotifers/day, fecundity: 1.49 eggs/female and productivity: 43 x 10(3) rotifers/L/day. No significant effect of vitamin C was found when used as a paste preservative. We concluded that centrifugation is an effective harvesting method, and that freezing to -20 degrees C for four weeks (no vitamin added), may help maintain the nutritional quality of R. salina paste, similar to fresh microalgae and can be offered to Brachionus plicatilis.

  19. Zoo-heleoplankton structure in three artificial ponds of North-eastern Argentina.

    The aim of the present study was to compare the abundance and species richness of zoo-heleoplankton bigger than 53 microm in an annual cycle under similar climate conditions in three artificial ponds, in order to observe the changes during an annual cycle. Samples were taken monthly from June 1993 to July 1994 in Corrientes, Argentina. The first pond (A) was covered an 80% by Eichhornia crassipes (Mart.), the second one (B) with bloom of Microcystis aeruginosa (Kurtzing) and the last one (C) with organic matter deposited in the bottom. The water was more acidic at pond A, and the water at pond B contained more dissolved oxygen concentration than the water at the other two ponds. The zoo-heleoplankton densities varied between 20-1728 ind.l(-1) at pond A, 42-4082 ind.l(-1) at pond B and 148-2447 ind.l(-1) at pond C. The maximum zoo-heleoplankton abundance was found in the pond with cyanobacteria bloom during Autumn 1994 and the minimum abundance was found in the one with a predominance of E. crassipes. The rank of species richness was pond A > pond B > pond C. Rotifera was the most abundant group in pond A whereas the larval stages of Copepoda were abundant in the other two ponds. Anuraeopsis navicula Rousselt 1910 was the dominant population in the pond with macrophytes prevalence. Brachionus calyciflorus Pallas 1776 and larval stage of Copepoda had variable proportions in the pond with cyanobacteria bloom. Thermocyclops decipiens (Kiefer 1929) was present during the annual cycle only in the pond with organic matter deposited in the bottom. The succession of taxa was observed in the pond with coverage of aquatic macrophytes and with cyanobacteria bloom. Differences in species richness and low similarity in zoo-heleoplankton between ponds were determined by differences in the quality of the water in relation to the presence of macrophytes, cyanobacteria, organic matter deposited in the bottom and fish predation. Multiple regression analysis (stepwise) revealed that water transparency, dissolved oxygen and conductivity were the environmental variables that explained more than 42% of variability in the abundance of the dominant species.

  20. Exposure and effects of sediment-spiked fludioxonil on macroinvertebrates and zooplankton in outdoor aquatic microcosms.

    Information from effects of pesticides in sediments at an ecosystem level, to validate current and proposed risk assessment procedures, is scarce. A sediment-spiked outdoor freshwater microcosm experiment was conducted with fludioxonil (lipophilic, non-systemic fungicide) to study exposure dynamics and treatment-related responses of benthic and pelagic macroinvertebrates and zooplankton. Besides blank control and solvent control systems the experiment had six different treatment levels (1.7-614mga.s./kg dry sediment) based around the reported 28-d No Observed Effect Concentration (NOEC) for Chironomus riparius (40mga.s./kg dry sediment). Twelve systems were available per treatment of which four were sacrificed on each of days 28, 56 and 84 after microcosm construction. Fludioxonil persisted in the sediment and mean measured concentrations were 53-82% of the initial concentration after 84days. The dissipation rate increased with the treatment level. Also exposure concentrations in overlying water were long-term, with highest concentrations 28days after initiation of the experiment. Sediment-dwelling Oligochaeta and pelagic Rotifera and Cladocera showed the most pronounced treatment-related declines. The most sensitive sediment-dwelling oligochaete was Dero digitata (population NOEC 14.2mga.s./kg dry sediment). The same NOEC was calculated for the sediment-dwelling macroinvertebrate community. The most sensitive zooplankton species was the cladoceran Diaphanosoma brachyurum (NOEC of 1.6μga.s./L in overlying water corresponding to 5.0mga.s./kg dry sediment). At the two highest treatments several rotifer taxa showed a pronounced decrease, while the zooplankton community-level NOEC was 5.6μga.s./L (corresponding to 14.2mga.s./kg dry sediment). Zooplankton taxa calanoid Copepoda and Daphnia gr. longispina showed a pronounced treatment-related increase (indirect effects). Consequently, an assessment factor of 10 to the chronic laboratory NOECs of Chironomus riparius (sediment) and Daphnia magna (water) results in a regulatory acceptable concentration that is sufficiently protective for both the sediment-dwelling and pelagic organisms in the microcosms.   1. Limnological study of Lake Shastina, Siskiyou County, California

