  1. The Bxb1 recombination system demonstrates heritable transmission of site-specific excision in Arabidopsis

    Background The mycobacteriophage large serine recombinase Bxb1 catalyzes site-specific recombination between its corresponding attP and attB recognition sites. Previously, we and others have shown that Bxb1 has catalytic activity in various eukaryotic species including Nicotiana tabacum, Schizosaccharomyces pombe, insects and mammalian cells. Results In this work, the Bxb1 recombinase gene was transformed and constitutively expressed in Arabidopsis thaliana plants harboring a chromosomally integrated attP and attB-flanked target sequence. The Bxb1 recombinase successfully excised the target sequence in a conservative manner and the resulting recombination event was heritably transmitted to subsequent generations in the absence of the recombinase transgene. In addition, we also show that Bxb1 recombinase expressing plants can be manually crossed with att-flanked target transgenic plants to generate excised progeny. Conclusion The Bxb1 large serine recombinase performs site-specific recombination in Arabidopsis thaliana germinal tissue, producing stable lines free of unwanted DNA. The precise site-specific deletion produced by Bxb1 in planta demonstrates that this enzyme can be a useful tool for the genetic engineering of plants without selectable marker transgenes or other undesirable exogenous sequences.   2. Transgene autoexcision in switchgrass pollen mediated by the Bxb1 recombinase

    Background Switchgrass (Panicum virgatum L.) has a great potential as a platform for the production of biobased plastics, chemicals and energy mainly because of its high biomass yield on marginal land and low agricultural inputs. During the last decade, there has been increased interest in the genetic improvement of this crop through transgenic approaches. Since switchgrass, like most perennial grasses, is exclusively cross pollinating and poorly domesticated, preventing the dispersal of transgenic pollen into the environment is a critical requisite for the commercial deployment of this important biomass crop. In this study, the feasibility of controlling pollen-mediated gene flow in transgenic switchgrass using the large serine site-specific recombinase Bxb1 has been investigated. Results A novel approach utilizing co-transformation of two separate vectors was used to test the functionality of the Bxb1/att recombination system in switchgrass. In addition, two promoters with high pollen-specific activity were identified and thoroughly characterized prior to their introduction into a test vector explicitly designed for both autoexcision and quantitative analyses of recombination events. Our strategy for developmentally programmed precise excision of the recombinase and marker genes in switchgrass pollen resulted in the generation of transgene-excised progeny. The autoexcision efficiencies were in the range of 22-42% depending on the transformation event and assay used. Conclusion The results presented here mark an important milestone towards the establishment of a reliable biocontainment system for switchgrass which will facilitate the development of this crop as a biorefinery feedstock through advanced biotechnological approaches.   3. Accuracy and efficiency define Bxb1 integrase as the best of fifteen candidate serine recombinases for the integration of DNA into the human genome

    Background Phage-encoded serine integrases, such as φC31 integrase, are widely used for genome engineering. Fifteen such integrases have been described but their utility for genome engineering has not been compared in uniform assays. Results We have compared fifteen serine integrases for their utility for DNA manipulations in mammalian cells after first demonstrating that all were functional in E. coli. Chromosomal recombination reporters were used to show that seven integrases were active on chromosomally integrated DNA in human fibroblasts and mouse embryonic stem cells. Five of the remaining eight enzymes were active on extra-chromosomal substrates thereby demonstrating that the ability to mediate extra-chromosomal recombination is no guide to ability to mediate site-specific recombination on integrated DNA. All the integrases that were active on integrated DNA also promoted DNA integration reactions that were not mediated through conservative site-specific recombination or damaged the recombination sites but the extent of these aberrant reactions varied over at least an order of magnitude. Bxb1 integrase yielded approximately two-fold more recombinants and displayed about two fold less damage to the recombination sites than the next best recombinase; φC31 integrase. Conclusions We conclude that the Bxb1 and φC31 integrases are the reagents of choice for genome engineering in vertebrate cells and that DNA damage repair is a major limitation upon the utility of this class of site-specific recombinase.     Microwave spectroscopy has been restricted to the investigation of small molecules in the last years. However, with the advent of FTMW and CP-FTMW spectroscopies coupled with laser vaporization techniques it has turned into a very competitive methodology in the studies of moderate-size molecules. In particular, the studies of relatively large molecular aggregates are very interesting, being a bridge between microsystems and molecular bulk. Here, we present the study of two pentarmers of difluoromethane (CH_2F_2)_5 and the water clusters (CH_2F_2)_1...(H_2O)_2, (CH_2F_2)_2...(H_2O)_1 and (CH_2F_2)_2...(H_2O)_2 stabilized by weak hydrogen bonds networks (O-H...F, C-H...F and C-H...O interactions).   5. Whole Tibetan Hull-Less Barley Exhibit Stronger Effect on Promoting Growth of Genus Bifidobacterium than Refined Barley In Vitro.

