Although the social and political changes accompanying the transition from the Neolithic through Copper Age, between the 4th and 3rd millennia cal BCE, in southwestern Iberia are reasonably well understood, much less is known about whether population movements and dietary changes accompanied these transformations. To address this question, human dental remains from the Middle through Late Neolithic site of Feteira II (3600-2900 cal BCE) and the Late Neolithic site of Bolores (2800-2600 cal BCE) in the Portuguese Estremadura were used to examine diet (microwear) and affinity (dental non-metrics). Microwear features were not found to be significantly different between Feteira II and Bolores, suggesting that the emergence of social complexity during this period did not result in large-scale changes in subsistence practices during the period of use at these sites. Using the Arizona State University Dental Anthropology System and supporting statistics, no significant difference between the samples from Feteira II and Bolores was observed, suggesting that no population replacement occurred between the Middle Neolithic and Late Neolithic/Copper Age. However, at Bolores there is some indication that there may have been demographic exchanges between southern Iberian and North African populations during the Late Neolithic/Copper Age. Problems of the dataware of the systems with the use of optoelectronic means of the linear displacement are considered in the article. The classification of the known physical effects, realized by the means of information-measuring systems, is given. The organized analysis of information flows in technical systems from the standpoint of determination of inaccuracies of measurement and management was conducted. In spite of achieved successes in automation of machine-building and instruments-building equipment in the field of dataware of the technical systems, there are unresolved problems, concerning the qualitative aspect of the production process. It was shown that the given problem can be solved using optoelectronic lazer information-measuring systems. Such information-measuring systems are capable of not only executing the measuring functions, but also solving the problems of management and control during processing, thereby guaranteeing the quality of final products.

  7. Physiological Monitoring of Optically Trapped Cells: Studying the Effects of Confinement by 1064 NM Lazer Tweezers Using Microfluorometry

    NASA Astrophysics Data System (ADS)

    Liu, Yagang

A novel technique that combines microfluorometric detection and optical laser trapping has been developed for in-situ assessing the physiological state of an optically trapped biological sample. This optical diagnostic technique achieves high sensitivity (>30 dB signal -to-noise ratio) and high spatial resolution (~ 1 μm) over a broad spectral range (>400 nm). The fluorescence spectra derived from exogenous fluorescent probes, including laurdan, acridine orange, propidium iodide and Snarf, are used to assess the effects of optical confinement with respect to temperature, DNA structure, cell viability, and intracellular pH, respectively. In the latter three cases, fluorescence is excited via a two-photon absorption process, using the cw laser trap itself as the fluorescence excitation source. This enables the cw near infrared laser trapping beam to be used simultaneously as an optical diagnostic probe as well as an optical micromanipulator. Using microfluorometry, a temperature increase of less than several degrees centigrade was measured for test samples, including liposomes, Chinese hamster ovary (CHO) cells and human sperm cells that were held stationary by 1064 nm optical tweezers having a power density of ~10^7 W/cm^2. Additional physiological monitoring experiments indicated that there is no observable denaturation of DNA, or change of intracellular pH under typical continuous wave laser trapping conditions (P <= 400 mW). Under some circumstances, however, it was possible to achieve a decrease in cell viability with cw trapping, as monitored by a live/dead vital stain. In comparison, significant DNA denaturation and cellular physiological changes (e.g. cell death) were observed when a Q-switched pulsed laser at a threshold of ~30mu J/pulse was used as trapping source. These results generally support the conclusion that cw laser trapping at 1064 nm wavelength is a safe, non-invasive process and should prove to be of great value for understanding the mechanisms of laser microirradiation effects on living cells held stationary in a near-infrared trapping beam.

Hepatocytes derived from embryonic stem cells (ESCs) are expected to be useful for basic research and clinical applications. However, in several studies, genetic methods used to detect and obtain them are difficult and pose major safety problems. Therefore, in this study, we established a novel detection system for hepatocytes by using indocyanine green (ICG), which is selectively taken up by hepatocytes, based on nongenetic manipulation. ICG has maximum light absorption near 780 nm, and it fluoresces between 800 and 900 nm. Making use of these properties, we developed flow cytometry equipped with an excitation lazer of 785 nm and specific bandpass filters and successfully detected ESC-derived ICG-positive cells that were periodic acid-Schiff positive and expressed hepatocyte phenotypic mRNAs. These results demonstrate that this detection system based on nongenetic manipulation with ICG will lead to isolate hepatocytes generated from ESCs and provide the appropriate levels of stability, quality, and safety required for cell source for cell-based therapy and pharmaceutical studies such as toxicology.

Cycle safety helmets are designed to prevent head injury. Although most commercially available helmets conform to one of several national and international standards, individual designs differ widely, particularly in relation to face coverage. A method was developed to assess the potential for the differing designs to protect the face from injury. A nonimpact test was assessed, using digitized image-processing software (Digithurst Ltd.) to measure the shadow cast by a helmet rim under a collimated plane light source onto the face of a mannequin headform. Twelve helmet designs available internationally were tested and ranked with respect to the direct protection conferred (area of the face directly covered by the helmet) and indirect protection (area of the face shaded). The three highest-ranking helmets for direct protection (Rosebank Stackhat, Asphalt Warrior, and Lazer Voyager) also ranked the highest for indirect protection. These helmets were more inferiorly extended and were of a more bulky construction. It was concluded that the dimensions of cycle helmets in relation to face coverage are crucial in influencing the extent to which facial protection is conferred. International test standards need urgent revision to ensure that face coverage is optimized. Lower-face protection could be achieved through incorporation of a lower-face bar to cycle helmets.