    Lake Shastina provides water for irrigation in Shasta Valley, as well as recreation. Presently, its shoreline is being developed for summer homes. Surface water constituted more than 90 percent of the approximately 50,000 acre-foot (62-cubic hectometre) inflow to Lake Shastina in the 1972 water year. Controlled outflow is via the Montague Main Canal; however, leakage from the lake through volcanic rocks to the northwest was estimated to be greater than the measured outflow. Appreciable annual changes in the quantity of water in storage in the lake are related mainly to variations in annual inflow.From June through August the lake was thermally stratified. In the spring and summer the epilimnion was often supersaturated with oxygen, while at the same time the hypolimnion was undersaturated and 'often devoid of dissolved oxygen. Vertical stratification of carbon dioxide, carbonate, bicarbonate, hydrogen ion, nitrogen, and phosphorus was also recorded during the spring and summer. Orthophosphate, total phosphorus, and total nitrogen concentrations (organic, ammonium, and nitrate) were highest in the hypolimnion during the period of thermal stratification.Ten-inch (25-centimetre) core samples from the reservoir bottom were chemically analyzed at 0.8-inch (2-centimetre) intervals. The concentrations ranged from 6.3 to 28.9 milligrams per gram of iron, 0.07 to 0.43 milligrams per gram of manganese, 0.4 to 2.7 milligrams per gram of organic nitrogen plus ammonium, and 0.06 to 1.3 milligrams per gram of total phosphorus. Organic matter in the cores ranged from 4 to 14 percent.Green algae and diatoms were the dominant algal types, reaching maximum concentrations of about 7 and 30 million cells per litre, respectively. These phytoplankton occurred near the surface during thermally stratified periods, but were distributed at greater depths during nonthermally stratified periods. Blue-green algae were present only in the spring samples, and reached a maximum concentration of about 5 million cells per litre.Zooplankton numbers were greatest in March, July, and September, with lesser concentrations in June. Three major zooplankton groups, Cladocera, Copepoda, and Rotifera, were present. The major groups of benthic organisms were Oligochaeta, Chironomidae, and Chaoborus, with numbers ranging from 3350, 890, and 8450 per square metre, respectively.A discussion on algal control is included.

  2. Moderately lower temperatures greatly extend the lifespan of Brachionus manjavacas (Rotifera): thermodynamics or gene regulation?

    Environmental temperature greatly affects lifespan in a wide variety of animals, but the exact mechanisms underlying this effect are still largely unknown. A moderate temperature decrease from 22°C to 16°C extends the lifespan of the monogonont rotifer Brachionus manjavacas by up to 163%. Thermodynamic effects on metabolism contribute to this increase in longevity, but are not the only cause. When rotifers are exposed to 16°C for four days and then transfered to 22°C, they survive until day 13 at nearly identical rates as rotifers maintained at 16°C continuously. This persistence of the higher survival for nine days after transfer to 22°C suggests that low temperature exposure alters the expression of genes that affect the rate of aging. The relative persistence of the gene regulation effect suggests that it may play an even larger role in slowing aging than the thermodynamic effects. The life extending effects of these short-term low temperature treatments are largest when the exposure happens early in the life cycle, demonstrating the importance of early development. There is no advantage to lowering the temperature below 16°C to 11° or 5°C. Rotifers exposed to 16°C also displayed increased resistance to heat, starvation, oxidative and osmotic stress. Reproductive rates at 16°C were lower than those at 22°C, but because they reproduce longer, there is no significant change in the lifetime fecundity of females. To investigate which genes contribute to these effects, the expression of specific temperature sensing genes was knocked down using RNAi. Of 12 genes tested, RNAi knockdown of four eliminated the survival enhancing effects of the four-day cold treatment: TRP7, forkhead box C, Y-box factor, and ribosomal protein S6. This demonstrates that active gene regulation is an important factor in temperature mediated life extension, and that these particular genes play an integral role in these pathways. As a thermoresponsive sensor, TRP7 may be responsible for triggering the signaling cascade contributing to temperature mediated life extension. The TRP genes may also provide especially promising candidates for targeted gene manipulations or pharmacological interventions capable of mimicking the effects of low temperature exposure. These results support recent theories of aging that claim rate of aging is determined by an actively regulated genetic mechanism rather than an accumulation of molecular damage. PMID:26939542

