  1. Neuropharmacological and neuroprotective activities of some metabolites produced by cell suspension culture of Waltheria americana Linn.

    Waltheria americana is a plant used in Mexican traditional medicine to treat some nervous system disorders. The aims of the present study were to isolate and determine the neuropharmacological and neurprotective activities of metabolites produced by a cell suspension culture of Waltheria americana. Submerged cultivation of W. americana cells provided biomass. A methanol-soluble extract (WAsc) was obtained from biomass. WAsc was fractionated yielding the chromatographic fractions 4WAsc-H 2 O and WAsc-CH 2 Cl 2 . For the determination of anticonvulsant activity in vivo, seizures were induced in mice by pentylenetetrazol (PTZ). Neuropharmacological activities (release of gamma amino butyric acid (GABA) and neuroprotection) of chromatographic fractions were determined by in vitro histological analysis of brain sections of mice post mortem. Fraction 4WAsc-H 2 O (containing saccharides) did not produce neuronal damage, neurodegeneration, interstitial tissue edema, astrocytic activation, nor cell death. Pretreatment of animals with 4WAsc-H 2 O and WAsc-CH 2 Cl 2 from W. americana cell suspensions induced an increase in: GABA release, seizure latency, survival time, neuroprotection, and a decrease in the degree of severity of tonic/tonic-clonic convulsions, preventing PTZ-induced death of up to 100% of animals of study. Bioactive compounds produced in suspension cell culture of W. americana produce neuroprotective and neuropharmacological activities associated with the GABAergic neurotransmission system.   2. Genome size, cytogenetic data and transferability of EST-SSRs markers in wild and cultivated species of the genus Theobroma L. (Byttnerioideae, Malvaceae)

    The genus Theobroma comprises several trees species native to the Amazon. Theobroma cacao L. plays a key economic role mainly in the chocolate industry. Both cultivated and wild forms are described within the genus. Variations in genome size and chromosome number have been used for prediction purposes including the frequency of interspecific hybridization or inference about evolutionary relationships. In this study, the nuclear DNA content, karyotype and genetic diversity using functional microsatellites (EST-SSR) of seven Theobroma species were characterized. The nuclear content of DNA for all analyzed Theobroma species was 1C = ~ 0.46 pg. These species presented 2n = 20 with small chromosomes and only one pair of terminal heterochromatic bands positively stained (CMA+/DAPI− bands). The small size of Theobroma ssp. genomes was equivalent to other Byttnerioideae species, suggesting that the basal lineage of Malvaceae have smaller genomes and that there was an expansion of 2C values in the more specialized family clades. A set of 20 EST-SSR primers were characterized for related species of Theobroma, in which 12 loci were polymorphic. The polymorphism information content (PIC) ranged from 0.23 to 0.65, indicating a high level of information per locus. Combined results of flow cytometry, cytogenetic data and EST-SSRs markers will contribute to better describe the species and infer about the evolutionary relationships among Theobroma species.   4. Biological studies on Brazilian plants used in wound healing.

    n-Hexanic and ethanolic extracts from twelve plants (Brugmansia suaveolens Brecht. et Presl., Eupatorium laevigatum Lam., Galinsoga parviflora Cav., Iresine herbstii Hook., Kalanchöe tubiflora Hamet-Ahti, Petiveria alliacea L., Pluchea sagittalis (Lam.) Cabrera, Piper regnellii DC., Schinus molle L., Sedum dendroideum Moç et Sessé ex DC., Waltheria douradinha St. Hill., Xanthium cavanillesii Schouw.) used in traditional South Brazilian medicine as wound healing agents were investigated in various biological assays, targeting different aspects in this complex process. The extracts were investigated on NF-kappaB DNA binding, p38alpha MAPK, TNF-alpha release, direct elastase inhibition and its release as well as on caspase-3. Fibroblasts migration to and proliferation into the wounded monolayers were evaluated in the scratch assay, the agar diffusion test for antibacterial and the MTT assay for cytotoxic effects. The hydrophilic extracts from Galinsoga parviflora, Petiveria alliacea, Schinus molle, Waltheria douradinha and Xanthium cavanillesii as well as the lipophilic extract of Waltheria douradinha turned out to be the most active ones. These results increase our knowledge on the wound healing effects of the investigated medicinal plants. Further studies are necessary to find out the effective secondary metabolites responsible for the observed effects.

