Background Due to the indiscriminate use of antimicrobial drugs, the emergence of human pathogenic microorganisms resistant to major classes of antibiotics has been increased and has caused many clinical problems in the treatment of infectious diseases. Thus, the aim of this study was to evaluate for the first time the in vitro antimicrobial activity and brine shrimp lethality of extracts and isolated compounds from Zeyheria tuberculosa (Vell.) Bur., a species used in Brazilian folk medicine for treatment of cancer and skin diseases. Methods Using the disc diffusion method, bioautography assay and brine shrimp toxicity test (Artemia salina Leach), we studied the antimicrobial activity and lethality of extracts and isolated compounds against three microorganisms strains, including Gram-positive (Staphylococcus aureus) and Gram-negative (Pseudomonas aeruginosa) bacteria and yeasts (Candida albicans). Results In this study, the extracts inhibited S. aureus (8.0 ± 0.0 to 14.0 ± 0.0 mm) and C. albicans (15.3 ± 0.68 to 25.6 ± 0.4 mm) growth. In the brine shrimp test, only two of them showed toxic effects (LC50 29.55 to 398.05 μg/mL) and some extracts were non-toxic or showed weak lethality (LC50 705.02 to > 1000 μg/mL). From these extracts, four flavones [5,6,7,8-tetramethoxyflavone (1), 5,6,7-trimethoxyflavone (2), 4'-hydroxy-5,6,7,8-tetramethoxyflavone (3), and 4'-hydroxy-5,6,7-trimethoxyflavone (4)] were isolated through bioassay-guided fractionation and identified based on the 1D and 2D NMR spectral data. By bioautography assays, compounds 1 [S. aureus (16.0 ± 0.0 mm) and C. albicans (20.0 ± 0.0 mm)] and 3 [S. aureus (10.3 ± 0.6 mm) and C. albicans (19.7 ± 0.6 mm)] inhibited both microorganisms while 2 inhibited only S. aureus (11.7 ± 0.6 mm). Compound 4 did not restrain the growth of any tested microorganism. Conclusion Our results showed that extracts and isolated flavones from Z. tuberculosa may be particularly useful against two pathogenic microorganisms, S. aureus and C. albicans. These results may justify the popular use this species since some fractions tested had antimicrobial activity and others showed significant toxic effects on brine shrimps. However, in order to evaluate possible clinical application in therapy of infectious diseases, further studies about the safety and toxicity of isolated compounds are needed. PMID:19450272

New sequences and external morphology of Trachycephalus typhonius are also analyzed to assess the status of Ecuadorian and Peruvian populations. The phylogeny shows unequivocally that Ecnomiohyla tuberculosa is nested within the genus Tepuihyla , tribe Lophiohylini. This position was unexpected because the remaining species of Ecnomiohyla belong to the tribe Hylini. To solve the paraphyly of the genus Ecnomiohyla , Ecnomiohyla tuberculosa is transferred to the genus Tepuihyla . Comparisons of DNA sequences, external morphology, and advertisement calls between populations of Ecnomiohyla tuberculosa from Ecuador and Peru indicate that the Peruvian population represents an undescribed species. The new species is described and a species account is provided for Ecnomiohyla tuberculosa . Trachycephalus typhonius is paraphyletic relative to Trachycephalus cunauaru , Trachycephalus hadroceps , and Trachycephalus resinifictrix . The phylogenetic position of populations from western Ecuador indicates that they represent a species separate from Trachycephalus typhonius sensu stricto . We resurrect the name Hyla quadrangulum ( Trachycephalus quadrangulum comb. n. ) for those populations. Amazonian populations of " Trachycephalus typhonius " from Ecuador and Peru are genetically and morphologically distinct from Trachycephalus typhonius sensu stricto and are conspecific with the holotype of Hyla macrotis . Therefore, we also resurrect Hyla macrotis , a decision that results in Trachycephalus macrotis comb. n.

The gross and histopathologic lesions of meningoencephalitis tuberculosa in a 4-year-old Holstein cow showing clinical signs compatible with bovine spongiform encephalopathy are described in this report. Grossly, numerous gray to yellow, firm and caseous nodules were seen on the ventral surfaces of the brain and in the lateral and fourth ventricles. Histopathologically, foci of caseation and dystrophic mineralization were surrounded by multinucleated giant cells, epitheloid macrophages, plasma cells, lymphocytes and fibrous proliferation. Ziehl-Neelsen stains of the lesions revealed masses of slender acid-fast bacilli in the necrotic centers of lesions and within surrounding giant cells.

We report the case of an 81-year-old man diagnosed with liver cirrhosis complicated by spontaneous bacterial peritonitis and septic shock. Mycobacterium tuberculosis complex was isolated from the ascites, sputum, and blood culture 1 month after the patient died. Clinicians should be aware of the unusual diagnosis of sepsis tuberculosa gravissima presenting with tuberculous peritonitis, which is easily misdiagnosed as spontaneous bacterial peritonitis and Gram-negative bacillus sepsis in patients with cirrhosis. Clinicians should cautiously evaluate the patient's sputum, gastric contents, urine, cerebrospinal fluid, and bone marrow for early diagnosis of disseminated tuberculosis in patients with a high degree of suspicion of this diagnosis. Three new compounds, tuberazines A–C (1–3), and eleven known compounds (4–14) were obtained from the ethanolic extract of Taiwanese zoanthid Palythoa tuberculosa. Compounds 1–4 are rare marine natural products with a pyrazine moiety, and compound 5 is a tricyclic tryptamine derivative isolated from nature for the first time. The structures of all isolated metabolites were determined by analyzing their IR, Mass, NMR, and UV spectrometric data. The absolute configuration of 1 was confirmed by comparing the trend of experimental electronic circular dichroism (ECD) with calculated ECD spectra. The anti-lymphangiogenic activities of new compounds were evaluated in human lymphatic endothelial cells (LECs). Of these, new compound 3 displayed the most potent anti-lymphangiogenesis property by suppressing cell growth and tube formation of LECs. PMID:29385063

The use and management of "angico" (Anadenanthera colubrina (Vell.) Brenan) by a rural community in northeastern Brazil was examined. By employing different techniques of data collection and population structure analysis, it was determined that this species had multiple uses within the local community (especially as timber and for other wood products), and that local management of this species is based on simple maintenance and harvesting of individuals in agroforest homegardens. The study of the population structure of this tree species indicated that management and conservation strategies must include the participation of the local community. PMID:16420708
Analysis of identified iron meteoroids: Possible relation with M-type Earth-crossing asteroids?

We have used two different techniques to analyze the U. S. Prairie Network (PN) fireballs in order to search for possible nickel-iron meteoroids. The first approach used is that of ReVelle and Rajan which is similar to the analysis carried out earlier by Wetherill and ReVelle in a series of papers relating first to the chondrites and later to fireballs of cometary origin. The second approach is a new technique developed by Ceplecha and co-workers that can simultaneously determine the presence and location of gross fragmentation events and also determine an effective ablation parameter during the fireball entry. Using this combined approach we have determined that seven fireballs among the 287 that were analyzed are likely to be iron in composition. Using the method of Ceplecha we have determined that none of these objects experienced any gross fragmentation events during their entry to the atmosphere and most of the meteoroids also exhibited rather large ablation coefficients during entry as well, a feature that is also characteristic of the ReVelle and Rajan approach. For all of these objects for which we currently have available data, we have determined that gross fragmentation events did not occur during the entry.

The aim of the present study was to perform an in vitro analysis of the antimicrobial and antiproliferative potential of an extract from Anadenanthera colubrina (Vell.) Brenan (angico) and chemically characterize the crude extract. Antimicrobial action was evaluated based on the minimum inhibitory concentration (MIC), minimum bactericidal/fungicidal concentration, and the inhibition of formation to oral biofilm. Cell morphology was determined through scanning electron microscopy (SEM). Six strains of tumor cells were used for the determination of antiproliferative potential. The extract demonstrated strong antifungal activity against Candida albicans ATCC 18804 (MIC = 0.031 mg/mL), with similar activity found regarding the ethyl acetate fraction. The extract and active fraction also demonstrated the capacity to inhibit the formation of Candida albicans to oral biofilm after 48 hours, with median values equal to or greater than the control group, but the difference did not achieve statistical significance (P > 0.05). SEM revealed alterations in the cell morphology of the yeast. Regarding antiproliferative activity, the extract demonstrated cytostatic potential in all strains tested. The present findings suggest strong antifungal potential for Anadenanthera colubrina (Vell.) Brenan as well as a tendency toward diminishing the growth of human tumor cells. PMID:25093029

Cabralea canjerana (Vell.) Mart. is a tree that occurs frequently in secondary forests of Maringá, Paraná, Brazil and presents a valuable wood. Its flowering time occurs from August to October and the anthesis occurs during the night. Its flowers are visited by Lepidoptera-Noctuidae. The flowers are unisexual and solitary or arranged in panicles. The perianth presents a papillose epidermis with striate cuticle and a parenchymatic mesophyll. Ten stamens constitute the androecium and are arranged in a staminal tube with anthers. The anthers present epidermis, endothecium, two median layers and secretory tapetum with binucleate cells. The semi-inferior ovary presents anatropous, bitegmic and crassinucleate ovules. The nectaries are located in the base of the ovary and staminal tube and they present papillose epidermis with stomata and secretory parenchyma with a conspicuous phloematic tissue.

