An interferometric imaging biosensor using weighted spectrum analysis to confirm DNA monolayer films with attogram sensitivity.

Interferometric imaging biosensors are powerful and convenient tools for confirming the existence of DNA monolayer films on silicon microarray platforms. However, their accuracy and sensitivity need further improvement because DNA molecules contribute to an inconspicuous interferometric signal both in thickness and size. Such weaknesses result in poor performance of these biosensors for low DNA content analyses and point mutation tests. In this paper, an interferometric imaging biosensor with weighted spectrum analysis is presented to confirm DNA monolayer films. The interferometric signal of DNA molecules can be extracted and then quantitative detection results for DNA microarrays can be reconstructed. With the proposed strategy, the relative error of thickness detection was reduced from 88.94% to merely 4.15%. The mass sensitivity per unit area of the proposed biosensor reached 20 attograms (ag). Therefore, the sample consumption per unit area of the target DNA content was only 62.5 zeptomoles (zm), with the volume of 0.25 picolitres (pL). Compared with the fluorescence resonance energy transfer (FRET), the measurement veracity of the interferometric imaging biosensor with weighted spectrum analysis is free to the changes in spotting concentration and DNA length. The detection range was more than 1µm. Moreover, single nucleotide mismatch could be pointed out combined with specific DNA ligation. A preliminary assessment of a medium-enthalpy geothermal resource in Nagu (Tibet) people's republic of China

The Nagqu geothermal field is a single-phase, liquid-dominated system at reservoir conditions, having a high gas content. This field is located at an elevation of about 4,500 m (asl), in the vicinity of the City of Nagqu, which is one of the most important cities of Tibet.The reservoir rock is made of a highly fractured, low-permeability sedimentary sequence. During the implementation of the study described in this paper, fluid production was mainly obtained from two out of four possible productive wells. The main fault systems are located in a NE-SW and E-W directions, which seem to control fluid movement atmore » depth. The geothermal field is restricted to a small area where hydrothermal manifestations are located. Reservoir temperature is 114 C, gas content is in the range of 0.5 to 0.6% by mass, being mainly CO{sub 2}. Reservoir transmissivity in the area of the wells is very high. Reservoir response to changes in flow rate in any of the producing wells could be detected almost immediately in the observation wells, which were distant between 300 to 900 m, depending on the production-observation well arrangement. Calcium carbonate scaling was present in all producing wells. This deposition was controlled by the CO{sub 2} partial pressure. Description of well testing results is provided, as well as the thermodynamics and geochemistry of reservoir fluids.« less

Clarifying functional roles: algal removal by the surgeonfishes Ctenochaetus striatus and Acanthurus nigrofuscus

The lined bristletooth, Ctenochaetus striatus, and the brown surgeonfish, Acanthurus nigrofuscus, are among the most abundant surgeonfishes on Indo-Pacific coral reefs. Yet, the functional role of these species has been the focus of an ongoing debate lasting at least six decades. Specifically, to what extent are C. striatus herbivorous like the visually similar A. nigrofuscus? To address this question, we used natural feeding surfaces, covered with late successional stage reef-grown algal turfs, to examine turf algal removal by the two species. Surfaces exposed to C. striatus in laboratory experiments exhibited no significant reductions in turf length or area covered by turfing algae. In marked contrast, A. nigrofuscus reduced turf length by 51% and area covered by turfing algae by 15% in 1 h. The gut contents of specimens from the reef revealed that A. nigrofuscus predominantly ingests algae (the dominant item in 79.6-94.7% of gut content quadrats), while C. striatus ingests detritus and sediments (dominant in 99.6-100% of quadrats). Therefore, C. striatus ingests detritus and sediment, leaving mature algal turfs relatively intact, while A. nigrofuscus directly removes and ingests turf algae. The function of C. striatus differs from cropping herbivorous surgeonfishes such as A. nigrofuscus. On coral reefs, C. striatus brush detrital aggregates from algal turfs, removing microorganisms, organic detritus and inorganic sediment. Confusion over the functional role of C. striatus may stem from an inability to fit it into a single functional category.