  5. Olfactory Response and Host Plant Feeding of the Central American Locust Schistocerca piceifrons piceifrons Walker to Common Plants in a Gregarious Zone.

    The Central American locust (CAL) Schistocerca piceifrons piceifrons Walker is one of the most harmful plant pests in the Yucatan Peninsula, where an important gregarious zone is located. The olfactory response and host plant acceptance by the CAL have not been studied in detail thus far. In this work, the olfactory response of the CAL to odor of various plant species was evaluated using an olfactometer test system. In addition, the host plant acceptance was assessed by the consumption of leaf area. Results showed that the CAL was highly attracted to odor of Pisonia aculeata. Evaluation of host plant acceptance showed that the CAL fed on Leucaena glauca and Waltheria americana, but not on P. aculeata or Guazuma ulmifolia. Analysis of leaf thickness, and leaf content of nitrogen (N) and carbon (C) showed that the CAL was attracted to plant species with low leaf C content.

  6. Xylocopa bees in tropical coastal sand dunes: use of resources and their floral syndromes.

    Large bees such as species from Xylocopa Latreille are usually associated with pollination in tropical sand dune areas, which frequently present shrubby herbaceous vegetation adapted to conditions of high salinity, high solar radiation and strong winds. We report on the diversity of Xylocopa and the plants they visited to collect nectar and pollen, focusing on the floral syndromes they present in these plants and on the breadth of the trophic niche in a tropical sand dune fragment over the year. The field work was carried out monthly in Baixio (Bahia, Brazil; Northern Coast Environmental Protection Area) from April 2008 to March 2009, over two consecutive days, from 06:30 AM to 05:00 PM. The medium-large body sized Xylocopa (Neoxylocopa) cearensis Ducke and Xylocopa (Schonnherria) subcyanea Pérez were noticeable for their frequency, constancy on the flowers and sharing of plant species. Xylocopa spp. visited plants with flowers of different shapes, colors, inflorescence arrangement and syndromes. However, their resource collections were mainly concentrated on Cuphea brachiata, Waltheria cinerascens, Croton sellowii and Chamaecrista ramosa, which may be considered key species for Xylocopa spp. maintenance in coastal sand dune and restinga environments in Northeast Brazil.

  7. Traditional medicine in north Côte-d'Ivoire: screening of 50 medicinal plants for antibacterial activity.

    Sixty-seven crude ethanol extracts from 50 plants (31 families), which are used in North Côte-d'Ivoire as traditional remedies for bacterial diseases, were screened for in vitro activity against Gram negative (Escherichia coli and Pseudomonas aeruginosa) and Gram positive (Staphylococcus aureus, Enterococcus faecalis, Streptococcus pyogenes and Bacillus subtilis) bacteria. Thirty-one extracts showed antibacterial activity only on Gram positive bacteria. Of these, 10 extracts from 10 plant species had a promising level of activity against bacteria including strains resistant to antibiotics such as aminosides, penicillin M, macrolides, lincosamide and streptrogramin B. The most active was Erythrina senegalensis DC (Fabaceae) followed by Bobgunnia madagascariensis (Desv.) J.H. Kirkbr. & Wiersema (Caesalpinaceae), Waltheria lanceolata R. Br. ex Mast. (Sterculiaceae), Uapaca togoensis Pax. (Euphorbiaceae), Ximenia americana L. (Olacaceae), Khaya senegalensis (Ders.) A. Juss. (Meliaceae), Lannea acida A. Rich. (Anacardiaceae), Cissus populnea Guill. & Perr. (Vitaceae), Keetia hispida (Benth.) Bridson (Rubiaceae) and Ficus thonningii (Miq.) A. Rich. (Moraceae). This is the first report of the antibacterial potency of these 10 plant species on a range of bacteria. The results provided evidence that some of the studied plants might indeed be potential sources of new antibacterial agents, also against some antibiotic-resistant strains.

