  1. Identification of a novel inhibitor of isocitrate lyase as a potent antitubercular agent against both active and non-replicating Mycobacterium tuberculosis.

    Screen and identify novel inhibitors of isocitrate lyase (ICL) as potent antitubercular agents against Mycobacterium tuberculosis and determine their inhibitory characteristics, antitubercular activities and mechanisms of action. Recombinant ICL of M. tuberculosis was expressed and purified, which was used for high-throughput screening (HTS) and the following experiments. A total of 71,765 compounds were screened to identify ICL inhibitors which were then evaluated for their roles as potent antitubercular agents. To determine the inhibitory characteristics of the agents against latent M. tuberculosis in persistent infections, a macrophage model (mouse J774A.1 cell) infected with Mycobacterium marinum BAA-535 strain was built and assessed. The potent antitubercular agents were identified using the macrophage model. Then, the inhibitory intensity and mode of the agents that exhibit on ICL protein of M. tuberculosis were analyzed, and the interaction mechanisms were preliminarily clarified according to the parameters of enzyme kinetics, circular dichroism experiments, fluorescence quenching assay, and molecular docking. The previously established ICL inhibitor screening model was evaluated to be suitable for HTS assay. Of the 71,765 compounds, 13 of them were identified to inhibit ICL effectively and stably. IMBI-3 demonstrated the most significant inhibitory activity with IC50 of 30.9 μmol/L. Its minimum inhibitory concentration (MIC) for M. tuberculosis, including extensively drug-resistant tuberculosis (XDR-TB) and multidrug-resistant tuberculosis (MDR-TB), were determined in the range of 0.25-1 μg/mL. When IMBI-3 is used in combination with isoniazid, the colony-forming units (CFU) counting of latent M. tuberculosis in J774A.1 macrophage cells decreased significantly as IMBI-3 concentration increased. This new class of models is constrained by a variety of Antarctic data sets (e.g. proxy reconstructions of past ice extent, GPS estimates of vertical land motion) and provides a correction that is approximately one half to one third of that obtained from the far-field based models. As a consequence, this newer class of models gives Antarctic ice mass balance of approximately -81 ± 33 Gt/yr, or 0.225 ± 0.092 mm/yr contribution to sea-level rise.     Fluctuations in the mass of ice stored in Antarctica and Greenland are of considerable societal importance. The Ice Sheet Mass Balance Inter-Comparison Exercise (IMBIE) is a joint-initiative of ESA and NASA aimed at producing a single estimate of the global sea level contribution to polar ice sheet losses. Within IMBIE, estimates of ice sheet mass balance are developed from a variety of satellite geodetic techniques using a common spatial and temporal reference frame and a common appreciation of the contributions due to external signals. The project brings together the laboratories and space agencies that have been instrumental in developing independent estimates of ice sheet mass balance to date. In its first phase, IMBIE involved 27 science teams, and delivered a first community assessment of ice sheet mass imbalance to replace 40 individual estimates. The project established that (i) there is good agreement between the three main satellite-based techniques for estimating ice sheet mass balance, (ii) combining satellite data sets leads to significant improvement in certainty, (iii) the polar ice sheets contributed 11 ± 4 mm to global sea levels between 1992 and 2012, and (iv) that combined ice losses from Antarctica and Greenland have increased over time, rising from 10% of the global trend in the early 1990's to 30% in the late 2000's. Demand for an updated assessment has grown, and there are now new satellite missions, new geophysical corrections, new techniques, and new teams producing data. The period of overlap between independent satellite techniques has increased from 5 to 12 years, and the full period of satellite data over which an assessment can be performed has increased from 19 to 40 years. It is also clear that multiple satellite techniques are required to confidently separate mass changes associated with snowfall and ice dynamical imbalance - information that is of critical importance for climate modelling.   4. Research of building information extraction and evaluation based on high-resolution remote-sensing imagery

    Building extraction currently is important in the application of high-resolution remote sensing imagery. At present, quite a few algorithms are available for detecting building information, however, most of them still have some obvious disadvantages, such as the ignorance of spectral information, the contradiction between extraction rate and extraction accuracy. The purpose of this research is to develop an effective method to detect building information for Chinese GF-1 data. Firstly, the image preprocessing technique is used to normalize the image and image enhancement is used to highlight the useful information in the image. Secondly, multi-spectral information is analyzed. Subsequently, an improved morphological building index (IMBI) based on remote sensing imagery is proposed to get the candidate building objects. Furthermore, in order to refine building objects and further remove false objects, the post-processing (e.g., the shape features, the vegetation index and the water index) is employed. To validate the effectiveness of the proposed algorithm, the omission errors (OE), commission errors (CE), the overall accuracy (OA) and Kappa are used at final. The proposed method can not only effectively use spectral information and other basic features, but also avoid extracting excessive interference details from high-resolution remote sensing images. Compared to the original MBI algorithm, the proposed method reduces the OE by 33.14% .At the same time, the Kappa increase by 16.09%. In experiments, IMBI achieved satisfactory results and outperformed other algorithms in terms of both accuracies and visual inspection

  6. Upper secondary school students' compliance with two Internet-based self-help programmes: a randomised controlled trial.

