Textured digital elevation models (TDEMs) have valuable use in precision agriculture, situational awareness, and disaster response. However, scientific-quality models are expensive to obtain using conventional aircraft-based methods. The cost of creating an accurate textured terrain model can be reduced by using a low-cost (<$20k) UAV system fitted with ladar and electro-optical (EO) sensors. A texel camera fuses calibrated ladar and EO data upon simultaneous capture, creating a texel image. This eliminates the problem of fusing the data in a post-processing step and enables both 2D- and 3D-image registration techniques to be used. This paper describes formation of TDEMs using simulated data from a small UAV gathering swaths of texel images of the terrain below. Being a low-cost UAV, only a coarse knowledge of position and attitude is known, and thus both 2D- and 3D-image registration techniques must be used to register adjacent swaths of texel imagery to create a TDEM. The process of creating an aggregate texel image (a TDEM) from many smaller texel image swaths is described. The algorithm is seeded with the rough estimate of position and attitude of each capture. Details such as the required amount of texel image overlap, registration models, simulated flight patterns (level and turbulent), and texture image formation are presented. In addition, examples of such TDEMs are shown and analyzed for accuracy.

Formation of a Textured Digital Elevation Model (TDEM) has been useful in many applications in the fields of agriculture, disaster response, terrain analysis and more. Use of a low-cost small UAV system with a texel camera (fused lidar/digital imagery) can significantly reduce the cost compared to conventional aircraft-based methods. This paper reports continued work on this problem reported in a previous paper by Bybee and Budge, and reports improvements in performance. A UAV fitted with a texel camera is flown at a fixed height above the terrain and swaths of texel image data of the terrain below is taken continuously. Each texel swath has one or more lines of lidar data surrounded by a narrow strip of EO data. Texel swaths are taken such that there is some overlap from one swath to its adjacent swath. The GPS/IMU fitted on the camera also give coarse knowledge of attitude and position. Using this coarse knowledge and the information from the texel image, the error in the camera position and attitude is reduced which helps in producing an accurate TDEM. This paper reports improvements in the original work by using multiple lines of lidar data per swath. The final results are shown and analyzed for numerical accuracy.

Texel sheep are renowned for their exceptional meatiness. To identify the genes underlying this economically important feature, we performed a whole-genome scan in a Romanov x Texel F2 population. We mapped a quantitative trait locus with a major effect on muscle mass to chromosome 2 and subsequently fine-mapped it to a chromosome interval encompassing the myostatin (GDF8) gene. We herein demonstrate that the GDF8 allele of Texel sheep is characterized by a G to A transition in the 3' UTR that creates a target site for mir1 and mir206, microRNAs (miRNAs) that are highly expressed in skeletal muscle. This causes translational inhibition of the myostatin gene and hence contributes to the muscular hypertrophy of Texel sheep. Analysis of SNP databases for humans and mice demonstrates that mutations creating or destroying putative miRNA target sites are abundant and might be important effectors of phenotypic variation.

Two trials were conducted to determine the energy requirements of feedlot Texel crossbred lambs. In a comparative slaughter trial, thirty 11/16 Texel x 5/16 Ile de France crossbred noncastrated male lambs, weaned at 42 d of age (16.2 +/- 2.1 kg of shrunk BW; SBW), were used. Five lambs were randomly chosen and slaughtered after 10 d of experimental management and diet adaptation (baseline group). Fifteen lambs then were fed for ad libitum intake and slaughtered at 25, 30, or 35 kg of SBW. The remaining 10 lambs were randomly assigned to 2 levels of DMI, either 70 or 55% of the ad libitum intake, and were slaughtered concomitantly with lambs of the 35 kg of SBW group. Total body N, fat, and energy contents were determined. In a digestibility trial, 6 Texel x Ile de France crossbred lambs (30.4 +/- 2.6 kg of SBW) were housed in metabolic cages and used in a replicated 3 x 3 Latin square experiment to evaluate the energetic value of the diet at different feed intake levels. Net and ME requirements for maintenance were 58.6 and 91 kcal/kg(0.75) of SBW, respectively. Consequently, partial efficiency of energy use for maintenance was 0.64. Body fat content varied from 72.7 to 125.9 g/kg of empty BW, respectively, for 13.1 and 28.2 kg of empty BW. Net energy requirements for growth of lambs at 15 and 35 kg of SBW at an ADG of 250 g were 424 and 553 kcal/d, respectively. Partial efficiency of energy use for growth was 0.47. Texel x Ile de France crossbred growing lambs used in this study showed decreased nutritional requirements than those reported by most nutritional systems.

For the consumer, tenderness, juiciness and flavour are often described as the most important factors for meat eating quality, all of which have a close association with intramuscular fat (IMF). X-ray computed tomography (CT) can measure fat, muscle and bone volumes and weights, in vivo in sheep and CT predictions of carcass composition have been used in UK sheep breeding programmes over the last few decades. This study aimed to determine the most accurate combination of CT variables to predict IMF percentage of M. longissimus lumborum in Texel lambs. As expected, predicted carcass fat alone accounted for a moderate amount of the variation (R(2)=0.51) in IMF. Prediction accuracies were significantly improved (Adj R(2)>0.65) using information on fat and muscle densities measured from three CT reference scans, showing that CT can provide an accurate prediction of IMF in the loin of purebred Texel sheep. Postfabrication carcass component weights of 517 crossbred wether lambs were analyzed to evaluate 4 terminal-sire breeds. Wethers were produced over 3 yr from single-sire matings of 22 Columbia, 22 USMARC-Composite (Composite), 21 Suffolk, and 17 Texel rams to adult Rambouillet ewes. Lambs were reared to weaning in an extensive western rangeland production system and finished in a feedlot on a high-energy finishing diet. When wethers reached a mean BW of 54.4, 61.2, or 68.0 kg, they were transported to The Ohio State University abattoir for harvest. After refrigeration for approximately 24 h, chilled carcass weight (CCW) was measured, carcasses were fabricated according to Style A of Institutional Meat Purchase Specifications, and postfabrication weights were recorded. At comparable numbers of days on feed, Suffolk-sired lambs had heavier (P < 0.04) neck, breast, shoulder, foreshank, rack, loin, leg, sirloin, roast-ready rack, trimmed loin, and boneless leg cuts than progeny of the other sire breeds. Boneless sirloins were heavier (P < 0.01) for Suffolk-sired than Composite-sired lambs but did not differ from those for Columbia- or Texel-sired lambs. Columbia- and Suffolk-sired lambs had heavier (P < 0.01) hindshanks than Texel-sired lambs. Suffolk-sired lambs had heavier (P < 0.01) high-value cuts (rack, loin, leg, and sirloin) and trimmed high-value cuts than progeny of the other sire breeds. Cutting loss (CCW - wholesale cut weights) and high-value trimming loss were greatest (P < 0.02) for Suffolk-sired lambs and least for Texel- and Composite-sired lambs. Sire breed did not affect (P > 0.06) flank weight. Data adjusted to comparable CCW reduced the number of significant sire-breed effects and changed sire-breed rankings of carcass component weights, for which sire breeds differed. After adjusting, Suffolk-sired lambs had lighter (P < 0.05) loins than Columbia- and Composite-sired lambs, Composite-sired lambs had heavier (P < 0.05) high-value cuts than Suffolk-sired lambs, and Suffolk- and Columbia-sired lambs had heavier (P < 0.05) necks than Texel-sired lambs. At predicted backfat thickness of 6.6 mm, Composite-sired lambs had a greater (P < 0.05) percentage of high-value cuts than Suffolk-sired lambs before but not after trimming. Producers can use these results to select terminal-sire breeds that will complement their production system and improve lamb value.