    The gut microbiota has recently become a new route for research at the intersection of diet and human health. The aim of this study was to investigate whether whole Tibetan hull-less barley (WHB) and refined Tibetan hull-less barley (RHB) caused differentiation of the fecal microbiota in vitro. The microbiota-accessible ingredients in the 2 barley samples were studied using an in vitro enzymatic digestion procedure. After in vitro digestion, insoluble dietary fiber, phenolic compounds, proteins, and β-glucans were 93.2%, 103.4%. 18.8%, and 10.2% higher provided by WHB flour as compared with RHB flour based on the same mass amount. However, due to the significantly higher content of insoluble dietary fiber, WHB digesta had lower percentage contents of fast fermentable substrates including dietary fiber and starch as compared with RHB digesta. The results of Next-generation sequencing of the bacterial 16SrRNA gene showed that both WHB and RHB fermentation had significantly promoted the growth of Bifidobacterium and inhibited the growth of pathogenic bacteria such as Dorea, Escherichia, Oscillopira, and Ruminococcus. Moreover, in response to WHB fermentation, the relative abundance of Bifidobacterium increased by 78.5% and 92.8% as compared with RHB and fructo-oligosaccharides (FOs). Both WHB and RHB are good sources of fermentable dietary fiber with the ability to yield high concentration of short chain fatty acids (SCFAs) as compared to FOs. However, the higher fraction of soluble fiber in RHB digesta increase higher amounts of SCFA compared with WHB digesta. Our findings shed light on the complex interactions of whole cereals with gut microbiota and the possible impact on host health. Until now, only few reports have regarded the impact of in vitro digestion in components of whole grain with complex food matrix.   6. Force Field Model of Periodic Trends in Biomolecular Halogen Bonds

    The study of the noncovalent interaction now defined as a halogen bond (X-bond) has become one of the fastest growing areas in experimental and theoretical chemistry—its applications as a design tool are highly extensive. The significance of the interaction in biology has only recently been recognized, but has now become important in medicinal chemistry. We had previously derived a set of empirical potential energy functions to model the structure-energy relationships for bromines in biomolecular X-bonds (BXBs). Here, we have extended this force field for BXBs (ffBXB) to the halogens (Cl, Br, and I) that are commonly seen to form stable X-bonds. The ffBXB calculated energies show a remarkable one-to-one linear relationship to explicit BXB energies determined from an experimental DNA junction system, thereby validating the approach and the model. The resulting parameters allow us to interpret the stabilizing effects of BXBs in terms of well-defined physical properties of the halogen atoms, including their size, shape, and charge, showing periodic trends that are predictable along the Group VII column of elements. Consequently, we have established the ffBXB as accurate computational tool that can be applied to, for example, for the design of new therapeutic compounds against clinically important targets and new biomolecular based materials.     The Waste Handling Building Ventilation System provides heating, ventilation, and air conditioning (HVAC) for the contaminated, potentially contaminated, and uncontaminated areas of the Monitored Geologic Repository's (MGR) Waste Handling Building (WHB). In the uncontaminated areas, the non-confinement area ventilation system maintains the proper environmental conditions for equipment operation and personnel comfort. In the contaminated and potentially contaminated areas, in addition to maintaining the proper environmental conditions for equipment operation and personnel comfort, the contamination confinement area ventilation system directs potentially contaminated air away from personnel in the WHB and confines the contamination within high-efficiency particulate air (HEPA) filtration units. Themore » contamination confinement areas ventilation system creates airflow paths and pressure zones to minimize the potential for spreading contamination within the building. The contamination confinement ventilation system also protects the environment and the public by limiting airborne releases of radioactive or other hazardous contaminants from the WHB. The Waste Handling Building Ventilation System is designed to perform its safety functions under accident conditions and other Design Basis Events (DBEs) (such as earthquakes, tornadoes, fires, and loss of the primary electric power). Additional system design features (such as compartmentalization with independent subsystems) limit the potential for cross-contamination within the WHB. The system provides status of important system parameters and equipment operation, and provides audible and/or visual indication of off-normal conditions and equipment failures.     Polar bears (Ursus maritimus) are being impacted by climate change and increased exposure to pollutants throughout their northern circumpolar range. In this study, we quantified concentrations of total mercury (THg) in the hair of polar bears from Canadian high- (southern Beaufort Sea, SBS) and sub- (western Hudson Bay, WHB) Arctic populations. Concentrations of THg in polar bears from the SBS population (14.8 ± 6.6 μg g(-1)) were significantly higher than in polar bears from WHB (4.1 ± 1.0 μg g(-1)). On the basis of δ(15)N signatures in hair, in conjunction with published δ(15)N signatures in particulate organic matter and sediments, we estimated that the pelagic and benthic food webs in the SBS are ∼ 4.7 and ∼ 4.0 trophic levels long, whereas in WHB they are only ∼ 3.6 and ∼ 3.3 trophic levels long. Furthermore, the more depleted δ(13)C ratios in hair from SBS polar bears relative to those from WHB suggests that SBS polar bears feed on food webs that are relatively more pelagic (and longer), whereas polar bears from WHB feed on those that are relatively more benthic (and shorter). Food web length and structure accounted for ∼ 67% of the variation we found in THg concentrations among all polar bears across both populations. The regional difference in polar bear hair THg concentrations was also likely due to regional differences in water-column concentrations of methyl Hg (the toxic form of Hg that biomagnifies through food webs) available for bioaccumulation at the base of the food webs. For example, concentrations of methylated Hg at mid-depths in the marine water column of the northern Canadian Arctic Archipelago were 79.8 ± 37.3 pg L(-1), whereas, in HB, they averaged only 38.3 ± 16.6 pg L(-1). We conclude that a longer food web and higher pelagic concentrations of methylated Hg available to initiate bioaccumulation in the BS resulted in higher concentrations of THg in polar bears from the SBS region compared to those inhabiting the western