Recently, in Brazil, it has appeared a new and unusual "plague" in lazer and commercial fishing. It is caused by the parasitic larval phase of certain native bivalve mollusks of fresh water known as "Naiades" and its involves the presence of big bivalve of fresh water, mainly Anodontites trapesialis, in the tanks and dams of the fish creation. These bivalve mollusks belong to the Unionoida Order, Mycetopodidae Family. The objective of the present work was to analyze the shells of these mollusks to verify the possibility of use as bioindicators for heavy metals in freshwater. The mollusks shells were collected in a commercial fishing at Londrina-PR. A qualitative analysis was made to determine the chemical composition of the shells and verify a possible correlation with existent heavy metals in the aquatic environment. In the inner part of the shells were identified the elements Ca, P, Fe, Mn and Sr and in the outer part were identified Ca, P, Fe, Mn, Sr and Cu. The Ca ratio of the outer part by inner part of the analyzed shells is around of 1, as expected, because Ca is the main compound of mollusks shells. The ratio of P, Fe, Mn, and Sr to the Ca were constant in all analyzed shells, being close to 0.015. The ratio Cu/Ca varied among the shells, showing that this mollusk is sensitive to concentration of this element in the aquatic environment.

    2016-04-01

Four occurred debris flows in the Dolomites area (North-Eastern Italian Alps) are modeled by back-analysis. The four debris flows events are those occurred at Rio Lazer (Trento) on the 4th of November 1966, at Fiames (Belluno) on the 5th of July 2006, at Rovina di Cancia (Belluno) on the 18th of July 2009 and at Rio Val Molinara (Trento) on the 15th of August 2010. In all the events, runoff entrained sediments present on natural channels and formed a solid-liquid wave that routed downstream. The first event concerns the routing of debris flow on an inhabited fan. The second event the deviation of debris flow from the usual path due to an obstruction with the excavation of a channel in the scree and the downstream spreading in a wood. The third event concerns the routing of debris flow in a channel with an ending the reservoir, its overtopping and final spreading in the inhabited area. The fourth event concerns the routing of debris flow along the main channel downstream the initiation area until spreading just upstream a village. All the four occurred debris flows are simulated by modeling runoff that entrained debris flow for determining the solid-liquid hydrograph. The routing of the solid-liquid hydrograph is simulated by a bi-phase cell model based on the kinematic approach. The comparison between simulated and measured erosion and deposition depths is satisfactory. Nearly the same parameters for computing erosion and deposition were used for all the four occurred events. The maps of erosion and deposition depths are obtained by comparing the results of post-event surveys with the pre-event DEM. The post-event surveys were conducted by using different instruments (LiDAR and GPS) or the combination photos-single points depth measurements (in this last case it is possible obtaining the deposition/erosion depths by means of stereoscopy techniques).

Three occurred events of debris flows are documented and modeled by back-analysis. The three debris flows events are those occurred at Rio Lazer on the 4th of November 1966, at Fiames on the 5th of July 2006 and at Rovina di Cancia on the 18th of July 2009. All the three sites are located in the North-Eastern Italian Alps. In all the events, runoff entrained sediments present on natural channels and formed a solid-liquid wave that routed downstream. The first event concerns the routing of debris flow on an inhabited fan. Map of deposition pattern of sediments are built by using post-events photos through stereoscopy techniques. The second event concerns the routing of debris flow along the main channel descending from Pomagagnon Fork. Due to the obstruction of the cross-section debris flow deviated from the original path on the left side and routed downstream by cutting a new channel on the fan. It dispersed in multiple paths when met the wooden area. Map of erosion and deposition depths are built after using a combination of Lidar and GPS data. The third event concerns the routing of debris flow in the Rovina di Cancia channel that filled the reservoir built at the end of the channel and locally overtopped the retaining wall on the left side. A wave of mud and debris inundated the area downstream the overtopping point. Map of erosion and deposition depths are obtained by subtracting two GPS surveys, pre and post event. All the three occurred debris flows are simulated by modeling runoff that entrained debris flow for determining the solid-liquid hydrograph downstream the triggering areas. The routing of the solid-liquid hydrograph was simulated by a bi-phase cell model based on the kinematic approach. The comparison between simulated and measured erosion and deposition depths is satisfactory. The same parameters for computing erosion and deposition were used for the three occurred events.

Investigating the prevalence and factors associated with minor psychiatric disorders (MPDs) in Hospital housekeeping workers. A cross-sectional study carried out in 2013 with workers from the cleaning service of a public university hospital in Rio Grande do Sul, Brazil. Data were collected through a form containing sociodemographic, occupational, habits and health variables. The Self-Reporting Questionnaire-20 was used in order to evaluate MPDs. The study population consisted of 161 workers. The overall prevalence of suspected MPD was 29.3%. The chances of suspected MPDs were higher in workers with Effort-Reward Imbalance, those who did not have time or who occasionally had time for leisure activities, and those taking medications. The prevalence of MPDs was similar to that found in the literature for health workers. Therefore, we consider it important to include these workers in institutional programs for continuing health education.