Several photogrammetry-based methods have been proposed that the derive three-dimensional (3-D) information from digital images from different perspectives, and lidar-based methods have been proposed that merge lidar point clouds and texture the merged point clouds with digital imagery. Image registration alone has difficulty with smooth regions with low contrast, whereas point cloud merging alone has difficulty with outliers and a lack of proper convergence in the merging process. This paper presents a method to create 3-D images that uses the unique properties of texel images (pixel-fused lidar and digital imagery) to improve the quality and robustness of fused 3-D images. The proposed method uses both image processing and point-cloud merging to combine texel images in an iterative technique. Since the digital image pixels and the lidar 3-D points are fused at the sensor level, more accurate 3-D images are generated because registration of image data automatically improves the merging of the point clouds, and vice versa. Examples illustrate the value of this method over other methods. The proposed method also includes modifications for the situation where an estimate of position and attitude of the sensor is known, when obtained from low-cost global positioning systems and inertial measurement units sensors.

TM-QTL is a quantitative trait locus (QTL) on ovine chromosome 18 (OAR18) known to affect loin muscling in Texel sheep. Previous work suggested that its mode of inheritance is consistent with paternal polar overdominance, but this has yet to be formally demonstrated. This study used purebred Texel sheep segregating for TM-QTL to confirm its presence in the chromosomal region in which it was first reported and to determine its pattern of inheritance. To do so, this study used the first available data from a Texel flock, which included homozygote TM-QTL carriers (TM/TM; n=34) in addition to homozygote non-carriers (+/+; n=40 and, heterozygote TM-QTL-carriers inheriting TM-QTL from their sire (TM/+; n=53) or their dam (+/TM; n=17). Phenotypes included a wide range of loin muscling, carcass composition and tissue distribution traits. The presence of a QTL affecting ultrasound muscle depth on OAR18 was confirmed with a paternal QTL effect ranging from +0.54 to +2.82 mm UMD (s.e. 0.37 to 0.57 mm) across the sires segregating for TM-QTL. Loin muscle width, depth and area, loin muscle volume and dissected M. longissimus lumborum weight were significantly greater for TM/+ than +/+ lambs (+2.9% to +7.9%; P<0.05). There was significant evidence that the effect of TM-QTL on the various loin muscling traits measured was paternally polar overdominant (P<0.05). In contrast, there was an additive effect of TM-QTL on both live weight at 20 weeks and carcass weight; TM/TM animals were significantly (P<0.05) heavier than +/+ (+11.1% and +7.3%, respectively) and +/TM animals (+11.9% and +11.7%, respectively), with TM/+ intermediate. Weights of the leg, saddle and shoulder region (corrected for carcass weight) were similar in the genotypic groups. There was a tendency for lambs inheriting TM-QTL from their sire to be less fat with slightly more muscle than non-carriers. For example, carcass muscle weight measured by live animal CT-scanning was 2.8% higher in TM/TM than +/+ lambs (P<0.05), carcass muscle weight measured by carcass CT-scanning was 1.36% higher in TM/+ than +/+ lambs (P<0.05), and weight of fat trimmed from the carcass cuts was significantly lower for TM/+ than +/+ lambs (-11.2%; P<0.05). No negative effects of TM-QTL on carcass traits were found. Optimal commercial use of TM-QTL within the sheep industry would require some consideration, due to the apparently different mode of action of the two main effects of TM-QTL (on growth and muscling).

Quality measures of lamb longissimus dorsi were evaluated in 514 crossbred wether lambs to assess sire breed differences. Wethers were produced over 3 yr from single-sire matings of 22 Columbia, 22 U.S. Meat Animal Research Center (USMARC)-Composite (Composite), 21 Suffolk, and 17 Texel rams to adult Rambouillet ewes. Lambs were reared to weaning in an extensive western rangeland production system and finished in a feedlot on a high-energy finishing diet. One of three harvest groups were randomly assigned to each lamb, and lambs were transported to The Ohio State University abattoir when the mean BW of wethers remaining in the feedlot reached 54.4, 61.2, or 68.0 kg. After harvest, subjective lean quality scores were assigned and LM pH (immediately after and 24 h after harvest), color (quantified as Minolta L*, a*, and b*), intramuscular fat (IMF), cooking loss percentage, and Warner-Bratzler shear force (WBSF) were determined. Statistical models included fixed effects of sire breed, year of birth, and harvest group and random effects of sire (nested within sire breed and year) and maternal grandsire. Year and harvest group were significant (P < 0.04) for all traits, except harvest group was not for WBSF and lean quality (P > 0.28). At comparable numbers of days on feed, Texel-sired wethers had the greatest (more desirable; P < 0.01) subjective lean quality scores while Composite- and Suffolk-sired lambs were intermediate and Columbia-sired lambs had the lowest (less desirable). Minolta L* values were greater (P < 0.02) for Texel- than Columbia-sired lambs, although this difference is not visually discernible by humans. No significant (P > 0.05) sire breed effects were detected for LM pH at or 24 h after harvest, Minolta a* and b*, IMF, percentage of cooking loss, and WBSF at comparable numbers of days on feed. At comparable chilled carcass weight, significant (P < 0.01) sire breed effects were detected for subjective lean quality score. Texel-sired lambs had greater scores than Columbia- and Suffolk-sired lambs, but Composite-sired lambs did not differ from lambs sired by the other sire breeds. Sire breed effects were not detected (P > 0.15) for LM pH at or 24 h after harvest, Minolta L*, a*, and b*, cooking loss percentage, IMF, and WBSF at comparable chilled carcass weight. Therefore, results of the present study indicated that the 4 sire breeds produced crossbred lamb carcasses that had similar and desirable LM quality measurements, meeting consumer expectations.