    Knowledge is limited regarding methylsulfone (MeSO{sub 2})-polychlorinated biphenyl (PCB), and especially MeSo{sub 2}-2,2-bis(chlorophenyl)-1,1-dichloroethylene (DDE), metabolites in cetacean species. The authors hypothesized that the ability of beluga whale (Delphinapterus leucas) to biotransform PCB and DDE compounds, and to form and degrade their MeSO{sub 2}-PCB and -DDE metabolites, is related to the capacity for xenobiotic metabolism. Adipose biopsies were collected from male and female beluga whale from distinct populations in the St. Lawrence River estuary (STL) and western Hudson Bay (WHB), Canada, which are contrasted by the exposure to different levels of cytochrome P450 enzyme-inducing, chlorinated hydrocarbon contaminants. The PCBs, DDTs, DDEs,more » 28 MeSO{sub 2} metabolites of 14 meta-para chlorine-unsubstituted PCBs, and four MeSO{sub 2} metabolites of 4,4{prime}- and 2,4{prime}-DDE were determined. The mean concentrations of total ({Sigma}-) MeSO{sub 2}-PCB in male STL beluga (230 ng/g), and ratios of {Sigma}-MeSO{sub 2}-PCB to {Sigma}-PCB (0.05) and {Sigma}-precursor-PCB (0.17) were approximately twofold higher, whereas the {Sigma}-precursor-PCB to {Sigma}-PCB ratio was approximately twofold lower, than in male WHB beluga. Both populations had a low formation capacity for MeSO{sub 2}-PCBs with {le} six chlorines (<4% of {Sigma}-MeSO{sub 2}-PCBs). The congener patterns were dominated by trichloro- and tetrachloro-MeSO{sub 2}-PCBs, and tetrachloro- and pentachloro-MeSO{sub 2}-PCBs in WHB and STL animals, respectively. In addition to 2- and 3-MeSO{sub 2}-4,4{prime}-DDE, two unknown MeSO{sub 2}-2,4{prime}-DDEs were detected. The mean 3-MeSO{sub 2}-4,4{prime}-DDE concentration in STL beluga (1.2 ng/g) was much greater than in WHB animals. The concentrations of 4,4{prime}-DDE, and not 3-MeSO{sub 2}-4,4{prime}-DDE, increased with age in male STL animals. Individual differences in male sexual behavior, independent of gonadal steroids, are prevalent across a wide range of species, including man. However, the genetic mechanisms underlying steroid-independent male sexual behavior are poorly understood. A high proportion of B6D2F1 hybrid male mice demonstrates steroid-independent male sexual behavior (identified as "maters"), providing a mouse model that opens up avenues of investigation into the mechanisms regulating male sexual behavior in the absence of gonadal hormones. Recent studies have revealed several proteins that play a significant factor in regulating steroid-independent male sexual behavior in B6D2F1 male mice, including amyloid precursor protein (APP), tau, and synaptophysin. The specific goals of our study were to determine whether steroid-independent male sexual behavior was a heritable trait by determining if it was dependent upon the behavioral phenotype of the B6D2F1 sire, and whether the differential expression of APP, tau, and synaptophysin in the medial preoptic area found in the B6D2F1 sires that did and did not mate after gonadectomy was similar to those found in their male offspring. After adult B6D2F1 male mice were bred with C57BL/6J female mice, they and their male offspring (BXB1) were orchidectomized and identified as either maters or "non-maters". A significant proportion of the BXB1 maters was sired only from B6D2F1 maters, indicating that the steroid-independent male sexual behavior behavioral phenotype of the B6D2F1 hybrid males, when crossed with C57BL/6J female mice, is inherited by their male offspring. Additionally, APP, tau, and synaptophysin were elevated in in the medial preoptic area in both the B6D2F1 and BXB1 maters relative to the B6D2F1 and BXB1 non-maters, respectively, suggesting a potential genetic mechanism for the inheritance of steroid-independent male sexual behavior.     Adequate concentrations of ATP are required to preserve physiological cell functions and protect tissue from hypoxic damage. Decreased oxygen concentration results in ATP synthesis relying increasingly on the presence of phosphocreatine. The lack of ATP through hypoxic insult to neurons that generate or regulate respiratory function, would lead to the cessation of breathing (apnea). It is not clear whether creatine plays a role in maintaining respiratory phrenic nerve (PN) activity during hypoxic challenge. The aim of the study was to test the effects of exogenously applied creatine or creatine pyruvate in maintaining PN induced respiratory rhythm against the deleterious effects of severe hypoxic insult using Working Heart-Brainstem (WHB) preparations of juvenile Swiss type mice. WHB's were perfused with control perfusate or perfusate containing either creatine [100μM] or creatine pyruvate [100μM] prior to hypoxic challenge and PN activity recorded throughout. Results showed that severe hypoxic challenge resulted in an initial transient increase in PN activity, followed by a reduction in that activity leading to respiratory apnea. The results demonstrated that perfusing the WHB preparation with creatine or creatine pyruvate, significantly reduced the onset of apnea compared to control conditions, with creatine pyruvate being the more effective substance. Overall, creatine and creatine pyruvate each produced time-dependent degrees of protection against severe hypoxic-induced disturbances of PN activity.     The Site Generated Radiological Waste Handling System handles radioactive waste products that are generated at the geologic repository operations area. The waste is collected, treated if required, packaged for shipment, and shipped to a disposal site. Waste streams include low-level waste (LLW) in solid and liquid forms, as-well-as mixed waste that contains hazardous and radioactive constituents. Liquid LLW is segregated into two streams, non-recyclable and recyclable. The non-recyclable stream may contain detergents or other non-hazardous cleaning agents and is packaged for shipment. The recyclable stream is treated to recycle a large portion of the water while the remaining concentrated wastemore » is packaged for shipment; this greatly reduces the volume of waste requiring disposal. There will be no liquid LLW discharge. Solid LLW consists of wet solids such as ion exchange resins and filter cartridges, as-well-as dry active waste such as tools, protective clothing, and poly bags. Solids will be sorted, volume reduced, and packaged for shipment. The generation of mixed waste at the Monitored Geologic Repository (MGR) is not planned; however, if it does come into existence, it will be collected and packaged for disposal at its point of occurrence, temporarily staged, then shipped to government-approved off-site facilities for disposal. The Site Generated Radiological Waste Handling System has equipment located in both the Waste Treatment Building (WTB) and in the Waste Handling Building (WHB). All types of liquid and solid LLW are processed in the WTB, while wet solid waste from the Pool Water Treatment and Cooling System is packaged where received in the WHB. There is no installed hardware for mixed waste. The Site Generated Radiological Waste Handling System receives waste from locations where water is used for decontamination functions.     This study presented the construct of Work-Health Balance (WHB) and the design and validation of the Work-Health Balance Questionnaire (WHBq). More and more workers have a long-standing health problem or disability (LSHPD). The management of health needs and work demands is crucial for the quality of working life and work retention of these workers. However, no instrument exists measuring this process. The WHBq assesses key factors in the process of adjusting between health needs and work demands. We tested the reliability and validity of 38 items with cross-sectional data from a sample of 321 Italian workers (mean age = 45 ± 11 years) using exploratory factor analysis (EFA), Rasch analyses, and the correlations with other relevant variables. The instrument ultimately consisted of 17 items that reliably measured three factors: work-health incompatibility, health climate, and external support. These dimensions were associated with well-being in the workplace, dysfunctional behaviors at work, and general psychological health. A higher level on the WHB index was associated with lower levels of presenteeism, emotional exhaustion, workaholism, and psychological distress and with higher levels of job satisfaction and work engagement, supporting the construct validity of the instrument. The WHBq shows good psychometric characteristics and strong and theoretically consistent relationships with important and well-known variables.     To evaluate relevant optical properties of esthetic direct restorative materials focusing on whitened and translucent shades. Enamel (E), body (B), dentin (D), translucent (T) and whitened (Wh) shades for E (WhE) and B (WhB) from a restorative system (Filtek Supreme XTE, 3M ESPE) were evaluated. Samples (1 mm thick) were prepared. Spectral reflectance (R%) and color coordinates (L*, a*, b*, C* and h°) were measured against black and white backgrounds, using a spectroradiometer, in a viewing booth, with CIE D65 illuminant and d/0° geometry. Scattering (S) and absorption (K) coefficients and transmittance (T%) were calculated using Kubelka-Munk's equations. Translucency (TP) and opalescence (OP) parameters and whiteness index (W*) were obtained from differences of CIELAB color coordinates. R%, S, K and T% curves from all shades were compared using VAF (Variance Accounting For) coefficient with Cauchy-Schwarz inequality. Color coordinates and optical parameters were statistically analyzed using one-way ANOVA, Tukey's test with Bonferroni correction (α=0.0007). Spectral behavior of R% and S were different for T shades. In addition, T shades showed the lowest R%, S and K values, as well as the highest T%, TP an OP values. In most cases, WhB shades showed different color and optical properties (including TP and W*) than their corresponding B shades. WhE shades showed similar mean W* values and higher mean T% and TP values than E shades. When using whitened or translucent composites, the final color is influenced not only by the intraoral background but also by the color and optical properties of multilayers used in the esthetic restoration. The system provides the capability to transfer between normal and emergency power. The system provides emergency power via independent and physically separated distribution feeds from the normal supply.   18. AIR DISPERSION MODELING AT THE WASTE ISOLATION PILOT PLANT