Previous analyses of meteoroid compositional groupings have utilized the end height of fireballs as a diagnostic tool. From an observational perspective this definition is straight forward, but from a theoretical viewpoint there are problems with using this operational definition. In order to realistically assess the estimated geometric uncertainty of + 1 km in the observed end height, a theoretical definition of the end height of meteoritic fireballs is proposed using the results from the integral radiation efficiency model of ReVelle. Three photographed and recovered meteorites are used as a calibration for this proposed definition. This definition was used to evaluate the end height of all fireballs that were deduced by Wetherill and ReVelle as being meteoritic. In almost all cases the theoretical values are lower than the observed values, in some cases as much as 5 km lower. A preliminary summary of results are given.

An Analysis of the Atmospheric Trajectories of the Famous Meteorite Producing Fireballs

In the present study the modern analytical model of the atmospheric entry (Gritsevich, 2007) is applied to well-known real impacts. These are Pribram, Lost City, Innisfree, and Neuschwanstein meteorite falls. For each of the events preatmospheric mass is calculated using the data of actual observations, by selecting the parameters describing deceleration and ablation of fireballs along the luminous segment of the trajectory. Mathematical model is based on the best fitting of the observational data by the analytical solution of the equations of meteor physics. Other estimations of preatmospheric mass for Pribram, Lost City, Innisfree, and Neuschwanstein fireballs are also presented. The majority of them considerably differs from each other. The greater difference is appreciable between the estimations received by dynamic and photometric methods. In the paper (Ceplecha, ReVelle, 2005) the mixed approach is presented. However, the found terminal mass has been accepted by authors (Ceplecha, ReVelle, 2005) as the given condition. When this study was prepared, the terminal mass has been calculated analytically. And it is exceptionally important, that results have appeared quite close to their real values.

In order to produce explants of Mandevilla illustris (Vell) Woodson for the "Cerrado in vitro", the Germplasm Bank of UNAERP, we carried out a micropropagation protocol using MS or MS/3 medium supplemented with different concentrations of 6-benzyladeninepurine (BA), Zeatin or 2-isopentenyladenine for nodal segment growth, and alpha-naphthaleneacetic acid, indole-3-butyric acid (IBA) or 1,4 dithiothreitol for rooting. For nodal segments, all the cytokinins tested yielded similar results. However, 2.22 micro M BA is more economical to use. MS/3 medium supplemented with 0.49 micro M IBA was the most appropriate medium for rooting, resulting in 29% rooted explants. The crude aqueous extract from the subterranean system (SS) of M. illustris was assayed for its inhibitory action on the enzymatic activity of Crotalus durissus terrificus snake venom, isolated basic phospholipase A2 (CB) and crotoxin. It totally inhibited the phospholipase activity of crude Cdt venom and CB toxin and inhibited the phospholipase activity of crotoxin by 49%. The toxic action of both the crude venom and crotoxin was partially inhibited-there was a prolonged survival time and a 40.0% decrease in lethality.

The widespread overuse of antibiotics in aquaculture has led to the emergence of antibiotic-resistance shrimp pathogens, the negative impact on shrimp gut microbiota, and the presence of antimicrobial residues in aquaculture products, with negative consequences on human health. Alternatively, probiotics have positive effects on immunological responses and productive performance of aquatic animals. In this study, three probiotic bacteria, (Bacillus licheniformis MAt32, B. subtilis MAt43 and B. subtilis subsp. subtilis GAtB1), isolated from the Anadara tuberculosa were included in diets for juvenile shrimp, Litopenaeus vannamei, to evaluate their effects on growth, survival, disease prevalence, and immune-related gene expression. Shrimp naturally infected with WSSV and IHHNV were fed with the basal diet (control, T1) and diets supplemented with four levels of bacilli probiotic mix (1:1:1) at final concentration of (T2) 1 × 10 6 , (T3) 2 × 10 6 , (T4) 4 × 10 6 , and (T5) 6 × 10 6 CFU g -1 of feed. The specific growth rate of shrimp was significantly higher in T2 than in T1 (control) treatment, and the final growth as well as the survival were similar among treated groups. The prevalence of WSSV and IHHNV infected shrimp was reduced in T2 and T4 treatments, respectively, compared with control. The mRNA expression of proPO gene was higher in treatment T4 than control. The LvToll1 gene was significantly up-regulated in treatments T4 and T5 compared to control. The SOD gene was up-regulated in treatment T5 compared to control. In contrast, the mRNA expression of the Hsp70 gene was down-regulated in treatments T4 and T5 respect to control, and the TGase gene remained unaffected by the level of bacillus probiotic mix. As conclusion, the bacilli probiotic mix (Bacillus spp.) enhanced immune-related gene expression in WSSV and IHHNV naturally infected shrimp. This is the first report of probiotic potential of bacteria isolated from A. tuberculosa on the immune response and viral prevalence in shrimp Litopenaeus vannamei. Copyright © 2016 Elsevier Ltd. Two new secondary metabolites, the phenylpropanoid 3-(2',4',5'-trimethoxyphenyl)propanoic acid (1) and the sesquiterpene (+)-1beta,4beta,6alpha-trihydroxyeudesmane (2) were isolated from the heartwood of Cordia trichotoma Vell., along with the known sesquiterpenes (-)-1beta,4beta,7alpha-trihydroxyeudesmane (3) and (+)-1beta,4beta,11-trihydroxyoppositane (4). Their structures were elucidated by means of spectroscopic data interpretation, mainly 1D and 2D NMR and mass spectrometry.

The Tobari Lagoon also receives effluents from nearby shrimp farms and untreated municipal sewage. Surface sediment samples and six different species of filter feeders (Crassostrea corteziensis, Crassostrea gigas, Chione gnidia, Anadara tuberculosa, Chione fluctifraga, and Fistulobalanus dentivarians) were collected during the dry and the rainy seasons and analyzed to determine concentrations of cadmium (Cd), copper (Cu), mercury (Hg), lead (Pb), and zinc (Zn). Seasonal variations in metal concentrations in sediment were evident, especially for Cd, Cu, Hg, and Zn. The total and bioavailable concentrations of the five metals are not elevated in comparison to other areas around the world. The percentages of bioavailable respect to total concentrations of the metals varied from 0.6 % in Hg to 50.2 % for Cu. In the organisms, Hg showed the lowest concentrations (ranged from 0.22 to 0.65 μg/g) while Zn showed the highest (ranged from 36.6 to 1,702 μg/g). Linear correlations between the levels of Cu, Pb, and Zn in the soft tissues of C. fluctifraga and C. gnidia, and A. tuberculosa and C. gnidia were found. Seasonal and interspecies variations in the metal levels in filter feeders were found; F. dentivarians, C. corteziensis, and C. gigas exhibited the highest levels, could be used as biomonitors of metals contamination in this area.

Solanum cernuum Vell. (Solanaceae) is a Brazilian medicinal plant, traditionally known as "panaceia". Its folk name is probably due to its wide range of applications in traditional medicine including the treatment of ulcers. To evaluate the gastroprotective activities of the hydroethanolic extract (ESC) of S. cernuum and its major isolated compounds using in vivo gastric ulcer models. The ESC extract was obtained by maceration followed by percolation of the dried and powdered leaves of S. cernuum in ethanol:water (7:3). The major compounds in the extract were isolated by applying various preparative chromatographic techniques. The gastroprotective activity was evaluated in mice using different gastric ulcer-induced models. The anti-Helicobacter pylori activity was performed using the agar-well diffusion and broth microdilution methods. The ESC extract showed gastroprotective effects in the assay of acute gastric ulcer-induced by HCl/EtOH, nonsteroidal anti-inflammatory drug, and acetic acid-induced chronic ulcer protocols. The results also demonstrated that the gastroprotection induced by ESC extract is related to the activity of nitric oxide and endogenous sulfhydryls, which are important gastroprotective factors. The ESC extract and the alkaloid cernumidine did not show activity against H. pylori in the concentrations tested. The present study showed that the crude extract of S. cernuum possessed gastroprotective activity which corroborating the traditional use of this plant for the treatment of gastric ulcers. The isolated flavonoids, quercitrin and afzelin as well as the phenylpropanoid, isoferulic acid are suggested to be the compounds responsible for the gastroprotective activity of S. cernuum extract. Anadara tuberculosa is one of the most important bivalves along the Western Pacific coast because of its commercial value. Nevertheless, the variability in growth, long-life span, natural mortality and reproductive parameters of this mangrove cockle has not yet been described. The aim of this study was to analyze these life-history traits in three areas of the Southern coast of Ecuador. Empirical and length-based methods were used to estimate these biological parameters. Body size data were collected from the commercial fishery between 2004 and 2011 in landing ports near to the Archipelago of Jambeli [Puerto Bolivar (PB), Puerto Jeli (PJ) and Puerto Hualtaco (PH)]. The von Bertalanffy growth parameters for combined sex were estimated between 70.87 to 93.45mm for L(infinity) and 0.22 to 0.80/year for k. The growth indices (PHI') ranged from 3.17 to 3.85, while the overall growth performance (OGP) ranged from 5.03 to 5.82. The mean of long-life span (t(max)), size and age at maturity (L50% and t50%) were estimated in 7.71 +/- 2.53 years, 39.13 +/- 2.24mm and 1.46 +/- 0.56 years for PB; 9.51 +/- 2.85 years, 37.78 +/- 1.95mm and 1.37 +/- 0.41 years for PJ and 5.81 +/- 2.11 years, 39.73 +/- 3.31mm and 0.94 +/- 0.41 years for PH. Natural mortality (M) ranged from 0.46 to 1.28/year. We concluded that significant intra-specific variation was observed in a temporal scale in PHI' and OGP indices as well as L50% and M. Therefore, temporal changes in these life-history traits should be taken into account when assessing the status of the mangrove cockle fishery.