Reproduction is an important trait in sheep breeding as well as in other livestock. However, despite its importance the genetic mechanisms of litter size in domestic sheep (Ovis aries) are still poorly understood. To explore genetic mechanisms underlying the variation in litter size, we conducted multiple independent genome-wide association studies in five sheep breeds of high prolificacy (Wadi, Hu, Icelandic, Finnsheep, and Romanov) and one low prolificacy (Texel) using the Ovine Infinium HD BeadChip, respectively. We identified different sets of candidate genes associated with litter size in different breeds: BMPR1B, FBN1, and MMP2 in Wadi; GRIA2, SMAD1, and CTNNB1 in Hu; NCOA1 in Icelandic; INHBB, NF1, FLT1, PTGS2, and PLCB3 in Finnsheep; ESR2 in Romanov and ESR1, GHR, ETS1, MMP15, FLI1, and SPP1 in Texel. Further annotation of genes and bioinformatics analyses revealed that different biological pathways could be involved in the variation in litter size of females: hormone secretion (FSH and LH) in Wadi and Hu, placenta and embryonic lethality in Icelandic, folliculogenesis and LH signaling in Finnsheep, ovulation and preovulatory follicle maturation in Romanov, and estrogen and follicular growth in Texel. Taken together, our results provide new insights into the genetic mechanisms underlying the prolificacy trait in sheep and other mammals, suggesting targets for selection where the aim is to increase prolificacy in breeding projects. The effects of several commercial meat starter cultures on degradation of polychlorinated biphenyls (PCBs) in dry fermented sausages over 28days of drying/ripening were investigated. The sausage batter was prepared according to a classic recipe and spiked with a standard solution of a PCB congener mixture. With addition of different commercial meat starter cultures, five experimental groups were prepared: no further addition; and separate addition of each of four starter cultures: Texel DCM-1, Texel LM-30, Biostar Sprint, and SM-181. Samples were taken at the beginning of fermentation (zero time), and after 4, 7, 14, 21 and 28days. PCB residues were extracted with hexane. The PCB contents were determined using gas chromatography-mass spectrometry. The PCB levels were reduced in all of the experimental groups tested, where addition of starter culture Biostar Sprint (Lactobacillus sakei, Staphylococcus carnosus, Staphylococcus xylosus) showed the highest PCB degradation rates. Lamb meat is often perceived by consumers as fatty, and consumption has decreased in recent decades. A lean growth index was developed in the UK for terminal sire breeds to increase carcass lean content and constrain fat content at a constant age end point. The purposes of this study were 1) to evaluate the effects of index selection of terminal sires on their crossbred offspring at finishing and 2) to evaluate its effectiveness within terminal sire breeds. Approximately 70% of lambs marketed in the UK have been sired by rams of breeds typically thought of as specialized terminal sires. The most widely used are Charollais, Suffolk, and Texel. These breeds participated in sire referencing schemes from the early 1990s by sharing rams among flocks selected on the lean growth index. From 1999 to 2002 approximately 15 "high" and 15 "low" lean growth index score rams were selected from within their sire referencing schemes and mated to Welsh and Scottish Mule ewes. Their crossbred offspring were commercially reared on 3 farms in the UK. Lambs were finished to an estimated 11% subcutaneous fat by visual evaluation. At finishing, lambs were weighed, ultrasonically scanned, and assessed for condition score and conformation. Records were obtained for 6356 lambs on finishing BW (FWT), ultrasonic muscle depth (UMD), ultrasonic fat depth, overall condition score (OCS), and conformation of gigot, loin, and shoulder. Ultrasonic fat depth was log transformed (logUFD) to approach normality. High-index-sired lambs were heavier at finishing (1.2±0.2 kg) with thicker UMD (0.7±0.2 mm) and less logUFD (0.08±0.01 mm; P<0.05). There were no differences in OCS or conformation based on the sire index or breed (P>0.08). Suffolk-sired lambs were heavier than Charollais (1.0±0.3 kg), which were heavier than Texel (0.9±0.3 kg; P<0.001). Texel-sired lambs had thicker UMD than Charollais (0.7±0.2 mm; P<0.001) but were not different than Suffolk. Charollais-sired lambs had greater logUFD than both Texel (0.098±0.016 mm) and Suffolk (0.061±0.017 mm) sired lambs (P<0.001). Within a breed, high- and low-index-sired lambs differed in performance with the exceptions of FWT and UMD in Suffolks. Index selection produced heavier and leaner lambs at finishing. Producers have flexibility in choosing the terminal sire that best fits their production system.

Earth Observations taken by the Expedition 15 Crew
2007-04-30
ISS015-E-05977 (1 May 2007) --- Den Helder, Netherlands is featured in this image photographed by an Expedition 15 crewmember on the International Space Station. The city and harbor of Den Helder in the northern Netherlands has been the home port of the Dutch Royal Navy for over 175 years. Its favorable location provides access to the North Sea, and has made it an important commercial shipping port in addition to its strategic role. Bright red agricultural fields to the south of Den Helder indicate another noteworthy aspect of the region--commercial farming of tulips and hyacinth. This image is an oblique view--the camera is oriented at an angle relative to "straight down"--of the Den Helder region taken from the space station, which was located to the southeast, near Dulmen, Germany (approximately 225 kilometers away in terms of ground distance) when the image was acquired. In addition to the manmade structures of the Den Helder urban area (reddish gray to gray street grids) and dockyards to the east of the city, several striking geomorphic features are visible. The extensive gray mudflats, with their prominent branching pattern (top right), indicate that this image was acquired at low tide, and suggest the general low elevation of the region. Parallel wave patterns along the mudflats and in the Marsdiep strait are formed as water interacts with the sea bottom between Den Helder and Texel Island during tidal flow. Some ship wakes are also visible. According to scientists, the bright white-gray triangular region at the southern tip of Texel Island (bottom center) is a dune field, consisting mainly of eolian (windborne) sands deposited during the last Ice Age. Subsequent sea level rise and shoreline processes have mobilized and re-deposited these sands into their current configuration -- including a new dune field island to the southwest of Texel (bottom center).