    Psychiatric symptoms and stress are on the increase among Swedish adolescents. We aimed to study the potential effect and feasibility of two Internet-based self-help programmes, one mindfulness based (iMBI) and the other music based in a randomised controlled trial that targeted adolescents. A total of 283 upper secondary school students in two Swedish schools were randomised to either a waiting list or one of the two programmes, on their own incentive, on schooltime. General psychiatric health (Symptoms Checklist 90), sleep quality (Pittsburgh Sleep Quality Index), and perceived stress (Perceived Stress Scale) were assessed before and after the interventions. In total, 202 participants answered the questionnaires. Less than 20 logged into each intervention and only 1 performed a full intervention (iMBI). No significant differences in any of the scales were found between those who logged in and those who did not. The potential effect of Internet-based self-help programmes was not possible to examine due to low compliance rates. Adolescents seem to have a very low compliance with Internet-based self-help programmes if left to their own incentive. There were no associations between the psychiatric and stress-related symptoms at baseline and compliance in any of the intervention groups, and no evidence for differences in compliance in relation to the type of programme. Additional studies are needed to examine how compliance rates can be increased in Internet-based self-help mindfulness programmes in adolescents, as the potentially positive effects of mindfulness are partly related to compliance rates.

  7. Variability in Annual and Average Mass Changes in Antarctica from 2004 to 2009 using Satellite Laser Altimetry

    We present a new record of Antarctic ice thickness changes, reconstructed from ICESat laser altimetry observations, from 2004-2009, at over 100,000 locations across the Antarctic Ice Sheet (AIS). This work generates elevation time series at ICESat groundtrack crossover regions on an observation-by-observation basis, with rigorous, quantified, error estimates using the SERAC approach (Schenk and Csatho, 2012). The results include average and annual elevation, volume and mass changes in Antarctica, fully corrected for glacial isostatic adjustment (GIA) and known intercampaign biases; and partitioned into contributions from surficial processes (e.g. firn densification) and ice dynamics. The modular flexibility of the SERAC framework allows for the assimilation of multiple ancillary datasets (e.g. GIA models, Intercampaign Bias Corrections, IBC), in a common framework, to calculate mass changes for several different combinations of GIA models and IBCs and to arrive at a measure of variability from these results. We are able to determine the effect these corrections have on annual and average volume and mass change calculations in Antarctica, and to explore how these differences vary between drainage basins and with elevation. As such, this contribution presents a method that compliments, and is consistent with, the 2012 Ice sheet Mass Balance Inter-comparison Exercise (IMBIE) results (Shepherd 2012). Additionally, this work will contribute to the 2016 IMBIE, which seeks to reconcile ice sheet mass changes from different observations,, including laser altimetry, using a different methodologies and ancillary datasets including GIA models, Firn Densification Models, and Intercampaign Bias Corrections.

    The Ice Sheet Mass Balance Inter-Comparison Exercise (IMBIE) is a community effort, jointly supported by ESA and NASA, that aims to provide a consensus estimate of ice sheet mass balance from satellite gravimetry, altimetry and mass budget assessments, on an annual basis. The project has five experiment groups, one for each of the satellite techniques and two others to analyse surface mass balance (SMB) and glacial isostatic adjustment (GIA). The basic premise for the exercise is that individual ice sheet mass balance datasets are generated by project participants using common spatial and temporal domains to allow meaningful inter-comparison, and this controlled comparison in turn supports aggregation of the individual datasets over their full period. Participation is open to the full community, and the quality and consistency of submissions is regulated through a series of data standards and documentation requirements. The second phase of IMBIE commenced in 2015, with participant data submitted in 2016 and a combined estimate due for public release in 2017. Data from 48 participant groups were submitted to one of the three satellite mass balance technique groups or to the ancillary dataset groups. The individual mass balance estimates and ancillary datasets have been compared and combined within the respective groups. Following this, estimates of ice sheet mass balance derived from the individual techniques were then compared and combined. The result is single estimates of ice sheet mass balance for Greenland, East Antarctica, West Antarctica, and the Antarctic Peninsula. The participants, methodology and results of the exercise will be presented in this paper.

  9. Using satellite laser ranging to measure ice mass change in Greenland and Antarctica

    A least squares inversion of satellite laser ranging (SLR) data over Greenland and Antarctica could extend gravimetry-based estimates of mass loss back to the early 1990s and fill any future gap between the current Gravity Recovery and Climate Experiment (GRACE) and the future GRACE Follow-On mission. The results of a simulation suggest that, while separating the mass change between Greenland and Antarctica is not possible at the limited spatial resolution of the SLR data, estimating the total combined mass change of the two areas is feasible. When the method is applied to real SLR and GRACE gravity series, we find significantly different estimates of inverted mass loss. There are large, unpredictable, interannual differences between the two inverted data types, making us conclude that the current 5×5 spherical harmonic SLR series cannot be used to stand in for GRACE. However, a comparison with the longer IMBIE time series suggests that on a 20-year time frame, the inverted SLR series' interannual excursions may average out, and the long-term mass loss estimate may be reasonable.