    One concern at the Waste Isolation Pilot Plant (WIPP) is the amount of alpha-emitting radionuclides or hazardous chemicals that can become airborne at the facility and reach the Exclusive Use Area boundary as the result of a release from the Waste Handling Building (WHB) or from the underground during waste emplacement operations. The WIPP Safety Analysis Report (SAR), WIPP RCRA Permit, and WIPP Emergency Preparedness Hazards Assessments include air dispersion calculations to address this issue. Meteorological conditions at the WIPP facility will dictate direction, speed, and dilution of a contaminant plume of respirable material due to chronic releases or duringmore » an accident. Due to the paucity of meteorological information at the WIPP site prior to September 1996, the Department of Energy (DOE) reports had to rely largely on unqualified climatic data from the site and neighboring Carlsbad, which is situated approximately 40 km (26 miles) to the west of the site. This report examines the validity of the DOE air dispersion calculations using new meteorological data measured and collected at the WIPP site since September 1996. The air dispersion calculations in this report include both chronic and acute releases. Chronic release calculations were conducted with the EPA-approved code, CAP88PC and the calculations showed that in order for a violation of 40 CFR61 (NESHAPS) to occur, approximately 15 mCi/yr of 239Pu would have to be released from the exhaust stack or from the WHB. This is an extremely high value. Hence, it is unlikely that NESHAPS would be violated. A site-specific air dispersion coefficient was evaluated for comparison with that used in acute dose calculations. The calculations presented in Section 3.2 and 3.3 show that one could expect a slightly less dispersive plume (larger air dispersion coefficient) given greater confidence in the meteorological data, i.e. 95% worst case meteorological conditions.   1. A Novel System for Simultaneous or Sequential Integration of Multiple Gene-Loading Vectors into a Defined Site of a Human Artificial Chromosome