Predicting Regulatory Compliance in Beer Advertising on Facebook.

The prevalence of alcohol advertising has been growing on social media platforms. The purpose of this study was to evaluate alcohol advertising on Facebook for regulatory compliance and thematic content. A total of 50 Budweiser and Bud Light ads posted on Facebook within 1 month of the 2015 NFL Super Bowl were evaluated for compliance with a self-regulated alcohol advertising code and for thematic content. An exploratory sensitivity/specificity analysis was conducted to determine if thematic content could predict code violations. The code violation rate was 82%, with violations prevalent in guidelines prohibiting the association of alcohol with success (Guideline 5) and health benefits (Guideline 3). Overall, 21 thematic content areas were identified. Displaying the product (62%) and adventure/sensation seeking (52%) were the most prevalent. There was perfect specificity (100%) for 10 content areas for detecting any code violation (animals, negative emotions, positive emotions, games/contests/promotions, female characters, minorities, party, sexuality, night-time, sunrise) and high specificity (>80%) for 10 content areas for detecting violations of guidelines intended to protect minors (animals, negative emotions, famous people, friendship, games/contests/promotions, minorities, responsibility messages, sexuality, sunrise, video games). The high prevalence of code violations indicates a failure of self-regulation to prevent potentially harmful content from appearing in alcohol advertising, including explicit code violations (e.g. sexuality). Routine violations indicate an unwillingness to restrict advertising content for public health purposes, and statutory restrictions may be necessary to sufficiently deter alcohol producers from repeatedly violating marketing codes. Violations of a self-regulated alcohol advertising code are prevalent in a sample of beer ads published on Facebook near the US National Football League's Super Bowl. Overall, 16 thematic content areas demonstrated high specificity for code violations. Alcohol advertising codes should be updated to expressly prohibit the use of such content. © The Author 2017. Mapping soil magnetic susceptibility and mineralogy in Ukraine

Soil suatainable planning is fundamental for agricultural areas. Soil mapping and modeling are increasingly used in agricultural areas in the entire world (Brevik et al., 2016). They are beneficial to land managers, to reduce soil degradation, increase soil productivity and their restoration. Magnetic susceptibility (MS) methods are low cost and accurate for the developing maps of agricultural areas.. The objective of this work is to identify the minerals responsible for MS increase in soils from the two study areas in Poltava and Kharkiv region. The thermomagnetic analyses were conducted using the KLY-4 with an oven apparatus. The hysteresis parameters were measured with the Rotating Magnetometer at the Geophysical Centre Dourbes, Belgium. The results showed that all of samples from Kharkiv area and the majortity of the samples collected in Poltava area represent the pseudo single domain (PSD) zone particles in Day plot. According to Hanesch et al. (2006), the transformation of goethite, ferrihydrite or hematite to a stronger ferrimagnetic phase like magnetite or maghemite is common in strongly magnetic soils with high values of organic carbon content. In our case of thermomagnetic study, the first peak on the heating curve near 260 ˚C indicates the presence of ferrihydrite which gradually transforms into maghemite (Jordanova et al., 2013). A further decrease in the MS identified on the heating curve may be related to the transformation of the maghemite to hematite. A second MS peak on the heating curve near 530 ˚C and the ultimate loss of magnetic susceptibility near 580 ˚C were caused by the reduction of hematite to magnetite. The shape of the thermomagnetic curves suggests the presence of single domain (SD) particles at room temperature and their transformation to a superparamagnetic (SP) state under heating. Magnetic mineralogical analyses suggest the presence of highly magnetic minerals like magnetite and maghemite as well as slightly magnetic goethite, ferrihydrite, and hematite. Pseudosingle-domain, single-domain, and superparamagnetic grains of pedogenic origin dominate in the chernozem soils of the Kharkiv and Poltava region. References Brevik, E. Seasonal changes of the mineral contents in the rumen of wild Yeso sika deer (Cervus nippon yesoensis).

