  1. Overexpression of soluble ADAM33 promotes a hypercontractile phenotype of the airway smooth muscle cell in rat

    A disintegrin and metalloproteinase 33 (ADAM33) has been identified as a susceptibility gene for asthma, but details of the causality are not fully understood. We hypothesize that soluble ADAM33 (sADAM33) overexpression can alter the mechanical behaviors of airway smooth muscle cells (ASMCs) via regulation of the cell's contractile phenotype, and thus contributes to airway hyperresponsiveness (AHR) in asthma. To test this hypothesis, we either overexpressed or knocked down the sADAM33 in rat ASMCs by transfecting the cells with sADAM33 coding sequence or a small interfering RNA (siRNA) that specifically targets the ADAM33 disintegrin domain, and subsequently assessed the cells formore » stiffness, contractility and traction force, together with the expression level of contractile and proliferative phenotype markers. We also investigated whether these changes were dependent on Rho/ROCK pathway by culturing the ASMCs either in the absence or presence of ROCK inhibitor (H1152). The results showed that the ASMCs with sADAM33 overexpression were stiffer and more contractile, generated greater traction force, exhibited increased expression levels of contractile phenotype markers and markedly enhanced Rho activation. Furthermore these changes were largely attenuated when the cells were cultured in the presence of H-1152. However, the knock-down of ADAM33 seemed insufficient to influence majority of the mechanical behaviors of the ASMCs. Taken together, we demonstrated that sADAM33 overexpression altered the mechanical behaviors of ASMCs in vitro, which was most likely by promoting a hypercontractile phenotype transition of ASMCs through Rho/ROCK pathway. This revelation may establish the previously missing link between ADAM33 expression and AHR, and also provide useful insight for targeting sADAM33 in asthma prevention and therapy. - Highlights: • sADAM33 overexpression enhances the stiffness, traction force and contractility of ASMCs. • sADAM33 overexpression promotes a hypercontractile phenotype of ASMCs. • The hypercontractile phenotype transition was largely mediated via Rho/ROCK pathway. • ADAM33 knock-down had little effect on the stiffness, traction force and contractility of ASMCs.« less

    Ashra (All-sky Survey High Resolution Air-shower detector) is a project to build an unconventional optical telescope complex that images very wide field of view, covering 80% of the sky, yet with the angle pixel resolution of 1.2 arcsin, sensitive to the blue to UV light with the use of image intensifier and CMOS technology. The project primarily aims to observe Cherenkov and fluorescence lights from the lateral and longitudinal developments of very-high energy cosmic rays in the atmosphere. It can also be used to monitor optical transients in the wide field of sky. In 2004 we built prototype telescopes to verify and develop techniques at Haleakala in Hawaii, needed for the development of the full-scale telescopes. Construction of the main detector station has begun at Mauna Loa on the Hawaii Island in the summer of 2005. The pilot observation data have been taken. We will present the project status, and expected scientific impacts on the observational objectives such as optical transients, unidentified TeV gamma- ray and PeV neutrino sources, and the propagation of EeV cosmic rays.     During the studies of aeolian forms in the central part of the Lubaczów-Jaworów Plateau, in the borderland between Poland and Ukraine (Fig. 1), a special attention was given to lithologic features of deposits building a ridge dune, which is situated to the NE of Wola Arłamowa village. These deposits are characterized by distinct cyclic variability. This fact determined us to undertake investigations in order to reconstruct aerodynamic conditions under which such deposit variability had appeared. These investigations included: a) geomorphological mapping of the ridge dune and its immediate vicinity, b) identification of texture and structure of deposits building the form, c) measurements of scale and frequency of lithofacies, d) measurements of structural directional elements, and e) identification of primary deposition environments according to the classification by Hunter (1977) and Borówka (2001), described according to the suggestions published by Zieliński and Issmer (2008). Based on the lithologic features of dune building material (Figs 2, 3), we distinguished two deflation-accumulation cycles. The first one is recorded within the south-eastern slope of the dune, and it is characterized by the occurrence of sand sets with high-angle inclined stratification, which are separated by reactivation surfaces (Figs 2D, 3B). Sand was deposited on the leeward slope by wind from a predominant direction. The formation of reactivation surfaces is regarded as a result of the leeward slope exposure to winter strong wind from a direction completely different from the predominant one (McKee 1966; Borówka 1980; Hunter and Rubin 1983; Zieliński et al. in press). Therefore, we can assume that this change of wind direction occurred in annual rhythm. The second cycle was most completely recorded in the crest zone of the dune. It consists, from bottom to top, of the following lithofacies: RC→MR→PB→DT (Figs 2D, 3A). This cycle evidences wind from a slightly variable direction but with increasing velocity that resulted in increased deposition, and after exceeding the threshold value — increased deflation (Fig. 4). The nature of deposition can be compared with the described in literature wedge stratification (McKee 1966; McKee et al. 1971; Izmaiłow 2001). During this cycle the aerodynamic conditions changed in considerably shortened periods — from several days to several weeks. The authors suppose that this variability was associated with deepening, travelling to the east, and filling up of dynamic lows.