    Human artificial chromosomes (HACs) are gene-delivery vectors suitable for introducing large DNA fragments into mammalian cells. Although a HAC theoretically incorporates multiple gene expression cassettes of unlimited DNA size, its application has been limited because the conventional gene-loading system accepts only one gene-loading vector (GLV) into a HAC. We report a novel method for the simultaneous or sequential integration of multiple GLVs into a HAC vector (designated as the SIM system) via combined usage of Cre, FLP, Bxb1, and φC31 recombinase/integrase. As a proof of principle, we first attempted simultaneous integration of three GLVs encoding EGFP, Venus, and TdTomato into a gene-loading site of a HAC in CHO cells. These cells successfully expressed all three fluorescent proteins. Furthermore, microcell-mediated transfer of HACs enabled the expression of those fluorescent proteins in recipient cells. We next demonstrated that GLVs could be introduced into a HAC one-by-one via reciprocal usage of recombinase/integrase. Lastly, we introduced a fourth GLV into a HAC after simultaneous integration of three GLVs by FLP-mediated DNA recombination.     Abstract Engineering mammalian cell lines that stably express many transgenes requires the precise insertion of large amounts of heterologous DNA into well-characterized genomic loci, but current methods are limited. To facilitate reliable large-scale engineering of CHO cells, we identified 21 novel genomic sites that supported stable long-term expression of transgenes, and then constructed cell lines containing one, two or three ‘landing pad’ recombination sites at selected loci. By using a highly efficient BxB1 recombinase along with different selection markers at each site, we directed recombinase-mediated insertion of heterologous DNA to selected sites, including targeting all three with a single transfection. We used this method to controllably integrate up to nine copies of a monoclonal antibody, representing about 100 kb of heterologous DNA in 21 transcriptional units. Because the integration was targeted to pre-validated loci, recombinant protein expression remained stable for weeks and additional copies of the antibody cassette in the integrated payload resulted in a linear increase in antibody expression. Overall, this multi-copy site-specific integration platform allows for controllable and reproducible insertion of large amounts of DNA into stable genomic sites, which has broad applications for mammalian synthetic biology, recombinant protein production and biomanufacturing.   4. Preliminary Studies of the Immunomodulator Effect of the Bougainvillea xbuttiana Extract in a Mouse Model