    The GRACE mission has done an admirable job of measuring large-scale mass changes over Greenland since its launch in 2002. However before that time, measurements of large-scale ice mass balance were few and far between, leading to a lack of baseline knowledge. High-quality Satellite Laser Ranging (SLR) data existed a decade earlier, but normally has too low a spatial resolution to be used for this purpose. I demonstrate that a least squares inversion technique can reconstitute the SLR data and use it to measure ice loss over Greenland. To do so, I first simulate the problem by degrading today's GRACE data to a level comparable with SLR, then demonstrating that the inversion can re-localize Greenland's contribution to the low-resolution signal, giving an accurate time series of mass change over all of Greenland which compares well with the full-resolution GRACE estimates. I then utilize that method on the actual SLR data, resulting in an independent 1994-2014 time series of mass change over Greenland. I find favorable agreement between the pure-SLR inverted results and the 2012 Ice-sheet Mass Balance Inter-comparison Exercise (IMBIE) results, which are largely based on the "input-output" modeling method before GRACE's launch.

  12. Recent Changes in Ices Mass Balance of the Amundsen Sea Sector

    The glaciers flowing into the Amundsen Sea Embayment (ASE) sector of West Antarctica were confirmed in the Ice Sheet Mass Balance Inter-comparison Exercise (IMBIE) to be the dominant contributors to the current Antarctic ice mass loss, and recently recognized to be undergoing marine ice sheet instability. Here, we investigate their regional ice mass balance using a time series of satellite and airborne data combined with model output products from the Regional Atmospheric and Climate Model (RACMO). Our dataset includes laser altimetry from NASA's ICESat-1 satellite mission and from Operation IceBridge (OIB) airborne surveys, satellite radar altimetry data from ESA's Envisat mission, time-variable gravity data from NASA/DLR's GRACE mission, surface mass balance products from RACMO, ice velocity from a combination of international synthetic aperture radar satellites and ice thickness data from OIB. We find a record of ice mass balance for the ASE where all the analyzed techniques agree remarkably in magnitude and temporal variability. The mass loss of the region has been increasing continuously since 1992, with no indication of a slow down. The mass loss during the common period averaged 91 Gt/yr and accelerated 20 Gt/yr2. In 1992-2013, the ASE contributed 4.5 mm global sea level rise. Overall, our results demonstrate the synergy of multiple analysis techniques for examining Antarctic Ice Sheet mass balance at the regional scale.   13. Partitioned learning of deep Boltzmann machines for SNP data.

    Learning the joint distributions of measurements, and in particular identification of an appropriate low-dimensional manifold, has been found to be a powerful ingredient of deep leaning approaches. Yet, such approaches have hardly been applied to single nucleotide polymorphism (SNP) data, probably due to the high number of features typically exceeding the number of studied individuals. After a brief overview of how deep Boltzmann machines (DBMs), a deep learning approach, can be adapted to SNP data in principle, we specifically present a way to alleviate the dimensionality problem by partitioned learning. We propose a sparse regression approach to coarsely screen the joint distribution of SNPs, followed by training several DBMs on SNP partitions that were identified by the screening. Aggregate features representing SNP patterns and the corresponding SNPs are extracted from the DBMs by a combination of statistical tests and sparse regression. In simulated case-control data, we show how this can uncover complex SNP patterns and augment results from univariate approaches, while maintaining type 1 error control. Time-to-event endpoints are considered in an application with acute myeloid leukemia patients, where SNP patterns are modeled after a pre-screening based on gene expression data. The proposed approach identified three SNPs that seem to jointly influence survival in a validation dataset. This indicates the added value of jointly investigating SNPs compared to standard univariate analyses and makes partitioned learning of DBMs an interesting complementary approach when analyzing SNP data.   14. Final Report: Comparison of the primary (national) standards of low-pressure gas flow

    The EURAMET.M.FF-K6 comparison was organized for the purpose of determination of the degree of equivalence of the primary (national) standards for low-pressure gas flow measurement over the range (2 to 100) m3/h and was performed simultaneously with CCM.FF-K6.2011 with the same transfer standard. A rotary gas meter G65 was used as a transfer standard. The measurements were provided by prescribed reference conditions. Fifteen laboratories from EURAMET participated in this key comparison - SMU, Slovakia; PTB, Germany; CEM, Spain; LNE-LADG, France; VSL, Netherlands; CMI, Czech Republic; BEV, Austria; MKEH, Hungary; GUM Poland; SP, Sweden; METAS, Switzerland; DMDM, Serbia; TUBITAK-UME, Turkey; EIM, Greece; IMBiH, Bosnia-Herzegovina. The EURAMET.M.FF-K6 is linked to the CCM.FF-K6.2011 by correcting the results of three linking laboratories (Slovakia SMU, Germany PTB and France LNE LADG). This correction provides an estimate of what would have been the result from the EURAMET.M-FF-K6 participants, if they had actually participated in CCM.FF-K6.2011. According to the evaluation 93.7 % of the results were consistent with KCRV, 3.4 % of the results were in the warning level and 2.9 % of the results were inconsistent.