This paper concerns the likely origin of three mutations with large effects on ovulation rate identified in the Belclare and Cambridge sheep breeds; two in the BMP15 gene (FecXG and FecXB) and the third (FecGH) in GDF9. All three mutations segregate in Belclare sheep while one, FecXB, has not been found in the Cambridge. Both Belclare and Cambridge breeds are relatively recently developed composites that have common ancestry through the use of genetic material from the Finnish Landrace and Lleyn breeds. The development of both composites also involved major contributions from exceptionally prolific ewes screened from flocks in Ireland (Belclare) and Britain (Cambridge) during the 1960s. The objective of the current study was to establish the likely origin of the mutations (FecXG, FecXB and FecGH) through analysis of DNA from Finnish Landrace and Lleyn sheep, and Galway and Texel breeds which contributed to the development of the Belclare breed. Ewes with exceptionally high prolificacy (hyper-prolific ewes) in current flocks on Irish farms were identified to simulate the screening of ewes from Irish flocks in the 1960s. DNA was obtained from: prolific ewes in extant flocks of Lleyn sheep (n = 44) on the Lleyn peninsula in Wales; hyper-prolific ewes (n = 41); prolific Galway (n = 41) ewes; Finnish Landrace (n = 124) and Texel (n = 19) ewes. The FecXG mutation was identified in Lleyn but not in Finnish Landrace, Galway or Texel sheep; FecXB was only found among the hyper-prolific ewes. The FecGH mutation was identified in the sample of Lleyn sheep. It was concluded from these findings that the Lleyn breed was the most likely source of the FecXG and FecGH mutations in Belclare and Cambridge sheep and that the FecXB mutation came from the High Fertility line that was developed using prolific ewes selected from commercial flocks in Ireland in the 1960′s and subsequently used in the genesis of the Belclare. A Therapeutic Nipple Shield (TNS) was previously developed to respond to the global need for new infant therapeutic delivery technologies. However, the release efficiency for the same Active Pharmaceutical Ingredient (API) from different therapeutic matrices within the TNS formulation has not yet been investigated. To address this, in-vitro release of elemental zinc into human milk from two types of Texel non-woven fibre mats of varying thickness and different gram per square meter values, placed inside the TNS was explored and compared to the release from zinc-containing rapidly disintegrating tablets. In-vitro delivery was performed by means of a breastfeeding simulation apparatus, with human milk flow rates and suction pressure adjusted to physiologically relevant values, and release was quantified using Inductively Coupled Plasma Optical Emission Spectrometry (ICP-OES). It was found that a total recovery of 62-64 % elemental zinc was obtained after the human milk had passed through the fibre insert, amounting to a 20-48% increase compared to previous zinc delivery studies using rapidly disintegrating tablets within the TNS. This indicates that non-woven Texel fibre mats were identified as the superior dosage form for oral zinc delivery into human milk using a TNS. Copyright © 2018 Elsevier B.V. All rights reserved.

This paper concerns the likely origin of three mutations with large effects on ovulation rate identified in the Belclare and Cambridge sheep breeds; two in the BMP15 gene (FecX(G) and FecX(B)) and the third (FecG(H)) in GDF9. All three mutations segregate in Belclare sheep while one, FecX(B), has not been found in the Cambridge. The objective of the current study was to establish the likely origin of the mutations (FecX(G), FecX(B) and FecG(H)) through analysis of DNA from Finnish Landrace and Lleyn sheep, and Galway and Texel breeds which contributed to the development of the Belclare breed. The FecX(G) mutation was identified in Lleyn but not in Finnish Landrace, Galway or Texel sheep; FecX(B) was only found among the hyper-prolific ewes. The FecG(H) mutation was identified in the sample of Lleyn sheep. It was concluded from these findings that the Lleyn breed was the most likely source of the FecX(G) and FecG(H) mutations in Belclare and Cambridge sheep and that the FecX(B) mutation came from the High Fertility line that was developed using prolific ewes selected from commercial flocks in Ireland in the 1960's and subsequently used in the genesis of the Belclare.

A cellular automata model is used to analyze the effects of groundwater levels and sediment supply on aeolian dune development occurring on sand flats close to inlets. The model considers, in a schematized and probabilistic way, aeolian transport processes, groundwater influence, vegetation development, and combined effects of waves and tides that can both erode and accrete the sand flat. Next to three idealized cases, a sand flat adjoining the barrier island of Texel, the Netherlands, was chosen as a case study. Elevation data from 18 annual LIDAR surveys was used to characterize sand flat and dune development. Additionally, a field survey was carried out to map the spatial variation in capillary fringe depth across the sand flat. Results show that for high groundwater situations, sediment supply became limited inducing formation of Coppice-like dunes, even though aeolian losses were regularly replenished by marine import during sand flat flooding. Long dune rows developed for high sediment supply scenarios which occurred for deep groundwater levels. Furthermore, a threshold depth appears to exist at which the groundwater level starts to affect dune development on the inlet sand flat. The threshold can vary spatially depending on external conditions such as topography. On sand flats close to inlets, groundwater is capable of introducing spatial variability in dune growth, which is consistent with dune development patterns found on the Texel sand flat.
Polwarth and Texel ewe parturition duration and its association with lamb birth asphyxia.

The objective of the present study was to test the hypothesis that parturition duration is related to birth asphyxia in lambs and that asphyxia affects newborn lamb viability and vigor. Two sire and dam genotypes (Texel: TX; Polwarth: PW) and their crosses were represented in the study. Eighty lambs (25 PW sire × PW dam, 13 TX × TX, 25 TX × PW, and 17 PW × TX) born to 69 grazing ewes were used. At birth, the log₁₀ length of the second stage of parturition, birth weight, placental weight, and several body measurements were recorded on all lambs, and jugular blood samples were analyzed with the i-Stat Portable Clinical Analyzer (Abbott, Montevideo, Uruguay). A modified Apgar viability score at birth and lamb behavior during their first hour of life were recorded. Brain weight, muscle:bone ratio, and bone density were recorded in 20 male lambs (5 from each breed group) that were euthanized and dissected 24 h after birth. Data were analyzed by linear regression, least squares ANOVA, and ordinal and binary logistic regressions. Mean blood gas and acid-base variables were 7.21 ± 0.09 for pH, 18.4 ± 9.8 mmHg for partial pressure of oxygen, 53 ± 12.5 mmHg for partial pressure of carbon dioxide, and -4 ± 5.1 mmol/L for extracellular fluid base excess. Parturition duration increased with birth weight (P < 0.001) and was shorter in TX ewes (P < 0.001), female lambs (P < 0.05), twins (P < 0.09), and twin females (sex × litter size interaction, P < 0.02). Twenty-six (32.9%) lambs were born asphyxiated (pO₂ < 10 mmHg or pH <7.1). Parturition duration increased the risk of asphyxia (P < 0.001), decreased the viability score (P < 0.001), and increased the latency to suckle the udder (P < 0.05). Twin-born lambs presented at birth a 16-fold greater risk of asphyxia (P < 0.01) and reduced placental efficiency (P < 0.05). Texel-sired lambs appeared immature at birth, with less bone density (P < 0.05), smaller brain (P < 0.05), shorter forelimbs (P < 0.05), greater anterior (P < 0.001) and posterior (P < 0.05) neck circumference, and greater muscle:bone ratio (P < 0.05). Immaturity may explain greater TX biotype survival. Together these results demonstrate that a relationship exists between parturition duration, neonatal viability and behavior, and acid-base balance values in single- and twin-born lambs, suggesting that birth asphyxia is an important risk factor in perinatal lamb mortality.