  5. The effect of concentration of tackifying agent on adhesive and skin-protective properties of ceramide 2-containing hydrocolloid dressings.

    In the treatment of pressure ulcers and leg ulcers it is necessary to achieve an effective balance between adhesive and skin-protective properties. We speculated that addition of a tackifying agent (TA) to ceramide 2-containing hydrocolloid dressings would increase their adhesiveness under dry conditions and reduce their adhesiveness under wet conditions because dry tack converts to wet tack after water absorption. We prepared ceramide 2-containing hydrocolloid dressings with varying amounts of TA. Basic characteristics of the test ceraminde dressings, such as initial tack force and peeling force, were evaluated using standard methods. Peeling force and stratum corneum (SC) removal on healthy human skin were also evaluated at 20 minutes, 7 hours, and 72 hours. In addition, the effect of 10 repeated applications on transepidermal water loss (TEWL) was investigated on the skin of hairless mice under dry and wet conditions. Statistical analyses were performed using one-way analysis of variance followed by Dunnett's multiple comparison test. A p-value of <0.05 was considered statistically significant. On a stainless steel substrate, initial tack force and 180° peeling force increased as TA content increased. Twenty minutes after application on human skin, peeling force and SC removal increased with increasing TA content. When TA contents were over 10%, significant differences in peeling force and SC removal were obtained compared with ceramide 2-containing hydrocolloid dressings without TA (p<0.05). However, a TA content-dependent increase in peeling force was not evident 7 hours and 72 hours after application. Under dry conditions, TEWL increased with repeated application and peeling. Conversely, no significant increases in TEWL were evident under wet conditions after 10 repeated applications and peelings. Our data demonstrate that the initial attachment of ceramide 2-containing hydrocolloid dressings to the skin increases with addition of TA.   6. Evaluation and Management of the Gas and Rock Outburst Hazard in the Light of International Legal Regulations

    As part of the present article, the Authors analyzed relevant legal rules that are in force in countries where the gas and coal outburst hazard occurs (Australia, the Czech Republic, China, Germany, Poland, Russia, and Ukraine). Similarities and differences between particular solutions were highlighted. As the subject of the analysis were the original legal standards, the article incorporates the parameter symbols and units adopted in the discussed regulations. W ramach niniejszej pracy poddano analizie obowiązujące rozwiązania prawne z krajów, gdzie występuje zagrożenie wyrzutami gazu i węgla (Australia, Czechy, Chiny, Niemcy, Polska, Rosja, Ukraina). Zwrócono uwagę na podobieństwa i różnice w poszczególnych rozwiązaniach. Ze względu na analizę oryginalnych rozwiązań legislacyjnych w opracowaniu pozostawione zostały stosowane w odpowiednich przepisach oznaczenia parametrów oraz ich jednostki. Istnieje duże podobieństwo w obrębie przepisów w krajach europejskich. Podobieństwo to dotyczy zarówno ogólnej koncepcji przepisów, w których dąży się do maksymalnego sformalizowania nakładanych wymogów, łącznie ze wskazaniem konkretnych parametrów górniczych oraz ich wartości kryterialnych, na bazie których klasyfikowane są pokłady węglowe, bądź ich części, do poszczególnych klas zagrożenia wyrzutowego. Podobne podejście w ustawodawstwie w zakresie bezpieczeństwa w górnictwie reprezentują Chiny. W całkowitej opozycji do przepisów europejskich i chińskich znajdują się przepisy amerykańskie i australijskie. W przepisach tych wyraźnie zaakcentowany jest fakt, iż charakter ryzyka wyrzutowego jest bardzo lokalny, więc trudno go uogólniać nie tylko między kopalniami, ale również w poszczególnych wyrobiskach. Jedno, niezmienne podejście do zarządzania ryzykiem, zgodnie z przepisami australijskimi, jest więc niewłaściwe. Przepisy te nie podają szczegółowego opisu, w jaki sposób prowadzić zarządzanie ryzykiem wyrzutu, ale dają natomiast zarys tego, jakie elementy muszą być uwzględnione w rozwoju zarządzaniu ryzykiem. Kategoryzacja zagrożenia wyrzutowego w krajach europejskich i w Chinach ma wiele cech wspólnych. Ustalane są najczęściej dwie, lub trzy kategorie zagrożenia. Często incydenty wyrzutów, bądź poważnych symptomów mogących świadczyć o zagrożeniu są podstawą do zaliczenia pokładu do najwyższej kategorii zagrożenia. Dominującym parametrem w podstawowej ocenie zagrożenia wyrzutowego jest zawartość metanu w pokładzie węgla. Występuje ona w aktach prawnych wszystkich krajów, które narzucają konkretne sposoby określana stanu zagrożenia, jedynie przepisy chińskie nie obligują do badań zawartości gazu w węglu. W przepisach chińskich pojawia się pomiar ciśnienia gazu.