    Bougainvillea xbuttiana is used as an analgesic in folk medicine in Mexico. The purpose of the present study was to determine the effects of the ethanolic extract from B. xbuttiana on macrophages activities. The phytochemical screening was performed for determine the presence of alkaloids, flavonoids, triterpenes, and saponins. The effects of B. xbuttiana were analyzed using the macrophages activities as determined by the H2O2 release, spreading and phagocytic index, vacuoles formation percentage, and mediators production. The viability percentage was determined in live cells after fixing and staining with crystal violet. The presence of H2O2 in macrophages was performed by using the peroxidase-phenol red solution. The cytokine production was determined by two assays, ELISA for detection of IL-6, IL-10, and IFN-γ and biological assay for TNF detection. The results showed that the Bxb extract dose-dependent manner produces (a) an increase in levels of H2O2 and spreading and vacuoles formation percentages, (b) a decrease in phagocytic index and in the amounts of TNF, IL-6, and IFN-γ, and (c) an increase significant in IL-10 and NO production. This study indicates that the ethanolic extract from Bougainvillea xbuttiana was able to activate macrophages. The combination of these results suggests that this extract has an immunomodulator effect. PMID:25861362

  5. Enrichment of PCDDs/PCDFs in the cooling system of municipal solid waste incineration plants.

    This study measured the levels of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDDs/PCDFs), destroyed or formed in combustors and re-synthesized in cooling systems. For the proper control of PCDDs/PCDFs in municipal solid waste (MSW) incinerators, three grate-type MSW incinerators were selected, two of which had boilers, and one of which had a water spray tower (WST) as a cooling system. At the combustor outlets, dusts were in the range of 1640-4270 mg/Sm3 and PCDDs/PCDFs were in the range of 0.103-2.619 ng-TEQ/Sm3, showing the different values according to the grate structure of combustor and the flow direction of flue gas. After the flue gases passed through the cooling system, PCDDs/PCDFs at the waste heat boiler (WHB) outlets were enriched to levels that were 10.8-13.6 times higher than those at the furnace outlets, but PCDDs/PCDFs at the WST outlet was reduced to 5% of the level found at the furnace outlet. The emission patterns, such as the ratio of PCDFs to PCDDs, the ratio of gaseous-phase to particulate-phase PCDDs/PCDFs, and the compositional percentiles of each 2,3,7,8-substituted congener varied according to the types of air pollution control devices (APCDs). Reducing re-synthesis in the cooling system rather than enhancing the removal efficiencies of the APCDs seems to be more effective for lowering the levels of PCDDs/PCDFs in MSW incineration plants.

  6. Anti-parallel versus Component Reconnection at the Earth Magnetopause

    Magnetic reconnection at the Earth's magnetopause is discussed and has been observed as anti-parallel and component reconnection. While anti-parallel reconnection occurs between magnetic field lines of (ideally) exactly opposite polarity, component reconnection (also known as the tilted X-line model) predicts the location of the reconnection line to be anchored at the sub-solar point and extend continuously along the dayside magnetopause, while the ratio of the IMF By/Bz component determines the tilt of the X-line relative to the equatorial plane.A reconnection location prediction model known as the Maximum Magnetic Shear Model combines these two scenarios. The model predicts that during dominant IMF By conditions, magnetic reconnection occurs along an extended line across the dayside magnetopause but generally not through the sub-solar point (as predicted in the original tilted X-line model). Rather, the line follows the ridge of maximum magnetic shear across the dayside magnetopause. In contrast, for dominant IMF Bz (155° < tan-1(By/Bz) < 205°) or dominant Bx (|Bx|/B > 0.7) conditions, the reconnection location bifurcates and traces to high-latitudes, in close agreement with the anti-parallel reconnection scenario, and does not cross the dayside magnetopause as a single tilted reconnection line. Using observations from the Magnetospheric MultiScale missions during a magnetopause crossing when the IMF rotated from an dominate IMF BZ to a dominant IMF BY field we will investigate when the transition between the anti-parallel and tilted X-line scenarios occurs.