Global warming, and nutrient and sediment runoff from coastal development, both exert increasing pressures on coastal coral reefs. The objective of this study was to resolve the question of whether coastal eutrophication may protect corals from thermal stress by improving their nutritional status, or rather diminish their thermal tolerance through the synergy of dual stressors. A review of previous studies on the topic of combined trophic status and heat exposure on the thermal tolerance of corals reveals a broad range of outcomes, including synergistic, additive and antagonistic effects. We conducted a 90-day long experiment exposing corals to realistic levels of elevated nutrients and sediments, and heat stress. Colonies of two common scleractinian corals (Acropora millepora and Montipora tuberculosa) were kept in coastal seawater, or coastal seawater that was further organically and nutrient enriched (OE), and/or enriched with nitrate. Batches of OE were created daily, facilitating nutrient uptake, plankton succession and organic enrichment as observed in coastal waters. After 10 days of acclimation, 67% of the colonies had their temperature gradually increased from 27° to 31.2°C. After 3–7 weeks of heat stress, colonies of both species had significantly greater reductions in fluorescence yields and lower survival in OE than without addition of OE. Furthermore, photophysiological recovery was incomplete 31–38 days after ending the heat stress only in the OE treatments. Nitrate alone had no measurable effect on survival, bleaching and recovery in either species. Skeletal growth rates were reduced by 45% in heat-stressed A. millepora and by 24% in OE-exposed M. tuberculosa. We propose a conceptual trophic framework that resolves some of the apparently contradictory outcomes revealed by the review. Our study shows that management actions to reduce coastal eutrophication can improve the resistance and resilience of vulnerable coastal coral reefs to warming temperatures. PMID:23349876

A red tide event occurred in El Salvador from August 2001 to January 2002. National health authorities usually measured toxin levels in Ostrea iridescens, however other species were analyzed during this microalgae bloom: Anadara similis, Anadara tuberculosa and Modiolus sp. El Salvador authorities consider 400 mouse units/100 g the highest value that is safe for human health. During this period toxin levels in 0. iridescens and Modiolus sp. increased from values under 400 to 3977 and 15,468 mouse units/100 g, respectively. Persistent and higher levels were recorded in oyster and mussel banks on the west part of the country. The Ministry of Health and Social Assistance treated 41 slight to moderate intoxications associated to bivalve mollusks consumption.

The first host plant record for Anastrepha fumipennis Lima (Diptera: Tephritidae) in Geissospermum laeve (Vell.) Baill (Apocynaceae) and for A. nascimentoi Zucchi found in Cathedra bahiensis Sleumer (Olacaceae) was determined in a host plant survey of fruit flies undertaken at the “Reserva Natural da Companhia Vale do Rio Doce”. This reserve is located in an Atlantic Rain Forest remnant area, in Linhares county, state of Espírito Santo, Brazil. The phylogenetic relationships of Anastrepha species and their hosts are discussed. The occurrence of these fruit fly species in relation to the distribution range of their host plants is also discussed. PMID:20302458

Vernonia herbacea (Vell.) Rusby, a native species from the Brazilian Cerrado, accumulates about 80% of fructans in the rhizophores, the underground reserve organs. Besides their role as reserve, fructans have been recognized as protective compounds against drought. This physiological function attributed to fructans seems consistent with the wide occurrence of these carbohydrates in the cerrado, a biome that undergoes seasonal drought. The aim of this work was to analyze fructan composition and the activities of the enzymes involved in fructan synthesis, sucrose:sucrose 1-frutosyltransferase (1-SST) and fructan:fructan 1-frutosyltransferase (1-FFT), and depolymerization, fructan 1-exohydrolase (1-FEH) in plants submitted to water suppression. The plants were divided into 3 groups receiving 3 treatments: daily watering (control), water suppression for 23 days (WS) and re-watering after 15 days (RW). Samples were taken at the beginning of the experiment (Time 0) and after 3, 7, 11, 15, 17 and 23 days of water suppression. 1-SST and 1-FFT activities increased at the beginning of the water restriction period, coinciding with a decrease in 1-FEH activity, the onset of the reduction in soil water potential and in leaf water potential. Increases in 1-FEH and invertase activities led to a high yield of reducing sugars at the 23rd day after water suppression, and together with 1-FFT, 1-FEH also seemed to act in the redistribution of fructan molecules after re-watering. The increase in reducing sugars and in the fructo-oligo:fructo-polysaccharide ratio were associated to the maintenance of rhizophore turgor. Considering that WS plants showed changes in fructan metabolism that favored water retention and absorption after re-watering, the occurrence of osmotic adjustment mechanisms is suggested, reinforcing the hypothesis of fructans as protective agents against abiotic stresses, such as drought. Palytoxin (PTX), C129H223N3O54, a highly toxic substance isolated from zoanthids of Palythoa tuberculosa, inhibited (Na,K)-ATPase (ATP phosphohydrolase, EC 3.6.1.3) prepared from guinea pig heart and hog cerebral cortex in a dose-dependent manner at concentrations greater than 10(-8) M. In the presence of Na (100 mM) and K (20 mM), PTX showed potency nearly equal to that of ouabain. When the ATPase was activated by the various Na concentrations at a constant K concentration, both PTX and ouabain inhibited the ATPase activity noncompetitively. On the other hand, when K concentration was changed at a constant Na concentration, PTX caused a competitive inhibition in all ranges of K concentrations employed, whereas ouabain caused a competitive inhibition at low concentrations and a noncompetitive inhibition at high concentrations.

The present study reports the phylogenetic relationship of six zoanthid species belonging to three genera, Isaurus, Palythoa, and Zoanthus identified using systematic computational analysis of mtDNA gene sequences. All six species are first recorded from the coasts of Kathiawar Peninsula, India. Genus: Isaurus is represented by Isaurus tuberculatus, genus Zoanthus is represented by Zoanthus kuroshio and Zoanthus sansibaricus, while genus Palythoa is represented by Palythoa tuberculosa, P. sp. JVK-2006 and Palythoa heliodiscus. Results of the present study revealed that among the various species observed along the coastline, a minimum of 99% sequence divergence and a maximum of 96% sequence divergence were seen. An interspecific divergence of 1-4% and negligible intraspecific divergence was observed. These results not only highlighted the efficiency of the COI gene region in species identification but also demonstrated the genetic variability of zoanthids along the Saurashtra coastline of the west coast of India.

Abstract Two additions and four new species are described from Brazil for the large Geminata clade (Solanum: Solanaceae) bringing the total diversity in the group to 149 species, with 44 of these occurring in Brazil. New species are described from Brazil: Solanum amorimii S.Knapp & Giacomin, sp. nov. from Bahia and adjacent Minas Gerais states, Solanum filirhachis Giacomin & Stehmann, sp. nov. from Espirito Santo, Solanum psilophyllum Stehmann & Giacomin, sp. nov. from Minas Gerais and Solanum verticillatum S.Knapp & Stehmann, sp. nov. from São Paulo, Rio de Janeiro and Minas Gerais. Modern character-rich descriptions and lectotypifications are provided for Solanum apiahyense Witasek and Solanum lacteum Vell. All are illustrated, mapped and assessed for conservation status. We also provide a brief analysis of the diversity and endemism of the Geminata clade in Brazil and a key to all 44 Brazilian species. PMID:25878553

Pollen grain morphology of Bromeliaceae species collected in areas of the Atlantic Rainforest of southeastern Brazil was studied. The following species were analyzed: Aechmea bambusoides L.B.Sm. & Reitz, A. nudicaulis (L.) Griseb., A. ramosa Mart. ex Schult.f., Ananas bracteatus (Lindl.) Schult.f., Billbergia distachia (Vell.) Mez, B. euphemiae E. Morren, B. horrida Regel, B. zebrina (Herb.) Lindl., Portea petropolitana (Wawra) Mez, Pitcairnia flammea Lindl., Quesnelia indecora Mez, Tillandsia polystachia (L.) L., T. stricta Sol., T. gardneri Lindl., T. geminiflora Brongn. and Vriesea grandiflora Leme. Light and scanning electron microscopy were used and the species were grouped into three pollen types, organized according to aperture characteristics: Type I - pantoporate pollen grains observed in P. petropolitana, Type II - 2-porate pollen grains, observed in the genera Ananas, Aechmea and Quesnelia, and Type III - 1-colpate pollen grains, observed in the genera Billbergia, Pitcairnia, Tillandsia and Vriesea. Pollen data led to the construction of an identification key. The results showed that the species analyzed can be distinguished using mainly aperture features and exine ornamentation, and that these characteristics may assist in taxonomic studies of the family.

