Embedded turbulence model in numerical methods for hyperbolic conservation laws

The paper describes the use of numerical methods for hyperbolic conservation laws as an embedded turbulence modelling approach. Different Godunov-type schemes are utilized in computations of Burgers' turbulence and a two-dimensional mixing layer. The schemes include a total variation diminishing, characteristic-based scheme which is developed in this paper using the flux limiter approach. The embedded turbulence modelling property of the above methods is demonstrated through coarsely resolved large eddy simulations with and without subgrid scale models. Copyright
EDITORIAL: Special Issue on Light Sources

The papers in this Special Issue of Journal of Physics D: Applied Physics originate from the 11th International Symposium on the Science and Technology of Light Sources (LS:11) held at Fudan University, Shanghai, China, during 20 24 May 2007. Abstracts of all papers were published in the conference book Light Sources 2007 (Sheffield: FAST-LS) edited by Muqing Liu and R Devonshire. Special issues were produced after LS:9 and LS:10 and have proved to be well-cited and important sources of information for this community. The Symposia occur at three-year intervals. In this one over 200 papers were presented—the majority as posters—with ample time provided for active discussion. As all submitted papers had to be refereed in the normal way for J. Phys. D: Appl. Phys., I was concerned that too many submissions would overwhelm the small number of referees available in this area. To ensure a broad spread of interests and opinions, I invited 10 senior colleagues to give me their recommendations about who should be asked to submit papers for this Special Issue. The criteria were that the work should be new, complete and within the scope of the journal. As a result of their suggestions 42 authors were asked to submit papers. Not all authors were able to submit a manuscript in time and some, at my request, combined their work into a single paper. The 28 papers published here are the result of that process. The issue starts with a comprehensive review by Benilov of the remarkable progress that has been made in the past 15 years in understanding the behaviour of cathode and anode terminations in arcs. It is fair to say that we now have a fundamental understanding of the formerly baffling behaviour of spot and diffuse terminations, at least in the quasi-steady state. A number of following papers cover applications of this theory, extensions to time dependence and examination of the effects of the different gaseous atmospheres in which lighting arcs operate. Mercury has very great advantages for efficient discharge lighting, including high volatility, high electron collision cross-section and its 3P levels, which are a major factor in the high efficiency generation of UV. Unfortunately, it is somewhat toxic and therefore unwelcome in the environment. Most of the mercury used in lighting is in fluorescent lamps and these produce (at high efficiency) most of the artificial light on this planet. Finding an alternative is proving a difficult but nevertheless active research field (see the recent Special Cluster on Mercury-Free Discharges for Lighting, edited by M Haverlag, in J. Phys. D: Appl. Phys. volume 40, issue 13). The other approach is to reduce the amount of mercury in each lamp. Much has already been done but pressure for further reduction continues; Corrazza and Boffito review the current status. Developments in the behaviour of fluorescent lamp electrodes and in the ignition of fluorescent lamps are amongst the papers presented on this most important discharge. Major advances in the performance of LEDs have been made in recent years, although currently most LED lighting is used for signalling and decorative lighting; this is because LEDs cannot yet produce the light levels needed for general illumination at costs and efficiencies that compete with discharge lamps. Because of their specialized technology, papers from this area are somewhat under-represented in this issue. Papers on OLEDs and the specialized field of LED measurement are included. HID (high intensity discharge) lamps operate close to LTE at high temperatures and pressures in corrosive atmospheres. Research on materials has been very important for their development and is represented here in two papers. Diagnostics of HID have been crucial to securing agreement between models and experiment in HID lamps. Two papers are presented here on the spectroscopy of self-reversed lines, which provide very reliable results in lamps that typically have relatively poor optical quality. These are followed by further papers on emission spectroscopy. Amongst other diagnostics, the use of x-ray methods has proved of great importance in lamps with diffusing ceramic envelopes. Developments and applications of this technique are reported in three papers. Comparison of model and experiment on the behaviour of metal-halide and mercury HID arcs is covered in further papers with particular emphasis on instabilities and the interaction of diffusion and convection in the flows. The understanding of breakdown and ignition of HID lamps—so critical to reliable operation—has made substantial progress as exemplified by the final two papers. Many other interesting posters on these subjects and others were presented at the LS:11 meeting. I hope when some of this work is brought to a conclusion that it will also be published. Discharge lamps still provide the best combination of high luminous flux and efficiency for illumination. In the next few years it will be interesting to see how strongly this position will be challenged by the rapidly improving solid state light sources. My task in preparing this Special Issue would have been impossible without the help of my unnamed colleagues who spent considerable time and energy in short-listing possible papers; they know who they are and I am extremely grateful to them.

The purpose of this study was to summarize the historical changes in the list of plasma fractionation products (PFP) placed on the Model List of Essential Medicines (EML) issued by the World Health Organization (WHO). PFP such as albumin, blood coagulation factors, and immunoglobulins are derived from blood collected from thousands of people. PFP have been listed since the first edition of the EML (1977). However, the PFP listed on the EML have changed dramatically because EML's selection process has changed from experience-based to evidence-based. For example, albumin, which had been listed since the 2nd edition (1979), was deleted in the 11th edition (2000) because of the uncertainty of its efficacy. Human immunoglobulin normal, which had been deleted from the 13th edition (2003), was relisted in the 15th edition (2007). Moreover, the WHO has issued several resolutions and guidelines regarding PFP production, quality, and safety in order to promote the establishment of blood programmes in every nation. The focus of WHO's EML selection process has changed over 30 years. In the 20th century, WHO mainly focused on PFP efficacy, quality, and safety problems. However, currently the focus is on the problem of PFP accessibility, especially in developing countries. Therefore, it would be important to know how to capitalize on established knowledge and production technology to increase the accessibility of PFP worldwide.