Objectives were to document effects of the Texel myostatin mutation (MSTN) on growth and carcass traits and also test whether or not interactions with the callipyge mutation (CLPG) could be detected. Twelve rams heterozygous at both loci on the two different chromosomes were mated to 215 terminal-sire type composite crossbred ewes genotyped as non-carriers for both loci. A total of 365 lambs were born, 362 of those were genotyped and 236 lambs contributed carcass data to estimate effects and interactions among the four genotype combinations produced. The four genotype combinations were defined as follows: ++/++ for wild-type at both loci; ++/C+ for wild-type at MSTN and heterozygous at CLPG; M+/++ for heterozygous at MSTN and wild-type at CLPG; and M+/C+ for heterozygous at both loci. The two independently segregating sire-derived alleles represent different breed-of-origin contrasts at each locus (Texel vs. composite origin for MSTN and Dorset vs. Texel origin for CLPG). Birth weight was recorded on all lambs, and subsequent body weights were adjusted to 56 (weaning), 70, and 140 d of age. Within sire-sex-genotype subgroups, naturally reared lambs were assigned to one of eight slaughter groups accounting for variation in birth date. Lambs were serially slaughtered at weekly intervals, 30 lambs per group, from roughly 26 to 33 wk of age. In addition to standard carcass traits, subjective leg scores were assigned and widths of carcasses were measured at the widest points of the shoulder and rump. Differences in birth weight were detected (P < 0.01) for the combination of the two loci and birth type, with single-born differences among genotypes exceeding differences among twin born progeny. Those interaction differences among genotypes were not as important at weaning (P = 0.36). Impact on growth rate differences among the genotypes during the post-weaning period were variable and dependent on sex of the lamb (P < 0.01). A synergistic interaction between MSTN and CLPG was observed for leg muscling scores (P < 0.05) but no other measures of carcass shape were affected. One copy of MSTN had a more modest impact on fat deposition and muscle conformation than did CLPG and did not interact (all values P > 0.20). Although some non-additive interactions that vary by trait and sex were detected, in general the data are consistent with the two mutations acting on muscle growth through independent pathways.

The discovery and identification of Ovis aries (sheep) miRNAs will further promote the study of miRNA functions and gene regulatory mechanisms. To explore the microRNAome (miRNAome) of sheep in depth, samples were collected that included eight developmental stages: the longissimus dorsi muscles of Texel fetuses at 70, 85, 100, 120, and 135 days, and the longissimus dorsi muscles of Ujumqin fetuses at 70, 85, 100, 120, and 135 d, and lambs at 0 (birth), 35, and 70 d. These samples covered all of the representative periods of Ovis aries growth and development throughout gestation (about 150 d) and 70 d after birth. Texel and Ujumqin libraries were separately subjected to Solexa deep sequencing; 35,700,772 raw reads were obtained overall. We used ACGT101-miR v4.2 to analyze the sequence data. Following meticulous comparisons with mammalian mature miRNAs, precursor hairpins (pre-miRNAs), and the latest sheep genome, we substantially extended the Ovis aries miRNAome. The list of pre-miRNAs was extended to 2,319, expressing 2,914 mature miRNAs. Among those, 1,879 were genome mapped to unique miRNAs, representing 2,436 genome locations, and 1,754 pre-miRNAs were mapped to chromosomes. Furthermore, the Ovis aries miRNAome was processed using an elaborate bioinformatic analysis that examined multiple end sequence variation in miRNAs, precursors, chromosomal localizations, species-specific expressions, and conservative properties. Taken together, this study provides the most comprehensive and accurate exploration of the sheep miRNAome, and draws conclusions about numerous characteristics of Ovis aries miRNAs, including miRNAs and isomiRs. There is considerable interest in locally adapted breeds of livestock as reservoirs of genetic diversity that may provide important fitness traits for future use in agriculture. In marginal areas, these animals contribute to food security and extract value from land unsuitable for other systems of farming. In England, close to 50% of the national sheep flock is farmed on grassland designated as disadvantaged areas for agricultural production. Many of these areas are in the uplands, where some native breeds of sheep continue to be commercially farmed only in highly localised geographical regions to which they are adapted. This study focuses on three of these breeds, selected for their adaptation to near identical environments and their geographical concentration in regions close to one another. Our objective has been to use retrotyping, microsatellites and single nucleotide polymorphisms to explore the origins of the breeds and whether, despite their similar adaptations and proximity, they are genetically distinctive. We find the three breeds each have a surprisingly different pattern of retrovirus insertions into their genomes compared with one another and with other UK breeds. Uniquely, there is a high incidence of the R0 retrotype in the Herdwick population, characteristic of a primitive genome found previously in very few breeds worldwide and none in the UK mainland. The Herdwick and Rough Fells carry two rare retroviral insertion events, common only in Texels, suggesting sheep populations in the northern uplands have a historical association with the original pin-tail sheep of Texel Island. Microsatellite data and analyses of SNPs associated with RXFP2 (horn traits) and PRLR (reproductive performance traits) also distinguished the three breeds. Significantly, an SNP linked to TMEM154, a locus controlling susceptibility to infection by Maedi-Visna, indicated that all three native hill breeds have a lower than average risk of infection to the lentivirus. Dune slacks are important habitats, with many endangered plant species. A series of eleven dune slacks of 1-42years old was studied in SW-Texel, the Netherlands, with the EU-habitat directive species Liparis loeselii present in all except the youngest and oldest. Analysis of aerial photographs revealed that new slacks are currently formed every 4-5years. In each slack, topsoil and vegetation data were collected in 2010 and 2014-2015. During succession, vegetation changed from brackish pioneer stages to dune slacks with L. loeselii and Parnassia palustris and ultimately grassland species. Differences between dune slacks and sampling periods were mostly significant. Herb cover and soil C increased with slack age, and over the five year study period, while bare sand, bulk density and pH decreased. The annual pH-decrease was 0.055 and 0.075 for pH-H2O and pH-KCl respectively, and annual C-increase 0.16% and 35gm(-2). Liparis loeselii was only present between pHH2O 5.8-7.5 and pHKCl 5.6-7.6, and only occurred at C-content below 4.3%. In lime-poor dunes, environmental conditions thus become unsuitable approximately 34years after the start of succession. In the dune slacks, Liparis loeselii established within 6years, showed peak values after 11-16years, and declined until conditions became unsuitable. Rejuvenation may occur after large storms with fresh sand deposits. However, even with further succession, the present populations are not endangered and probably last until 2040. With new dune slacks every 5years, L. loeselii occurs in approximately eight different dune slacks at the same time, ensuring viable populations also in the future. This shows that adverse effects of succession can be counteracted by dynamics on local and landscape scale. Copyright © 2016 Elsevier B.V. All rights reserved.