Successive applications of insecticides to control Plutella xylostella L. (Lepidoptera: Plutellidae) have resulted in the emergence of resistant populations of this insect. A novel control measure for this target insect could be the use of botanical insecticides derived from plant tissues. Hence, we experimentally tested aqueous extracts of Alibertia edulis (Rich.), Alibertia intermedia (Mart.), and Alibertia sessilis (Vell.) K. Schum. found in the Brazilian savannah in order to investigate their potential to disrupt the life cycle of P. xylostella. Aqueous extracts of the leaves of A. intermedia and A. sessilis negatively affected the development of P. xylostella in all stages of the life cycle, prolonging the larval stage and causing mortality in the larval or pupal stages. Treatments with A. intermedia and A. sessilis extracts caused the lowest fecundity and the number of hatched larvae. The harmful effects of these aqueous extracts on the life cycle of P. xylostella may be attributable to the flavonoids and other phenolic compounds present in A. intermedia and A. sessilis. These aqueous botanical extracts are low in toxicity when compared to non-aqueous pesticides, and may emerge as an effective approach for control of populations of P. xylostella. PMID:29165339

We investigated whether there were consistent differences in the physiological and anatomical traits and phenotypic variability of an invasive (Prosopis juliflora (Sw.) DC.) and native species (Anadenanthera colubrina (Vell.) Brenan) in response to seasonality in a tropical dry forest. The water potential, organic solutes, gas exchange, enzymes of the antioxidant system, products of oxidative stress and anatomical parameters were evaluated in both species in response to seasonality. An analysis of physiological responses indicated that the invasive P. juliflora exhibited higher response in net photosynthetic rate to that of the native species between seasons. Higher values of water potential of the invasive species than those of the native species in the dry season indicate a more efficient mechanism for water regulation in the invasive species. The invasive species exhibits a thicker cuticle and trichomes, which can reduce transpiration. In combination, the increased epidermal thickness and the decreased thickness of the parenchyma in the dry season may contribute to water saving. Our data suggest a higher variability in anatomical traits in the invasive species as a response to seasonality, whereas physiological traits did not present a clear pattern of response. © The Author 2017. The bark of Anadenanthera peregrina (L.) Speg and Anadenanthera colubrina (Vell.) Brenan were characterized in relation to anatomical and chemical features. The barks were similar and included a thin conducting phloem, a largely dilated and sclerified non-conducting phloem, and a rhyridome with periderms with thin phellem interspersed by cortical tissues. Only small differences between species were observed that cannot be used alone for taxonomic purposes. The summative chemical composition of A. peregrina and A. colubrina was respectively: 8.2% and 7.7% ash; 28.8% and 29.3% extractives; 2.4% and 2.6% suberin; and 18.9% lignin. The monosaccharide composition showed the predominance of glucose (on average 82% of total neutral sugars) and of xylose (9%). The ethanol-water extracts of A. peregrina and A. colubrina barks included a high content of phenolics, respectively: total phenolics 583 and 682 mg GAE/g extract; 148 and 445 mg CE/g extract; tannins 587 and 98 mg CE/g extract. The antioxidant activity was 238 and 269 mg Trolox/g extract. The barks of the Anadenanthera species are a potential source of polar extractives that will represent an important valorization and therefore contribute to improve the overall economic potential and sustainability of A. peregrina and A. colubrina PMID:29281656

In the Brazilian Cerrado (neotropical savanna), the development of bud-bearing underground systems as adaptive structures to fire and dry periods can comprise an important source of buds for this ecosystem, as already demonstrated in the Brazilian Campos grasslands and North American prairies. Asteraceae species from both woody and herbaceous strata have subterranean organs that accumulate carbohydrates, reinforcing the adaptive strategy of these plants to different environmental conditions. This study aims to analyse the morpho-anatomy of underground systems of six species of Asteraceae (Mikania cordifolia L.f. Willd., Mikania sessilifolia DC, Trixis nobilis (Vell.) Katinas, Pterocaulon alopecuroides (Lam.) DC., Vernonia elegans Gardner and Vernonia megapotamica Spreng.), to describe these structures and to verify the occurrence and origin of shoot buds, and to analyse the presence of reserve substances. Individuals sampled in Cerrado areas in São Paulo State showed thick underground bud-bearing organs, with adventitious or lateral roots and presence of fructans. Xylopodium was found in all studied species, except for Trixis nobilis, which had stem tuber. The presence of fructans as reserve, and the capacity of structures in the formation of buds indicate the potential of herbaceous species of Asteraceae in forming a viable bud bank for vegetation regeneration in the Brazilian Cerrado.

Laser Raman and Fourier transform infrared spectroscopies were applied in the investigation of conformational features of a chymotrypsin inhibitor (SPC), inactive on trypsin, isolated from Schizolobium parahyba, a Leguminosae of the Cesalpinoidae family, found in tropical and subtropical regions. As a serine protease inhibitor, its importance is related to the control of proteolytic activity, which in turn is involved in a wide range of critically important biotechnological issues, such as blood coagulation, tumour cell growth, and plant natural defences against predators. SPC is a 20 kDa molecular mass monomeric protein, with two disulfide bonds. Its complete aminoacid primary sequence has not yet been determined. We analysed protein backbone conformation for the lyophylized solid and for an evaporated film, through Raman scattering and FTIR, respectively. The presence of significant amounts of disordered structures and of non-negligible contributions from α-helical and β-sheet structures were reckoned in both cases. The geometries of the disulfide bonds were defined: a gauche-gauche-gauche geometry was verified for one of the two bridges and a transient gauche-gauche-trans/trans-gauche-trans geometry has been indicated for the second one.Two out of the three tyrosine residues were shown to be in external location in the solid protein, as well as the only tryptophan residue.

The presented paper considered the pollen morphology of thirteen species belonging to seven genera of the Fabaceae family occurring in the Pau-de-Fruta Special Protection Area (SPA), Diamantina, state of Minas Gerais, Brazil. The pollen grains of six species of Chamaecrista [C. cathartica (Mart.) H.S. Irwin & Barneby, C. debilis Vogel, C. flexuosa (L.) Greene, C. hedysaroides (Vogel) H.S. Irwin & Barneby, C. glandulosa (L.) Greene, and C. papillata H.S. Irwin & Barneby] have a similar morphology, characterized by three long colporated apertures with a central constriction. The species share specific morphological features regarding pollen size, endoaperture type (circular, lalongate or lolongate) and SEM ornamentation patterns of the exine (rugulate with perforations or perforate). Andira fraxinifolia Benth., Dalbergia miscolobium Benth, Galactia martii DC, Periandra mediterranea (Vell.) Taub., Senna rugosa (G.Don) H.S. Irwin & Barneby and Zornia diphylla (L.) Pers showed different pollen types in small to large size; oblate spheroidal to prolate form; colpus or colporus apertures; circular, lalongate or lolongate endoapertures and distinctive SEM ornamentation patterns of the exine (perforate, microreticulate, reticulate or rugulate with perforations). Only Stryphnodendron adstringens (Mart.) Coville presents polyads. The pollen morphology variation of these species allowed the Fabaceae family to be characterized as eurypalynous in the SPA Pau-de-Fruta.

Food security is the condition where the food fulfilment is managed well for the country till the individual. Indonesia is one of the country which has the commitment to create the food security becomes main priority. However, the food necessity becomes common thing means that it doesn’t care about nutrient standard and the health condition of family member, so in the fulfilment of food necessity also has to consider the disease suffered by the family member, one of them is pulmonary tuberculosa. From that reasons, this research is conducted to know the factors which influence on household food security status which suffered from pulmonary tuberculosis in the coastal area of Surabaya by using binary logistic regression method. The analysis result by using binary logistic regression shows that the variables wife latest education, house density and spacious house ventilation significantly affect on household food security status which suffered from pulmonary tuberculosis in the coastal area of Surabaya, where the wife education level is University/equivalent, the house density is eligible or 8 m2/person and spacious house ventilation 10% of the floor area has the opportunity to become food secure households amounted to 0.911089. While the chance of becoming food insecure households amounted to 0.088911. The model household food security status which suffered from pulmonary tuberculosis in the coastal area of Surabaya has been conformable, and the overall percentages of those classifications are at 71.8%.