The rumen contents were collected from 36 wild Yeso sika deer (Cervus nippon yesoensis) captured by deer culling or by hunting in the spring, summer, autumn and winter in Hokkaido, Japan. Botanical classification was conducted, and the contents of mineral (calcium (Ca), phosphorus (P), potassium (K), sodium (Na), iron (Fe), copper (Cu) and zinc (Zn)) were measured. The animals were captured around pastures or fallow field areas in the Kushiro area. The rumen contents consisted of grasses and Sasa sp. leaves regardless of the season. Leaves and bark were ingested in the spring, autumn and winter. The macro-mineral contents in the rumen showed seasonal changes. In the summer, the Ca, K and P contents were high, and the Na content was low. There were no seasonal changes in the Fe content. The P, Na and Fe contents were higher than the animals' requirements. Quantitative determination of carbonaceous particle mixing state in Paris using single-particle mass spectrometer and aerosol mass spectrometer measurements

Single-particle mixing state information can be a powerful tool for assessing the relative impact of local and regional sources of ambient particulate matter in urban environments. However, quantitative mixing state data are challenging to obtain using single-particle mass spectrometers. In this study, the quantitative chemical composition of carbonaceous single particles has been determined using an aerosol time-of-flight mass spectrometer (ATOFMS) as part of the MEGAPOLI 2010 winter campaign in Paris, France. Relative peak areas of marker ions for elemental carbon (EC), organic aerosol (OA), ammonium, nitrate, sulfate and potassium were compared with concurrent measurements from an Aerodyne high-resolution time-of-flight aerosol mass spectrometer (HR-ToF-AMS), a thermal-optical OCEC analyser and a particle into liquid sampler coupled with ion chromatography (PILS-IC). ATOFMS-derived estimated mass concentrations reproduced the variability of these species well (R2 = 0.67-0.78), and 10 discrete mixing states for carbonaceous particles were identified and quantified. The chemical mixing state of HR-ToF-AMS organic aerosol factors, resolved using positive matrix factorisation, was also investigated through comparison with the ATOFMS dataset. The results indicate that hydrocarbon-like OA (HOA) detected in Paris is associated with two EC-rich mixing states which differ in their relative sulfate content, while fresh biomass burning OA (BBOA) is associated with two mixing states which differ significantly in their OA / EC ratios. Aged biomass burning OA (OOA2-BBOA) was found to be significantly internally mixed with nitrate, while secondary, oxidised OA (OOA) was associated with five particle mixing states, each exhibiting different relative secondary inorganic ion content. Externally mixed secondary organic aerosol was not observed. These findings demonstrate the range of primary and secondary organic aerosol mixing states in Paris. Examination of the temporal behaviour and chemical composition of the ATOFMS classes also enabled estimation of the relative contribution of transported emissions of each chemical species and total particle mass in the size range investigated. Only 22% of the total ATOFMS-derived particle mass was apportioned to fresh, local emissions, with 78% apportioned to regional/continental-scale emissions.

The foliar and biochemical traits of Azadirachta indica A. Juss from fly ash (FA) dumping site in Badarpur thermal power plant (BTPP) New Delhi, India was studied. Three different experimental sites were selected at different distances from the thermal power plant. Ambient suspended particulate matter (SPM) and plant responses such as leaf pigments (chlorophyll a, chlorophyll b, and carotenoids), total chlorophyll, net photosynthetic rate, stomatal index (SI), stomatal conductance (SC), intercellular carbon dioxide concentration [CO2]i, net photosynthetic rate (NPR), nitrogen, nitrate, nitrate reductase activity, proline, protein, reducing sugar and sulphur content were measured. Considerable reduction in pigments (chlorophyll a, chlorophyll b and carotenoids), and total chlorophyll was observed at fly ash dumping site. Fly ash stress revealed the inhibitory effect on Nitrate reductase activity (NRA), Nitrate, soluble protein, and reducing sugar content, whereas stimulatory effect was found for the stomatal index, nitrogen, proline, antioxidants and sulphur content in the leaves. Under fly ash stress, stomatal conductance was low, leading to declining in photosynthetic rate and increase in the internal CO2 concentration of leaf. Single leaf area (SLA), leaf length and leaf width also showed a declining trend from control to the polluted site. Microplankton trace element contents: implications for mineral limitation of mesozooplankton in an HNLC area