The performance of 3D rendering of Graphics Processing Unit that convents 3D vector stream into 2D frame with 3D image effects significantly impact users’ gaming experience on modern computer systems. Due to the high texture throughput in 3D rendering, main memory bandwidth becomes a critical obstacle for improving the overall rendering performance. 3D stacked memory systems such as Hybrid Memory Cube (HMC) provide opportunities to significantly overcome the memory wall by directly connecting logic controllers to DRAM dies. Based on the observation that texel fetches significantly impact off-chip memory traffic, we propose two architectural designs to enable Processing-In-Memory based GPUmore » for efficient 3D rendering.« less

2016-01-01
In this study, we report a complete mitochondrial (mt) genome sequence of the Texel ewe, Ovis aries. The total genome is 16,615 bp in length and its overall base composition was estimated to be 33.68% for A, 27.36% for T, 25.86% for C, and 13.10% for G indicating an AT-rich (61.04%) feature in the O. aries mtgenome. It contains a total of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes and a control region (D-loop region). Comparisons with other publicly available sheep mitogenomes revealed a bunch of nucleotide diversity. This complete mitgenome sequence would enlarge useful genomic information for further studies on sheep evolution and domestication that will enhance germplasm conservation and breeding programs of O. aries.

The objective of this work was to study nutritional strategies for decreasing methane production by ruminants fed tropical diets, combining in vitro and in vivo methods. The in vitro approach was used to evaluate the dose effect of condensed tannins (CT) contained in leaves of Gliricidia sepium, Leucaena leucocephala, and Manihot esculenta (39, 75, and 92 g CT/kg DM, respectively) on methane production and ruminal fermentation characteristics. Tannin-rich plants (TRP) were incubated for 24 h alone or mixed with a natural grassland hay based on Dichanthium spp. (control plant), so that proportions of TRP were 0, 0.25, 0.5, 0.75, and 1.0. Methane production, VFA concentration, and fermented OM decreased with increased proportions of TRP. Numerical differences on methane production and VFA concentration among TRP sources may be due to differences in their CT content, with greater effects for L. leucocephala and M. esculenta than for G. sepium. Independently of TRP, the response to increasing doses of CT was linear for methane production but quadratic for VFA concentration. As a result, at moderate tannin dose, methane decreased more than VFA. The in vivo trial was conducted to investigate the effect of TRP on different ruminal microbial populations. To this end, 8 rumen-cannulated sheep from 2 breeds (Texel and Blackbelly) were used in two 4 × 4 Latin square designs. Diets were fed ad libitum and were composed of the same feeds used for the in vitro trial: control plant alone or combined with pellets made from TRP leaves at 44% of the diet DM. Compared to TRP, concentration of Ruminococcus flavefaciens was greater for the control diet and concentration of Ruminococcus albus was least for the control diet. The methanogen population was greater for Texel than for Blackbelly. By contrast, TRP-containing diets did not affect protozoa or Fibrobacter succinogenes numbers. Hence, TRP showed potential for mitigating methane production by ruminants. These findings suggest that TRP fed as pellets could be used to decrease methane production.

Coastal waters up to 90 km off the coast of the Netherlands and Germany were investigated with the RV 'Friedrich Heincke' at 21 stations in July 1987. Hydrographic, chemical and biological parameters were measured simultaneously, such as temperature, salinity, oxygen saturation, adenosine triphosphate (ATP) concentrations and electron transport system (ETS) activities. The mean surface temperature was 16°C, salinity distribution showed a tongue of high salinity in the Southern Bight and coastal areas of low values by river input from the Rhine and Elbe. The mean oxygen concentration was 102.4%, with the highest percentages northwest of Helgoland and the lowest near Scheveningen and Cuxhaven. The ecosystem parameters of ATP-biomass and respiratory ETS-activity showed a distribution pattern corresponding to the inflow of clear Channel water and nutrient-rich river water. Highest biomass values and respiratory activities were found near Texel and northwest of Helgoland. The distribution patterns of microplankton species were more patchy. Distribution patterns are shown of phytoplankton species such as Ceratium fusus, Phaeocystis spec., Rhizosolenia shrubsolei, and Mesodinium rubrum and various groups of microplankton. The highest numbers of Ceratium were found near Helgoland, of Phaeocystis near Terschelling and Cuxhaven, of Rhizosolenia near Texel and Langeoog and of Mesodinium near the Dutch coast and north of Borkum. Concentrations of bacteria were determined both by cultivation and epifluorescence microscopy methods. The numbers obtained by the pour-plate method varied much more than those determined by the direct-count method. At all stations, oil-degrading bacteria were found. The numbers of bacteria able to degrade mineral oil were higher than the numbers of bacteria able to degrade a tetradecane-pentadecane mixture. Biological oxygen demand (BOD) was measured with and without oil additions after incubation of 5, 10 and 15 days and a potency to degrade oil was shown. Beside the mapping of respiratory activity by the ETS-method, the real oxygen uptake at in situ temperatures was measured in a few samples with short incubation. An ETS/R ratio of 4.1 was estimated.
Detection of pigment network in dermatoscopy images using texture analysis

Dermatoscopy, also known as dermoscopy or epiluminescence microscopy (ELM), is a non-invasive, in vivo technique, which permits visualization of features of pigmented melanocytic neoplasms that are not discernable by examination with the naked eye. ELM offers a completely new range of visual features. One such prominent feature is the pigment network. Two texture-based algorithms are developed for the detection of pigment network. These methods are applicable to various texture patterns in dermatoscopy images, including patterns that lack fine lines such as cobblestone, follicular, or thickened network patterns. Two texture algorithms, Laws energy masks and the neighborhood gray-level dependence matrix (NGLDM) large number emphasis, were optimized on a set of 155 dermatoscopy images and compared. Results suggest superiority of Laws energy masks for pigment network detection in dermatoscopy images. For both methods, a texel width of 10 pixels or approximately 0.22 mm is found for dermatoscopy images. PMID:15249068
The callipyge mutation and other genes that affect muscle hypertrophy in sheep
2005-01-01
Genetic strategies to improve the profitability of sheep operations have generally focused on traits for reproduction. However, natural mutations exist in sheep that affect muscle growth and development, and the exploitation of these mutations in breeding strategies has the potential to significantly improve lamb-meat quality. The best-documented mutation for muscle development in sheep is callipyge (CLPG), which causes a postnatal muscle hypertrophy that is localized to the pelvic limbs and loin. Enhanced skeletal muscle growth is also observed in animals with the Carwell (or rib-eye muscling) mutation, and a double-muscling phenotype has been documented for animals of the Texel sheep breed. However, the actual mutations responsible for these muscular hypertrophy phenotypes in sheep have yet to be identified, and further characterization of the genetic basis for these phenotypes will provide insight into the biological control of muscle growth and body composition. Effect of liquid nitrogen pre-treatment on various types of wool waste fibres for biogas production.