  8. Do chromosome numbers reflect phylogeny? New counts for Bombacoideae and a review of Malvaceae s.l.

    • Whole genome duplication (WGD) and specific polyploidy events marked turning points for angiosperm genome structure and evolution. Therefore, cytogenetic studies of polyploidy-prone groups such as the tropical Malvaceae and plant formations such as as the Brazilian Cerrado have gained further importance. We present new chromosome counts for Cerrado Bombacoideae and revised chromosome numbers for the Malvaceae s.l., compare these between subfamilies, and relate them to phylogenetic signal.• We studied the chromosome number of Eriotheca candolleana, E. gracilipes, E. pubescens, Pachira glabra, Pseudobombax longiflorum, and P. tomentosum. We also compared Eriotheca species ploidy levels using flow cytometry. We compiled chromosome numbers for 557 species of Malvaceae s.l., including 37 Bombacoideae species. We included this information in a phylogenetic reconstruction based on chloroplast matK-trnK DNA to evaluate chromosome evolution of the Malvaceae s.l. and the Bombacoideae in particular.• The Cerrado Bombacoideae presented consistently high chromosome numbers. Numbers for Eriotheca species were among the highest and varied among populations. Flow cytometry analyses showed similar 1Cx DNA for all cytotypes and indicated neopolyploidy. Chromosome numbers differed between subfamilies, with the lowest numbers in the Malvoideae and Byttnerioideae and the highest in Tilioideae. Chromosome numbers had significant phylogenetic signal for Bombacoideae but not for Malvoideae or Malvaceae s.l.• Clearly distinct chromosome numbers allied to monophyly provide some support for a circumscription of the Bombacoideae and distinction within the Malvaceae. The phylogenetic signal for chromosome number supports the idea of an ancient WGD and further neopolyploidy events as important evolutionary trends for the Bombacoideae. © 2014 Botanical Society of America, Inc.

  9. Prediction of the Passive Intestinal Absorption of Medicinal Plant Extract Constituents with the Parallel Artificial Membrane Permeability Assay (PAMPA).

    At the early drug discovery stage, the high-throughput parallel artificial membrane permeability assay is one of the most frequently used in vitro models to predict transcellular passive absorption. While thousands of new chemical entities have been screened with the parallel artificial membrane permeability assay, in general, permeation properties of natural products have been scarcely evaluated. In this study, the parallel artificial membrane permeability assay through a hexadecane membrane was used to predict the passive intestinal absorption of a representative set of frequently occurring natural products. Since natural products are usually ingested for medicinal use as components of complex extracts in traditional herbal preparations or as phytopharmaceuticals, the applicability of such an assay to study the constituents directly in medicinal crude plant extracts was further investigated. Three representative crude plant extracts with different natural product compositions were chosen for this study. The first extract was composed of furanocoumarins (Angelica archangelica), the second extract included alkaloids (Waltheria indica), and the third extract contained flavonoid glycosides (Pueraria montana var. lobata). For each medicinal plant, the effective passive permeability values Pe (cm/s) of the main natural products of interest were rapidly calculated thanks to a generic ultrahigh-pressure liquid chromatography-UV detection method and because Pe calculations do not require knowing precisely the concentration of each natural product within the extracts. The original parallel artificial membrane permeability assay through a hexadecane membrane was found to keep its predictive power when applied to constituents directly in crude plant extracts provided that higher quantities of the extract were initially loaded in the assay in order to ensure suitable detection of the individual constituents of the extracts.