Fractionation of the MeOH extract from leaves of Piper cernuum Vell. (Piperaceae) afforded six phenylpropanoid derivatives: 3',4'-dimethoxydihydrocinnamic acid (1), piplaroxide (2), methyl 4'-hydroxy-3',5'-dimethoxy cinnamate (3), 3',4',5'-trimethoxydihydrocinnamic acid (3), dihydropiplartine (5), and piplartine (6). The structures of isolated metabolites were characterized by NMR and MS spectral data analysis. The chemical composition of essential oil from the leaves was determined using GC/LREIMS followed by the determination of Kovats indexes. This procedure allowed the identification of nineteen terpenoids, with β-elemene (7), bicyclogermacrene (8), germacrene D (9), and (E)-caryophyllene (10) as the main compounds. Compounds 1 and 3-6 displayed no in vitro cytotoxicity against cancer cell lineages B16F10-Nex2, U87, HeLa, HL-60, HCT, and A2058 while 2 showed moderate activity against B16F10-Nex2 and HL-60 lines. Otherwise, compounds 7-10 displayed high cytotoxic activity. Evaluation against non-tumorigenic HFF cells indicated a reduced selectivity of compounds 7-10 to tumoral cells. No antileishmanial activity on macrophages infected with L. (L.) amnazonensis was found for the crude MeOH extract and compounds 1-6. The crude essential oil and compounds 7-10 reduced parasitism and eliminated the majority of infected and non-infected cells at 50 μg/mL.

International trade in Brazilian rosewood, Dalbergia nigra (Vell.) Allemão ex Benth., is regulated by the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). One problem in enforcing these regulations is the difficulty in distinguishing the wood of D. nigra from that of a closely-related but unregulated species, Dalbergia spruceana Benth. Using LC-MS to analyse methanol extracts of xylaria specimens, we identified a chemical marker for D. nigra heartwood, and determined its structure as the neoflavonoid 6-hydroxy-7-methoxy-4-(4-methoxyphenyl)-2H-1-benzopyran-2-one (4'-O-methylmelanettin; dalnigrin), using spectroscopic techniques. Dalnigrin was present in all nine available heartwood specimens of D. nigra, but it was not detected in extracts of 59 other heartwood samples representing 15 species of Dalbergia, including D. spruceana. Five other phenolic compounds were also isolated from D. nigra heartwood and similarly identified as the neoflavonoids 3'-hydroxymelanettin, melanettin, melannein and dalbergin, and the isoflavone caviunin. In extracts of D. spruceana heartwood, pseudobaptigenin was identified by LC-MS to be a major phenolic component that was not detected in wood extracts of D. nigra. We conclude that chemical analysis, in combination with anatomical investigation, can provide persuasive evidence to support the positive identification of untreated heartwood of D. nigra. 2010 Elsevier Ltd. In the present investigation, four macrophytes, namely Typha latifolia (L.), Lemna minor (L.), Eichhornia crassipes (Mart.) Solms-Laubach, and Myriophyllum aquaticum (Vell.) Verdc, were evaluated for their heavy metal (Cu, Pb, Hg, and Zn) hyperaccumulation potential under laboratory conditions. Tolerance analyses were performed for 7 days of exposure at five different treatments of the metals mixture (Cu +2 , Hg +2 , Pb +2 , and Zn +2 ). The production of chlorophyll and carotenoids was determined at the end of each treatment. L. minor revealed to be sensitive, because it did not survive in all the tested concentrations after 72 hours of exposure. E. crassipes and M. aquaticum displayed the highest tolerance to the metals mixture. For the most tolerant species of aquatic macrophytes, The removal kinetics of E. crassipes and M. aquaticum was carried out, using the following mixture of metals: Cu (0.5 mg/L) and Hg, Pb, and Zn 0.25 mg/L. The obtained results revealed that E. crassipes can remove 99.80% of Cu, 97.88% of Pb, 99.53% of Hg, and 94.37% of Zn. M. aquaticum withdraws 95.2% of Cu, 94.28% of Pb, 99.19% of Hg, and 91.91% of Zn. The obtained results suggest that these two species of macrophytes could be used for the phytoremediation of this mixture of heavy metals from the polluted water bodies.
Gross and microscopic morphology of lesions in Cnidaria from Palmyra Atoll, Central Pacific

We conducted gross and microscopic characterizations of lesions in Cnidaria from Palmyra Atoll, Central Pacific. We found growth anomalies (GA) to be the most commonly encountered lesion. Cases of discoloration and tissue loss were rare. GAs had a focal or multi-focal distribution and were predominantly nodular, exophytic, and umbonate. In scleractinians, the majority of GAs manifested as hyperplasia of the basal body wall (52% of cases), with an associated absence or reduction of polyp structure (mesenteries and filaments, actinopharynx and tentacles), and depletion of zooxanthellae in the gastrodermis of the upper body wall. In the soft corals Sinularia sp. and Lobophytum sp., GAs exclusively manifested as prominent hyperplasia of the coenenchyme with an increased density of solenia. In contrast to scleractinians, soft coral GAs displayed an inflammatory and necrotizing component with marked edema of the mesoglea, accompanied by infiltrates of variably-sized granular amoebocytes. Fungi, algae, sponges, and Crustacea were present in some scleractinian GAs, but absent in soft coral GAs. Fragmentation of tissues was a common finding in Acropora acuminata and Montipora cf. dilatata colonies with tissue loss, although no obvious causative agents were seen. Discoloration in the zoanthid, Palythoa tuberculosa, was found to be the result of necrosis, while in Lobophytum sp. discoloration was the result of zooxanthellar depletion (bleaching). Soft corals with discoloration or tissue loss showed a marked inflammatory response, however no obvious causative organisms were seen. Lesions that appeared similar at the gross level were revealed to be distinct by microscopy, emphasizing the importance of histopathology.

Variations in the inulin contents have been detected in rhizophores of Vernonia herbacea during the phenological cycle. These variations indicate the occurrence of active inulin synthesis and depolymerization throughout the cycle and a role for this carbohydrate as a reserve compound. 1-Fructan exohydrolase (1-FEH) is the enzyme responsible for inulin depolymerization, and its activity has been detected in rhizophores of sprouting plants. Defoliation and low temperature are enhancer conditions of this 1-FEH activity. The aim of the present work was the cloning of this enzyme. Rhizophores were collected from plants induced to sprout, followed by storage at 5 degrees C. A full length 1-FEH cDNA sequence was obtained by PCR and inverse PCR techniques, and expressed in Pichia pastoris. Cold storage enhances FEH gene expression. Vh1-FEH was shown to be a functional 1-FEH, hydrolyzing predominantly beta-2,1 linkages, sharing high identity with chicory FEH sequences, and its activity was inhibited by 81% in the presence of 10 mM sucrose. In V. herbacea, low temperature and sucrose play a role in the control of fructan degradation. This is the first study concerning the cloning and functional analysis of a 1-FEH cDNA of a native species from the Brazilian Cerrado. Results will contribute to understanding the role of fructans in the establishment of a very successful fructan flora of the Brazilian Cerrado, subjected to water limitation and low temperature during winter.

Vernonia herbacea (Vell.) Rusby (Asteraceae) is a perennial herb native to the cerrado vegetation of tropical areas in Brazil, which accumulates inulin in the underground reserve organs. The aim of this paper was to determine whether the inulin extracted from V. herbacea could replace commercial inulin for the measurement of glomerular filtration rate (GFR). Underground organs of vegetative plants were collected from a preserved area of the Brazilian cerrado. The inulin fraction utilized was obtained by ethanol precipitation after discarding the high molecular mass fructans in the freeze-thawing precipitate. GFR was determined in male Wistar rats anesthetized with inactin (100 mg/kg), which received intravenously commercial inulin obtained from Dahlia sp (Sigma) or Vernonia herbacea inulin (30 mg/100 g) as a priming dose and 0.05 mg min-1 100 g-1 as a sustaining dose in isotonic saline at the rate of 0/055 ml/min. Clearance was determined during 3 periods, with urine collected from the bladder and blood from the carotid artery. There was no significant difference in the GFR measured by clearance in inulin from both sources even when the plasma concentration of inulin from V. herbacea was doubled. The mean arterial pressure did not vary after the application of both inulins, indicating that they do not produce systemic side effects. The filtered load and the excreted amount of inulin from V. herbacea were equal, showing that the substance is not influenced by tubular function. These results demonstrate that the inulin from V. herbacea can substitute for imported inulin for the determination of GFR and in experiments of kidney microperfusion as a marker of tubular water reabsorption.
[Tuberculin reactivity among ninth-grade schoolchildren in the city of Havana, Cuba].