The importance of developing a better understanding of positive emotion regulation in both healthy and clinical populations is now recognised. This special edition brings together leading figures in the positive emotion regulation field and has contributions characterizing positive phenomena, differentiating them from negative phenomena, and evaluating underlying psychological mechanisms that drive these phenomena. This commentary reviews these articles to highlight challenges and opportunities for this emerging field, including the need to better characterize positive phenomena, to be more explicit about how the links between negative and positive phenomena are conceptualised, to evaluate more robustly underlying mechanisms, to standardize measurement of positive constructs, and to ensure that these scientific findings lead to meaningful changes in real-world policy and practice.

A major cause of visual disorders is dysfunction and/or loss of the light-sensitive cells of the retina, the photoreceptors. To develop better treatments for patients, we need to understand how inherited retinal disease mutations result in the dysfunction of photoreceptors. New advances in the field of stem cell and gene editing research offer novel ways to model retinal dystrophies in vitro and present opportunities to translate basic biological insights into therapies. This brief review will discuss some of the issues that should be taken into account when carrying out disease modelling and gene editing of retinal cells. We will discuss (i) the use of human induced pluripotent stem cells (iPSCs) for disease modelling and cell therapy; (ii) the importance of using isogenic iPSC lines as controls; (iii) CRISPR/Cas9 gene editing of iPSCs; and (iv) in vivo gene editing using AAV vectors. Ground-breaking advances in differentiation of iPSCs into retinal organoids and methods to derive mature light sensitive photoreceptors from iPSCs. Furthermore, single AAV systems for in vivo gene editing have been developed which makes retinal in vivo gene editing therapy a real prospect. Genome editing is becoming a valuable tool for disease modelling and in vivo gene editing in the retina.

Book Review: Reconstructing Quaternary Environments

A third edition of this, the foremost Quaternary textbook, is most welcome, coming seventeen years after the 1997 second edition (which was 13 years after the first). The general impression is one of advancement, not least because of the extensive updating of literature cited and examples used, with the status maintained of an impressive compendium of a specialism with a very wide subject base. Some changes are cosmetic, with chapter and section headers having a more modern style and a profusion of new colour photographs and diagrams. Some of the latter are redrawn from black and white figures in the previous edition, although not all have been improved, as some are smaller and have been simplified. For example, black and white Fig. 3.10 of the Second Edition compares very favourably with the much smaller colour 3.17 in this latest volume (erratic sources). On the plus side, the number change, for a figure that appears in the same place within the chapter, shows that the latest edition is considerably better illustrated than its predecessor, perhaps accounting for a significant proportion of the increased page total (up from 446 to 538).

We present the regulatory framework for embryo research in the UK, and explain why CRISPR is not a significant break with the past. We discuss the ethical issues arising from CRISPR applications beyond human embryos, namely the use of gene drive-engineered mosquitoes to eradicate diseases, engineering nonhuman animals to harvest organs for human transplant and engineering crops. We discuss the experiments that have demonstrated the technical feasibility of cultivating embryos in vitro for up to 14 days, and possibly beyond this limit, and the ethical issues arising from the proposal to extend the limit beyond 14 days. The Clustered Regularly Interspaced Short Palindromic Repeat (CRISPR) and CRISPR-associated protein (Cas) system has enabled an accurate and efficient means to edit the human genome. Rapid advances in this technology could results in imminent clinical application, and with favourable anatomical and immunological profiles, ophthalmic disease will be at the forefront of such work. There have been a number of breakthroughs improving the specificity and efficacy of CRISPR/Cas-mediated genome editing. Similarly, better methods to identify off-target cleavage sites have also been developed. With the impending clinical utility of CRISPR/Cas technology, complex ethical issues related to the regulation and management of the precise applications of human gene editing must be considered. This review discusses the current progress and recent breakthroughs in CRISPR/Cas-based gene engineering, and outlines some of the technical issues that must be addressed before gene correction, be it in vivo or in vitro, is integrated into ophthalmic care. We outline a clinical pipeline for CRISPR-based treatments of inherited eye diseases and provide an overview of the important ethical implications of gene editing and how these may influence the future of this technology. Genome editing in livestock: Are we ready for a revolution in animal breeding industry?

Genome editing is a powerful technology that can efficiently alter the genome of organisms to achieve targeted modification of endogenous genes and targeted integration of exogenous genes. Current genome-editing tools mainly include ZFN, TALEN and CRISPR/Cas9, which have been successfully applied to all species tested including zebrafish, humans, mice, rats, monkeys, pigs, cattle, sheep, goats and others. The application of genome editing has quickly swept through the entire biomedical field, including livestock breeding. Traditional livestock breeding is associated with rate limiting issues such as long breeding cycle and limitations of genetic resources. Genome editing tools offer solutions to these problems at affordable costs. Generation of gene-edited livestock with improved traits has proven feasible and valuable. For example, the CD163 gene-edited pig is resistant to porcine reproductive and respiratory syndrome (PRRS, also referred to as "blue ear disease"), and a SP110 gene knock-in cow less susceptible to tuberculosis. Given the high efficiency and low cost of genome editing tools, particularly CRISPR/Cas9, it is foreseeable that a significant number of genome edited livestock animals will be produced in the near future; hence it is imperative to comprehensively evaluate the pros and cons they will bring to the livestock breeding industry. Only with these considerations in mind, we will be able to fully take the advantage of the genome editing era in livestock breeding.