  7. A Role for the p38 Mitogen-activated Protein Kinase Pathway in Myocardial Cell Growth, Sarcomeric Organization, and Cardiac-specific Gene Expression

    Three hallmark features of the cardiac hypertrophic growth program are increases in cell size, sarcomeric organization, and the induction of certain cardiac-specific genes. All three features of hypertrophy are induced in cultured myocardial cells by α1- adrenergic receptor agonists, such as phenylephrine (PE) and other growth factors that activate mitogen- activated protein kinases (MAPKs). In this study the MAPK family members extracellular signal–regulated kinase (ERK), c-jun NH2-terminal kinase (JNK), and p38 were activated by transfecting cultured cardiac myocytes with constructs encoding the appropriate kinases possessing gain-of-function mutations. Transfected cells were then analyzed for changes in cell size, sarcomeric organization, and induction of the genes for the A- and B-type natriuretic peptides (NPs), as well as the α-skeletal actin (α-SkA) gene. While activation of JNK and/or ERK with MEKK1COOH or Raf-1 BXB, respectively, augmented cell size and effected relatively modest increases in NP and α-SkA promoter activities, neither upstream kinase conferred sarcomeric organization. However, transfection with MKK6 (Glu), which specifically activated p38, augmented cell size, induced NP and α-Ska promoter activities by up to 130-fold, and elicited sarcomeric organization in a manner similar to PE. Moreover, all three growth features induced by MKK6 (Glu) or PE were blocked with the p38-specific inhibitor, SB 203580. These results demonstrate novel and potentially central roles for MKK6 and p38 in the regulation of myocardial cell hypertrophy. PMID:9314533

  8. Development of a high efficiency integration system and promoter library for rapid modification of Pseudomonas putida KT2440

    Pseudomonas putida strains are highly robust bacteria known for their ability to efficiently utilize a variety of carbon sources, including aliphatic and aromatic hydrocarbons. Recently, P. putida has been engineered to valorize the lignin stream of a lignocellulosic biomass pretreatment process. Nonetheless, when compared to platform organisms such as Escherichia coli, the toolkit for engineering P. putida is underdeveloped. Heterologous gene expression in particular is problematic. Plasmid instability and copy number variance provide challenges for replicative plasmids, while use of homologous recombination for insertion of DNA into the chromosome is slow and laborious. Furthermore, heterologous expression efforts to date typicallymore » rely on overexpression of exogenous pathways using a handful of poorly characterized promoters. In order to improve the P. putida toolkit, we developed a rapid genome integration system using the site-specific recombinase from bacteriophage Bxb1 to enable rapid, high efficiency integration of DNA into the P. putida chromosome. We also developed a library of synthetic promoters with various UP elements, -35 sequences, and -10 sequences, as well as different ribosomal binding sites. We tested these promoters using a fluorescent reporter gene, mNeonGreen, to characterize the strength of each promoter, and identified UP-element-promoter-ribosomal binding sites combinations capable of driving a ~150-fold range of protein expression levels. One additional integrating vector was developed that confers more robust kanamycin resistance when integrated at single copy into the chromosome. This genome integration and reporter systems are extensible for testing other genetic parts, such as examining terminator strength, and will allow rapid integration of heterologous pathways for metabolic engineering.« less

    Programmable Logic Controller or PLC today plays an important role in most industrial control systems. PLC usage can be encountered in almost all fields of industry, not only in the manufacturing world but also on many other things such as elevators in office buildings, hotels hospitals, and others. PLC is an electronic control tool that operates in logic that its programming can be modified with relative ease. As with any controller in general, the PLC processes input signals to further discharge output according to the desired program. PLC usage is very broad because of its high reliability, can be reprogrammed or modified with relative ease, and very helpful in the tracking troubleshooting. One type of existing PLC is Allen Bradley PLC. Allen Bradley PLC program is commonly used in various industries. PLC Allen Bradley (AB) has several types, and one of them is the type of Micrologic 1400. In this study we design a system as a comparison with the conventional system. For that to explore the use of a PLC program which will be supported by a simulator tool, including a program to RSLogic 500, how to programming, monitoring via RSView32, and modification. It is expected to understand the application aspect of operation and programming of this specific PLC and its potential. The purpose of this research is to design water filling automation system by using Allen Bradley Micrologic 1400 type 1766-L32BXB PLC, empowering the use of Allen Bradley Micrologic 1400 PLC and to regulate the desired process to obtain efficiency and effectiveness compared with conventional system arrangement using Relay.