To determine the proportion of 14-year-old schoolchildren in the city of Havana, Cuba, with a positive tuberculin skin test, as an indicator of the prevalence of tuberculosis infection among them. Using single-stage cluster sampling, 1 936 Mantoux (tuberculin) tests were carried out with ninth-grade students (cohort born in 1985) during the 1999- 2000 school year in 20 basic high schools randomly selected in Havana. The tests were performed according to the standard technique recommended by the World Health Organization, and they were read after 72 hours. The percentage of skin tests that were positive and the average diameter of the indurations were calculated for the cohort overall and for the two genders. The means and the percentages were compared using the chi-square test, with 95% confidence intervals. The computer software used was Epi Info version 6.0. Of the tests read, 96% of them were negative (0-4 mm), 2.5% were doubtful (5-9 mm), and 1.5% were positive (>/=10 mm). The percentage of reactivity was 0.1% when a cutoff value of 15 mm was used. The mean diameter of the indurations was 0.41 mm. No statistically significant difference was found between the genders. In this study the proportion of schoolchildren with tuberculin reactivity, using an induration-diameter cutoff point of 10 mm, was very low (1.5%), and it was much lower (0.1%) when a cutoff point of 15 mm was used. The skin reactions with an induration diameter of >/=10 mm could be the expression of a natural infection if one takes into account the low frequency of bacillary tuberculosis in Cuba and that there is an inverse relationship between the time elapsed from the BCG vaccination and the intensity of the response to tuberculin. Therefore, that would mean that in this case (l)the point (.)prevalence of tuberculosis infection in this group of schoolchildren wou d be 1.5% ResumenObjetivos. Determinar la proporción de personas que reaccionan a la tuberculina como indicador de la prevalencia de infección tuberculosa en escolares de 14 años de Ciudad de La Habana, Cuba. Métodos. Mediante un muestreo monoetápico por conglomerados se aplicaron 1 936 pruebas de Mantoux (tuberculina) a estudiantes de noveno grado del curso escolar 1999-2000 (cohorte de los nacidos en 1985) de 20 escuelas secundarias básicas seleccionadas aleatoriamente en Ciudad de La Habana, Cuba. Las pruebas se realizaron según la técnica estándar recomendada por la Organización Mundial de la Salud y se evaluaron a las 72 horas. Se calcularon los porcentajes de reactividad y el diámetro medio de las induraciones cutáneas en la cohorte y según el sexo. Las comparaciones de medias y porcentajes se realizaron mediante la prueba de ji al cuadrado con 95% de confiabilidad. Se usó el programa Epi Info v. 6.0. Resultados. Noventa y seis por ciento de las pruebas leídas fueron negativas (0-4 mm), 2,5% fueron dudosas (5-9 mm) y 1,5% resultaron positivas ( >/= 10 mm). El porcentaje de reactividad disminuyó a 0,1% cuando se utilizó un valor de corte de 15 mm. El diámetro medio de las induraciones fue de 0,41 mm y no se detectó ninguna diferencia estadísticamente significativa en función del sexo. Conclusiones. La proporción de personas que reaccionaron a la tuberculina, usando como punto de corte un diámetro de induración cutánea de 10 mm, fue muy baja (1,5%) en este estudio y mucho más baja (0,1%) cuando se utilizó 15 mm como punto de corte. Si se toma en cuenta la baja frecuencia de tuberculosis bacilífera en Cuba y que hay una relación inversa entre el tiempo transcurrido desde la aplicación de la vacuna a base del bacilo de Calmette-Guérin (BCG) y la intensidad de la respuesta a la tuberculina, las reacciones cutáneas con un diámetro de induración >/= 10 mm podrían ser la expresión de una infección natural, por lo que, en ese caso, la prevalencia puntual de infección tuberculosa en este grupo de escolares sería de 1,5%.

Background Understanding how people of diverse cultural backgrounds have traditionally used plants and animals as medicinal substances during displacements is one of the most important objectives of ethnopharmacological studies. An ethnopharmacological survey conducted among migrants living in the Southeast Atlantic Forest remnants (Diadema, São Paulo, Brazil) is presented herein. Methods Ethnographical methods were used to select and interview the migrants, and botanical and zoological techniques were employed to collect the indicated resources. Results We interviewed five migrants who described knowledge on 12 animals and 85 plants. Only 78 plants were present in Diadema, they belong to 37 taxonomic families; 68 were used exclusively for medicinal purposes, whereas 10 were reported to be toxic and/or presented some restriction of use. These taxa were grouped into 12 therapeutic categories (e.g., gastrointestinal disturbances, inflammatory processes or respiratory problems) based on the 41 individual complaints cited by the migrants. While the twelve animal species were used by the migrants to treat nine complaints; these were divided into six categories, the largest of which related to respiratory problems. None of the animal species and only 57 of the 78 plant species analysed in the present study were previously reported in the pharmacological literature; the popular knowledge concurred with academic findings for 30 of the plants. The seven plants [Impatiens hawkeri W. Bull., Artemisia canphorata Vill., Equisetum arvensis L., Senna pendula (Humb. & Bonpl. ex Willd.) H.S. Irwin & Barneby, Zea mays L., Fevillea passiflora Vell. and Croton fuscescens Spreng)] and the two animals (Atta sexdens and Periplaneta americana) that showed maintenance of use among migrants during their displacement in Brazilian territory, have not been studied by pharmacologists yet. Conclusions Thus, they should be highlighted and focused in further pharmacology and phytochemical studies, since the persistence of their uses can be indicative of bioactive potentials. Palytoxin (PLTX) is a complex marine toxin produced by zoanthids (i.e. Palythoa), dinoflagellates (Ostreopsis) and cyanobacteria (Trichodesmium). PLTX outbreaks are usually associated with Indo-Pacific waters, however their recent repeated occurrence in Mediterranean-European Atlantic coasts demonstrate their current worldwide distribution. Human sickness and fatalities have been associated with toxic algal blooms and ingestion of seafood contaminated with PLTX-like molecules. These toxins represent a serious threat to human health. There is an immediate need to develop easy-to-use, rapid detection methods due to the lack of validated protocols for their detection and quantification. We have developed an immuno-detection method for PLTX-like molecules based on the use of microspheres coupled to flow-cytometry detection (Luminex 200™). The assay consisted of the competition between free PLTX-like compounds in solution and PLTX immobilized on the surface of microspheres for binding to a specific monoclonal anti-PLTX antibody. This method displays an IC50 of 1.83 ± 0.21 nM and a dynamic range of 0.47-6.54 nM for PLTX. An easy-to-perform extraction protocol, based on a mixture of methanol and acetate buffer, was applied to spiked mussel samples providing a recovery rate of 104 ± 8% and a range of detection from 374 ± 81 to 4430 ± 150 μg kg(-1) when assayed with this method. Extracts of Ostreopsis cf. siamensis and Palythoa tuberculosa were tested and yielded positive results for PLTX-like molecules. However, the data obtained for the coral sample suggested that this antibody did not detect 42-OH-PLTX efficiently. The same samples were further analyzed using a neuroblastoma cytotoxicity assay and UPLC-IT-TOF spectrometry, which also pointed to the presence of PLTX-like compounds. Therefore, this single detection method for PLTX provides a semi-quantitative tool useful for the screening of PLTX-like molecules in different matrixes. Copyright © 2015 Elsevier B.V. Fast Prediction of Blast Damage from Airbursts: An Empirical Monte Carlo approach

The February 15, 2013 Chelyabinsk airburst was the first modern bolide whose associated shockwave caused blast damage at the ground (Popova et al., 2013). Near-Earth Object (NEO) impacts in the Chelyabinsk-size range (~20 m) are expected to occur every few decades (Boslough et al., 2015) and therefore we expect ground damage from meteoric airbursts to be the next planetary defense threat to be confronted. With pre-impact detections of small NEOs certain to become more common, decision makers will be faced with estimating blast damage from impactors with uncertain physical properties on short timescales.High fidelity numerical bolide entry models have been developed in recent years (eg. Boslough and Crawford, 2008; Shuvalov et al., 2013), but the wide range in a priori data about strength, fragmentation behavior, and other physical properties for a specific impactor make predictions of bolide behavior difficult. The long computational running times for hydrocode models make the exploration of a wide parameter space challenging in the days to hours before an actual impact.Our approach to this problem is to use an analytical bolide entry model, the triggered-progressive fragmentation model (TPFM) developed by ReVelle (2005) within a Monte Carlo formalism. In particular, we couple this model with empirical constraints on the statistical spread in strength for meter-scale impactors from Brown et al (2015) based on the observed height at maximum bolide brightness. We also use the correlation of peak bolide brightness with total energy as given by Brown (2016) as a proxy for fragmentation behaviour. Using these constraints, we are able to quickly generate a large set of realizations of probable bolide energy deposition curves and produce simple estimates of expected blast damage using existing analytical relations.We validate this code with the known parameters of the Chelyabinsk airburst and explore how changes to the entry conditions of the observed bolide may have modified the blast damage at the ground. We will also present how this approach could be used in an actual short-warning impact scenario.