Mesozooplankton production in high-nutrient low-chlorophyll regions of the ocean may be reduced if the trace element concentrations in their food are insufficient to meet growth and metabolic demands. We used elemental microanalysis (SXRF) of single-celled plankton to determine their trace metal contents during a series of semi-Lagrangian drift studies in an HNLC upwelling region, the Costa Rica Dome (CRD). Cells from the surface mixed layer had lower Fe:S but higher Zn:S and Ni:S than those from the subsurface chlorophyll maximum at 22–30 m. Diatom Fe:S values were typically 3-fold higher than those in flagellated cells. The ratios of Zn:C in flagellates and diatoms were generally similar to each other, and to co-occurring mesozooplankton. Estimated Fe:C ratios in flagellates were lower than those in co-occurring mesozooplankton, sometimes by more than 3-fold. In contrast, Fe:C in diatoms was typically similar to that in zooplankton. RNA:DNA ratios in the CRD were low compared with other regions, and were related to total autotrophic biomass and weakly to the discrepancy between Zn:C in flagellated cells and mesozooplankton tissues. Mesozooplankton may have been affected by the trace element content of their food, even though trace metal limitation of phytoplankton was modest at best. PMID:27275029

To describe the activities performed by people involved in clinical decision support (CDS) at leading sites. We conducted ethnographic observations at seven diverse sites with a history of excellence in CDS using the Rapid Assessment Process and analyzed the data using a series of card sorts, informed by Linstone's Multiple Perspectives Model. We identified 18 activities and grouped them into four areas. Area 1: Fostering relationships across the organization, with activities (a) training and support, (b) visibility/presence on the floor, (c) liaising between people, (d) administration and leadership, (e) project management, (f) cheerleading/buy-in/sponsorship, (g) preparing for CDS implementation. Area 2: Assembling the system with activities (a) providing technical support, (b) CDS content development, (c) purchasing products from vendors (d) knowledge management, (e) system integration. Area 3: Using CDS to achieve the organization's goals with activities (a) reporting, (b) requirements-gathering/specifications, (c) monitoring CDS, (d) linking CDS to goals, (e) managing data. Area 4: Participation in external policy and standards activities (this area consists of only a single activity). We also identified a set of recommendations associated with these 18 activities. All 18 activities we identified were performed at all sites, although the way they were organized into roles differed substantially. We consider these activities critical to the success of a CDS program. A series of activities are performed by sites strong in CDS, and sites adopting CDS should ensure they incorporate these activities into their efforts.

Single trial discrimination of individual finger movements on one hand: A combined MEG and EEG study☆

It is crucial to understand what brain signals can be decoded from single trials with different recording techniques for the development of Brain-Machine Interfaces. A specific challenge for non-invasive recording methods are activations confined to small spatial areas on the cortex such as the finger representation of one hand. Here we study the information content of single trial brain activity in non-invasive MEG and EEG recordings elicited by finger movements of one hand. We investigate the feasibility of decoding which of four fingers of one hand performed a slight button press. With MEG we demonstrate reliable discrimination of single button presses performed with the thumb, the index, the middle or the little finger (average over all subjects and fingers 57%, best subject 70%, empirical guessing level: 25.1%). EEG decoding performance was less robust (average over all subjects and fingers 43%, best subject 54%, empirical guessing level 25.1%). Spatiotemporal patterns of amplitude variations in the time series provided best information for discriminating finger movements. Non-phase-locked changes of mu and beta oscillations were less predictive. Movement related high gamma oscillations were observed in average induced oscillation amplitudes in the MEG but did not provide sufficient information about the finger's identity in single trials. Importantly, pre-movement neuronal activity provided information about the preparation of the movement of a specific finger. Our study demonstrates the potential of non-invasive MEG to provide informative features for individual finger control in a Brain-Machine Interface neuroprosthesis. PMID:22155040

Dental fluorosis in children in areas with fluoride-polluted air, high-fluoride water, and low-fluoride water as well as low-fluoride air: a study of deciduous and permanent teeth in the Shaanxi province, China.