2018-05-01
This study investigated the role of liquid nitrogen (LN 2 ) in increasing microbial accessibility of wool proteins for biogas production. It involves a mechanical size reduction of four different types of raw wool fibres, namely, Blackface, Bluefaced Leicester, Texel and Scotch Mule, in presence of liquid nitrogen, followed by the determination of the methane production potential of the pre-treated wool fibres. The highest methane yield, 157.3 cm 3 g -1 VS, was obtained from pre-treated Scotch mule wool fibre culture, and represented more than 80% increase when compared to the yield obtained from its raw equivalent culture. The increase in biogas yield was attributed to the effectiveness of LN 2 in enhancing particle size reduction and the consequent increase in wool solubility and bioavailability. Results also showed that LN 2 pre-treatment can enhance size reduction but has limited effect on the molecular structure. The study also showed that the biogas potential of waste wool fibres varies with the type and source of wool.

Use of lamb BW or chilled carcass weights (CCW), live-animal ultrasound or direct carcass measurements of backfat thickness (BF; mm) and LM area (LMA; cm(2)), and carcass body wall thickness (BWall; mm) to predict carcass yield and value was evaluated using 512 crossbred lambs produced over 3 yr by mating Columbia, U.S. Meat Animal Research Center Composite, Suffolk, and Texel rams to adult Rambouillet ewes. Lambs were harvested at 3 BW endpoints within each year. The predictive value of 3 to 5 additional linear measurements of live-animal or carcass size and shape was also evaluated. Residual correlations (adjusted for effects of year, breed, and harvest group) between ultrasound and direct measurements were 0.69 for BF and 0.65 for LMA. Increasing ultrasound or carcass LMA had positive effects (P < 0.001) on yield of chilled carcass (i.e., on dressing percentage) and, at comparable CCW, on weight of high-value cuts (rack, loin, leg, and sirloin) before trimming (HVW), weight of trimmed high-value cuts (trimmed rack and loin and trimmed boneless leg and sirloin; TrHVW), and carcass value before (CVal) and after (TrCVal) trimming of high-value cuts. By contrast, ultrasound and direct measures of BF had positive effects on yields of CCW and on HVW and CVal but large negative effects on TrHVW and TrCVal. After adjusting for BW at scanning, increases of 1 mm in ultrasound BF or 1 cm(2) in ultrasound LMA were associated with changes of US$-0.32 (P < 0.10) and $1.62 (P < 0.001), respectively, in TrCVal. Carcass BWall was generally superior to carcass BF as a predictor of TrHVW and TrCVal. Carcass LMA was superior to ultrasound LMA but carcass BF was inferior to ultrasound BF for prediction of carcass yield and value. Increasing LMA thus would be expected to improve carcass yield and value. Addition of linear measurements of live-animal or carcass size and shape to the prediction model reduced residual SD (RSD) for TrHVW and TrCVal by 0.4 to 2.2%, but subsequent removal of ultrasound or direct measures of BF and LMA from the prediction model increased RSD by 7.4 to 12.2%. Measurements of CCW, LMA, BF, and BWall would thus be appropriate to support programs for value-based marketing of lamb carcasses and are superior to systems based only on measurements of size and shape in unribbed carcasses.

Linear measurements on live lambs and carcasses can be used to characterize sheep breeds and may have value for prediction of carcass yield and value. This study used 512 crossbred lambs produced over 3 yr by mating Columbia, U.S. Meat Animal Research Center (USMARC) Composite, Suffolk, and Texel rams to adult Rambouillet ewes to assess sire-breed differences in live-animal and carcass shape and to evaluate the value of shape measurements as predictors of chilled carcass weight (CCW), weight of high-value cuts (rack, loin, leg, and sirloin; HVW), weight of trimmed high-value cuts (trimmed rack and loin and trimmed, boneless leg and sirloin; TrHVW), and estimated carcass value before (CVal) and after trimming of high-value cuts (TrCVal). Lambs were produced under extensive rangeland conditions, weaned at an average age of 132 d, fed a concentrate diet in a drylot, and harvested in each year in 3 groups at target mean BW of 54, 61, and 68 kg. Canonical discriminant analysis indicated that over 93% of variation among sire breeds was accounted for by the contrast between tall, long, less-thickly muscled breeds with greater BW and CCW (i.e., the Columbia and Suffolk) compared with shorter, more thickly muscled breeds with smaller BW and CCW. After correcting for effects of year, harvest group, sire breed, and shipping BW, linear measurements on live lambs contributed little to prediction of CCW. Similarly, after accounting for effects of CCW, linear measurements on live animals further reduced residual SD (RSD) of dependent variables by 0.2 to 5.7%, with generally positive effects of increasing live leg width and generally negative effects of increasing heart girth. Carcass measurements were somewhat more valuable as predictors of carcass merit. After fitting effects of CCW, additional consideration of carcass shape reduced RSD by 2.1, 3.6, 9.5, and 2.2% for HVW, TrHVW, CVal, and TrCVal, respectively. Effects of increasing carcass leg width were positive for HVW, TrHVW, and TrCVal. We also observed positive effects of increasing carcass length on TrCVal and negative effects of increasing cannon bone length on HVW and CVal. Increasing shoulder width had positive effects on CVal but negative effects on TrHVW. Differences in lamb and carcass shape were significantly associated with carcass yield and value, but the additional accuracy associated with use of these measurements was modest relative to that achieved from use of only shipping BW or CCW.

To test the hypothesis that sheep live weight (LW) could be used to improve enteric methane (CH4) emission calculations, mature ewes of 4 different breeds representative of the UK sheep industry were studied: Welsh Mountain, Scottish Blackface, Welsh Mule and Texel (n = 8 per breed). The ewes were housed and offered ad libitum access to fresh cut pasture of three different types, varying in digestibility: (a) a relatively high digestibility monoculture of perennial ryegrass (Lolium perenne), (b) a medium digestibility permanent pasture comprising a range of grass species, and (c) a relatively low digestibility native grassland pasture comprising mainly Molinia caerulea. Individual LW, feed dry matter intake (DMI), and CH4 emissions in chambers were measured. The linear functional relationship between DMI and CH4 emissions was positive (r = 0.77) with little breed effect. The relationships between LW and DMI, and LW and CH4 emissions were also positive but weaker, regardless of pasture type. It is concluded that change to LW was a poor indicator of DMI and has limited value in the prediction of enteric CH4 emissions from mature ewes. PMID:26647754
Genetic variations in the digestibility in sheep of selected whole-crop cereals used as silages.

Whole-plant winter cereals could be of great interest if used as silages for ruminant feeding as opposed to summer crops in that they would spare water resources or valorize low-input management. This study aimed to compare the feeding value of rye, barley, wheat (two genotypes) and triticale (six genotypes). The cereals were sown in October and harvested as silage in June. Forages were offered to Texel castrated sheep in order to evaluate the organic matter digestibility (OMd). The OMd of the wheat cultivars was higher (61.6%, P<0.05) than those of barley (57.2%) and rye (54.7%) but no different from that of triticale (60.6%). Within the triticale genotypes, OMd ranged from 54.7 to 62.3%. The presence of rough barbs should explain the relatively low intake of the cereals with the exception of wheat. Winter cereals provide good-quality forage for feeding ruminants. Wheat has a higher nutritional value than barley and rye and a wide variability for digestibility seems to exist within the triticale cultivars. Such variability in a species known for its ability to be cropped under limiting conditions should be explored in much greater depth as it could result in providing farmers with genotypes of good quality with an acceptable yield at a lower cost.