A color-tunable luminescent material was prepared based on the composition of functionalized graphitic carbon nitride (g-C{sub 3}N{sub 4}) and europium (III). The functionalized g-C{sub 3}N{sub 4} layers not only behave as multifunctional supports including ligand coordinated with europium (III) and a support structure for the formation of the luminescent material, but exhibit excitation wavelength-dependent luminescence, thus the energy transfer between the functionalized g-C{sub 3}N{sub 4} and europium (III) can match very well by controlling the emission wavelength of functionalized g-C{sub 3}N{sub 4}. The as-prepared materials was comprehensively characterized via X-ray photoelectron spectroscopy, Fourier Transform Infrared spectroscopy, X-ray scattering techniques, Ultravioletmore » and Visible spectrophotometer, fluorescence spectrophotometer, thermogravimetric analysis, etc. The luminescent material exhibits multi-color emissions which are consistent with the characteristic emissions of europium (III) and functionalized g-C{sub 3}N{sub 4}, and the photoluminescence quality and density of the europium (III) can be greatly enhanced. Cordia ecalyculata Vell. and Echinodorus grandiflorus (Cham. & Schltdl.) Micheli are extensively used in Brazil as therapeutic preparations for indigenous groups and the general population. These plants have been used in the folk medicine as: tonic, diuretic, anti-inflammatory, appetite suppressants, for the treatment of snake bites, and weight loss. In this study, it was verified the possible cytotoxic and genotoxic effects of the crude extracts of. Cordia ecalyculata and Echinodorus grandiflorus, as well as their effectiveness in treating obesity. The Micronucleus Test was used for the evaluation of possible clastogenic and aneugenic effects, and the Comet Assay was used for the evaluation of single-strand and double-strand DNA breaks. The cytotoxic effects of the crude extracts were verified by PCE/NCE ratio. Swiss mice (Mus musculus) were used as the experimental model. It was observed a significant (P<0.05) increase, dose-independent, in the average frequency of micronucleated erythrocytes in peripheral blood in mice treated with either the Cordia ecalyculata or Echinodorus grandiflorus extracts, in comparison with the negative control. There were no significant differences (P>0.05) in the frequency of micronucleated polychromatic erythrocytes for both extract treatment. We observed that treatment with the Cordia ecalyculata extract at concentrations of 1000 and 2000 mg/kg bw resulted in a PCE/NCE ratio that was larger (P<0.05) than the negative control. After 15 days of daily treatment, a dose of 2000 mg/kg bw of either phytotherapeutic did not reduce body mass gain or the amount of food consumed by Swiss mice when compared with the negative control (P>0.05). The results of this study allowed us to infer that the crude extracts of Cordia ecalyculata and Echinodorus grandiflorus do not display cytotoxic or genotoxic activities. However, they do possess weak clastogenic activity (without significance) on peripheral blood cells. Contrary to commonly held beliefs it was also found in this study that the extracts are not effective for obesity treatments. Copyright 2009 Elsevier Ireland Ltd. All rights reserved.

Lithraea molleoides (Vell.) Engl. (Anacardiaceae) is a medicinal plant traditionally used in South America to treat various ailments, including diseases of the digestive system. To evaluate the in vivo antiulcer and antimicrobial activities against Helicobacter pylori of L. molleoides and its isolated compounds. Methanolic extract 250 and 500 mg/kg, (LmE 250 and LmE 500, respectively) and infusions, 10 g and 20 g en 100mL (LmI 10 and LmI 20, respectively) of L. molleoides was evaluated for antiulcer activity against 0.6N HCl, 0.2N NaOH, 200mg/kg acetilsalicilic acid and absolute ethanol-induced gastric ulcers in rats. The degree of erosion in the glandular part of the stomach was assessed from a scoring system. Acute toxicity in mice was also evaluated. The antiulcer effect of the isolated compounds (catechol, mannitol, rutin, gallic acid, ferulic acid and caffeic acid, 100mg/kg) was evaluated against absolute ethanol-induced gastric ulcers in rats. The anti-Helicobacter pylori activity of L. molleoides and isolated compounds was performed using broth dilution methods. The LmE 250, LmE 500, LmI 10 and LmI 20 produced significant inhibition on the ulcer index in 0.6N HCl, 0.2N NaOH, 200mg/kg acetilsalicilic acid and absolute ethanol- induced gastric ulcers in rats. The isolated compounds, catechol, mannitol, rutin, ferulic acid and caffeic acid were active in absolute ethanol- induced gastric ulcers in rats. L. molleoides and different compounds showed antimicrobial activity in all strains tested. The lowest MIC value (0. 5 μg/mL) was obtained with catechol in six of eleven strains assayed. No signs of toxicity were observed with doses up to 2g/kg in an acute toxicity assay. These findings indicate that L. molleoides displays potential antiulcerogenic and antimicrobial activities and the identification of active principles could support the use of this plant for the treatment of digestive affections. Different fragmentation models are applied to a number of events, including the entry of TC3 2008 asteroid in order to reproduce existing observational data. Keywords meteoroid entry fragmentation modeling 1 Introduction Fragmentation is a very important phenomenon which occurs during the meteoroid entry into the atmosphere and adds more drastic effects than mere deceleration and ablation. Modeling of bolide fragmentation (100 106 kg in mass) may be divided into several approaches. Detail fitting of observational data (deceleration and/or light curves) allows the determination of some meteoroid parameters (ablation and shape-density coefficients, fragmentation points, amount of mass loss) (Ceplecha et al. 1993; Ceplecha and ReVelle 2005). Observational data with high accuracy are needed for the gross-fragmentation model (Ceplecha et al. 1993), which is used for the analysis of European and Desert bolide networks data. Hydrodynamical models, which describe the entry of the meteoroid including evolution of its material, are applied mainly for large bodies (>106 kg) (Boslough et al. 1994; Svetsov et al. 1995; Shuvalov and Artemieva 2002, and others). Numerous papers were devoted to the application of standard equations for large meteoroid entry in the attempts to reproduce dynamics and/or radiation for different bolides and to predict meteorite falls. These modeling efforts are often supplemented by different fragmentation models (Baldwin and Sheaffer, 1971; Borovi.ka et al. 1998; Artemieva and Shuvalov, 2001; Bland and Artemieva, 2006, and others). The fragmentation may occur in different ways. For example, few large fragments are formed. These pieces initially interact through their shock waves and then continue their flight independently. The progressive fragmentation model suggests that meteoroids are disrupted into fragments, which continue their flight as independent bodies and may be disrupted further. Similar models were suggested in numerous papers, beginning with Levin (1956) and initial interaction of fragments started to be taken into account after the paper by Passey and Melosh (1980). The progressive fragmentation model with lateral spreading of formed fragments is widely used (Artemieva and Shuvalov, 1996; Nemtchinov and Popova, 1997; Borovi.ka et al. 1998; Bland and Artemieva, 2006).
The Orbital Distribution of Earth-crossing Asteroids and Meteoroids

The relationship between meteorites and Earth-crossing asteroids and between individual meteorites and meteor showers has been the subject of debate for some time. Recently, links have been claimed between certain meteorites and meteoroid complexes [e.g., 1] and it has been suggested that some meteorites are members of orbital "streams" [2]. It is difficult to evaluate these ideas because of the lack of appropriate measureable properties in the meteorites themselves. Cosmic ray exposure ages provide one approach but most cosmogenic nuclides have large halflives and hence generally reflect the long term radiation exposure of the body rather than the short term orbital evolution leading up to Earth impact. Here we use natural thermoluminescence (TL) data to determine the "average" perihelion of ordinary chondrites among the modern falls over periods of time of less than 10^3-10^5 years prior to Earth impact. The level of natural TL of a meteorite (at a given glow curve temperature) is a function of buildup through radiation dose (which, in turn, is a function of depth or "shielding" and external cosmic ray flux) and decay through thermal draining [3]. The shallow TL vs. depth profile observed in lunar cores [4] can, after correction for irradiation geometry, be used to to calculate TL profiles in meteoroid-sized bodies. Our new calculations indicate a range of natural TL of only about 15% in large meteoroid-sized bodies and an even smaller range in smaller bodies. The "half-life" of TL is far greater than the solar/cosmic ray flux cycle and hence variations in the external flux over time are expected to have only very minor effects. It is thus possible to calculate an "irradiation temperature" for a meteorite using its natural TL level, which can be shown through decay calculations to largely reflect the perihelion of the meteoroid body. The time period over which this irradiation temperature is averaged is a function of the temperature (perihelion); the period is <10^3 years at 0.7 AU, <10^4 years at 0.9 AU, <10^5 years at 1.0 AU and <5 x 10^6 years at >1.2 AU. Conversion of irradiation temperatures to perihelion requires that a value for meteoroid albedo be assumed; in the present analysis we assume such bodies have an albedo of ~0.2. For a collection of H, L, and LL modern falls (Fig. 1) we find a perihelion distribution that is fairly similar to those suggested by other observations. Perihelia for modern falls tend to be concentrated between 0.9 and 1.0 AU. However, our distribution is most similar to that found for ordinary chondrite-like fireballs by Wetherill and ReVelle [5] in that we find a large number of meteorites with perihelia between 0.8 and 0.9 AU. About 10% of modern falls have perihelia <0.7 AU and about 5% of modern falls apparently struck the Earth prior to reaching thermal equilibrium after evolving from orbits with perihelia >1.0 AU and thus have 2 calculated perihelia >1.0 AU. We observe no apparent direct link between any group of meteorites and individual Q asteroids (Fig. 1). Our data support the conclusions of orbital calculations for Lost City [6] and Innisfree, [7] which suggest that Lost City had a perihelion of ~1.0 AU for at least the last 10^5 years and that Innisfree had a perihelion as low as 0.8 AU within the last 10^5 years. Our data do not support a direct link between Farmington (with a TL-derived perihelion of 0.82) and the Taurid meteoroid complex (perihelion of ~0.4) [1]. We also observe a tendency for meteorites with large cosmic ray exposure ages (>35 Ma) to have shallower perihelia, typically close to 0.8 AU, than those with relatively short cosmic ray exposure ages, which tend to have perihelia between 0.85-1.0 AU. In summary, we find that natural TL data can be used to quantitatively derive the perihelia of meteorite falls. These data are free of potential observational biases (such as those present in, for example, time-of-fall data) and assumptions of orbital parameters (which are a problem for orbits calculated from the radiants of observed falls). Potential applications of these data include investigation of possible meteorite "streams" [2] and exploring questions such as whether different types and classes of meteorites have different orbital distributions [8]. Climate and Streamflow Reconstruction on the São Francisco Basin, Brazil, Using Tree-Ring Data