    The Assembly Transfer System (ATS) receives, cools, and opens rail and truck transportation casks from the Carrier/Cask Handling System (CCHS). The system unloads transportation casks consisting of bare Spent Nuclear Fuel (SNF) assemblies, single element canisters, and Dual Purpose Canisters (DPCs). For casks containing DPCs, the system opens the DPCs and unloads the SNF. The system stages the assemblies, transfer assemblies to and from fuel-blending inventory pools, loads them into Disposal Containers (DCs), temporarily seals and inerts the DC, decontaminates the DC and transfers it to the Disposal Container Handling System. The system also prepares empty casks and DPCs formore » off-site shipment. Two identical Assembly Transfer System lines are provided in the Waste Handling Building (WHB). Each line operates independently to handle the waste transfer throughput and to support maintenance operations. Each system line primarily consists of wet and dry handling areas. The wet handling area includes a cask transport system, cask and DPC preparation system, and a wet assembly handling system. The basket transport system forms the transition between the wet and dry handling areas. The dry handling area includes the dry assembly handling system, assembly drying system, DC preparation system, and DC transport system. Both the wet and dry handling areas are controlled by the control and tracking system. The system operating sequence begins with moving transportation casks to the cask preparation area. The cask preparation operations consist of cask cavity gas sampling, cask venting, cask cool-down, outer lid removal, and inner shield plug lifting fixture attachment. Casks containing bare SNF (no DPC) are filled with water and placed in the cask unloading pool. The inner shield plugs are removed underwater.     Questions surround the mechanism of mutually exclusive expression by which Plasmodium falciparum mediates activation and silencing of var genes. These encode PfEMP1 proteins, which function as cytoadherent and immunomodulatory molecules at the surface of parasitized erythrocytes. Current evidence suggests that promoter silencing by var introns might play a key role in var gene regulation. To evaluate the impact of cis-acting regulatory regions on var silencing, we generated P. falciparum lines in which luciferase was placed under the control of an UpsA var promoter. By utilizing the Bxb1 integrase system, these reporter cassettes were targeted to a genomic region that was not in apposition to var sub-telomeric domains. This eliminated possible effects from surrounding telomeric elements and removed the variability inherent in episomal systems. Studies with highly synchronized parasites revealed that the UpsA element possessed minimal activity in comparison with a heterologous (hrp3) promoter. This may well result from the integrated UpsA promoter being largely silenced by the neighboring cg6 promoter. Our analyses also revealed that the DownsA 3’ untranslated region further decreased the luciferase activity from both cassettes, whereas the var A intron repressed the UpsA promoter specifically. By applying multivariate analysis over the entire cell cycle, we confirmed the significance of these cis-elements and found the parasite stage to be the major factor regulating UpsA promoter activity. Additionally, we observed that the UpsA promoter was capable of nucleating reversible silencing that spread to a downstream promoter. We believe these studies are the first to analyze promoter activity of Group A var genes which have been implicated in severe malaria, and support the model that var introns can further suppress var expression.     The Pool Water Treatment and Cooling System is located in the Waste Handling Building (WHB), and is comprised of various process subsystems designed to support waste handling operations. This system maintains the pool water temperature within an acceptable range, maintains water quality standards that support remote underwater operations and prevent corrosion, detects leakage from the pool liner, provides the capability to remove debris from the pool, controls the pool water level, and helps limit radiological exposure to personnel. The pool structure and liner, pool lighting, and the fuel staging racks in the pool are not within the scope of themore » Pool Water Treatment and Cooling System. Pool water temperature control is accomplished by circulating the pool water through heat exchangers. Adequate circulation and mixing of the pool water is provided to prevent localized thermal hotspots in the pool. Treatment of the pool water is accomplished by a water treatment system that circulates the pool water through filters, and ion exchange units. These water treatment units remove radioactive and non-radioactive particulate and dissolved solids from the water, thereby providing the water clarity needed to conduct waste handling operations. The system also controls pool water chemistry to prevent advanced corrosion of the pool liner, pool components, and fuel assemblies. Removal of radioactivity from the pool water contributes to the project ALARA (as low as is reasonably achievable) goals. A leak detection system is provided to detect and alarm leaks through the pool liner. The pool level control system monitors the water level to ensure that the minimum water level required for adequate radiological shielding is maintained. Through interface with a demineralized water system, adequate makeup is provided to compensate for loss of water inventory through evaporation and waste handling operations.     handling cell is located adjacent to the canister transfer cell and is interconnected to the transfer cell by means of the off-normal canister transfer tunnel.