  3. Joint inhibition of TOR and JNK pathways interacts to extend the lifespan of Brachionus manjavacas (Rotifera)

    The TOR kinase pathway is central in modulating aging in a variety of animal models. The target of rapamycin (TOR) integrates a complex network of signals from growth conditions, nutrient availability, energy status, and physiological stresses and matches an organism’s growth rate to the resource environment. Important problems remaining are to identify the pathways that interact with TOR and characterize them as additive or synergistic. One of the most versatile stress sensors in metazoans is the Jun-N-terminal Kinase (JNK) signalling pathway. JNK is an evolutionarily conserved stress-activated protein kinase that is induced by a range of stressors, including UV irradiation, reactive oxygen species, DNA damage, heat, and bacterial antigens. JNK is thought to interact with the TOR pathway, but its effects on TOR are poorly understood. We used the rotifer Brachionus manjavacas as a model animal to probe the regulation of TOR and JNK pathways and explore their interaction. The effect of various chemical inhibitors was examined in life table and stressor challenge experiments. A survey of 12 inhibitors revealed two, rapamycin and JNK inhibitor, that significantly extended lifespan of B. manjavacas. At 1 μM concentration, exposure to rapamycin or JNK inhibitor extended mean rotifer lifespan by 35% and maximum lifespan by 37%. Exposure to both rapamycin and JNK inhibitor simultaneously extended mean rotifer lifespan 65% more than either alone. Exposure to a combination of rapamycin and JNK inhibitors conveyed greater protection to starvation, UV and osmotic stress than either inhibitor alone. RNAi knockdown of TOR and JNK gene expression was investigated for its ability to extend rotifer lifespan. RNAi knockdown of the TOR gene resulted in 29% extension of mean lifespan compared to control and knockdown of the JNK gene resulted in 51% mean lifespan extension. In addition to lifespan, we quantified mitochondria activity using the fluorescent marker Mitotracker and lysosome activity using Lysotracker. Treatment of rotifers with JNK inhibitor enhanced mitochondria activity nearly 3-fold, whereas rapamycin treatment had no significant effect. Treatment of rotifers with rapamycin or JNK inhibitor reduced lysosome activity in 1, 3 and 8 day old animals, but treatment with both inhibitors did not produce any additive effect. We conclude that inhibition of TOR and JNK pathways significantly extends the lifespan of B. manjavacas. These pathways interact so that inhibition of both simultaneously acts additively to extend rotifer lifespan more than inhibition of either alone. PMID:24486130

    Background Mate choice is of central importance to most animals, influencing population structure, speciation, and ultimately the survival of a species. Mating behavior of male brachionid rotifers is triggered by the product of a chemosensory gene, a glycoprotein on the body surface of females called the mate recognition pheromone. The mate recognition pheromone has been biochemically characterized, but little was known about the gene(s). We describe the isolation and characterization of the mate recognition pheromone gene through protein purification, N-terminal amino acid sequence determination, identification of the mate recognition pheromone gene from a cDNA library, sequencing, and RNAi knockdown to confirm the functional role of the mate recognition pheromone gene in rotifer mating. Results A 29 kD protein capable of eliciting rotifer male circling was isolated by high-performance liquid chromatography. Two transcript types containing the N-terminal sequence were identified in a cDNA library; further characterization by screening a genomic library and by polymerase chain reaction revealed two genes belonging to each type. Each gene begins with a signal peptide region followed by nearly perfect repeats of an 87 to 92 codon motif with no codons between repeats and the final motif prematurely terminated by the stop codon. The two Type A genes contain four and seven repeats and the two Type B genes contain three and five repeats, respectively. Only the Type B gene with three repeats encodes a peptide with a molecular weight of 29 kD. Each repeat of the Type B gene products contains three asparagines as potential sites for N-glycosylation; there are no asparagines in the Type A genes. RNAi with Type A double-stranded RNA did not result in less circling than in the phosphate-buffered saline control, but transfection with Type B double-stranded RNA significantly reduced male circling by 17%. The very low divergence between repeat units, even at synonymous positions, suggests that the repeats are kept nearly identical through a process of concerted evolution. Information-rich molecules like surface glycoproteins are well adapted for chemical communication and aquatic animals may have evolved signaling systems based on these compounds, whereas insects use cuticular hydrocarbons. Conclusion Owing to its critical role in mating, the mate recognition pheromone gene will be a useful molecular marker for exploring the mechanisms and rates of selection and the evolution of reproductive isolation and speciation using rotifers as a model system. The phylogenetic variation in the mate recognition pheromone gene can now be studied in conjunction with the large amount of ecological and population genetic data being gathered for the Brachionus plicatilis species complex to understand better the evolutionary drivers of cryptic speciation.