The São Francisco River crosses the most drought-prone region of Brazil and regional economic dynamics are dependent on the water availability in the basin. The seasonally dry forests are widely distributed in the basin, where Cedrela fissilis Vell (cedro) are frequently found. This semi-arid region provides a favorable setting where the deciduous cedro trees form well-defined semi-ring porous annual rings that can be exactly crossdated and used to build climate sensitive chronologies. Therefore, we have developed chronologies of cedro from seasonally dry forest fragments of three sites located in the middle-sector of the São Francisco River basin and south limit of the Brazilian Drought Polygon. The samples were analyzed according to standard procedures: sample preparation, ring count, crossdating and measurement of the tree rings. Dating quality was tested using the computer program COFECHA and ring-width time series where detrended and standardized to produce the final index chronology using the ARSTAN program. The results show that crossdating within and among trees from different sites demonstrate the potential to expand the spatial sampling. The tree-ring chronologies are sensitive with wet season precipitation totals (October - March), and can explain approximately 40% of the variance (1961-2015). Significant correlation was also observed with total annual discharge of the Rio São Francisco River measured at Barra (r=0.48; 1961-2015). However, the correlation disapears after 1993 (r=0.64 for 1961-1993, but r=-0.004 for 1994-2015) and we suspect that the stream gage at Barra has been impacted by human activity. Tree-ring chronologies can provide important information on climate and streamflow variability of São Francisco River, where hydrological records are often short and discontinuous. This chronology is now being extended with 150-yr old trees from the region and may be used to reconstruct climate and streamflow records back to the pre-instrumental period, in order to help understanding the impact and magnitude of hidroclimatic changes over the third largest Brazilian basin. Infrasonic detection performance in presence of nuisance signal

The infrasound network of the International Monitoring System (IMS) consists of sixty stations deployed all over the World by the Comprehensive Nuclear-Test-Ban Treaty Organization (CTBTO). The IMS has been designed to reliably detect, at least by two stations, an explosion greater than 1 kiloton located anywhere on the Earth [1]. Each station is an array of at least four microbarometers with an aperture of 1 to 3 km. The first important issue is to detect the presence of the signal of interest (SOI) embedded in noise. The detector is commonly based on the property that the SOI provides coherent observations on the sensors but not the noise. The statistic of test, called F-stat [2], [5], [6] , calculated in a time cell a few seconds, is commonly used for this purpose. In this paper, we assume that a coherent source is permanently present arriving from an unknown direction of arrivals (DOA). The typical case is the presence of microbaroms or the presence of wind. This source is seen as a nuisance signal (NS). In [4], [3] authors assume that a time cell without the SOI (CH0) is available, whereas a following time cell is considered as the cell under test (CUT). Therefore the DOA and the SNR of the NS can be estimated. If the signal-to-noise ration SNR of the NS is large enough, the distribution of the F-stat under the absence of SOI is known to be a non central Fisher. It follows that the threshold can be performed from a given value of the FAR. The major drawback to keep the NS is that the NS could hide the SOI, this phenomena is similar to the leakage which is a well-known phenomena in the Fourier analysis. An other approach consists to use the DOA estimate of the NS to mitigate the NS by spatial notch filter in the frequency domain. On this approach a new algorithm is provided. To illustrate, numerical results on synthetical and real data are presented, in term of Receiver Operating Characteristic ROC curves. Mapping biological ideas: Concept maps as knowledge integration tools for evolution education

Many students leave school with a fragmented understanding of biology that does not allow them to connect their ideas to their everyday lives (Wandersee, 1989; Mintzes, Wandersee, & Novak, 1998; Mintzes, Wandersee, & Novak, 2000a). Understanding evolution ideas is seen as central to building an integrated knowledge of biology (Blackwell, Powell, & Dukes, 2003; Thagard & Findlay, 2010). However, the theory of evolution has been found difficult to understand as it incorporates a wide range of ideas from different areas (Bahar et al., 1999; Tsui & Treagust, 2003) and multiple interacting levels (Wilensky & Resnick, 1999; Duncan & Reiser, 2007; Hmelo-Silver et al., 2007). Research suggests that learners can hold a rich repertoire of co-existing alternative ideas of evolution (for example, Bishop & Anderson, 1990; Demastes, Good, & Peebles, 1996; Evans, 2008), especially of human evolution (for example, Nelson, 1986; Sinatra et al., 2003; Poling & Evans, 2004). Evolution ideas are difficult to understand because they often contradict existing alternative ideas (Mayr, 1982; Wolpert, 1994; Evans, 2008). Research suggests that understanding human evolution is a key to evolution education (for example, Blackwell et al., 2003; Besterman & Baggott la Velle, 2007). This dissertation research investigates how different concept mapping forms embedded in a collaborative technology-enhanced learning environment can support students' integration of evolution ideas using case studies of human evolution. Knowledge Integration (KI) (Linn et al., 2000; Linn et al., 2004) is used as the operational framework to explore concept maps as knowledge integration tools to elicit, add, critically distinguish, group, connect, and sort out alternative evolution ideas. Concept maps are a form of node-link diagram for organizing and representing connections between ideas as a semantic network (Novak & Gowin, 1984). This dissertation research describes the iterative development of a novel biology-specific form of concept map, called Knowledge Integration Map (KIM), which aims to help learners connect ideas across levels (for example, genotype and phenotype levels) towards an integrated understanding of evolution. Using a design-based research approach (Brown, 1992; Cobb et al., 2003), three iterative studies were implemented in ethically and economically diverse public high schools classrooms using the web-based inquiry science environment (WISE) (Linn et al., 2003; Linn et al., 2004). Study 1 investigates concept maps as generative assessment tools. Study 1A compares the concept map generation and critique process of biology novices and experts. Findings suggest that concept maps are sensitive to different levels of knowledge integration but require scaffolding and revision. Study 1B investigates the implementation of concept maps as summative assessment tools in a WISE evolution module. Results indicate that concept maps can reveal connections between students' alternative ideas of evolution. Study 2 introduces KIMs as embedded collaborative learning tools. After generating KIMs, student dyads revise KIMs through two different critique activities (comparison against an expert or peer generated KIM). Findings indicate that different critique activities can promote the use of different criteria for critique. Results suggest that the combination of generating and critiquing KIMs can support integrating evolution ideas but can be time-consuming. As time in biology classrooms is limited, study 3 distinguishes the learning effects from either generating or critiquing KIMs as more time efficient embedded learning tools. Findings suggest that critiquing KIMs can be more time efficient than generating KIMs. Using KIMs that include common alternative ideas for critique activities can create genuine opportunities for students to critically reflect on new and existing ideas. Critiquing KIMs can encourage knowledge integration by fostering self-monitoring of students' learning progress, identifying knowledge gaps, and distinguishing alternative evolution ideas. This dissertation research demonstrates that science instruction of complex topics, such as human evolution, can succeed through a combination of scaffolded inquiry activities using dynamic visualizations, explanation activities, and collaborative KIM activities. This research contributes to educational research and practice by describing ways to make KIMs effective and time efficient learning tools for evolution education. Supporting students' building of a more coherent understanding of core ideas of biology can foster their life-long interest and learning of science.