Dietary lipid effect, as a consequence of protein supplement, on lamb m. longissimus dorsi fatty acid composition was investigated, with emphasis on biohydrogenation intermediates. Crossbred lambs (White Swedish Landrace × Texel) were fed a barley-based diet without (CON) or with protein supplements including peas (PEA), rapeseed cake (RC) or hempseed cake (HC). The HC diet resulted in the highest muscle 22:6n-3 proportion, with the RC diet being similar (P<0.05). Protein supplement did not affect the c9,t11 conjugated linoleic acid (CLA) proportion, however the HC diet increased some minor CLA isomers, including t10,c12 CLA (P<0.05). The t10-18:1 and total trans-18:1 were lowest for the RC diet (P<0.05), likely relating to rumen conditions and precursor availability. The saturated, monounsaturated and branched-chain fatty acids were largely unaffected by protein supplement. In conclusion, feeding the RC diet lowered the t10-18:1 and total trans-18:1 in meat, and modestly increased 22:6n-3 content. The direction of these changes would be beneficial, making the RC diet the preferred protein supplement; however the magnitude of the changes in the present experiment may not be sufficient to have an impact on human health. Myostatin and Pax7 have been well documented individually, however, the mechanism by which Myostatin regulates Pax7 is seldom reported. Here, based on muscle transcriptome analysis in Texel (Myostatin mutant) and Ujumqin (wild type) sheep across the five fetal stages, we constructed and examined the Myostatin-Pax7 pathways in muscle. Then we validated the signals by RNAi in the proliferating and differentiating sheep myoblasts in vitro at mRNA, protein, and cell morphological levels. We reveal that Myostatin signals to Pax7 at least through Ezh2, Src, and Akt during the sheep myoblast proliferation and differentiation. Other signals such as p38MAPK, mTOR, Erk1/2, Wnt, Bmp2, Smad, Tgfb1, and p21 are most probably involved in the Myostatin-affected myogenic events. Myostatin knockdown significantly reduces the counts of nucleus and myotube, but not the fusion index of myoblasts during cell differentiation. In addition, findings also indicate that Myostatin is required for normal myogenic differentiation of the sheep myoblasts, which is different from the C2C12 myoblasts. We expand the regulatory network of Myostatin-Pax7 pathways and first illustrate that Myostatin as a global regulator participates in the epigenetic events involved in myogenesis, which contributes to understand the molecular mechanism of Myostatin in regulation of myogenesis. Albendazole (ABZ), widely used benzimidazole anthelmintic, administered to animals enters via excrements into environment and may impact non-target organisms. Moreover, exposure of lower development stages of helminths to anthelmintics may also encourage the development of drug-resistant strains of helminths. In present project, the kinetics of ABZ (10 mg kg(-1) p.o.) and its metabolite (ABZ.SO, ABZSO2) elimination in faeces from treated Texel lambs were studied using UHPLC/MS/MS with the aim to find out their concentrations achievable in the environment. Consequently, the effect of these compounds on lower development stages of Barber's pole worm (Haemonchus contortus) and on germination of white mustard (Sinapis alba) seeds was evaluated. The results showed that ABZ concentrations in faeces excreted in 4-60 h after treatment were above the concentrations lethal for H. contortus eggs. Moreover, pre-incubation with sub-lethal doses of ABZ and ABZ.SO did not increase the resistance of H. contortus eggs and larvae to anthelmintics. On the other hand, concentrations of ABZ and ABZ.SO in faeces are so high that might have negative influence on non-target soil invertebrates. As neither ABZ nor its metabolites affect the germination of mustard seeds, phytoremediation could be considered as potential tool for detoxification of ABZ in the environment.

More than half of the annual catch of about 30 million lugworms Arenicola marina from the Dutch Wadden Sea originates from digging machines which make 40-cm deep guilles in a few restricted tidal-flat areas (Texel, Balgzand) in the westernmost part of the Wadden Sea. Four successive years (1978-1982) of frequent disturbance by a lugworm dredge of one of the 15 sampling stations involved in a long-term study of the dynamics of the macrozoobenthos on Balgzand allowed a study of long-term effects of mechanical lugworm digging. Within an area of about 1 km 2, a near-doubling of the annual lugworm mortality rate resulted in a gradual and substantial decline of the local lugworm stock from more than twice the overall Balgzand mean at the start of the 4-year digging period to a value close to this mean at the end of the period (when the dredge moved to a richer area). Simultaneously, total zoobenthic biomass declined even more by the almost complete extinction of the population of larger gaper clams Mya arenaria that initially comprised half of the total biomass. Of the other, mostly short-lived, species only Heteromastus filiformis showed a clear reduction during the dredging period. Recovery of the biomass of the benthos took several years, particularly by the slow re-establishment of a Mya population with a normal size and age structure.
Ultramicroscopic examination of the ovine tonsillar epithelia.
Casteleyn, Christophe; Cornelissen, Maria; Simoens, Paul; Van den Broeck, Wim
2010-05-01
As solid morphological knowledge of ovine tonsillar epithelia might contribute to a better understanding of the pathogenesis of several diseases including prion diseases, the epithelia of all tonsils of 7 one-year-old Texel sheep were examined using scanning and transmission electron microscopy. Major parts of the pharyngeal and tubal tonsils were covered by pseudostratified columnar ciliated epithelia that were interrupted by patches of epithelium containing cells with densely packed microfolds or microvilli, and cells with both microvilli and cilia. Smaller parts were covered by either flattened polygonal cells with densely packed microvilli or microfolds, squamous epithelial cells, or patches of reticular epithelium. The palatine and paraepiglottic tonsils were mainly lined by squamous epithelial cells with apical microplicae or short knobs. Additionally, regions of reticular epithelium containing epithelial cells with apical microvilli were seen. The lingual tonsil was uniformly covered by a keratinized squamous epithelium and devoid of microvillous cells and patches of reticular epithelium. The rostral half of the tonsil of the soft palate was lined by a pseudostratified columnar ciliated epithelium with characteristics of the pharyngeal and tubal tonsils. The epithelium of the caudal part resembled the epithelia of the palatine and paraepiglottic tonsils. Putative M cells, mainly characterized by apical microvilli or microfolds and a close association with lymphoid cells, seem manifestly present on the nasopharyngeal tonsils. The reticular epithelium of the palatine and paraepiglottic tonsils also harbor cells with small apical microvilli. The exact nature of these presumptive M cells should, however, be elucidated in functional studies.