The aim of the study was to assess dental fluorosis (DF) in the deciduous and permanent teeth of children in areas with high-F coal (area A) and high-F water (area C) compared to children from area B, with low-F water and coal. 596 children were examined. DF was assessed by TF-score. F-content of indoor air, drinking water, coal, tea, rice, and maize was analyzed. F-content of air and coal ranged from 3.2 microg/m(3) and 25.8 mg/kg (area B), 3.8 microg/m(3) and 36.3 mg/kg (area C) to 56.8 microg/m(3) and 713.1 mg/kg (area A). Likewise, mean F-content of water ranged from approximately 0.50 mg/l (areas A and B) to 3.64 mg/l (area C). F-content of tea leaves was similar in all three areas. Maize and rice contained <5 mg F/kg. Prevalence of primary teeth with DF was 49.1%, 2.0%, and 66.8% in areas A, B, and C, respectively. Similarly, DF was found in 96.7% (area A), 19.6% (area B), and 94.4% (area C) of the permanent teeth. Severe fluorosis (TF > or = 5) was found in area A (47.0%) and area C (36.1%) (p<0.01). Early erupting teeth had slightly higher mean TF-scores in area A than in area C. DF was prevalent in both dentitions in areas A and C. Similarity in percentages of DF may indicate that indoor air with approximately 60 microg F/m(3) and drinking water with 3.6 mg F/L are similarly toxic to developing permanent teeth. The percentage of deciduous teeth with DF was significantly lower in area A compared to area C. Where low-F coal and low-F water were used (area B), approximately 20% of permanent teeth had DF, indicating a relatively low tolerance to fluoride in Chinese children brought up under the present living conditions.

Single cell versus large population analysis: cell variability in elemental intracellular concentration and distribution.

The quantification of elemental concentration in cells is usually performed by analytical assays on large populations missing peculiar but important rare cells. The present article aims at comparing the elemental quantification in single cells and cell population in three different cell types using a new approach for single cells elemental analysis performed at sub-micrometer scale combining X-ray fluorescence microscopy and atomic force microscopy. The attention is focused on the light element Mg, exploiting the opportunity to compare the single cell quantification to the cell population analysis carried out by a highly Mg-selective fluorescent chemosensor. The results show that the single cell analysis reveals the same Mg differences found in large population of the different cell strains studied. However, in one of the cell strains, single cell analysis reveals two cells with an exceptionally high intracellular Mg content compared with the other cells of the same strain. The single cell analysis allows mapping Mg and other light elements in whole cells at sub-micrometer scale. A detailed intensity correlation analysis on the two cells with the highest Mg content reveals that Mg subcellular localization correlates with oxygen in a different fashion with respect the other sister cells of the same strain. Influence of molybdenum on the creep properties of nickel-base superalloy single crystals

The Mo content of an alloy series based on Ni-6 wt pct Al-6 wt pct Ta was systematically varied from 9.8 to 14.6 wt pct, in order to ascertain the influence of Mo on the creep properties of single crystals. The optimum initial gamma-gamma prime microstructure for raft development and creep strength was established in each alloy before testing. It was found that, as the Mo content increased from 9.8 to 14.0 percent, the magnitude of the lattice mismatch increased; upon reaching 14.6 percent, a degradation of mechanical properties occurred due to the precipitation of a third phase. These results suggest that small refractory metal content and initial gamma-prime variations can profoundly affect mechanical properties.

Information Content of Aerosol Retrievals in the Sunglint Region

We exploit quantitative metrics to investigate the information content in retrievals of atmospheric aerosol parameters (with a focus on single-scattering albedo), contained in multi-angle and multi-spectral measurements with sufficient dynamical range in the sunglint region. The simulations are performed for two classes of maritime aerosols with optical and microphysical properties compiled from measurements of the Aerosol Robotic Network. The information content is assessed using the inverse formalism and is compared to that deriving from observations not affected by sunglint. We find that there indeed is additional information in measurements containing sunglint, not just for single-scattering albedo, but also for aerosol optical thickness and the complex refractive index of the fine aerosol size mode, although the amount of additional information varies with aerosol type.
